Variegated tobacco leaves, white on one side of the midrib andgreen on the other, were detached from the stem and incubatedunder water stress or turgid conditions for 4 days to determineany changes in the levels of free amino acids. Drastic changes in the free amino acid composition occurredin the green tissue during the water stress period, but onlyvery small changes in the white tissue. During that time, themost striking difference between the two tissues was the largeamount of proline accumulated in the green tissue, but not inthe white. An exogenous supply of sucrose increased the prolinecontent in both tissues during water stress. An exogenous supplyof glutamic acid increased the proline and asparagine contentsin the green tissue, but it increased only the asparagine contentin the white tissue during water stress. 反相高效液相色谱法测定烟叶中的游离氨基酸   总被引:3,自引:1,他引:3  
用不同浓度的乙醇溶液提取烟样中的游离氨基酸 ,结果显示 ,存在最佳的乙醇溶液浓度 ,使烟样中被提取的游离氨基酸总量最大 ;对比了活性炭、乙醚、5 %磺基水杨酸、阳离子交换柱的纯化效果 ,发现阳离子交换柱的纯化效果较其它三种试剂要好。在提取和纯化之后 ,采用OPA、FMOC联合在线衍生反相高效液相色谱法测定了烟样中的游离氨基酸 ,该方法使烟样中的氨基酸和亚氨基酸能被同时测定 ,并且分析方法的重现性和回收率均令人满意。最后用该方法对云南B2 F98(上部、橘黄、二等烟叶 ,98年产 )烟叶中的游离氨基酸进行了测定 ,有 15种氨基酸被测出 ,其中Pro含量最高 ,约占总量的 2 5 % ,Thr含量最低 ,约占总量的 1%。  相似文献

Wheat prolamines were preincorporated into the endosperm of maize seed, and concentrations of free and bound amino acids were measured in the endosperm and seedlings during seed germination. Incorporation of foreign prolamines lowered the rate of decrease of free proline concentration in both the endosperm (sprouting and shoots) and the embryo (sprouting). The seedlings at the stage of shoots were characterized by higher concentrations of aspartic acid, glutamic acid, and tyrosine. At the stage of transition to autotrophic nutrition, the pool of amino acids that are physiologically important for plant growth was greater in maize seedlings with incorporated wheat prolamines.  相似文献

利用OPA、FMOC联合在线衍生反相高效液相色谱法测定白肋烟在110℃、120℃、130℃、140℃、150℃分别烘焙12m in和16 m in后游离氨基酸的含量,研究烘焙温度和烘焙时间对白肋烟中游离氨基酸的影响。结果显示整个烘焙过程是蛋白质的降解和M ailard反应相结合的动态过程,蛋白质的降解主要发生在110℃~120℃的低温区;而M ailard反应主要发生在130℃~150℃高温区。实验结果还表明烘焙时间对白肋烟在烘焙后游离氨基酸的含量有重要影响,烘焙时间的延长使烟叶中的游离氨基酸均有较大幅度降低,其中影响最明显的是Pro、Asp和Asn。  相似文献

Shikimate, anthranilate, indole, l -tryptophan, phenylpyruvate, l -p henylalanine, p-hydroxyphenylpyruvate or l -tyrosine were added to suspension-cultured Nicotiana tabacum (tabacco) and Daucus carota (carrot) tissues and incubated for 24 hours. Uptake of each compound was substantial as measured by its decrease in the medium. The levels of free tryptophan, phenylalanine and tyrosine were determined in the tissues after the 24 hours incubation. Shikimate did not change the aromatic animo acid levels in carrot tissue, but did increase all three in tobacco (3-fold or more), indicating a less stringent feedback control in tobacco. Anthranilate and indole increased the tissue tryptophan levels in both species by at least 17-fold, showing that the flow from anthranilate and indole to tryptophan was apparently unhindered by enzymatic control mechanisms. When tryptophan levels were elevated in both carrot and tobaccotissues by anthranilate, indole or tryptophan addition, there was also an increase in free phyenylalanine and tyrosine. This might be due to the reversal of phenylalanine and tyrosine feedback inhibition of chorismate mutase by the high tryptophan in the tissue. Chorismate mutase activity in tobacco crude extracts could be inhibited by 66–90% by 1 mM phenylalanine and /or tyrosine. Tryptophan at 1 mM stimulated the enzyme activity by 1/3 and completely reversed the phenylalanine and/or tyrosine inhibition of enzyme activity. Chorsimate mutase activity amino acids under a variety of conditions. Phenylpyruvate increased the phenylalanine levels and p-hydroxyphenylpyruvate increased the tyrosine levels in carrot and tobacco tissues indicating that there was no feedback control of the last step in phenylalanine and tyrosine biosynthesis.  相似文献

Changes in the levels of protein and free amino acids in theseeds and placentae of Nicotiana tabacum were studied duringseed development. Seed maturation was completed 24 days afteranthesis. During maturation, protein rapidly accumulated inthe seeds between the 6th and 18th day, along with an appreciablecompositional change in the protein amino acids as the proportionsof glutamic acid and arginine increased. The amount of freeamino acids in the seeds gradually decreased throughout maturation.The major free amino acid on the 6th day after anthesis wasglutamine, which then drastically decreased between the 6thand 12th day with increases of glutamic acid, proline, arginineand alanine. The latter amino acids decreased thereafter untilthe 24th day. On the other hand, the amount and composition of the proteinsin the placentae did not change significantly throughout seedmaturation. In the early stage of development, the major freeamino acids in the placentae were glutamine, asparagine andglutamic acid, while in the later stage asparagine was mostabundant. The present study was undertaken to select the most preferred deproteinizing agent for microbiological or chromatographic determination of free amino acids in plasma. The highest quantities of amino acids were obtained from samples treated with picric acid, ethanol or sulfosalicylic acid. It was also found from examining the recoveries of 14C-labeled amino acids added to plasma following treatment with picric acid, ethanol and sulfosalicylic acid that one kind of deproteinizing agent is not equally effective with all amino acids. Picric acid treatment gave the most reliable results for determination of amino acids except the basic amino acids, alanine and tryptophan. Sulfosalicylic acid was the most recommended deproteinizing agent for basic amino acids and ethanol was good for the assay of alanine and tryptophan. The response of d-amino acids to the deproteinizing agents tested was not always similar to that of their l-isomers.  相似文献

目的:通过检测外源氨基酸胁迫下植物体内的游离氨基酸含量,探讨氨基酸胁迫下植物生长受抑制的机制。方法:小麦胚接种在加入D-丙氨酸、D-丝氨酸和L-缬氨酸等3种外源氨基酸的培养基中,其发芽和生长会受到强烈抑制。取材胁迫植株和对照植株,提取并用HPLC方法检测游离氨基酸含量,分析主要氨基酸特征的改变。结果:作为处理的胁迫氨基酸在体内有数倍到数十倍的增高,其他氨基酸,尤其是同族及相近族氨基酸的量也出现较大改变,有增加也有减少,有些氨基酸甚至检测不出。结论:在外源氨基酸胁迫下,植物可直接吸收这些氨基酸,胁迫氨基酸在体内的积累。至少影响到了部分氨基酸的合成,使细胞内正常游离态氨基酸的数量增加或减少,这些变化引起代谢失调,进而引起生长抑制。  相似文献

The free amino acid content of the hippocampus, obtained at postmortem, has been analysed in cases of Alzheimer's disease and compared with normal cases. There were no significant differences in the levels of 23 amino acids including the transmitter candidates γ-aminobutyric, glutamic or aspartic acids. This finding is interpreted in relation to present knowledge of transmitter pathways in the region of the hippocampus. A tendency for some amino acids to be increased in the Alzheimer group reached statistical significance for arginine. This observation is consistent with increased proteolytic or peptidase activity in Alzheimer's disease.  相似文献

采用氨基酸自动分析仪测定荔枝核水提取液、醇提取液样品。在醇提取液中游离氨基酸的含量高于水提取液,在两种提取液中共检出了21种游离氨基酸(其中4种为未知氨基酸,6种必需氨基酸);已知游离氨基酸的质量分数为307μg.kg-1(醇提取液)和269μg.kg-1(水提取液),总游离氨基酸的质量分数为500μg.kg-1,必需氨基酸的量占总游离氨基酸量的44%。结论:荔枝核中的游离氨基酸具有利用价值。  相似文献

In our searching program for novel sorbicillin related compounds, three novel compounds, spirosorbicillinols A (1), B (2), and C (3), were isolated from the fermentation broth of the USF-4860 strain isolated from a soil sample. The planar structures of compounds 13 were determined from spectroscopic evidence and degradation reaction, and that of 1 was the same as that of 2. The relative stereochemistries of compounds 13 were determined by 1H-1H coupling constants, the elucidation of HMBC and NOESY spectra in detail. 1 and 2 were stereoisomers at C8 position, each other. We propose that compounds 1 and 2 were formed by exo and endo intermolecular Diels-Alder reaction between sorbicillinol as a diene and scytolide (proposed precursor-1) as a dienophile, respectively. Similarly, we propose that compound 3 was formed by an endo intermolecular Diels-Alder reaction between sorbicillinol and proposed precursor-2.  相似文献

Chemical information is used in a variety of contexts including habitat recognition, foraging and predator avoidance, and a plethora of studies have shown that an individual's distinct chemical profile can mediate interactions with conspecifics. Interestingly, recent work has revealed that diet alone is sufficient to change the chemical profile of individuals, and with it, the way other individuals perceive and interact with them. Free amino acids are known to be utilised across species in a range of contexts, including during predator–prey and host–parasite interactions, and have been widely postulated to underpin diet‐mediated social interactions, especially in fish. However, so far no empirical evidence has been presented to support this suggestion. Using an established behavioural assay in three‐spined sticklebacks (Gasterosteus aculeatus), we aimed to assess association preferences in groups of fish fed experimental diets which differed by a single free amino acid. Our results demonstrate that free amino acids alone are sufficient to mediate interindividual association preferences, raising the possibility that such a mechanism may be widespread among aquatic animals.  相似文献
