DAPI and Feulgen stains were used as specific DNA markers for studying the mitosis process in Naegleria fowleri. Both DAPI and Feulgen stains reacted with DNA in the nuclei of the amoebae. Representative figures of N. fowleri mitotic nuclei with a defined arrangement according to the phase of the cell cycle were observed. A notable characteristic is that the nucleolus is present throughout the stages of mitosis. During metaphase, several deeply stained DNA condensations following an elongated pattern were observed, corresponding almost certainly to tightly grouped chromosomes. Ultrastructural observations demonstrated that the nucleus divides by cryptomitosis, a process in which the nuclear membrane does not disappear during the mitosis. Centrioles were not found, and a spindle of microtubules was observed running the length of the nucleus from pole to pole however, they did not come to a focal point. 相似文献

A 4 1/2 year old female was treated for embryonal rhabdomyosarcoma of the left orbit in 1975 with radiation (59.5 Gy in 5 weeks), followed by chemotherapy. An electroretinogram (ERG) in March, 1988 revealed cone responses 3% of normal and no rod responses in the left eye, and normal responses in the right eye. The eye was enucleated in April 1988. In the fovea no choroidocapillaris was seen at the intact Bruch's membrane, and the pigment epithelium was preserved only in small patches. No photoreceptor cells were seen in the areas devoid of pigment epithelial cells. The parafoveal and peripheral (30 degrees eccentricity) retina was better preserved. The thickness of the layer of rods and cones and of Henle's fiber layer was reduced. Very few outer segments were present. Macrophages had invaded the retinal tissue in moderate numbers. The retinal vessels were ensheathed by several layers of collagen fibrils. The spatial densities of pigment epithelial, cone, rod, and bipolar cells had been reduced. The optic nerve contained a total number of 1,022,000 nerve fibers. 相似文献

Ultrastructural examination of the IIN1b nerve to the dorsal longitudinal flight muscle of Manduca sexta L. verified the presence of neurosecretory processes. Subspherical and irregular vesicles were found where the nerve enters the muscle, while spherical vesicles were found in the proximal region only. A dorsal unpaired median (DUM) cell, the median nervous system, and two or more peripheral cells are the sources of these neurosecretory inclusions. Light the electron microscopy CoCl2 backfills of the transverse nerve produced intensification of a peripheral neuron (#1) and processes in nerves IIN1a and IIN1b. Similar backfills of nerve IIN1b produced intensification of a DUM cell, a second peripheral neuron (#2), and processes in the transverse nerve and nerve IIN1a. Neuron #1 contained large spherical electron-dense vesicles while neuron #2 contained smaller subspherical vesicles. These cells were situated upon the link and/or transverse nerves. Based on these results, we suspect central and peripheral neurosecretory processes reach nerve IIN1b as follows: the link nerve projects prothoracic median nervous system and neuron #2 processes, nerve IIN1a projects neuron #1 processes, and nerve IIN1 projects mesothoracic DUM cell processes, although this latter pathway was less clear. 相似文献

The anatomy of electroplaques of Narcine brasiliensis has been studied by scanning electron microscopy. The ventral faces of the plaques are innervated with an abundance of branching nerves in close contact with the underlying (postsynaptic) membrane. Large numbers of troughs and evaginations of the postsynaptic membrane serve to increase its surface area. Fibroblasts are closely associated with the dorsal faces which appear covered with a matrix of fibrous material. Well-washed dorsal surfaces of single electroplaques are free of this matrix. The dorsal surface area is considerably increased by tubules extending into, and projections out of, the surface. 相似文献

We report here the first cloning of a chalcone flavonone isomerase gene (CHI) from maize. Northern blot experiments indicate that the maize CHI gene (ZmCHI1) is regulated in the pericarp by the P gene, a myb homologue. The ZmCHI1 gene encodes a 24.3 kDa product 55% and 58% identical to CHI-A and CHI-B from Petunia, respectively. This maize CHI gene has four exons and an intron-exon structure identical to the CHI-B gene of Petunia hybrida. RFLP mapping data indicate that some inbred lines contain two additional CHI-homologous sequences, suggesting an organization more complex than that found in Petunia or bean. The possibility that the additional CHI-homologous sequences are responsible for the lack of CHI mutants in maize will be discussed. 相似文献

International Journal for Parasitotogy14:151–157. During histogenesis of the protoscolices of Echinococcus multilocularis, the apical portion of the protoscolex consisting of the suckers, rostellum and hook region develops as an introversion and invagination within the tissue of the basal portion. In vitro incubation of protoscolices in evagination fluid stimulates the emergence of the apical portion. The initiation of evagination is first detected by a surface change in the basal portion. The smooth contour of this surface which lacks microtriches becomes transformed into tegumental indentations that form transverse and longitudinal furrows within the basal tegument as the protoscolices contract and expand, respectively. An orifice formed at the site or junction where the apical portion is invaginated begins to expand laterally in order to allow emergence of the suckers. The hooks are arranged within the invaginated protoscolex with blades directed towards the basal orifice, the handles directed towards the peduncle and the guards directed laterally. This arrangement persists throughout the evagination of the suckers and rostellum until the apical dome of the hook region emerges, thereby rotating the blades laterally in the direction of the peduncle and rotating the handles and guards medially to assume a coronal arrangement. Evagination is an asynchronous event and therefore allows observation of individual protoscolices in various stages of emergence. 相似文献

Cephalaspideans are a group of opisthobranch gastropods that comprises carnivorous and herbivorous species, allowing an investigation of the relationship between these diets and the morphofunctional features of the salivary glands. In this study, the salivary glands of the carnivorous cephalaspidean Philinopsis depicta were observed by light and electron microscopy. The secretory epithelium of these ribbon-shaped glands is formed by ciliated cells, granular cells and cells with apical vacuole. In ciliated cells the nucleus and most cytoplasmic organelles are located in the wider apical region and a very thin stalk reaches the base of the epithelium. These cells possess significant amounts of glycogen. Granular cells are packed with electron-dense secretory granules and also contain several cisternae of rough endoplasmic reticulum and Golgi stacks. The other type of secretory cell is mainly characterized by the presence of a large apical vacuole containing secretion. These cells possess high amounts of rough endoplasmic reticulum cisternae and several Golgi stacks. Vesicles with peripheral electron-dense material are also abundant, and seem to fuse to form the apical vacuole. The available data point out to a significant difference between the salivary glands of carnivorous and herbivorous cephalaspidean opisthobranchs, with an intensification of protein secretion in carnivorous species. 相似文献

The final stages of spermiogenesis in ticks occur in the female genital tract. Scanning electron microscopy was used to follow the morphologic changes that occur in the sperm during this post-ejaculatory spermiogenesis in the African soft tick, Ornithodoros moubata, and to determine a time sequence for its occurrence in vivo. Characteristic features of the maturing and mature cell described include (1) differentiation and detachment of the operculum, (2) changes in cell shape corresponding to different developmental stages, (3) passive migration of the nucleus and acrosome from an anterior to a posterior position, and (4) eversion of that portion of the acrosomal canal containing the nucleus and acrosome. A possible fate for the remainder of the acrosomal canal is suggested by extrusion and detachment of spherical structures, the ‘posterior bubbles’, from the posterior end of the mature supermatozoon. A mechanism for cellular elongation resulting from contractions of the outer sheath is proposed. 相似文献

Tn554, a transposon in Staphylococcus aureus that specifies resistance to erythromycin and spectinomycin, exhibits a high preference for a single chromosomal insertion site. If this site is already occupied by a copy of Tn554, the transposition of a second element is inhibited 100- to 1000-fold. This report defines the locus of the inhibitory activity and presents both a functional and a restriction map of Tn554. Fragments containing parts of Tn554 were cloned on an autonomously replicating plasmid. Those clones containing the "left" end of Tn554 strongly inhibited the transposition of an incoming, intact copy of Tn554. Analysis of deleted derivatives of these clones defined a locus tnpI, which is both necessary and sufficient for transpositional inhibition. This locus consists of the terminal 89 bp of the "left" end of Tn554. It is suggested that this terminal sequence acts to titrate a factor required for transposition. 相似文献
