Glutathione S-transferase (GST) is a phase II enzyme that functions as a detoxicant by catalyzing the conjugation of reduced glutathione with a variety of xenobiotics via cysteine thiol. Molecular genetic approaches using gene biomarkers show substantial relevance as sensitive biomarkers for the indication of pollution levels. In order to use GSTs as molecular biomarkers for marine pollution monitoring, we cloned and sequenced the full-length cDNA of seven GST genes from the marine polychaete Perinereis nuntia. The deduced amino acid sequence of Pn-GSTs showed a high similarity to those of other species that clustered into the same clades in a phylogenetic analysis. In addition, to evaluate Pn-GSTs as useful biomarkers on effects after cadmium (Cd) exposure, we exposed sublethal concentrations of Cd (5, 50, and 500 μg/L) to P. nuntia, and they showed relatively different but significantly increases, depending on exposure time and Cd concentrations. Particularly, Pn-GST-omega and Pn-GST-sigma genes were highly sensitive with a clear dose-dependent manner on mRNA expression. The total GST activities also have significantly increased levels at higher concentrations of Cd exposure. These results indicate that Pn-GSTs play important roles in Cd-induced oxidative stress in terms of the physiological changes relating to metabolism and cell protection, and those genes would have great potential as molecular biomarkers to monitor marine environmental health.  相似文献

The exploitation of non-invasive samples has been widely used in genetic monitoring of terrestrial species. In aquatic ecosystems, non-invasive samples such as feces, shed hair or skin, are less accessible. However, the use of environmental DNA (eDNA) has recently been shown to be an effective tool for genetic monitoring of species presence in freshwater ecosystems. Detecting species in the marine environment using eDNA potentially offers a greater challenge due to the greater dilution, amount of mixing and salinity compared with most freshwater ecosystems. To determine the potential use of eDNA for genetic monitoring we used specific primers that amplify short mitochondrial DNA sequences to detect the presence of a marine mammal, the harbor porpoise, Phocoena phocoena, in a controlled environment and in natural marine locations. The reliability of the genetic detections was investigated by comparing with detections of harbor porpoise echolocation clicks by static acoustic monitoring devices. While we were able to consistently genetically detect the target species under controlled conditions, the results from natural locations were less consistent and detection by eDNA was less successful than acoustic detections. However, at one site we detected long-finned pilot whale, Globicephala melas, a species rarely sighted in the Baltic. Therefore, with optimization aimed towards processing larger volumes of seawater this method has the potential to compliment current visual and acoustic methods of species detection of marine mammals.  相似文献

Sponges are a dominant component of benthic assemblages in hard substratum environments across the world, but despite the importance of sponges, they are generally poorly represented in most marine monitoring programmes. There is considerable need to develop effective monitoring tools to monitor changes in sponge assemblages. Morphological monitoring has been proposed as a suitable method to monitor sponges and while morphological monitoring has already taken place at Skomer Marine Reserve (MNR), Wales, here we investigate whether species level and morphological level data sets are correlated with respect to temporal variation. Furthermore, we examine the environmental factors that correlate with the patterns of temporal variability. Both species and morphological data sets revealed significant seasonal changes and spatial variation in sponge assemblages; these data sets were highly correlated and explained by a number of environmental factors. We conclude that morphological monitoring of sponge assemblages may represents a cost-effective method for assessing temporal and spatial variation in sponges, where full species level monitoring is not possible, as patterns identified from morphological data were a suitable surrogate of species-level data.  相似文献

Benthic invertebrates are good indicators of aquatic ecosystem health. Yet, environmental monitoring and assessment of community changes in relation to both natural and human sources of disturbance involves considerable efforts for sample processing and time-consuming identifications of organisms, which make challenging large-scale and continuous monitoring programs required under the current regulatory frameworks at European scale. The use of higher taxa (e.g. families) as surrogates for species is a mainstream approach to reduce cost and time associated to fine taxonomic resolution in environmental studies, especially concerning macro-invertebrate communities. However, this approach of ‘taxonomic sufficiency’ simply relies on the static grouping of organisms in taxa belonging to a single higher taxonomic level irrespective of their ecological relevance or difficulties in their taxonomic identification, leading to unnecessary losses of taxonomic detail or ecological information. A new approach, namely the Best Practicable Aggregation of Species (BestAgg), has been recently developed as an alternative procedure for species surrogacy. BestAgg is based on aggregating species in the minimum number of surrogates sufficient to depict species-level community patterns consistently, while capitalizing on ecological information. Although the approach has been successfully applied to marine and freshwater invertebrate assemblages, its effectiveness in transitional water systems, where the complex and highly variable environmental conditions may affect the performance of surrogacy methods, still remain untested. Here, we applied the BestAgg approach to quantifying spatio-temporal patterns of variability of macro-invertebrate assemblages from Mediterranean coastal lagoons (Northern Adriatic Sea). Surrogates were defined using species-level data from a representative lagoon system, which served as pilot study. Then, they were used to analyze macro-invertebrate assemblages in two independent lagoons in the same geographic area. Results showed that BestAgg surrogates were effective in depicting multivariate patterns of macro-invertebrate assemblages as at species level over and beyond potential variations among the investigated lagoons. The use of families, following a more classic approach based on taxonomic sufficiency, also provided results comparable to those obtained using species. However, with respect to families, BestAgg surrogates allowed an estimated timesaving of 40% higher while still retaining an equivalent amount of information on species-level patterns. More importantly, BestAgg allowed exploiting different criteria of species aggregation, leading to a set of surrogates more aligned with ecological/functional characteristics of organisms, suggesting that BestAgg approach may provide a fresh perspective for optimizing trade-offs between pragmatism and the need for relevant ecological information in environmental assessment and monitoring.  相似文献

本文系统地总结了近10年中国研究人员在遗传、物种、生态系统3个层次上对海洋生物多样性研究的重要进展, 并使用VOSviewer软件对近10年中国近海生物多样性的研究成果进行文献计量分析。近年来, 中国研究人员借助新的研究方法和手段, 比如分子生物学和流式细胞术等, 可以在物种多样性水平进行更准确和快速的分类鉴定, 借此在中国近海发现了较多新的物种; 通过多学科交叉融合, 更多的是在生态系统水平探讨海洋生物多样性, 也为今后海洋生态系统的修复提供了科学依据。目前中国的海洋生物多样性研究紧跟国际科技前沿和步伐, 在深海、海山和极端环境生物类群等新兴领域有了长足发展, 新物种的发现不断更新了原有认识, 对典型海洋生态系统的监测和部分入侵物种的整治有了长足的进步。中国近海生物多样性高, 监测数据全, 通过整合空间数据资料和时间序列变化, 进行更广更深的宏观生态模式分析研究十分必要。通过探究生物多样性的多重胁迫因子及其交互作用, 可为优化海洋生物多样性的保护和管理提供帮助。  相似文献

There is a multitude of ecosystem service classifications available within the literature, each with its own advantages and drawbacks. Elements of them have been used to tailor a generic ecosystem service classification for the marine environment and then for a case study site within the North Sea: the Dogger Bank. Indicators for each of the ecosystem services, deemed relevant to the case study site, were identified. Each indicator was then assessed against a set of agreed criteria to ensure its relevance and applicability to environmental management. This paper identifies the need to distinguish between indicators of ecosystem services that are entirely ecological in nature (and largely reveal the potential of an ecosystem to provide ecosystem services), indicators for the ecological processes contributing to the delivery of these services, and indicators of benefits that reveal the realized human use or enjoyment of an ecosystem service. It highlights some of the difficulties faced in selecting meaningful indicators, such as problems of specificity, spatial disconnect and the considerable uncertainty about marine species, habitats and the processes, functions and services they contribute to.  相似文献

Mangroves are subject to the effects of tides and fluctuations in environmental conditions, which may reach extreme conditions. These ecosystems are severely threatened by human activities despite their ecological importance. Although mangroves are characterized by a highly specialized but low plant diversity in comparison to most other tropical ecosystems, they support a diverse microbial community. Adapted microorganisms in soil, water, and on plant surfaces perform fundamental roles in nutrient cycling, especially nitrogen and phosphorus. Cyanobacteria contribute to carbon and nitrogen fixation and their cells act as phosphorus storages in ecosystems with extreme or oligotrophic environmental conditions such as those found in mangroves. As the high plant productivity in mangroves is only possible due to interactions with microorganisms, cyanobacteria may contribute to these ecosystems by providing fixed nitrogen, carbon, and herbivory-defense molecules, xenobiotic biosorption and bioremediation, and secreting plant growth-promoting substances. In addition to water, cyanobacterial colonies have been detected on sediments, rocks, decaying wood, underground and aerial roots, trunks, and leaves. Some mangrove cyanobacteria were also found in association to algae or seagrasses. Few studies on mangrove cyanobacteria are available, but together they have reported a substantial number of species in these ecosystems. However, the cyanobacterial diversity in this biome has been traditionally underestimated. Though mangrove communities generally host cyanobacterial taxa commonly found in marine environments, unique microhabitats found in mangroves potentially harbor several undescribed cyanobacterial taxa. The relevance of cyanobacteria for mangrove conservation is highlighted in their use for the recovery of degraded mangroves as biostimulants or in bioremediation.  相似文献

Predicting the host range for herbivores has been a major aim of research into plant-herbivore interactions and an important model system for understanding the evolution of feeding specialization. Among many terrestrial insects, host range is strongly affected by herbivore phylogeny and long historical associations between particular herbivore and plant taxa. For small herbivores in marine environments, it is known that the evolution of host use is sculpted by several ecological factors (e.g., food quality, value as a refuge from predators, and abiotic forces), but the potential for phylogenetic constraints on host use remains largely unexplored. Here, we analyze reports of host use of herbivorous amphipods from the family Ampithoidae (102 amphipod species from 12 genera) to test the hypotheses that host breadth and composition vary among herbivore lineages, and to quantify the extent to which nonpolar secondary metabolites mediate these patterns. The family as a whole, and most individual species, are found on a wide variety of macroalgae and seagrasses. Despite this polyphagous host use, amphipod genera consistently differed in host range and composition. As an example, the genus Peramphithoe rarely use available macrophytes in the order Dictyotales (e.g., Dictyota) and as a consequence, display a more restricted host range than do other genera (e.g., Ampithoe, Cymadusa, or Exampithoe). The strong phylogenetic effect on host use was independent of the uneven distribution of host taxa among geographic regions. Algae that produced nonpolar secondary metabolites were colonized by higher numbers of amphipod species relative to chemically poor genera, consistent with the notion that secondary metabolites do not provide algae an escape from amphipod herbivory. In contrast to patterns described for some groups of phytophagous insects, marine amphipods that use chemically rich algae tended to have broader, not narrower, host ranges. This result suggests that an evolutionary advantage to metabolite tolerance in marine amphipods may be that it increases the availability of appropriate algal hosts (i.e., enlarges the resource base).  相似文献

Logging can significantly change the structure of rainforest communities. To better understand how logging drives this change, butterflies and environmental variables were assessed within both unlogged and logged forest in Indonesian Borneo. In the whole dataset, we found local environmental variables and geographic distance combined captured 53.1% of the variation in butterfly community composition; 29.6% was associated with measured local environmental variables, 13.6% with geographic distance between sites, and 9.9% with covariation between geographic distance and environmental variables. The primary axis of variation in butterfly community composition represented a disturbance gradient from unlogged to logged forest. Subsequent axes represented gradients influenced by variables such as canopy cover and total tree density. There were significant associations between environmental variables and geographic range and larval host plant use of species. Specifically, butterflies using trees as larval host plants and those with distributions limited to Borneo were more likely to be present in unlogged forest. By contrast, species that tended to be more abundant in logged forest were those with widespread distributions and those using lianas and grasses as larval host plants. The results of this study highlight the importance of environmental variables and disturbance, e.g., selective logging, in structuring rainforest community diversity. Moreover, they confirm how species traits, such as larval food use and geographic distributions can determine patterns of species abundance following environmental change.  相似文献

The marine environment is becoming increasingly contaminated by environmental pollutants with the potential to damage DNA, with marine sediments acting as a sink for many of these contaminants. Understanding genotoxic responses in sediment-dwelling marine organisms, such as polychaetes, is therefore of increasing importance. This study is an exploration of species-specific and cell-specific differences in cell sensitivities to DNA-damaging agents in polychaete worms, aimed at increasing fundamental knowledge of their responses to genotoxic damage. The sensitivities of coelomocytes from three polychaetes species of high ecological relevance, i.e. the lugworm Arenicola marina, the harbour ragworm Nereis diversicolor and the king ragworm Nereis virens to genotoxic damage are compared, and differences in sensitivities of their different coelomic cell types determined by use of the comet assay. A. marina was found to be the most sensitive to genotoxic damage induced by the direct-acting mutagen methyl methanesulfonate (MMS), and showed dose-dependent responses to MMS and the polycyclic aromatic hydrocarbon benzo(a)pyrene. Significant differences in sensitivity were also measured for the different types of coelomocyte. Eleocytes were more sensitive to induction of DNA damage than amoebocytes in both N. virens and N. diversicolor. Spermatozoa from A. marina showed significant DNA damage following in vitro exposure to MMS, but were less sensitive to DNA damage than coelomocytes. This investigation has clearly demonstrated that different cell types within the same species and different species within the polychaetes show significantly different responses to genotoxic insult. These findings are discussed in terms of the relationship between cell function and sensitivity and their implications for the use of polychaetes in environmental genotoxicity studies.  相似文献

An investigation of a parasite species that is broadly host- and habitat-specific and exhibits alternative transmission strategies was undertaken to examine intraspecific variability and if it can be attributed to cryptic speciation or environmentally induced plasticity. Specimens of an acanthocephalan parasite, Leptorhynchoides thecatus, collected throughout North America were analysed phylogenetically using sequences of the cytochrome oxidase I gene and the internal transcribed spacer region. Variation in host use, habitat use, and transmission were examined in a phylogenetic context to determine if they were more likely phylogenetically based or due to environmental influences. Results indicated that most of the variation detected can be explained by the presence of cryptic species. The majority of these species have narrow host and microhabitat specificities although one species, which also may comprise a complex of species, exhibits broad host and habitat specificity. Alternate transmission pathways only occurred in two of the cryptic species and correlate with host use patterns. Taxa that mature in piscivorous piscine hosts use a paratenic fish host to bridge the trophic gap between their amphipod intermediate host and piscivorous definitive host. One potential example of environmentally induced variation was identified in three populations of these parasites, which differ on their abilities to infect different host species.  相似文献

Cross-taxon congruence has been suggested as an efficient tool in conservation planning and biodiversity monitoring. It corresponds to the degree to which patterns in assemblage structure in a set of sites are similar among different taxonomic groups. If different groups ordinate the sampling sites similarly, they are concordant and this information is frequently used as an indicative that one group could be used as a surrogate for the other. Using spatiotemporal benthic data from a biodiverse coastal ecosystem (the Araçá Bay in southeast Brazil), we first assessed if macro- and meiofaunal assemblages inhabiting marine soft-sediments show concordant patterns. Then, we used multi- and univariate analysis to examine the relationship of both groups with the environment. Macro- and meiofaunal assemblages exhibited strong congruent patterns in all periods analyzed. Moreover, both groups responded to similar environmental features and their ordination patterns matched those generated by the environmental data. These results show that the patterns of concordance are probably mediated by similar responses to environmental gradients. Overall, our results suggest the potential for use of the surrogacy approach in conservation planning and monitoring purposes of marine benthic biodiversity, and that macro- and meiofauna can be used as a surrogate for each other.  相似文献

Trematode larvae must generally invade a molluscan intermediate host, usually a gastropod, before they can reach reproductive maturity in another definitive host. The research literature to date has focused almost exclusively on the documented specificity between particular trematode species and particular molluscan hosts; little attention has been paid to gastropod species that do not appear to serve as hosts. We sampled Rhode Island and Massachusetts populations of the marine gastropod Crepidula fornicata to determine whether this widespread species serves as a first intermediate host for trematodes. We also sampled from the same habitat populations of Littorina littorea and Ilyanassa obsoleta, gastropods known to serve as first intermediate hosts for several trematode species. All individuals were examined by dissection for the presence of sporocysts, rediae, or developing cercariae. Although 4-28% of L. littorea (N=112) and I. obsoleta (N=84) were infected by larvae of at least one trematode species, no individuals of C. fornicata sampled from the same locations were so infected (N=136). A survey of the Biological Abstracts computer database indicates that snails in only about 10% of marine gastropod families are known to serve as first intermediate hosts for trematodes. We suggest that more attention be paid to marine gastropods that appear not to be infected by trematode miracidia. Such species may productively serve as new models for understanding trematode host specificity and gastropod resistance to infection.  相似文献

Baited Underwater Video (BUV) systems have become increasingly popular for assessing marine biodiversity. These systems provide video footage from which biologists can identify the individual fish species present. Here we explore the relevance of spatial dependence and marine park boundaries while estimating the distribution and habitat associations of the commercially and recreationally important snapper species Chrysophrys auratus in Moreton Bay Marine Park during a period when new Marine National Parks zoned as no-take or “green” areas (i.e., areas with no legal fishing) were introduced. BUV studies typically enforce a minimum distance among BUV sites, and then assume that observations from different sites are independent conditional on the measured covariates. In this study, we additionally incorporated the spatial dependence among BUV sites into the modelling framework. This modelling approach allowed us to test whether or not the incorporation of highly correlated environmental covariates or the geographic placement of BUV sites produced spatial dependence, which if unaccounted for could lead to model bias. We fitted Bayesian logistic models with and without spatial random effects to determine if the Marine National Park boundaries and available environmental covariates had an effect on snapper presence and habitat preference. Adding the spatial dependence component had little effect on the resulting model parameter estimates that emphasized positive association for particular coastal habitat types by snapper. Strong positive relationships between the presence of snapper and rock habitat, particularly rocky substrate composed of indurated freshwater sediments known as coffee rock, and kelp habitat reinforce the consideration of habitat availability in marine reserve design and the design of any associated monitoring programs.  相似文献

Evolutionary responses of herbivores to their host plants depend not only on selection from plants, but also on the genetic basis of traits relating to host use. The genetic basis of such traits has been investigated extensively among terrestrial insect herbivores, but has received almost no attention among marine herbivores. We tested whether performance traits in the herbivorous marine amphipod Peramphithoe parmerong display heritable variation and, for the first time for a marine herbivore, whether selection has resulted in local adaptation to host plants on two spatial scales. Peramphithoe parmerong displayed heritable genetic variation for survival on two host macroalgae, the high-quality Sargassum linearifolium and the poor-quality Padina crassa, and for growth on S. linearifolium. Differences in performance on different hosts thus have the potential to select for differential use of hosts by this amphipod. Despite this potential, there was no evidence among field populations of local adaptation to host algae on either scale tested: between hosts within a site or among sites differing in algal species composition. Within a site, amphipods were not more likely to prefer or perform better on the host on which they were collected. Similarly, amphipods collected from sites in which P. crassa was present were not more likely to perform well on this host than amphipods collected from sites where this alga was not found. Ecological factors that may explain the persistence of P. parmerong on P. crassa and the possibility of phylogenetic constraints on host use by P. parmerong are discussed.  相似文献

Heavy-metal concentrations were determined in tissues of different species of benthic invertebrates collected in the Casey region (Australian Antarctic Territory) where an old waste-disposal tip site is a source of contamination. The species studied included the bivalve Laternula elliptica, starfish Notasterias armata, heart urchins Abatus nimrodi and A. ingens and gammaridean amphipod Paramoera walkeri. The specimens were collected at both reference and contaminated locations where lead was the priority element and copper was the next most important in terms of increased concentrations. The strong association between a gradient of contamination and concentrations in all species tested indicates that they are reflecting well the environmental changes, and that they appear as appropriate biological indicators of heavy-metal contamination. Aspects of the biology of species with different functional roles in the marine ecosystem are discussed in relation to their suitability for wider use in Antarctic monitoring programmes. For example, in terms of heavy-metal bioaccumulation, the bivalve appears as the most sensitive species to detect contamination; the starfish provides information on the transfer of metals through the food web while the heart urchin and gammarid give indications of the spatial and temporal patterns of the environmental contamination. The information gathered about processes of contaminant uptake and partitioning among different tissues and species could be used in later studies to investigate the behaviour and the source of contaminants.  相似文献

Ecological risk assessment (EcoRA) provides both a process and a framework to evaluate the potential for adverse ecological effects occurring as a result of exposure to contaminants or other stressors. EcoRA begins with problem formulation/hazard identification, progresses to effects and exposure assessment, and culminates with risk characterization (an estimate of the incidence and severity of any adverse effects likely to occur). Key components of EcoRA include determining: stressors/contaminants of concern; sensitive, exposed biota; and, appropriate tests and organisms for evaluating effects. Aquatic oligochaetes are not generally used directly in EcoRA because of three major perceptions. First, EcoRA personnel are generally not familiar with or comfortable using this group of organisms. Second, there is believed to be a paucity of widely accepted toxicity tests with these organisms. Third, their taxonomy is considered difficult and uncertain. In fact, aquatic oligochaetes potentially have great utility and relevance to EcoRAs because of factors including: their importance in the aquatic food chain (e.g. prey to fauna including fish and waterfowl; as a vector for contaminant movement through the food chain from bacteria); many species are widely distributed and well studied; representatives include fresh, estuarine and marine species; as a group, they range from sensitive to insensitive over a wide range of environmental insults; they have a long history of use in pollution monitoring and assessment; and, relevant toxicity and biaccumulation tests exist. Toxicity testing under defined conditions is appropriate for problem formulation while more realistic testing for effects assessment (e.g. microcosms) is logistically easier with this group of organisms than with others due to their relatively small size. The importance of aquatic oligochaetes for EcoRA, in particular of sediments, is particularly compelling.  相似文献
