Forest floor vegetation is an important component of forest biodiversity, and numerous studies have shown that N input alters the vegetation. In some cases, however, the effects of experimental N addition have been small or absent. Two alternative hypotheses have been suggested: (a) competition from the tree layer confounds the response to N, or (b) N response in areas with high background deposition is limited by N saturation. Neither of these hypotheses has so far been explicitly tested. Here, we compile data on forest floor vegetation from N addition experiments, in which the forest had been clear-cut, along an N deposition gradient ranging from 4 to 16 kg ha?1 year?1 in Sweden. We analyzed the effects of N addition and its interaction with N deposition on common species and thereby tested the second hypothesis in an environment without the confounding effects of the tree layer. The results show that the effects of the experimental N addition are significantly influenced by background N deposition: the N addition effects are smaller in areas with high N deposition than in areas with low N deposition, despite the fact that the highest N deposition in this study can be considered moderate from an international perspective. The results are important when assessing the reliability of results from N addition experiments on forest floor vegetation in areas with moderate to high background N deposition. We conclude that the interacting effects of N addition and N deposition need to be included when assessing long-term N sensitivity of plant communities.  相似文献

A current trend in single-particle electron microscopy is to compute three-dimensional reconstructions with ever-increasing numbers of particles in the data sets. Since manual--or even semi-automated--selection of particles represents a major bottleneck when the data set exceeds several thousand particles, there is growing interest in developing automatic methods for selecting images of individual particles. Except in special cases, however, it has proven difficult to achieve the degree of efficiency and reliability that would make fully automated particle selection a useful tool. The simplest methods such as cross correlation (i.e., matched filtering) do not perform well enough to be used for fully automated particle selection. Geometric properties (area, perimeter-to-area ratio, etc.) and the integrated "mass" of candidate particles are additional factors that could improve automated particle selection if suitable methods of contouring particles could be developed. Another suggestion is that data be always collected as pairs of images, the first taken at low defocus (to capture information at the highest possible resolution) and the second at very high defocus (to improve the visibility of the particle). Finally, it is emphasized that well-annotated, open-access data sets need to be established in order to encourage the further development and validation of methods for automated particle selection.  相似文献

An indirect approach was used to estimate exposure to background levels of atmospheric benzene for a selection of Ontario non‐smoking urban sub‐populations. Activity codes obtained from nationally representative time‐budget surveys were allocated to five general microenvironments (home, work (or school), outdoors, commuting, and other indoors) and further combined with inhalation rates corresponding to specific levels of physical activity in order to develop average activity patterns for six sub‐populations believed to be differently exposed to atmospheric benzene in urban areas (children, teenagers, office workers, outdoor workers, transit workers, and seniors). These activity patterns were then combined with representative concentrations of benzene measured in the selected microenvironments in Ontario in order to evaluate exposure. Two metrics were used for this purpose, integrated exposure and potential average daily dose (intake). Potential lifetime average daily doses were also estimated for three composite sub‐groups representing average office, outdoor, and transit workers. A probabilistic approach using a Monte‐Carlo sampling procedure was used in order to estimate possible ranges of exposures experienced by the various sub‐populations. Results obtained suggested that the highest daily integrated exposure (mean: 131 μg‐hrs/m3) was associated with the average transit worker while comparable levels were estimated for the other sub‐populations investigated (mean: 77–86 μg‐hrs/m3). These levels corresponded to 24‐hours time‐weighted average (TWA)‐equivalent concentrations of 5.5 μg/m3 and 3.5 μg/m3, respectively. Statistical distributions of integrated exposures obtained with the probabilistic approach indicated levels as high as 343 μg‐hrs/m3 (97.5th percentile) in the case of the average transit worker, corresponding to TWA‐equivalents in excess of 15 μg/m3. When levels of physical activities and inhalation rates were integrated in the calculation of exposure, the highest potential average daily dose was found to be associated with the average child (mean: 3.1 μg/kg‐day; 97.5th percentile: 6.0 μg/kg‐day) whereas comparable amounts were estimated for teenager and transit workers (mean: 2.1 μg/kg‐day; 97.5th: 4.1 and 6.9 μg/kg‐day, respectively). Indoor microenvironments (home, office/school, other indoors) were identified as the major contributors to total exposure and intake of benzene (≥50%) although their relative importance varied depending on the exposure metric utilized. Potential lifetime average daily doses estimated for transit workers varied from 2.1 (mean) to 5.4 (97.5th) μg/kg‐day. This was slightly higher than those estimated for the average office and outdoor workers (mean: 1.5–1.7 μg/kg‐day). These projections suggest that average non‐smoking children and teenagers are the most exposed sub‐populations among those investigated to background levels of atmospheric benzene as a result of their daily activities. However, these projections must be interpreted with caution in view of uncertainties associated with some of the assumptions adopted, the limited database used to document benzene levels, and as a result of unaccounted sources of emissions which, under certain circumstances, can significantly modify these conclusions.  相似文献

Normal waking mentation is the outcome of the combined action of both electrophysiological and neurochemical antagonistic and complementary activating and inhibitory influences occurring mainly in the cerebral cortex. The chemical ones are supported principally by acetylcholine, and noradrenaline and serotonin, respectively. During rapid eye movement (REM) sleep, the monoaminergic silence - except dopaminergic ongoing activity - disrupts this equilibrium and seems to be responsible for disturbances of mental activity characteristic of dreaming. This imbalance could cause disconnectivity of cortical areas, failure of latent inhibition and possibly the concomitant prefrontal dorsolateral deactivation. Moreover, the decrease of prefrontal dopaminergic functioning could explain the loss of reflectiveness in this sleep stage. All these phenomena are also encountered in schizophrenia. The psychotic-like mentation of dreaming (hallucinations, delusions, bizarre thought processes) could result from the disinhibition of dopamine influence in the nucleus accumbens by the noradrenergic and serotonergic local silence and/or the lifting of glutamate influence from the prefrontal cortex and hippocampus. We hypothesize that, during REM sleep, the increase of dopamine and the decrease of glutamate release observed in nucleus accumbens reach the threshold values at which psychotic disturbances arise during wakefulness. Whatever the precise mechanism, it seems that the functional state of the prefrontal cortex and nucleus accumbens is the same during dreaming sleep stage and in schizophrenia. The convergent psychological, electrophysiological, tomographic, pharmacological and neurochemical criteria of REM sleep and schizophrenia suggest that this sleep stage could become a good neurobiological model of this psychiatric disease.  相似文献

The interaction of background disease processes with environmental induced diseases has long been an issue of considerable interest and debate with respect to its impact on risk assessment. Whether and to what extent these processes should be considered independent or additive to background has been the principal focus of debate. The concept of hormesis, a biphasic dose response characterized by a low dose stimulation and a high dose inhibition, as framed within the context of post-conditioning, reveal the occurrence of a third type of “background” possibility, that of “subtraction to background”. This novel application of the hormesis concept, which is framed within the biological context of post-conditioning adaptive processes, offers considerable implications for the assessment of aging and environmental risk assessment.  相似文献

The equilibrium constants and the respective standard Gibbs energy changes for hydrolysis of some β-lactam antibiotics have been determined. Native and immobilized penicillin amidase (EC 3.5.1.11) from Escherichia coli has been used as a catalyst. The values of standard Gibbs energy changes corresponding to the pH-independent product of equilibrium concentrations (ΔG0c = ? RT ln Kc) have been calculated. The differences in the structure of the antibiotics nucleus hardly ever affect the value of the pH-independent component of the standard Gibbs energy change (ΔG0c) and value of apparent standard Gibbs energy change at a fixed pH (ΔG0′c). At the same time, the value of ΔG0c is more sensitive to the structure of the acyl moiety of the antibiotic; when ampicillin is used instead of benzylpenicillin, ΔG0c increases by ~6.3 kJ mol?1 (1.5 kcal mol?1). pH-dependences of the apparent standard Gibbs energy changes for hydrolysis of β-lactam antibiotics have been calculated. The pH-dependences of ΔG0′c for hydrolysis of all β-lactam antibiotics have a similar pattern. The thermodynamic pH optimum of the synthesis of these compounds is in the acid pH range (pH < 5.0). The breakage of the β-lactam ring leads to a sharp decrease in the ΔG0′c value and a change in the pattern of the pH-dependence. For example, at pH 5.0 ΔG0′c decreases from 14.4 kJ mol?1 for benzylpenicillin to ?1.45 kJ mol?1 for benzylpenicilloic acid. The reason for these changes is mainly a considerable increase in the pK of the amino group of the nucleus of the antibiotic and, as a consequence, a decrease in the component of standard Gibbs energy change, corresponding to the ionization of the system. The thermodynamic potentials of the enzymatic synthesis of semisynthetic penicillins and cephalosporins on the basis of both free acids and their derivatives (N-acylated amino acids, esters) are discussed. It is shown that with esters of the acids, a high yield of the antibiotic can, in principle, be achieved at higher pH values.  相似文献

BackgroundArsenic (As) causes oxidative stress through generation of reactive oxygen species. 8-Oxo-7,8-dihydro-2′-deoxyguanosine (8-oxodG), a sensitive marker of oxidative DNA damage, has been associated with As exposure in some studies, but not in others, possibly due to population-specific genetic factors.ObjectivesTo evaluate the association between As and 8-oxodG in urine in a population with a low urinary monomethylated As (%MMA) and high dimethylated As (%DMA), as well as the genetic impact on (a) 8-oxodG concentrations and (b) the association between As and 8-oxodG.Materials and methodsWomen (N = 108) in the Argentinean Andes were interviewed and urine was analyzed for arsenic metabolites (ICPMS) and 8-oxodG (LC–MS/MS). Twenty-seven polymorphisms in genes related to oxidative stress and one in As(+III)methyltransferase (AS3MT) were studied.ResultsMedian concentration of 8-oxodG was 4.7 nmol/L (adjusted for specific weight; range 1.6–13, corresponding to 1.7 μg/g creatinine, range 0.57–4.8) and of total urinary As metabolites (U-As) 290 μg/L (range 94–720; 380 μg/g creatinine, range 140–1100). Concentrations of 8-oxodG were positively associated with %MMA (strongest association, p = 0.013), and weakly associated with U-As (positively) and %DMA (negatively). These associations were strengthened when taking ethnicity into account, possibly reflecting genetic differences in As metabolism and genes regulating oxidative stress and DNA maintenance. A genetic influence on 8-oxodG concentrations was seen for polymorphisms in apurinic/apyrimidinic endonuclease 1 (APEX1), DNA-methyltransferases 1 and 3b (DNMT1, DNMT3B), thioredoxin reductase 1 (TXNRD1) and 2 (TXNRD2) and glutaredoxin (GLRX).ConclusionDespite high As exposure, the concentrations of 8-oxodG in this population were low compared with other As-exposed populations studied. The strongest association was found for %MMA, stressing that some inconsistencies between As and 8-oxodG partly depend on population variations in As metabolism. We found evidence of genetic impact on 8-oxodG concentrations.  相似文献

Two-color microarrays are a powerful tool for genomic analysis, but have noise components that make inferences regarding gene expression inefficient and potentially misleading. Background fluorescence, whether attributable to nonspecific binding or other sources, is an important component of noise. The decision to subtract fluorescence surrounding spots of hybridization from spot fluorescence has been controversial, with no clear criteria for determining circumstances that may favor, or disfavor, background subtraction. While it is generally accepted that subtracting background reduces bias but increases variance in the estimates of the ratios of interest, no formal analysis of the bias-variance trade off of background subtraction has been undertaken. In this paper, we use simulation to systematically examine the bias-variance trade off under a variety of possible experimental conditions. Our simulation is based on data obtained from 2 self versus self microarray experiments and is free of distributional assumptions. Our results identify factors that are important for determining whether to background subtract, including the correlation of foreground to background intensity ratios. Using these results, we develop recommendations for diagnostic visualizations that can help decisions about background subtraction.  相似文献

Performance ratings were obtained by the clinical supervisors of four graduated classes of McMaster University medical students during internship. The supervisors detected no difference in performance between the graduates who met the "traditional" admissions criteria (both an undergraduate grade point average of 3.1 or greater on a 4-point scale and previous training in biology, general and organic chemistry, and physics) and those who lacked one or both of these prerequisites. These data suggest that medical schools can expand their admissions criteria without fearing that their graduates will perform less well as interns because of a lack of traditional academic preparation for medical school.  相似文献

During the past two decades, molecular biologists and geneticists have deconstructed intracellular signaling pathways in individual cells, revealing a great deal of crosstalk among key signaling pathways in the animal kingdom. Fewer examples have been reported in plants, which appear to integrate multiple signals on the promoters of target genes or to use gene family members to convey signal-specific output. For both plants and animals, the question now is whether the "crosstalk" is biologically relevant or simply noise in the experimental system. To minimize such noise, we suggest studying signaling pathways in the context of intact organisms with minimal perturbation from the experimenter.  相似文献

In the present paper we correlate clinical data, as well as histopathological, immunohistochemical and molecular biology methods, with the occurrence of both forms of inflammatory bowel disease (IBD) i.e. ulcerative colitis and Crohn's disease. We found that patients with a history of Epstein-Barr virus (EBV) or cytomegalovirus (CMV) infections, as well as steroid treatment, had increased susceptibility to the development of IBD. The diagnosis of IBD was confirmed by histopathology. Previous infections by EBV and CMV, as well as M. tuberculosis, were proved by PCR-based techniques and in situ hybridization. We found PCR-proved latent viral infections in 30-50% of the IBD patients we studied. However, we were unable to prove the presence of viral antigens by immunohistochemistry for EBV or CMV. We found positive correlations between the presence of anti-CMV IgG, as well as PCR-positive results for M. tuberculosis with an ulcerative colitis diagnosis. Additionally, up to 80% of IBD patients used steroids, which was found to be correlated with a diagnosis of Crohn's disease. Our data may support the theory that IBD could be related to previous viral infections and the use of steroids.  相似文献

This study examined the extent to which socioemotional well-being mediated the relationship between language status and achievement, while exploring variability in this relationship based on informant (student versus teacher reports of socioemotional problems) and native language background (Spanish-speaking English language learners [ELLs] versus ELLs from Asian-language backgrounds). Participants included 9,046 fifth-grade students from the Early Childhood Longitudinal Study-Kindergarten Cohort of 1998. Results from structural equation modeling showed that Spanish-speaking ELLs reported more social-emotional problems as compared to English-monolingual (EM) and Asian-language ELL classmates in third grade, which partially explained their lower achievement levels in fifth grade. The model accounted for approximately 34% of the variance in students’ academic achievement in fifth grade (R2 = .343, p < .001). When comparing ELL and EM students, results differed when using teachers’ versus students’ reports of socioemotional well-being. For both Spanish-speaking and Asian-language ELLs, teachers perceived fewer social and emotional difficulties than the students themselves reported in comparison to their EM classmates.  相似文献

This study was conducted with a purpose to examine whether the T102C polymorphism of the serotonin receptor 2A (HTR2A) gene moderates the association between parental education and children's school achievement across nine compulsory school years. The study was carried out in a population‐based sample of Finnish students (aged 9, 12 and 15 years, n = 982). It was found that the HTR2A gene was not related to the school achievement at any school level, but moderated the association between maternal education and the children's grade point averages. The T/T genotype carriers benefited most from high‐maternal education, and suffered from a low one more than the carriers of the other variants of the HTR2A gene. The present finding may at least partly answer the important question why academic outcomes of environmental interventions vary even at the same intelligence levels of the students.  相似文献

Abstract

This paper reports on an examination of whether the effects of farm background on socioeconomic differentials in fertility are diminished among nonfarm couples. The data are for a sample of white ever‐married women belonging to the 1901–1910 birth cohorts. The research provides another test of the two‐generation‐urbanite hypothesis first advanced by Goldberg. Unlike a number of other studies, the findings do not support the hypothesis. Reasons for the lack of support are discussed, and methodological weaknesses of previous studies are identified.  相似文献

A novel model for the regulation of cell excitability has recently been proposed. It originates from the observation that the background K(+) channel K2P1 (TWIK1) may be silenced by sumoylation in Xenopus oocytes and that inactivation of the putative sumoylation site (mutation K274E) gives rise to robust current expression in transfected COS-7 cells. Here, we show that only the mutation K274E, and not K274R, is associated with an increase of K2P1 current density, suggesting a charge effect of K274E. Furthermore, we failed to observe any band shift by western blot analysis that would confirm an eventual sumoylation of K2P1 in COS-7 cells and oocytes.  相似文献

Hundreds of studies have assessed variation in the degree to which people experience disgust toward substances associated with pathogens, but little is known about the mechanistic sources of this variation. The current investigation uses olfactory perception and threshold methods to test whether it is apparent at the cue-detection level, at the cue-interpretation level, or both. It further tests whether relations between disgust sensitivity and olfactory perception are specific to odors associated with pathogens. Two studies (N's = 119 and 160) of individuals sampled from a Dutch university each revealed that pathogen disgust sensitivity relates to valence perceptions of odors found in pathogen sources, but not to valence perceptions of odors not associated with pathogens, nor to intensity perceptions of odors of either type. Study 2, which also assessed olfactory thresholds via a three-alternative forced-choice staircase method, did not reveal a relation between pathogen disgust sensitivity and the ability to detect an odor associated with pathogens, nor an odor not associated with pathogens. In total, results are consistent with the idea that pathogen disgust sensitivity relates to how olfactory pathogen cues are interpreted after detection, but not necessarily to the ability to detect such cues.  相似文献

Abstract

Soybean [Glycine max (L.) Merr.] oil typically contains 2–4 % stearic acid. Oil with at least 20 % stearic acid is desirable because of its improved baking properties and health profile. This study identifies two new sources of high stearic acid and evaluates the interaction of high stearic and oleic acid alleles. TCHM08-1087 and TCHM08-755, high stearic acid ‘Holladay’ mutants, were crossed to FAM94-41-3, a line containing a point mutation in a seed-specific isoform of a Δ9-stearoyl-acyl carrier protein-desaturase (SACPD-C). F2-derived lines were evaluated for fatty acid content in four field environments. Sequencing of SACPDs in TCHM08-1087 and TCHM08-755 revealed distinct deletions of at least one megabase encompassing SACPD-C in both lines. After genotyping, the additive effect for stearic acid was estimated at +1.8 % for the SACPD-C point mutation and +4.1 % for the SACPD-C deletions. Average stearic acid in lines homozygous for the deletions was 12.2 %. A FAM94-41-3-derived line and TCHM08-1087-11, a selection from TCHM08-1087, were crossed to S09-2902-145, a line containing missense mutations in two fatty acid desaturases (FAD2-1A and FAD2-1B). F1 plants were grown in a greenhouse and individual F2 seed were genotyped and phenotyped. No interaction was observed between either FAD2-1A or FAD2-1B and any of the SACPD-C mutant alleles. Seed homozygous mutant for SACPD-C/FAD2-1A/FAD2-1B contained 12.7 % stearic acid and 65.5 % oleic acid while seed homozygous for the SACPD-C deletion and mutant for FAD2-1A and FAD2-1B averaged 10.4 % stearic acid and 75.9 % oleic acid.  相似文献

The interplay of plant resistance mechanisms and bacterial pathogenicity is very complex. This applies also to the interaction that takes place between the pathogenPseudomonas syringae pv.lachrymans (Smith et Bryan) and the cucumber (Cucumis sativus L.) as its host plant. Research onP. syringae pv.lachrymans has led to the discovery of specific factors produced during pathogenesis, i.e. toxins or enzymes. Similarly, studies on cucumber have identified the specific types of plant resistance expressed, namely Systemic Acquired Resistance (SAR) or Induced Systemic Resistance (ISR). This paper presents a summary of the current state of knowledge about this particular host-pathogen interaction, with reference to general information about interactions ofP. syringae pathovars with host plants.  相似文献
