The objectives of this experiment were to compare estrous synchronization responses and AI pregnancy rates of beef heifers using protocols that included either CIDR or MGA as the progestin source. The hypotheses tested were that: (1) estrous synchronization responses after (a) progestin removal, and (b) PGF(2alpha); and, (2) AI pregnancy rates, do not differ between heifers synchronized with either progestin source. At the start of the experiment (Day 0) in both years, heifers were assigned randomly to receive, MGA supplement for 14 days (MGA-treated; n=79) or CIDR for 14 days (CIDR-treated; n=77). On Day 14 progestin was removed and heifers were observed for estrus up to and after PGF(2alpha) on Days 31 and 33 for CIDR-treated and MGA-treated heifers, respectively. Heifers that exhibited estrus within 60h after PGF(2alpha) were inseminated by AI 12h later; the remaining heifers were inseminated at 72h after PGF(2alpha) and given GnRH (100mug). More (P<0.05) CIDR-treated heifers exhibited estrus within 120h after progestin removal than MGA-treated heifers. Intervals to estrus after progestin removal were shorter (P<0.05) for CIDR-treated heifers than MGA-treated heifers. More (P<0.05) CIDR-treated heifers exhibited estrus and were inseminated within 60h after PGF(2alpha) than MGA-treated heifers. Pregnancy rates did not differ (P>0.10) between MGA-treated (66%) and CIDR-treated (62%) heifers. In conclusion, the use of CIDR as a progestin source in a 14-day progestin, PGF(2alpha), and timed AI and GnRH estrous synchronization protocol was as effective as the use of MGA to synchronize estrus and generate AI pregnancies in beef heifers.  相似文献

The effectiveness of treatments to induce estrus in prepubertal beef heifers was evaluated. Angus x Hereford (n = 148) and Brahman x Hereford (n = 148) heifers were sorted after weaning by body weight into light and heavy weight blocks. Heifers were assigned to diets, calculated to reach a target weight of 55% or 65% of their projected mature weight by the start of breeding. Cyclicity was determined after a 160-d observation period and from concentrations of progesterone in serum determined 10 d before and on the day that treatments began to induce puberty. The remaining nonpubertal heifers, with concentrations of progesterone in serum of less than 1 ng/ml (0 or 10 d before treatment), were assigned randomly within breed and nutrition group to either a melengestrol acetate + saline (MGA+S) or MGA + gonadotropin-releasing hormone (MGA+GnRH) treatment. Prepubertal Angus x Hereford heifers (n = 11) and Brahman x Hereford heifers (n = 49) were fed 0.5 mg MGA for 7 d. Forty-eight hours after MGA, heifers were injected with 500 ug s.c. GnRH or 5 ml of saline. Blood samples were collected from all prepubertal heifers every 3 d after GnRH or saline for 30 d. There was no difference between treatments in the proportion of heifers that exhibited estrus by Day 7 after treatment. However, a larger (P<0.05) proportion of MGA+S-treated heifers exhibited estrus within 14 d after treatment than MGA+GnRH-treated heifers (87 vs 63%). Among heifers that exhibited estrus during that time period, the proportion with increased progesterone was higher (P<0.10) for the MGA+GnRH group than for the MGA+S group (71 vs 41%, Day 7; 79 vs 54%, Day 14). There was no difference in conception rate at first service between treatment groups. Thirty-seven and 53%, respectively, of the MGA+S and MGA+GnRH-treated heifers had short estrous cycles after treatment, and 44 and 50%, respectively, of those short cycles were repeated. Pregnancy rates at the end of 45 d were numerically higher for MGA+S heifers than for MGA+GnRH treated counterparts (63 vs 53%).  相似文献

This experiment was conducted to compare pregnancy rates in postpartum beef cows resulting from fixed-time AI (FTAI) after treatment with controlled internal drug release (CIDR)-based protocols to synchronize estrus. Cows assigned to the Show-Me-Synch (n=167) protocol received a CIDR from d 0 to 14, and prostaglandin F(2α) (PGF(2α)) on d 30. Cows assigned to 7-d CO-Synch+CIDR (n=177) received a CIDR and gonadotropin releasing hormone (GnRH) on d 23. On d 30, CIDRs were removed and PGF(2α) was administered. Blood sampling occurred on d -10 and 0 of treatment to determine estrous cyclicity status (progesterone ≥0.5 ng/mL estrous cycling). Treatments were balanced on age, DPP and BCS. Estrous detection was performed using HeatWatch from PGF(2α) to FTAI. Artificial insemination was performed at predetermined fixed times (72 h, Show-Me-Synch; 66h, 7-d CO-Synch+CIDR) and all cows were administered GnRH at FTAI. This experiment was conducted over a two year period; no differences were found between years so the data were pooled for further analysis. Pregnancy rate resulting from FTAI did not differ (P>0.10) between technicians or AI sires. Pregnancy rate resulting from FTAI was similar between treatments (P=0.20); however, cows that exhibited estrus prior to FTAI had a higher pregnancy rate (P<0.01) than for those that did not. Pregnancy rate at the end of the breeding period was similar between treatments (P=0.28). In summary, FTAI pregnancy rates were similar among postpartum beef cows following treatment with either a short- or long-term CIDR-based estrous synchronization protocol.  相似文献

Two experiments were conducted to compare pregnancy rates when GnRH or estradiol were given to synchronize ovarian follicular wave emergence and ovulation in an MGA-based estrus synchronization program. Crossbred beef cattle were fed melengestrol acetate (MGA, 0.5 mg per day) for 7 days (designated days 0-6, without regard to stage of the estrous cycle) and given cloprostenol (PGF; 500 microg intramuscular (im)) on day 7. In Experiment 1, lactating beef cows (n=140) and pubertal heifers (n=40) were randomly allocated to three groups to receive 100 microg gonadorelin (GnRH), 5 mg estradiol-17beta and 100 mg progesterone (E+P) in canola oil or no treatment (control) on day 0. All cattle were observed for estrus every 12 h from 36 to 96 h after PGF. Cattle in the GnRH group that were detected in estrus 36 or 48 h after PGF were inseminated 12 h later; the remainder were given 100 microg GnRH im 72 h after PGF and concurrently inseminated. Cattle in the E+P group were randomly assigned to receive either 0.5 or 1.0 mg estradiol benzoate (EB) in 2 ml canola oil im 24 h after PGF and were inseminated 30 h later. Cattle in the control group were inseminated 12 h after the first detection of estrus; if not in estrus by 72 h after PGF, they were given 100 microg GnRH im and concurrently inseminated. In the absence of significant differences, all data for heifers and for cows were combined and the 0.5 and 1.0 mg EB groups were combined into a single estradiol group. Estrus rates were 57.6, 57.4 and 60.0% for the GnRH, E+P and control groups, respectively (P=0.95). The mean (+/-S.D.) interval from PGF treatment to estrus was shorter (P<0.001) and less variable (P<0.001) in the E+P group (49.0+/-6.1 h) than in either the GnRH (64.2+/-15.9 h) or control (66.3+/-13.3 h) groups. Overall pregnancy rates were higher (P<0.005) in the GnRH (57.6%) and E+P (55.7%) groups than in the control group (30.0%) as were pregnancy rates to fixed-time AI (47.5, 55.7 and 28.3%, respectively). In Experiment 2, 122 crossbred beef heifers were given either 100 microg GnRH or 2 mg EB and 50 mg progesterone in oil on day 0 and subsequently received either 100 microg GnRH 36 h after PGF and inseminated 14 h later or 1 mg EB im 24 h after PGF and inseminated 28 h later in a 2 x 2 factorial design. Pregnancy rates were not significantly different among groups (41.9, 32.2, 33.3 and 36.7% in GnRH/GnRH, GnRH/EB, EB/GnRH and EB/EB groups, respectively). In conclusion, GnRH or estradiol given to synchronize ovarian follicular wave emergence and ovulation in an MGA-based synchronization regimen resulted in acceptable pregnancy rates to fixed-time insemination.  相似文献

High-fertility (control cows) and low-fertility (cows and heifers not pregnant after two consecutive breeding seasons — twice-open) cyclic bovine females were treated with a single injection of 1000 IU of human chrionic gonadotropin (HCG) or 100 μg of gonadotropin releasing hormone (GnRH) to enhance and/or hasten corpus luteum formation and progesterone secretion, and improve conception rate in the low-fertility females. Hormone treatments were administered to 38 parous control cows, 34 twice-open parous cows, and 27 twice-open nonparous heifers immediately after natural mating by a fertile bull. Blood samples were collected on Days 3, 6, 9, 12, and 18 after mating for determination of systemic progesterone concentrations. Pregnancy rate at necrospy approximately 33 days after mating (range 31–37) was higher in control cows (73.0%) than in twice-open cows (48.4%; P<0.05) or twice-open heifers (34.6%; P<0.01). Pregnancy rate was not affected by the HCG or GnRH treatment. The HCG treatment increased plasma progesterone concentrations in twice-open heifers but not in control or twice-open cows. Progesterone was unaffected by the GnRH treatment. Systemic progesterone concentrations were higher in control than in twice-open females but did not differ between pregnant and nonpregnant females of Days 3, 6, 9 and 12 after mating. Enhanced gonadotropin stimulation at estrus by injection of either HCG or GnRH did not increase pregnancy rate or systemic progesterone concentrations (except in HCG-treated twice-open heifers) in low- or high-fertility females. Lower pregnancy rates in twice-open females were not associated directly with the lower systemic progesterone concentrations.  相似文献

The objective was to synchronize follicular wave emergence among cattle for synchronization of estrus and ovulation, and to determine pregnancy rate after AI at observed estrus. At random stages of the estrous cycle, a controlled internal drug release device (CIDR-B) was inserted intravaginally (Day 0) in 67 cross-bred beef heifers, and they were randomly allocated to receive either no further treatment (Control; n = 18); 5 mg of estradiol-17beta and 100 mg of progesterone im (E/P; n = 16); 100 microg im of GnRH (GnRH; n = 16); or transvaginal ultrasound-guided follicular ablation of all follicles > or = 5 mm (FA; n = 17). All heifers received a luteolytic dose of PGF (repeated 12 h later), and CIDR-B were removed on Days 9, 8, 6 or 5, in Control, E/P, GnRH or FA groups, respectively, so the dominant follicle of the induced wave was exposed to exogenous progesterone for a similar period of time in each group. Mean (+/- SEM) intervals (and range, in days) from treatment to follicular wave emergence in these groups were 3.5 +/- 0.6 (-2 to 8), 3.4 +/- 0.1 (3 to 4), 1.5 +/- 0.3 (-1 to 4), and 1.0 +/- 0.1 (0 to 2), respectively. Although the interval was longest (P<0.01) in the E/P and Control groups, it was least variable (P<0.01) in the E/P and FA groups. Intervals (and range, in days) from CIDR-B removal (and first PGF treatment) to estrus were 2.3 +/- 0.2 (1.5 to 4.5), 2.2 +/- 0.2 (1.5 to 3.0), 2.1 +/- 0.1,(1.5 to 3.5), and 2.5 +/- 0.1 (2.0 to 3.5), and to ovulation were 3.5 +/- 0.2 (2.5 to 5.5), 3.4 +/- 0.1 (3.0 to 4.5), 3.5 +/- 0.1 (2.5 to 4.5), and 3.8 +/- 0.1 (3.0 to 4.5), for Control, E/P, GnRH and FA groups, respectively (ns). The proportion of heifers displaying estrus was higher in the Control than in the FA group (94% versus 65%, P<0.05) and intermediate in EP and GnRH groups (87% and 75%). Heifers were inseminated approximately 12 h prior to ovulation (based on estrous behavior and ultrasound examinations). Pregnancy rates were 78%, 80%, 69% and 65% for Control, E/P, GnRH and FA groups, respectively (P=0.73). Results support the hypothesis that synchronous follicular wave emergence results in synchronous follicle development and, following progesterone removal, synchronous estrus and ovulation with high pregnancy rates to AI. The synchrony of estrus and ovulation in the E/P, GnRH and FA groups suggest that these treatments, in combination with CIDR-B, could be adapted to fixed-time insemination programs.  相似文献

Pregnancy rates were compared in lactating dairy cows (n = 1083) assigned to protocols for resynchronization of ovulation based on stages of the estrous cycle, or presence of ovarian cysts or anestrus. Cows were detected not pregnant by ultrasonography 30 d after a previous AI (study day 0) and classified as diestrus, metestrus, proestrus, with ovarian cysts or anestrus. Cows in diestrus (January-May) were assigned to either Ovsynch (GnRH day 0, PGF2alpha day 7, GnRH day 9, and timed-AI [TAI] 16 h later; n = 96), or Quicksynch (PGF2alpha day 0, estradiol cypionate [ECP] day 1, AI at detected estrus [AIDE] on day 2, or TAI on day 3; n = 96). Cows in diestrus (June-December) were assigned to either Ovsynch (n = 156) or Modified Quicksynch (PGF2alpha day 0, ECP day 1, AIDE days 2 and 3, and to Ovsynch on day 4 if not detected in estrus; n = 142). Cows in metestrus were assigned either to Ovsynch (n = 68), Heatsynch (GnRH day 0, PGF2alpha day 7, ECP day 8, AIDE day 9, or TAI day 10; n = 62), or GnRH + Ovsynch (GnRH on day 0, followed by Ovsynch on day 8; n = 64). Cows in proestrus, with ovarian cysts, or anestrus were assigned to either Ovsynch (proestrus n = 89, ovarian cysts n = 97, anestrus n = 8) or GnRH + Ovsynch (proestrus n = 87, ovarian cysts n = 109, anestrus n = 9). Pregnancy rate was evaluated 30, 55 and 90 d after resynchronized AI. For cows in diestrus (January-May), pregnancy rates were higher for Ovsynch (35.9, 29.2 and 26.0%) than for Quicksynch (21.7, 16.7 and 15.6%). For cows in diestrus (June-December), pregnancy rates were similar for Ovsynch (34.4, 24.0 and 23.6%) and Modified Quicksynch (27.1, 26.2 and 21.6%). For cows in metestrus, pregnancy rates were higher for GnRH + Ovsynch (33.3, 24.5 and 20.3%) than for Heatsynch (20.3, 12.9 and 9.8%). For cows with ovarian cysts, pregnancy rates were higher for GnRH + Ovsynch (30.3, 26.6 and 22.9%) than for Ovsynch (20.2, 18.5 and 14.7%). Assignment to resynchronization protocols based on the stages of the estrous cycle, or presence of ovarian cysts improved pregnancy rates.  相似文献

In Experiment III, a single insemination dose (2.1 × 106 sperm) 12 h after estrus detection (n = 193), a double dose at 12 h (n = 193), or a double dose involving insemination 12 and 24 h after estrus detection (n = 190) did not affect P/AI (87/193 = 45.1%, 85/193 = 44.0%, and 94/190 = 49.5%, respectively; P = 0.51). However, P/AI was influenced by the number of AI service (First, 115/208 = 55.3%a; Second, 94/204 = 46.1%a; and Third, 57/165 = 34.8%b; P = 0.004). In Experiment IV, the P/AI of heifers inseminated from 12 to 16 h after the onset of estrus (40/106 = 37.7%) was less (P = 0.03) than those inseminated from 16.1 to 20 h (85/164 = 51.8%), and 20.1 to 24 h (130/234 = 55.6%). However, the P/AI for heifers inseminated from 24.1 to 30 h (61/134 = 45.5%) did not differ from that of any other interval. In conclusion, in Jersey heifers inseminated with sex-sorted semen, P/AI was not significantly affected by giving GnRH at detection of estrus or a double insemination dose, but it was higher with AI 16.1 to 24 h vs. 12 to 16 h after the onset of estrus.  相似文献

Two experiments evaluated long-term controlled internal drug release (CIDR) insert-based protocols to synchronize estrus and compare differences in their potential ability to facilitate fixed-time artificial insemination (FTAI) in beef heifers. In Experiment 1 estrous cycling heifers (n = 85) were assigned to one of two treatments by age and body weight (BW). Heifers with T1 received a CIDR from days 0 to 14, gonadotropin releasing hormone (GnRH) on day 23, and prostaglandin F (PG) on day 30. Heifers with T2 received a CIDR from days 2 to 16, GnRH on day 23, and PG on day 30. Ovaries were evaluated by ultrasonography on days 23 and 25 to determine ovulatory response to GnRH. In Experiment 2 heifers (n = 353) were assigned within reproductive tract scores by age and BW to one of four treatments. Heifers in T1 and T2 received the same treatments described in Experiment 1. Heifers in T3 and T4 received the same treatments as T1 and T2, respectively, minus the addition of GnRH. In Experiments 1 and 2, heifers were fitted with HeatWatch transmitters for estrous detection and AI was performed 12 h after estrus. In Experiment 1 heifers assigned to T1 had larger dominant follicles at GnRH compared to T2 (P < 0.01) but response to GnRH, estrous response after PG, mean interval to estrus, and variance for interval to estrus after PG did not differ (P > 0.10). AI conception and final pregnancy rate were similar (P > 0.50). In Experiment 2 estrous response after PG did not differ (P > 0.70). Differences in mean interval to estrus and variance for interval to estrus (P < 0.05) differed based on the three-way interaction of treatment length, GnRH, and estrous cyclicity status. AI conception and final pregnancy rates were similar (P > 0.10). In summary, the greater estrous response following PG and resulting AI conception and final pregnancy rates reported for heifers assigned to the two treatments in Experiment 1 and among the four treatments in Experiment 2 suggest that each of these long-term CIDR-based protocols was effective in synchronizing estrus in prepubertal and estrous cycling beef heifers. However, the three-way interaction involving treatment length, GnRH, and estrous cyclicity status in Experiment 2 clearly suggests that further evaluation of long-term CIDR-based protocols is required with and without the addition of GnRH and on the basis of estrous cyclicity status to determine the efficacy of these protocols for use in facilitating FTAI.  相似文献

The objective was to determine whether timed artificial insemination (TAI) 56 h after removal of a Controlled Internal Drug Release (CIDR, 1.38 g of progesterone) insert would improve AI pregnancy rate in beef heifers compared to TAI 72 h after CIDR insert removal in a 5-days CO-Synch + CIDR protocol. Angus cross beef heifers (n = 1098) at nine locations [WA (5 locations; n = 634), ID (2 locations; n = 211), VA (one location; n = 193) and WY (one location; n = 60)] were included in this study. All heifers were given a body condition score (BCS; 1-emaciated; 9-obese), and received a CIDR insert and 100 μg of gonadorelin hydrochloride (GnRH) on Day 0. The CIDR insert was removed and two doses of 25 mg of dinoprost (PGF) were given, first dose at CIDR insert removal and second dose 6 h later, on Day 5. A subset of heifers (n = 629) received an estrus detector aid at CIDR removal. After CIDR removal, heifers were observed thrice daily for estrus and estrus detector aid status until they were inseminated. Within farm, heifers were randomly allocated to two groups and were inseminated either at 56 h (n = 554) or at 72 h (n = 544) after CIDR removal. All heifers were given 100 μg of GnRH at AI. Insemination 56 h after CIDR insert removal improved AI pregnancy rate compared to insemination 72 h (66.2 vs. 55.9%; P < 0.001; 1 - β = 0.94). Locations, BCS categories (≤ 6 vs. > 6) and location by treatment and BCS by treatment interactions did not influence AI pregnancy rate (P > 0.1). The AI pregnancy rates for heifers with BCS ≤ 6 and > 6 were 61.8 and 60.1%, respectively (P > 0.1). The AI pregnancy rates among locations varied from 54.9 to 69.2% (P > 0.1). The AI pregnancy rate for heifers observed in estrus at or before AI was not different compared to heifers not observed in estrus [(65.4% (302/462) vs. 52.7% (88/167); P > 0.05)]. In conclusion, heifers inseminated 56 h after CIDR insert removal in a 5-days CO-Synch + CIDR protocol had, on average, 10.3% higher AI pregnancy rate compared to heifers inseminated 72 h after CIDR insert removal.  相似文献

Estrus synchronization contributes to optimizing the use of time, labor, and financial resources by shortening the calving season, in addition to increasing the uniformity of the calf crop. We determined whether acceptable pregnancy rates could be achieved after synchronization of ovulation and fixed-time artificial insemination (AI) in peripuberal replacement beef heifers using gonadotropin-releasing hormone (GnRH) and PGF2alpha. Crossbred heifers from two herds (MH, n=239; SS, n=330) were wintered at a single location. After a prebreeding examination revealed that 55 heifers had a reproductive tract score (RTS) of 1 (infantile reproductive tracts), they were culled and the remaining heifers were assigned randomly to one of three treatment groups: administration of 25mg PGF2alpha i.m. on Days -12 and 0 followed by estrus detection and insemination between 10 and 14 h after an observed estrus (Control; n=173); administration of 100 microg GnRH i.m. on Day -6, followed by 25 mg PGF2alpha i.m. on Day 0, then fixed-time AI and administration of 100 microg GnRH i.m. on Day +2 (GPG; n=172); and, treatment as for group GPG in addition to administration of 100 microg GnRH i.m. on Day -12 (GGPG; n=169). Bulls were introduced 10 days after AI for 60 days to breed heifers which did not conceive after AI (clean-up bulls). On Days -12, -6, and 0 transrectal ultrasonography was used to monitor ovarian structures in a subset of heifers (30 per treatment). At 30-35 days after AI, ultrasound was used to determine the presence of a viable fetus. Presence of a fetus and stage of pregnancy were determined via palpation per rectum 61-63 days after the conclusion of the breeding season. Heifers in the MH herd (309+/-1.9 kg) were heavier (P<0.001) than those in the SS herd (283+/-1.7 kg) at initiation of the breeding season. Synchronized pregnancy rates were greater (P<0.05) in GGPG (25.4%) and GPG (22.1%) than Control (12.7%) heifers. Pregnancy rates were 9, 21, 32, or 31% for heifers with RTS of 2, 3, 4, or 5, respectively. The average diameter of 22 follicles induced to ovulate in heifers treated with GnRH (GPG and GGPG treatments) was 14.2+/-0.8 mm (range=10.0-23.6 mm). In conclusion, a fixed-time ovulation synchronization program using GnRH and PGF2alpha improved pregnancy rates in peripuberal, lightweight replacement beef heifers.  相似文献

Yearling Bos indicus × Bos taurus heifers (n = 410) from three locations, were synchronized with either the Select Synch/CIDR+timed-AI (SSC+TAI) or 7-11+timed-AI (7-11+TAI) treatments. On Day 0 of the experiment, within each location, heifers were equally distributed to treatments by reproductive tract score (RTS; Scale 1-5: 1 = immature, 5 = estrous cycling) and body condition score. The 7-11+TAI treatment consisted of melengestrol acetate (0.5 mg/head/d) from Days 0 to 7, with PGF (25 mg im) on Day 7, GnRH (100 μg im) on Day 11, and PGF (25 mg im) on Day 18. The SSC+TAI heifers received the same carrier supplement (without MGA) from Days 0 to 7, and on Day 11 they were given 100 μg GnRH and an intravaginal CIDR (containing 1.38 g progesterone). The CIDR were removed on Day 18, concurrent with 25 mg PGF im For both treatments, estrus was visually detected for 1 h twice daily (0700 and 1600 h) for 72 h after PGF, with AI done 6 to 12 h after a detected estrus. Non-responders were timed-AI and received GnRH (100 μg im) 72 to 76 h post PGF. The 7-11+TAI heifers had a greater (P < 0.05) estrous response (55.2 vs 41.9%), conception rate (47.0 vs 31.3%), and synchronized pregnancy rate (33.5 vs 24.8%) compared to SSC+TAI heifers, respectively. Heifers exhibiting estrus at 60 h (61.7%) had a greater (P < 0.05) conception rate compared to heifers that exhibited estrus at ≤ 36 (35.3%), 48 (31.6%), and 72 h (36.2%), which were similar (P > 0.05) to each other. As RTS increased from ≤ 2 to ≥ 3, estrous response, conception rate, synchronized pregnancy rate, and 30 d pregnancy rate all increased (P < 0.05), irrespective of synchronization treatment. In conclusion, the 7-11+TAI treatment yielded greater synchronized pregnancy rates compared to SSC+TAI treatment in yearling Bos indicus × Bos taurus heifers.  相似文献

A synthetic gonadotropin-releasing hormone (GnRH) agonist (fertirelin acetate, FA) was administered to beef cattle within 12 h after onset of estrus (Day = 0) to study effects on subsequent endocrine responses and fertility. In Study 1, 16 crossbred beef heifers were injected with either 100 mug FA (n = 8) or saline (n = 8) at 6 or 12 h (n = 7; n = 9) after onset of estrus. Concentrations of luteinizing hormone (LH) over time were affected (P<0.01) by the interaction of treatment and interval from onset of estrus to treatment. Heifers treated with FA at 6 h after onset of estrus exhibited the greatest increase in LH after treatment. There was no effect of treatment, interval from onset of estrus to treatment or treatment by interval interaction on duration of the estrous cycle, on concentrations of progesterone from Days 1 through 14 posttreatment, or on concentrations of progesterone prior to subsequent estrus (Day -10 through 0, posttreatment estrus). In summary, FA administered to beef cattle within 12 h after onset of estrus effectively increased peripheral plasma concentration of LH, but this increase had no effect on subsequent luteal function as measured by duration of the estrous cycle or concentrations of plasma progesterone. In Study 2, 86 parous beef cows were bred artificially to one of two bulls following natural or prostaglandin F(2)alpha induced estrus. Cows received either no treatment or 50 or 100 mug FA at the time of AI. There was no effect of treatment, breed, parity, technician, service sire or interactions on conception rate (mean = 76.7%). Although not significant, the numerical pattern of conception rate among experimental groups (control = 71.4%, 50 mug FA = 76.7%, 100 mug FA = 82.1%) supports further investigation of this GnRH agonist with larger numbers of cattle.  相似文献

Anestrous and lactating Bos taurus by Bos indicus crossbred cows with minimum body condition were studied to determine the efficacy of GnRH+PGF 2alpha combinations for induction of estrus and/or ovulation on pregnancy rate during the months of the year when temperatures are greater. On day 0 (start of treatment), cows were assigned randomly to either treatment or control groups. Treated cows (n = 74) received i.m. 200 microg of GnRH on day 0 and 150 microg of PGF 2alpha 7 days later (day 7). On day 7, treated cows were equally distributed to each of three protocols: (1) Select Synch (n = 25), artificial insemination (AI) 12 h after exhibiting estrus from day 7 (PGF 2alpha injection) until day 12; (2) Ovsynch (n = 24), 200 microg of GnRH at 48 h after PGF 2alpha (day 9) + timed-AI (TAI) 16-20 h later; (3) CO-Synch (n = 25), 200 microg of GnRH + TAI at 48 h after PGF 2alpha (day 9). Control cows (n = 25) received no treatment + AI 12 h after exhibiting estrus from days 0 to 12. Detection of estrus was performed daily during the early morning and evening hours from days 0 to 7 in all the cows, and from days 7 to 12 in the cows treated with Select Synch and in the control group, with the aid of a sterilized bull. Palpation per rectum and transrectal ultrasonography were used on days -30, -20, -10 and 0 to confirm anestrus (absence of CL and no signs of estrus at each evaluation) but with ovarian follicles > or = 10 mm on day 0. Pregnancy rate was 0% for Select Synch, 21% for Ovsynch and 28% for CO-Synch (P < 0.05). In conclusion, the Ovsynch and CO-Synch protocols resulted in greater pregnancy rates compared with the Select Synch protocol in Bos taurus/Bos indicus cows with minimum body condition that were anestrous and lactating during the summer months in a tropical environment.  相似文献

Twelve mature Angus bulls possessing similar semen quality were used to study the effects of single and multiple services and single-sire and multiple-sire mating on pregnancy rate of beef heifers. Heifers were observed for natural estrus twice daily and mated within 6 hr of onset of estrus. Pregnancy rate was determined by rectal palpation at 60 days post-mating. Pregnancy rate of heifers receiving one service per estrus averaged 62.1% (n=124) and pregnancy rate of heifers receiving multiple services (single-sire) per estrus averaged 62.9% (n=105). Pregnancy rate per estrus of heifers mated to three sires (multiple-sire, one service per sire; 74.0%, n=123) was significantly higher than that of heifers mated to a single sire (62.9%). The range in pregnancy rate per bull for single-sire matings was large (0 to 95%), but this range was reduced among the same bulls used for multiple-sire matings (68 to 84%). These results indicate that pregnancy rate to one service did not differ (P>.10) from pregnancy rate to multiple services among single-sire matings, but multiple-sire matings increased (P<.05) pregnancy rate per estrus by 11% over that obtained with single-sire matings.  相似文献

Dairy heifers were used to compared the effects of two timed AI + controlled internal drug release (CIDR) protocols (5-day vs. a modified 7-day) on: (1) luteal regression to initiate a new ovarian follicular wave; (2) ovarian response to the initial GnRH injection; and (3) pregnancy outcomes. Holstein heifers (N = 543) were assigned randomly to two treatments: (1) 25 mg PGF (im) and a CIDR insert on Day −7 followed by 100 μg of GnRH (GnRH-1) on Day −5 and 25 mg PGF (im) at CIDR insert removal (7-day [7D]) on Day 0; or (2) 100 μg GnRH (GnRH-1) and insertion of a CIDR on Day −5 and 25 mg PGF (im) at CIDR removal (5-day [5D]) on Day 0. Insemination with frozen-thawed conventional or gender-biased semen occurred after detected estrus from Days 0 to 2 or by appointment at 72 h after PGF when a second 100-μg dose of GnRH was given. Blood was collected on Days −7, −5, 0, and 3 to determine concentrations of progesterone and incidence of luteolysis. Ovaries were scanned on Days −5 and 0. Luteolysis in the 7D treatment by 48 h after the initial PGF was greater (P < 0.01) than what occurred spontaneously in the 5D treatment (36.2% vs. 19.7%, respectively). Incidence of ovulation after GnRH-1 on Day −5 was greater (P < 0.05) for 7D than for 5D heifers, but the proportion of heifers with an induced CL on Day 0 did not differ between treatments. Heifers inseminated after detected estrus (166/543, 30.6%) on Days 0, 1, and 2 had greater (P < 0.05) pregnancy per AI (P/AI) at 32 days post AI than after timed AI (38.2% vs. 28.3%) on Day 3. Pregnancy P/AI, however, was greater (P < 0.05) for 7D heifers inseminated at estrus (46.5%) than for 7D heifers receiving the timed AI (26.8%) and differed (P < 0.05) from all 5D heifers regardless of insemination time at estrus (30.5%) or at timed AI at 72 h (29.9%). At the Florida location in which conventional and sexed semen were used during two breeding clusters, P/AI using sexed semen (43.9%, N = 56) did not differ from that of conventional semen (21.2%, N = 50). Remaining replicates of sexed semen produced similar P/AI at the other two locations (sexed = 27.6%, N = 71; and sexed = 31.9%, N = 215). We concluded that the modified 7-day CO-Synch + CIDR program produced more P/AI in heifers inseminated at estrus than a standard 5-day CO-Synch + CIDR program, but when timed AI occurred at 72 h after PGF and CIDR insert removal, P/AI did not differ between programs.  相似文献

The objectives were (1) to determine the effects of gonadorelin hydrochloride (GnRH) injection at controlled internal drug release (CIDR) insertion on Day 0 and the number of PGF2α doses at CIDR removal on Day 5 in a 5-day CO-Synch + CIDR program on pregnancy rate (PR) to artificial insemination (AI) in heifers; (2) to examine how the effect of systemic concentration of progesterone and size of follicles influenced treatment outcome. Angus cross beef heifers (n = 1018) at eight locations and Holstein dairy heifers (n = 1137) at 15 locations were included in this study. On Day 0, heifers were body condition scored (BCS), and received a CIDR. Within farms, heifers were randomly divided into two groups: at the time of CIDR insertion, the GnRH group received 100 μg of GnRH and No-GnRH group received none. On Day 5, all heifers received 25 mg of PGF2α at the time of CIDR insert removal. The GnRH and No-GnRH groups were further divided into 1PGF and 2PGF groups. The heifers in 2PGF group received a second dose of PGF2α 6 hours after the administration of the first dose. Beef heifers underwent AI at 56 hours and dairy heifers at 72 hours after CIDR removal and received 100 μg of GnRH at the time of AI. Pregnancy was determined approximately at 35 and/or 70 days after AI. Controlling for herd effect (P < 0.06), the treatments had significant effect on AI pregnancy in beef heifers (P = 0.03). The AI-PRs were 50.3%, 50.2%, 59.7%, and 58.3% for No-GnRH + PGF + GnRH, No-GnRH + 2PGF + GnRH, GnRH + PGF + GnRH, and GnRH + 2PGF + GnRH groups, respectively. The AI-PRs were ranged from 50% to 62.4% between herds. Controlling for herd effects (P < 0.01) and for BCS (P < 0.05), the AI pregnancy was not different among the treatment groups in dairy heifers (P > 0.05). The AI-PRs were 51.2%, 51.9%, 53.9%, and 54.5% for No-GnRH + PGF + GnRH, No-GnRH + 2PGF + GnRH, GnRH + PGF + GnRH, and GnRH + 2PGF + GnRH groups, respectively. The AI-PR varied among locations from 48.3% to 75.0%. The AI-PR was 43.5%, 50.4%, and 64.2% for 2.5 or less, 2.75 to 3.5, and greater than 3.5 BCS categories. Numerically higher AI-PRs were observed in beef and dairy heifers that exhibited high progesterone concentrations at the time of CIDR insertion (>1 ng/mL, with a CL). In addition, numerically higher AI-PRs were also observed in heifers receiving CIDR + GnRH with both high and low progesterone concentration (<1 ng/mL) initially compared with heifers receiving a CIDR only with low progesterone. In dairy heifers, there were no differences in the pregnancy loss between 35 and 70 days post-AI among the treatment groups (P > 0.1). In conclusion, GnRH administration at the time of CIDR insertion is advantageous in beef heifers, but not in dairy heifers, to improve AI-PR in the 5-day CIDR + CO-Synch protocol. In addition, in this study, both dairy heifers that received either one or two PGF2α doses at CIDR removal resulted in similar AI-PR in this study regardless of whether they received GnRH at CIDR insertion.  相似文献

A study was conducted to evaluate the effectiveness of treatment with Syncro-Mate-B (SMB) and gonadotropin releasing-hormone (GnRH) on inducing a fertile estrus and/or ovulation in peripuberal beef heifers. Two hundred and twenty-three, 13 to 15 month peripuberal beef heifers, primarily of Angus or Simmental breeding, were alternately allotted by weight and age within breed to a non-treated control group (C) and three groups which received the SMB treatment regime. Treated groups differed in that one group was inseminated approximately 12 hr after detected in estrus (T) while two other groups were inseminated at a fixed time after implant removal (T-G-I42, T-I48). Heifers in group T-G-I42 received 125 mug GnRH at 30 hr, and were inseminated 42 hr after implant removal. The remaining group of SMB-treated heifers received no additional hormone therapy but were inseminated 48 hr after implant removal (T-I48). Age had no significant effect on estrous response or pregnancy rate nor were there significant age by weight interactions on these parameters. Weight was a significant source of variation on interval from start of breeding to pregnancy (P<.01) and the effect on interval to estrus approached statistical significance (P<.0.9). In control heifers, increased weight had a positive effect on estrous response during the first 21 days of breeding (P<.05). Conversely, weight had no effect on estrous response during the 5-day synchronized period in heifers treated with SMB and observed for estrus (group T). Cumulative estrous response in control heifers was 10, 47, 48 and 55% in 5, 21, 27 and 45 days of breeding, respectively. Cumulative estrous response in SMB-treated heifers obsrved for estrus (group T) was 88, 91, 91 and 91% in 5, 21, 27 and 45 days of breeding, respectively. Pregnancy rate after 5 days of breeding was 9, 27, 18 and 30% for heifers in groups C, T, T-G-I42 and T-I48, respectively. Pregnancy rate after 21 days of breeding for heirfers in groups C, T, T-G-I42 and T-I48, respectively, was 33, 38, 21 and 41%; after 27 days of breeding was 36, 43, 40 and 48%; after 45 days of breeding was 48, 61, 51 and 69% and after the total 88-day breeding season was 59, 68, 71 and 72%. While a large percentage of peripuberal beef heifers with adequate age but insufficient weight were induced into estrus with the SMB treatment, pregnancy rates were low. Timed insemination programs with or without GnRH do not appear to be effective in improving pregnancy response in non-cyclic beef heifers.  相似文献

Two experiments evaluated a modified delivery of prostaglandin F2alpha (PGF2alpha) after a melengestrol acetate (MGA) treatment in Angus and Bos indicus x Bos taurus (BI) heifers. Experiment 1 was replicated three times with yearling BI heifers (n = 695). Heifers received MGA (0.5 mg head(-1) day(-1)) for 14 days. In Replications 1 and 2, heifers received either 25 mg of PGF2alpha im 19 days after MGA (single) or 12.5 mg of PGF2alpha im 19 and 20 days after MGA (split). In Replication 3, heifers received the same treatments, with PGF2alpha initiated either 18 or 19 days after MGA. Estrus was detected for 72 h after PGF2alpha, with AI commencing 8-12 h after a detected estrus. Heifers not observed in estrus by 72 h were timed-AI concomitant with GnRH (100 microg im). Heifers from Replication 2 (n = 146) had blood samples collected at the initial PGF2alpha and at timed-AI to determine corpus luteum (CL) regression by evaluating plasma progesterone concentrations. The interval from MGA withdrawal to PGF2alpha did not have a significant effect on any variable in Replication 3 and there were no treatment by replication effects for any variables, therefore data were pooled. Modifying the PGF2alpha treatment from a single treatment to two treatments on consecutive days increased (P < 0.05) 72 h estrous response (43.2% versus 50.1%), timed-AI (23.9% versus 33.5%) and total-AI pregnancy rates (34.5% versus 42.5%), and CL regression (79.1% versus 92.5%), respectively. In Experiment 2, yearling Angus (n = 66) and 2-year-old BI (n = 68) heifers were synchronized as per Experiment 1 (with the initial PGF2alpha 19 days after MGA). Neither breed nor PGF2alpha treatment effected (P > 0.05) 72 h estrous response, total-AI pregnancy rate, or CL regression rate. In conclusion, treating yearling BI heifers with split treatments of PGF2alpha (given on two consecutive days) improved estrous response and pregnancy rates by increasing PGF2alpha-induced luteolysis.  相似文献
