Experimental and theoretical works were performed for the separation of large polyelectrolyte, such as DNA, in a column packed with gel particles under the influence of an electric field. Since DNA quickly orient in the field direction through the pores, this paper presents how intraparticle convection affects the residence time distribution of DNAs in the column. The concept is further illustrated with examples from solid-liquid systems, for example, from chromatography showing how the column efficiency is improved by the use of an electric field. Dimensionless transient mass balance equations were derived, taking into consideration both diffusion and electrophoretic convection. The separation criteria are theoretically studied using two different Peclet numbers in the fluid and solid phases. These criteria were experimentally verified using two different DNAs via electrophoretic mobility measurements, which showed how the separation position of the DNAs varies in the column in relation to the Peg/Pef values of an individual DNA. The residence time distribution was solved by an operator theory and the characteristic method to yield the column response.  相似文献

以新疆蓝刺头(Echinops ritro L.)全草为研究材料,通过硅胶柱色谱,Sephadex LH-20柱色谱,重结晶等技术对新疆蓝刺头化学成分进行分离纯化,通过理化性质分析及1H-NMR,13C-NMR等技术对化合物结构进行鉴定.结果表明,共分离出5个化合物,分别是三萜类化合物蒲公英甾醇乙酰酯(化合物1)、蒲公英甾醇(化合物2)、黄酮苷类化合物金丝桃苷(化合物3)、胡萝卜苷(化合物4)与β-豆甾醇葡萄糖苷(化合物5).其中化合物1,2,5首次从该种植物中分离,化合物3为首次从该属植物中分离.  相似文献

Complete mineralization of 50 µM of pentachlorophenol (PCP) was achieved anaerobically under continuous flow conditions using two columns connected in series with a hydraulic retention time of 14.2 days, showing the highest reported mineralization rate yet of 3.5 µM day?1. The first column, when injected with a reductive PCP dechlorinating consortium, dechlorinated PCP to mainly phenol and traces of 3‐chlorophenol (3‐CP) using lactate supplied continuously as an electron donor. The second column, with an anaerobic phenol degrading consortium, decomposed phenol and 3‐CP under iron‐reducing conditions with substantial fermentative degradation of organic compounds. When 20 mM of lactate was introduced into the first column, the phenol degradation activity of the second column was lost in a short period of time, because the amorphous Fe(III) oxide (FeOOH) that had been packed in the column before use was depleted by lactate metabolites, such as acetate and propionate, flowing into the second column from the first column. The complete mineralization of PCP was maintained for a long period by reducing the lactate concentration to 4 mM, effectively extending the longevity of second‐column activity with no depletion of FeOOH for more than 200 pore volumes (corresponding to 3,000 days). The carbon balance showed that 50 µM PCP and 4 mM lactate in the influent had transformed to CO2 (81%) and CH4 (3%) and had contributed to biomass growth (8%). A comparison of the microbial consortia introduced into the columns and those flowing out from the columns suggested that the introduced population did not flow out during the experiments, although the microbial composition of the phenol column was considered to be affected by the inflow of microbes from the PCP dechlorination column. These results suggest that a sequential combination of reductive dechlorinating and anaerobic oxidizing consortia is useful for anaerobic remediation of chlorinated aromatic compounds in a microbial permeable reactive barrier. Acutin, a thrombin-like enzyme was purified from Agkistrodon acutus venom in three steps by DEAE-Sepharose CL-6B, Superose 12 column on FPLC and Mono-Q column chromatographies. Its first 15 N-terminal amino acid residues sequence was then determined and the acutin cDNA was isolated from venom gland total RNA using RT-PCR. Determination of its nucleotide sequence allowed elucidation of the amino acid sequence of mature peptide for the first time. The mature acutin has 233 amino acids and its amino acid sequence exhibits significant homology with those of thrombin-like enzymes from crotaline snakes venoms. Based on the homology, the catalytic residues and disulfide bridges of acutin were deduced to be as follows: catalytic residues, His41, Asp84 and Ser179; and disulfide bridges, Cys7-Cys139, Cys26-Cys42, Cys74-Cys231, Cys118-Cys185, Cys150-Cys164, Cys175-Cys200. The recombinant acutin has been expressed in E. coli and purified by affinity column. The renatured recombinant acutin is reported for the first time to have the activity of clotting fibrinogen and arginine-esterase.  相似文献

Effects of operational and system parameters on process scale-up of murine immunoglobulin (IgG2a kappa) purification using Protein A affinity chromatography are investigated. Parameters studied are those related to sample application, elution, ligand concentration on support, and column size change. Between sample application velocities of 0.004 and 0.030 cm/s (16-108 cm/h), the product concentration profiles in eluate did not show significant differences. With given system parameters, the retention time and bandwidth of the peak could be predicted by moment theory. The mean equilibrium constant during elution showed a strong effect of pH between 2 and 4. Within the range of protein A concentrations studied, 0.15-1.22 mg/mL of gel, the column capacity shows a linear relationship with the concentration of protein A immobilized. Dimensional scale-up of the column in the radial direction increased the total purification capacity linearly as the cross-sectional area increased without increasing pressure drop; however, the product concentration was diluted. Scale-up of the column dimension in the axial direction enables higher concentrations of product in the eluate, although the retention time increases linearly as the gel height increases.  相似文献

反复采用硅胶柱色谱法、Sephadex LH-20柱色谱法、ODS柱色谱法、反复重结晶等方法对南山茶果皮的化学成分进行分离纯化,并通过理化常数测定和波谱分析等方法进行结构鉴定。从南山茶果皮中分离并得到了10个化合物,分别鉴定为槲皮素(1)、山奈酚(2)、原儿茶酸(3)、山柰酚-3-O-β-D-葡萄糖基-(6→1)-2′′′,4′′′-O-二乙酰基-α-L-鼠李糖基-(3→1)-2′′′′,3′′′′,4′′′′-O-三乙酰基-α-L-鼠李糖苷(4)、山柰酚-3-O-β-D-葡萄糖基-(6→1)-4′′′-O-乙酰基-α-L-鼠李糖基-(3→1)-3′′′′-O-乙酰基-α-L-鼠李糖苷(5)、3-甲氧基鞣花酸(6)、鞣花酸(7)、羽扇豆烷醇(8)、β-谷甾醇(9)和胡萝卜苷(10)。化合物2、6、7均为首次从南山茶中分离得到,其中化合物6、7为首次从山茶属植物中分离得到。  相似文献

Purification of proteins is commonly a multiple-step process involving size exclusion, ion exchange, affinity, hydrophobic, and other modes of chromatography. In an effort to circumvent the laborious process of collecting the solutes from each column and reintroducing them onto a second column, a valving system is described that directs the samples eluted from a high-performance liquid chromatographic column through a detector with a high-pressure cell into either a second column or into storage loops of a multiloop value. This multiloop value is referred to as a high-pressure fraction collector. After development of the first column is complete, a second solvent can be directed to the second column or high-pressure fraction collector to elute the solutes back through the detector and onto any other column in the system. The process of eluting a sample from a column through a single detector and directing it to the high-pressure fraction collector or any other column in the system may be repeated a number of times. Such valving systems make it possible to chromatograph a single protein component on two or three columns in a short time.  相似文献

Mathematical theory is developed to describe the transient state of isoelectric focusing (pH-gradient electrophoresis) in a linear pH gradient under highly idealized conditions. This theory makes it possible to predict the concentration profile (distribution) for the protein or other amphoteric species of interest as a function of time, when the sample is applied in a zone of infinitesimal thickness at one end of the column, or in a uniform distribution throughout the column. Further, the position of the centroid, and the second moment around the mean, σ2, (square of the standard deviation of peak width) are described as a function of time, irrespective of the initial distribution of the protein in the column. Three arbitrary stages of the “focusing” experiment are considered: (1) Focusing, wherein the sample is applied to a preformed pH gradient; (2) Defocusing, which occurs when the electrical field is abolished after an arbitrary time (usually after the concentration profile has begun to approach its steady state) and diffusion is allowed to occur. (3) Refocusing, which occurs after the electrical field is reapplied. Although stages 1 and 3 are conceptually identical aside from the difference in initial conditions, they may differ in several important respects in practice, both with regard to technical problems of measurement, and with regard to the closeness of conditions to the stated assumptions.This theory should make it possible to predict the time necessary to achieve any desired degree of focusing, i.e., approach to the steady-state distribution. Further, this theory and the techniques of analytical scanning isoelectric focusing provide the basis for measurement of the apparent diffusion coefficient (D), the derivative of velocity with respect to position, and if the field strength is known, the slope of the mobility-pH curve at the isoelectric point, {dMd(pH)}.  相似文献

Zone-interference chromatography is a new method for studying macromolecular interactions (S. Endo and A. Wada, Anal. Biochem. 124 (1982) 372). This method is a new style of affinity chromatography which requires no preparation of affinity-column materials but utilizes the velocity difference in a column between interacting molecular species. Using the stochastic theory on the behavior of solute molecules, both the association and the dissociation rate constants can be analytically obtained from the degree of deformation of elution patterns, i.e., the change of the first and second moments. In order to verify the present theory, computer simulation of elution profiles by the extended plate theory and a binding experiment between glutamate dehydrogenase and ADP have been carried out.  相似文献

Gas-vacuolate heterotrophic bacteria from marine habitats are reported here for the first time. They have been isolated from Antarctic sea ice microbial communities and the underlying water column. The predominant gas-vacuolate bacterium from the sea ice is filamentous and pigmented, whereas those of the water column are unicellular and nonpigmented. The highest concentrations of bacteria in sea ice were found in conjunction with the highest algal (chlorophyll a) concentrations.  相似文献

1. Communities of competing sessile organisms are often modelled using Markov chains. Sensitivity analysis of the stationary distribution of these models tells us how we expect the abundance of each organism to respond to changes in interactions between species. This is important for conservation and management. 2. Markov models for such communities have usually been formulated in discrete time. Each column of the discrete-time transition matrix must sum to 1 (column stochasticity). Sensitivity analysis therefore involves defining a pattern of compensation that maintains column stochasticity as a single transition probability changes. There is little biological theory about the appropriate compensation pattern, but the usual choices involve changing only the elements of a single column of the transition matrix. 3. I argue that if the underlying dynamics occur in continuous time, each transition probability is the net outcome of direct and many indirect interactions. 4. Determining the consequences of changing a single direct interaction will often be of interest. I show how this can be achieved using a continuous-time model. The resulting discrete-time compensation pattern is quite different from those that have been considered elsewhere, with changes occurring in many columns. 5. I also show how to determine which direct interactions are being changed under any discrete-time compensation pattern.  相似文献

The potential antitumor principles, maytansine and maytanprine were isolated from the overground parts of Gymnosporia diversifolia for the first time. The method of separation was modified by application of dry column chromatography, low pressure column chromatography and other separation techniques. Besides, six known compounds were also isolated and identificated as dulcitol, friedelin, β-amyrin, β-sitosterol, kaempferitrin and kaempferol-7-O-rhamnoside respectively. Satellite-linked dive recorders were attached to 53 harbor seal pups in Prince William Sound (PWS) and at Tugidak Island, Alaska, during 1997–1999. We used generalized additive models and bootstrap techniques to describe pup diving behavior during their first year of life. Pups increased their ability to dive during the first 3–6 mo, as indicated by increases in proportion of time in the water (time wet) and maximum dive depth achieved by a pup each day (max-depth) values. Time wet and/or max-depth later decreased, suggesting a seasonal component to diving behavior. Monthly time wet varied from an overall minimum of 0.68 at tagging in July to a maximum of 0.89 in November. Pups spent half of their time wet swimming in water <25 m deep, the shallowest 30% of the available water column. They spent only 5% of their time swimming in the deepest 30% of the available water column, at depths >60–70 m. This strongly suggests they were not feeding on or near bottom during their first year. Average max-depths and deepest actual dives were similar for PWS and Tugidak pups. PWS pups dove deeper sooner and spent less time wet than Tugidak pups during the first few months after tagging, probably as a result of regional bathymetric differences. Diving behavior and body condition suggest that food availability was not likely a major factor in the population decline in PWS during the period of this study.  相似文献

A one-dimensional ultrafiltration problem of fluid flow through a soft permeable tissue or gel under high pressure and compressive strain is solved. A finite deformation biphasic theory is used to model the behavior of the soft porous permeable solid matrix. This theory includes a Helmholtz free energy function which depends on the three principal invariants (I, II, III) of the right Cauchy-Green tensor and which satisfies the Baker-Ericksen inequalities on the principal stresses and strains. The dependence of the porosity phi f and the solidity phi s on deformation is deduced and a generalization of the exponential strain-dependent functional form for the permeability, k = k0 exp (M epsilon), of Lai and Mow (Biorheology 103, 111-123, 1980) is proposed. In this one-dimensional problem, we show that the dependence of the permeability on phi f, phi s, and III is equivalent to its dependence on hydration as proposed by Fatt and Goldstick (J. Colloid Sci. 20, 962-988, 1965). The exact solution of the ultrafiltration problem is derived and asymptotic and numerical methods are used to evaluate it. For high pressures and finite strains, the solution provides some surprising effects. The theory predicts that a material starting with a homogeneous porosity will have a strongly non-homogeneous porosity throughout the column during ultrafiltration. The resulting change in pore size through the filtration column may be very important in understanding its filtration characteristics. It is also found that there is a long delay time, up to 10 to 15 min, before the filtration velocity reaches an equilibrium. In filtration experiments where the rate of mass transport across the tissue or column of gel is important, sufficient time must be allowed for the steady state to be reached.  相似文献
