Nitrogen dioxide radical (NO·2) is known as a toxic agent produced in the metabolism of nitrates and nitrites. By the use of the pulse radiolysis technique, the mechanism of the reaction of NO·2 radical with hydroxycinnamic acid derivatives (HCA) was studied and the rate constants have been measured. The rate constants were found to be 7.4 × 108, 7.2 × 108, 8.6 × 108 dm3 mol-1s-1 for ferulic acid, sinapic acid and caffeic acid, respectively. The reactions produce the corresponding phenoxyl radical.  相似文献

The bifunctional hypoxia-specific cytotoxin RB90745, has a nitroimidazole moiety attached to an imidazo[1,2,-a]quinoxaline mono-N-oxide with a spacer/linking group. The reduction chemistry of the drug was studied by pulse radiolysis using the one electron reductant CO2˙-. As N-oxides and nitro compounds react with CO2˙- at diffusion controlled rates, initial reaction produced a mixture of the nitro radical (λmax 410 nm) and the N-oxide radical (λmax 550 nm) in a few microseconds. Subsequently an intramolecular electron transfer (IET) was observed (k = 1.0 ± 0.25 × 103 s-1 at pH 5-9), from the N-oxide to the more electron-affinic nitro group. This was confirmed by the first order decay rate of the radical at 550 nm and formation at 410 nm, which was independent of both the concentration of the parent compound and the radicals. The rates of electron transfer and the decay kinetics of the nitro anion radicals were pH dependent and three different pKaS could be estimated for the one electron reduced species: 5.6 (nitroimidazole group) and 4.3, and 7.6 (N-oxide function). The radicals react with oxygen with rate constants of 3.1 × 107 and 2.8 × 106 dm3 mol-1 s-1 observed at 575 nm and 410 nm respectively. Steady state radiolysis studies indicated four electron stoichiometry for the reduction of the compound.  相似文献

n-Propyl gallate reacts with the superoxide radical anion in aqueous solution (k = 5.1 × 105 mol-1 dm3s-1). The spectrum of the transient species so formed has been measured (absorbance maximum at 550nm, ε = 1360mol-1dm3cm-1). Electron or H atom transfer processes as well as proton abstraction have been excluded as possible mechanisms, and it is proposed that an addition reaction takes place.  相似文献

The rate constant for the reaction of NO with ·O2- was determined to be (6.7 ± 0.9) × 109 1 mol-1 s-1, considerably higher than previously reported. Rate measurements were made from pH 5.6 to 12.5 both by monitoring the loss of ·O2- and the formation of the product -OONO. The decay rate of -OONO, in the presence of 0.1 moll-1 formate, ranges from 1.2s-1 at pH 5 to about 0.2s-1 in strong base, the latter value probably reflecting catalysis by formate.  相似文献

Allopurinol has been employed as a “specific” inhihitor of xanthine oxidase in studies of hypoxic/ reoxygenation injury. Pulse radiolysis was used to establish rate constants for the reactions of allopurinol and its major metabolite oxypurinol with hydroxyl radicals: values were (1.45 ± 0.241 × 109 M-1 s-1 for allopurinol and (4.95 ± 0.84) × 109 M-1 s-1 for oxypurinol. These rate constants show that, in view of the amounts of allopurinol that have been used in animal studies. hydroxyl radical scavenging by this molecule could contribute to its biological actions. especially if animals are pre-treated with allopurinol. so allowing oxypurinol to form. The ability of allopurinol to protect tissues not containing xanthine oxidase against reoxygenation injury may be related to radical scavenging by allopurinol and oxypurinol.  相似文献

The absorption spectra of polyadenylic acid (polyA) radicals in N20 saturated aqueous solution have been measured as a function of time (up to 15 s) following an 0.4μS electron pulse. The spectra and their changes were analysed by comparison with those from monomeric adenine derivatives (nucleosides and nucleotides) which had been studied by Steenken.1

The reaction of OH· radicals with the adenine moiety in poly A results in the formation of two hvdroxvl adducts at the positions C-4 [polyA40H·] and C-8 [polyA80H·]. Each OH-adduct undergoes a unimol-ecular transformation reaction before any bimolecular or other unimolecular decay occurs. These reactions are characterized by different rate constants and pH dependencies. The polyA40H· adduct undergoes a dehydration reaction to yield a neutral N6 centered radical (rate constant Kdeh= 1.4 × 104s-1 at pH7.3). This reaction is strongly inhibited by H+. In comparison with the analogous reactions in adenosine phosphates, the kinetic pK value for its inhibition is two pH units higher. This shift is the result of the counter ion condensation or double-strand formation. The polyA80H· adduct undergoes an imidazole ring opening reaction to yield an enol type of formamidopyrimidine radical with the resulting base damage (kr.o. = 3.5 × 104 s -1 at pH7.3). This reaction in contrast is strongly catalysed by H+and OH-, similar as for adenosine but different compared to the nucleotides.  相似文献

The reactivity of 5-amino-2,3-dihydro-phthalazine-1,4-dione (luminol) and phthalic hydrazide with hydroxyl radicals was studied. HO·-radicals were generated by the Fenton reaction as well as by water radiolysis. Both luminol and phthalic hydrazide react with hydroxyl radicals under intense chemiluminescence (CL) emission. However, exclusively the CL arising from phthalic hydrazide oxidation can be quenched by competition (e.g. by the addition of carbohydrates), whereas luminol CL is enhanced.

The reactivities of both compounds with HO·-radicals were further studied by time-resolved spectroscopy (pulse radiolysis), competition methods, NMR spectroscopy and mass spectrometry. Whereas only slight differences were detectable by pulse radiolysis, the analysis of competition kinetics in the presence of p-nitroso-dimethylaniline (NDMA) gave a two-fold-enhanced reactivity for luminol (4.8 × 109l mol-1 s-1) in comparison to phthalic hydrazide (2.0 × 109l mol-1s-1).

NMR and mass spectrometric analyses revealed significant differences in the reactivity of HO·-radicals: whereas in luminol solutions hydroxylation of the aromatic ring system predominated, hydroxylated products were not detectable upon irradiation of phthalic hydrazide. A hypothetical mechanism is proposed which may explain the observed differences.  相似文献

Nitrogen dioxide (NO2) is a key biological oxidant. It can be derived from peroxynitrite via the interaction of nitric oxide with superoxide, from nitrite with peroxidases, or from autoxidation of nitric oxide. In this study, submicromolar concentrations of NO2 were generated in < 1 μs using pulse radiolysis, and the kinetics of scavenging NO2 by glutathione, cysteine, or uric acid were monitored by spectrophotometry. The formation of the urate radical was observed directly, while the production of the oxidizing radical obtained on reaction of NO2 with the thiols (the thiyl radical) was monitored via oxidation of 2,2′-azino-bis-(3-ethylthiazoline-6-sulfonic acid). At pH 7.4, rate constants for reaction of NO2 with glutathione, cysteine, and urate were estimated as 2 × 107, 5 × 107, and 2 × 107 M−1 s−1, respectively. The variation of these rate constants with pH indicated that thiolate reacted much faster than undissociated thiol. The dissociation of urate also accelerated reaction with NO2 at pH > 8. The thiyl radical from GSH reacted with urate with a rate constant of 3 × 107 M−1 s−1. The implications of these values are: (i) the lifetime of NO2 in cytosol is < 10 μs; (ii) thiols are the dominant ‘sink’ for NO2 in cells/tissue, whereas urate is also a major scavenger in plasma; (iii) the diffusion distance of NO2 is 0.2 μm in the cytoplasm and < 0.8 μm in plasma; (iv) urate protects GSH against depletion on oxidative challenge from NO2; and (v) reactions between NO2 and thiols/urate severely limit the likelihood of reaction of NO2 with NO• to form N2O3 in the cytoplasm.  相似文献

The free radical scavenging properties of retinyl ascorbate (RA-AsA) were determined by monitoring the decomposition of 2,2-diphenyl-1-picrylhydrazyl (DPPH) as a function of time and in comparison with ascorbic acid (AsA), ascorbic acid palmitate (AsA-Pal), retinoic acid (RA), retinol (ROL) and retinol palmitate (Rol-Pal). The rate constant of RA-AsA (mean3±SD) was 4.9±0.3 M-1 s-1, and indicated greater potency as an antioxidant compared to the rest of the test compounds (AsA 3.4±0.4 M-1 s-1, AsA-Pal, 2.9±0.2 M-1 s-1, RA 1.4±0.3 M-1 s-1, ROL 1.3±0.1 M-1 s-1, Rol-Pal exhibited insignificant activity). The decomposition rate constant of DPPH, 5±0.6 × 10-8 M-1 s-1, in ethanol and BHA, 154±3 M-1 s-1 were both used as control. The compound RA-2-carboxy-2-hydroxy-ethanoate was isolated by prep-TLC and was identified, by 13C and 1HNMR spectroscopy, as the major by-product from the reaction of RA-AsA with DPPH, which was also found to be potent antioxidant, 2.1±0.2 M-1 s-1. This suggests that oxidation of AsA moiety did not lead to the production of erythrulose species, which could cause deleterious modifications of cellular proteins.  相似文献

Crocin in aqueous solution is oxidized by ferrylmyoglobin, MbFe(IV)=O, in a second order reaction with k = 183 1 · mol-1 · s-1, AH298 = 55.0 kJ · mol-1, and ΔLS298 = -17 J · mol-1 K-1 (pH = 6.8, ionic strength 0.16 (NaCl), 25°C), as studied by stopped-flow spectroscopy. The reaction has 1:1 stoichiometry to yield metmyoglobin, MbFe(III), and has AGo = -11 kJ · mol-1, as calculated from the literature value E0 = +0.85 V (pH = 7.4) vs. NHE for MbFe(IV)=O/MbFe(III) and from the half-peak potential +0.74 V (vs. NHE in aqueous 0.16 NaCl, pH = 7.4) determined by cyclic voltammetry for the one-electron oxidation product of crocin, for which a cation radical structure is proposed and which has a half-peak potential of +0.89 V for its formation from the two-electron oxidation product of crocin. The fer-rylmyoglobin protein-radical, MbFe(IV)=O, reacts with crocin with 2:l stoichiometq to yield MbFe(IV)= 0, as determined by ESR spectroscopy, in a reaction faster than the second order protein-radical generating reaction between H2O2 and MbFe(III), for which latter reaction k = 137 L · mol-1 · s-1, ΔH298 = 51.5 kJ · mol-1, and ΔH298 = -31 J · mol-1 · K-1 (pH = 6.8, ionic strength = 0.16 (NaCI), 25°C) was determined. Based on the difference between the stoichiometry for the reaction between crocin and each of the two hypervalent forms of myoglobin, it is concluded in agreement with the determined half peak reduction potentials, that the crocin cation radical is less reducing compared to crocin, as the cation radical can reduce the protein radical but not the iron(IV) centre in hypervalent myoglobin.  相似文献

Rate constants determined by the stopped-flow method for four protein-protein reactions at 25°C, pH's in the range 5.8–7.5. I = 0.10 M (NaCI), are as follows: cytochrome c(II) with plastocyanin, PCu(II). 1.5 × 106 M−1 sec−1, pH 7.6; high-potential iron-sulfur protein (Hipip) with PCu(II), 3.7 × 105 M−1sec−1. pH 5.8; cytochrome c(II) with azurin, ACu(ll). 6.4 × 103 M−1sec−1, pH 6.1; Hipip with ACu(II), 2.2 × 105 M−1sec−1, pH 5.8. Activation parameters have been determined for all four reactions; they indicate higher enthalpy requirements and less negative entropy requirements for the PCu(II) as opposed to ACu(II) reactions. Equilibrium constants K for association prior to electron transfer are < 150 M−1 for the cytochrome c(II) reduction of PCu(II) (estimated charges 8 + and 9-,respectively), and < 300 M−1 for the other reactions, indicating no favorable interactions. Rate constants have been analyzed in terms of the simple Marcus theory, which has previously given an excellent fit to thirteen protein-protein reactions considered by Wherland and Pecht. No similar correlation exists in the present studies, and calculated rate constants differ by orders of magnitude from experimentally determined values.  相似文献

Enzymatic acylation of rutin and esculin with aromatic, aliphatic and aryl-aliphatic acids using Candida antarctica lipase in tert amyl alcohol as solvent was investigated under low water content. Whatever the acyl donor used, the conversion yields and initial rates for esculin were higher than for rutin. For a given flavonoid, the performance of these reactions depended on the acyl donor structures. For aliphatic acids, conversion yields and initial rates of both flavonoids were respectively in the ranges of 68-90% and of 9.5×10-2-72×10-2 mmol l-1 h-1. For aromatic acids, the reaction occurred only with the aryl subgroup (cinnamic, hydrocinnamic, 3,4-dihydroxyhydrocinnamic and 4-hydroxyphenyl acetic acids) and was drastically influenced by the presence of side chain and substitution patterns of the aromatic ring. Except for hydrocinnamic acid (75%, 23.4×10-2 mmol l-1 h-1), with these acids the conversion yields and initial rates were lower and in the range of 10-45% and of 0.7×10-2 to 12.1×10-2 mmol l-1 h-1. Unsaturation of the side chain of the hydrocinnamic acid decreased the esculin conversion rate from 75 to 13% and initial rate from 23.4 to 1.76×10-2 mmol l-1 h-1. The presence of hydroxyl or nitro-groups on the aromatic ring of the aryl aliphatic acid also reduced conversion yields and initial rates. Even without a spacer, the non-phenolic ring acid (quinic acid) was reactive and lead to conversion yields of about 20 and 23% respectively for rutin and esculin.  相似文献

The kinetics of O·-2 reaction with semi-oxidized tryptophan radicals in lysozyme, Trp·(Lyz) have been investigated at various pHs and conformational states by pulse radiolysis. The Trp·(Lyz) radicals were formed by Br·-2 oxidation of the 3-4 exposed Trp residues in the protein. At pH lower than 6.2, the apparent bimolecular rate is about 2 × 108M-1s-1; but drops to 8 × 107M-1s-1 or less above pH 6.3 and in CTAC micelles. Similarly, the apparent bimolecular rate constant for the intermolecular Trp·(Lyz) + Trp·(Lyz) recombination reaction is about (4-7 × 106M-1s-1) at/or below pH 6.2 then drops to 1.3-1.6 × 106M-1s-1 at higher pH or in micelles. This behavior suggests important conformational and/or microenvironmental rearrangement with pH, leading to less accessible semioxidized Trp· residues upon Br·-2 reaction. The kinetics of Trp·(Lyz) with ascorbate, a reducing species rather larger than O·-2 have been measured for comparison. The well-established long range intramolecular electron transfer from Tyr residues to Trp radicals-leading to the repair of the semi-oxidized Trp·(Lyz) and formation of the tyrosyl phenoxyl radical is inhibited by the Trp·(Lyz)+O·-2 reaction, as is most of the Trp·(Lyz)+Trp·(Lyz) reaction. However, the kinetic behavior of Trp·(Lyz) suggests that not all oxidized Trp residues are involved in the intermolecular recombination or reaction with O·-2. As the kinetics are found to be quite pH sensitive, this study demonstrates the effect of the protein conformation on O·-2 reactivity. To our knowledge, this is the first report on the kinetics of a protein-O·-2 reaction not involving the detection of change in the redox state of a prosthetic group to probe the reactivity of the superoxide anion.  相似文献

The reaction of FeII oxalate with hydrogen peroxide and dioxygen was studed for oxalate concentrations up to 20 mM and pH 2-5, under which conditions mono- and bis-oxalate comlexes (FeII(ox) and FeII(ox)22-) and uncomplexed Fe2+ must be considered. The reaction of FeII oxalate with hydrogen peroxide (Fe2+ + H2O2 → Fe3+ + *OH + OH-) was monitored in continuous flow by ESR with t-butanol as a radical trap. The reaction is much faster than for uncomplexed Fe2+ and a rate constant, k = 1 × 104 M-1 s-1 is deduced for FeII(ox). The reaction of FeII oxalate with dioxygen is strongly pH dependent in a manner which indicates that the reactive species is FeII(ox)22-, for which an apparent second order rate constant, k = 3.6 M-1 s-1, is deduced. Taken together, these results provide a mechanism for hydroxyl radical production in aqueous systems containing FeII complexed by oxalate. Further ESR studies with DMPO as spin trap reveal that reaction of FeII oxalate with hydrogen peroxide can also lead to formation of the carboxylate radical anion (CO2*-), an assignment confirmed by photolysis of FeIII oxalate in the presence of DMPO.  相似文献

The tetrazolium dyes MTS and XTT were reduced to their soluble formazans by superoxide radical anions (O2_) produced by the oxidation of xanthine by xanthine oxidase under standard conditions. These reactions were compared to the well-known reductions of NBT and cytochrome c by the xanthine/xanthine oxidase system. Reduction of the dyes was completely inhibited by superoxide dismutase (SOD). Rate constants for the reaction of MTS and XTT with O2_: were estimated at 1.3 × .1 × 105 M-1s-1 and 8.6 × .8 × 104 M-1s-1 respectively. The stable MTS and XTT formazans have high extinction coefficients in the visible range which enable sensitive detection and quantification of superoxide radicals, avoiding some of the problems inherent in assays based on production of the insoluble NBT formazan. MTS and XTT have considerable potential both for the quantitative assay of radical production in living tissues and for the assay of superoxide dismutase activity in tissue extracts. Implications for the interpretation of cell culture growth assays which employ these dyes are discussed.  相似文献

Carotenoicls in chloroform and carbon tetrachloriclc photobleach upon nanosecond laser flash photolysis in two steps: instantaneously and in a second-order reaction. The rate constant for second-order reaction (first-order in a solvent derived radical and first-order in (excess) ccirotenoid) is largest for carotenes (9.8·108 M-1 s-1 for β-carotene), intermediate for hydroxylated carotenoids, and smallest for carbonyl containing carotenoids (1.0·108 M-1 s-1 for astaxanthin) in chloroform at 20°C. Near infrared, ibsorbing transients are formed concomitant with pliotohleaching in chloroform (not detected in cxbon tetrachloride). A species formed instantaneously is tentatively identified as either a carotenoid/solvent adduct or an ion-pair. A second species is formed by decay of the instantaneously formed species and is identified as the carotenoid radical cation. This species is formed in a first-order reaction with a rate constant of approx. 5·104 s-1 and absorbing at longer wavelength than the precursor. The lifetime (second-order decay) of the interniediates appears to be longest for the carotenoids with the longest conjugated system. The results indicate that carotenes are better antioxidants than xantliophylls as the carotenes, at least in the present lipophilic solvents, react faster with free radicals.  相似文献

Fungal laccase oxidized derivatives of hydroxycinnamic acid. The rates decreased in the order sinapic acid > ferulic acid ≥p-coumaric acid. The laccase oxidized sinapyl alcohol faster than coniferyl alcohol. The rates of oxidation of the hydroxycinnamic acid derivatives by an isoenzyme of peroxidase from horseradish decreased in the order p-coumaric acid > ferulic acid ≥ sinapic acid. The peroxidase oxidized coniferyl alcohol much faster than sinapyl alcohol. The laccase and the peroxidase predominantly oxidized (a) ferulic acid in a reaction mixture that contained p-coumaric acid and ferulic acid, (b) sinapic acid in a mixture of p-coumaric acid plus sinapic acid, and (c) sinapic acid in a mixture of ferulic acid plus sinapic acid. In a reaction mixture that contained both coniferyl and sinapyl alcohols, both fungal laccase and horseradish peroxidase predominantly oxidized sinapyl alcohol. From these results, it is concluded (1) that the p-hydroxyphenyl radical can oxidize guaiacyl and syringyl groups and produce their radicals and (2) that the guaiacyl radical can oxidize the syringyl group under formation of its radical; and that (3) in both cases the reverse reactions are very slow.  相似文献

Phototactic responses of light-adapted zoeae IV, glaucothoe, and first stage juveniles of the red king crab to three intensities of white light were quantitatively measured under laboratory conditions. All stages observed were photopositive to all light intensities tested, except for late glaucothoe (10 days since moulting) which did not respond to light stimuli. Phototactic response changed in the early life history of the red king crab. The extent of photopositive movement decreased after each metamorphosis. Peak phototactic response in zoea IV were observed at a light intensity of 1.9 × 1013 q cm-2 s-1, in early glaucothoe at 1.1 × 1010 q cm-2 s-1 and in juveniles at 1.3 × 109 q cm-2 s-1. The data on behavioural responses to light may provide a better understanding of the early life history, survival and recruitment of the red king crab and assist the development of feasible methods and techniques for aquaculture of this species.  相似文献

The kinetics of several processes involving the potential antioxidant role of urate in physiological systems have been investigated by pulse radiolysis. While the monoanionic urate radical, ·UH-, can be produced directly by oxidation with ·Br-2 or ·OH, it can also be generated by oxidation with the neutral tryptophan radical, ·Trp, with a rate constant of 2 × 107 M-1s-1. This radical, ·UH-, reacts with ·O-2 with a rate constant of 8 × 108 M-1s-1. Also, ·UH- is reduced by flavonoids, quercetin and rutin in CTAB micelles at rate constants of 6 × 106 M-1s-1 and 1 × 106 M-1s-1, respectively. These results can be of value by providing reference data useful in further investigation of the antioxidant character of urate in more complex biological systems.  相似文献

Experimental evidence is provided that selenomethionine oxide (MetSeO) is more readily reducible than its sulfur analogue, methionine sulfoxide (MetSO). Pulse radiolysis experiments reveal an efficient reaction of MetSeO with one-electron reductants, such as e-aq (k = 1.2 × 1010M-1s-1), CO·-2 (k = 5.9 × 108 M-1s-1) and (CH3)2) C·OH (k = 3.5 × 107M-1s-1), forming an intermediate selenium-nitrogen coupled zwitterionic radical with the positive charge at an intramolecularly formed Se N 2σ/1σ* three-electron bond, which is characterized by an optical absorption with λmax at 375 nm, and a half-life of about 70 μs. The same transient is generated upon HO· radical-induced one-electron oxidation of selenomethionine (MetSe). This radical thus constitutes the redox intermediate between the two oxidation states, MetSeO and MetSe. Time-resolved optical data further indicate sulfur-selenium interactions between the Se N transient and GSH. The Se N transient appears to play a key role in the reduction of selenomethionine oxide by glutathione.  相似文献
