Growth rates of two clones of the freshwater planktonic diatom Asterionella formosa Hass. were measured under conditions in which external silicon concentrations controlled growth. Clone AfOH2 from Lake Ohrid, Yugoslavia, had a higher maximum growth rate (μmax= 1.11 doublings/day) and apparent half-saturation constant (Ksi] + Sio= 1.93 μM Si) than clone L262 from Lake Windermere, England. (μmax= 0.61 doublings/day; Ksi+ Sio= 1.09 μM Si). Klim, the silicon concentration at μ= 0.9 μmax, is 13.8 μM Si for clone AfOH2 and 6.5 μM Si for clone L262. These values agree well with published field observations showing A. formosa populations decreasing below 0.5 mg/l SiO2 (= 8.4 μM Si). Calculations of yield gave a range of 0.5–1.5 μM Si/106 cells for clone AfOH2 and 0.6–1.9 μM Si/106 cells for clone L262.  相似文献

Tracer techniques using the stable isotope 30Si were used to measure rates of silicic acid uptake and silica dissolution in silicon replete and silicon depleted populations of 2 clones of the marine diatom Thalassiosira pseudonana Hasle & Heimdal. Uptake kinetics were describable using the Michaelis-Menten equation for enzyme kinetics, and no threshold concentration for uptake was evident. The maximum specific uptake rate of the estuarine clone 3H (0.062–0.092 · h?1) was higher than that of the Sargasso Sea clone 13-1 (0.028–0.031 · h?1), but half-saturation constants for uptake by the 2 clones were not measurably different (0.8–2.3 μM for 3H; 1.4–1.5 μM for 13-1). There was little or no light dependence of uptake in populations grown under optimal light conditions prior to the experiment. Exponentially growing populations released silicic acid to the medium by dissolution of cellular silica at rates ranging from 6.5 to 15% of the maximum uptake rate.  相似文献

A silicon requirement for growth is demonstrated for an alga other than a diatom. Concentrations of less than 1 μM silicate greatly decreased the growth rate of Synura petersenii Korshikov and caused morphological changes. Half of maximum growth rate (μmax= 1.12 divisions/day) appeared at a concentration of only 0.23 μM silicate. Germanium dioxide inhibited, growth considerably; the degree of inhibition varying from none to almost 100% depending upon both Si and Ge concentrations. The amount of silicon deposited in the external scales of S. petersenii is comparable per unit area to that of diatoms. S. petersenii is able to deplete the medium of silicate to very low levels. The feasibility of batch culture techniques for this kind of work is discussed briefly.  相似文献

The effect of temperature on the silicon limited growth and nutrient kinetics of Stephanodiscus minutus Grun. was examined using batch and semicontinuous culture methods. Short-term batch culture methods gave maximum growth rates which were essentially constant over the temperature range of 10° to 20°C (μ3= 0.71–0.80 d?1). The half-saturation constant for growth (Ks) was significantly lowest at 10°C (Ks= 0.31 μM Si; 0.22–0.41), and higher at both 15°C (Ks= 1.03 μM Si; 0.68–1.47) and 20°C (Ks= 0.88 μM Si; 0.60–1.22). Two methods were used to evaluate the semicontinuous experiments. The Droop relationship showed that the minimum cell quota was about 1.50 × 10?7 nmol Si cell?1, but there was much overlap in the results at all three temperatures. The Monod growth relationship for the semicontinuous experiments gave estimates of Ks which were lowest at 15°C (Ks= 0.12 μM Si), and higher at 10°C (Ks= 0.68 μM Si) and 20°C (Ks= 1.24 μM Si), although 95% confidence intervals overlapped. The maximum growth rate estimates for the semicontinuous experiments were similar at 10° and 15°, and higher at 20°C, but the number of points used in making the calculations makes the results less reliable than those from batch cultures. Generally, there were no consistent significant differences in the silicon limited growth of S. minutus over the temperature range studied. Our values of Ks for S. minutus are the lowest recorded for a freshwater diatom, and are consistent with the distribution of this species in nature. Generally, this species becomes abundant in areas with high phosphorus loading and very low silicon levels (low Si:P loading rates). Stephanodiscus species are also fossil indicators of eutrophication in north temperate lakes.  相似文献

Phaeodactylum tricornutum Bohlin, the one diatom known to lack a silicon requirement for growth, and the prasinophyte Platymonas sp. are two representatives of a taxonomically diverse group of planktonic algae that have been reported to take up Si without a demonstrable requirement for the element. For both species, removal of Si from solution during growth in batch culture has at least two components; true biological uptake throughout the growth of the culture, and spontaneous inorganic precipitation of a solid silicate phase–probably Mg2Si3O8 (sepiolite)–under the elevated pH conditions that prevail late in batch growth. It is not clear to what extent previous observations of Si uptake by algae without siliceous frustules may be influenced by inorganic, non-cellular precipitation. The kinetics of true cellular uptake of Si are similar in Phaeodalylum and Platymonas, and different from those reported for the Si-requiring diatoms. Uptake follows hyperbolic saturation kinetics in both species, with half-saturation concentrations of 97.4 μM in Phaeodactylum and 80.9 μM in Platymonas, as compared to ca. 1–6 μM in diatoms that form siliceous frustules. Uptake by Phaeodactylum and Platymonas is not substrate-saturated until the dissolved Si concentration of the medium exceeds 200 μM. Concentrations this high do not occur in the surface layer of the ocean, and the kinetics suggest that both species deposit much less silica in nature than they can be induced to deposit in culture.  相似文献

Silicon uptake kinetics of the diatom Phaeodactylum tricornutum (Bohlin) were examined at pH 8.8 ± 0.1 and pH 9.1 ± 0.1. Uptake follows hyperbolic saturation kinetics at both pH's, but at the higher pH the half-saturation constant for uptake is 11.8 μM, as opposed to 54.8 μM at the lower pH. When the uptake rate is examined as a function of the calculated concentration of the monovalent conjugate base, SiO(OH)3?, the half-saturation constant for uptake is 6.6 μM at either pH.  相似文献

The blue-green alga (Cyanobacterium) Synechococcus leopoliensis (Racib.) Komarek was grown in dissolved inorganic carbon [DIC]-limited chemostats over the entire range of growth rates. At each growth rate, the kinetics of photosynthesis with respect to [DIC] and the maximal rate of photosynthesis (Pmax) were determined. The half-saturation constant for [DIC]-limited photosynthesis (K1/2DIC) for cells growing below 1.7 d?1 was constant (4.7 μM) whereas for growth rates between 1.7 d?1 and 2.1 d?1max) the kinetics of photosynthesis were multiphasic with an apparent K1/2DIC between 1.5–2.0 mM. Pmax increased in a linear fashion with growth rate for growth rates below 1.7 d?1. No trend in Pmax was apparent for growth rates greater than 1.7 d?1. These kinetic parameters were used to predict a growth rate versus [DIC] relationship. Results show that the Monod relationship is a physiologically valid expression of growth as a function of [DIC] provided (K1/2DIC) remains constant. The major change in (K1/2DIC) as μ approaches μmax results in the conclusion that two separate and distinct Monod equations must be used to describe growth as a function of DIC over the entire growth range. These results point to a major discontinuity in the μ vs. [DIC] curve at 1.7 d?1 which corresponds to the change from high to low affinity photosynthetic kinetics. We believe these results account for the previously described deficiencies of the Monod equation in describing [DIC]-limited algal growth.  相似文献

We used batch cultures of three strains of the unicellular synurophyte Mallomonascaudata to investigate the effects of nitrate, phosphate, silicate and light intensity on population growth and growth rate. The three strains were isolated from three different reservoirs in Kyungpook Province, Korea. For all three strains, we observed high population growth under all nutrient concentrations studied, except at nitrate concentration below 0.8 μM. The maximum growth rate (μmax) occurred at 8.2 μM or 16.5 μM nitrate, depending on the strain, and at 11.5 μM phosphate. Silicate concentration had no effect on growth rate. With respect to light intensity, the maximum population growth and maximum growth rates (μmax) occured between 42 and 104 μmol m?2 s?1 depending on strain and culture temperature. Population growth of these three strains under batch culture occurred over a wide range of nutrient and light intensities, but there seemed to be strain‐specific differences that may represent adaptations to local environments.  相似文献

The biochemical properties of Spirulina platensis in an internally illuminated photobioreactor (IlPBR) were investigated under different light-emitted diode (LED) wavelengths; blue (λmax= 450 and 460 nm), green (λmax= 525 nm), red (λmax = 630 and 660 nm), and white (6,500K), with various light intensities (200, 500, 1,000, and 2,000 μmol/m2/sec) were examined. The highest specific growth rate, maximum biomass, and phycocyanin productivity occurred under the red LEDs (0.39/day, 0.10 g/L/day, and 0.14 g/g-cell/day, respectively) at 1,000 μmol/m2/sec; the lowest growth rate was obtained under blue LEDs. Indeed, the size of trichomes was changed into short form under blue LEDs at all light intensities or all LEDs at 2,000 μmol/m2/sec for the first 2 days after inoculation, and S. platensis did not grow in the IlPBR under the dark condition. These results provide a base for different approaches for designing the pilot scale photobioreactor and developing cost-effective light sources.  相似文献

The characteristics of phosphate uptake and photosynthetic capacity were studied in P-limited populations of Euglena gracilis Klebs (Z), using both P-limited batch cultures in stationary phase and cyclostat cultures grown on 14:10 LD. P uptake obeyed Michaelis-Menten kinetics between 0 and 150 μM PO4 under both growth conditions. The value of Vmax was 35% lower in the dark than in the light in the stationary phase cells. The value of K8 was not affected by light conditions, and uptake was completely inhibited in the presence of 1 mm KCN. P uptake (at 2.0 μM PO4) and photosynthetic capacity showed diel periodicity with peak rates occurring just before the beginning of the dark period for P uptake, and 8 h into the light period for photosynthetic capacity. Vmax for P uptake increased by a factor of 1.5 over the light period, whereas K8 remained constant at 1.4 μM PO4. These patterns were displayed by both nondividing stationary phase cells and populations in which less than a third of the cells divided each day, indicating that the rhythmicity is not coupled to cell division.  相似文献

Amphora coffeaeformis (Ag.) Kütz. var. perpusilla (Grun.) Cleve took up glucose by an inducible transport system. The system was induced by d -fructose, d -mannose, as well as glucose. Some d -pentoses also induced a glucose uptake system but it may not be the same one as that induced by hexose. d -fructose, d -mannose and 2-deoxy-d -glucose inhibited 2 mM glucose uptake at equimolar concentration, but d -pentoses did not. The uptake system decayed in ca. 5 h in the absence of glucose. The half-saturation constant for uptake, K8 was ca. 0.1 mM glucose with a maximum uptake rate, Vmax= 0.4 nmol/106 cells-min?1.  相似文献

Short-term (within 5 min) and long-term (≤2 h) rates of nitrate uptake were determined for the marine diatoms, Nitschiella longissima (Cleve), Skeletonema costatum (Greville) Cleve and Asterionella japonica (Cleve). Pigment levels, cell carbon, nitrogen and cell volume were also determined for cells in the logarithmic and stationary phases of growth. For each species, one clone isolated from oligotrophic coastal water and one clone isolated from eutrophic coastal water were compared. Long-term NO3? uptake typically followed saturation kinetics describable by the Michaelis-Menten expression. Under experimental conditions, half-saturation constants ranged from 0.6 to 2.2 μM NO3?. Generally, the oligotrophic clones had lower Ks and Vmax (on a per cell basis) than their eutrophic counterparts, though this was only statistically significant in one pair of clones. Eutrophic and oligotrophic clones also differed in their short-term response to nutrient addition; oligotrophic clones showed greatest rate of uptake at the lowest nitrate addition while uptake by eutrophic clones increased with increasing nitrate concentration. However, all clones had very similar Vmax values expressed on a dry weight basis. Under N-starvation, cellular C and pigment levels (and N to a lesser extent) generally declined more in eutrophic than in oligotrophic clones. While the differences between inshore and offshore clones were not great, the results are consistent with the hypothesis that eutrophic waters support algae which grow faster and are less conservative biochemically than cells in oligotrophic waters.  相似文献

The applicability of Bordetella sp. Sulf-8 to degrade Hydrogen Sulfide (H2S) gas in a biotrickling system was investigated. The isolate is a heterotrophic gram-negative, catalase- and oxidase-positive, rod-shaped bacterium which can metabolize thiosulfate or sulfide into sulfate. The mesophilic Bordetella sp. Sulf-8 can grow within a wide pH range using yeast as carbon source, with or without the presence of sulfur. In batch experiments, kinetic constants such as maximum specific growth rate (μ max = 0.12 1/h), saturation constant (K S = 0.017 g/L), and specific sulfur removal rate (88 mg S/g cells h) were obtained. In biotrickling experiments removal efficiencies were satisfactory, but the system performance was observed to be more influenced by empty bed residence time than by H2S feed gas concentration. Critical and maximum elimination capacities were 78.0 and 94.5 g H2S/m3 day, respectively. Macrokinetic analysis of the biotrickling system revealed maximum H2S removal rate V max = 15.97 g S/kg media-day and half saturation constant K S′ = 12.45 ppmv.  相似文献

Heterocapsa circularisquama is a harmful dinoflagellate whose first bloom in Hiroshima Bay, Japan, appeared in 1992. As suggested by the authors’ group, in the Seto Inland Sea including Hiroshima Bay, oligotrophication particularly the reduction of phosphate starting 1980 is severe. The bloom caused serious damage to the bay's extensive oyster culture. In the present study, the uptake kinetics of nitrate, ammonia, and phosphate by this species were experimentally investigated. The maximum uptake rate (ρmax) and the half‐saturation constant (Ks) were 0.41 pmol cell?1 h?1 and 4.45 μM, respectively, for nitrate, 2.02 pmol cell?1 h?1 and 11.1 μM for ammonium, and 0.079 pmol cell?1 h?1 and 1.79 μM for phosphate. The maximum specific uptake rates (Vmax) for nitrate, ammonia, and phosphate were estimated to be 8.95, 44.1, and 21.3 day?1, respectively. A comparison of Vmax/Ks, which is also an index of affinity to nutrients, between this species and others suggested that H. circularisquama can utilize nitrate and ammonia efficiently, but not phosphate. Considering both reports describing that H. circularisquama has the ability to utilize dissolved organic phosphorus (DOP) and the DOP concentration is higher than phosphate in Hiroshima Bay, it was concluded that H. circularisquama became dominant due to the phosphate reduction measure.  相似文献

In this report the biokinetic parameter μmax, m and Ymax for the denitrification of a medium containing acetat as C-source and a soft coal gasification water containing volatile fatty acids as main components have been compared. A model of growth and substrate utilization for ferrous-iron-oxidizing bacteria attached to the disks of a rotating biological contactor was developed and tested. The model describes attached bacterial growth as a saturation function in which the rate of substrate utilization is determined by a maximum substrate oxidation rate constant (P), a half-saturation constant (Ks), and the concentration of substrate within the rotating biological contactor (S1). The maximum oxidation rate constant was proportional to flow rate, and the substrate concentration in the reactor varied with influent substrate concentration (S0). The model allowed the prediction of metabolic constants and included terms for both constant and growth-rate-dependent maintenance energies. Estimates for metabolic constants of the attached population of acidophilic, chemolithotrophic, iron-oxidizing bacteria limited by ferrous iron were: maximum specific growth rate (μmax), 1.14 h−1; half-saturation constant (Ks) for ferrous iron, 54.9 mg/liter; constant maintenance energy coefficient (m1), 0.154 h−1; growth-rate-dependent maintenance energy coefficient (m′), 0.07 h−1; maximum yield (Yg), 0.063 mg of organic nitrogen per mg of Fe(II) oxidized.  相似文献

A mathematical model was developed to describe the biodegradation kinetics of perchlorate in the presence of nitrate and oxygen as competing electron acceptors. The rate of perchlorate degradation is described as a function of the electron donor (acetate) degradation rate, the concentration of the alternate electron acceptors, and rates of biomass growth and decay. The kinetics of biomass growth are described using a modified Monod model, and inhibition factors are incorporated to describe the influence of oxygen and nitrate on perchlorate degradation. In order to develop input parameters for the model, a series of batch biodegradation studies were performed using Azospira suillum JPLRND, a perchlorate-degrading strain isolated from groundwater. This strain is capable of utilizing oxygen, nitrate, or perchlorate as terminal electron acceptors. The maximum specific growth rate (μmax) and half-saturation constant (K S don) for the bacterium when utilizing either perchlorate or nitrate were similar; 0.16 per h and 158 mg acetate/L, respectively. However, these parameters were different when the strain was growing on oxygen. In this case, μmax and K S don were 0.22 per h and 119 mg acetate/L, respectively. The batch experiments also revealed that nitrate inhibits perchlorate biodegradation by this strain. This finding was incorporated into the model by applying an inhibition coefficient (K i nit) value of 25 mg nitrate/L. Combined with appropriate groundwater transport models, this model can be used to predict perchlorate biodegradation during in situ remediation efforts.  相似文献

Three diatom species, Stephanodiscus hantzschii (Ehr.) Grun., Asterionella formosa Hass. and Fragilaria crotonensis Kitt. Hass. were isolated from Lake Maarsseveen where they are dominant and show a successional sequence. The physiological responses of each species to temperature and limitation by silicon and phosphorus were determined over the temperature range of 5° to 20° C using short-term batch culture methods. Stephanodiscus hantzschii had higher maximum growth rates than the other two species at all temperatures, and the maximum growth rates of all species increased with increasing temperature. Temperature affected not only maximum growth rates but also half-saturation constants (Ks) and the minimum cell quotas. S. hantzschii had low silicon requirements for growth under Si-limiting conditions, and A. formosa and F. crotonensis had higher and nearly identical silicon requirements. The Ks values for silicon for S. hantzschii were essentially constant from 5° to 20° C but varied greatly for the other two species. A. formosa had the lowest requirements for growth under phosphorus limitation, F. crotonensis was intermediate and S. hantzschii had the highest growth requirements for phosphorus. The K1 values for phosphorus were constant over the temperature range for both A. formosa and F. crotonensis and were much higher and variable for S. hantzschii. Nutrient competition experiments were performed in continuous cultures at four temperatures and various Si:P ratios. The results generally, but not always, confirmed the predictions based on the Monod relationships for each species. Results not in agreement with predictions were usually because of similar physiological properties of A. formosa and F. crotonensis or because of decreased loss rates for F. crotonensis due to wall growth. In cultures with all three species phosphorus-limited (Si:P > 75), A. formosa often dominated as predicted, although F. crotonensis was sometimes the most abundant species. As predicted, S. hantzschii never dominated at high Si:P ratios. At intermediate Si:P ratios when A. formosa and F. crotonensis were both Si-limited and S. hantzschii P-limited, all three species coexisted because A. formosa and F. crotonensis have almost identical silicon requirements, although sometimes F. crotonensis was more abundant than predicted. At 10°C the results agreed best with the predictions; A. formosa dominated at high Si:P ratios and S. hantzschii dominated as predicted at low Si:P ratios when all three species were Si-limited.  相似文献

Uptake and assimilation of nitrogen and phosphorus were studied in Olisthodiscus luteus Carter. A diel periodicity in nitrate reductase activity was observed in log and stationary phase cultures; there was a 10-fold difference in magnitude between maximum and minimum rates, but other cellular features such as chlorophyll a, carbon, nitrogen, C:N ratio (atoms) · cell?1 were less variable. Ks values (~2 μM) for uptake of nitrate-N and ammonium-N were observed. Phosphorus assimilated · cell?1· day?1 varied with declining external phosphorus concentrations; growth rates <0.5 divisions · day?1 were common at <0.5 μM PO4-P. Phosphate uptake rates (Ks= 1.0–1.98 μM) varied with culture age and showed multiphasic kinetic features. Alkaline phosphatase activity was not detected. Comparisons of the nutrient dynamics of O. luteus to other phytoplankton species and the ecological implications as related to the phytoplankton community of Narragansett Bay (Rhode Island) are discussed.  相似文献

Silicic acid transport was studied in the photosynthetic diatom Navicula pelliculosa (Bréb.) Hilse using [68Ge] germanic acid (68Ge(OH)4) as a tracer of silicic acid (Si(OH)4). The initial uptake rate of Si(OH)4 was dependent on cell number, pH, temperature, light and was promoted by certain monovalent cations in the medium. Na+ was more effective than K+, whereas Li+ and NH+4 were ineffective at promoting uptake. Uncouplers and inhibitors of oxidative phosphorylation and of photophosphorylation reduced uptake by 40–99% of control values. Uptake was also especially sensitive to the sulfhydryl blocking agents at 10?5 M and to the ionophorous compound valinomycin (10?7 M) which inhibited uptake by 82%. The Si(OH)4 transport system displayed Michaelis-Menten-type saturation kinetics with kinetic parameters of KS= 4.4 p. mol Si(OH)4· 1?1, Vmax= 334 pmol Si(OH)4· 106 cells?1· min?1. Calculations of the acid soluble silicic acid pool size based on 60 s uptake at 20 μM Si(OH)4 suggested that intracellular levels of Si could reach 20 mM and as much as 5 mM could exist as free silicic acid, representing maintenance of a 250-fold concentration gradient compared with the medium. Efflux from preloaded cells was dependent on temperature and the Si(OH)4 concentration of the external medium. In the presence of 100 μMM “cold” Si(OH)4, approximately 30% of the Si(OH)4 in preloaded cells was exchanged in 20 min. The initial uptake rate of Si(OH)4 in logarithmic phase cells was constant, but the uptake rate increased in a linear fashion for 6 h in stationary phase cells. These results suggest that the first step in silica mineralization by diatoms is the active transmembrane transport of Si(OH)4 by an energy dependent, saturable, membrane-carrier mechanism which requires the monovalent cations Na+ and K+ and is sensitive to sulfhydryl blocking agents. Silicic acid transport activity also appears to be regulated during different growth stages of the diatom.  相似文献
