SYNOPSIS. Two morphologically distinct types of intranuclear inclusions are found in ameboid cells of the protostelid mycetozoan Protostelium zonatum. One type of inclusion is a coiled tubular structure which in cross section appears as cisternae and oval to elliptical vesicles 40–60 nm in diameter. These tubular and vesicular structures are formed by a unit membrane that is connected directly with the inner nuclear membrane. The other type of inclusion is a membrane-bound structure that contains amorphous and/or fibrous material. These inclusions usually are present at several locations in a nucleus. No similar structures occur in the cytoplasm.  相似文献

The first data of Protostelids from the southwest of Europe are presented in this paper. A total of 21 species were identified from samples collected in Somiedo Biosphere Reserve (Spain). This is the highest species richness recorded to date for Europe or for a latitude this high (>40°). Seven species (Cavostelium apophysatum, Endostelium zonatum, Microglomus paxillus, Protosporangium fragile, Protostelium okumukumu, Soliformovum expulsum and Schizoplasmodiopsis micropunctata) are new records for Europe. Approximately 65 % of the microscopic protostelid species described in the world have been found in this Reserve, a fact that increases the biological value of this protected area and suggests that Spain is an excellent location to study this group. A microhabitat study has been carried out finding differences in species composition and abundance between ground litter, aerial litter, and bark substrates. Comments on the distribution and ecology of the species, as well as illustrations of some species are included.  相似文献

Spermatogenesis and the morphology of mature sperm in the free-living chromadorid Paracyatholaimus pugettensis from the Sea of Japan were studied using transmission electron microscopy. In spermatocytes fibrous bodies (FBs) appear; in spermatids, the synthetic apparatus is located in the residual body, whereas the main cell body (MCB) houses the nucleus, mitochondria, and FBs. The nucleus of the spermatid consists of a loose fibrous chromatin that is not surrounded by a nuclear envelope; centrioles lie in the perinuclear cytoplasm. The plasma membrane of the spermatid MCB forms numerous filopodia. Immature spermatozoa from the proximal part of the testis are polygonal cells with a central nucleus. The latter is surrounded by mitochondria and FBs with poorly defined boundaries. The immature spermatozoa bear lamellipodia all along their surface. Mature spermatozoa are polarized cells with an anterior pseudopodium, which is filled with filaments that make up the cytoskeleton; the MCB houses a nucleus that is surrounded by mitochondria and osmiphilic bodies. In many ultrastructural characteristics, the spermatozoa of P. Pugettensis are similar to those of most nematode species studied so far (i.e., they are ameboid, have no acrosome, axoneme, or nuclear envelope). On the other hand, as in other chromadorids, no aberrant membrane organelles were observed during spermatogenesis of P. ABSTRACT. Electron microscope observations of the trophic and cyst stages of the protostelid Protostelium pyriformis Olive & Stoianovitch, 1969 indicate a perinuclear golgi-vesicular region in the amoeboid cell. Within this region is a microtubule organizing center (MTOC) and associated microtubules which radiate into the cytoplasm. The cyst wall is revealed as having three distinct regions, certain areas of the wall possibly corresponding to exit pores. Protostelium pyriformis shares many similarities in trophic cell structure with other eumycetozoans and members of the non-mycetozoan family Acanthamoebidae.  相似文献

Changes in mutagenicity during crude oil degradation by fungi   总被引:5,自引:0,他引:5  
Two fungal strains, Cunninghamella elegans and Penicillium zonatum, that grow with crude oil as a sole carbon source were exposed to three crude oils that exhibit a range of mutagenic activity. At regular time intervals following fungal incubation with the various crude oils, extracts were tested for the presence of mutagenic activity using the spiral Salmonella assay. When the most mutagenic of the oils, Pennsylvania crude oil, was degraded by C. elegans or by P. zonatum, its mutagenicity was significantly reduced; corresponding uninoculated (weathered) controls of Pennsylvania crude remained mutagenic. West Texas Sour crude oil, a moderately mutagenic oil, exhibited little change in mutagenicity when incubated with either C. elegans or P. zonatum. Swanson River Field crude oil from Cook Inlet, Alaska is a slightly mutagenic oil that became more mutagenic when incubated with C. elegans; weathered controls of this oil showed little change in mutagenicity. Mycelial mat weights measured during growth on crude oils increased corresponding to the biodegradation of about 25% of the crude oil.  相似文献

Five morphological categories of hemocyte (prohemocyte, ameboid plasmatocyte, spindle-shaped plasmatocyte, oenocytoid, and granular cell) were observed by light and electron microscopy of hemolymph from Tipula paludosa larvae. In addition, vast numbers of membrane-bounded granule-containing fragments were present, and were also found in the larval hemolymph of four other Tipula spp. The fragments appear to be derived from the granular cells, which readily fragment. Granules in the granular cells and the fragments appear to have similar substructure. Melanization and coagulation of the hemolymph occur rapidly on exposure to air; the granular cell fragments may be concerned with one or both of these processes.  相似文献

Abstract

Embryological researches on SENECIO VULGARIS L. var. THYRRENUS Fiori. — Male gametophyte, development of tapetal cells and female gametophyte have been studied in Senecio vulgaris L. var thyrrenus Fiori.

1) The development of male gametophyte is normal. Divisions of the microspore mother cells are of the simultaneous type. The division of the generative nucleus has never been observed till the pollen grain was in the anther.

2) The tapetal cells follow a very simple development. The nucleus of each cell divides only twice starting at the same time with the meiotic divisions of pollen mother cells but ending much earlier; subsequently, as usually happens with the Asteraceae, the ameboid involution of the tapetum begins. Endomitosis or any other process which leads to a polyploidy not due to nuclear fusion, has never been observed.

3) The female gametophyte is eight nucleate of the normal type (Polygonum). At maturity it shows only three antipodal cells whose nucleus undergoes at first, two or three divisions. Only later these new nuclei, always within the antipodal cell, may fuse in a polyvalent one.  相似文献

SYNOPSIS. Two new species of the simple mycetozoan genus Schizoplasmodium—S. ovatum and S. gracile—are described, the former having ovate spores and the latter spherical ones. All stages of the life cycle are multinucleate. At sporulation the holozoic plasmodium segments into prespore cells which develop directly into long-stalked, single-spored sporocarps. Sexual reproduction was not observed. A new order—the PROTOSTELIDA—is proposed for this and related genera, which are believed to comprise a phylogenetically linked series, beginning with the amoebo-flagellate genus Cavostelium and evolving through Schizoplasmodium to Protostelium. The two families Cavostelidae (Cavostelium) and Protostelidae (Schizoplasmodium, Protostelium) are recognized.  相似文献

Summary The protostelidPlanoprotostelium aurantium Olive andStoianovitch has trophic cells which are either amoebae or flagellates. The general morphology and ultrastructure are consistent with what has been reported for otherEumycetozoa (protostelids, myxomycetes, and dictyostelids). The flagellar apparatus structure has the same basic pattern as that of other flagellate eumycetozoans. It shares with all these an anteriorly directed flagellum and centriole and microtubule arrays (MTA) 2–4. Unlike more primitive species which have two centrioles per flagellar apparatus,P. aurantium has only one. Also, the flagellar apparatus is independent of the nucleus inP. aurantium, not linked to it as in the primitive species. These features are useful in explaining the differences in swimming behavior betweenP. aurantium and biflagellate species. Evidence is presented to show thatP. aurantium is closely related to the non-flagellateProtostelium mycophaga Olive andStoianovitch.This research represents part of a Ph.D. dissertation presented to the University of North Carolina.  相似文献

Illus. 1962.—Two new species of this simplest genus of the cellular slime molds are described: P. fimicola , a species with spherical spores and gelatinizing stalks occurring on dung, and P. arachispora , an elongate-spored species from dead plant material. The first has been found in North Carolina and the West Indies, while the second species has been found only once in North Carolina. Protostelium mycophaga var. major , from Connecticut, has distinctly larger spores than the original species. Further data on the distribution and development of P. mycophaga are presented. The genus is transferred to a family of its own, the Protosteliaceae .  相似文献

Social behavior in the myxobacterium Myxococcus xanthus involves epicellular, peritrichous appendages called fibrils. These are polysaccharide organelles containing a set of tightly adhering proteins. It is proposed that cell-cell contact is perceived by the fibrils and is mediated by the action of a fibrillar ADP-ribosyl transferase. Fibrils or fibril-like organelles have also been found on a variety of other gram-negative bacteria and at least one archaeon, and may mediate cell-cell contact between the bacteria themselves or between the bacteria and their eukaryotic host cells. We have previously observed that mouse peritoneal macrophages cultured for 48 h and treated with colchicine to depolymerize cytoplasmic microtubules become ameboid and cease to migrate by gliding on the substratum. We have now found that when such cells were further exposed to both colchicine and cytochalasin B, the induced ameboid movements were reversibly inhibited. Cells treated concomitantly with both drugs did not become motionless, but exhibited a remnant motility that took the form of zeiosis (blebbing). The zeiotic blebs contained ribosomes and fibrous material, but lacked organized microfilament arrays and rarely included other cytoplasmic organelles. Zeiosis appears to be a form of surface movement independent both of cytoplasmic microtubules and of the cytochalasin-sensitive contractile system. These observations imply an additional mechanism that can reversibly alter the form of the cell.  相似文献

Gymnodimium breve Davis, an unarmored marine dinoflagellate has a cell covering (theca) composed of four membranes. The inner two membranes represent a vesicular layer and in tangential section, the theca appears composed of polygonal areas. Unusual threat ridges are located in the cingular region between the epi- and hypocone. This osmotically sensitive species is extremely vesiculate with dispersed areas of cytoplasm containing typical eukaryotic organelles as well as other organelles found only in dinoflagellates. The non-vesiculated cytoplasm is continuous in serial sections. The chloroplasts can contain either quasi-radial or parallel lamellae typically consisting of three thylakoids each. The pyrenoid is multiple-stalked and lacks a starch cap. The dinophycean pusule is simple and similar to those found in several unarmored marine species. The nucleus is typically dinophycean but the chromosomes appear to lack nonfibrillar material.  相似文献

Summary The structure of guppy (Poecilia reticulata) spermatogonia and spermatocytes has been studied using electron microscopy. The spermatogonia, situated at the apex of the seminiferous tubule, are almost all surrounded by a network of Sertoli cells; they have very diffuse chromatin and one or two large nucleoli. The cytoplasm contains relatively few organelles, although annulate lamellae are found. The mitochondria have few cristae and are concentrated at one pole of the cell; they are sometimes found with intermitochondrial cement. These spermatogonia are separated from each other, having no intercellular bridges or inclusion in Sertoli cells, and are relatively undifferentiated; they correspond to stem cells. The spermatogonia beneath the apex are organized into cysts. First-generation spermatogonia are more dense and heterogeneous, their nuclei becoming smaller and their chromatin becoming denser during successive generations. In spermatocytes, the synaptinemal complex exists as a modified form until metaphase. The concentration of organelles in the cytoplasm increases and the organelles become more diversified as spermatogenesis progresses. Many cytoplasmic bridges are observed (several per cell), indicating that the cells remain in contact after several divisions. These changes in germ cell structure have been related to some of the characteristic features of spermatogenesis in guppy, e.g. the large number of spermatogonial generations and the complexity of spermiogenesis.  相似文献

Abstract Among the brown algae, species of the Fucaceae (Pelvetia, Fucus and Ascophyllum) were found to have a ‘photosynthetic buffering’ system, allowing the algae to carry out oxygen production without a concomitant uptake of inorganic carbon. This system was not found in other brown algae examined (e.g. Halidrys, Laminaria and Desmarestia) nor in 16 examined species of red and green algae. Pelvetia, Fucus and Ascophyllum belong to the littoral algae which are periodically emersed. In the Fucaceae, the meristodermal cells were found to have a special organization of organelles. Towards the outer cell wall there was a prominent layer of mitochondria while the chloroplasts were concentrated towards the inner and side walls. Between the mitochondria and the chloroplasts there was a large number of physodes. This arrangement of organelles was not found in the other brown algae examined nor in red or green algae. The significance of this organization of the mitochondria is discussed in connection with the function of the ‘photosynthetic buffering’ system.  相似文献

Autophagy is a conserved process in eukaryotic cells for degradation of cellular proteins and organelles. In filamentous fungi, autophagic degradation of organelles such as peroxisomes, mitochondria, and nuclei occurs in basal cells after the prolonged culture, but its mechanism is not well understood. Here, we functionally analyzed the filamentous fungus Aspergillus oryzae AoAtg26, an ortholog of the sterol glucosyltransferase PpAtg26 involved in pexophagy in the yeast Pichia pastoris. Deletion of Aoatg26 caused a severe decrease in conidiation and aerial hyphae formation, which is typically observed in the autophagy-deficient A. oryzae strains. In addition, cup-shaped AoAtg8-positive membrane structures were accumulated in the Aoatg26 deletion strain, indicating that autophagic process is impaired. Indeed, the Aoatg26 deletion strain was defective in the degradation of peroxisomes, mitochondria, and nuclei. Taken together, AoAtg26 plays an important role for autophagic degradation of organelles in A. oryzae, which may physiologically contribute to the differentiation in filamentous fungi.  相似文献

Summary The fine structure of granulosa lutein cells from three crabeater seals, Lobodon carcinophagus, and two leopard seals, Hydrurga leptonyx, has been studied from early through mid-pregnancy. Analysis of the arrangement and modifications of the cytoplasmic organelles and inclusions has revealed three types of lutein cells throughout the corpus. Type I cell typically possesses a central nucleus and cytoplasm containing very large amounts of smooth and/or fenestrated endoplasmic cisternae which frequently extend from the juxta-nuclear to the periphery of the cell. Type II cell contains a central or eccentric nucleus, moderate amounts of peripheral, smooth and fenestrated cisternae which often form large and concentric membranous whorls, numerous mitochondria and small lipid droplets. Frequently these cells show polarity in the arrangement of the cytoplasmic organelles and inclusions. Type III cell contains predominant large lipid droplets, many mitochondria, and small amounts of smooth and fenestrated cisternae. In light microscopy the type I cell is evenly granular, while the type III cell is highly vacuolated. Type II cells have both granular and vacuolated conditions. Ultrastructural features of type I and II cells suggest that they probably secrete most of the steroids, whereas the primary role of the type III cells appear to be lipid storage.This research was supported by National Science Foundation, Grant No. 1325 from the Office of Antarctic Biology.  相似文献
