Lauderia annulata Cleve is probably unique among marine centric diatoms in possessing an elongate dumbbell-shaped nucleus. A lobe at, each end of the nucleus lies adjacent to each valve during interphase and each lobe resembles a typical eucaryotic nucleus. The central portion of the thin strand, which passes through the vacuole connecting the two lobes is Feulgen positive and is nuclear membrane bounded. A group of micro tubules occurs in this strand clustered excentrically between, the nuclear membrane and the tonoplast. Evidence for the coordinated functioning of both nuclear lobes is suggested, by the aggregation of chloroplasts around, both lobes when shade-adapted cells are exposed, to intense white or blue (400–500 nm) light.  相似文献

Summary Glenodinium foliaceum Stein (Pyrrophyta) has a single mesocaryotic nucleus which contains numerous typically dinoflagellate chromosomes and one or more nucleoli with a structure similar to that of nucleoli in higher organisms. In addition this organism possesses another nucleus-like organelle which is here termed the eucaryotic nucleus. This is a polymorphic body which varies in shape from ovoid to a branched filamentous form. As with the mesocaryotic nucleus it is surrounded by a perforated envelope. The organelle contains granular material and usually several nucleoli which again appear to have the typical form of nucleoli. No other dinoflagellate is known in which two nuclei of differing types are found. The function and significance of the presence of the two nuclei is discussed.  相似文献

The fine structure of the binucleate, fucoxanthin-containing dinoflagellate Peridinium foliaceum (Stein) Biechler was re-examined for evidence of an endosymbiout. The eucaryotic nucleus, chloroplasts and associated ribosome-dense cytoplasm were separated by a single invaginating membrane from the rest of the dinoflagellate cell. The triple membrane-enclosed eyespot, mesocaryotic nucleus, trichocysts and accumulation bodies resided in the dinoflagellate cytoplasm. These observations suggest that P. foliaceum contains a membrane-bound endosymbiont, similar to that already described for the closely related species. P. balticum (Levander) Lemmermann.  相似文献

A rice (Oryza sativa L.) cDNA clone coding for the cytoplasmic ribosomal protein L5, which associates with 5 S rRNA for ribosome assembly, was cloned and its nucleotide sequence was determined. The primary structure of rice L5, deduced from the nucleotide sequence, contains 294 amino acids and has intriguing features some of which are also conserved in other eucaryotic homologues. These include: four clusters of basic amino acids, one of which may serve as a nucleolar localization signal; three repeated amino acid sequences; the conservation of glycine residues. This protein was identified as the nuclear-encoded cytoplasmic ribosomal protein L5 of rice by sequence similarity to other eucaryotic ribosomal 5 S RNA-binding proteins of rat, chicken, Xenopus laevis, and Saccharomyces cerevisiae. Rice L5 shares 51 to 62% amino acid sequence identity with the homologues. A group of ribosomal proteins from archaebacteria including Methanococcus vanniellii L18 and Halobacterium cutirubrum L13, which are known to be associated with 5 S rRNA, also related to rice L5 and the other eucaryotic counterparts, suggesting an evolutionary relationship in these ribosomal 5 S RNA-binding proteins.  相似文献

Summary The detailed fine structure of a pigmented marine member of the Cryptophyceae is reported for the first time. The flagella are similar to those of other cryptomonads which have been studied but the root system is probably less complicated. The ejectosomes (trichocysts) have the typical structure but are present in fewer numbers than in many other members of the class. The chloroplasts contain numerous lamellae each composed of two rather dense thylakoids. A single large pyrenoid is attached to a chloroplast and on a short spur beyond this is the centrally placed eyespot. The chloroplasts, pyrenoid and eyespot are all enclosed by an endoplasmic reticulum sheath and starch grains are laid down between this sheath and the chloroplasts. The interphase nucleus is irregular in shape but has the typical eucaryotic appearance. At the anterior end of the cell are numerous coated vesicles which may have an excretory function. The fine structure of Chroomonas is discussed in relation to the ultrastructure of other algae.

Die Geißeln gleichen denen der anderen, bisher untersuchten Cryptomonaden; nur die Art der Einfügung ist vielleicht etwas einfacher. Die Ejectosomen (Trichocysten) haben die übliche Struktur, sind aber weniger zahlreich als bei den anderen Mitgliedern der Familie und Klasse. Die Chromatophoren enthalten zahlreiche Lamellen, deren jede aus zwei ziemlich dichten Thylakoiden zusammengesetzt ist. Ein einzelnes pyrenoid sitzt an den Chromatophoren. Ein zentraler Augenfleck ist an einem kurzen Fortsatz derselben befestigt. Chromatophoren, Pyrenoid und Augenfleck sind gemeinsam von einer endoplasmatischen netzigen Hülle (endoplasmatisches Reticulum) umhüllt. Zwischen dieser Hülle und den Chromatophoren werden Stärkekörner abgelagert. Der Zellkern ist in der Interphase unregelmäßig gestaltet. Er hat das übliche Aussehen der eukaryotischen Lebewesen. Am Vorderende sind zahlreiche umhüllte Bläschen, die wohl Abscheidungsorganellen sind. Der Feinbau von Chroomonas wird mit dem anderer Algen verglichen.

A method of preparation for electron microscopy of fern spores in early stages of germination is presented. The cytochemistry and fine structure of Onoclea spores during the early stages of germination are described. The cytoplasm of the hydrated spore is filled with lipid droplets, protein granules and chloroplasts. During the early stages of development ribosomes and mitochondria increase in the area surrounding the central nucleus, and a new peripheral wall forms around the protoplast. Microtubules and large, branching mitochondria are associated with the nucleus during migration from its original central position in the spore to the proximal face and then to one end of the spore. There is no morphological polarization of cytoplasmic organelles of the spore before migration of the nucleus.  相似文献

The general fine structure of the giant coenocyte Caulerpa prolifera (Forsskål) Lamouroux is presented. The cytoplasm forms a parietal layer throughout the plant without any regular membranous separations within organs or between organs. The vacuome is similar in structure. There are distinct patterns of organelle distribution in the highly polar cytoplasm of rhizoids and blades. The organelles are compared to those of other members of the division Chlorophyta and are typical. Amyloplast structure is compared to that of chloroplasts. A possible developmental sequence from chloroplast buds through an unusual circular body is suggested.  相似文献

The 5' cap structure of eucaryotic mRNA plays a pivotal role in mRNA metabolism. This report demonstrates that anti-sense oligonucleotides equipped with 3'-overhanging nucleotides modulate the amount of recombinant human eucaryotic initiation factor-4E that binds to a 5'-capped oligoribonucleotide. The degree of inhibition or enhancement of protein binding is dependent upon the number and sequence of overhanging nucleotides. A 45% inhibition of complexation was observed by the addition of one 3'-overhanging guanosine residue. Addition of a second residue (+2/GN) resulted in a higher degree of inhibition, 77-88%. In contrast, addition of one adenosine residue enhanced the formation of the eucaryotic initiation factor-4E-m7GpppRNA complex by 213%. Modulation of protein interactions with the 5'-cap structure is likely to effect several biological events, including pre-mRNA processing, transport of the mRNA from the nucleus to the cytoplasm and translation of the target mRNA. This targeting strategy in anti-sense chemistry may have practical applications in experimental biology and medicine.  相似文献

Summary The genes corresponding to the L10 and L12 equivalent ribosomal proteins (L10e and L12e) ofEscherichia coli have been cloned and sequenced from two widely divergent species of archaebacteria,Halobacterium cutirubrum andSulfolobus solfataricus. The deduced amino acid sequences of the L10e and L12e proteins have been compared to each other and to available eubacterial and eucaryotic sequences. We have identified the hyman P0 protein as the eucaryotic L10e. The L10e proteins from the three kingdoms were found to be colinear. The eubacterial L10e protein is much shorter than the archaebacterial-eucaryotic proteins because of two large deletions, one internal and one at the carboxy terminus. The archaebacterial and eucaryotic L12e proteins were also colinear; the eubacterial protein is homologous to the archaebacterial and eucaryotic L12e proteins, but has suffered rearrangement through what appear to be gene fusion events. Intraspecies comparisons between L10e and L12e sequences indicate the archaebacterial and eucaryotic L10e proteins contain a partial copy of the L12e protein fused to their carboxy terminus. In the eubacteria most of this fusion has been removed by the carboxy terminal deletion. Within the L12e-derived region, a 26-amino acid-long internal modular sequence reiterated thrice in the archaebacterial L10e, twice in the eucaryotic L10e, and once in the eubacterial L10e was discovered. This modular sequence also appears to be present as a single copy in all L12e proteins and may play a role in L12e dimerization, L10e–L12e complex formation, and the function of L10e–L12e complex in translation. From these sequence comparisons a model depicting the evolutionary progression of the L10e and L12e genes and proteins from the primordial state to the contemporary archaebacterial, eucaryotic, and eubacterial states is presented.  相似文献

The ultrathin structure of amoeboid flagellate Thaumatomastix sp. is considered. The cell is surrounded by two-layered triangular scales. They are formed on the surface of mitochondria. Pseudopodia grabbing bacteria run from ventricular furrow, which is armored with two longitudinal bands of microtubules. Heterodynamic flagella run from small flagellar pocket. Long back flagellum has thin mastigonemes. Proximal area of short flagellum is covered with flat oval scales. Transitional flagellant zone has no spiral or other additional elements. Transverse plate is localized above cell surface. Kinetosomes are parallel to each other. Vesicular nucleus and Golgi apparatus have typical structure. Oval mitochondria contain tubular cristae. Within cytoplasm, extrusive organelles (kinetocysts) containing amorphous material and capsule were found. The latter consists of muff and cylinder. Plasmodial and cystic phases of development have not been discovered. Contractile vacuole is absent. The resemblance between Thaumatomastix sp. and other thaumatomonads has been discussed.  相似文献

Gymnodinium aeruginosum has the usual fine structure of a dinoflagellate but does not seem to contain a well elaborated peduncle or a microtubular basket. Naked cells are surrounded by a single large amphiesmal vesicle. It houses an endosymbiont with typical blue-green cryptophycean chloroplasts (generally only one), cryptophycean starch grains in the periplastidal cytoplasm without a nucleomorph, and two membranes separating the periplastidal cytoplasm from the cryptophycean cytoplasm which contains mitochondria, ER, vesicles and ribosomes, but no eukaryotic nucleus. The endosymbiont is surrounded by a single membrane. Possible ways of the acquisition of the endosymbiont and the problem of the existence of ribosomes within a compartment without nucleus are discussed.Devoted to Prof. Dr.L. The fine structure of 3 members of the genus Cryptomonas, C. rostrella, C. reticulata, and C. calceiformis, has been examined. These species exhibit the features typical of the class Cryptophyceae. The presence of subpellicular trichocysts causes a regular folding of the periplast, and larger trichocysts are present within the gullet. The plastid contains thylacoids arranged in pairs and a prominent pyrenoid; both structures are enclosed in 4 membranes, the outermost of which is a rough endoplasmic reticulum. The nucleus, Golgi apparatus and the Corps de Maupas occupy characteristic positions within the cell, and are the most characteristic features. All these structures indicate that this algal class occupies a unique phylogenetic position.  相似文献

Accumulating evidence suggests that unicellularArchezoa are the most primitive eukaryotes and their nuclei are of significance to the study of evolution of the eukaryotic nucleus. Nuclear matrix is an ubiquitous important structure of eukaryotic nucleus; its evolution is certainly one of the most important parts of the evolution of nucleus. To study the evolution of nuclear matrix, nuclear matrices ofArchezoa are investigated.Giardia lamblia cells are extracted sequentially. Both embedment-free section EM and whole mount cell EM of the extracted cells show that, like higher eukaryotes, this species has a residual nuclear matrix in its nucleus and rich intermediate filaments in its cytoplasm, and the two networks connect with each other to form a united network. But its nuclear matrix does not have nucleolar matrix and its lamina is not as typical as that of higher eukaryotes; Western blotting shows that lamina ofGiardia and two otherArchezoa Entanzoeba invadens andTrichomonas vaginali all contain only one polypeptide each which reacts with a mammalia anti-lamin polyclonal serum and is similar to lamin B (67 ku) of rnammiia in molecular weight. According to the results and references, it is suggested that nuclear matrix is an early acquisition of the eukaryotic nucleus, and it and the “eukaryotic chromatin” as a whole must have originated very early in the process of evolution of eukaryotic cell, and their origin should be an important prerequisite of the origin of eukaryotic nucleus: in the lamin (gene) family, B-type lamins (gene) should be the ancestral typz and that A-type lamins (gene) might derive therefrom. Amoebae of Naegleria fowleri nN68 became elongated flagellated cells 150 to 180 min after subculture to non-nutrient buffer. N. fowleri NF69 did not become elongated or flagellated under these conditions. Electron microscopic examination of N. fowleri confirmed that it is a typical eucaryotic protist with a distinct nuclear envelope and prominent nucleolus, numerous vacuoles and cytoplasmic inclusions, pleomorphic mitochondria, and some rough endoplasmic reticulum. During incubation in non-nutrient buffer, both strains lost ultraviolet-absorbing material to the medium, and the number of vacuoles decreased. In strain nN68, basal bodies, a rootlet, and flagella are formed quickly after an initial lag of 90 min. Initially, the rootlet is not associated with the nucleus but they become associated subsequent at the leading end of the elongated cell. In elongated cells, the rootlet lies in a furrow or groove extending the length of the nucleus. Flagella of N. fowleri nN68 exhibit the typical 9 + 2 arrangement of filaments and are surrounded by a sheath which is continuous with the plasma membrane. The enflagellation process in N. fowleri can be manipulated reproducibly.  相似文献

The spermatozoon and some spermatid stages of Siboglinum (Pogonophora) have been examined by light and electron microscopy. In the spermatozoon a helical acrosome, a helical nucleus and a “body” with axonema follow each other in normal sequence. Head and tail are joined by a very short neck region containing two modified centrioles. The posterior portion of the nucleus is surrounded by a mitochondrial sheath consisting of three tightly wound mitochondrial helices. In the main portion of the tail the 9+2 unit is sorrounded by a granular sheath of dense material. In the neck region a centriole adjunct develops into a dense substance containing about nine rods. At an early stage, when the centriolar apparatus and flagellum become associated with the nucleus, three large mitochondria with fairly regular cristae are seen at the base of the nucleus. A well developed Golgi apparatus is present in early stages. Rows of microtubules are observed encircling the spermatid nucleus. Compared with the primitive type of spermatozoon the pogonophore sperm shows elongated and specialized nucleus, acrosome and mitochondria. It is concluded that the ancestral form must have had a fairly primitive spermatozoon and that evolution has proceeded towards a modified sperm with complicated spiral structure in connection with the evolution of a modified biology of fertilization, viz. specialized spermatophores. It is not known how the spermatophore discharges the spermatozoa nor how the spermatozoa find their way to the eggs. Two kinds of sperms are produced in the gonads of Siboglinum. The atypical sperm is smaller than the typical one.  相似文献

A new armored dinoflagellate species, Heterocapsa psammophila Tamura, Iwataki et Horiguchi sp. nov. is described from Kenmin‐no‐hama beach, Hiroshima, Japan using light and electron microscopy. This dinoflagellate possesses the typical thecal plate arrangement of the genus Heterocapsa, Po, cp, 5′, 3a, 7′′, 6c, 5s, 5′′′, 2′′′′; and the 3‐D body scales of Heterocapsa on the plasma membrane. The cell shape is ovoidal. The spherical nucleus and the pyrenoid are situated in the hypotheca and the epitheca, respectively. The ultrastructure of H. psammophila is typical of dinoflagellates and the pyrenoid is invaginated by cytoplasmic tubules. H. psammophila is distinguished from all other hitherto‐described Heterocapsa species by the cell shape, the relative position of the nucleus and pyrenoid and the structure of the body scale. The habitat and behavior of this new species in culture suggest that the organism is truly a sand‐dwelling species.  相似文献

The fine structure of the primary nucleus in Bryopsis hypnoides was compared with that of the primary nucleus in Acetabularia calyculus and Batophora oerstedii. In Bryopsis the mature primary nucleus contains a peripheral reticulum composed of 4–6 layers of 200 A diameter fibrils. The nuclear membrane has numerous nuclear pores that are located adjacent to the openings in the layers of the reticulum. Each nuclear pore is constituted of peripheral globular elements and a central granule. Although pores of similar structure are present in the nuclear envelope of Acetabularia and Batophora, a fibrillar reticulum is absent.  相似文献

Spermatogenesis and the structure of mature spermatozoa were studied using TEM in a free-living marine chromadorid nematode Neochromadora poecilosoma from the Sea of Japan. In spermatocytes, fibrous bodies (FB) develop; in spermatids, the synthetic apparatus lies in the residual body, while the nucleus, mitochondria, and FB are located in the main cell body (MCB). The nucleus consists of a diffuse chromatin of fibrous structure, which is not enclosed in a nuclear envelope. In the spermatid stage, the development of FB is completed, and immature spermatozoa from the proximal region of the testis do not show any structural differences from the MCB of spermatids. The mature spermatozoa are polarized cells. They attach to the uterus wall by a pseudopod filled with filaments of the cytoskeleton; in the MCB of spermatozoon, there is a nucleus surrounded by mitochondria and osmiophilic bodies. The spermatozoa of N. poecilosoma show typical ultrastructure features of sperm cells found in most studied nematodes (amoeboid nature and the absence of axoneme, acrosome, and nuclear envelope). However, no aberrant organelles characteristic of nematode spermatozoa were found throughout sperm development in N. poecilosoma and other chromadorids.  相似文献
