The common quail (Coturnix coturnix) is a popular game species for which restocking with farm-reared individuals is a common practice. In some areas, the number of released quails greatly surpasses the number of wild breeding common quail. However, common quail are difficult to raise in captivity and this casts suspicion about a possible hybrid origin of the farmed individuals from crosses with domestic Japanese quail (C. japonica). In this study we used a panel of autosomal microsatellite markers to characterize the genetic origin of quails reared for hunting purposes in game farms in Spain and of quails from an experimental game farm which was founded with hybrids that have been systematically backcrossed with wild common quails. The genotypes of these quail were compared to those of wild common quail and domestic strains of Japanese quail. Our results show that more than 85% of the game farm birds were not common quail but had domestic Japanese quail ancestry. In the experimental farm a larger proportion of individuals could not be clearly separated from pure common quails. We conclude that the majority of quail sold for restocking purposes were not common quail. Genetic monitoring of individuals raised for restocking is indispensable as the massive release of farm-reared hybrids could represent a severe threat for the long term survival of the native species.  相似文献

Many cases of introgressive hybridization have been reported among birds, particularly following introduction to the natural environment of individuals belonging to non-native similar taxa. This appears to be the case for common quail (Coturnix coturnix) in France where wild populations artificially come into contact with domesticated Japanese quail (Coturnix japonica) raised for meat and egg production but sometimes released for hunting purposes. In order to highlight the possible existence of gene flows between both taxa, a comparison of nuclear (25 microsatellite loci) and mitochondrial (sequencing and RFLP) DNA polymorphisms was performed on 375 common quails (from France, Spain and Morocco) and 140 Japanese quails (from France and Japan). Genetic diversity was assessed, and analyses (Factorial Correspondence Analysis, Bayesian admixture) of molecular polymorphisms revealed clear differentiation between the two taxa, making it possible to detect for hybrids among quails sampled in the wild. Eight birds expected to be common quail were found to be two pure Japanese quail, one probable backcross to C. japonica, three F1/F2 hybrids, and two probable backcrosses to Coturnix coturnix. These results show that Japanese quails were released and suggest that the two taxa hybridize in the wild. They confirm the urgent need for preventing the release of pure Japanese or hybrid quails to preserve the genetic integrity of C. coturnix. The tools developed for this study should be useful for accurate monitoring of wild quail populations within the framework of avifauna management programs.  相似文献

In Europe, farm‐reared quail used for restocking purposes are often hybrids between the Common Quail Coturnix coturnix and the Japanese Quail Coturnix japonica. These hybrids interbreed with wild Common Quail populations and suffer higher rates of nest predation, which would suggest that the two quail types have different nest‐site selection patterns. We monitored 103 radiotagged nesting females (77 wild Common Quail over 18 breeding seasons and 26 hybrid quail over six breeding seasons) and analysed their nest placement behaviour. Our results did not provide any evidence that the quail types showed different nest‐site selection trends, as they all preferred to nest near cereal field margins. The higher nest predation rate experienced by hybrids might therefore be explained by less effective anti‐predator behaviour among hybrid females during incubation. As no edge effect on nest predation was observed, the preference for nesting close to cereal margins could be due to the greater food resources of field margins. Moreover, Common Quail nest predation decreased with a rise in barley straw production, supporting evidence that nest cover plays an important role in the prevention of nest predation in this species. No such relationship was found for hybrid quail, but this result is not conclusive, as the data on barley straw production for hybrid quail covered a much smaller range of interannual variability. Barley straw production in Catalonia decreased significantly between 1992 and 2012, possibly as a result of a change in the barley varieties used. This practice could entail a conservation threat for ground‐nesting bird species.  相似文献

Since the 1970s, the decline of the occidental populations of European Quail Coturnix c. coturnix , a partial migrant Galliform species, has driven the release of domestic Japanese Quail Coturnix c. japonica into the wild as game birds. Recent experiments suggest that reproductive isolating mechanisms have not developed to prevent hybridization between European and Japanese Quail. To assess the potential impact of hybridization between the subspecies in the wild, this study aims to compare the migratory characteristics of captive European Quail, domestic Japanese Quail, F1 hybrids and a backcross. In a laboratory experiment, birds hatched in summer and reared under a natural photoperiod were transferred during winter to an artificial photoperiod (light–dark) of 14 : 10 h. Behavioural (migratory restlessness, crowing activity in males) and physiological (sexual development, fat accumulation, body mass) measures were recorded. Domestic Japanese Quail showed no migratory tendency in response to the increased day length. F1 hybrids showed rapid sexual development and few of them exhibited the typical profile of a migrant bird. Backcrosses showed great variation in their response, comparable with those of the European Quail. Therefore, hybridization could lead to an increased proportion of quail showing sedentary rather than migratory behaviour. Furthermore, hybrids that do show migratory behaviour have the potential to facilitate Japanese gene flow into areas where the release of domestic Quail as game birds is not yet practised.  相似文献

Genetic coadaptability of wild Japanese quail, wild Common quail and Domestic quail populations in China was studied using 7 microsatellite DNA markers and Monte Carlo method to test genetic disequilibrium. The molecular effects of genetic coadaptability were analyzed through a new statistical model of neutral site. The results showed that genetic coadaptability dominated the genetic disequilibrium of the three quail populations, and totally 16.67%, 9.66% and 10.05% of non-allelic combinations were in the genetic disequilibrium in wild Japanese quail, wild Common quail and Domestic quail populations, respectively. Genetic coadaptability existed at almost all the tested sites. In the molecular point of view, genetic coadaptability plays an important role of keeping lots of polymorphisms in natural populations. Therefore, it is another key factor to the genetic disequilibrium in the population except for linkage. The results enrich the conceptions and connotations of genetic disequilibrium, and help us know more about genetic coadaptability and its effects, and lay a foundation of evaluation and protection of wild quail genetic resources in China.  相似文献

Cytological maps of lampbrush macrobivalents of the Japanese quail (Coturnix coturnix japonica) were constructed. Investigation of chiasmata allowed determination of the meiotic frequency of reciprocal genetic recombination (crossing over) in Japanese quail females. The total chiasma number in bivalents of Japanese quail oocyte nuclei was determined to be 53-58. Macrobivalents 1-5 and Z of the Japanese quail had on average 3.3 chiasmata per bivalent, and microbivalents, 1.0-1.1 chiasmata per bivalent. The chiasmata (crossover) frequency in Japanese quail females was lower than in chicks. In macrochromosomes of Japanese quail females, one crossover occurred per 43.9 Mb, and in chicken, per 30.0 Mb. Judging from chiasma frequency, the genetic length of the Japanese quail genome is likely to be 2650-2900 cM. Crossover frequency in the species was 0.023 per Mb in macrobivalents and 0.07-0.08 Mb in microbivalents and for the total genome, 0.041 crossovers per Mb. The genetic length of one Mb (theta) in female Japanese quails was 1.14 cM in macrochromosomes, 3.60-4.12 cM in microchromosomes, and about 1.96-2.15 cM averaged over the genome.  相似文献

Domestic fowl or chicken (Gallus gallus) and Japanese quail (Coturnix japonica) belong to the family Phasianidae. The exchange of marker information between chicken and quail is an important step towards the construction of a high-resolution comparative genetic map in Phasianidae, which includes several poultry species of agricultural importance. We tested chicken microsatellite markers to see if they would be suitable as genetic linkage markers in Japanese quail. Twenty-six per cent (31/120) of chicken primers amplified individual loci in Japanese quail and 65% (20/31) of the amplified loci were found to be polymorphic. Eleven of the polymorphic loci were excluded as uninformative because of the lack of amplification in some individuals or high frequency of nonspecific amplification. The sequence information of the remaining nine loci revealed six of them to contain microsatellites that were nearly identical with those of the orthologous regions in chicken. For these six loci, allele frequencies were estimated in 50 unrelated quails. Although the very few chicken markers that do work well in quail could be used as anchor points for a comparative mapping, most chicken markers are not useful for studies in quail. Therefore, more effort should be committed to developing quail-specific markers rather than attempting to adapt chicken markers for work in quail.  相似文献

Aim Information on the movements of bird populations in extensive landscapes is needed to assess environmental effects and conservation strategies over appropriate temporal and spatial scales. The common quail (Coturnix coturnix) is a game bird that breeds mainly in dense cereal crops. These crops cover huge areas of the Iberian Peninsula. The aim of this work is to relate cereal mowing, which causes rapid and massive habitat loss for the common quail, to population movements of the species during its breeding season. Location Spain, southern Europe. Methods We used ring recoveries to analyse, using circular statistics, the orientation of movements by male common quails in the northern half of Spain. Forward stepwise multiple regressions were applied to correlate (1) the Julian day of cereal harvesting at 770 locations with the respective latitude, longitude and elevation, and (2) the number of hunted birds with hunting pressure, breeding densities and mean mowing date by province. Finally, data concerning the number of quails hunted at the end of the breeding season were compared by province and year using two‐way ANOVA. Results Our results show that during the breeding season in the northern half of Spain, male quails orient their movements towards higher areas, and mainly to the northern Castilian Plateau. These are the areas of Spain where cereals are harvested latest. Moreover, records show that the number of quails hunted is significantly higher in these areas than in other areas of the country, independently of hunting pressure and breeding densities, and it is extremely high in the northern Castilian Plateau. Main conclusions Cereal mowing combined with some Spanish geographical characteristics acts as a funnel, forcing quail populations to concentrate in certain areas of Spain at the end of the breeding season. This implies that the number of quails hunted in these areas may be high, and that they therefore constitute priority conservation areas.  相似文献

In line with the Gifu University''s initiative to map the Japanese quail genome, a total of 100 Japanese quail microsatellite markers isolated in our laboratory were evaluated in a population of 20 unrelated quails randomly sampled from a colony of wild quail origin. Ninety-eight markers were polymorphic with an average of 3.7 alleles per locus and a mean heterozygosity of 0.423. To determine the utility of these markers for comparative genome mapping in Phasianidae, cross-species amplification of all the markers was tested with chicken and guinea fowl DNA. Amplification products similar in size to the orthologous loci in quail were observed in 42 loci in chicken and 20 loci in guinea fowl. Of the cross-reactive markers, 57.1% in chicken and 55.0% in guinea fowl were polymorphic when tested in 20 birds from their respective populations. Five of 15 markers that could cross-amplify Japanese quail, chicken, and guinea fowl DNA were polymorphic in all three species. Amplification of orthologous loci was confirmed by sequencing 10 loci each from chicken and guinea fowl and comparing with them the corresponding quail sequence. The microsatellite markers reported would serve as a useful resource base for genetic mapping in quail and comparative mapping in Phasianidae.  相似文献

Hunting wild African harlequin quails (Coturnix delegorguei delegorguei) using traditional methods in Western Kenya has been ongoing for generations, yet their genetic diversity and evolutionary history are largely unknown. In this study, the genetic variation and demographic history of wild African harlequin quails were assessed using a 347bp mitochondrial DNA (mtDNA) control region fragment and 119,339 single nucleotide polymorphisms (SNPs) from genotyping‐by‐sequencing (GBS) data. Genetic diversity analyses revealed that the genetic variation in wild African harlequin quails was predominantly among individuals than populations. Demographic analyses indicated a signal of rapid demographic expansion, and the estimated time since population expansion was found to be 150,000–350,000 years ago, corresponding to around the Pliocene–Pleistocene boundary. A gradual decline in their effective population size was also observed, which raised concerns about their conservation status. These results provide the first account of the genetic diversity of wild African harlequin quails of Siaya, thereby creating a helpful foundation in their biodiversity conservation.  相似文献

Endangered species are often maintained in captivity to serve as a safeguard in the event of an extirpation of natural populations. However, wild and captive populations can rapidly diverge in genetic and phenotypic characteristics, including behavior, when maintained in isolation and under different environmental conditions. Here, we compare two populations of the endangered Leon Springs pupfish (Cyprinodon bovinus): a captive population maintained at the Southwestern Native Aquatic Resources and Recovery Center (SNARRC) and the only remaining wild population. Wild fish in the natural Texan Diamond Y desert spring are remnants of a past reintroduction of captive SNARRC fish, which occurred in 1998. The fifteen years of isolation have led to genetic and morphologic differences between the two populations. To assess the behavioral implications of such divergence, we released previously captive C. bovinus into natural habitat and quantified behaviors under natural conditions and in semi-controlled mesocosms and then replicated these conditions with the established natural population. Both populations exhibited similar levels of reproduction, foraging, and agonistic behavior. Despite divergence in genetic and morphometric characteristics, overall behavioral patterns of C. bovinus remained consistent in similar environments. This stability suggests that captive C. bovinus could again be successful establishing in their natural habitat. It is our hope that this investigation will help focus future conservation efforts towards monitoring the persistence of reintroduced C. bovinus in addition to providing a framework for other recovery plans reintroducing captive stock into ancestral habitats.  相似文献

Transferrin types in the Japanese quail Coturnix coturnix japonica are controlled by a single autosomal locus Tf with at least two codominant alleles TfB and Tfc. The frequencies of TfB and Tfc in a commercial population of the domestic quail were 1.00 and 0.00, respectively.
Previous studies demonstrated that in the domestic populations of the Japanese quail three electrophoretic patterns AB, B and BC existed in egg white conalbumin and that the electrophoretic variation of conalbumin occurred in parallel with that of serum transferrin. Furthermore, from the preliminary mating experiments the transferrin-conalbumin variation was proposed to be under the control of at least two codominant alleles 77s and Tfc at an autosomal locus (Kimura et al., 1977, 1978).
The present study was designed (1) to report a large amount of family data from three generations in order to support the previously published hypothesis on genetic control of electrophoretic patterns of transferrin, and (2) to survey the gene constitution of transferrin in a commercial population of the domestic quail.
Sera were added with iron, heated for 5 min at 65°C (Stratil, 1967), then were investigated by means of horizontal starch gel electrophoresis. Hydrolysed starch from Connaught Medical Laboratories, Toronto, was used. A discontinuous citrate/Tris/LiOH/borate buffer system (pH 8.0) of Ferguson & Wallace (1961) was employed. Studies on the genetic diversity and relatedness of zoo populations are crucial for implementing successful breeding programmes. The European wildcat, Felis s. silvestris, is subject to intensive conservation measures, including captive breeding and reintroduction. We here present the first systematic genetic analysis of the captive population of Felis s. silvestris in comparison with a natural wild population. We used microsatellites and mtDNA sequencing to assess genetic diversity, structure and integrity of the ex situ population. Our results show that the ex situ population of the European wildcat is highly structured and that it has a higher genetic diversity than the studied wild population. Some genetic clusters matched the breeding lines of certain zoos or groups of zoos that often exchanged individuals. Two mitochondrial haplotype groups were detected in the in situ populations, one of which was closely related to the most common haplotype found in domestic cats, suggesting past introgression in the wild. Although native haplotypes were also found in the captive population, the majority (68%) of captive individuals shared a common mtDNA haplotype with the domestic cat (Felis s. catus). Only six captive individuals (7.7%) were assigned as wildcats in the STRUCTURE analysis (at K = 2), two of which had domestic cat mtDNA haplotypes and only two captive individuals were assigned as purebred wildcats by NewHybrids. These results suggest that the high genetic diversity of the captive population has been caused by admixture with domestic cats. Therefore, the captive population cannot be recommended for further breeding and reintroduction.  相似文献

To evaluate genetic variability in two populations of the wild quail Cofurnix coturnix japonica , proteins and enzymes were examined by starch gel electrophoresis.
Rare variants so far not observed in domestic quail were found in the following five enzymes; aspartate aminotransferase, acid phosphatase, pancreatic esterase, isocitrate dehydrogenase and lactate dehydrogenase. The proportion of polymorphic loci ( P poly) and the expected average heterozygosity ( H ) in one of the two populations were estimated to be 0.484 (15/31) and 0.085, respectively. Those in another population were 0.433 (13/30) and 0.086, respectively. The genetic distance (Nei, 1975) between the two wild quail populations was D = 0.0074. D values of 0.0321 and 0.0189 were estimated between the laboratory quail population previously examined (Kimura et al., 1982) and each of these two wild populations.  相似文献

To provide information on the extent to which domestication has altered the natural levels of genetic variability of the wild quail, electrophoretic variability at 32 loci was estimated in a quail population established by capturing wild quail and keeping them for 15 years in a domestic environment without any artificial selection. The gene frequencies of the domesticated quail population were very similar to those of the commercial and laboratory quail populations and different from those of the wild quail populations previously reported by Kimura et al . Understanding the mechanisms that lead organisms to be separated into distinct species remains a challenge in evolutionary biology. Interspecific hybridization, which results from incomplete reproductive isolation, is a useful tool to investigate such mechanisms. In birds, interspecific hybridization is relatively frequent, despite the fact that closed species exhibit morphological and behavioural differences. Evolution of behaviour is difficult to investigate on a large timescale since it does not ‘fossilize’. Here I propose that calls of hybrid non-songbirds that develop without the influence of learning may help in understanding the gradual process that leads to vocal divergence during speciation. I recorded crows produced by the European quail (Coturnix c. coturnix), the domestic Japanese quail (Coturnix c. japonica) and their hybrids (F1, F2 and backcrosses). Most crowing patterns were intermediate to those of the parental species; some were similar to one or the other parental species, or not present in either parental species. I also observed vocal changes in hybrid crows during the breeding season and from one year to the other. This vocal variability resembles those observed during the ontogeny of the crow in quails. It is likely that similar mechanisms involved in vocal changes during ontogeny might have driven vocal divergence in the species of Palearctic quails. I suggest that hybrid crows might have resembled those produced by intermediary forms of quails during speciation.  相似文献

Coturnix chinensis (blue-breasted quail) has been classically grouped in Galliformes Phasianidae Coturnix, based on morphologic features and biochemical evidence. Since the blue-breasted quail has the smallest body size among the species of Galliformes, in addition to a short generation time and an excellent reproductive performance, it is a possible model fowl for breeding and physiological studies of the Coturnix japonica (Japanese quail) and Gallus gallus domesticus (chicken), which are classified in the same family as blue-breasted quail. However, since its phylogenetic position in the family Phasianidae has not been determined conclusively, the sequence of the entire blue-breasted quail mitochondria (mt) genome was obtained to provide genetic information for phylogenetic analysis in the present study. The blue-breasted quail mtDNA was found to be a circular DNA of 16,687 base pairs (bp) with the same genomic structure as the mtDNAs of Japanese quail and chicken, though it is smaller than Japanese quail and chicken mtDNAs by 10 bp and 88 bp, respectively. The sequence identity of all mitochondrial genes, including those for 12S and 16S ribosomal RNAs, between blue-breasted quail and Japanese quail ranged from 84.5% to 93.5%; between blue-breasted quail and chicken, sequence identity ranged from 78.0% to 89.6%. In order to obtain information on the phylogenetic position of blue-breasted quail in Galliformes Phasianidae, the 2,184 bp sequence comprising NADH dehydrogenase subunit 2 and cytochrome b genes available for eight species in Galliformes [Japanese quail, chicken, Gallus varius (green junglefowl), Bambusicola thoracica (Chinese bamboo partridge), Pavo cristatus (Indian peafowl), Perdix perdix (gray partridge), Phasianus colchicus (ring-neck pheasant), and Tympanchus phasianellus (sharp-tailed grouse)] together with that of Aythya americana (redhead) were examined using a maximum likelihood (ML) method. The ML analyses on the first/second codon positions, the third codon positions, and amino acid sequence consistently demonstrated that blue-breasted quail and Japanese quail are in the same phylogenetic cluster.  相似文献

Background  

Mass spectrometry and proteomic analyses have become powerful tools for the analysis of proteins and peptides. Investigation of proteins contained in the various layers of the avian eggshell has focused entirely on domesticated species. It has been widely assumed that this existing research can inform the study of wild bird species despite the fact that the vast majority of the diversity in avian species (~95%) exists outside the Orders to which domestic and poultry species belong. Museum collections offer a potentially valuable source of material for studying composition of wild avian eggshell matrix proteins. We used museum and fresh eggshells of common quails Coturnix coturnix to compare the protein composition of their organic matrices. Four eggs of domestic chickens were analysed simultaneously as a control for comparison to the fresh and museum quail eggs. The determination of the proteins was carried out using enzymatic cleavage followed by high-performance mass spectrometry.  相似文献
