Total root production (∑P), total root loss (∑L), net root production. (NP), and biomass production were determined for seedlings of Betula papyrifera and Acer rubrum in ambient and elevated CO2 environments. ∑P, ∑L, and NP were calculated from sequential, independent observations of root length production through plexiglass windows. Elevated CO2 increased ∑P, ∑L, and NP in seedlings of Betula papyrifera but not Acer rubrum. Root production and loss were qualitatively similar to whole-plant growth responses to elevated CO2. Betula showed enhanced ∑P, ∑L, and biomass with elevated CO2 but Acer did not. However, the observed effects of CO2 on root production and loss did not alter the allometric relationship between root production and root loss for either Acer or Betula. Thus, in this experiment, elevated CO2 did not affect the relationship between root production and root loss. The results of this study have important implications for the potential effects of elevated CO2 on root dynamics. Elevated CO2 may lead to increases in root production and in root loss (turnover) where the changes in root turnover are largely a function of the magnitude of root production increases.  相似文献

Summary Egg production and development of the carabid beetles Notiophilus biguttatus F. and Notiophilus rufipes Curtis were studied in relation to temperature. Adult beetles and larvae were fed springtails of the species Orchesella cincta (Linné). The rate of egg production, at the different temperatures did not differ between the species. As observed in N. biguttatus egg production at a daily fluctuating temperature regime is high compared to egg production at the constant temperatures. Food supply not only has a strong effect on egg production but also on the relation of egg production with temperature, as shown in N. biguttatus. Development times in N. rufipes are longer than in N. biguttatus particularly at low temperatures. The temperature during development has a distinct effect on adult body size. As observed in N. biguttatus body size and fecundity show a strong positive correlation.  相似文献

The zooplankton community in the highly eutrophic Lake Kasumigaura was investigated and its relation to a bloom of Microcystis was analyzed. The zooplankton community was dominated by small cladocerans, whose biomass and production became highest in summer, when Microcystis bloomed. The high cladoceran production is considered to depend on the production of colonial Microcystis, because the production of nannoplankton was apparently too low to ensure the cladoceran production. Microcystis cells were unsuitable as food for the cladocerans inhabiting Lake Kasumigaura, but became utilizable when decomposed. Decomposed Microcystis may be the main food for Cladocera in the lake in summer. High water temperatures occurring in summer probably promoted decomposition of the Microcystis, leading to increased production of the small cladocerans.  相似文献

The effect of different cultivation temperatures (30 and 37 °C) and pH of the media (5.5, 7.5, 8.5) on the biofilm production was compared in Pseudomonas aeruginosa, Klebsiella pneumoniae, and Vibrio cholerae non-O1 and O1 using the crystal-violet test for estimation of quantitative production of the biofilm. Decrease (46.4–98.4 %) in the biofilm production was observed at 37 °C in 8 of the tested strains (P. aeruginosa three strains, K pneumoniae two, V. cholerae non-O1 two, and V. cholerae O1 one strain) compared with the production at 30 °C. On the other hand, five strains (P. aeruginosa 1, K. pneumoniae 3, V. cholerae non-O1 1) exhibited under these conditions a higher biofilm production (103–143 %). However, this difference was not significant (p = 0.196). Increased pH lead to a higher biofilm production using all media tested. In P. aeruginosa the biofilm production at pH 8.5 was 139–244 %, at pH 7.5 136–164 % in comparison with pH 5.5. Similarly, in K. pneumoniae the biofilm production increased to 151–319 % at pH 8.5 while with the drop of pH to 7.5 the biofilm production was 113–177 % compared with pH 5.5. In V. cholerae non-O1 and O1 the biofilm production reached 204–329 % at pH 8.5, and 123–316 % at pH 7.5 (compared with the production at pH 5.5). An increase in biofilm production represented an average of 169 % (p = 0.001) at pH change from 5.5 to 7.5, with the rise of pH from 5.5 to 8.5 caused an average difference of 229 % (p = 0.001).  相似文献

Fig trees ( Ficus spp.) and Agaonine fig‐wasps participate in an obligate mutualism. Fig wasps can only develop within fig inflorescences (syconia) and they are the only organisms capable of pollinating fig flowers. Other non‐pollinating wasps that lay eggs by inserting their ovipositors from the outside can also develop in syconia. These parasitic wasps may be parasitoids of either pollinating or other non‐pollinating wasps, or form galls in fig flowers or other tissues. Depending on this interaction, parasitic wasps may have various effects on the production of pollinating wasps and seeds. Wasps in the genus Idarnes, which parasitize New World figs (subgenus Urostigma), have an effect on wasp production but not on seed production. Heterandrium spp., which have short ovipositors and lay on external flowers, are infrequent and no effect on seed production has been documented. In the Colombian Andes, Idarnes spp. and Heterandrium spp. are the most frequent parasites of the Ficus andicola Pegoscapus sp. mutualism, affecting 62 and 43 percent of syconia, respectively. Controlling for other factors that influence wasp and seed production, such as number of foundresses, syconium size and tree, we found that Idarnes reduced pollinator production by almost half but did not reduce seed production, whereas Heterandrium reduced seed production by 40 percent, and marginally affected pollinator production. Our results provide the first clear documentation of Heterandrium spp. impact on fig seed production. Whether the relative abundance of this genus is a generalized phenomenon in montane forest remains to be determined.  相似文献

The genusScrophularia in the Iberian Peninsula and Balearic Islands comprises two sections,Scrophularia andCanina G. Don. Analyses were carried out on flower production, flower duration and their sexual phases, pollen and nectar production together with observations on their pollinators. Nectar production is correlated with corolla size and pollen production with anther size. The taxa of sect.Scrophularia show greater nectar and pollen production than those of sect.Canina. Also, those of the first section produce more ovules per ovary than those of the second group, production being correlated with the ovary size.Some observations on floral and reproductive biology in some species ofScrophularia from the Iberian Peninsula and the Balearic Islands. Filamentous fungi are important cell factories for large-scale enzyme production. However, production levels are often low, and this limitation has stimulated research focusing on the manipulation of genes with predicted function in the protein secretory pathway. This pathway is the major route for the delivery of proteins to the cell exterior, and a positive relationship between the production of recombinant enzymes and the unfolded protein response (UPR) pathway has been observed. In this study, Aspergillus nidulans was exposed to UPR-inducing chemicals and differentially expressed genes were identified by RNA-seq. Twelve target genes were deleted in A. nidulans recombinant strains producing homologous and heterologous GH10 xylanases. The knockout of pbnA (glycosyltransferase), ydjA (Hsp40 co-chaperone), trxA (thioredoxin) and cypA (cyclophilin) improved the production of the homologous xylanase by 78, 171, 105 and 125% respectively. Interestingly, these deletions decreased the overall protein secretion, suggesting that the production of the homologous xylanase was specifically altered. However, the production of the heterologous xylanase and the secretion of total proteins were not altered by deleting the same genes. Considering the results, this approach demonstrated the possibility of rationally increase the production of a homologous enzyme, indicating that trxA, cypA, ydjA and pbnA are involved in protein production by A. nidulans.  相似文献

Production of extracellular amylase and protease in Vibrio parahaemolyticus was repressed by various carbohydrates present in the medium. In addition, the protease production was repressed very strongly by peptones or casamino acids. Cyclic adenosine 3′, 5′-monophosphate (cyclic AMP) added exogenously could reverse the repression of amylase production, but not that of protease production irrespective of the “repressors” used. Mutants of V. parahaemolyticus, which resembled the reported cya (adenylate cyclase) and crp (cyclic AMP receptor protein) mutants of Escherichia coli and related organisms, were examined for the exoenzyme production. Amylase production in the mutants was defective, while their protease production was not defective, but rather accentuated as compared with that in the parental strain. These findings strongly suggest that amylase production is subject to catabolite repression mediated by cyclic AMP, whereas protease production is controlled by a repression mechanism which mimics in part, but may be distinct from catabolite repression.  相似文献

Native to propionibacteria, the Wood–Werkman cycle enables propionate production via succinate decarboxylation. Current limitations in engineering propionibacteria strains have redirected attention toward the heterologous production in model organisms. Here, we report the functional expression of the Wood–Werkman cycle in Escherichia coli to enable propionate and 1-propanol production. The initial proof-of-concept attempt showed that the cycle can be used for production. However, production levels were low (0.17 mM). In silico optimization of the expression system by operon rearrangement and ribosomal-binding site tuning improved performance by fivefold. Adaptive laboratory evolution further improved performance redirecting almost 30% of total carbon through the Wood–Werkman cycle, achieving propionate and propanol titers of 9 and 5 mM, respectively. Rational engineering to reduce the generation of byproducts showed that lactate (∆ldhA) and formate (∆pflB) knockout strains exhibit an improved propionate and 1-propanol production, while the ethanol (∆adhE) knockout strain only showed improved propionate production.  相似文献

Aims: To investigate the transmission of Salmonella spp. between production animals (pigs and cattle) and wildlife on production animal farms in Denmark. Methods and Results: In the winter and summer of 2001 and 2002, 3622 samples were collected from Salmonella-infected and noninfected herds of pigs and cattle and surrounding wildlife. Salmonella was detected in wildlife on farms carrying Salmonella-positive production animals and only during the periods when Salmonella was detected in the production animals. The presence of Salmonella Typhimurium in wild birds significantly correlated to their migration pattern and food preference. Conclusions: Salmonella was transmitted from infected herds of production animals (cattle and pigs) to wildlife that lived amongst or in close proximity to them. Significance and Impact of the Study: Salmonella in animal food products is associated with the occurrence of Salmonella in primary animal production. Strategies to control the introduction and spread of infection should include wildlife management, as the nearby wildlife may act as reservoirs for Salmonella spp. and/or may be passive carriers of the bacteria.  相似文献

The purpose of this study was to develop and validate a habitat-specific production simulation model to quantify annual benthic microalgal production in North Inlet estuary, South Carolina. Using hourly measurements of incident irradiance during 1990–1991 as the forcing function, the simulation model was used to obtain hourly estimates of areal benthic microalgal gross primary production in five habitat types. The model, which was validated using actual measurements of production, showed good (r2= 0.63, P < 0.001) agreement between observed and predicted production in the short Spartina alterniflora Loisel zone habitats showed the highest mean hourly production (61.1 mg C m?2 h?1) while intertidal mudflats had the maximum hourly rate (166.9 mg C m?2 h?1). Daily production was highly variable, primarily due to daily fluctuations in irradiance. Annual estimates of habitat-specific production were multiplied by the mates of habitat-specific production were multiplied by the known area of each habitat type to determine total microalgal production for the estuary (3.423 × 109 g C yr?1). Short Spartina zone habitats provided 45% of total microalgal annual production, followed by intertidal mudflats (22%), tall Spartina zones (18%), shallow subtidal (13%) and microalgal production exceeds phytoplankton and microalgal production but is less than Spartina production.  相似文献

The fungus Fusarium oxysporum Schlecht f. sp. tulipae Apt. can produce ethylene abundantly in vitro when grown in Pratt's liquid medium with glucose as the only organic substrate. This production starts after a lag phase of about 4 days, and peak production occurs when mycelium weight has reached its maximum value. For several days the rate of production is more or less linearly dependent on pO2. The total production is also dependent on the oxygen concentration, but pure oxygen inhibits the total production by about 50% as compared with 21% oxygen. The high production in shake cultures, as compared with the low production in stagnant cultures, is probably the result of a better oxygen supply in the culture medium. The mycelium weight proved not to be a valid referential basis for the production of ethylene.  相似文献

Summary Protein-secreting procaryotic host organisms are currently being sought as alternatives to Escherichia coli for recombinant processing. In this study we examined how manipulation of the cultivation conditions can enhance heterologous protein production by Streptomyces lividans. The recombinant S. lividans used in this study expressed and excreted a Flavobacterium enzyme capable of hydrolyzing organophosphates. Initial shake-flask studies demonstrated that supplementing Luria-Bertani medium with moderate amounts of glucose (30 g/l), led to improved enzyme production. In fermentor studies with controlled pH, a further twofold increase in production was observed when glucose was fed continuously as compared to batch cultivation. This improved production in the glucose-fed culture may be related to a reduced accumulation of acids. Continuous feeding of both glucose and tryptone led to a further sixfold increase in production. In addition to enhancing production 25-fold, the efficiency of enzyme production and the specific activity of the excreted enzyme were also improved by glucose and tryptone feeding. These results demonstrate that in addition to genetic manipulations, optimization of cultivation conditions can lead to significant improvements in the production of heterologous proteins from Streptomyces. Turkey’s natural and ecological situations are very suitable for aquaculture. Turkey also has a wide variety of freshwater and marine species comprising trout, carp, sea bass, sea bream, turbot, mussel, crayfish, etc. The total production of fish and shellfish was 646,310 tons in 2008. The contribution of freshwater catch to total fishery production is relatively small. Capture fisheries production amounted to 494,124 tons whilst aquaculture production was 152,186 tons in the same year. In Turkey, Engraulis encrasicholus (anchovy) is the main caught sea fish species. Fisheries in the Black Sea are the most important fishery by far and show the greatest variations in total catch. Alburnus tarichii (a local species belonging to Cyprinidae) and Cyprinus carpio (the common carp) are the most important species caught from freshwaters. Aquaculture is going to play an increasingly important role in the Turkish economy, as fishery products are the only products of animal origin that can be exported to the EU. There has been a fast increase in the aquaculture production in Turkey with the implementation of scientific and technological modernization. For example, total aquaculture production for 1986 and 2008 was 3,075 and 152,186 tons, respectively. The percentage of aquaculture in total fish production has been rising every year. The ratio of cultured fish production to total fish production was 1.5% in 1990 s, 13.57% in 2000 and more than 20% in 2005. It was 23.55% in 2008. Trouts are the main cultured freshwater fish species. Raceways and floating cages are employed in culture of trout. Carps are also important cultured freshwater fish species. Sea bass and gilthead sea bream are grown marine fish species. Floating cages, off-shore and earthen ponds are used for marine fish species culture. There has been an increase in fishery exports and imports in recent years. It was more than US500 million in 2008, but that of 2004 was just over US 500 million in 2008, but that of 2004 was just over US 233 million. However, aquaculture production is still far away from the production targets and fisheries sector is not an important part of the economy at present.  相似文献

The combined effects of grazing and nutrient regeneration by Daphnia and Eudiaptomus on the growth of Rhodomonas and heterotrophic bacteria was assessed experimentally. The responses of Rhodomonas and bacteria to the grazers were measured as net specific growth rate over the entire experimental periods, as well as production and specific production at the end of the experiments. Both zooplankton species had a negative effect on Rhodomonas net specific growth rate due to grazing and a positive effect on specific primary production due to nutrient regeneration. Daphnia had no effect on bacterial net specific growth rate, bacterial production or specific bacterial production in one of two experiments. In the other experiment, however, both bacterial growth rate and production decreased as a result of grazing. Furthermore, Daphnia had a negative effect on specific bacterial production, but Eudiaptomus had a positive effect on all bacterial parameters due to nutrient regeneration, probably of phosphorus. Positive effects of copepods on bacterial growth has previously been attributed to trophic cascades via protozoa. However, the present experiments show that regeneration of nutrients, especially phosphorus, may account for a large part of the stimulation of bacterial growth.  相似文献

We investigated the in vivo relevance of the impact of sarA and saeRS on protease production using derivatives of the USA300 strain LAC. The results confirmed that mutation of saeRS or sarA reduces virulence in a bacteremia model to a comparable degree. However, while eliminating protease production restored virulence in the sarA mutant, it had little impact in the saeRS mutant. Additionally, constitutive activation of saeRS (saeRSC) enhanced the virulence of LAC and largely restored virulence in the isogenic sarA mutant. Based on these results, together with our analysis of the representative virulence factors alpha toxin, protein A (Spa), and extracellular nucleases, we propose a model in which the attenuation of saeRS mutants is defined primarily by decreased production of such factors, while constitutive activation of saeRS increases virulence, and reverses the attenuation of sarA mutants, because it results in both increased production and decreased protease‐mediated degradation of these same factors. This regulatory balance was also apparent in a murine model of catheter‐associated infection, with the results suggesting that the impact of saeRS on nuclease production plays an important role during the early stages of these infections that is partially offset by increased protease production in sarA mutants.  相似文献

SUMMARY 1. A 2‐year study of the life histories, production dynamics and resource utilisation of five mayfly species was undertaken in two forest streams in Hong Kong [Tai Po Kau Forest Stream (TPKFS) and Shing Mun River (SMR)]. Afronurus sp. and Cinygmina sp. (Heptageniidae), Procloeon sp. and Baetiella pseudofrequenta (Baetidae), and Choroterpes sp. (Leptophlebiidae) were abundant in both streams and contributed more than 50% of the total mayfly populations. 2. All species had asynchronous larval development with recruitment occurring throughout the year. Mean annual production (all mayflies combined) was 3.1 and 2.0 g dry weight m?2 year?1 in SMR and TPKFS, respectively – the higher value at SMR reflecting greater mayfly densities – with more than 70% of production occurring during the wet season. Mayfly production varied between years, decreasing by 5% in TPKFS and 43% in SMR during 1996–97, reflecting lower densities of heptageniids relative to 1995–96. Annual biomass turnover rates (P/B) were high in both sites ranging from 27.2 to 94.6 in TPKFS (Cinygmina sp. and Procloeon sp.) and from 31.8 to 109.8 in SMR (Cinygmina sp. and B. pseudofrequenta). 3. Patterns of daily production in both streams showed that Afronurus sp., Cinygmina sp. and Choroterpes sp. were most productive during the wet season, while Procloeon sp. maintained high production levels throughout the year. The highest daily production of B. pseudofrequenta occurred during the wet season in TPKFS, but in the dry season at SMR. Temporal overlap in production and hence resource utilisation in both streams, calculated using the proportional similarity index (PS), ranged from 0.39 to 0.81. It was highest (0.63–0.81) between pairs of species of Heptageniidae and Baetidae, and lowest between Choroterpes sp. and other mayflies (0.39–0.61). No clear temporal segregation was observed among any species. However, when using the fraction of production attributable to each food, lower PS values were obtained for all species in both sites. In SMR, trophic segregation may have occurred between the two species pairs Procloeon sp.–Cinygmina sp. and Procloeon sp.–Choroterpes sp. (PS=0.17 and 0.03, respectively). 4. A combination of production data and information on the stable isotope signature of mayflies revealed that, during both the wet and dry seasons, more than 50% of total mayfly production in TPKFS was derived from autochthonous foods. In SMR, 68% of production was supported by allochthonous foods during the wet season, and 72% by autochthonous sources in the dry season. Considering that more than 70% of the total production occurred in the wet season, the trophic basis of mayfly production in SMR is mostly allochthonous (58%) while in TPKFS it is mainly of autochthonous origin (66%). The year‐round importance of autochthonous foods in shaded streams such as TPKFS is surprising, but the wet season contribution of allochthonous foods (especially in SMR) may have resulted from depletion of algal biomass during spates.  相似文献
