Utilization of enzymes for environmental applications   总被引:6,自引:0,他引:6  
Enzymes are powerful tools that help sustain a clean environment in several ways. They are utilized for environmental purposes in a number of industries including agro-food, oil, animal feed, detergent, pulp and paper, textile, leather, petroleum, and specialty chemical and biochemical industry. Enzymes also help to maintain an unpolluted environment through their use in waste management. Recombinant DNA technology, protein engineering, and rational enzyme design are the emerging areas of research pertaining to environmental applications of enzymes. The future will also see the employment of various technologies including gene shuffling, high throughput screening, and nanotechnology. This article presents an overview of the enzymatic applications in pollution control and the promising research avenues in this area.  相似文献

Abstract

Biotechnology, defined as the technological application that uses biological systems and living organisms, or their derivatives, to create or modify diverse products or processes, is widely used for healthcare, agricultural and environmental applications. The continuity in industrial applications of biotechnology enables the rise and development of the bioeconomy concept. Bioeconomy, including all applications of biotechnology, is defined as translation of knowledge received from life sciences into new, sustainable, environment friendly and competitive products. With the advanced research and eco-efficient processes in the scope of bioeconomy, more healthy and sustainable life is promised. Knowledge-based bioeconomy with its economic, social and environmental potential has already been brought to the research agendas of European Union (EU) countries. The aim of this study is to summarize the development of knowledge-based bioeconomy in EU countries and to evaluate Turkey’s current situation compared to them. EU-funded biotechnology research projects under FP6 and FP7 and nationally-funded biotechnology projects under The Scientific and Technological Research Council of Turkey (TUBITAK) Academic Research Funding Program Directorate (ARDEB) and Technology and Innovation Funding Programs Directorate (TEYDEB) were examined. In the context of this study, the main research areas and subfields which have been funded, the budget spent and the number of projects funded since 2003 both nationally and EU-wide and the gaps and overlapping topics were analyzed. In consideration of the results, detailed suggestions for Turkey have been proposed. The research results are expected to be used as a roadmap for coordinating the stakeholders of bioeconomy and integrating Turkish Research Areas into European Research Areas.  相似文献

Purpose

The introduction of renewable materials into automotive applications is perceived as an innovative lightweight solution. Wood-based materials are advantageous in that they have potentially lower environmental impacts as compared with other materials such as steel. However, using wood per se does not automatically ensure more sustainability. Few prospective sustainability assessment methods or studies on the use of wood-based materials in automotive applications have been carried out, although these are needed to reduce unintended, negative sustainability effects and to support sustainable oriented research and innovation. Therefore, this study was conducted to assess the potential sustainability effects and consequences of introducing a wood-based component into an automotive application.

Methods

A combination of methods was used to analyze the potential sustainability effects when introducing wood into automotive applications. This prospective life cycle sustainability analysis solely relied on secondary data. The environmental impacts were analyzed using a simplified environmental life cycle assessment on the product level. A multi-regional input-output-based assessment was conducted to model the country-specific environmental and socioeconomic consequences. The potential shift in social risks and opportunities on a national scale was analyzed by conducting a generic social life cycle assessment. Various aspects of each approach differ, with each providing a specific perspective of the system under study.

Results and discussion

The results indicate that implementing wood into automotive application can have environmental, social, and economic benefits, according to most of the indicators analyzed. Mostly due to the product weight reduction due to the use of a wood-based component, the results show that environmental impacts decrease. Some possible consequences of using wood-based materials are increased value added and increasing the number of jobs in European countries. Similarly, the social risks and opportunities are shifted from countries all over the world to European countries, which perform better than developing countries according to several indicators. However, some indicators, such as migrant acceptance or local supplier quantity, perform better in the current situation.

Conclusions

The presented case study is particularly notable, because the results clearly indicate the advantages of using wood-based materials in automotive applications, although the application of such relatively holistic and complex approaches often may lead to rather indifferent pictures. Policy makers, researchers, and companies can apply this combination of methods that rely solely on generic data to obtain both feasible and informative results. These methods also allow users to link the product level assessment with a regional and social perspective and screen critical topics to support sustainability research and innovation.

Purpose

The purpose of this study is to provide an integrated method to identify the resource consumption, environmental emission, and economic cost for mechanical product manufacturing from economic and ecological dimensions and ultimately to provide theoretical and data support of energy conservation and emission reduction for mechanical product manufacturing.

Methods

The applied research methods include environmental life cycle assessment (LCA) and life cycle cost (LCC). In life cycle environmental assessment, the inventory data are referred from Chinese Life Cycle Database and midpoint approach and EDIP2003 and CML2001 models of life cycle impact assessment (LCIA) are selected. In life cycle cost assessment, three cost categories are considered. The proposed environment and cost assessment method is based on the theory of social willingness to pay for potential environmental impacts. With the WD615 Steyr engine as a case, life cycle environment and cost are analyzed and evaluated.

Results and discussion

The case study indicates that, in different life cycle phases, the trend of cost result is generally similar to the environmental impacts; the largest proportion of cost and environmental impact happened in the two phases of “material production” and “component manufacturing” and the smallest proportion in “material transport” and “product assembly.” The environmental impact category of Chinese resource depletion potential (CRDP) accounted for the largest proportion, followed by global warming potential (GWP) and photochemical ozone creation potential (POCP), whereas the impacts of eutrophication potential (EP) and acidification potential (AP) are the smallest. The life cycle “conventional cost” accounted for almost all the highest percentage in each phase (except “material transport” phase), which is more than 80% of the total cost. The “environmental cost” and “possible cost” in each phase are relatively close, and the proportion of which is far below the “conventional cost.”

Conclusions

The proposed method enhanced the conventional LCA. The case results indicate that, in a life cycle framework, the environment and cost analysis results could support each other, and focusing on the environment and cost analysis for mechanical product manufacturing will contribute to a more comprehensive eco-efficiency assessment. Further research on the life cycle can be extended to phases of “early design,” “product use,” and “final disposal.” Other LCIA models and endpoint indicators are advocated for this environmental assessment. Environmental cost can also be further investigated, and the relevant social willingness to pay for more environmental emissions is advocated to be increased.

Abstract

Enzymes of the subtilisin family, of which proteinase K is a member, have been studied extensively on account of their numerous biological applications. Specific inhibitors of the proteinases are of immense importance in regulating their activity so as to protect the cells against uncontrolled proteolysis. Using the specific design principles of peptides containing dehydro-Alanine (ΔAla), generated by our theoretical calculations, we present here the design of an inhibitor of proteinase K. Energy minimization and molecular modeling of the interaction of the designed tetrapeptide with the recognition site of proteinase K indicate that it is an effective inhibitor.  相似文献

Abstract

Microreaction technology is an interdisciplinary area of science and engineering. It has attracted the attention of researchers from different fields in the past few years and consequently, several microreactors have been developed. Enzymes are organic catalysts used for the production useful substances in an environmentally friendly way, and have high potential for analytical applications. However, relatively few enzymatic processes have been commercialized because of problems in the stability of enzyme molecule, and the cost and efficiency of the reactions. Thus, there have been demands for innovation in process engineering particularly for enzymatic reactions, and microreaction devices can serve as efficient tools for the development of enzyme processes. In this review, we summarize the recent advances of microchannel reaction technologies and focus our discussion on enzyme microreactors. We discuss the manufacturing process of microreaction devices and the advantages of microreactors compared with the conventional reactors. Fundamental techniques for enzyme microreactors and important applications of this multidisciplinary technology in chemical processing are also included in our topics.  相似文献

Introduction: The mass spectrometry society of Japan, Japanese proteomics society, and Asia–Oceania human proteome organization held the conference ‘Mass Spectrometry and Proteomics 2018’ in Osaka, Japan, on May 15–18, 2018. This international conference focused on cutting edge technologies and their applications in a variety of research fields such as agriculture, material science, environmental factors, and clinical applications. An overview of the conference and a summary of the major lectures are reported here.

Background

High-throughput sequencing involves library preparation and amplification steps, which may induce contamination across samples or between samples and the environment.

Methods

We tested the effect of applying an inline-index strategy, in which DNA indices of 6 bp were added to both ends of the inserts at the ligation step of library prep for resolving the data contamination problem.

Results

Our results showed that the contamination ranged from 0.29 to 1.25% in one experiment and from 0.83 to 27.01% in the other. We also found that contamination could be environmental or from reagents besides cross-contamination between samples.

Conclusions

Inline-index method is a useful experimental design to clean up the data and address the contamination problem which has been plaguing high-throughput sequencing data in many applications.

Our current global environmental challenges include the reduction of harmful chemicals and their derivatives. Bioremediation has been a key strategy to control the massive presence of chemicals in the environment. Enzymes including the phenoloxidases, laccases and tyrosinases, are increasingly being investigated as “green products” in the removal of many chemical contaminants in waters and soils. Both phenoloxidases are widespread in nature and attractive biocatalysts due to their ability to use readily available molecular oxygen as sole cofactor for their catalytic elimination of a large number of chemicals. Taking advantage of their catalytic potentials, remarkable advances have been made in the engineering of laccases to produce suitable biocatalysts in environmental applications. Studies about novel strategies of laccase immobilization and insolubilization for the treatment of chemical contaminants were provided. Likewise, tyrosinases are gaining increasing interest in environmental applications due to their catalytic similarities with laccases although they remain far less investigated to date. This disparity was addressed in this review along with the molecular features and catalytic mechanism of tyrosinases relevant in environmental applications. A perspective on the future use of laccases and tyrosinases in bioremediation was discussed.  相似文献

Purpose

To reduce the environmental impact of the building sector, environmental targets considering the full life cycle of buildings can be supportive. In recent years, various benchmarks based on Life Cycle Assessment (LCA) have been developed as part of regulations, labelling systems, sustainability rating tools and research studies. The objective of this paper is to critically analyse 23 existing benchmarking systems focusing on the benchmark methodology but also on the benchmark applications and communication.

Methods

The critical literature review consists of two parts. In a first part, the choices related to the assessment method, functional equivalent, definition of benchmark values, benchmark scope, benchmark applications and benchmark communication are compared. In the second part, benchmark values are compiled from literature and statistically analysed.

Results and discussion

The comparative analysis allows to identify the main approaches and methods used in benchmarking systems. For each evaluation aspect, the strengths and weaknesses of the various approaches are highlighted. The statistical analysis provides insight in the spread of benchmark values. Important variations are found between the literature sources which can be explained by differences in benchmark approach, scope, system boundaries and applications.

Conclusions

Based on the comparative analysis, recommendations are formulated for the development of LCA benchmarks for the building sector. The results of the statistical analysis furthermore provide reference values which can be used for the validation of future benchmarks. For global warming, the statistical values for the full life cycle impacts (i.e. embodied and operational impacts) range from about 15 up to 35 kg CO2 eq/m2.a.

Azole resistance is a concern for the management of diseases caused by Aspergillus fumigatus in humans. Azole fungicide use in the environment has been identified as a possible cause for development of resistance, which increases the complexity and number of stakeholders involved in this emerging problem. A workshop was held in Amsterdam early 2019 in which stakeholders, including medical and agricultural researchers, representatives from the government, public health, fungicide producers and end-users, reviewed the current evidence supporting environmental selection for resistance and to discuss which research and measures are needed to retain the effectiveness of the azole class for environmental and medical applications. This paper provides an overview of the latest insights and understanding of azole resistance development in the clinical setting and the wider environment. A One Health problem approach was undertaken to list and prioritize which research will be needed to provide missing evidence and to enable preventive interventions.  相似文献

Abstract

The COST Action CM1201 “Biomimetic Radical Chemistry” has been active since December 2012 for 4 years, developing research topics organized into four working groups: WG1 – Radical Enzymes, WG2 – Models of DNA damage and consequences, WG3 – Membrane stress, signalling and defenses, and WG4 – Bio-inspired synthetic strategies. International collaborations have been established among the participating 80 research groups with brilliant interdisciplinary achievements. Free radical research with a biomimetic approach has been realized in the COST Action and are summarized in this overview by the four WG leaders.  相似文献

Background and AimsReproductive isolation and local establishment are necessary for plant speciation. Polyploidy, the possession of more than two complete chromosome sets, creates a strong postzygotic reproductive barrier between diploid and tetraploid cytotypes. However, this barrier weakens between polyploids (e.g. tetraploids and hexaploids). Reproductive isolation may be enhanced by cytotype morphological and environmental differentiation. Moreover, morphological adaptations to local conditions contribute to plant establishment. However, the relative contributions of ploidy level and the environment to morphology have generally been neglected. Thus, the extent of morphological variation driven by ploidy level and the environment was modelled for diploid, tetraploid and hexaploid cytotypes of Campanula rotundifolia agg. Cytotype distribution was updated, and morphological and environmental differentiation was tested in the presence and absence of natural contact zones.MethodsCytotype distribution was assessed from 231 localities in Central Europe, including 48 localities with known chromosome counts, using flow cytometry. Differentiation in environmental niche and morphology was tested for cytotype pairs using discriminant analyses. A structural equation model was used to explore the synergies between cytotype, environment and morphology.Key ResultsTremendous discrepancies were revealed between the reported and detected cytotype distribution. Neither mixed-ploidy populations nor interploidy hybrids were detected in the contact zones. Diploids had the broadest environmental niche, while hexaploids had the smallest and specialized niche. Hexaploids and spatially isolated cytotype pairs differed morphologically, including allopatric tetraploids. While leaf and shoot morphology were influenced by environmental conditions and polyploidy, flower morphology depended exclusively on the cytotype.ConclusionsReproductive isolation mechanisms vary between cytotypes. While diploids and polyploids are isolated postzygotically, the environmental niche shift is essential between higher polyploids. The impact of polyploidy and the environment on plant morphology implies the adaptive potential of polyploids, while the exclusive relationship between flower morphology and cytotype highlights the role of polyploidy in reproductive isolation.  相似文献

BackgroundThe lack of new anthelmintic agents is of growing concern because it affects human health and our food supply, as both livestock and plants are affected. Two principal factors contribute to this problem. First, nematode resistance to anthelmintic drugs is increasing worldwide and second, many effective nematicides pose environmental hazards. In this paper we address this problem by deploying a high throughput screening platform for anthelmintic drug discovery using the nematode Caenorhabditis elegans as a surrogate for infectious nematodes. This method offers the possibility of identifying new anthelmintics in a cost-effective and timely manner.Conclusions/SignificanceThe challenge of anthelmintic drug discovery is exacerbated by several factors; including, 1) the biochemical similarity between host and parasite genomes, 2) the geographic location of parasitic nematodes and 3) the rapid development of resistance. Accordingly, an approach that can screen large compound collections rapidly is required. C. elegans as a surrogate parasite offers the ability to screen compounds rapidly and, equally importantly, with specificity, thus reducing the potential toxicity of these compounds to the host and the environment. We believe this approach will help to replenish the pipeline of potential nematicides.  相似文献

Abstract

Microstructural post mortem changes to skeletal tissues by microorganisms are driven by several factors including the death history of an animal, its decomposition trajectory, and the depositing environment itself. The study we describe here brings together material from recent and fossil contexts that are depositionally distinct from a terrestrial-marine transitionary shoreline environment. We compare these changes with those of marine environments previously identified in the Mary Rose material, and those of continental waters (lakes) previously identified in the Cerro de la Garita (Concud) site, and we document this against bacterially related changes observed from terrestrial contexts. A new microstructural change identified in material from terrestrial sites is also described relating to rootlet damage. By considering microstructural change in skeletal tissues, it is maybe possible to ascribe environmental context, or, to better understand the complexity of material presented by transitionary environments.  相似文献

【背景】磷转化微生物对于自然环境中的磷循环具有重要作用。此类微生物通过对环境中磷素的溶解矿化和吸收转运等作用促进资源高效利用,减少环境污染,对粮食安全和生态系统稳定也具有积极的影响。【目的】探究近年来国内外关于磷转化微生物的发展热点及趋势,展示该领域的知识结构和知识演化过程,为后续研究提供可行性的参考和启示。【方法】应用Cite Space可视化软件对2002-2022年发表在中国知网(CNKI)和WebofScience(WOS)数据库中的文献通过关键词共现分析、关键词聚类分析、关键词突现分析、发文数量分析、国家分布、作者合作共现及机构合作共现等方式对磷转化微生物的研究现状及新兴趋势进行分析。【结果】最终共筛选出887篇有效文献。磷转化微生物研究在2016年后开始蓬勃发展,近几年增速加快;中国和美国学者是此领域重要的研究力量;中国是发文量最多的国家,中国科学院是发文量最多的机构;热门的研究领域为菌株的分类与筛选、代谢途径、营养元素循环以及环境污染与生态保护。【结论】磷转化微生物具有广阔的发展前景。目前,生物炭及磷转化微生物的响应机制正在成为新的研究热点。此类技术在未来的广泛应用是必然...  相似文献

BackgroundNanomaterials (NMs) are often exposed to a broad range of biomolecules of different abundances. Biomolecule sorption driven by various interfacial forces determines the surface structure and composition of NMs, subsequently governs their functionality and the reactivity of the adsorbed biomolecules. Isothermal titration calorimetry (ITC) is a nondestructive technique that quantifies thermodynamic parameters through in-situ measurement of the heat absorption or release associated with an interaction.Scope of reviewThis review highlights the recent applications of ITC in understanding the thermodynamics of interactions between various nanoparticles (NPs) and biomolecules. Different aspects of a typical ITC experiment that are crucial for obtaining accurate and meaningful data, as well as the strengths, weaknesses, and challenges of ITC applications to NP research were discussed.Major conclusionsITC reveals the driving forces behind biomolecule–NP interactions and the effects of the physicochemical properties of both NPs and biomolecules by quantifying the crucial thermodynamics parameters (e.g., binding stoichiometry, ΔH, ΔS, and ΔG). Complimentary techniques would strengthen the interpretation of ITC results for a more holistic understanding of biomolecule–NP interactions.General significanceThe thermodynamic information revealed by ITC and its complimentary characterizations is important for understanding biomolecule–NP interactions that are fundamental to the biomedical and environmental applications of NMs and their toxicological effects.
