Microalgae are attracting much attention as superior biodiesel producers. In particular, under stressful conditions, they accumulate organic compounds consisting entirely of carbon and hydrogen. The aim of this work was to increase intracellular fatty acid content in Dunaliella tertiolecta (Chlorophyceae), Nannochloropsis oculata (Eustigmatophyceae), and Porphyridium cruentum (Rhodophyceae) using a combination of nitrogen starvation and chemical inhibitors of carbohydrate biosynthesis. These microalgae were subjected to nitrogen starvation and their physiological changes were then observed over time. In D. tertiolecta, no significant change in total fatty acid content was detected on day 3.5 relative to the initial total fatty acid content (day 0), while total carbohydrate content dramatically increased as the nitrogen starvation period was extended. In N. oculata, total fatty acid content rapidly increased, reaching up to nearly 40% of the DCW at day 3.5. However, total carbohydrate content exhibited a gradual reduction throughout the experiment. In P. cruentum, total carbohydrate content increased up to 43% of DCW on day 3.5 and total fatty acid content increased slightly under nitrogen depletion. These data suggest that different eukaryotic microalgae use different storage products under stressful conditions. Among the three strains, D. tertiolecta showed decreased total carbohydrate content and enhanced total fatty acid content following inhibition of carbohydrate synthesis by dichlorophenyl dimethylurea and cyclohexane diamine tetra acetic acid. The results demonstrate the possibility of furthering our understanding of the fatty acid and carbohydrate biosynthesis metabolic network that responds to environmental changes in microalgae.  相似文献

实验针对三株海洋富油微藻:球等鞭金藻(Isochrysis galbana CCMM5001)、一种等鞭金藻(Isochrysis sp. CCMM5002)和一种微拟球藻(Nannochloropsis sp. CCMM7001),研究了它们在通入0.03%(空气)、5%、10%三个浓度CO2培养条件下的生长特性,同时考察了其总油脂及中性脂的累积情况。结果显示,富碳培养有利于这三株海洋微藻的生长,但最适生长的CO2浓度不同。球等鞭金藻(Isochrysis galbana CCMM5001)和等鞭金藻(Isochrysis sp. CCMM5002)在通入10% CO2时具有最大产率,分别达到(182.287.07) mg/(Ld)和(164.227.10) mg/(Ld),而微拟球藻在通入5%时具有最大产率,达到(122.251.17) mg/(Ld),随着CO2浓度的增加,三株海洋微藻的总脂含量和中性脂含量有明显提高。在通入10% CO2条件下,球等鞭金藻(Isochrysis galbana CCMM5001)、等鞭金藻(Isochrysis sp. CCMM5002)和微拟球藻(Nannochloropsis sp. CCMM7001)的总脂含量分别达到(45.154.03)%、(47.151.20)%和(41.201.69)%;从中性脂的累积规律来看,三株藻均在平台期的累积达到最大,脂肪酸分析结果表明三株藻种适合制备生物柴油的C14-C18系脂肪酸相对含量在不同CO2条件下基本保持不变,维持在90%左右。实验结果显示,研究的藻株作为富油高固碳优良藻株,具备用于海洋生物质能耦合CO2减排开发的潜力。    相似文献

This study demonstrates the production of algal biodiesel from Dunaliella tertiolecta, Nannochloropsis oculata, wild freshwater microalgae, and macroalgae lipids using a highly efficient continuous catalytic process. The heterogeneous catalytic process uses supercritical methanol and porous titania microspheres in a fixed bed reactor to catalyze the simultaneous transesterification and esterification of triacylglycerides and free fatty acids, respectively, to fatty acid methyl esters (biodiesel). Triacylglycerides and free fatty acids were converted to alkyl esters with up to 85% efficiency as measured by 300 MHz 1H NMR spectroscopy. The lipid composition of the different algae was studied gravimetrically and by gas chromatography. The analysis showed that even though total lipids comprised upwards of 19% of algal dry weight the saponifiable lipids, and resulting biodiesel, comprised only 1% of dry weight. Thus highlighting the need to determine the triacylglyceride and free fatty acid content when considering microalgae for biodiesel production.  相似文献

Biodiesel production from microalgae is a promising approach for energy production; however, high cost of its process limits the use of microalgal biodiesel. Increasing the levels of triacylglycerol (TAG) levels, which is used as a biodiesel feedstock, in microalgae has been achieved mainly by nitrogen starvation. In this study, we compared effects of sulfur (S) and nitrogen (N) starvation on TAG accumulation and related parameters in wild-type Chlamydomonas reinhardtii CC-124 mt(-) and CC-125 mt(+) strains. Cell division was interrupted, protein and chlorophyll levels rapidly declined while cell volume, total neutral lipid, carotenoid, and carbohydrate content increased in response to nutrient starvation. Cytosolic lipid droplets in microalgae under nutrient starvation were monitored by three-dimensional confocal laser imaging of live cells. Infrared spectroscopy results showed that relative TAG, oligosaccharide and polysaccharide levels increased rapidly in response to nutrient starvation, especially S starvation. Both strains exhibited similar levels of regulation responses under mineral deficiency, however, the degree of their responses were significantly different, which emphasizes the importance of mating type on the physiological response of algae. Neutral lipid, TAG, and carbohydrate levels reached their peak values following 4 days of N or S starvation. Therefore, 4 days of N or S starvation provides an excellent way of increasing TAG content. Although increase in these parameters was followed by a subsequent decline in N-starved strains after 4 days, this decline was not observed in S-starved ones, which shows that S starvation is a better way of increasing TAG production of C. reinhardtii than N starvation.  相似文献

Microalgae have been widely reported as a promising source of biofuels, mainly based on their high areal productivity of biomass and lipids as triacylglycerides and the possibility for cultivation on non-arable land. The isolation and selection of suitable strains that are robust and display high growth and lipid accumulation rates is an important prerequisite for their successful cultivation as a bioenergy source, a process that can be compared to the initial selection and domestication of agricultural crops. We developed standard protocols for the isolation and cultivation for a range of marine and brackish microalgae. By comparing growth rates and lipid productivity, we assessed the potential of subtropical coastal and brackish microalgae for the production of biodiesel and other oil-based bioproducts. This study identified Nannochloropsis sp., Dunaniella salina and new isolates of Chlorella sp. and Tetraselmis sp. as suitable candidates for a multiple-product algae crop. We conclude that subtropical coastal microalgae display a variety of fatty acid profiles that offer a wide scope for several oil-based bioproducts, including biodiesel and omega-3 fatty acids. A biorefinery approach for microalgae would make economical production more feasible but challenges remain for efficient harvesting and extraction processes for some species.  相似文献

Nannochloropsis oceanica F&M-M24 is able to increase its lipid content during nitrogen starvation to more than 50% of the total biomass. We investigated the ultrastructural changes and the variation in the content of main cell biomolecules that accompany the final phase of lipid accumulation. Nitrogen starvation induced a first phase of thylakoid disruption followed by chloroplast macroautophagy and formation of lipid droplets. During this phase, the total amount of proteins decreased by one-third, while carbohydrates decreased by 12–13%, suggesting that lipid droplets were formed by remodelling of chloroplast membranes and synthesis of fatty acids from carbohydrates and amino acids. The change in mitochondrial ultrastructure suggests also that these organelles were involved in the process. The cell wall increased its thickness and changed its structure during starvation, indicating that a disruption process could be partially affected by the increase in wall thickness for biomolecules recovery from starved cells. The wall thickness in strain F&M-M24 was much lower than that observed in other strains of N. oceanica, showing a possible advantage of this strain for the purpose of biomolecules extraction. The modifications following starvation were interpreted as a response to reduction of availability of a key nutrient (nitrogen). The result is a prolonged survival in quiescence until an improvement of the environmental conditions (nutrient availability) allows the rebuilding of the photosynthetic apparatus and the full recovery of cell functions.

Thirty microalgal strains were screened in the laboratory for their biomass productivity and lipid content. Four strains (two marine and two freshwater), selected because robust, highly productive and with a relatively high lipid content, were cultivated under nitrogen deprivation in 0.6-L bubbled tubes. Only the two marine microalgae accumulated lipid under such conditions. One of them, the eustigmatophyte Nannochloropsis sp. F&M-M24, which attained 60% lipid content after nitrogen starvation, was grown in a 20-L Flat Alveolar Panel photobioreactor to study the influence of irradiance and nutrient (nitrogen or phosphorus) deprivation on fatty acid accumulation. Fatty acid content increased with high irradiances (up to 32.5% of dry biomass) and following both nitrogen and phosphorus deprivation (up to about 50%). To evaluate its lipid production potential under natural sunlight, the strain was grown outdoors in 110-L Green Wall Panel photobioreactors under nutrient sufficient and deficient conditions. Lipid productivity increased from 117 mg/L/day in nutrient sufficient media (with an average biomass productivity of 0.36 g/L/day and 32% lipid content) to 204 mg/L/day (with an average biomass productivity of 0.30 g/L/day and more than 60% final lipid content) in nitrogen deprived media. In a two-phase cultivation process (a nutrient sufficient phase to produce the inoculum followed by a nitrogen deprived phase to boost lipid synthesis) the oil production potential could be projected to be more than 90 kg per hectare per day. This is the first report of an increase of both lipid content and areal lipid productivity attained through nutrient deprivation in an outdoor algal culture. The experiments showed that this marine eustigmatophyte has the potential for an annual production of 20 tons of lipid per hectare in the Mediterranean climate and of more than 30 tons of lipid per hectare in sunny tropical areas.  相似文献

Three Isochrysis galbana strains, one I. galbana "substrain", and one Nannochloropsis sp. strain were grown in nutrient replete conditions at 15, 20, 25, and 30°C. The interactive effect of temperature and isolate on growth rate, cell volume, production, dry weight, protein content, carbohydrate content, and total lipid content was examined. Nannochloropsis sp., growth rate had no relation with temperature; all I. galbana isolates increased linearly with temperature, with no significant differences. Cell volume and dry weight decreased linearly with temperature, with significant isolate and species differences. Biovolume-production did not vary significantly with temperature, but significant isolate and species differences occurred. Although significant temperature isolate interactions existed, no consistent trends in the change of protein, carbohydrate, and lipid with temperature occurred. We indicate a need to consider temperature induced affects in application using microalgae isolates, at various taxonomic levels, as some intraspecific differences were as great as interspecific differences.  相似文献

Microalgae are a rich source of high value compounds such as carbohydrates, lipids, proteins and bioactive compounds. In particular, microalgae have been identified as a potentially important resource for carbon-capture and as a feedstock for green biofuels. Successful cultivation of microalgae can occur under a variety of nutrient and environmental conditions with each condition producing a unique distribution of compounds. In order to steer the cultivation towards a particular distribution of compounds, rapid and accurate methods for compound identification are required. Current methods for determining the absolute quantity of each component are time consuming and arduous making cultivation optimization impractical. High-resolution magic angle spinning (HR-MAS) nuclear magnetic resonance (NMR) spectroscopy offers a robust and rapid screening method capable of ascertaining the absolute quantity of each component with minimal sample manipulation. Sample preparation consists of harvested, centrifuged and freeze-dried whole-cell Nannochloropsis granulata from large-scale photobioreactors being accurately weighed and rehydrated with deuterium oxide and placed in an HR-MAS rotor. One-dimensional HR-MAS NMR spectra were recorded under quantitative conditions to determine the lipid and carbohydrate profile of the microalgae. The total time per sample for preparation, data acquisition and analysis was approximately 1 h. Changes in resonance profiles corresponding to varying proportions of saturated and polyunsaturated fatty acids were correlated to the time of harvest. In addition, standard two dimensional experiments were used to identify the major carbohydrate components. HR-MAS NMR spectroscopy has been used to profile the lipid and carbohydrate content of N. granulata and we have begun to establish methodologies for quality analysis/quality control for cultivation of various microalgal strains.  相似文献

Three species of microalgae were grown in mass culture to investigate the influence of culture technique and growth phase on the production of 20:5(n?3) and 22:6(n?3). These polyunsaturated fatty acids (PUFA) are considered to be essential in many marine animals diets for high growth and survival rates. The species of microalgae examined wereNannochloropsis oculata, Pavlova lutheri andIsochrysis sp. (clone T.Iso). All batch cultures (logarithmic and stationary phase) and semi-continuous cultures (logarithmic phase) examined contained high levels of the long-chain (n?3) PUFA, but production could be maximised by harvesting at specific times and growth phases. Maximum cellular content (pg cell-1) of long-chain PUFA was found in logarithmic phase batch cultures ofN. oculata and in stationary phase cultures ofP. lutheri. The cellular content of PUFA in cultures ofIsochrysis sp. did not change significantly with culture technique or growth phase. Alternatively, stationary phase cultures of all three species showed increased proportions (%) and cellular contents of triacylglycerols, and saturated and monounsaturated fatty acids with correspondingly decreased proportions of polar lipids and most PUFA relative to logarithmic phase cultures. The exception was the proportion and cellular content of 22:6(n?3) inP. lutheri which increased with triacylglycerol content. The mass of long-chain (n?3) PUFA per volume of culture was significantly higher in stationary phase cultures due to the higher cell counts per volume. These findings indicate that the opportunity exists to maximise PUFA production by microalgae with the potential to improve animal growth and reduce production costs in mariculture operations and may be of use in the large scale culture and harvesting of microalgae for the biotechnology industry.  相似文献

Summary The effect of cyclopropene fatty acids, sterculic and malvalic, on the lipids of yeasts grown under nitrogen limiting, lipid accumulating, conditions was studied. The ratio of stearic to oleic acid showed a dose response effect, with an increase in stearic acid content as the dose of cyclopropene fatty acid increased, and a corresponding reduction in oleic acid. Linoleic and linolenic acids were not affected to the same extent. These effects are shown for the yeasts Candida sp. 107, Trichosporon cutaneum, and Rhodosporidium toruloides.  相似文献

Cell growth and lipid production of a marine microalga Nannochloropsis oceanica DUT01 were investigated, and fresh medium replacement with different ratios to promote long term cell growth and lipid accumulation was also tested. The highest lipid content reached 64% in nitrogen deplete f/2 medium containing 37.5 mg/L NaNO3 combined with 1/5 fresh medium replacement, however, the highest lipid titer (0.6 g/L) and lipid productivity (31 mg/L/d) were achieved using BG11 medium containing 1.5 g/L NaNO3, taking advantage of 1/5 fresh medium replacement as well, which corresponded to the maximum biomass production of 1.4 g/L, highlighting the importance of high biomass accumulation for efficient lipid production. When biomass compositions were monitored throughout the culture, decreased protein content was found to be coupled with increased lipid production, whereas relatively stable carbohydrate content was observed. The fatty acids in the lipid of N. oceanica DUT01 comprise over 65% saturated fatty acids and monounsaturated acids (i.e. palmitic acid (C16:0) and oleic acid (C18:1)), suggesting that N. oceanica DUT01 is a promising candidate for biodiesel production. Interestingly, very high content of hexadecadienoic acid (C16:2, about 26–33%) was produced by DUT01, which distinguished this microalga with other microalgae strains reported so far.  相似文献

The green alga Hematococcus pluvialis accumulates large amounts of the antioxidant astaxanthin under inductive stress conditions, such as nitrogen starvation. The response to nitrogen starvation and high light leads to the accumulation of carbohydrates and fatty acids as well as increased activity of the tricarboxylic acid cycle. Although the behavior of individual pathways has been well investigated, little is known about the systemic effects of the stress response mechanism. Here we present time-resolved metabolite, enzyme activity, and physiological data that capture the metabolic response of H. pluvialis under nitrogen starvation and high light. The data were integrated into a putative genome-scale model of the green alga to in silico test hypotheses of underlying carbon partitioning. The model-based hypothesis testing reinforces the involvement of starch degradation to support fatty acid synthesis in the later stages of the stress response. In addition, our findings support a possible mechanism for the involvement of the increased activity of the tricarboxylic acid cycle in carbon repartitioning. Finally, the in vitro experiments and the in silico modeling presented here emphasize the predictive power of large scale integrative approaches to pinpoint metabolic adjustment to changing environments.  相似文献

The high content of lipids in microalgae (>60% w/w in some species) and of carbohydrates in seaweed (up to 75%) have promoted intensive research towards valorisation of algal components for the production of biofuels. However, the exploitation of the carbohydrate fraction to produce a range of chemicals and chemical intermediates with established markets is still limited. These include organic acids (e.g. succinic and lactic acid), alcohols other than bioethanol (e.g. butanol), and biomaterials (e.g. polyhydroxyalkanoates). This review highlights current and potential applications of the marine algal carbohydrate fractions as major C-source for microbial production of biomaterials and building blocks.  相似文献

The lipid profile of seven species of unicellular eukaryotic microalgae grown under controlled conditions was studied with emphasis on the hydrocarbons and the fatty acids as part of a search for oil-producing algae. Green, slow-growing colonies of Botryococcus braunii Kutz contained the highest lipid content of 45% based on the organic weight, with an increase to 55% under nitrogen deficiency and with no effect of sodium chloride stress. Ankistrodesmus sp. Thomas, Dunaliella spp., Isochrysis sp., Nannochloris sp. Thomas, and Nitzschia sp. Chapman contained an average of 25% lipids under nitrogen sufficient conditions. Nitrogen deficiency resulted in significant increase in the lipid content in all species but Dunaliella spp., which produced a higher content of carbohydrates. Significant low amounts of acyclic hydrocarbons were detected only in Botryococcus braunii Kutz and not in the other algae. The major hydrocarbon fractions in nitrogen deficient Botryococcus braunii Kutz, Dunaliella salina Thomas, Isochrysis spp. and Nannochloris sp. Thomas were cyclic and branched polyunsaturated components which were identified as various isoprenoid derivatives. The polar lipid composition of glycolipids and phospholipids of all species investigated was fairly typical of photosynthetic eukaryotic algae. Fatty acid composition was species specific, with changes occurring in the relative amounts of individual acids of cells cultivated under different conditions and growth phases. All species synthesized C14:0, C16.0, C18:1, C18:2 and C18:3 fatty acids; C 16:4 in Ankistrodesmus sp. Thomas; C18:4 and C 22.6 in Isochrysis sp.; C16:2, C16:3 and C20:5 in Nannochloris sp. Thomas; C16:2, C16:3 and C20:5 in Nitzschia sp. Chapman. Nitrogen deficiency and salt stress induced accumulation of C18:1 in all treated species and to a lesser extent in Botryococcus braunii Kutz. The low production of hydrocarbons under optimal growth conditions and the high production of hydrocarbons under limited growth conditions cannot support the notion that microalgae can be utilized as biosolar energy converters for the production of liquid fuel, but point to the availability of a variety of neutral and polar lipid products.  相似文献

Microalgal biomass seems to be a promising feedstock for biofuel generation. Microalgae have relative high photosynthetic efficiencies, high growth rates, and some species can thrive in brackish water or seawater and wastewater from the food- and agro-industrial sector. Today, the main interest in research is the cultivation of microalgae for lipids production to generate biodiesel. However, there are several other biological or thermochemical conversion technologies, in which microalgal biomass could be used as substrate. However, the high protein content or the low carbohydrate content of the majority of the microalgal species might be a constraint for their possible use in these technologies. Moreover, in the majority of biomass conversion technologies, carbohydrates are the main substrate for production of biofuels. Nevertheless, microalgae biomass composition could be manipulated by several cultivation techniques, such as nutrient starvation or other stressed environmental conditions, which cause the microalgae to accumulate carbohydrates. This paper attempts to give a general overview of techniques that can be used for increasing the microalgal biomass carbohydrate content. In addition, biomass conversion technologies, related to the conversion of carbohydrates into biofuels are discussed.  相似文献

The lipid characteristics of microalgae are known to differ between species and change with growth conditions. This work provides a methodology for lipid characterization that enables selection of the optimal strain, cultivation conditions, and processing pathway for commercial biodiesel production from microalgae. Two different microalgal species, Nannochloropsis sp. and Chlorella sp., were cultivated under both nitrogen replete and nitrogen depleted conditions. Lipids were extracted and fractionated into three major classes and quantified gravimetrically. The fatty acid profile of each fraction was analyzed using GC–MS. The resulting quantitative lipid data for each of the cultures is discussed in the context of biodiesel and omega‐3 production. This approach illustrates how the growth conditions greatly affect the distribution of fatty acid present in the major lipid classes and therefore the suitability of the lipid extracts for biodiesel and other secondary products. A number of business opportunities may arise from microalgae and wastewater treatment becoming an integrated system, as biofuels and high-added value products could be obtained simultaneously. This study, performed under controlled and non-controlled conditions, aimed at cultivating Chlorococcum sp. using a digestate from pig manure as culture medium and assessing its growth and biochemical composition for further applications. Under controlled conditions, cultures of Chlorococcum sp. were established testing various digestate dilutions (v/v). It was found that all tested dilutions (up to 8% v/v) promoted a higher biomass density, compared to the control culture in modified Bold’s Basal Medium (modified BBM). Under non-controlled conditions, it was found that the biomass productivity using the digestate diluted 5.6% v/v (23.4 mg L?1 day?1) was statistically similar to the one obtained using modified BBM (26.4 mg L?1 day?1). The volatile fatty acids contained in the digestate might have allowed mixotrophic growth for Chlorococcum sp. The intracellular lipid content in Chlorococcum sp. remained constant throughout the experiments in both, treatment and control cultures, while carbohydrates increased from 20 to 45% of the cell dry weight in the treatment and from 20 to 42% in the control one. It was concluded that conditions of nitrogen starvation and fluctuating irradiance and temperature benefit carbohydrate accumulation in this strain, since intracellular carbohydrate content increased nearly two-fold during this period. Additionally, the obtained biomass has the potential to be used as feedstock for bioethanol production. This system can meet the concept of a microalgae-based biorefinery, in which biofuels and high-added value products are produced from microalgae and wastewater.  相似文献
