Meristematic growth and loss of distal tissue from blades of two ecologically important species in the south-east Pacific, Lessonia nigrescens and Lessonia trabeculata, was evaluated during 1 year. Comparative growth was determined by a hole-punch method, loss of distal tissue from the blades was determined by subtracting final blade length (with loss) from expected blade lengths (without loss); growth and tissue loss were transformed to fresh biomass units for calculation of inter-algae differences. The results showed that blade elongation rate increased at the beginning of spring, and declined towards the end of summer, with mean values between 0.40 and 0.08 cm day−1 for L. nigrescens, and 0.65–0.17 cm day−1 for L. trabeculata. Loss of distal tissue varied seasonally when examined as length units for both species; with mean values between 0.24 and 0.10 cm day−1 for L. nigrescens, and 0.51–0.25 cm day−1 for L. trabeculata. Variations in fresh biomass units were only observed in Lessonia trabeculata, increasing in spring, with mean values to 0.13 g (fresh weight) day−1. Annual growth and loss of distal tissue were higher in L. trabeculata (0.41 and 0.39 cm day−1, respectively) than in L nigrescens (0.19 and 0.15 cm day−1). When growth and tissue loss were considered as fresh biomass, monthly gains significantly outweighed loss of distal tissue in both species, but parallel results based on length data followed a different trend. L. trabeculata released about 50% of its growth biomass as particulate organic matter, while the comparative value for L. nigrescens was about 20%.  相似文献

The Valdavara-1 cave, located in the westernmost part of the Iberian Peninsula, is an archaeological site that has been excavated since 2007. At least two main sedimentary units have been identified: an upper one, 30 cm-thick, that has yielded many fragments of ceramic characteristic of the Recent Prehistory (with a radiocarbon age of 4.490 ± 40 years BP), and a lower one, 90 cm-thick, separated from the upper one by an erosive discontinuity and late Pleistocene in age (radiocarbon ages between 13.770 ± 70 and 15.120 ± 70 years BP). The small-vertebrate assemblages recovered from the sieving-washing of all the sediment from the excavation campaigns include a total of at least 34 taxa (six amphibians, nine squamate reptiles, five insectivores, two chiropterans and 12 rodents). The materials from this locality constitutes the first mention in northwestern Spain for Microtus (Iberomys) cabrerae and Micromys minutus, currently absent from this area, as well as the first mention in the fossil record for Chioglossa lusitanica, Discoglossus galganoi and Chalcides striatus. In both layers, such small-vertebrate associations suggest a patchy landscape dominated by humid meadows and woodland areas, with the existence of water areas in the vicinity of the cave. The climate shows a more continental pattern during the late Pleistocene, as evidenced by the presence of Microtus arvalis, Sorex minutus and Rana temporaria, and was milder during the Recent Prehistory, with the occurrence of typically Mediterranean taxa such as Crocidura russula, Chioglossa lusitanica, Discoglossus galganoi, Rana iberica, Chalcides striatus and Timon lepidus. The amphibian and squamate associations suggest that the climate change recorded at the Valdavara-1 cave between the latest Pleistocene and Holocene levels may correspond to a difference of 0.8 °C in terms of mean annual temperature, although the difference is 2.8 °C for the mean temperature of the coldest month.  相似文献

The bimetallic [M1M2(tren)2(CAn?)]m+ series, where M = GaIII or CrIII and CA is the chloranilate ligand which can take on diamagnetic (CAcat,cat)4? or paramagnetic (CAsq,cat)3? forms, comprises an electronically diverse series of compounds ranging from the closed-shell [Ga2(tren)2(CAcat,cat)]2+ to the S = 5/2 ground state of [Cr2(tren)2(CAsq,cat)]3+. This report deals with the interpretation of the EPR and ENDOR spectra of [Ga2(tren)2(CAsq,cat)](BPh4)2(BF4) (2) and the related derivative [Ga2(tren)2(DHBQ)](BPh4)2(BF4) (2a) (where DHBQ is the fully deprotonated trianionic form of 2,5-dihydroxy-1,4-benzoquinone) in an effort to further characterize the electronic structure of this radical species. The X-band (~9.5 GHz) EPR spectrum of complex 2 acquired in a butyronitrile/propionitrile glass at 4 K reveals a rhombic g-tensor with gxx = 2.0100, gyy = 2.0097, and gzz = 2.0060 with hyperfine interactions due to spin delocalization onto the two Ga nuclei (axx = 4.902 G, ayy = 4.124 G, azz = 3.167 G); the origin of the hyperfine coupling was confirmed by analysis of the room temperature spectra of complexes 2 and 2a. The low-temperature spectrum of complex 2 also indicates the presence of a triplet electronic state characterized by a g-value of 2.009 and axial zero-field splitting of D = 150 G (0.012 cm?1) as determined from measurements carried out at both X- and W-band (~95 GHz) frequencies. This triplet state is believed to arise due to a weak intermolecular Heisenberg exchange interaction between two aggregating complexes. ENDOR measurements on complex 2a at 20 K allowed for a determination of the magnitude of hyperfine coupling to the protons associated with the radical bridge as well as providing a rare example of an ENDOR signal arising from coupling to a gallium nucleus. Finally, these results were combined with literature data on the free semiquinone form of the bridging ligand in order to assess the extent to which density functional theory can predict unpaired spin density distribution in a complex molecule of this type. Although differences between theory and experiment were noted, DFT was able to provide a reasonably accurate picture of the electronic structure of this system as well as provide insight into the spin polarization mechanism(s) responsible for the observed hyperfine interactions.  相似文献

Numerous taxa show ancient intertropical disjunct distributions. Many can be explained by well-known processes of historical vicariance, such as the breakup of Gondwanaland. Others, such as Asian–Neotropical divergences are not as well understood. To clarify the phylogenetic position and understand biogeographic and temporal origins of the geographically disjunct and morphologically unique genera of hinged-teeth snakes, Scaphiodontophis (n = 1) and Sibynophis (n = 9; Colubridae), we inferred a time-calibrated phylogeny with additional 107 taxa representing the superfamily Colubroidea using four genes (c-mos, cyt-b, ND2, RAG-1; 3085 bp). We used this tree to estimate ancestral areas for the group. The results show that Scaphiodontophis is sister to Sibynophis, both originated in the late Eocene/Oligocene in Asia and likely dispersed through Beringia to the New World, but unlike other snake groups left no extant species in temperate North America. Current recognition of Scaphiodontophiinae renders Colubrinae paraphyletic, and we resurrect the previously named subfamily Sibynophiinae to encompass both genera and use the tribes Sibynophiini (Sibynophis) and Scaphiodontophiini (Scaphiodontophis) to highlight the geographically distinct areas occupied by these taxa. These results suggest that intercontinental dispersal with extinction in intermediate areas can explain puzzling patterns of ancient intertropical disjunct distributions.  相似文献

PurposeAngiogenesis, a multistep process that results in new blood vessel formation from preexisting vasculature is essential for both the growth of solid tumour and for metastasis. Stimulation of vascular endothelial growth factor receptor (VEGFR), a transmembrane glycoprotein, results in mitogenesis. Within this family of receptors, VEGFR 2/kinase-insert-domain containing receptor appears to be principally upregulated during tumorigenesis. The aim of this study was to determine the expression of VEGFR-2/kinase-insert-domain containing receptor (KDR) and its correlation with angiotensin receptor type 1 (AT1-R) and clinical factors in endometrial carcinoma.MethodsThe expression of KDR and AT1-R was studied in endometrial carcinoma and normal endometrium by Real-time RT-PCR and Western blot analysis in 136 samples. The expression profile was correlated with the clinicopathological characteristics of endometrial adenocarcinoma.ResultsWe noted a significant correlation between the expression of KDR and AT1-R in tumour grade G1, G2 and G3 (Rs = 0.50; p = 0.002, Rs = 0.69; p = 0.0001, Rs = 0.52; p = 0.005, respectively). In stage I and stage II carcinoma, a significant correlation was also found between the expression of KDR and AT1-R (Rs = 0.70, p = 0.0001, Rs = 0.67; p = 0.001, respectively). Moreover significant correlation was observed between both KDR and AT1-R in tissue with different myometrial invasion (Rs = 0.54, p = 0.0001, Rs = 0.68; p = 0.0001; respectively for tumours with invasion into the inner half and invasion into the outer half).ConclusionsBasing on received correlation between AT1-R and KDR expression and previous results we speculate that angiotensin through AT1-R modulates KDR expression and thus have influence on local VEGF level. However, further studies are required to clarify the biological interaction between KDR, AT1-R and other hormonal regulators in endometrial carcinoma.  相似文献

Little is known about how the growth of individual Gambierdiscus species responds to environmental factors. This study examined the effects of temperature (15–34 °C), salinity (15–41) and irradiance (2–664 μmol photons m−2 s−1) on growth of Gambierdiscus: G. australes, G. belizeanus, G. caribaeus, G. carolinianus, G. carpenteri, G. pacificus and G. ruetzleri and one putative new species, Gambierdiscus ribotype 2. Depending on species, temperatures where maximum growth occurred varied between 26.5 and 31.1 °C. The upper and lower thermal limits for all species were between 31–34 °C and 15–21 °C, respectively. The shapes of the temperature vs. growth curves indicated that even small differences of 1–2 °C notably affected growth potentials. Salinities where maximum growth occurred varied between 24.7 and 35, while the lowest salinities supporting growth ranged from <14 to 20.9. These data indicated that Gambierdiscus species are more tolerant of lower salinities than is generally appreciated. Growth of all species began to decline markedly as salinities exceed 35.1–39.4. The highest salinity tested in this study (41), however, was lethal to only one species, Gambierdiscus ribotype 2. The combined salinity data indicated that differences in salinity regimes may affect relative species abundances and distributions, particularly when salinities are <20 and >35. All eight Gambierdiscus species were adapted to relatively low light conditions, exhibiting growth maxima at 50–230 μmol photons m−2 s−1 and requiring only 6–17 μmol photons m−2 s−1 to maintain growth. These low light requirements indicate that Gambierdiscus growth can occur up to 150 m depth in tropical waters, with optimal light regimes often extending to 75 m. The combined temperature, salinity and light requirements of Gambierdiscus can be used to define latitudinal ranges and species-specific habitats, as well as to inform predictive models.  相似文献

Tree-ring research in Ireland has typically been dominated by Quercus species, particularly Quercus petraea and Quercus robur. Recent years have seen a greater focus on multi-species reconstructions in Ireland but, due to difficulties with the hardness of the wood, missing/pinched rings and fused stems, Taxus baccata has not been included in these investigations. Despite these difficulties, a 31-tree, 204-year T. baccata chronology was successfully constructed from Killarney National Park, southwest Ireland. The chronology exhibits promising dendroclimatological potential, with climatic responsiveness equivalent to that of the other major Irish tree taxa, including Quercus. The chronology shows the strongest relationship with May–June precipitation from Muckross House synoptic station (1970–2007; r = 0.521, p < 0.01) and Valentia Observatory (1941–2007; r = 0.545, p < 0.01). November–April temperatures also exhibited a strong relationship with the chronology post-1970 (r = 0.605, p < 0.01 for Muckross House, r = 0.567, p < 0.01 for Valentia Observatory), but this relationship is not time stable and breaks down for the pre-1970 Valentia Observatory record. The long-lived nature of T. baccata, the exceptional preservation of wood and rings in this hard softwood species, as well as its prominence in Irish archaeology, all point to the potential to expand this chronology both spatially and temporally, and demonstrate T. baccata's potential in multi-site and multi-species tree-ring studies in the region.  相似文献

Paleoceanographic variability at southern high latitude Ocean Drilling Program (ODP) Site 747 was investigated in this study through the interval which spans the Middle Miocene Climate Transition (MMCT). Between 15.0 and 12.2 million years ago (Ma), foraminiferal δ18O records derived from both benthic (Cibicidoides spp.) and planktonic taxa (Globorotalia praescitula and Globigerina bulloides) reveal a history of changes in water column thermal and salinity structure and a strong imprint of seasonality. Prior to the MMCT, in the interval between 14.35 and 13.9 Ma, G. bulloides displays relatively high δ18O values similar to those of G. praescitula, interpreted to indicate weakening of the thermocline and/or increased seasonality with cooler early-spring and/or late-fall temperatures. Following this interval, G. bulloides δ18O values diverge significantly from benthic and G. praescitula values, with G. bulloides values remaining relatively low for at least 600 kyr following the benthic foraminiferal δ18O shift during the MMCT at ~ 13.9 Ma. This divergence in δ18O records occurs in direct association with the Mi3 cooling and glaciation event and may suggest: (1) a strengthening of the vertical temperature gradient, with greater cooling of deep waters than surface waters, (2) changes in the depth habitat of G. bulloides, (3) changes in the dominant season of G. bulloides calcification, (4) modification of surface-water δ18O values in association with enhanced sea-ice formation, (5) increased surface-water carbonate ion concentration, and/or (6) a significant decrease in surface-water salinity across the MMCT. The first of these possible scenarios is not likely, particularly in light of recent Mg/Ca evidence for significant surface-water cooling in the Southern Ocean associated with the MMCT. Of the remaining possibilities, we favor a change in surface salinity to explain the observed trends in δ18O values and hypothesize that surface salinity may have decreased by up to 2 salinity units at ~ 13.9 Ma. In this scenario, the development of a lower-salinity Antarctic surface layer coincided with regional cooling of both surface and deep waters of the Southern Ocean during the Mi3 glaciation of East Antarctica, and contributed into the dominance of Neogloboquadrina spp. between 13.8 and 13.2 Ma. Additionally, the distinct patterns observed in planktonic foraminiferal δ18O records spanning the MMCT correspond with changes in the vertical δ13C gradient between planktonic and benthic foraminiferal records and major changes in planktonic foraminiferal assemblages at Site 747, providing further evidence of the environmental significance of this climatic transition.  相似文献

Crepis capillaris, Hieracium pilosella, and Hypochaeris radicata were investigated for the influence of the altitude of the collection site on the content of phenolics within the flowering heads. These three taxa from the Lactuceae tribe of the Asteraceae family originate from Europe and are now widespread within New Zealand. Flowering heads collected from different altitudes ranging from 180 m to 1060 m (C. capillaris), from 190 to 1290 m (H. pilosella), and from 20 m to 1290 m (H. radicata), respectively, were extracted and analysed by high performance liquid chromatography. Results showed a positive correlation between the altitude of the growing site and the contents of flavonoids and phenolic acids for all investigated taxa. The altitudinal effect was, however, partially concealed by geographic differences between coastal and inland collection sites, with the inland collections containing higher concentrations of flavonoids and phenolic acids than plants collected from the coast. The results are discussed in the light of a putative UV-B protective function of the quantified compounds and of the immigration histories of the three species at hand.  相似文献

The alkaloid rich extracts from an acid/base extraction of bulb material of Haemanthus coccineus L., H. montanus Baker and H. sanguineus Jacq. revealed that two montanine type Amaryllidaceae alkaloids, montanine (1) and coccinine (2) were the major alkaloid constituents. Together these two alkaloids constituted 88, 91 and 98% of the total alkaloid extract from each species respectively. GC–MS analysis revealed that H. coccineus and H. sanguineus had a relative abundance of coccinine (74 and 91% respectively) to montanine (14 and 7% respectively); whereas H. montanus had 20% coccinine and 71% montanine. The three extracts and two isolated alkaloids were evaluated for binding to the serotonin transporter protein (SERT) in vitro. Affinity to SERT was highest in H. coccineus (IC50 = 2.0 ± 1.1 μg/ml) followed by H. montanus (IC50 = 6.8 ± 1.0 μg/ml) and H. sanguineus (IC50 = 28.7 ± 1.1 μg/ml). Montanine (IC50 = 121.3 ± 3.6 μM or 36.56 ± 1.14 μg/ml; Ki = 66.01 μM) was more active than coccinine (IC50 = 196.3 ± 3.8 μM or 59.15 ± 1.08 μg/ml; Ki = 106.8 μM), both of which were less active than the total alkaloid extracts of each species investigated. The possible synergistic effects of two coccinine/montanine mixtures (80:20 and 20:80) were investigated, however the mixtures gave similar activities as the pure compounds and did not show any increase in activity or activity similar to the total alkaloid extracts. Thus the considerably higher activity observed in the total alkaloid extracts is not correlated to the relative proportions of coccinine and montanine in the extracts and thus are likely to be due to more potent unidentified minor constituents. Both alkaloids exhibited low binding affinity to P-glycoprotein (P-gp) as demonstrated by low inhibition of calcein-AM efflux in the MDCK-MDR1 cell line. This indicates that P-gp efflux will not be limiting for blood–brain-barrier passage of the alkaloids.  相似文献

The purpose of this study was to examine whether muscle architecture of the long head of biceps femoris (BF) and semitendinosus (ST) muscles varies along their length. The ST and BF muscles were dissected and removed from their origins in eight cadaveric specimens (age range 67.8–73.4 years). One-way analysis of variance designs were used to compare fascicle length (FL), pennation angle (PA) and muscle thickness (MT) between proximal, mid-belly and distal positions. Tendon and muscle length properties were also quantified. For the BF muscle, one-way analysis of variance tests showed a higher PA (23.96 ± 3.82°) and FL (7.12 ± 0.48 cm) proximally than distal positions (PA = 17.78 ± 1.95° and FL = 6.35 ± 0.89 cm, respectively). For the ST, there was a significantly (p < 0.05) lower PA (8.81 ± 1.22°) and FL (13.10 ± 1.54 cm) proximally than distally (PA = 14.69 ± 1.09° and FL = 15.49 ± 2.30 cm, respectively). Muscle thickness significantly increased from distal to more proximal positions (p < 0.05). These data suggest that the ST and BF architecture is not uniform and that measurement of these parameters largely depends on the measurement site. Modeling these muscles by assuming a uniform architecture along muscle length may yield less accurate representation of human hamstring muscle function.  相似文献

Vallisneria spinulosa is a dominant submerged macrophyte in lakes of the middle–lower reaches of the Yangtze River. Allozyme variation, clonal diversity and population genetic structure were investigated for a total of 396 individuals sampled from 10 extant populations. V. spinulosa maintained high levels of genetic variation both at the species (P = 46.2, A = 1.69, He = 0.23) and at the population level (P = 46.2, A = 1.58, He = 0.21). Although aquatic macrophytes commonly exhibit low genetic variation within populations, the obligately outcrossing mating system of V. spinulosa and pervasive gene flow likely account for the high levels of diversity maintained within populations. All V. spinulosa populations contained high clonal diversity with a mean proportion of distinguishable genotypes of 0.57 and a mean Simpson's diversity index of 0.95, indicating that populations were founded sexually or that successful seedling recruitment occurred after initial colonization. Partitioning of genetic diversity revealed a surprisingly low population differentiation (GST = 0.06) as compared to other hydrophilous angiosperms. No evidence of isolation-by-distance was found (r = 0.056, P = 0.312), suggesting that gene flow was not restricted geographically. The UPGMA cluster analysis revealed that several widely separated populations grouped together, suggesting long-distance gene flow among populations. The high vagility of V. spinulosa and extensive hydrologic connectivity among populations have facilitated long-distance gene flow and resulted in the pattern of population genetic structure in V. spinulosa.  相似文献

Out of some isolated Heterorhabditis bacteriophora from Korea, ecological study on two isolates which had different geographical features was investigated. That is, effects of temperature and dose on the pathogenicity and reproduction of two Korean isolates of H. bacteriophora were investigated using Galleria mellonella larvae in the laboratory. The median lethal dose (LD50) decreased with increasing temperature, but increased at 35 °C. The optimal temperatures for infection were 30 °C for H. bacteriophora Jeju strain and 24 °C for H. bacteriophora Hamyang strain. The median lethal time, LT50 of H. bacteriophora Hamyang strain was recorded at 13 °C to 35 °C and that of H. bacteriophora Jeju strain was recorded at 18 °C to 30 °C. The number of established nematodes in G. mellonella larvae was significantly different depending on temperature and dose. When G. mellonella larvae were exposed to 300 infective juveniles (IJs), mortality of G. mellonella gradually increased with exposure time with H. bacteriophora Jeju strain but not with H. bacteriophora Hamyang strain. 87.5% mortality of G. mellonella was recorded by H. bacteriophora Hamyang strain after 1440 min whereas 100% mortality was recorded by H. bacteriophora Jeju strain after 4320 min. The time from infection to the first emergence of nematodes decreased with increasing temperature. Duration of emergence of the two strains in the White traps also decreased with increasing temperature. The highest progeny numbers of H. bacteriophora Jeju strain were 264,602 while those of H. bacteriophora Hamyang strain were 275,744 at the rate of 160 IJs at 24 °C.  相似文献

The phylogeny of the family Sciaridae is reconstructed, based on maximum likelihood, maximum parsimony, and Bayesian analyses of 4809 bp from two mitochondrial (COI and 16S) and two nuclear (18S and 28S) genes for 100 taxa including the outgroup taxa. According to the present phylogenetic analyses, Sciaridae comprise three subfamilies and two genus groups: Sciarinae, Chaetosciara group, Cratyninae, and Pseudolycoriella group + Megalosphyinae. Our molecular results are largely congruent with one of the former hypotheses based on morphological data with respect to the monophyly of genera and subfamilies (Sciarinae, Megalosphyinae, and part of postulated “new subfamily”); however, the subfamily Cratyninae is shown to be polyphyletic, and the genera Bradysia, Corynoptera, Leptosciarella, Lycoriella, and Phytosciara are also recognized as non-monophyletic groups. While the ancestral larval habitat state of the family Sciaridae, based on Bayesian inference, is dead plant material (plant litter + rotten wood), the common ancestors of Phytosciara and Bradysia are inferred to living plants habitat. Therefore, shifts in larval habitats from dead plant material to living plants may have occurred within the Sciaridae at least once. Based on the results, we discuss phylogenetic relationships within the family, and present an evolutionary scenario of development of larval habitats.  相似文献

The isolation and culturing of toxic Gambierdiscus species is desirable for obtaining reliable sources of toxins responsible for Ciguatera Fish Poisoning (CFP) in order to provide reference materials and purified standards for chemical analyses and bioassays. Gambierdiscus polynesiensis CAWD212, G. pacificus CAWD213 and G. australes CAWD149 and 216 have been isolated from samples of sea water from the Cook Islands. G. polynesiensis produced ciguatoxins (CTX; 18.2 pg per cell) and G. australes CAWD149 produced maitotoxin (MTX; 8.3 pg per cell) and in both extracts putative MTX-3 was detected. G. pacificus also produced putative MTX-3. The toxicities of extracts of G. polynesiensis and G. pacificus were similar by i.p. injection (LD50s of 1.0 mg/kg and 0.8 mg/kg respectively), but when administered by gavage, the MTX-3 analogue producing G. pacificus (LD50 251 mg/kg) was 30-times less toxic than the CTX-producing G. polynesiensis (LD50 7.9 mg/kg). Consumption of fish contaminated by CFP has caused illness and even death in the Cook Islands and throughout the Pacific region and is now considered a globally neglected disease. Large-scale cultures of these organisms are now available, and isolation of the toxins they produce is in progress.  相似文献

Secoisolariciresinol diglucosides (SDGs) (S,S)-SDG-1 (major isomer in flaxseed) and (R,R)-SDG-2 (minor isomer in flaxseed) were synthesized from vanillin via secoisolariciresinol (6) and glucosyl donor 7 through a concise route that involved chromatographic separation of diastereomeric diglucoside derivatives (S,S)-8 and (R,R)-9. Synthetic (S,S)-SDG-1 and (R,R)-SDG-2 exhibited potent antioxidant properties (EC50 = 292.17 ± 27.71 μM and 331.94 ± 21.21 μM, respectively), which compared well with that of natural (S,S)-SDG-1 (EC50 = 275.24 ± 13.15 μM). These values are significantly lower than those of ascorbic acid (EC50 = 1129.32 ± 88.79 μM) and α-tocopherol (EC50 = 944.62 ± 148.00 μM). Compounds (S,S)-SDG-1 and (R,R)-SDG-2 also demonstrated powerful scavenging activities against hydroxyl [natural (S,S)-SDG-1: 3.68 ± 0.27; synthetic (S,S)-SDG-1: 2.09 ± 0.16; synthetic (R,R)-SDG-2: 1.96 ± 0.27], peroxyl [natural (S,S)-SDG-1: 2.55 ± 0.11; synthetic (S,S)-SDG-1: 2.20 ± 0.10; synthetic (R,R)-SDG-2: 3.03 ± 0.04] and DPPH [natural (S,S)-SDG-1: EC50 = 83.94 ± 2.80 μM; synthetic (S,S)-SDG-1: EC50 = 157.54 ± 21.30 μM; synthetic (R,R)-SDG-2: EC50 = 123.63 ± 8.67 μM] radicals. These results confirm previous studies with naturally occurring (S,S)-SDG-1 and establish both (S,S)-SDG-1 and (R,R)-SDG-2 as potent antioxidants and free radical scavengers for potential in vivo use.  相似文献

Senegal has since 2003 used sulphadoxine-pyrimethamine (SP) for Intermittent Preventive Treatment (IPT) of malaria in risk groups. However, the large-scale IPT strategy may result in increasing drug resistance. Our study investigated the possible impact of SP-IPT given to infants and children on the prevalence of SP-resistant haplotypes in the Plasmodium falciparum genes Pfdhfr and Pfdhps, comparing sites with and without IPTi/c. P. falciparum positives samples (n = 352) were collected from children under 5 years of age during two cross-sectional surveys in 2010 and 2011 in three health districts (two on IPTi/c and one without IPTi/c intervention) located in the southern part of Senegal. The prevalence of SP-resistance-related haplotypes in Pfdhfr and Pfdhps was determined by nested PCR followed by sequence-specific oligonucleotide probe (SSOP)–ELISA. The prevalence of the Pfdhfr double mutant haplotypes (CNRN and CICN) was stable between years at < 10% in the control group (P = 0.69), while it rose significantly in the IPTi/c group from 2% in 2010 to 20% in 2011 (P = 0.008). The prevalence of the Pfdhfr triple mutant haplotype (CIRN) increased in both groups, but only significantly in the IPTi/c group from 41% to 65% in 2011 (P = 0.005). Conversely, the Pfdhps 437G mutation decreased in both groups from 44.6% to 28.6% (P = 0.07) and from 66.7% to 47.5% (P = 0.02) between 2010 and 2011 in the control and the IPTi/c groups, respectively. Combined with Pfdhfr, there was a weak trend for decreasing prevalence of quadruple mutants (triple Pfdhfr + Pfdhps 437G) in both groups (P = 0.15 and P = 0.34). During the two cross-sectional surveys, some significant changes were observed in the SP-resistance-related genes. However, since these changes were observed in the two groups, the IPTi/c strategy does only seem to have limited impact on resistance development and other factors as well. However, continuous monitoring will be needed, due to the up-scaling of the IPTi/c strategy in Senegal according to WHO recommendations.  相似文献

It is known that arsenate and phosphate (P) share the same transporters in plants, and arbuscular mycorrhizal fungi (AMF) influence the expressions of Pi transporters (OsPT1–13) in rice. In order to study the effects of AMF on arsenate accumulation in rice, Glomus intraradices (AH01) was inoculated to rice and treated with different levels of arsenate (0, 2 and 8 μM). Results revealed that OsPT11 was increased whereas OsPT2 decreased (P < 0.05) in mycorrhizal plants. The increased expression of OsPT11 was one of the most important factors that led to the significantly higher P concentration (P < 0.05) in plant tissues, which compensated the down-regulation of OsPT2. The symbiosis of G. intraradices with rice slightly decreased (P > 0.05) the arsenate concentration in plant tissues but markedly enhanced (P < 0.05) plant biomass. The higher P content in mycorrhizal plants led to the higher P/As molar ratio (P < 0.05) and lower As uptake ratio (P < 0.05) in mycorrhizal plants treated by 2 μM arsenate. Mycorrhizal plants under such an arsenate treatment took up less As by per unit of root dry mass. The inoculation of G. intraradices was not able to transform the inorganic As to organic As. Further studies should be conducted focusing on the transport activities of each Pi transporter using yeast or oocyte expression system to identify which Pi transporters are responsible for the accumulation of arsenate.  相似文献

Two new ectocarpic arbuscular mycorrhizal fungal species, Glomus drummondii and G. walkeri (Glomeromycota), found in maritime sand dunes of northern Poland and those adjacent to the Mediterranean Sea are described and illustrated. Mature spores of G. drummondii are pastel yellow to maize yellow, globose to subglobose, (58–)71(–85) μm diam, or ovoid, 50–80 × 63–98 μm. Their wall consists of three layers: an evanescent, hyaline, short-lived outermost layer, a laminate, smooth, pastel yellow to maize yellow middle layer, and a flexible, smooth, hyaline innermost layer. Spores of G. walkeri are white to pale yellow, globose to subglobose, (55–)81(–95) μm diam, or ovoid, 60–90 × 75–115 μm, and have a spore wall composed of three layers: a semi-permanent, hyaline outermost layer, a laminate, smooth, white to pale yellow middle layer, and a flexible, smooth, hyaline innermost layer. In Melzer's reagent, only the inner- and outermost layers stain reddish white to greyish rose in G. drummondii and G. walkeri, respectively. Both species form vesicular–arbuscular mycorrhizae in one-species cultures with Plantago lanceolata as the host plant. Phylogenetic analyses of the ITS and parts of the LSU of the nrDNA of spores placed both species in Glomus Group B sensu Schüßler et al. [Schüßler A, Schwarzott D, Walker C, 2001.
