The aim of the experiment was to evaluate the effects of GnRH and/or eCG and progestin type (implant versus CIDR) on the induction of estrus and pregnancy rate following laparoscopic AI (LAI) with frozen semen. In the first trial, ewes (n = 129) were treated with norgestomet implants for 14 days. At implant removal ewes received eCG (400 IU) and/or GnRH (25 μg) 36 h after removal, resulting in control, eCG, GnRH, and eCG/GnRH groups (n = 30–34/group). In trial 2, ewes (n = 36) were treated with intravaginal fluorogestone acetate sponges (FGA) or CIDR for 12 days. After withdrawal, half of the ewes from each progestin group received eCG (400 IU), resulting in sponge, sponge/eCG, CIDR and CIDR/eCG groups (n = 8–10/group). In both trials, estrous activity was assessed using a vasectomized ram from the time of progestin removal to laparoscopic AI with frozen semen 58–60 h (trial 1) or 54–56 h (trial 2) following cessation of treatment. In trial 1, GnRH decreased (P < 0.05) the percentage of ewes in estrus (GnRH, 75.8% versus control, 93.8% versus eCG/GnRH, 94.1%), however pregnancy rates were similar in all groups (control, 53.1%; eCG, 70.0%; GnRH, 51.5%; eCG/GnRH, 55.9%, respectively). In trial 2, neither the type of progestin nor eCG treatment effected the percentage of ewes in estrus (sponge, 75.0%; sponge/eCG, 100.0%; CIDR, 100.0%; CIDR/eCG, 90.0%). However, pregnancy rates following LAI were higher (P < 0.05) when ewes were treated with eCG (progestin + eCG, 73.7% versus progestin alone, 41.2%). Results demonstrate that the source of progestin does not influence the expression of estrus or the proportion of ewes pregnant following LAI. When progestin treatment protocols are used in combination with eCG, pregnancy rates can be increased. A dose of GnRH near the end of progestin treatment may decrease the estrous response, by inducing ovulation before normal expression of estrus.  相似文献

The purpose of the study was to investigate the effects of different doses of equine chorionic gonadotropin (eCG) treatment on follicular development, ovulation and pregnancy rate during the breeding season in fat-tailed Chall ewes. Seventy-two cycling (62.5 ± 2.5 kg), multiparous Iranian Chall ewes were used in the trial. The ewes were randomly allocated to 6 groups (n = 12/group). Estrus was synchronized with the aid of controlled intravaginal drug release (CIDR) devices, inserted for 14 days. At the time of CIDR removal (day 14), the ewes received i.m. either 0 (control group, G0), 450 (G450), 550 (G550), 650 (G650), 750 (G750) or 850 (G850) IU eCG. Vasectomized rams were used to detect estrus in the ewes from 24 h after CIDR removal. Ovarian follicular activity was monitored with the aid of transrectal ultrasonography on the day of CIDR insertion (day 0) and daily from the day of eCG treatment (day 14), until estrus (day 16). During these days, blood samples were collected for the determination of plasma progesterone and estradiol concentrations. Laparoscopic intrauterine inseminations were conducted 54–60 h after CIDR removal. The number of CL's and pregnancy diagnosis was recorded using ultrasonography 7 and 54 days following AI, respectively. Half of ewes in control group and most of the ewes treated with eCG showed signs of estrus within 36 h of CIDR removal. The ewes in groups G750 and G850 recorded the highest number of large follicles at estrus and CL's 7 days later. The pregnancy rate in groups G550 (75.0%) and G650 (75.0%) was higher (P < 0.05) than that in the other groups. The ovarian response and estradiol concentration, as well as pregnancy rate showed that 550 or 650 IU eCG treatment is the most effective doses in improving the pregnancy rate in Iranian Chall ewes.  相似文献

The aim of this experiment was to investigate the effects of increased group size on eating- and resting behaviour, aggression and feed intake in housed ewes. During an initial period of 14 days 36 adult (2–6 years old) ewes of the domestic Norwegian Dala breed were divided into four groups of 9. In the second period (14 days), these ewes were merged into one group of 36 ewes. This experiment was repeated with a second batch of ewes, but this time starting with a group of 36 individuals in the first period, then splitting them up into four groups of 9 ewes in the second period. From 24 h video recordings we scored activity behaviours using instantaneous sampling every 10 min. Aggressive interactions were continuously observed the first 10 min every hour during the 24 h (4 h in total). A mixed statistical procedure with group size, day, batch and the interactions between them were included as fixed effects, whereas individual and group were specified as random effects.Ewes in large groups (36) had a larger variation in lying time at day one (P < 0.01), less synchronized lying (P < 0.05) and eating behaviour (P < 0.01), and spent less time queuing at the feed barrier (P < 0.001) compared to in the small group size (9). There were no effects of group size on aggressive interactions or feed intake.In conclusion, a larger group size decreased synchrony in resting and feeding behaviour and reduced the time spent queuing in front of the feed barrier. It is possible that the aggression level in sheep is more sensitive to changes in space allowance than to changes in group size per se.  相似文献

The fertility obtained in sheep after the use of intravaginal progesterone devices is related to the content of progesterone of the device. The hypothesis of this study was that the reproductive response of anoestrous ewes to the ram-effect could be improved by the administration of oestradiol-17β in conjunction with CIDRs treatment—using previously used CIDRs in a 5-day progestagen priming. Therefore, the objective was to determine if oestradiol-17β treatment increases fertility of anoestrous ewes primed with used CIDRs and stimulated by the ram-effect. The hypothesis was tested with CIDRs that had been previously used for 12 or 18 days. The trial was performed during the non-breeding season using 158 Corriedale ewes. Ewes had been isolated from rams since Day −35 (Day 0 = introduction of the rams). A CIDR (0.3 g progesterone, InterAg, Hamilton, New Zealand) was inserted on Day −5 in all ewes with CIDR that had been previously used for 12 days (n = 62) or 18 days (n = 96). Also on Day −5, 29 and 53 ewes that had received CIDRs of 12 or 18 days, respectively, received an intra-muscular treatment of 50 μg of oestradiol-17β (E groups). The ewes that did not receive the oestradiol-17β treatment remained as the control group (C group). Overall the treatment groups were thus: C12 (n = 33), C18 (n = 43), E12 (n = 29), and E18 (n = 53). On Day 0 all CIDRs were withdrawn, and ewes were placed with 18 rams and 20 ewes hormonally induced to exhibit oestrus. Sexual receptivity of ewes treated with CIDRs was estimated from marks on the rumps of the ewes daily from Day 0 to Day 5, and the pregnancy status diagnosed with transrectal ultrasonography on Day 40. The percentage of ewes exhibiting oestrus and pregnancy rates were lower in ewes synchronized with previously used CIDRs for 18 days, compared to those used for 12 days. The responses of ewes in oestrus were 39.4, 14.0, 65.5, and 32.1% for the C12, C18, E12, and E18 groups respectively, with pregnancy rates of 30.3, 14.0, 34.5, and 17.0%. Administration of oestradiol-17β increased the frequency of oestrous response in ewes that were treated with CIDRs previously used for 12 days (P < 0.05), but not in those treated with CIDRs used for 18 days. It could be concluded that the administration of oestradiol-17β only improved the percentage of ewes responding to oestrus when CIDRs previously used for 12 days were used for 5 days before the introduction of rams. No positive effect on fertility was observed irrespective of the period during which CIDR had been previously used.  相似文献

An experiment was conducted to examine the changes in colostral production and composition in Karakul ewes fed at different nutritional levels. Pregnant ewes were randomly allotted to one of four groups (n = 20 per group) 90 days after artificial insemination. Three groups were fed with total mixed rations supplying 90, 100 and 110% of the NRC requirements, respectively, and the last group served as the control feeding on natural vegetations until parturition. Blood samples were taken from the jugular vein during late pregnancy, and soon after lambing. Fifteen singleton ewes from each group were hand-milked completely at 1, 12 and 24 h post-lambing. Samples of colostrum were obtained and the lambs were fed with their ewe's colostrum (30 g/kg BW) at each milking. Blood samples were taken from the lambs before receiving their first colostrum, and again at 24 and 48 h post-lambing. Ewes on 110% NRC diet produced significantly (P < 0.01) more colostrum than did the ewes in other groups. Feeding level did not have a significant effect on the IgG level of the colostrums and blood sera of ewes and lambs. Mean IgG concentration in the ewe serum was greatest during late gestation (1–2 months before lambing) and were significantly reduced at 1 h post-lambing. Non-immunoglobulin composition of colostrum was not significantly affected by the treatment. There was a significant correlation between IgG levels of the ewe serum and colostrum (r = 0.64, P < 0.0001). Mean weight of the ewes on the control ration at lambing and the mean birth weight of their lambs were significantly lower as compared with other dietary groups. The results suggested that hand-feeding of Karakul ewes on poor rangeland vegetation with 110% NRC for 2 months before parturition increased colostral production, decreased the ewe loss and increased lamb birth weight which is beneficial to lamb survival.  相似文献

To examine the effects of maternal supranutritional selenium (Se) and nutrient restriction during mid and late gestation on placental characteristics and fetal liver glycogen, ewes received either adequate Se (ASe) or high Se (HSe) prior to breeding. On d 64 of gestation, ASe and HSe ewes remained at 100% of requirements (controls; CON) or were restricted (RES; 60% of requirements). On d 135 of gestation, fetal weight (P  0.08) was greatest in both HSe and CON ewes. Placentome number, mass, and caruncular and cotyledonary weight were not different (P  0.17) among treatments. Fetal mass:placental mass ratio was less (P = 0.06) in RES compared to CON ewes. Compared to ASe, HSe exhibited increased (P  0.08) cellular proliferation and DNA concentration and decreased (P = 0.07) cellular size in cotyledonary tissue. Nutritional restriction decreased (P  0.08) cotyledonary protein concentration and cellular size. VEGF receptor 1 (Flt) mRNA in cotyledonary tissue was greater in HSe compared with ASe ewes (P = 0.06) and in RES compared with CON ewes (P = 0.08). There was no effect of diet on caruncular growth variables (P  0.13) or on placental vascularity (P  0.11). Progesterone was greater (P  0.08) in ASe–RES ewes compared to all groups at d 90 and ASe–CON and HSe–CON at d 104. Although fetal glucose and cortisol concentrations were not affected by diet, fetal liver glycogen was greater (P = 0.04) in ASe–RES compared to ASe–CON and HSe–RES ewes with HSe–CON being intermediate. Both Se and nutritional plane may impact placental function and fetal growth, as fetal weight and liver glycogen are altered despite similar placental vascularity measurements.  相似文献

The objective of this study was to evaluate the effect of crossbreeding Awassi ewes with either Charollais or Romanov sires, on pre-weaning lamb production as reflected by reproductive performance and milk production in the ewes. Two hundred and five, 3- to 6-year-old multiparous ewes of three breeds were allocated to three groups [Awassi (A; n = 56), F1 Romanov × Awassi (RA; n = 78) and F1 Charollais × Awassi (CA; n = 71)]. Upon lambing, ewes and their offspring were placed in a large pen in which they remained until the end of the trial. Body weight (BW) and body condition score (BCS) of the ewes and the BW of the lambs were recorded weekly from birth to weaning at 70 days of age. Milk production was recorded weekly from parturition to weaning. Pregnancy rates were not influenced by breed-type. Multiple births and the fecundity of ewes were greater (P < 0.05) in the RA group, compared to the A and CA groups—while the weaning percentage was similar between the breeds. Lamb birth and weaning weights were similar, while the kg lamb born per kg ewe lambed was greater (P < 0.05) in the RA group, compared to the CA group. The CA ewes recorded a higher BW (P < 0.01) and BCS (P < 0.01) than the other breed groups. Awassi ewes produced more milk (P < 0.01) throughout the observation period. Milk ash percentage was higher (P < 0.05) in the CA group, while the percentage of crude protein and dry matter was similar for the different groups. Results of the present study indicate that crossbreeding of Awassi ewes with Charollais and Romanov breeds decreased milk production in the progeny, without affecting lamb growth. The crossbreeding of Awassi with Charollais resulted in improved BW and BCS, while the crossbreeding of Awassi ewes with Romanov resulted in improved reproductive performance of the crossbreds. Crossbreeding Awassi ewes with exotic rams can thus be conducted to increase the number of lambs produced per ewe.  相似文献

In the current study follicular dynamics, pituitary function, ovulatory response and luteal activity of 30 maiden Barbarine sheep were analyzed according to oestrus occurrence and lambing outcome after oestrus synchronisation with cloprostenol. Animals were retrospectively classified in three groups named as O? (n = 7, ewes not displaying oestrus), O+L? (n = 7, ewes showing oestrus but failing to lamb) and O+L+ (n = 16; ewes showing oestrus and lambing thereafter). All the sheep ovulated and daily transrectal ultrasonographies revealed that preovulatory follicles were present at cloprostenol injection in all the animals. In sheep O+L+ and O+L?, 50% and 57% of the ovulatory follicles were the largest follicles at cloprostenol treatment (mean size of 4.1 ± 0.26 mm and 4.3 ± 0.74 mm, respectively). In O? ewes, the same percentage was higher (86%, P < 0.05 when compared to group O+L+; mean size of 4.0 ± 0.46 mm). The number of large follicles and the final diameter of the ovulatory follicles at oestrous tended thereafter to be higher in group O+L+ (1.4 ± 0.1 and 6.4 ± 0.2) than in groups O+L? (1 ± 0.2 and 5.7 ± 0.36) and O? (0.9 ± 0.2 and 5.9 ± 0.5, respectively). Conversely, the number of medium follicles at oestrus detection was higher in the group O+L? (2.1 ± 0.3, P < 0.05) than in the other two groups (1 ± 0.2 and 1 ± 0.3 for O+L+ and O? respectively). Timing of preovulatory LH surge was earlier for ewes O? (24.0 ± 4.75, P < 0.05) than for sheep O+L+ and O+L? (37.9 ± 2.45 h and 38.0 ± 4.75 h, respectively) and 94% of O+L+ ewes had a LH surge between 16 h and 64 h after cloprostenol injection compared to 57% in O+L? and O? groups (P < 0.05). Thus, maiden Barbarine sheep failing to display oestrus or conceive showed alterations in their follicular dynamics and, thereafter, pituitary function and ovulatory response.  相似文献

We estimated the effect of estrus synchronization on reproduction, production and economic outcomes in 272 beef heifers randomly allocated to a synchronized Test group or an unsynchronized Control group. The Test group received AI upon estrus detection for 6 days followed by PGF2 treatment of heifers that had not shown estrus by day 6 (PGF/6). In both groups AI was continued for 50 days, followed by a 42-day bull breeding period. Heifers were followed through their second breeding season and until they had weaned their first calves. Synchronization resulted in a reduction in median days to first insemination (8 vs. 11 in the Test and Control groups, respectively, P < 0.01) and median days to calving of calves born to AI (14 vs. 20, P = 0.04). There was no significant difference in pregnancy rate to the AI period (60.0% vs. 51.8%, P = 0.18), final pregnancy rate (82.2% vs. 83.2%, P = 0.87) or pregnancy rate to the subsequent breeding season (96.0% vs. 95.0%, P = 1.00). Although mean calf weaning mass was not significantly different (207.0 kg vs. 201.4 kg, P = 0.32), the total mass of calves weaned in this study was 14,843 kg vs. 13,060 kg and the benefit: cost ratio for synchronization was 2.8. It was therefore concluded that a PGF/6 protocol may affect the total mass of calves weaned by changing days to calving, weaning rate, the ratio of male: female calves born and/or the birth mass of calves.  相似文献

Two experiments were conducted to evaluate the effects of the administration of 125 mg rbST at the time of breeding on fertile and subfertile superovulated ewes – in terms of the ovulation rate, embryo yield and embryonic development. In addition, the effects of the treatment on the plasma progesterone concentrations and interferon-tau (IFNT) expression in the embryos were evaluated in the fertile animals. In the first experiment, carried out on fertile superovulated ewes, the treatment significantly increased the ovulation rate (p < 0.05) and shifted the embryo distribution towards more advanced stages of development (p < 0.01). Plasma progesterone concentrations increased faster in the treated ewes, being significantly different (p < 0.05) between groups at day 6 post-breeding. In the second experiment, embryonic development was more advanced in fertile, but not in subfertile superovulated ewes. The expression of IFNT was significantly (p < 0.01) decreased in blastocyst embryos obtained from rbST treated animals, compared with non-treated ewes. It is concluded that the administration of rbST to subfertile ewes at the time of breeding does not stimulate embryonic development as it does when administered to fertile animals.  相似文献

The objective of the present study was to determine the efficacy of the synchronization of estrus using short- and long-term progestagen treatments in ewes at the onset of the breeding season, and to evaluate the effect of the exogenous GnRH administration immediately prior to short-term progestagen treatment on the reproductive performance. A total of 240 Tahirova cross-bred ewes, aged 18–24 months, and 40 rams, aged 2–4 years-old, were used in the trial. Ewes were divided equally into 3 groups (n = 80 per group). Intravaginal progestagen sponges containing FGA (30 mg) were inserted in the ewes for 7 d in the FGA1 (short-term) and GnRH treatment groups, and for 12 d in the FGA2 group (long-term). The ewes in the GnRH group received 10.5 μg busereline acetate i.m. at the time of sponge insertion. Tiaprost tromethamol (PGF; 0.294 mg) and eCG (400 IU) were injected i.m. on the 6th day of progestagen treatment in the GnRH and FGA1 groups, and on the 11th day in the FGA2 group following sponge insertion. All ewes were hand-mated once at the detection of estrus. The estrous response, fertility rate, multiple birth rate and litter size recorded was 88.7, 87.3, 51.6% and 1.6 in the FGA1 group, 92.5, 71.6, 50.9% and 1.5 in the FGA2 group, and 96.2, 89.6, 71.0% and 1.8 in the GnRH group, respectively. No significant difference in estrous response between the groups was recorded, but the fertility rate in the FGA1 and GnRH groups was significantly (P < 0.05) higher than in the FGA2 group. The occurrence of multiple births and litter sizes were significantly (P < 0.05) higher in the GnRH group, compared to both the FGA1 and FGA2 groups, with the number of single lambs being significantly (P < 0.05) higher in the FGA1 (48.4%) and FGA2 (49.0%) groups than in the GnRH (29.0%) group. However, the differences recorded between any of the groups in terms of the number of twin and triplet lambs were insignificant. In conclusion, it can be said that estrous synchronization using the 12-d-FGA-eCG-PGF regimen could be replaced with the 7-d-FGA-eCG-PGF regime in sheep at the onset of the breeding season. However, the combination of GnRH with the latter regimen (7-d-GnRH-FGA-eCG-PGF) increased the multiple birth rate and litter size in the ewes.  相似文献

The cervical anatomy limits the use of trans-cervical artificial insemination (AI) and embryo transfer in ewes. Thus, the objective of the study was to evaluate the sub-arachnoid administration of ketamine as a tool to be used in cervical dilation of the ewe. The following experimental treatments were used: in Experiment 1, animals (n = 6) were treated with the sub-arachnoid administration of ketamine (KS, 1.5 mg kg−1); and in Experiment 2, one tablet of misoprostol was diluted in 1.5 ml saline and deposited near the cervical os, 5 h before the trial + oxytocin administered i.v. 5 min before the reproductive procedures (MO, 200 + 20 U.I., respectively). In Experiment 3, animals received sub-arachnoid ketamine + misoprostol/oxytocin (KSMO). The systemic effects, analgesia, sedation, and motor neuron block were evaluated before each experiment and at pre-determined time intervals thereafter. A subjective score system was used to assess pain and discomfort during the reproductive manipulations in the different experiments. The mean duration of analgesia in the sternal decubitus position without sedation was 30 ± 4 and 25 ± 7 min for the KS and KSMO treatments, respectively. All treatments provided sufficient cervical dilation for the passage of the insemination rod into the uterus. The ketamine sub-arachnoid treatment alone produced a complete cervical dilation, allowing easy passage of the rod. Further studies are needed to establish whether this anesthetic technique may interfere with fertility, pregnancy or lambing rate in ewes during AI or embryo transfer.  相似文献

In pregnant sheep at 120–128 days of gestational age, a study was done to evaluate the effect of the co-administration of verapamil (Vpm) and ivermectin (IVM) on the maternal and fetal disposition kinetics after intravenous administration of IVM. Ten pregnant Suffolk Down sheep of 63.5 ± 6.6 kg body weight (bw) were surgically prepared to insert polyvinyl catheters in the fetal femoral artery and vein and amniotic sac. The ewes were randomly assigned to two experimental groups. In Group 1 (control), 5 ewes were treated with an intravenous bolus of 0.2 mg IVM/kg bw. In Group 2 (Vpm-IVM), 5 ewes were subcutaneously treated with Vpm (2.5 mg/kg × 3 doses at 12 h intervals) and 0.2 mg/kg IVM by intravenous route. Maternal and fetal blood samples were taken before and after IVM administration during a 144 h post-treatment period. Samples were analyzed by liquid chromatography (HPLC). A non-compartmental pharmacokinetic analysis was performed and statistical differences were determined using the Mann–Whitney U-test.Significantly higher IVM levels in maternal plasma concentrations were determined after co-administration of Vpm/IVM compared with the group treated with IVM alone. Significant decreases in the volume of distribution and in the half-life of elimination (t½β) were observed in the Vpm/IVM treated group. A significantly faster (Tmax) and higher (Cmax) (P < 0.05) increase in IVM fetal plasma concentrations were observed in the group of fetuses from pregnant ewes treated with Vpm/IVM. The results of our study support the hypothesis that pharmacological blockage of P-glycoprotein with Vpm can increase the transfer of IVM to fetal circulation.  相似文献

Objective: Patients with 21-hydroxylase deficiency (21OHD) typically suffer from short stature due to early exposure to adrenal-derived androgen. The aim of this study was to investigate whether adding aromatase inhibitor (AI) to gonadotropin-releasing hormone (GnRH) analogue (GnRHa) and recombinant human growth hormone (rhGH) therapy would increase the height of patients with 21OHD.Methods: This retrospective study included 15 patients with 21OHD. The AI/GnRHa/rhGH group consisted of 9 patients, who were treated with AI for at least 12 months in addition to GnRHa/rhGH therapy. The other 6 patients, who received GnRHa/rhGH therapy only, were defined as the GnRHa/rhGH group.Results: Patients were 6.3 ± 1.7 years old, and 7/15 of patients were male. Among them, 12 patients exhibited simple virilization type, and 3 patients were salt-wasting type. In the AI/GnRHa/rhGH group, patients were 6.6 ± 2.0 years old when AI therapy was initiated. Their bone age was 5.9 ± 2.2 years ahead of their chronological age. They received the AI letrizole for an average of 25.1 months (range, 12 to 37 months). In the GnRHa/rhGH group, the patients were 5.9 ± 0.9 years old when they started GnRHa/rhGH therapy, and their bone age was 6.2 ± 1.7 years ahead of their chronological age. Patients received GnRHa/rhGH therapy for an average of 24.5 months (range, 12 to 41 months). The predicted final height increased from 145.9 ± 7.9 to 158.0 ± 8.4 cm in the AI/GnRHa/rhGH group (P = .001, compared with the baseline) and from 141.7 ± 2.7 to 150.7 ± 4.7 cm in the GnRHa/rhGH group (P = .001, compared with the baseline). Bone age progression was 0.15 ± 0.05 per year versus 0.44 ± 0.13 per year in the two groups, respectively (P = .032).Conclusion: Addition of letrizole to GnRHa/rhGH therapy significantly delays bone maturation and may increase the final height.  相似文献

The aim of this work was to determine the effect of two different milking methods (machine versus hand milking) on milk yield, milk composition and somatic cell counts (SCC) in mountainous Greek Boutsiko ewes. Ninety-six ewes were divided in two groups of 48 ewes each. Milk yield, lactation number (parity) and type of lambing were taken into consideration for the separation of the ewes. One group was milked by machine and the other by hand. Milk production, milk composition (with Milkoscan 104) and SCC (with Fossomatic) were performed every 14 days, in the morning and evening milking; during the experimental period seven samplings were performed in total. The results of this study have shown that ewes milked by machine gave a greater daily milk yield as compared to those milked by hand (698 ml versus 652 ml; P < 0,05) and less log SCC (5.37 versus 5.62; P < 0,05). The composition of milk did not differ between the two treatments. Also, no phenotypic correlations were established between SCC and milk production or composition, apart from a negative correlation, which was noted between SCC and the percentage of lactose. The results of this experiment have shown that ewes of the Boutsiko breed can be milked by machine without any problems.  相似文献

Loss of luteal progesterone secretion at the end of the ovine estrous cycle is via uterine PGF2α secretion. However, uterine PGF2α secretion is not decreased during early pregnancy in ewes. Instead, the embryo imparts a resistance to PGF2α. Prostaglandins E (PGE; PGE1 + PGE2) are increased in endometrium and uterine venous blood during early pregnancy in ewes to prevent luteolysis. Chronic intrauterine infusion of PGE1 or PGE2 prevents spontaneous or IUD, estradiol-17β, or PGF2α-induced premature luteolysis in nonbred ewes. The objective was to determine whether chronic intrauterine infusion of PGE1 or PGE2 affected mRNA for LH receptors, occupied and unoccupied receptors for LH in luteal and caruncular endometrium, and luteal function. Ewes received Vehicle, PGE1, or PGE2 every 4 h from days 10 to 16 of the estrous cycle via a cathether installed in the uterine lumen ipsilateral to the luteal-containing ovary.Jugular venous blood was collected daily for analysis of progesterone and uterine venous blood was collected on day-16 for analysis of PGF2α and PGE. Corpora lutea and caruncular endometrium were collected from day-10 preluteolytic control ewes and day-16 ewes treated with Vehicle, PGE1 or PGE2 for analysis of the mRNA for LH receptors and occupied and unoccupied receptors for LH. Luteal weights on day-16 in ewes treated with PGE1 or PGE2 and day-10 control ewes were similar (P  0.05), but were greater (P  0.05) than in day-16 Vehicle-treated ewes. Progesterone profiles on days 10–16 differed (P  0.05) among treatment groups: PGE1 > PGE2 > Vehicle-treated ewes. Concentrations of PGF2α and PGE in uterine venous plasma on day-16 were similar (P  0.05) in the three treatment groups. Luteal mRNA for LH receptors and unoccupied and occupied LH receptors were similar (P  0.05) in day-10 control ewes and day-16 ewes treated with PGE2 and were lower (P  0.05) in day-16 Vehicle-treated ewes. PGE2 prevented loss (P  0.05) of day-16 luteal mRNA for LH receptors and occupied and unoccupied LH receptors. Luteal and caruncular tissue mRNA for LH receptors and occupied and unoccupied LH receptors were greater (P  0.05) on day-16 of PGE1-treated ewes than any treatment group. mRNA for LH receptors and occupied and unoccupied receptors for LH in caruncules were greater (P  0.05) in day-16 Vehicle or PGE2-treated ewes than in day-10 control ewes. It is concluded that PGE1 and PGE2 share some common mechanisms to prevent luteolysis; however, only PGE1 increased luteal and endometrial mRNA for LH receptors and occupied and unoccupied LH receptors. PGE2 prevents a decrease in luteal mRNA for LH receptors and occupied and unoccupied receptors for LH without altering endometrial mRNA for LH receptors or occupied and unoccupied receptors for LH.  相似文献

Effects of supplementing willow stem cuttings to ewes grazing drought pastures upon plasma amino acid (AA) concentrations was studied on Massey University's Riverside Farm, near Masteron, on the East Coast of New Zealand. Ewes of similar age and weight (i.e., 59.0 ± 2.22 kg) were assigned to two groups of 7 each, either with (supplemented) or without (control) supplementation of willow, and experimental grazing was carried for 10 weeks from early February until mid April of 2005. Live weight (LW) was recorded fortnightly and body condition score (BCS) was monthly. Blood samples for quantification of plasma amino acids were collected at week 5 and 10. Both groups had a similar pre-grazing pasture mass (i.e., 2000 kg of dry matter/ha) and dead matter content (0.80) with the diet selected by the ewes containing a metabolisable energy (ME) of 8.3 MJ/kg DM, which is typical of drought conditions. The willow was readily eaten, with intake averaging 0.26 kg DM/ewe/d. Willow was of higher ME content than short drought pasture (i.e., 10.1 versus 8.4 MJ/kg DM) and contained condensed tannins at 40.8 ± 1.97 g/kg DM. Both groups of ewes lost live weight at about 50 g/d. Plasma concentration of 3-methyl histidine (88 versus 127 μmol/L) at week 5 and non-essential amino acids (1082 versus 1417 μmol/L) at week 5 and (1155 versus 1324 μmol/L) at week 10, were substantially lower (P<0.05) in willow supplemented versus control ewes, indicating that willow supplementation reduced catabolism of body proteins in ewes under drought feeding conditions.  相似文献

The ability of sheep to consume big sagebrush (Artemisia tridentata Nutt.), a shrub that contains terpenes, may be improved with supplemental energy and protein needed for detoxification processes. We compared feeding behavior of supplemented and unsupplemented ewes grazing separately in three 0.5 ha plots of 40 m × 120 m during October 2001. Supplemented ewes were offered a choice of a high-energy (50% corn and 50% beet pulp—3.62 Mcal/kg DE and 9.5% CP) and a high-protein (85% alfalfa and 15% soybean meal—2.89 Mcal/kg DE and 15% CP) supplement for 15 min/day. Supplemented ewes consumed 492 g (S.E. = 24.3) of the high-energy supplement and 140 g (S.E. = 9.8) of the high-protein supplement. The incidence of feeding on sagebrush was recorded using scan sampling at 10-min intervals from 09:00 to 17:00 h every other day for 10 days. Incidence of feeding on sagebrush increased gradually for both treatments from 0.4% on day 1 to 28% on day 10 (P < 0.01), but supplemented ewes increased feeding frequency on sagebrush to nearly twice that of unsupplemented ewes by the end of the trial. Averaged throughout the trial, incidence of eating sagebrush was twice as high for supplemented compared to unsupplemented ewes (22% versus 11% of the total time feeding, P < 0.01). The results of this study show that supplemental macronutrients increase use of sagebrush by ewes. Thus, strategic supplementation during fall and winter may be a way to increase intake of sagebrush by sheep and enhance and maintain biodiversity in areas of the sagebrush steppe where sagebrush is deemed too abundant.  相似文献

A new group of hybrid nitric oxide (NO) releasing anti-inflammatory (AI) coxib prodrugs (NO-coxibs) wherein the para-tolyl moiety present in celecoxib was replaced by a N-(4-nitrooxybutyl)piperidyl 15ab, or N-(4-nitrooxybutyl)-1,2,3,6-tetrahydropyridyl 17ab, NO-donor moiety was synthesized. All compounds released a low amount of NO upon incubation with phosphate buffered saline (PBS) at pH 7.4 (2.4–5.8% range). In comparison, the percentage NO released was higher (3.1–8.4% range) when these nitrate prodrugs were incubated in the presence of l-cysteine. In vitro COX-1/COX-2 isozyme inhibition studies showed this group of compounds are moderately more potent, and hence selective, inhibitors of the COX-2 relative to the COX-1 enzyme. AI structure–activity relationship data acquired showed that compounds having a MeSO2 COX-2 pharmacophore exhibited superior AI activity compared to analogs having a H2NSO2 substituent. Compounds having a MeSO2 COX-2 pharmacophore in conjunction with a N-(4-nitrooxybutyl)piperidyl (ED50 = 132.4 mg/kg po), or a N-(4-nitrooxybutyl)-1,2,3,6-tetrahydropyridyl (ED50 = 118.4 mg/kg po), moiety exhibited an AI potency profile that is similar to aspirin (ED50 = 128.7 mg/kg po) but lower than ibuprofen (ED50 = 67.4 mg/kg po).  相似文献

A total of 2567 indigenous Greek ewes (Chios, Vlachiki and Vlachiki×Chios breeds) were used to determine the optimum time for insemination, following synchronization of oestrus with MAP-impregnated intravaginal sponges and PMSG during non-breeding season. Within each breed group, the ewes were divided into three subgroups and submitted to a double blind cervical artificial insemination 48 and 60 h (subgroup I), 60 and 72 h (subgroup II) and 48 and 72 h (subgroup III) after sponge withdrawal. From the results of the present investigation it can be concluded that the conception rate in the Chios island breed is better than that in the Vlachiki×Chios breed, with the latter being better than that in the Vlachiki breed. Using fixed time for AI, a better conception rate is obtained when applied 48 and 72 h after sponge withdrawal for Chios and Chios×Vlachiki breeds, while for Vlachiki breed a better conception rate is obtained when fixed AI is applied 48 and 60 h after sponge withdrawal.  相似文献
