The changing levels of carbohydrates and the rates of photosynthesis in rooting Populus cuttings are described. The cuttings were planted in a sand medium, in controlled environment conditions. There was no evidence to support the view that failure to root in P. tremula was caused by insufficient carbohydrate reserves. In leafless hardwood cuttings, the carbohydrate levels were initially very high (14–19%) but fell rapidly (to 5–10%) as roots, callus or shoots developed. In leafy softwood cuttings, a steady accumulation of carbohydrate reserves was observed, rising from 5–10% on planting to 15–25% when roots had developed. One of the differences between the easy-to-root P. × euramericana and the hard-to-root P. tremula was the apparent downward transport of assimilates in P. × euramericana softwood cuttings.  相似文献

Summary One easy-to-root and one difficult-to-root species of the ornamental plant Grevillea were investigated for rooting potential in relation to peroxidase activity. In vitro-grown shoot segments of both species started to root 30 d after transplanting to rooting medium containing indole-3-butyric acid (IBA), however, fewer roots were found on fewer segments of the difficult-to-root species G. petrophioides compared to the easy-to-root species G. rondeau. Total peroxidase (POX) activity was measured during the rooting process. G. petrophioides showed higher total POX activity at the time point of adventitious root formation than G. rondeau. Isoelectric focusing electrophoresis showed that G. rondeau contained more acidic isoforms than G. petrophioides, but the basic isoforms were more prominent in the difficult-to-root species, especially at the time point of lateral root emergence. In addition, the ability of different hormones to induced POX activity in upper and lower stem segments of both species was tested. Indole-3-acetic acid (IAA), IBA and α-naphthaleneacetic acid induced POX activity in the upper stem segments of G. rondeau, whereas the same hormones led to the induction of POX activity in the lower stem segments of G. petrophioides. Similar to the results obtained with Grevillea, the difficult-to-root variety of Protea showed higher POX activity, especially in the middle stem part and the leaves. Feeding of radiolabeled IAA to the Grevillea stem segments resulted in the synthesis of three different compounds in both species. After 1h incubation no differences were found in the uptake of IAA and the appearance of other labeled compounds. However, after 2 and 4 h incubation IAA uptake was faster in the easy-to-root species and IAA was also metabolized to a higher extent in G. rondeau. Three metabolites were found, tentatively identified as IAA-aspartate, IBA, and an IBA conjugate.  相似文献

The relationship between the level of endogenous growth regulators and the rooting ability of dahlia cuttings was investigated. Easy-to-root vegetative cuttings were compared with difficult-to-root, flower bud bearing cuttings and the easy-to-root cv. Choot Hashani was compared with the difficnlt-to-root cv. Orpheo. No differences in the level of extracted or diffusable auxin, nor in the level of rooting cofactors, were found between easy-to-root and difficult-to-root cuttings. No activity could be found at the Rf zone corresponding to Cofactor 4 which is considered the main rooting cofactor in other plants. Higher inhibitor activity levels (assayed by the wheat coleoptile test and the mung bean rooting test) were found in difficult-to-root than in easily rooted cuttings. Furthermore, the inhibitor activity level in exadates from decapitated plants, was higher in esodates from plants which bore difficult-to-root cuttings than from plants bearing easy-to-root ones. Grafting of an easy-to-root plant onto a difficult-to-root stock caused inhibition of rooting in the cuttings taken from the scion. Results suggest that inhibitors formed in the root system, move upwards and accumulate in the plant, there causing the inhibition of rooting. Evidence from chromatographic comparisons and exogenous treatments with ABA indicate that the main inhibitor involved in the inhibition of rooting in dahlia cuttings is not ABA.  相似文献

Indole-3-butyric acid (IBA) greatly enhanced the rooting of an early-flowering variety of protea, Leucadendron discolor, but had very little effect on a late-flowering variety. IBA transport and metabolism were studied in both varieties after incubating the cuttings in 3H-IBA. More of the radio-label was transported to the leaves of the easy-to-root variety than the difficult-to-root (35–45% and 10%, respectively). IBA was metabolized rapidly by the cuttings of both varieties and after 24 h most of the label was in the new metabolite. However, free IBA (about 10%) was present in the cuttings during the whole period up to the time of root emergence (4 weeks). More free IBA was accumulated in the base of easy-to-root cuttings, while in the difficult-to-root variety most of the IBA was found in the leaves. The metabolite was identified tentatively as an ester conjugate with a glucose. It is possible that IBA-glucose serves as a source for free IBA, and the difference between the varieties is a consequence of the free IBA which is released, transported and accumulated in the site of a root formation.  相似文献

The present study investigated the interaction of growth irradiance (Qint) with leaf capacity for and kinetics of adjustment of the pool size of xanthophyll cycle carotenoids (sum of violaxanthin, antheraxanthin and zeaxanthin; VAZ) and photosynthetic electron transport rate (Jmax) after changes in leaf light environment. Individual leaves of lower‐canopy/lower photosynthetic capacity species Tilia cordata Mill. and upper canopy/higher photosynthetic capacity species Populus tremula L. were either illuminated by additional light of 500–800 µmol m?2 s?1 for 12 h photoperiod or enclosed in shade bags. The extra irradiance increased the total amount of light intercepted by two‐fold for the upper and 10–15‐fold for the lower canopy leaves, whereas the shade bags transmitted 45% of incident irradiance. In control leaves, VAZ/area, VAZ/Chl and Jmax were positively associated with leaf growth irradiance (Qint). After 11 d extra illumination, VAZ/Chl increased in all cases due to a strong reduction in foliar chlorophyll, but VAZ/area increased in the upper canopy leaves of both species, and remained constant or decreased in the lower canopy leaves of T. cordata. The slope for VAZ/area changes with cumulative extra irradiance was positively associated with Qint only in T. cordata, but not in P. tremula. Nevertheless, all leaves of P. tremula increased VAZ/area more than the most responsive leaves of T. cordata. Shading reduced VAZ content only in P. tremula, but not in T. cordata, again demonstrating that P. tremula is a more responsive species. Compatible with the hypothesis of the role of VAZ in photoprotection, the rates of photosynthetic electron transport declined less in P. tremula than in T. cordata after the extra irradiance treatment. However, foliar chlorophyll contents of the exposed leaves declined significantly more in the upper canopy of P. tremula, which is not consistent with the suggestion that the leaves with the highest VAZ content are more resistant to photoinhibition. This study demonstrates that previous leaf light environment may significantly affect the adaptation capacity of foliage to altered light environment, and also that species differences in photosynthetic capacity and acclimation potentials importantly alter this interaction.  相似文献

Summary An elite aspen hybrid (Populus × canescens × P. grandidentata) was transformed with Agrobacterium tumefaciens strain EHA105 that harbored a binary vector (pBI121) carrying the nptII gene under the nos promoter and tandem rolB-uidA (GUS) genes with the CaMV 35S or heat shock promoter. Among 32 independent kanamycin-resistant plants, 25 plants were confirmed by polymerase chain reaction and Southern blot analyses to contain all three genes, whereas five plants contained only nptII or/and uidA genes and two plants had both the rolB and nptII or uidA genes. Integration of the rolB gene significantly increased rooting ability of hardwood cuttings. Heat shock-rolB-transformed plants rooted at significantly higher percentage than the CaMV 35S-rolB-transformed plants. Heat shock treatment further enhanced rooting of heat shock-rolB-transformed plants. Exposure to exogenous auxin did not significantly increase the rooting percentage of transgenic hardwood cuttings, but increased the number of roots induced. This research shows great potential to improve rooting of hardwood cuttings of difficult-to-root woody plants which are commercially important to the horticultural and forestry industry. The transgenic plants with gain-of-function in hardwood-cutting rooting can facilitate research in the understanding of adventitious rooting from hardwood cuttings of recalcitrant woody plants.  相似文献

采用5对SSR引物对52个山杨杂种无性系进行了遗传多样性检测,结果表明在研究的5个位点上SSR标记多态位点百分率为100%,平均等位基因数为4.4个,有效等位基因数最多的位点是PTR7,最少的位点为PTR12;欧美山杨杂种的遗传多样性最丰富,相比之下,中美山杨杂种遗传变异最低;聚类分析表明,在一定的遗传距离基础上,欧美山杨杂种和欧洲山杨首先聚为一类,然后又与中美山杨杂种聚类,最后是中国山杨。研究表明来自芬欧美山杨杂种具有较高的遗传多样性,这对我国山杨遗传资源的扩大,以及未来山杨杂交育种,杂种优势的利用都是重要的。  相似文献

Proteomic analysis provides a powerful method of studying plant responses to stress at the protein level. In order to study stress-responsive molecular mechanisms for Populus × euramericana cv. ‘74/76’, one of the most important forest plantation tree species in subtropical and temperate regions, we analyzed the response of 2-year-old cuttings of P. × euramericana cv. ‘74/76’ to drought and high temperature using two-dimensional gel electrophoresis. More than 1,000 reproducible leaf proteins were detected in the controls and treatments, and 26 proteins were found to change notably in abundance. We identified 13 proteins affected by drought stress and 11 proteins affected by high temperature. These proteins are mainly involved in photosynthesis such as ribulose-1,5-bisphosphate carboxylase/oxygenase large subunit and putative photosystem I reaction center subunit II precursor, and detoxification (manganese superoxide dismutase and methionine sulfoxide reductase A). Furthermore, the level of the photosynthesis proteins affected greatly by the imposed stress conditions was consistent with the observed noticeable decrease in net photosynthesis rate. These studies provides a fundamental data for future research on responses to drought and high temperature, two major factors limiting the growth of forest trees during summer under recent climatic warming.  相似文献

Although fast‐growing Populus species consume a large amount of water for biomass production, there are considerable variations in water use efficiency (WUE) across different poplar species. To compare differences in growth, WUE and anatomical properties of leaf and xylem and to examine the relationship between photosynthesis/WUE and anatomical properties of leaf and xylem, cuttings of six poplar species were grown in a botanical garden. The growth performance, photosynthesis, intrinsic WUE (WUEi), stable carbon isotope composition (δ13C) and anatomical properties of leaf and xylem were analysed in these poplar plants. Significant differences were found in growth, photosynthesis, WUEi and anatomical properties among the examined species. Populus cathayana was the clone with the fastest growth and the lowest WUEi13C, whereas P. × euramericana had a considerable growth increment and the highest WUEi13C. Among the analysed poplar species, the highest total stomatal density in P. cathayana was correlated with its highest stomatal conductance (gs) and lowest WUEi13C. Moreover, significant correlations were observed between WUEi and abaxial stomatal density and stem vessel lumen area. These data suggest that photosynthesis, WUEi and δ13C are associated with leaf and xylem anatomy and there are tradeoffs between growth and WUEi. It is anticipated that some poplar species, e.g. P. × euramericana, are better candidates for water‐limited regions and others, e.g. P. cathayana, may be better for water‐abundant areas.  相似文献

A large number of simple sequence repeat (SSR) marker-containing genetic maps are available for several Populus species. For aspen however, no SSR-containing map has been published so far. In this study, genetic linkage mapping was carried out with an interspecific mapping pedigree of 61 full-sib hybrids of European × quaking aspen (Populus tremula L. × Populus tremuloides Michx.), using the two-way pseudo-testcross strategy. Amplified fragment-length polymorphism (AFLP) and SSR markers were used for mapping, resulting in the first SSR-containing genetic linkage maps for aspen. The maps allow comparisons with a Populus consensus map and other published genetic maps of the genus Populus. The maps showed good collinearity to each other and to the Populus consensus map and provide a direct link to the Populus trichocarpa genomic sequence. Sex as a morphological trait was assessed in the mapping population and mapped on a non-terminal position of linkage group XIX on the male P. tremuloides map.  相似文献

The effect of light on rooting of leafy cuttings of aspen (Populus tremula × tremuloides) and a willow hybrid (Salix caprea × viminalis) was investigated under controlled conditions in water culture. Two levels of irradiance were used, 40 and 8 W m?2. The lower level gave the best rooting of aspen cuttings, both when applied to the stock plants before the cuttings were taken and when given to the cuttings during the rooting period. Irradiation of the cutting base during the rooting period inhibited rooting almost completely in aspen and decreased the number of roots formed in the Salix hybrid. Net photosynthesis in the cuttings of Salix decreased considerably after excision and increased again after formation of roots. Indirect evidence indicated that photosynthesis was even more affected in aspen cuttings. The possible roles of carbohydrates and inhibitors in the light effects are discussed.  相似文献

Therooting responses of cuttings of difficult-to-root lilac (Syringavulgaris) and easy-to-root forsythia(Forsythia×intermedia)were compared. The rooting ability of lilac cuttings declined over the growingseason (May–June). There was also a decline in the initial concentrationof free IAA at the base of the cuttings, but there was not a tight relationshipbetween basal IAA concentration and rooting ability. Polar auxin transportability was measured in lilac and forsythia during the period of maximum growthby [3H]IAA application to stem internodal tissue. Transport abilitydeclined in lilac over this time period, particularly in terms of transportintensity and percentage of [3H]IAA transported. In contrast thechanges in polar auxin transport ability in forsythia were less marked. Thisdifference between species was maintained in winter hardwood cuttings, withforsythia tissue showing greater polar auxin transport ability than lilac. Theimportance of polar auxin transport for adventitious rooting was demonstratedinboth lilac and forsythia softwood cuttings by use of the polar transportinhibitor 2,3,5-triiodobenzoic acid (TIBA). Overall the results indicate thatdifferences in polar auxin transport ability between lilac and forsythiacontribute to differences in rooting ability.  相似文献

 In woody plants, chilling stress occurs during the early spring growth and can have important economic consequences. The aim of this study was to compare the reliability of two different experimental systems, 3-month-old softwood cuttings and in vitro-grown shoots, to study chilling effects in a poplar clone (Populus tremula×P. tremuloides cv. Muhs1). Different parameters were recorded: lignin content, sucrose concentration and protein and fresh weight variation, during a 2-week treatment at 10  °C. Two families of polypeptides of high molecular weight (110 and 116 kDa) were shown to accumulate in response to chilling in both cuttings and microshoots. For some of the parameters studied, i.e. appearance of some groups of polypeptides and reduction of fresh weight gain, both in vitro and ex vitro systems were suitable and produced similar results. In contrast, for some other observations, i.e. on sucrose concentration and lignin content, the systems led to different conclusions. While sucrose and lignin contents were shown to increase in cuttings submitted to chilling, no variation in lignin and only a small temporary peak of sucrose could be observed in microshoots kept under chilling. These parameters seem not to be suitable for studying the response of poplar in the in vitro system in such a short-term study. The effects of applying indole-3-butyric acid (IBA) for periods up to 48 h were examined in difficult-to-root microcuttings (from newly-established cultures) and in easy-to-root microcuttings (from long-term subcultures) of Jonathan apple (Malus X domestica Borkh). In easy-to-root material, 20% of the microcuttings produced roots in the absence of IBA, while 6 h exposure to 10 M IBA gave 100% rooting of microcuttings. In contrast, root formation in difficult-to-root material was IBA-dependent. Maximum rooting of these microcuttings (50%) required 24 h exposure to 10 M IBA.Variation in the endogenous levels of free indole-3-acetic acid (IAA) during the course of root induction was similar in microcuttings of both types but there were marked differences in endogenous abscisic acid (ABA) levels. In easy-to-root microcuttings ABA remained at a constant low level, but in difficult-to-root material ABA exhibited marked fluctuations and was present at higher concentrations than in easy-to-root microcuttings.  相似文献

Parasitoid complexes of two species of the genus Phyllonorycter (Ph. apparella and Ph. populifoliella) reared from 2 plants (Populus tremula and P. nigra) were studied in Ul’yanovsk Province, Russia. Twenty species of parasitoids are new for Ph. apparella and 12 are new for Ph. populifoliella. Minotetrastichus frontalis and Pnigalio agraules are the dominant species for both parasitoid complexes. The highest relative abundance was observed for Ph. apparella on Populus tremula (75%) and for Ph. populifoliella on P. nigra (61%). The parasitoid complexes of Ph. apparella and Ph. populifoliella show high similarity (the Jaccard coefficient is 0.65).  相似文献

The plant growth activity of vitamin D3 has been examined using well-established bioassays. Vitamin D3 was found to promote adventitious root formation in cuttings ofPopulus tremula and to promote the germination of lettuce (Lactuca sativa cv. Grand Rapids) seeds in the absence of light. A fast growing high density Populus plantation located in central Italy was exposed to elevated carbon dioxide for a period of three years. An elevated CO2 treatment (550 ppm), of 200 ppm over ambient (350 ppm) was provided using a FACE technique. Standing root biomass, fine root turnover and mycorrhizal colonization of the following Populus species was examined: Populus alba L., Populus nigra L., Populus x euramericana Dode (Guinier). Elevated CO2 increased belowground allocation of biomass in all three species examined, standing root biomass increased by 47–76% as a result of FACE treatment. Similarly, fine root biomass present in the soil increased by 35–84%. The FACE treatment resulted in 55% faster fine root turnover in P. alba and a 27% increase in turnover of roots of P. nigra and P. x euramericana. P. alba and P. nigra invested more root biomass into deeper soil horizon under elevated CO2. Response of the mycorrhizal community to elevated CO2 was more varied, the rate of infection increased only in P. alba for both ectomycorrhizal (EM) and arbuscular mycorrhizas (AM). The roots of P. nigra showed greater infection only by AM and the colonization of the root system of P. x euramericana was not affected by FACE treatment. The results suggest that elevated atmospheric CO2 conditions induce greater belowground biomass investment, which could lead to accumulation of assimilated C in the soil profile. This may have implications for C sequestration and must be taken into account when considering long‐term C storage in the soil.  相似文献

As a common tree species in northern China, Populus × euramericana “Neva” has an important practical value for the study of continuous cropping obstacles in poplar cultivation. Plant allelopathy is the main reason for continuous cropping obstacles, which are caused by allelochemicals, such as para-hydroxybenzoic acid (p-HB). The objective of this study was to investigate the effects of p-HB on the photosynthesis of poplar. Photosynthetic parameters of Populus × euramericana “Neva” poplar were determined in a pot culture experiment where five p-HB concentrations were used (0, 1, 2, 4, and 6 mmol L?1). Each seedling was treated with 4 L of p-HB solution every seven days, ten times in total. p-HB inhibited the photosynthesis of poplar significantly, as shown by a clear decline in the net photosynthetic rate. Our results indicated nonstomatal limitation responsible for the photosynthesis reduction.  相似文献

Effect of alginate matrix composition on regrowth performance of encapsulated microcuttings of hybrid aspen (Populus tremula L. × P. tremuloides Mincx.) was studied. Both high regrowth frequency and viability of explants were registered in all encapsulation mixtures tested. Some ingredients of the matrix (nutrient medium salts, sugars, growth regulators) significantly affected the initial development of the microcuttings. Sucrose appeared to play an important role in the starting stage of the regrowth event.  相似文献
