Abstract

The concentration of chlorophyll a, b, and total chlorophyll have been monitored on a seasonal basis in Brachythecium rutabulum. Total chlorophyll increases during summer full canopy conditions from 1.70 mg chl g?1 on 8 May to 11.1 mg chl g?1 on 11 October. Photosynthetic-illumination curves show that during this period light saturation declines from 200 μmol m?2s?1 to 30 μmol m?2s?1 by 6 July, and light compensation falls dramatically from 65 μmol m?2s?1 to 4 μmol m?2s?1. The data also appear to support the conclusion that there is concurrently an increase in the density of photosynthetic units by the end of September.  相似文献

SUMMARY

A four year pre-impoundment study of the catchment of the Hendrik Verwoerd Dam was conducted. The Orange River was the major source of dissolved chemicals and suspended sediments, while the Caledon River contributed 24 and 30% respectively, and the Kraai River 9 and 8% respectively. The upper reaches of the Orange River catchment contributed mainly to the dissolved chemical and sediment loads, in contrast to the minimal contribution of the lower reaches. This was in spite of the relatively stable geological formations in the upper reaches and the erodable formations in the lower reaches. Dry-wet seasonal variations occurred in nutrient and sediment concentrations and loads, except for the Bell River where constant values were measured. The water was characterized by magnesium and bicarbonate dominance. Adsorption by the suspended sediments was dominated by calcium (potassium in the Bell River) and zinc among the trace elements. Due to its low salinity and sodium adsorption ratio, the water was suitable for irrigation. High magnesium levels made it, however, less attractive for irrigational purposes.

The phosphorus loading rate at the inflow of the Hendrik Verwoerd Dam was only 0,55 g P m?2 a?1, which was well below the 1,39 g P m?2 a?1 critical rate for eutrophic conditions, applying the Vollenweider Model. Therefore, this impoundment was classified as oligo-mesotrophic.  相似文献

Three photosynthetic parameters of 7 species of marine diatoms were studied using Na214CO3 at 5–8 C using log phase axenic cultures. The cell volumes of the different species varied from 70 μm3 to 40 × 105μm3. The present experiment is consistent with the interpretation that the initial slope α (mg C · [mg chl a]?1· h?1· w?1· m2) of photosynthesis vs. light curves is controlled by self-shading of chlorophyll a in the cell. Pm, the rate of photosynthesis at light saturation (mg C · [mg cell, C]?1· h?1) and R, the intercept at zero light intensity (mg C · [mg cell C]?1· H?1) are both dependent on the ratio of surface area to volume of cell.  相似文献

Summary

Selected limnological attributes of the Okavango Delta panhandle were measured during a brief summer survey of “open-water” habitats extending from the permanent mainstream channel, through contiguous off-channel lagoons and still backwaters, to seasonally isolated floodplain lagoon and temporary pool biotopes on the left-bank of the Okavango River at Seronga.

Wide ranges in most determinants were evident along this profile:- temperature (27–34°C); conductivity(4–12.7mS m?1); pH(5.7–9.2); transparency(0.2–>2.5 m Secchi depth); dissolved oxygen (20–220% saturation); PO4-P (61–110 μg ??1): SiO2—Si (6.9–14.0 mg ??1): NH4-N (30–44 μg ??1): chlorophyll (1.3–183 μ.g ??1). Zooplankton was variably diverse (species richness from ≥ 3 to 20), comprising both euplanktonic (Bosmina, Ceriodaphnia, Daphnia, Diaphanosoma, Moina, Mesocyclops, Thermocyclops, Tropodiaptomus) and more typical epiphytic crustacean taxa (Alona, Macrothrix, Pleuroxus), along with various rotifers (Brachionus, Hexarthra, Keratella, Trichocera) and other taxa (Arcella, Ostracoda. Chaoborus). Abundance varied widely between habitats. The littoral macrozoobenthos showed surprisingly low diversity, and was dominated by freshwater shrimps (Caridina).

Substantial allochthonous inputs to the Okavango swamps were evident from the significant concentrations of total suspended solids (7.6–12.6 mg ??1 , organic content of 33–41%) carried by the mainstream Okavango River during the survey.  相似文献

Seasonal and interannual dynamics of phytoplankton content (chlorophyll a), organic matter, and nutrients, as well as water temperature and water transparency, have been analyzed for the weekly observations performed in the Ob River from February to November, 2012–2015. The period of intensive phytoplankton growth (>10 mg/m3 Chl. a) lasts 70–90 days at a long spring flood in average and high-water years; in the absence of flooding during a dry year it exceeds 110 days. Seasonal phytoplankton dynamics is characterized by two summer maxima in July–August and in September (Chl. a content ~25 mg/m3); during a dry year, there is also a peak in June (up to 37.3 mg/m3). Seasonal dynamics of Chl. a in summer–autumn low water is closely related to the water temperature; in high water and autumn low water it is closely related to water transparency; in summer it is closely related to BOD5; and, in the summer–autumn period, it is closely related with nitrate content. According to the annual average (5.7–11.1 mg/m3) and maximum (19.2–35.0 mg/m3) concentrations of chlorophyll a, the trophic status of the river corresponds to mesotrophic–eutrophic water and the water quality corresponds to the second class, i.e., “clean water.”  相似文献

SUMMARY. 1. Phytoplankton density (organisms ml?1), standing crop (chlorophyll a mg m?2) and primary productivity (mg C m?2 d?1) were measured during years 2 (1976) to 5 (1979) after impoundment on West Point Lake. 2. West Point waters had low alkalinity (<0.4 meq 1–1) and low conductivity (<75 μs cm?1 at 20°C) but N and P concentrations typically exceeded those considered apt to cause nuisance blooms of algae. Abiogenic turbidity was normally higher in the upstream areas of the reservoir than in the downstream areas and was several times higher in winter-spring than in summer-autumn due to increased rains and runoff. 3. Primary productivity varied greatly both temporally and spatially. A mean value of 684 mg C m?2 d?1 was well within the mesotrophic range and did not approach the highly eutrophic state predicted. Productivity increased from a low of 550 mg C m?2 d?1 in 1976 to high of 763 mg Cm?2d?1 in 1979. 4. Observed variation in both chlorophyll a and primary productivity was more predictable in the cool (December-March) than in the warm (June-September) season and with plant nutrient data than without it. With plant nutrient data in the cool season 84% and 86% of the variation (R2) in chlorophyll a and productivity, respectively, were accounted for by the regression equations. During the warm season, with plant nutrient data, regression equations accounted for 44% and 68% of the variation in chlorophyll a and productivity, respectively. Higher R2 values in cool seasons resulted from the overriding influence of abiogenic turbidity on phytoplankton communities.  相似文献

Cooking Lake (113°02′W, 53°26′N), a well-mixed, shallow (mean depth (1.59 m), eutrophic lake in Alberta, Canada, is characterized by eutrophic chlorococcalean and cyanophycean phytoplankton associations, and little change in standing crop with increasing depth. Standing crop and primary productivity are low during the winter but pronounced spring and summer maxima occur. Mean yearly areal standing crop (ΔB) and primary productivity (ΔA) were 212.4 mg m?2 chlorophyll a and 301.8 mg C h?1 m?2 respectively. Annual productivity was estimated at 1322 g C m?2. The mean increase in the extinction coefficient (?) per unit increase in standing crop (B) was 0.03 In units m?1. High non-algal light attenuation (?q) occurred avenging 41 which prevented the ratio B/? from attaining more than 65% of the theoretical maximum except once when algal self-shading occurred. Close correlations existed between B (mg m?3 chlorophyll a) and A max (mg h?1 m?3) ΔA and ΔB, ΔA and B, Amax, and Amax/?, and ΔA and Io′, (W m?2). The depth of the euphotic zone (Zeu) varied between 0.5 and 1 25 m; the average relationship between zeu and E was Zeu= 3.74/?, and the mean standing Crop found in the euphotic zone represented 55.2% of the theoretical maximum, The high ?q, values made the model of Tailing (1957) inapplicable to Cooking Lake. The Q10 value for the lake was 2.2. The maximum rate of photosynthesis per unit of population per h. Ømax, (mg C sag chlorophyll a?1 h?1) was more closely related to temperature than irradiance and ma depressed by pH values greater than 9.1. Growth of the phytoplankton was not nutrient limited: instead irradiance and temperature were more important. Indirect evidence that free CO2 limited photosynthetic rates, is provided by the Ømax: pH relationship.  相似文献

SUMMARY

An eighteen month study (January 1979 - June 1980) of the sediment, emergent and submerged plant-associated benthic macro-invertebrates of the western sector of the Touw River Flood-plain is reported for four study sites: Wilderness Lagoon, the Touw River and East and West Serpentine. Total invertebrate standing stocks on emergent vegetation were negligible. Those associated with submerged plant communities (essentially Potamogeton pectinatus L.), were generally very much greater than sediment-associated invertebrate standing stocks. The following mean biomasses were recorded for the study period: (a) sediments - Wilderness Lagoon, 19,25; Touw River, 13,06; West Serpentine, 4,37; East Serpentine, 4,68 g dry mass m?2; (b) - Potamogeton-associated fauna - Touw River, 128,9; West Serpentine, 91; East Serpentine, 35,78 g dry mass m?2. Of the 37 taxa recorded for Wilderness Lagoon, 33 for Touw River and 31 each for West and East Serpentine sites, the most consistently dominant species were the amphipods, Melita zeylanica Stebbing, Corophium triaenonyx Stebbing, and Grandidierella lignorum Barnard, the tanaid, Apseudes digitalis Brown, and the bivalve mollusc, Musculus virgiliae Barnard. However, suspension feeding Musculus was by far the most important taxon.

In addition to normal winter die-back of Potamogeton, submerged plant cutting activities in the Serpentine lead to an extended period of plant senescence, a prolonged recovery period at the West Serpentine station (between 4–5 months, compared with un-cut Touw River - approximately 2 months), almost complete failure of the plant at East Serpentine, and associated reductions or shifts in the invertebrate community standing stocks, composition and structure. Sediment-associated invertebrate standing stocks at both West and East Serpentine stations increased after Potamogeton harvesting, both immediately and in the long-term. However, these sediment fauna increases nowhere near compensated for the reductions of plant-associated faunal standing stocks due to cutting activities. The significance of these findings is discussed in terms of future management of the area, and comparisons with the situation in Swartvlei, where natural Potamogeton reduction occurred, and with other estuarine and coastal systems, are made.  相似文献

1. An extensive series of PONAR grabs was used to determine the distribution and abundance of unionid clams in the freshwater tidal Hudson River. 2. The five species of unionids collected were distributed very unevenly within the river. Mean river-wide density and biomass of unionids were 8.0m?2 and 6.2 g DM m?2 (shell-free)., respectively. 3. The environmental variables that we measured (water depth, distance from shore, sediment granulometry and organic content, presence or absence of macrophytes, and the chlorophyll a and particulate organic matter content of the water) explained little of the variation in abundance of unionids. 4. The distributions of the various species of clams did not differ significantly with respect to the environmental variables measured. 5. We estimate that unionids filter a significant amount of water (0.14m3 m?2 day?1, on average) in the Hudson River estuary, roughly equivalent in magnitude to downstream flushing. 6. We project that unionids will serve as a major substratum for the settlement of the zebra mussel (Dreissena polymorpha), which is now invading the estuary. We emphasize that unionids may play important non-trophic roles in large river ecosystems.  相似文献

The dry matter production in Polytrichum commune protonemata was increased when the light intensity was increased from 0 to 160 μE m?2 s?1, and at 160 μE m?2 s?1 production was about 200% of that found at 17 μE m?2 s?1. Production of chlorophyll (Chl) was increased by increasing light intensity from 0 to 17 μE m?2 s?1, but decreasing at light intensities above 17 μE m?2 s?1. At 160 μE m?2 s?1 the production of Chl was only about 50% of that at 17 μE m?2 s?1. The rate of CO2 fixation was low (0.31 μg CO2/mg Chi × h) at the light intensity of 17 μE m?2 s?1 as compared with that at 160 μE m?2 s?1 (0.83 μg CO2/mg Chi × h). Production of mono- (MGDG) and diglycosyl diglycerides (DGDG) was closely associated with that of chlorophylls. At the higher light intensity (160 μE m?2 s?1) production of glycolipids was about 60% of that at 17 μE m?2 s?1. Production of more polar lipids was less affected by light intensity. Light intensity also affected the fatty acid pattern of the lipid fractions. The effect was most pronounced in the MGDG fraction, where the proportion of C 18: 3ω3 + C 16: 3ω3 was higher at the higher light intensity.  相似文献

Abstract

The aim of this study was to quantify algal colonisation on anthropogenic surfaces (viz. building facades and roof tiles) using chlorophyll a (chl a) as a specific biomarker. Chl a was estimated as the initial fluorescence F0 of ‘dark adapted’ algae using a pulse-modulated fluorometer (PAM-2000). Four isolates of aeroterrestrial green algae and one aquatic isolate were included in this study. The chl a concentration and F0 showed an exponential relationship in the tested range between 0 and 400 mg chl a m?2. The relationship was linear at chl a concentrations <20 mg m?2. Exponential and linear models are presented for the single isolates with large coefficients of determination (exponential: r2 > 0.94, linear: r2 > 0.92). The specific power of this fluorometric method is the detection of initial algal colonisation on surfaces in thin or young biofilms down to 3.5 mg chl a m?2, which corresponds to an abundances of the investigated isolates between 0.2 and 1.5 million cells cm?2.  相似文献

Microscopic epilithic algae in the River Itchen at Otterbourne near Southampton and in the Ober Water in the New Forest were studied during 1984 and 1985. The River Itchen rises from chalk springs and has a steady pH near 8.2 and a mean alkalinity of 236 mg HCO3 1–1; at the study site the river is about 16 m wide and 20 cm deep, with a mean flow rate of 0.33 m s–1 and a discharge ranging through the year between 0.34 and 2.46 m3 s–1. The Ober Water, which drains sands and gravels, has a pH between 6.9 and 7.2 and a mean alkalinity of about 50 mg HCO3 1–1; at the study site it is about 6 m wide, with a mean flow rate of 0.27 m s–1 and a discharge ranging through the year between 0.08 and 1.0 m3 s–1.Epilithic algae removed from the pebbles that form the major part of the beds of both streams show seasonal changes in abundance and composition. Diatoms peaked in April/May and dominate the epilithic flora in both streams, comprising 70–95% of all algal cells; highest numbers of chlorophytes occurred in summer and cyanophytes increased in autumn. The species composition of the epilithic flora in the two streams was different, as was the population density; algal cell numbers ranged between 500 and 7000 cells mm–2 of stream floor in the River Itchen and between 8 and 320 cells mm–2 of stream floor in the Ober Water. The chlorophyll a content of epilithic algae in the River Itchen ranged between 115 and 415 mg m–2 of stream floor, representing an annual mean biomass of about 8 g m–2, whereas in the Ober Water a chlorophyll a content of 2.2 to 44 mg m–2 of stream floor was found, representing an annual mean biomass of about 1 g m–2. Cautious estimates of the annual production of epilithic algae in these streams suggest a value of about 600 g organic dry weight m–2 in the River Itchen and about 75 g m–2 in the Ober Water.  相似文献

Abstract

The characteristics of adenosine receptors and adenosine uptake sites in brain are presented. High affinity adenosine receptors of the A1 type bind [3H]cyclohexyladenosine ([3H]CHA) and [3 H]diethyl-phenyl-xanthine ([3H]DPX) with 10?9 potency while adenosine uptake sites are labeled 10?10 potency with [3 H]nitrobenzyl-thioinosine ([3H]NBI). NBI does not inhibit either [3H]CHA (agonist) or [3H]DPX (antagonist) binding to adenosine receptors in brain cortical membranes and conversely CHA and other adenosine receptor ligands are very poor inhibitors of [3H]NBI binding to adenosine uptake sites. A number of other differences between the receptor and uptake site are discussed which provide rather strong evidence that these two sites are quite distinct and that the labeled ligands used represent specific probes for each site.  相似文献

We investigated the productivity of nymphs of the mayfly Hexagenia limbata in Lake Waco, a central Texas reservoir, and assessed its association with chlorophyll content of the water. We hypothesized that food availability measured as chlorophyll content of the water may directly associate with growth of Hexagenia and predict population productivity. To test this, we compared production by mayfly populations at two stations in the same reservoir; a northern station receiving water input with high chlorophyll content, and a southern station receiving water with low chlorophyll content. Both stations had similar substrate type and abundant mayflies. Benthic samples were collected from October 1984 through September 1985, and dissolved oxygen and temperature of the water were monitored.Annual production (size-frequency method) was 1270 mg m–2 (P/B = 7.5) at the northern station and 1990 mg m–2 (P/B = 6.1) at the southern station. The mean standing crop was 323 mg m – 2 at the southern station and 169mg m–2 at the southern station. Densities of mayflies at the two stations were not significantly different.Mean chlorophyll concentration (total mg pigment) during the sampling period was 23.5 mg m–3 at the northern station and 16.7 mg m – 3 at the southern station. Therefore, the station with lower mean chlorophyll content had higher secondary productivity by Hexagenia. Conversely, the station with higher mean chlorophyll content had lower mayfly productivity. The productivity of the mayfly populations did not positively associate with the chlorophyll content of the water, and chlorophyll content did not predict the success of the population of Hexagenia. Variation in mayfly growth success was associated with differences in temperature and dissolved oxygen. The northern station with higher chlorophyll content and lower productivity had low dissolved oxygen and temperatures higher than optimum for growth.  相似文献

Periphytic biomass in a downriver riffle of the Grand River in Southern Ontario, Canada was measured with concrete-block glass slide samplers from May to December 1971. Average rate of accumulation of periphyton on glass slides was 266.2 mg/m2/day ash-free weight, 120.9 mg/m2/day carbon, 20.4 mg/m2/day nitrogen, or 1.11 mg/m2/day chlorophyll a and on concrete blocks was 590.0 mg/m2/day ash-free weight, 202.9 mg/m2/day carbon, 22.8 mg/m2/day nitrogen, or 2.95 mg/m2/day chlorophyll a. The standing biomass of periphyton on glass slides varied from a maximum of 21.6 g/m2 between May 28 and July 15 to a minimum of 0.7 g/m2 in December with an average of 8.6 g/m2. The biomass on concrete blocks ranged from a high of 132.5 g/m2 in August to a low of 29.9 g/m2 in October with a mean of 66.1 g/m2. The well established periphyton on concrete blocks towards the end of the study period was similar to that from native rocks with respect to biomass, carbon and nitrogen contents, and ratios of biomass to chlorophyll a. This suggests that concrete blocks are a better substrate than glass slides for measuring riverbed periphyton. By comparing biomass of periphyton on slides and on concrete blocks, the average rate of loss of periphyton from concrete blocks was estimated to be 2.9 g/m2/day, representing 63% of the mean total accumulation rate of the periphyton. The magnitude of the estimated total loss and the high standing biomass of the periphyton on concrete blocks manifest the importance of the periphyton as a source of organic matter in the river.  相似文献

This study reports the primary production of phytoplankton determined with a 13C tracer, and their related variables, in Lake Kasumigaura, a shallow, hyper-eutrophic lake, and the second largest lake in Japan. Measurements were conducted monthly from August 1981 to December 2013 at four stations within the lake. Monitoring was a component of the Lake Kasumigaura Long-term Environmental Monitoring program, conducted by the National Institute for Environmental Studies (NIES) since 1977. The program collects data on water quality, and plankton and benthic communities. Lake Kasumigaura is registered as a core site of the Japan Long-term Ecological Research Network (JaLTER), which is a member of the International Long-term Ecological Research Network (ILTER). This dataset includes daily primary production (Pzd gC m?2 d?1) and the six parameters required to calculate Pzd: maximum photosynthesis rate (P max gC gC?1 h?1); light irradiance at the junction of the initial slope (α (gC gC?1 h?1) (μmol photon m?2 s?1)?1) and P max of the photosynthesis vs. irradiance (P vs. E) curve (E k μmol photon m?2 s?1); attenuation coefficient of photosynthetically available radiation (PAR) (K PAR m?1); water depth at each sampling station (Z b , m); dissolved inorganic carbon (DIC mgC L?1) and particulate organic carbon concentrations (POC gC m?3); and chlorophyll a amounts (Chl.a μg L?1). Daily primary production was calculated by obtaining a P vs. E curve over a short-term incubation (approximately 1 h) in a water tank using in situ water temperature in the laboratory, based on the field conditions of the sampling date. The dataset has been used for ecological studies as well as for management studies on water quality and ecosystems. This dataset is unique among the available published papers concerning lakes or primary production in various ecosystems, collected over a long period of time and freely available.  相似文献

The building blocks fac-[99mTc{κ3-HB(timMe)3}(CO)3] and fac-[99mTc{κ3-R(μ-H)B(timMe)2}(CO)3] [R is H (4a), Ph (5a); timMe is 2-mercapto-1-methylimidazolyl] were obtained almost quantitatively by reacting fac-[99mTc(CO)3(H2O)3]+ with the corresponding scorpionate. These compounds cross the intact blood–brain barrier in mice, with significant retention in the case of 4a and 5a. Using 4a as the lead structure, we have synthesized the functionalized complexes fac-[M{κ3-H(μ-H)B(timBu-pip)2}(CO)3] [M is Re (8), 99mTc (8a); timBu-pip is methyl[4-((2-methoxyphenyl)-1-piperazinyl)butyl](2-mercapto-1-methylimidazol-5-yl)methanamide] and fac-[M{κ 3-H(μ-H)B(timMe)(timBu-pip)}(CO)3] [M is Re (9), 99mTc (9a)] and evaluated their potential as radioactive probes for the targeting of brain 5-HT1A serotonergic receptors. The Re complexes exhibit excellent affinity [IC50=0.172 ± 0.003 nM (8); IC50=0.65 ± 0.01 nM (9)] for the 5-HT1A receptor. The radioactive congeners (99mTc) have shown an initial brain uptake of 1.38 ± 0.46%ID g−1 (8a) and 0.43 ± 0.12%ID g−1 (9a), but suffer from a relatively fast washout.  相似文献

Light effect on cultures of microalgae has been studied mainly on single species cultures. Cyanobacteria have photosynthetic pigments that can capture photons of wavelengths not available to chlorophylls. A native Louisiana microalgae (Chlorella vulgaris ) and cyanobacteria (Leptolyngbya sp.) co‐culture was used to study the effects of light quality (blue–467 nm, green–522 nm, red–640 nm and white–narrow peak at 450 nm and a broad range with a peak at 550 nm) at two irradiance levels (80 and 400 μmol m?2 s?1) on the growth, species composition, biomass productivity, lipid content and chlorophyll‐a production. The co‐culture shifted from a microalgae dominant culture to a cyanobacteria culture at 80 μmol m?2 s?1. The highest growth for the cyanobacteria was observed at 80 μmol μmol m?2 s?1 and for the microalgae at 400 μmol m?2 s?1. Red light at 400 μmol m?2 s?1 had the highest growth rate (0.41 d?1), biomass (913 mg L?1) and biomass productivity (95 mg L?1 d?1). Lipid content was similar between all light colors. Green light had the highest chlorophyll‐a content (1649 μg/L). These results can be used to control the species composition of mixed cultures while maintaining their productivity.  相似文献

Effects of deficient (20mmol m?3) and sufficient (1000 mmol m?3) magnesium (Mg) supply and of varied light intensity (100 μmol m?2 s?1 to 580 μmol m?2 s?1) on paraquat-dependent chlorophyll destruction in bean (Phaseolus vulgaris) plants grown in nutrient solution were studied over a 12-d period using leaf discs or intact primary leaves. Treatment of leaf discs with 10mmol m 3 paraquat for 15h caused severe chlorophyll loss, especially with increasing light intensity. This chlorophyll destruction by paraquat was very much higher in Mg-sufficient than Mg-deficient leaves. The occurrence of paraquat resistance in Mg deficient leaves was already apparent after 6d growth in nutrient solution, i.e. before any decrease in chlorophyll or growth by Mg deficiency was evident. Also, following foliar application of paraquat (10–140 mmol m?3) to intact plants, Mg-deficient plants were much more resistant to paraquat, even following longer exposure duration (72 h) and four to 14 times higher paraquat concentrations than those received by Mg sufficient plants. From experiments where exogenous scavengers of superoxide radical (O2.-), hydroxyl radical (OH·) and singlet oxygen (1O2) were applied to leaf discs, it appears that O2.-, and partly, OH· are the main O2 species which contribute to chlorophyll destruction by paraquat. The results demonstrate that Mg-deficient bean plants become highly resistant to O2.--mediated and light-induced paraquat injury. The mode of this paraquat resistance is attributed to well-known stimulative effects of Mg deficiency on O2.- and H2O2 scavenging enzymes and antioxidants.  相似文献

Intraneuronal Site of Action for Imipramine in Rat Striatal Slices   总被引:2,自引:2,他引:0  
: The uptake of 3H-labeled imipramine ([3H]IMI) in rat corpus striatum slices was found to be strongly temperature-dependent. The accumulation was shown to be saturable and two apparent Km's were found: 2.2 × 10?8 and 3.5 × 10?7m . Once incorporated, the labeled drug was released from superfused slices by K+ (55 mm ) depolarization in the presence of calcium ions. Imipramine was also studied for its ability to induce the release of [3H]dopamine ([3H]DA) which had been previously accumulated by striatal slices. It was found that striatal slices superfused during 1 or 6 min with imipramine (10?6-10?4m ) release substantial amounts of radioactive dopamine, independently of the presence of Ca2+ in the medium. This release is completely abolished after reserpine pretreatment. It is proposed that imipramine enters the dopaminergic storage vesicles and displaces dopamine. An intraneuronal mechanism of action for imipramine is discussed.  相似文献
