采用去壁低渗方法,观察研究了玉兰Magnolia denudata有丝分裂和减数分裂的细胞学特征。实验结果证实玉兰存在两种染色体倍性,即2n=4x=76和2n=6x=114。通常,在木兰属甚至整个木兰科每个物种只具有一种染色体数目。玉兰有丝分裂间期核为复杂染色中心型,其中期染色体较小。玉兰在减数分裂中期I的构型表现出多样性,其中最主要的特点是比同源多倍体预期的二价体出现的频率更高些,其次是在减数分裂中期I可以观察到1或2个环状和(或)链状六价体。这些特征与同源异源六倍体或部分的异源六倍体种北美红杉Sequ  相似文献

Wild potato species are widely distributed in the Americas, where they spontaneously grow in very diverse habitats. These species - with low chromosome differentiation - form polyploid series with 2n = 2x, 3x, 4x and 6x (x =12). They are isolated in nature by external and internal hybridisation barriers that can be incomplete, allowing hybridisation in areas of sympatry. Nevertheless, most accessions in germplasm banks, regardless of genetic background of the sampled spontaneous populations, have been assigned specific categories based on morphological characters. To further investigate the extent of hybridisation in the group and for comparative purposes, pollen viability was estimated in (i) artificial hybrids between a commercial cultivar (Calén INTA) of the common potato (tetraploid Solanum tuberosum ssp. tuberosum) and the tetraploid cytotype of the related wild species S. gourlayi, and (ii) samples of plants (accessions) and inflorescences of natural populations from Argentina, tentatively classified as 'presumed hybrids' (S. infundibuliforme-S. gourlayi) and 'species' (S. infundibuliforme, S. gourlayi and S. chacoense). Regardless of origin, 98 out of 103 plants analysed had zero to 70% pollen viability (zero to 40% in eight of them). Pollen grains were of variable size and morphology and, in mostly male sterile plants, the only viable pollen grains were 2n and/or 4n. Furthermore, male sterile plants shared various abnormalities in meiosis I and II (unpaired chromosomes, unequal chromosome distribution, precocious/lagging chromosomes, parallel, tripolar, fused and multiple spindles, unequal size nuclei, dyads, triads and pentads in addition to normal tetrads, among others). These results provide novel evidence to support field observations of early potato botanists on the extent of spontaneous hybridisation in wild Argentinian potato populations, which is not reflected in the current taxonomy and has significant consequences for germplasm conservation and breeding.  相似文献

Chromosome number, meiotic behavior, and pollen viability were analyzed in 15 species of two genera, Vriesea and Aechmea, native to Rio Grande do Sul, Brazil. This study is the first cytogenetic analysis of these taxa. The chromosome numbers are all n = 25, consistent with the proposed base number of x = 25 for Bromeliaceae. All examined taxa displayed regular bivalent pairing and chromosome segregation at meiosis. Observed meiotic abnormalities include univalents in metaphase I; missing or extra chromosomes and precocious division of centromeres in metaphase II; laggards in telophase I and anaphase II/telophase II. The high pollen viability (>88%) reflects a regular meiosis.  相似文献

Campuloclinium macrocephalum DC. is a perennial herb widely distributed in the New World and introduced in South Africa, where it is commonly called “pompom weed”. This species is considered one of the most important weeds of Brazil and one of the problematic invasive plants of South Africa. The meiotic system can be studied to assess the ability of a weed to spread, but only few studies on C. macrocephalum have been realized. In this study, we examined the meiotic behavior and pollen fertility of 14 natural populations of C. macrocephalum from Argentina and Uruguay. Meiotic analysis revealed 2 triploid (2n = 3x = 30), 11 tetraploid (2n = 4x = 40) and 1 mixed population (2n = 2x = 20, 2n = 4x = 40). Both, triploid and tetraploid specimens showed a widely variable meiotic behavior with irregular chromosome pairing showing univalents, bivalents, trivalents (in triploids) and tetravalents (in tetraploids) at diacinesis of first meiotic division. Different abnormalities were observed, such as: laggard chromosomes, chromatin bridges, and out of plate chromosomes at metaphase I. During meiosis I (prophase), some cells showed the phenomenon of cytomixis or chromatin transfer between pollen mother cells. The meiotic indexes suggest that only four populations were normally fertile (over 90 % of fertile pollen), indicating meiotically stable plants. The remaining populations share variable pollen fertility, with triploids ranging from 46.64 to 54.83 % and tetraploids varying from 3.54 to 45.30 %. We suggest that polyploidy seems to be recurrent in C. macrocephalum, promoting partial sterility of pollen grains, generating large numbers of individuals by apomixis promoting invasion of crop fields. This study presents the meiotic behavior of this weed, these could be useful for future studies of biological control in areas with no natural enemies.  相似文献

We report the occurrence of cytomixis in wild populations of Himalayan poppy (Meconopsis aculeata Royle), which is considered to be an important and threatened medicinal plant growing in the high hills of the Himalayas. The impact of cytomixis on meiotic behaviour, reduced pollen viability and heterogeneous-sized pollen grains was also studied. Cytological studies in the seven wild populations from the high hills of Himachal Pradesh revealed that all the Himalayan populations exist uniformly at the tetraploid level (2n=56) on x=14. The phenomenon of chromatin transfer among the proximate pollen mother cells (PMCs) in six populations caused various meiotic abnormalities. Chromatin transfer also resulted in the formation of coenocytes, aneuploid, polyploid and anucleated PMCs. Among individuals that showed chromatin transfer, chromosome stickiness and interbivalent connections were frequently observed in some PMCs. The phenomenon of cytomixis in the species seems to be directly under genetic control; it affects the meiotic course considerably and results in reduced pollen viability.  相似文献

The genus Lolium comprises several species of economical importance in temperate countries, mostly due to their high nutritional value and adaptability to cold environments, including southern regions of Brazil. Recently, several diploid cultivars and populations, as well as synthetic tetraploid cultivars have been explored. In order to viabilize or to direct crossings, it is important that the accessions present regular meiosis, thus, producing viable pollen grains. In this way, this study aimed at investigating the meiosis of nine accessions of Lolium multiflorum used in breeding programs, seeking to evaluate its viability in future crossings. The meiosis of diploid plants (2n?=?2×?=?14) is more regular than the artificially tetraploidized genotypes (2n?=?4×?=?28). In the tetraploids, univalent, bivalent, and multivalent configurations were observed. The irregularities were associated to mixoploidy, irregular segregation of chromosomes, spindle disorders, asynchrony, micronuclei, and cellular fusion and formation of syncyte. The abnormalities affected the meiotic index of tetraploid cultivars, characterizing them as unstable. Nevertheless, both diploid and tetraploid genotypes are considered able to participate in crossings because, although there are abnormalities, they do not occur at levels that could affect the production of viable pollen grains.  相似文献

 This paper reports the occurrence of chromosome elimination during microsporogenesis in a Brazilian accession of Paspalum subciliatum. The accession was tetraploid (2n=4x=40) and meiosis was normal until diakinesis, with 20 regularly distributed bivalents. Starting at metaphase I, meiosis was very peculiar. In this phase, while ten bivalents were clustered in the equatorial plate, the other ten were still dispersed in the cytoplasm. In anaphase I the chromosomes showed different abilities to migrate to the poles. While one genome reached the poles in telophase I, the laggard was in metaphase or anaphase and was engulfed by extra nuclei. In the second division, behavior was the same. Our results show clear asynchrony in cell cycle, especially in some meiotic phases. Unfortunately we cannot explain the causes of the phenomenon, but this event shows once more that chromosome elimination serves as an incompatibility barrier preventing divergent genomes from coexisting in the same cellular system. In the cattail Typha latifolia the four haploid products of meiosis remain attached and form the flat tetrad of pollen grains. Gametophytic lethals arisen de novo in diploid cells of sporophyte must manifest themselves as pollen tetrads with two dead grains. This could allow to estimate the rate of recessive lethals arresting pollen grain development. We studied pollen samples collected from 44 sprouts in two populations in the vicinity of Novosibirsk. The anomalous tetrads T1, T2, T3, and T4 carrying one, two, three, and four dead grains, respectively, were detected in each sampled individual. The mean frequency of all anomalous tetrads in the two populations was 3.4% and 8.7%. The frequencies of tetrad classes varied widely among the individuals with correlation coefficient up to 0.94, but their ratios remained nearly constant. The majority of anomalous tetrads were presented by T1 and T2 classes (their sum comprising 72.7 and 74.0% in two populations), T1 being a little more abundant. The observed pattern of frequencies of tetrads with dead grains can be explained by errors of male meiosis such as chromosome non-disjunction in both meiotic divisions. The tetrads with two dead pollen grains may result mostly from non-disjunction in anaphase I, and those with one pollen grain from non-disjunction in anaphase II, thus making tetrad analysis ineffective for estimating the rate of gametophytic lethals.  相似文献

Jacobaea vulgaris subsp. vulgaris (syn. Senecio jacobaea subsp. jacobaea) constitutes an intricate polyploid complex distributed in Europe. Four cytotypes have been reported in this species, three with euploid (diploid, tetraploid and octoploid; 2n=20, 40 and 80) and one with aneuploid (2n=32) chromosome numbers. Here we report that the diploid chromosome number (2n=20) reported from Bulgaria is due to misidentification with Jacobaea aquatica. On the other hand, we have discovered a new, hexaploid (2n=6x=60) cytotype within J. vulgaris subsp. vulgaris using flow cytometry. The new cytotype occurs within four sympatric populations of otherwise tetraploid and octoploid plants in Pannonia (one locality in the eastern Czech Republic and two localities in southwestern Slovakia) and in Podillya (one locality in western Ukraine). The frequency of hexaploid individuals within 76 studied populations is very low (only 10 of 693 analysed plants), and hexaploids probably represent hybrids between tetraploid and octoploid plants. Three mixed populations with hexaploid plants were subjected to detailed morphological and pollen fertility analyses. Multivariate morphometric analysis reveals partial separation of tetraploid and octoploid plants, whereas hexaploid individuals are similar in morphology to octoploids. In comparison with tetraploids, octoploids and hexaploids exhibit slightly longer ray florets, involucral bracts and tubular florets and more hairy outer achenes. Hexaploid plants display larger pollen grains and lower pollen fertility compared to tetraploids and octoploids.  相似文献

对巴东木莲Manglietia patungensis及其近缘种乳源木莲M. yuyuanensis的花粉母细胞减数分裂过程的基本特征进行了比较研究。乳源木莲与巴东木莲的染色体数目和核型相同,但不经任何人为因素诱导,它们之间在减数分裂过程中的染色体行为上有明显差异。(1)巴东木莲减数分裂中期I构型为0.30IV+18.33II+0.15I,与乳源木莲构型19II不同,巴东木莲可能存在同臂内倒位杂合子,染色体结构存在一定的杂合性。(2)后期I和后期II染色体行为异常现象发生频率明显不同。以后期II为例,乳源木莲减数分裂相中有迟滞染色体的细胞占8.8%,迟滞染色体不超过2个;巴东木莲有迟滞染色体等异常现象的细胞占29.2%,迟滞染色体最高达11个,还出现染色体碎裂成断片现象。巴东木莲减数分裂过程中染色体组表现出染色体结构杂合变异和迟滞染色体与染色体的断裂频率很高的异常现象在一定程度上可能影响了雄配子体的发育。  相似文献

Present cytological investigations from the cold desert regions of Lahaul-Spiti and Kinnaur (India) record the first ever tetraploid (2n=32) chromosome count and cytomixis in Clematis orientalis L. var. acutifolia Hook. f. et Thoms. The phenomenon of cytomixis (9.33–29.80 %) involving chromatin transfer among 2–3 proximate pollen mother cells (PMCs) during male meiosis occurs through narrow and broad cytoplasmic channels from early prophase to tetrad stage. However, frequency of its occurrence during the later meiotic stages is rather low. Chromatin transfer results into the formation of hypo-, hyperploid and enucleated PMCs. Various meiotic abnormalities associated with cytomixis such as chromatin stickiness, pycnotic chromatin, interbivalent connections, out of plate bivalents, late disjunction of bivalents, and laggards and bridges resulted into some pollen sterility (16.33–49.30 %) and heterogeneous pollen grains size.  相似文献

Meiotic division and male gametophyte development were analyzed in one tetraploid (2n = 4x = 36) accession of Brachiaria decumbens cv. Basilisk that showed some pollen sterility. Meiotic process was typical of polyploids in that it consisted of multiple chromosome associations. Precocious chromosome migration to the poles, laggards, and micronucleus formation were abundant in both meiosis I and II and resulted in tetrads with micronuclei. After callose dissolution, microspores were released into the anther locule and had the semblance of being normal. Although each microspore initiated its differentiation by pollen mitosis, in 43.24% of the microspores, nuclear polarization was not observed and the typical hemispherical cell plate was not detected. Division was symmetric and microspores lacked differentiation between the vegetative and the generative cell. Both nuclei were of equal size, presented equal chromatin condensation, and had a spherical shape. After the first pollen mitosis and cytokinesis, each cell underwent a new symmetric mitosis without nuclear polarization. At the end of the second pollen mitosis, four equal nuclei were observed in each pollen grain. After the second cytokinesis, the cells gave rise to four equal-sized pollen grains with a similar tetrad configuration that initially remained together. Sterile pollen grains resulted from abnormal pollen mitosis. This anomaly may be explained by a mutation, probably affecting microtubule cytoskeleton formation. The importance of this male-sterile mutation for Brachiaria breeding programs is discussed.  相似文献

This is the first report on chromosome numbers and the reproductive behaviour in Stenodrepanum Harms, a rare endemic and monotypic legume genus from the arid and salty areas of central-western Argentina. Sixty individuals belonging to two populations from two salty areas ("salinas") were surveyed and included mostly triploid (2n = 3x = 36) and only two diploid (2n = 2x = 24) plants. Meiosis in diploids is regular, with bivalent pairing and uniform and viable pollen. In contrast, meiosis in triploids is characterized by high trivalent pairing, with irregularly shaped pollen and variation in cytoplasm content and stainability, which is in agreement with an unbalanced segregation occurring in anaphases I and II. However, different triploid plants/individuals showed various degrees of pollen fertility, which may be attributed to particular genotypes. Research on reproductive biology events indicates sexual cross-pollinated reproduction enhanced by protogyny in both cytotypes. All plants produced seeds, but seedlings were only recovered from diploid plants pollinated with triploids, and even those eventually perished. Chromosome counts in these seedlings revealed aneuploid chromosome numbers owing to the combination of unbalanced gametes.  相似文献

Houttuynia cordata (Saururaceae) is a leaf vegetable and a medicinal herb througout much of Asia. Cytomixis and meiotic abnormalities during microsporogenesis were found in two populations of H. cordata with different ploidy levels (2n = 38, 96). Cytomixis occurred in pollen mother cells during meiosis at high frequencies and with variable degrees of chromatin/chromosome transfer. Meiotic abnormalities, such as chromosome laggards, asymmetric segregation and polyads, also prevailed in pollen mother cells at metaphase of the first division and later stages. They were caused by cytomixis and resulted in very low pollen viability and male sterility. Pollen mother cells from the population with 2n = 38 showed only simultaneous cytokinesis, but most pollen mother cells from the population with 2n = 96 showed successive cytokinesis; a minority underwent simultaneous cytokinesis. Cytomixis and irregular meiotic divisions appear to be the origin of the intraspecific polyploidy in this species, which has large variations in chromosome numbers.  相似文献

Polyploidy is a major mechanism of speciation and adaptation, yet little is known about the origins of polyploids in natural species. I investigated gametic nonreduction and neopolyploid formation in natural tetraploid populations of Achillea borealis (Asteraceae), an autopolyploid complex consisting of tetraploid and hexaploid cytotypes. Cytological analyses of tetraploid populations revealed the occurrence of reduced (n=2x) as well as unreduced 'big' (2n=4x) and 'jumbo' (4n=8x) pollen grains, which were clearly distinguished by size. Production of unreduced pollen was monitored in two tetraploid populations in 1997 and 1998. Mean population-level frequencies of unreduced pollen ranged from 0.030 to 0.538%, with as few as one-third and as many as one-half of sampled plants producing unreduced grains. Eight individuals were found to produce >1% unreduced pollen, with highest observed frequencies of 7.0, 13.2 and 15.8%. Experimental crosses using high unreduced pollen producers as male parents generated viable seeds. However, the frequency of neohexaploids in the progeny of experimental crosses (0.388%) was similar to that observed in progeny of randomly selected, open-pollinated control parents (0.465%). These results suggest that unreduced eggs are the most likely source of new polyploids. In spite of the inefficiency of unreduced pollen in unilateral sexual polyploidization, the overall rate of neohexaploid formation (one in 233) was several orders of magnitude greater than estimates of genic mutation rates.  相似文献

 Cytogenetic studies carried out over a period of 2 consecutive years on a native Brazilian accession of Paspalum regnellii (2n=40) revealed a meiotic mutation that has not been previously reported for any other species. Among 13 inflorescences investigated during the first collection year, three presented anomalous meiotic behavior starting from metaphase I. At the beginning of this phase, the chromosomes occupied the entire equatorial plate in a membrane-to-membrane arrangement, and the spindle fibers, which were clearly visible, did not converge towards the poles. Degeneration of spindle fibers occurred at the end of metaphase I. Chromosome segregation did not occur and the bivalents were left scattered at random in the cytoplasm. Remnants of chromosome fibers could be seen close to the centromere during this stage. The bivalents gave origin to micronuclei in telophase I, with extremely wide variations in number and size among cells. With the absence of spindle formation during meiosis II, metaphase and anaphase II were not observed. Second cytokinesis occurred in prophase II cells after the occurrence of first cytokinesis. The final product of meiosis was completely abnormal, with a predominance of polyads with microspores of different sizes that resulted in abortive pollen grains.
