Responses of neurons of the lateral and medial parts of the preoptic regionRPO (LPO andMPO, respectively) and the supraoptic nucleus (SO) of the hypothalamus to an increase in systemic blood pressure were studied in acute experiments on ketamine-anesthetized cats. The blood pressure increase resulted from the injection of a 0.002% phenylephrine hydrochloride solution in the femoral vien. Responses of theRPO neurons to suprathreshold pressor stimulation were monophasic activation (48% of units), monophasic inhibition (10%), or two-phase activational-inhibitory complex (14%). The rest of the neurons did not respond to the systemic pressure increase. Pressor stimulations evoked the responses only of the first and third type inMPO neurons, and only of the first and second type inLPO neurons, whileSO neurons generated responses only of the first type. Hysteresis of a varying degree was manifested in the changes of neuronal spike activity accompanying shifts of the systemic pressure. In some neurons there was no strict correlation between the pressor effect and the onset of neuronal response. Osmosensitivity of a part of neurons was tested. Summary Male Wistar-Kyoto rats were given either tap water (control) or 3%-alanine (taurine-depleted) for three weeks. To prepare for the kidney function studies, the animals were then implanted with femoral vessels and bladder catheters. Two days after surgery, each rat was given an intravenous infusion of saline at the rate of 50l/min and urine samples were collected at specific time intervals. An isotonic saline solution (0.9% NaCl) was infused for determination of baseline parameters and was followed by the infusion of a hypotonic saline solution (0.45% NaCl). Two days later, the infusion protocol was repeated in the same animals; however, a hypertonic saline solution (1.8% NaCl) was substituted for the hypotonic saline solution. Renal excretion of fluid and sodium increased in the control, but not taurine-depleted, rats during the hypotonic saline infusion. Interestingly, diuretic and natriuretic responses were similar between the groups during hypertonic saline infusion. The results suggest that taurine-depletion in rats affects renal excretory responses to a hypotonic, but not a hypertonic, saline solution.  相似文献

Nociceptive responses were evoked in cats by electrical transcutaneous stimulation of the forepaw or electrical stimulation of respective brain structures; these responses could be modulated (intensified or suppressed) by combined electrical stimulation of different brain structures or by neurochemical influences upon these structures. Intensification of nociceptive responses was observed after stimulation of the noradrenergic orP-ergic systems localized in the ventral zone of the central gray (vl SGC) and the structures monosynaptically connected with the latter: the posterior and lateral hypothalamic nuclei (Hp andHl) and preoptic region (RPO). Similar effects were induced by suppression of the serotoninergic system concentrated within the dorsolateral central gray (dl SGC), dorsal raphe nucleus (Rd), and closely related structures: the ventromedial, dorsomedial, and paraventricular hypothalamic nuclei (Hvm, Hdm, andHpv), septum (Sep), basolateral amygdalar nucleus (Am bl), fields 3–4 of the hippocampus (CA3–4), and cingular cortex (GC). Suppression of the serotoninergic system resulted in a decrease in the levels of functioning of the met-enkephalin- and β-endorphinergic systems and facilitation of theP-ergic system. Moderation of nociceptive responses, i.e., an analgesic effect, was observed after either stimulation of the serotonin-, met-enkephalin-, and β-endorphinergic systems localized in thedl SGC, Rd, Hvm, Hdm, Sep, Am bl, CA3–4, andGC, or suppression of the noradrenergic system. The latter influence resulted in inhibition of theP-ergic system and a rise in the functional activity of the met-enkephalin- and β-endorphinergic systems. The composition of two antagonistic brain systems, nociceptive and antinociceptive, is considered. The antinociceptive system includes serotonin-, met-enkephalin-, and β-endorphinergic elements. Leu-enkephalin is a nonspecific activator of the met-enkephalin-, β-endorphin-, andP-ergic systems. The nociceptive system consists of thevl SGC, Hp, Hl, andRPO, while the antinociceptive system includes thedl SGC, Rd, Hvm, Hdm, Hpv, Sep, Am dl, CA3–4, andGC.  相似文献

Motor activity and changes in body temperature related to the alternation of stages of an estrous cycle were studied in albino rats; groups of animals with the intact preoptic region (RPO), with electrolytically destroyedRPO, and with lesion of this structure followed by a transplantation of the embryonicRPO tissue were compared. The effect of NaCl concentration gradients on the growth of a marine species,Vibrio pelagius, and two freshwater and estuarine species,Vibrio cholerae andAeromonas hydrophila, was determined in chemostat cultures containing a mineral medium withd-glucose as the sole source of carbon and energy. Complementary cell density profiles were obtained with the marine and the freshwater-estuarine species at the extreme ends of the NaCl concentration gradients. At the lower NaCl concentrations,V. cholerae andA. hydrophila attained maximal cell densities, while no growth or submaximal cell yields were characteristic ofV. pelagius; the reverse was true at the higher NaCl concentrations. At the intermediate concentrations (100–400 mM) all three organisms were able to grow with cell densities ranging from 80%–100% of the maximum. Competition experiments betweenV. cholerae andV. pelagius within this region (100, 150, and 300 mM) indicated that the lower NaCl concentrations favoredV. cholerae while the higher concentrations favoredV. pelagius. The results suggest that the responses ofV. pelagius, V. cholerae, andA. hydrophila to different concentrations of NaCl are important in determining the environmental distribution of these species.  相似文献

Ability of the freshwater leech, Poecilobdella viridis to withstand osmotic changes was investigated by following the fluctuations of the body weight in tap-water and in different salt concentrations. The salinity tolerance limit (lethal salt concentration) of this leech was found to be 1.54% NaCl which equilibrates approximately 51.359% sea-water. There was a significant weight loss in P. viridis when kept in both, hypo- and hypertonic media. It is concluded that volume regulation (through weight changes) was slight in hypotonic media whereas in hypertonic media there was an incessant decline in body weight. Adaptive significance of these findings is discussed.  相似文献

Transpiration and water absorption rates, stomatal and cuticular resistances to water vapour diffusion, were measured onPlantago maritima (halophyte) andPlantago lanceolata (glycophyte) grown in the presence of NaCl or Na2SO4. Water absorption was reduced in the presence of Na2SO4 and transpiration rate was increased when NaCl was added to the nutrient solution. The glycophyte daily water balance was more disturbed than that of the halophyte in the presence of Na2SO4.  相似文献

The osmotic shock process for the release of periplasmic recombinant human interferon-α2b from Escherichia coli was optimized using response surface method (RSM). The process parameters such as pH, buffer concentration and sucrose concentration in hypertonic solution, cell concentration to hypertonic solution, contact time of cells with hypertonic solution, temperature of hypertonic solution, cell concentration to hypotonic solution, contact time of cells with hypotonic solution and temperature of hypotonic solution were initially screened using Plackett Burman design. Further optimization was carried out using central composite design (one of the design in RSM) for sucrose concentration in hypertonic solution as well as cell concentration to hypertonic and hypotonic solutions. The optimal cell concentration was 0.05 g/mL in hypertonic solution and 0.2 g/mL in hypotonic solution. The use of hypertonic solution containing 18% sucrose with a combination of 100 mM Tris and 2.5 mM EDTA buffer (pH 8.0 and 25 °C) and cold water (4 °C) as a hypotonic solution gave the optimum release of interferon-α2b. Increased product concentration in the final solution resulted from the optimized process would reduce the downstream steps during purification. The concept of reuse of hypertonic solution was also demonstrated.  相似文献

In acute experiments on cats we studied convergence of the afferent temperature-related and glycemic signals on the neurons of the preoptic region (RPO1). Local heating and cooling (±7°C) of the skin on the contralateral forelimb pad were used for identification ofRPO neurons as thermosensitive units, while infusion of 0.1–0.4 ml of 5.5% glucose solution in the ipsilaterala. carotis revealed their sensitivity to shifts in the glucose concentration. More than half (57%) of glucosensitiveRPO neurons responded to the changes in peripheral temperature. The pattern of convergence and peculiarities of localization of the convergent neurons were studied. We propose the following hypothesis:RPO neurons receiving convergent thermal and glycemic inputs provide formation of integral controlling signals addressed to the ventromedial and lateral hypothalamic nuclei, which control the thermoregulatory food consumption.  相似文献

The tribePlucheeae (Benth.)A. Anderb., has been analysed cladistically by means of a computerized parsimony program (Hennig 86), using theArctotideae as outgroup. The results of the analysis are presented in a consensus tree and one cladogram. Four major monophyletic subgroups can be recognized: TheColeocoma group (3 genera), thePterocaulon group (3 genera), theLaggera group (6 genera), and thePluchea group (12 genera). All recognized genera are described and most genera are supplied with taxonomical notes including comments on their taxonomic status. Genera such asBlumea, Pluchea, andEpaltes are demonstrated to be unnatural assemblages.Monarrhenus andTessaria are both closely related to thePluchea complex. The old generic nameLitogyne Harv. has been taken up for one species ofEpaltes, the genusRhodogeron is reduced to a synonym ofSachsia, and the following new combinations are made;Litogyne gariepina (DC.)A. Acute experiments on cats showed that mild shifts in osmotic homeostasis (within physiological limits) result in modifications of the spike activity in a significant share of thermosensitive neurons of the Preoptic region (RPO); in particular, rhythmic aftereffects (RAE) are induced in these units. The neurons were identified as thermosensitive cells according to their responses to local heating/cooling (±7°C) of the pad surface of the contralateral forelimb. Osmotic stimulation was performed by infusions of 0.1–0.3 ml of 3.0 or 0.2% NaCl solutions in thea. carotis communis dextra. The RAE phenomenon more frequently developed in thermosensitive neurons after infusions of the hyportonic solution (13/34) than after hypotonic infusions (6/34), while in non-thermosensitive neurons the situation was reverse: hypotonic infusions induced RAE more frequently. Certain specificities were also observed in localization of the thermosensitive RPO neurons, in which osmotic stimulation-induced RAE was observed. The mechanisms of such induction and possible functional role of spike grouping in the activity of thermosensitive neurons are discussed.  相似文献

The phylogeny of the subfamilyRubioideae (Rubiaceae) was estimated from sequence variation in therps16 intron (cpDNA) in 143 ingroup and 5 outgroup taxa. The analysis largely confirms a recent one based onrbcL sequences, but branch support is often much stronger. Three of the traditional subfamilies are supported,Rubioideae, Cinchonoideae s. str., andIxoroideae s. l. while there is no support forAntirheoideae. TheRubioideae are the sister group of all otherRubiaceae and comprise the tribesAnthospermeae, Coccocypseleae, Cruckshanksieae, Coussareeae, Gaertnereae, Hedyotideae, Knoxieae, Morindeae, Ophiorrhizeae, Paederieae, Pauridiantheae, Perameae, Psychotrieae, Rubieae, Spermacoceae, Theligoneae, andUrophylleae. TheHamelieae andHillieae belong to theCinchonoideae. Rachicallis andSiemensia should be transferred from theHedyotideae to theCinchonoideae. ThePauridiantheae, Urophylleae, Ophiorrhizeae, andRaritebe form the basalmost subclade of theRubioideae. The second basalmost clade consists of the generaLasianthus andPerama. The third basalmost clade consists of the tribesCoussareeae, Coccocypseleae andCruckshanksieae, and the generaDeclieuxia andHindsia. The tribesKnoxieae, Anthospermeae, Argostemmateae, Paederieae, Theligoneae, Rubieae, Hedyotideae, andSpermacoceae are members of one clade. TheKnoxieae are monophyletic ifOtiophora, Otomeria, andPentas are included. The tribeAnthospermeae is supported as monophyletic, but its subtribes are not. ThePaederieae, together withTheligonum, form a paraphyletic grade basal to theRubieae. TheHedyotideae, includingSchismatoclada, form a grade at the base of theSpermacoceae. TheGaertnereae are monophyletic and distinct from thePsychotrieae. TheMorindeae are monophyletic and includeDamnacanthus andMitchella. Schradera is the sister group of theMorindeae. ThePsychotrieae are monophyletic when theGaertnereae, Lasianthus, andDeclieuxia are excluded. The recognition of a subtribeHydnophytineae leaves the rest of thePsychotrieae paraphyletic.Psychotria is paraphyletic with respect to all other genera of the tribe. Approximately 50 genera are here classified for the first time based on molecular data.  相似文献

Basil (Ocimum basilicum L., cultivar Genovese) plants were grown in Hoagland solution with or without 50 mM NaCl or 25 mM Na2SO4. After 15 days of treatment, Na2SO4 slowed growth of plants as indicated by root, stem and leaf dry weight, root length, shoot height and leaf area, and the effects were major of those induced by NaCl. Photosynthetic response was decreased more by chloride salinity than by sulphate. No effects in both treatments on leaf chlorophyll content, maximal efficiency of PSII photochemistry (F v/F m) and electron transport rate (ETR) were recorded. Therefore, an excess of energy following the limitation to CO2 photoassimilation and a down regulation of PSII photochemistry was monitored under NaCl, which displays mechanisms that play a role in avoiding PSII photodamage able to dissipate this excess energy. Ionic composition (Na+, K+, Ca2+, and Mg2+) was affected to the same extent under both types of salinity, thus together with an increase in leaves Cl, and roots SO4 2− in NaCl and Na2SO4-treated plants, respectively, may have resulted in the observed growth retardation (for Na2SO4 treatment) and photosynthesis activity inhibition (for NaCl treatment), suggesting that those effects seem to have been due to the anionic component of the salts.  相似文献

The effects of inhibitors [acetazolamide, an inhibitor of carbonic anhydrase; amiloride, an inhibitor of the Na channel; furosemide, an inhibitor of the Na/K/2Cl transporter; 4,4'-diisothiocyanostilbene-2,2'-disulfonic acid (DIDS), an inhibitor of the Cl channel] on the water response in the superior laryngeal nerve (SLN) were investigated using whole nerve recordings from the SLN of anesthetized and paralyzed rats. Changes in spontaneous activity in the SLN after i.v. injection of a hypo- or hypertonic solution were also investigated. The water response to higher concentration amiloride solutions (0.1, 1, 5 and 10 mM) were significantly smaller in comparison with the control, i.e. the water response to deionized water (88-59% of the control, Fisher's PLSD, P < 0.05). DIDS suppressed the water response significantly at concentrations of 0.5 and 2 mM by 18 and 33%, respectively (P < 0.05). Likewise, acetazolamide (2 mM) and furosemide (5 mM) significantly suppressed the water response by 9 and 40%, respectively (P < 0.05). An i.v. bolus injection of a hypertonic solution (1 ml of 1.5 M NaCl or 1.0 M mannitol) depressed spontaneous activity of the SLN. In contrast, an i.v. injection of a hypotonic solution (0.015 M NaCl) increased spontaneous activity. These results suggest that several ion transporters and ion channels, as well as carbonic anhydrase, that may exist in the dorsal surface in the epiglottis may regulate the water response in the SLN and that osmotic changes in the dorsal surface of the epiglottis and in the interstitial space can affect nerve activity in the SLN.  相似文献

The data presented in this paper characterize the immune responses in mice to the two related random terpolymers, poly(Glu57Lys38Tyr5) (GLT5) and poly (Glu55Lys34Tyr15) (GLT15), which are similar to the previously reported response against the terpolymer poly(Glu58Lys38Phe4) (GLØ). Responsiveness is linked to theH-2 d ,H-2 ja ,H-2 q andH-2 r haplotypes. The observed responsiveness of the recombinant strain B10.A(5R)tentatively maps theIr-GLT gene to the right of theIB subregion, i. e., in theIC subregion which codes for lymphocyte alloantigens. 1. Two clinical studies are reported which investigate: (1) the regulation of vasopressin release in moderate hypertensive subjects not under treatment compared to normotensives and, (2) the effects of antihypertensive treatment on vasopressin and on its osmoregulation in moderate hypertensives.2. In the first study two stimuli facilitating vasopressin release (active upright position and hypertonic saline infusion) and a stimulus inhibiting vasopressin release (hypotonic saline infusion) have been applied to 13 moderate essential hypertensives and 8 control normotensives. In the second study, limited to hypertensives, the effects on plasma vasopressin and other plasma and urine variables, of either acute (by clonidine, n = 6 or by sodium nitroprusside, n = 6) or chronic (antihypertensive treatment for 1 month, n = 8) blood pressure lowering, before and after the i.v. administration of a hypertonic NaCl solution, were investigated.3. Baseline plasma vasopressin was not different in hypertensive and in normotensive subjects. Upright posture and hypertonic challenge augmented, while hypotonic saline reduced plasma vasopressin levels with no difference between the two groups. Acute, but not chronic, lowering of blood pressure increased plasma vasopressin from 1.6 ± 0.63 to 3.4 ± 0.7 pg/mL (p < 0.05); administration of hypertonic saline further increased vasopressin to 10.8 ± 2.22 (p < 0.01) in the acute and to 6.0 ± 1.03 pg/mL (p < 0.01) in the chronic study.4. No significant alterations of the regulation of vasopressin have been found in moderate, uncomplicated hypertension. Moreover, acute lowering of blood pressure facilitated the release of vasopressin and its osmoregulation while a chronic antihypertensive treatment did not interfere with a normal control of vasopressin secretion.  相似文献

A phylogenetic analysis of theUlmaceae, Cannabaceae, Barbeyaceae, andBroussonetia of theMoraceae produced nine equally parsimonious trees with 127 steps. TheUlmoideae (Ulmaceae, sensuGrudzinskaya) are a monophyletic group and distinct from theCeltidoideae. The genusAmpelocera occupies an isolated taxonomic position among the celtidoids. The similarity ofAmpelocera to the fossil celtidoid flowerEoceltis of North America suggests thatAmpelocera posesses an archaic suite of characters, and occupies a primitive position among the celtidoids, theCannabaceae and theMoraceae. The relationships among the other celtidoid taxa,Cannabaceae, andBroussonetia are problematic. TheCannabaceae andBroussonetia of theMoraceae are nested within the celtidoids suggesting that this is a paraphyletic group. The close, but unresolved, relationship of the celtidoids to theMoraceae andCannabaceae observed in this analysis, and the appearance of the celtidoids in the fossil record prior to the ulmoids suggests that the evolutionary relationship of the ulmoids and celtidoids may be more distant than current taxonomic treatments reflect.  相似文献
