Most cardenolides used in the therapy of cardiac insufficiency are 5β-configured and thus the stereo-specific reduction of the Δ4,5-double bond of a steroid precursor is a crucial step in their biosynthesis. This step is thought to be catalysed by progesterone 5β-reductases. We report here on the isolation of 11 progesterone 5β-reductase (P5βR) orthologues from 5β-cardenolide-free and 5β-cardenolide-producing plant species belonging to five different angiosperm orders (Brassicales, Gentianales, Lamiales, Malvales and Solanales). Amino acid sequences of the P5βR described here were highly conserved. They all contain certain motifs qualifying them as members of a class of stereo-selective enone reductases capable of reducing activated CC double bonds by a 1,4-addition mechanism. Protein modeling revealed seven conserved amino acids in the substrate-binding/catalytic site of these enzymes which are all supposed to exhibit low substrate specificity. Eight P5βR genes isolated were expressed in Escherichia coli. Recombinant enzymes reduced progesterone stereo-specifically to 5β-pregane-3,20-dione. The progesterone 5β-reductases from Digitalis canariensis and Arabidopsis thaliana reduced activated CC double bonds of molecules much smaller than progesterone. The specific role of progesterone 5β-reductases of P5βRs in cardenolide metabolism is challenged because this class of enone reductases is widespread in higher plants, and they accept a wide range of enone substrates.  相似文献

Both the 5α, 6α- and 5β, 6β-dichloromethylene adducts (2a and 2b) of 3β-acetoxy-5-androsten-17-one (1) are produced when the latter is exposed to dichlorocarbene generated from chloroform and base by Phase Transfer Catalysis using ultrasound as a means of agitation. The 1H NMR substituent effects of 5α, 6α- and 5β, 6β-dichloromethylene on the angular methyl groups (Zürcher values) are given. Digitalis purpurea normal callus suspension culture is capable of metabolizing 5β-pregnane-3,20-dione (1) to 3β-hydroxy-5β-pregnan-20-one (2), 3α-hydroxy-5β-pregnan-20-one (3), 3β-hydroxy-5β-pregnan-20-one glucoside (7) and 3α-hydroxy-5β-pregnan-20-one glucoside (8). Digitalis purpurea habituated callus suspension culture is also capable of metabolizing 1 to 2, 3, 5β-pregnane-3β,20β-diol (5), (7), (8), 5β-pregnane-3β,20α-diol monoglucoside (9) and 5β-pregnane-3α,20α-diol monoglucoside (11). Furthermore, it was observed that 3β-hydroxy-5β-pregnan-20-one (2) is converted to 7, 9 and 11 by both suspension cultures. At the same time, 1, 3, 5 and 8 were detected in normal callus, while 5β-pregnane-3β,20α-diol (4) and 5β-pregnane-3β,20β-diol monoglucoside (10) were present in the habituated callus culture.  相似文献

We demonstrate that loss of succinate dehydrogenase 5 (SDH5) expression initiates epithelial-mesenchymal transition (EMT), which is visualized by the repression of E-cadherin and up-regulation of vimentin in lung cancer cell lines and clinical lung cancer specimens. In SDH5 knock-out mice, lung epithelial cells exhibited elevated mesenchymal markers, which is characteristic of EMT. Using a human lung xenograft-mouse model, we observed that knocking down endogenous SDH5 in human carcinoma cells leads to the development of multiple lymph node metastases. Moreover, our data indicate that SDH5 functions as a critical protein in regulating EMT by modulating the glycogen synthase kinase (GSK)-3β-β-catenin signaling pathway. These results reveal a critical role for SDH5 in EMT and suggest that SDH5 may be a prognostic biomarker and potential therapeutic target for lung cancer metastasis.  相似文献

17β-Nandrolone (17β-NT) is one of the most frequently misused anabolic steroids in meat producing animals. As a result of its extensive metabolism combined with the possibility of interferences with other endogenous compounds, detection of its illegal use often turns out to be a difficult issue. In recent years, proving the illegal administration of 17β-NT became even more challenging since the presence of endogenous presence of 17β-NT or some of its metabolite in different species was demonstrated. In bovines, 17α-NT can occur naturally in the urine of pregnant cows and recent findings reported that both forms can be detected in injured animals. Because efficient control must both take into account metabolic patterns and associated kinetics of elimination, the purpose of the present study was to investigate further some estranediols (5α-estrane-3β,17β-diol (abb), 5β-estrane-3α,17β-diol (bab), 5α-estrane-3β,17α-diol (aba), 5α-estrane-3α,17β-diol (aab) and 5β-estrane-3α,17α-diol (baa)) as particular metabolites of 17β-NT on a large number of injured (n=65) or pregnant (n=40) bovines. Whereas the metabolites abb, bab, aba and baa have previously been detected in urine up to several days after 17β-NT administration, the present study showed that some of the isomers abb (5α-estrane-3β,17β-diol) and bab (5β-estrane-3α,17β-diol) could not be detected in injured or pregnant animals, even at very low levels. This result may open a new way for the screening of anabolic steroid administration considering these 2 estranediols as biomarkers to indicate nandrolone abuse in cattle.  相似文献

Mena is an Ena/VASP family actin regulator with roles in cell migration, chemotaxis, cell-cell adhesion, tumor cell invasion, and metastasis. Although enriched in focal adhesions, Mena has no established function within these structures. We find that Mena forms an adhesion-regulated complex with α5β1 integrin, a fibronectin receptor involved in cell adhesion, motility, fibronectin fibrillogenesis, signaling, and growth factor receptor trafficking. Mena bound directly to the carboxy-terminal portion of the α5 cytoplasmic tail via a 91-residue region containing 13 five-residue "LERER" repeats. In fibroblasts, the Mena-α5 complex was required for "outside-in" α5β1 functions, including normal phosphorylation of FAK and paxillin and formation of fibrillar adhesions. It also supported fibrillogenesis and cell spreading and controlled cell migration speed. Thus, fibroblasts require Mena for multiple α5β1-dependent processes involving bidirectional interactions between the extracellular matrix and cytoplasmic focal adhesion proteins.  相似文献

Incubation of o-nitrophenyl 6-deoxy-β-d-xylo-hex-5-enopyranoside (1) with emulin β-d-glucosidase gave, instead of the expected 6-deoxy-d-xylo-hexos-5-ulose (3), o-nitrophenyl 6-deoxy-3-O-(6-deoxy-β-d-xylo-hex-5-enopyranosyl)-β-d-xylo-hex-5-enopyranoside (2) in high yield (≈90% under optimal conditions). The structure of 2 was established from spectroscopic data and by correlation with compounds synthesised definitively. The specificity of the transfer reaction is discussed as an argument for an acceptor or aglycon binding-site.  相似文献

Summary With growing cells of a Nocardia sp., isolated from soil, the degradation of 5-pregnene-3, 20-diol into 3-[5-oxo-7a-methyl-1 (1-hydroxo)-ethyl-3a-perhydroindane-4]-propionic acid was investigated. 5α-Androstane-3α, 16α 17β-triol was synthesized from 3β-hy-droxy-5-androsten-17-one. The procedure Involved catalytic hydrogenation of 3β-hydroxy-5-androsten-17-one to 3β-hydroxy-5α-androstan-17-one. This was followed by conversion of the 3β-hydroxy group to 3α-benzoyloxy group by the Mitsunobu reaction. Further treatment with isopropenyl acetate yielded 5α-androsten-16-ene-3α, 17-diol 3-benzoate 17-acetate. This was then converted to 3α, 17-dihydroxy-5α-androstan-16-one 3-benzoate 17-acetate via the unstable epoxide intermediate after treatment with m-cloroperoxybenzoic acid. LiAlH4 reduction of this compound formed 5α-androstane-3α, 16α, 17β-trlol. 1H and 13C NMR of various steroids are presented to confirm the structure of this compound.  相似文献

Oxysterols have been shown to induce apoptosis in a variety of cell lines. The mechanism of oxysterol-induced apoptosis is mainly known at the post-mitochondrial level. The aim of the present study was to compare the pathway of apoptosis induced by the oxysterols 7-hydroxycholesterol (7-OH) and cholesterol-5,6-epoxide (-epoxide) in U937 cells. To this end, we employed a range of inhibitors of apoptosis; a broad-spectrum caspase inhibitor, a specific caspase-3 inhibitor and an inhibitor of cytochromec release and the antioxidants; trolox, ebselen and resveratrol. The three inhibitors of apoptosis prevented cell death induced by 7-OH; however, in -epoxide-treated cells, the inhibitor of cytochromec release did not protect against apoptosis. The cellular antioxidant glutathione was depleted in 7-OH-treated cells but not in cells incubated with -epoxide. Trolox, a water-soluble synthetic analogue of -tocopherol, prevented 7-OH-induced apoptosis but did not protect against cell death induced by -epoxide. Ebselen and resveratrol did not protect U937 cells against apoptosis induced by either 7-OH or -epoxide. Our results suggest that differences occur in the pathways of apoptosis induced by 7-OH and -epoxide in U937 cells.  相似文献

作为开发新型实用性人绒毛膜促性腺激素(hCG)疫苗的一种尝试, 我们已构建若干组合靶抗原三个线性B- 细胞表位和外源强T- 细胞表位的基因工程hCG嵌合肽。为了检测用这些嵌合肽免疫的动物血清中是否能产生抗各表位的三种抗体,本研究选用能在大肠杆菌中高表达和与生物素亲和性强且特异(方便通过亲和层析纯化)的链霉亲和素为载体,分别构建了三种含β-hCG不同单一线性B_细胞表位(β5,β9和β8)的融合蛋白。在链霉亲和素基因下游多克隆区EcoRⅠ和Hind Ⅲ位点插入各表位编码基因片段(带TAA终止密码子)的pTSA-18重组质粒, 转化BL21(DE3)pLysS宿主菌后, 它们在IPTG诱导下均能以较高水平表达各自目的融合蛋白,而且它们的表达产物在Western blot鉴定中都能被抗各表位特异的多抗或单抗或抗报告表位单抗识别。用改良的制备性PAGE方法可以一步纯化电泳均一性高于95%的三个融合蛋白, 它们的收得率相对1L培养物约为5 mg。作为化学合成表位肽的替代物, β-hCG三个单一B- 细胞表位融合蛋白的可获得性将有助于所构建hCG基因工程嵌合肽以及其他hCG疫苗,也包括它的DNA疫苗的免疫原性分析。  相似文献

从鸽的骨髓和肝组织中扩增到2个禽β-防御素(AvBD)基因,在大肠杆菌中高效表达,检测其重组蛋白的抗菌活性。应用RT-PCR方法从鸽的骨髓和肝脏组织中扩增AvBD5基因,根据已发现的禽β-防御素和部分哺乳类动物β-防御素-5的氨基酸序列构建系统进化树,将该基因克隆到大肠杆菌原核表达载体pGEX-6p-1上进行原核表达,对该重组蛋白进行纯化,测定体外抗菌活性与理化特性。测序得到这2个基因的cDNA大小均为201 bp,编码66个氨基酸残基,内含6个位置保守的半胱氨酸残基。经遗传进化分析发现,该基因推导的两组氨基酸序列与鸭AvBD5的同源性最高,分别为87.9%和78.8%,这两组氨基酸序列的同源性为83.3%。因此,将其分别命名为鸽AvBD5α(骨髓)和鸽AvBD5β(肝脏)。进一步将这两个基因分别亚克隆到大肠杆菌pGEX-6p-1载体中进行原核表达。两个重组融合蛋白经纯化后,通过菌落计数法测定其体外抗菌活性,盐离子浓度对其抗菌活性的影响,及其融合蛋白的溶血活性。结果表明,重组鸽AvBD 5α、AvBD 5β融合蛋白的分子量约为32 kDa。具有明显的抗菌活性,对12种细菌均有不同程度的抑制作用,高盐浓度对其抗菌活性显著影响。此外,这两个重组蛋白对红细胞无显著溶血活性。  相似文献

A reliable radioimmunoassay for the determination of 5-androstene-3β, 17β-diol in plasma is described. Antisera were obtained by immunization of rabbits with 3β,17β-dihydroxy-5-androsten-16-one coupled to bovine serum albumin in position 16. The antiserum was characterized by titer, affinity, and specificity. Only dehydroepi-androsterone (24.3 %) and pregnenolone (2.7 %) showed a small crossreactivity. The assay method consisted of extraction with ether, thin-layer chromatography and endpoint determination.The reliability of the method was studied. The interassay variability was 7.5 % at a concentration of 1.22 μg/l. The limit of detection was 0.068 μg/l. Specificity was achieved by Chromatographic separation of the crossreacting steroids. Mass recovery experiments with 250 and 500 pg were performed, in which 99.0 ± 4.6 % of the smaller and 97.6 ± 11.3 % of the greater mass were recovered. In 45 healthy adult males plasma concentrations between 0.44 and 1.80 μg/l were found. The median was 1.06 μg/l. Stimulation of the Leydig cells with human chorionic gonadotropin (HCG) increased plasma concentrations by 93 % (average in 12 males). Therapeutic castration in 8 men caused an average decrease of 55.4 % in plasma values.  相似文献
