Binding of gossypol by gossypin and congossypin and their succinylated and sulfhydryl group-blocked derivatives has been measured. The binding by gossypin and congossypin is characterized by weak interaction. Succinylation of gossypin decreases the binding affinity whereas that of congossypin increases it. Blocking of sulfhydryl groups of both the proteins does not significantly affect gossypol binding, Succinylation dissociates gossypin and causes conformational changes whereas it does not dissociate congossypin but causes conformational changes. Sulfhydryl group blocking does not dissociate gossypin or congossypin, nor does it cause any conformational changes.  相似文献

In response to changes in electrolyte concentration and pH, erythrocyte ghosts can exhibit some of the characteristic shapes seen in the intact erythrocyte. These shape changes are accompanied by volume changes; both are reversible, not energy dependent, and not inhibited by sulfhydryl reagents. The volume reduction can also be seen in isolated Triton-free spectrin-actin lattices, showing that this network is capable of reversible contraction. The results suggest that reversible changes in size of the underlying cytoskeleton of the erythrocyte membrane can control cell shape.  相似文献

Pineal melatonin synthetic activity shows distinct diurnal characteristics. The circadian regulation of melatonin synthesis is provided by noradrenaline-releasing sympathetic nerves. The pineal noradrenaline content shows a circadian rhythmicity tidally related to the changes in melatonin synthesis rate. To evaluate possible circadian changes of pineal noradrenergic fibre arrangement, the nerve distribution in rat and guinea pig pineal glands was visualized by means of glyoxylic acid-induced histofluorescence. Histochemical findings at 08.00 h and 24.00 h did not exhibit any differences: in both species a dense, mainly perivascularly located network of fluorescent fibres was encountered. As indicated by the simultaneous intraneural presence of green-bluish and yellow fluorophores these fibres most likely contain noradrenaline and serotonin. Obviously circadian melatonin synthesis changes are not paralleled by changes in the distribution pattern of pineal sympathetic nerve fibers. Like other sympathetic innervation-related morphological parameters, histofluorescence does not accurately reflect circadian biochemical changes in the pineal gland.  相似文献

Arginine vasopressin (AVP) regulates aggression in male Syrian hamsters. In this study, we used radioligand receptor autoradiography to examine whether changes in agonistic behavior following acute and repeated social defeat are accompanied by changes in AVP V1a receptor binding. Social defeat produced high levels of submissive behavior and a loss of territorial aggression when hamsters were subsequently tested with a novel intruder, and repeated agonistic encounters produced similar behavioral changes in subordinates. AVP V1a receptor binding was not reduced by acute social defeat but was affected by repeated agonistic encounters. Dominants had significantly more AVP V1a receptor binding in lateral portions of the ventromedial hypothalamus (VMHL) than did their subordinate opponents, but subordinates were no different from controls. In contrast, receptor binding did not differ in most other brain regions examined. The changes in receptor binding appear to be independent of testosterone levels, as testosterone levels did not differ among dominants, subordinates, and controls. Our results suggest that changes in AVP V1a receptors do not account for the changes in agonistic behavior produced by acute social defeat but AVP V1a binding in the VMHL correlates with, and may modulate, the behavioral changes that occur following repeated experiences of victory.  相似文献

The role of oxygen radicals and lipid peroxidation in calcium-paradox injury in isolated perfused rat hearts was studied by examining the effects of mannitol and (or) allopurinol on this phenomenon. Myocardial changes due to calcium paradox were characterized by contractile failure, a rise in resting tension, and cell damage. These changes were also accompanied by increased lipid peroxidation, as indicated by an increase in malondialdehyde content. Mannitol (an effective quencher of hydroxyl radicals) treatment resulted in a dose-dependent decrease in lipid peroxidation but did not affect other changes due to calcium paradox. Allopurinol (an inhibitor of xanthine oxidase) neither affected lipid peroxidation nor modified any of the structure-function changes due to calcium paradox. These data demonstrate the occurrence of lipid peroxidation which, however, may not be involved in the observed structure-function changes due to calcium paradox. It is also suggested that in this experimental model, xanthine oxidase may not be the inducer of oxygen radicals or of lipid peroxidation.  相似文献

Wildfires have played a determining role in distribution, composition and structure of many ecosystems worldwide and climatic changes are widely considered to be a major driver of future fire regime changes. However, forecasting future climatic change induced impacts on fire regimes will require a clearer understanding of other drivers of abrupt fire regime changes. Here, we focus on evidence from different environmental and temporal settings of fire regimes changes that are not directly attributed to climatic changes. We review key cases of these abrupt fire regime changes at different spatial and temporal scales, including those directly driven (i) by fauna, (ii) by invasive plant species, and (iii) by socio-economic and policy changes. All these drivers might generate non-linear effects of landscape changes in fuel structure; that is, they generate fuel changes that can cross thresholds of landscape continuity, and thus drastically change fire activity. Although climatic changes might contribute to some of these changes, there are also many instances that are not primarily linked to climatic shifts. Understanding the mechanism driving fire regime changes should contribute to our ability to better assess future fire regimes.  相似文献

Serum LH and FSH concentrations were measured in adult male rats. Blockage of the ductuli efferentes by surgical ligation or treatment with the anti-fertility drug alpha-chlorohydrin did not produce any changes in serum LH or androgen-dependent organ weights. Serum FSH in both groups was significantly raised by 4 days after treatment and remained so throughout most of the experimental period (42 days) but did not attain the values obtained in castrates. The gonadotrophin changes were accompanied by an initial increase in the weight and turgidity of the testes which then became flaccid and atrophied. These changes were similar but not identical after both treatments. Androgen secretion was impaired in adult rats treated with ethylene dimethane sulphonate; serum LH values were similar to those of 14-day castrates, and serum FSH was also elevated but not to the level found in the castrates. Ligation of the ductuli efferentes did not produce any further changes in concentrations of either hormone in spite of the retention of the testicular products within the testes as reflected by an increase in weight. The results are consistent with the view that changes in serum FSH after blockage of the ductuli efferentes are related to disturbances in the production of inhibin from the seminiferous epithelium rather than retention of inhibin with the testicular tubules.  相似文献

The role of the proton gradient and external pH in the motility and chemotaxis of Bacillus subtilis was investigated. Presence of a substantial proton gradient is not necessary for motility or chemotaxis, as long as the electrical potential is sufficient to maintain motility. Changes in the proton gradient do, however, lead to changes in swimming behavior, and these changes are mediated by two processes. One is sensitive to external pH and probably operates through a pH receptor. The second is sensitive to changes in the proton gradient. When the level of the protonmotive force is high enough to maintain motiligy, changes in the components of the protonmotive force are sensed by the bacteria and lead to behavioral changes, but changes in the protonmotive force are not necessary for chemotaxis.  相似文献

Liver dysfunction was produced in rats by surgical portocaval anastomosis (PCA), and the time-course of changes in brain tryptophan and 5-HT metabolism studied in relation to plasma changes possibly influencing brain tryptophan concentration. Brain tryptophan and 5-hydroxyindolylacetic acid (5-HIAA) levels were increased greatly and maximally on the day after PCA and remained high. 5-HT changes were less marked but had a similar time-course. Plasma total tryptophan was little changed but plasma free tryptophan was raised. The latter change showed a similar time-course to that of brain tryptophan but was not large enough to account completely for it. Sham operation was followed by significant but transient increases in plasma free tryptophan, brain tryptophan and 5-HIAA but these were much smaller than after PCA. Brain tryptophan did not correlate with plasma total tryptophan either in control or PCA rats but it correlated significantly with plasma free tryptophan in both groups. However brain levels were much higher in PCA rats than in controls with similar plasma free tryptophan levels at all times from the first day after operation. The increase of brain tryptophan in anastomosed rats not accounted for by plasma free tryptophan was explained neither by insulin changes nor by an increase of the insulin/glucagon ratio nor by changes in plasma concentrations of those amino acids which compete with tryptophan for entry into brain. The results therefore indicate an unknown influence on brain tryptophan concentration in PCA rats. As tyrosine changes in brain and plasma after PCA were very similar to those of tryptophan this influence may not be specific to tryptophan. Results suggest that under the conditions used brain tryptophan concentrations of both PCA and control rats are more influenced by changes of plasma free tryptophan concentration than by changes of plasma concentrations of competing amino acids.  相似文献

Luminol-mediated chemiluminescence in neutrophils is stimulated by Sendai virus and by influenza virus; Lettré cells also exhibit chemiluminescence (less than 10% of that of neutrophils), which is stimulated by Sendai virus and by influenza virus. Virally induced permeability changes are not responsible for chemiluminescence, since (i) extracellular Ca2+ inhibits permeability changes but stimulates chemiluminescence, and (ii) influenza virus, which induces permeability changes at pH 5.3 but not at pH 7.4, induces chemiluminescence at either pH. Other agents [zymosan, N-formyl-L-methionyl-L-leucyl-L-phenylalanine, 4-phorbol 12-myristate 13-acetate (phorbol ester), A23187] likewise induce chemiluminescence in the absence of permeability changes.  相似文献

Abstract— Withdrawal of food or immobilization both led to changes in rat brain tryptophan metabolism. Brain tryptophan and 5-hydroxyindolylacetic acid concentrations both increased while changes in 5-hydroxytryptamine were much smaller. Changes were greater upon withdrawal of food. The brain tryptophan change did not appear merely to reflect an overall increase of brain amino acid concentrations, brain tyrosine concentration being only slightly increased by food withdrawal and significantly decreased upon immobilization. Plasma tryptophan did not increase. The changes in brain indole metabolism were not abolished by adrenalectomy. Results are discussed in relation to the regulation of brain serotonin metabolism.  相似文献

Pathological changes were found not only in the intestines but also in the liver, lungs, spleen, kidney and suprarenal glands. We studied epidermal growth factor (EGF) binding to renal basolateral membranes before and following unilateral nephrectomy. After 48 h unilateral nephrectomy there was a small increase in kidney cortex weight but EGF binding was unchanged, suggesting that alterations in EGF binding do not play a role in early renal hypertrophy. In contrast, 3 week unilateral nephrectomy was associated with a significant decrease in the Bmax of the high affinity binding sites for EGF without a change in the affinity constant. The changes in EGF binding seemed specific since binding for insulin was not changed by 3 week unilateral nephrectomy. The changes in EGF binding were not correlated with changes in Na-H antiporter activity elicited by unilateral nephrectomy but seemed inversely correlated with changes in renal cortical weight. Our results demonstrate that unilateral nephrectomy is not associated with changes in EGF binding in early stages, but is associated with a decrease in the number of high affinity binding sites after 3 weeks. This suggests that in the steady state, compensatory renal hypertrophy is associated with 'down regulation' of the EGF receptor.  相似文献

Mice with a smooth muscle cell (SMC)-specific deletion of Fibulin-4 (SMKO) show decreased expression of SMC contractile genes, decreased circumferential compliance, and develop aneurysms in the ascending aorta. Neonatal administration of drugs that inhibit the angiotensin II pathway encourages the expression of contractile genes and prevents aneurysm development, but does not increase compliance in SMKO aorta. We hypothesized that multidimensional mechanical changes in the aorta and/or other elastic arteries may contribute to aneurysm pathophysiology. We found that the SMKO ascending aorta and carotid artery showed mechanical changes in the axial direction. These changes were not reversed by angiotensin II inhibitors, hence reversing the axial changes is not required for aneurysm prevention. Mechanical changes in the circumferential direction were specific to the ascending aorta; therefore, mechanical changes in the carotid do not contribute to aortic aneurysm development. We also hypothesized that a published model of postnatal aortic growth and remodeling could be used to investigate mechanisms behind the changes in SMKO aorta and aneurysm development over time. Dimensions and mechanical behavior of adult SMKO aorta were reproduced by the model after modifying the initial component material constants and the aortic dilation with each postnatal time step. The model links biological observations to specific mechanical responses in aneurysm development and treatment.  相似文献

Histidine-rich glycoprotein (HRG) promoted the adhesion and morphological changes of human T-cell line MOLT-4 in a Con A-dependent manner. This morphological change-promoting activity was specific for HRG and the Arg23-Lys66 glycopeptide from human HRG. The carbohydrate chain at Asn45 was essential for this activity. The morphological changes of MOLT-4 cells caused by HRG and Con A (HRG/Con A) were not inhibited by phosphatidylinositol 3-kinase inhibitor, wortmannin or LY294002, while the changes by Con A alone were completely inhibited by these reagents, suggesting that HRG/Con A cooperate to activate leukocytes via a signaling pathway distinct from that by Con A alone. The morphological changes by Con A were associated with pseudopodia like structure. On the other hand, the morphological changes caused by HRG/Con A were associated not only with pseudopodia like structure but also with an increase of the F-actin-rich surface protrusions. Wortmannin inhibited only the formation of pseudopodia like structure.  相似文献

The response of bovine spermatozoa to media of different osmolalities and tonicities was studied by several methods. Packed cell volumes of cells in a dilute medium were greater than in a reference medium (similar to seminal plasma), and were less in a more concentrated medium, but the changes were not correlated quantitatively with the changed properties of the media. Specific effects on the configuration of the midpiece and tail were observed by phase-contrast microscopy. All these effects were elicited by changes in osmolality rather than tonicity. Electronic volume sensing and optical density measurements did not detect these changes in the cells. The morphological changes were not uniform in the cells of any one sample, some cells being unresponsive; in a small proportion of responsive cells the effects of raised osmolality appeared to be reversible.  相似文献

Bacterial chemoreceptors are embedded in the inner cell membrane in tight clusters. We show that changes in receptor methylation that generate large changes in kinase activity have relatively little effect on cluster morphology. Four pups were given three intravenous infusions of microfilariae over a 7-month period to determine if radiographic changes could be detected in the lungs while sensitivity to first stage microfilariae was being induced. Mild pathological changes occurred but these could not be detected on any of the radiographs. Radiographic changes described by others and 'Eosinophilic lungs' did not result from the immune response to the first stage larvae.  相似文献

Endometriosis is a poorly understood, estradiol-dependent condition associated with severe pelvic pains and defined by vascularized endometrial growths outside the uterus. Endometriosis is produced in cycling rats by autotransplanting pieces of uterus onto abdominal arteries where they develop into cysts. The surgery induces vaginal and abdominal muscle hyperalgesia, whose severity is greatest in proestrus and nearly absent in estrus. The cysts contain growth factors and cytokines and develop their own sympathetic and sensory C- and Adelta-fiber innervation. Here, we used quantitative immunostaining and protein array analyses to test the hypothesis that the innervation and growth factor/cytokine content of the cysts, but not uterine horn, contribute to proestrous-to-estrous changes in hyperalgesic severity. If so, these characteristics in the cysts, but not the uterine horn, should change with estrous stage. In cysts, the density of sympathetic (but not sensory) neurites and amounts of NGF and VEGF proteins (but not cytokines IL-1, IL-6, IL-10, or TNF-alpha) were greater in proestrus than estrus. These changes were accompanied by vascular changes. Both sympathetic and sensory fibers in both stages colabeled with TrkA, indicating that changes in NGF could act on both afferent and efferent fibers. In contrast with the cysts, no changes occurred in the uterine horn between proestrus and estrus. Together, these results suggest that coordinated proestrous-to-estrous changes in innervation and vascularization of the cysts contribute to similar changes in hyperalgesic severity. The findings also encourage consideration of endometriosis as a neurovascular condition.  相似文献
