Twenty-one strains of Renibacterium salmoninarum were degraded by acid methanolysis and the non-hydroxylated fatty acid esters released examined by thin-layer and gas chromatography. The fatty acid profiles were composed almost exclusively of methyl-branched fatty acids with 12-methyltetradecanoic ( anteiso -C15), 13-methyltetradecanoic ( iso -C15) and 14-methylhexadecanoic ( anteiso -C17) as major components. Polar lipids of the test strains were examined by two-dimensional thin-layer chromatography. All of the organisms possessed very characteristic polar lipid patterns consisting of diphosphatidylglycerol, two major and six or seven minor glycolipids, and two unidentified minor phospholipids. In all cases the major menaquinone components consisted of unsaturated menaquinones with nine isoprene units. The lipid data support the integrity of the genus Renibacterium and can be used to separate it from Corynebacterium and from coryneform bacteria which also contain lysine in the wall peptidoglycan.  相似文献

The types and amounts of polar lipids from the hair of monkey (Macacca fascicularis), dog (Canis familiaris), pig (Sus scrofa) and porcupine (Erethizon dorsatum) have been determined by quantitative thin-layer chromatography. The polar lipid content of the hair samples ranged from 0.6 to 1.6 wt%. Lipid compositions included ceramides (57-63% of the polar lipid by weight), glycosphingolipids (7-9%) and cholesteryl sulfate (22-29%). Several minor components (4-7%) remain unidentified. The results suggest that cholesteryl sulfate may be an important determinant of the cohesiveness of hair.  相似文献

Green and white leaves of the barley mutant line `albostrians' were compared for their polar lipid content and fatty acid composition. The mutant plastids of the white leaves have a double-layered envelope, but in contrast with the normal chloroplasts, lack 70 S ribosomes and thylakoids. In the green leaves, the amount of monogalactosyldiacylglycerol (MGDG) consistently exceeds the amount of digalactosyldiacylglycerol (DGDG) and the amount of galactolipids exceeds the amount of phospholipids. In contrast, in white leaves the amount of DGDG exceeds the amount of MGDG and the amount of phospholipids exceeds the amount of galactolipids. In white leaves, the galactolipid composition reflects the plastid envelope composition which is rich in DGDG, whereas in green leaves the galactolipid composition reflects the thylakoid composition which is rich in MGDG. These results demonstrate the likelihood that all the enzymes involved in galactolipid, sulfolipid and fatty acid synthesis are coded by the nuclear genome.  相似文献

The fatty acids and polar lipid compositions of leaves from nine alpine species were almost identical to that of plants growing in habitats with little seasonal variation in temperature. Furthermore each polar lipid had about the same fatty acid composition in all plant species studied. It is suggested that neither the relative proportions of different lipid classes nor the degree of saturation of individual classes are directly implicated in the adaptation of plant tissues to different climates.  相似文献

A characteristic feature of the domain archaea are the lipids forming the hydrophobic core of their cell membrane. These unique lipids are composed of isoprenoid side-chains stereospecifically ether linked to sn-glycerol-1-phosphate. Recently, considerable progress has been made in characterizing the enzymes responsible for the synthesis of archaeal lipids. However, little is known about their evolution. To better understand how this unique biosynthetic apparatus came to be, large-scale database surveys and phylogenetic analyses were performed. All characterized enzymes involved in the biosynthesis of isoprenoid side-chains and the glycerol phosphate backbone along with their assembly in ether lipids were included in these analyses. The sequence data available in public databases was complemented by an in-depth sampling of isoprenoid lipid biosynthesis genes from multiple genera of the archaeal order Halobacteriales, allowing us to look at the evolution of these enzymes on a smaller phylogenetic scale. This investigation of the isoprenoid biosynthesis apparatus of archaea on small and large phylogenetic scales reveals that it evolved through a combination of evolutionary processes, including the co-option of ancestral enzymes, modification of enzymatic specificity, orthologous and non-orthologous gene displacement, integration of components from eukaryotes and bacteria and lateral gene transfer within and between archaeal orders.  相似文献

Abstract In order to relate the benthic lipid composition to possible sources in the water column, the sestonic communities of a monomictic lake were profiled using their saponifiable polar lipid fatty acids, which were identified by capillary gas chromatography-mass spectrometry (GC-MS). The epilimnion, dominated by the dinoflagellate alga Ceratium hirundella , was characterized by C20:5 and C22:6 polyunsaturated fatty acids. The photic anoxic metalimnion supported a radically different community, dominated by photosynthetic sulfur-oxidizing bacteria ( Chromatium and Chloronema spp.) and a Synechococcus -like cyanobacterium, and was characterized by high concentrations of C16 and C18 monounsaturated fatty acids. The fatty acid compositions of the hypolimnetic seston and the sediment were qualitatively similar to that of the metalimnion. Methyl-branched acids, commonly found in eubacteria, increased with depth in the water column. The concentrations of several unusual fatty acids found in Desulfovibrio spp. Desulfobacter spp. and Desulfotomaculum spp. were inversely related to sulfate concentration in the metalimnion. After the water column mixed in the winter, steep gradients of respiratory terminal electron acceptors developed in the surface sediment and were reflected in the polar lipid fatty acid compositions. The results show that fatty acids derived from the membranes of epilimnetic phytoplankton were efficiently metabolized in the oxic portion of the water column. The fatty acids synthesized by prokaryotic microorganisms at and below the oxycline dominated the sediment. The polar lipid fatty acid composition of the sediment showed seasonal changes which can be associated with concentrations of terminal electron acceptors of microbial respiration, and thus with physiologically distinct bacterial groups.  相似文献

Studies were carried out on the lipid composition of a halotolerant Staphylococcus epidermidis isolated in pure culture from a growth medium for extreme halophiles containing 25% NaCl. The four major polar lipid components in this bacterium were found to be: (a) glycerophosphoryl diglucosyl diglyceride (10% by weight) with structure 3(1)-O-(-sn-glycerol-1-phosphoryl-6'-O=(beta-D glucopyranosyl-(1 leads to 6)- O-beta-D-glucopyranosyl)-1(3),2-diacyl-sn-glycerol; (b) diglucosyl diglyceride (15% by weight) with structure 3(1)-O-(beta-D-glucopyranosyl (1 leads to 6)-O-beta-D-glucopyranosyl)-1(3),2-diacyl-sn-glycerol; (c) monoglucosyl diglyceride (3% by weight) with structure 3(1)-O-(beta-D-glucopyranosyl)-1(3),2-diacyl-sn-glycerol, and (d) phosphatidylglycerol (60% by weight) with structure 1,2 diacyl-sn-glycero-3-phosphoryl-1'-sn-glycerol. Phosphatidic acid, cardiolipin, lysophosphatidylglycerol and three unidentified phospholipids were also detected in small amounts. Each lipid component had essentially the same fatty acid composition namely, anteiso-15:0 (60-75%), anteiso-17:0 (18-24%), iso-17:0 (8--10%), and small amounts of palmitic and stearic acids (2-5%). The fatty acids were non-randomly distributed in phosphatidylglycerol, the shorter chain anteiso 15:0 fatty acid being exclusively esterified to the 2-position and the longer chain anteiso- and iso-17:0 fatty acids at the 1-position. The fatty acid composition was not affected by increaseing NaCl content in the medium in the rande 0--15% but the proportion of anteiso-15:0 increased greatly when the salt concentration was increased to 25%. The proportions of ionic polar lipids were modified to give an increased net negative charge per mol ionic lipids when NaCl in the medium was increased from 15 to 25%, but the proportions of neutral glycolipids remained fairly constant.  相似文献

Ubiquinone, known as coenzyme Q, was shown to be the part of the metabolic pathways by Crane et al. in 1957. Its function as a component of the mitochondrial respiratory chain is well established. However, ubiquinone has recently attracted increasing attention with regard to its function, in the reduced form, as an antioxidant. In ubiquinone synthesis the para-hydroxybenzoate ring (which is the derivative of tyrosine or phenylalanine) is condensed with a hydrophobic polyisoprenoid side chain, whose length varies from 6 to 10 isoprene units depending on the organism. para-Hydroxybenzoate (PHB) polyprenyltransferase that catalyzes the condensation of PHB with polyprenyl diphosphate has a broad substrate specificity. Most of the genes encoding (all-E)-prenyltransferases which synthesize polyisoprenoid chains, have been cloned. Their structure is either homo- or heterodimeric. Genes that encode prenyltransferases catalysing the transfer of the isoprenoid chain to para-hydroxybenzoate were also cloned in bacteria and yeast. To form ubiquinone, prenylated PHB undergoes several modifications such as hydroxylations, O-methylations, methylations and decarboxylation. In eukaryotes ubiquinones were found in the inner mitochondrial membrane and in other membranes such as the endoplasmic reticulum, Golgi vesicles, lysosomes and peroxisomes. Still, the subcellular site of their biosynthesis remains unclear. Considering the diversity of functions of ubiquinones, and their multistep biosynthesis, identification of factors regulating their cellular level remains an elusive task.  相似文献

The polar acyl lipid composition was determined for samples of chloroplast thylakoids isolated from Pisum sativum plants grown at light intensities of 50 and 300 E·m-2·s-1 and from Aesculus hippocastanum leaves taken from shade or sun environments. Lighting conditions had no major effect on lipid class composition except for a small increase in the amount of monogalactosyldiacylglycerol relative to other lipids in low compared with high light and shade compared with sun conditions. The thylakoids from low light and shade environments also had, relative to those from high light and sun conditions, a substantial decrease in the level of trans-hexadecenoic acid in phosphatidyglycerol. In parallel with this there were lower lipid to chlorophyll ratios, higher overall fatty acid unsaturation, lower chlorophyll a to b ratios and increased relative levels of light harvesting chlorophyll a/b polypeptides as expected for an increase in the degree of thylakoid appression. 1. The syntheses of ubiquinone-9 and plastoquinone-9 were used as parameters respectively of mitochondrial and proplastid development in the germinating wheat grain. 2. The changes in the amounts of the tocopherols were also studied and the possible biological significance of these changes is discussed. During germination, the dimethyl tocopherols of the resting grain are probably not utilized for the synthesis of α-tocopherol. 3. It was demonstrated that ubiquinone synthesis, and hence probably mitochondrial development, in the aleurone cells during germination, is independent of control by gibberellic acid from the embryo. 4. The influence of light on the syntheses of the isoprenoid quinones in the etiolated wheat shoot was investigated. In particular, illumination did not stimulate the synthesis of either α-tocopherol or α-tocopherolquinone.  相似文献
