Inotropic effects of yttrium acetate (Y3+) on contractions of myocardium preparations of the frog Rana ridibunda, as well as on respiration and the inner membrane potential (ΔΨmito) of isolated rat heart mitochondria were studied. 2 mM yttrium in Ringer solution was found to significantly reduce the amplitude of myocardium contractions, evoked by electric stimulation, and increase the half-relaxation time (n = 5). In experiments with Ca2+, Y3+ decreased the Ca2+-dependent basal respiration rate in rat heart mitochondria, energized by glutamate and malate, impeded the reduction in respiration of these mitochondria operating in state 3 after Chance or uncoupled by 2,4-dinitrophenol, and inhibited a Ca2+-induced reduction in their inner membrane potential. The data obtained are important for better understanding the mechanism underlying Y3+ effects on the myocardial Ca2+-dependent processes. Possible mechanisms of the negative inotropic effect of Y3+ on myocardium and its influence on the Ca2+-dependent processes in rat mitochondria are discussed.  相似文献

The redox states of cytochrome a 3 in wheat leaves (Triticum aestivum L.), in light and in the dark were monitored by its reaction with CO, which resulted in the stimulation of the in vivo reduction of nitrate to nitrite by nitrate reductase under aerobic conditions. Illumination of the leaves for 10 min markedly stimulated the steady state reduction of cytochrome a 3, probably associated with a decreased energization of the mitochondria in light. In the dark, during steady state respiration of the tightly coupled mitochondria, cytochrome oxidase was in a more oxidised state than in the light, as judged by its reaction with CO. It is also likely that in light, intra mitochondrial NAD+ will be highly reduced on account of a high phosphate potential.  相似文献

The paper considers the effects of bedaquiline (BDQ), an antituberculous preparation of the new generation, on rat liver mitochondria. It was shown that 50?μM BDQ inhibited mitochondrial respiration measured with substrates of complexes I and II (glutamate/malate and succinate/rotenone systems respectively) in the states V3 and VDNP. At the same time, at concentrations below 50?μM, BDQ slightly stimulated respiration with substrates of complex I in the state V2. BDQ was also found to suppress, in a dose-dependent manner, the activity of complex II and the total activity of complexes II?+?III of the mitochondrial transport chain. It was discovered that at concentrations up to 10?μM, BDQ inhibited H2O2 production in mitochondria. BDQ (10–50?μM) suppressed the opening of Ca2+-dependent CsA-sensitive mitochondrial permeability transition pore. The latter was revealed experimentally as the inhibition of Ca2+/Pi-dependent swelling of mitochondria, suppression of cytochrome c release, and an increase in the Ca2+ capacity of the organelles. BDQ also decreased the rate of mitochondrial energy-dependent K+ transport, which was evaluated by the energy-dependent swelling of mitochondria in a K+ buffer and DNP-induced K+ efflux from the organelles. The possible mechanisms of BDQ effect of rat liver mitochondria are discussed.  相似文献

Lung mitochondria were isolated by differential centrifugation from pentobarbital-anesthetized male rats. One to three millimolar Mg2+-ATP increased the consumption of oxygen of lung mitochondria oxidizing 10 mM succinate > fourfold (P < 0.01) whereas ATP increased the respiration of liver mitochondria by < 35%. ATP also hyperpolarized partially uncoupled lung mitochondria in the presence of the mitochondria-specific antagonist, oligomycin. However, only 20% of the ATPase activity in the lung mitochondria was blocked by oligomycin compared to a blockade of 91% for liver mitochondria. We investigated the effect of reducing the non-mitochondrial ATPase activity in the lung preparation. A purer suspension of lung mitochondria from a Percoll gradient was inhibited 95% by oligomycin. The volume fraction identified as mitochondria by electron microscopy in this suspension (73.6± 3.5%) did not differ from that for liver mitochondria (69.1± 4.9%). ATP reduced the mean area of the mitochondrial profiles in this Percoll fraction by 15% (P <0.01) and increased its state 3 respiration with succinate as substrate by 1.5-fold (P < 0.01) with no change in the state 4 respiration measured after carboxyatractyloside. Hence, ATP increased the respiratory control ratio (state 3/state 4, P <0.01). In contrast, state 3 respiration with the complex 1-selective substrates, glutamate and malate, did not change with addition of ATP. The acceleration of respiration by ATP was accompanied by decreased production of H2O2. Thus ATP-dependent processes that increase respiration appear to improve lung mitochondrial function while minimizing the release of reactive oxygen species.  相似文献

It is known that permeability of the inner mitochondrial membrane is low to most univalent cations (K+, Na+, H+) but high to Tl+. Swelling, state 4, state 3, and 2,4-dinitrophenol (DNP)-stimulated respiration as well as the membrane potential (ΔΨmito) of rat liver mitochondria were studied in media containing 0–75 mM TlNO3 either with 250 mM sucrose or with 125 mM nitrate salts of other monovalent cations (KNO3, or NaNO3, or NH4NO3). Tl+ increased permeability of the inner mitochondrial membrane to K+, Na+, and H+, that was manifested as stimulation of the swelling of nonenergized and energized mitochondria as well as via an increase of state 4 and dissipation of ΔΨmito. These effects of Tl+ increased in the order of sucrose <K+ <Na+ ≤ NH4+. They were stimulated by inorganic phosphate and decreased by ADP, Mg2+, and cyclosporine A. Contraction of energized mitochondria, swollen in the nitrate media, was markedly inhibited by quinine. It suggests participation of the mitochondrial K+/H+ exchanger in extruding of Tl+-induced excess of univalent cations from the mitochondrial matrix. It is discussed that Tl+ (like Cd2+ and other heavy metals) increases the ion permeability of the inner membrane of mitochondria regardless of their energization and stimulates the mitochondrial permeability transition pore in low conductance state. The observed decrease of state 3 and DNP-stimulated respiration in the nitrate media resulted from the mitochondrial swelling rather than from an inhibition of respiratory enzymes as is the case with the bivalent heavy metals.  相似文献

In order to clarify the relationship between chill-induced disturbance in photosynthetic, respiratory electron transport and the metabolism of reactive oxygen species (ROS), leaf gas exchange, chlorophyll fluorescence quenching, respiration, and activities of superoxide dismutase (SOD) and ascorbate peroxidase (APX) were investigated in chloroplasts and mitochondria of cucumber (Cucumis sativus) leaves subjected to a chill (8 °C) for 4 d. Chilling decreased net photosynthetic rate (P N) and quantum efficiency of photosystem 2 (ΦPS2), but increased the ratio of ΦPS2 to the quantum efficiency of CO2 fixation (ΦCO2) and non-photochemical quenching (NPQ) in cucumber leaves. While chilling inhibited the activity of cytochrome respiration pathway, it induced an increase of alternative respiration pathway activity and the reduction level of Q-pool. Chilling also significantly increased O2 production rate, H2O2 content, and SOD and APX activities in chloroplasts and mitochondria. There was a more significant increase in SOD and APX activities in chloroplasts than in mitochondria with the increase of membrane-bound Fe-SOD and tAPX in chloroplasts being more significant than other isoenzymes. Taken together, chilling inhibited P N and cytochrome respiratory pathway but enhanced the photosynthetic electron flux to O2 and over-reduction of respiratory electron transport chain, resulting in ROS accumulation in cucumber leaves. Meanwhile, chilling resulted in an enhancement of the protective mechanisms such as thermal dissipation, alternative respiratory pathway, and ROS-scavenging mechanisms (SODs and APXs) in chloroplasts and mitochondria.  相似文献

Summary Rapid uptake of Ba2+ by respiring rat liver mitochondria is accompanied by a transient stimulation of respiration. Following accumulation of Ba2+, e.g. at a concentration of 120 nmol per mg protein, the mitochondria exhibit reduced rates of state 3 and uncoupler-stimulated respiration. ADP-stimulated respiration is inhibited at a lower concentration of Ba2+ than is required to affect uncoupler-stimulated respiration, suggesting a distinct effect of Ba2+ on mechanisms involved in synthesis of ATP. Ba2+, which has an ionic radius similar to that of K+, inhibits unidirectional K+ flux into respiring rat liver mitochondria. This effect on K+ influx is observable at concentrations of Ba2+, e.g. 23 to 37 nmol per mg protein, which cause no significant change in state 4 or uncoupler-stimulated respiration. The accumulated Ba2+ decreases the measuredV max of K+ influx, while having little effect on the apparentK m for K+. The inhibition of K+ influx by Ba2+ is seen in the presence and absence of mersalyl, an activator of K+ influx. In contrast, under the conditions studied, Ba2+ has no apparent effet on the rate of unidirectional K+ efflux. These data are consistent with the idea that K+ may enter and leave mitochondria via spearate mechanisms.  相似文献

The aim of the present work was to investigate the mechanisms of oxidative damage of the liver mitochondria under diabetes and intoxication in rats as well as to evaluate the possibility of corrections of mitochondrial disorders by pharmacological doses of melatonin. The experimental (30 days) streptozotocin‐induced diabetes mellitus caused a significant damage of the respiratory activity in rat liver mitochondria. In the case of succinate as a respiratory substrate, the ADP‐stimulated respiration rate V3 considerably decreased (by 25%, p < 0·05) as well as the acceptor control ratio (ACR) V3/V2 markedly diminished (by 25%, p < 0·01). We observed a decrease of the ADP‐stimulated respiration rate V3 by 35% (p < 0·05), with glutamate as substrate. In this case, ACR also decreased (by 20%, p < 0·05). Surprisingly, the phosphorylation coefficient ADP/O did not change under diabetic liver damage. Acute rat carbon tetrachloride‐induced intoxication resulted in considerable decrease of the phosphorylation coefficient because of uncoupling of the oxidation and phosphorylation processes in the liver mitochondria. The melatonin administration during diabetes (10 mg·kg‐1 body weight, 30 days, daily) showed a considerable protective effect on the liver mitochondrial function, reversing the decreased respiration rate V3 and the diminished ACR to the control values both for succinate‐dependent respiration and for glutamate‐dependent respiration. The melatonin administration to intoxicated animals (10 mg·kg−1 body weight, three times) partially increased the rate of succinate‐dependent respiration coupled with phosphorylation. The impairment of mitochondrial respiratory plays a key role in the development of liver injury under diabetes and intoxication. Brandt's voles (Lasiopodomys brandti) exposed to cold (5±1 °C) or warm (23±1 °C) showed some physiological and biochemical variations which might be important in adaptation to their environments. Cold acclimation induced increases in resting metabolic rate (RMR) and the serum triiodothyronine (T3) level, the state-4 respiration of liver and muscle mitochondria were activated after 7 days when animals exposed to cold, and the activity of cytochrome c oxidase (COX) of liver and muscle mitochondria tended to rise with cold exposure. RMR and T3 level decreased during warm acclimation. The state-4 respiration of liver mitochondria declined after 3 days and muscle after 7 days when animals exposed to warm, and the activities of COX of liver and muscle mitochondria tended to decrease with warm acclimation. The cold activation of liver and muscle mitochondrial respiration (regulated by T3) was one of the cytological mechanisms of elevating RMR. Both state-4 respiration and COX activity of brown adipose tissue (BAT) mitochondria increased significantly during cold acclimation and decreased markedly after acclimated to warm. The uncoupling protein 1 (UCP1) contents in BAT increased after exposure to cold and decreased after warm acclimation. Nonshivering thermogenesis (NST) plays an important role in the process of thermoregulation under cold acclimation for Brandt's voles. Changes in thermogenesis is a important way to cold adaptation for Brandt's voles in natural environments.  相似文献

The effects of oligomycin on photosynthesis and respiration in relation to ATP production in chloroplasts and mitochondria were investigated in protoplasts isolated from the detached pea (Pisum sativum L cv. Iłowiecki.) and barley (Hordeum vulgare L. cv. Gunilla) leaves treated 5 mM Pb(NO3)2. The oligomycin (OM), an inhibitor of oxidative phosphorylation at 0.1 μM concentration caused the inhibition of photosynthesis rate in the protoplasts from both the control and the Pb-treated pea leaves. The respiration rate and ATP/ADP ratio in the protoplasts and the activity of ATPase in mitochondria, were also diminished in the control protoplasts. These effects were not observed in the protoplasts and mitochondria isolated from the Pb-treated leaves. Oligomycin, an inhibitor of photophosphorylation at 10 μM concentration decreased ATPase activity in chloroplasts from both the control and the Pb- treated leaves. Using the method of rapid fractionation of barley protoplasts it was shown that the ATP/ADP ratio in the mitochondria from Pb-treated leaves was largely suppressed (from 1.8 to 0.4) by OM under nonphotorespiratory conditions (high CO2), whereas under photorespiratory conditions (low CO2) this ratio was high (5.3) and under OM decreased less (to 3.1). Our results indicate that oligomycin, in organelle isolated from Pb-treated leaves, had no inhibitory effect on the mitochondrial ATPase, whereas it inhibited chloroplasts ATPase. We suggest that Pb ions affected the catalytic cycle and/or conformational changes of ATPase in pea chloroplasts differently than in mitochondria. The differences in Pb responses may reflect fine mechanisms for the regulation of ATP production in the plant cells under stress conditions.  相似文献

Root respiration associated with symbiotic fixation in soybean (Glycine max [L.] Merr.) was estimated by four methods.

Averaged over the life of the plant, the root respires 5.8 milligrams C per milligram N accumulated from fixation. When nitrogenase (C2H2) activity and root respiration were decreased by treating roots briefly with 1.0 atmosphere O2, the respiration associated with nitrogenase was estimated as 2.10 micromoles CO2 per micromole C2H4.

When nitrogenase activity and respiration were decreased by addition of nitrate, the respiration associated with fixation was calculated as 2.90 micromoles CO2 per micromole C2H4. Removing nodules from roots decreased fixation and root respiration, and the ratio was 4.08 micromoles CO2 per micromole C2H4. When soybean plants were kept in prolonged darkness, then returned to light, the associated drop and recovery of respiration and nitrogenase activity had a ratio of 4.36 micromoles CO2 per micromole C2H2.

Effects of Tl+ were studied in experiments with isolated rat heart mitochondria (RHM) injected into 400 mOsm medium containing TlNO3 and a nitrate salt (KNO3 or NH4NO3) or TlNO3 and sucrose. Tl+ increased permeability of the inner membrane of the RHM to K+ and H+. This manifested as an increase of the non-energized RHM swelling, in the order of sucrose < K+ < NH4 +, respectively. After succinate administration, the swollen RHM contracted. The Tl+-induced opening of the mitochondrial permeability pore (MPTP) in Ca2+-loaded rat heart mitochondria increased both the swelling and the inner membrane potential dissipation, as well as decreased basal state and 2,4-dinitrophenol-stimulated respiration. These effects of Tl+ were suppressed by the MPTP inhibitors (cyclosporine A, ADP, bongkrekic acid, and n-ethylmaleimide), activated in the presence of the MPTP inducer (carboxyatractyloside) or mitoKATP inhibitor (5-hydroxydecanoate), but were not altered in the presence of mitoKATP agonists (diazoxide or pinacidil). We suggest that the greater sensitivity of heart and striated muscles, versus liver, to thallium salts in vivo can result in more vigorous Tl+ effects on muscle cell mitochondria.  相似文献

The effects of streptozotocin-induced diabetes mellitus upon mitochondria from rat skeletal muscle and kidney were examined. The rate of amino acid incorporation in vitro by isolated skeletal muscle mitochondria from diabetic animals was decreased by 50–60% from control values. Treatment of diabetic animals with insulin lowered blood glucose levels to control values and restored the rate of muscle mitochondrial protein synthesis in vitro to control levels. The rates of skeletal muscle mitochondrial protein synthesis were also decreased 23–27% by a 2-day fast. Comparison of the translation products synthesized by isolated muscle mitochondria from control and diabetic rats by dodecyl sulfate polyacrylamide-gel electrophoresis revealed a uniform decrease in the synthesis of all polypeptides. Aurintricarboxylic acid and pactamycin, inhibitors of chain initiation, blocked protein synthesis to a greater extent in muscle mitochondria from control as compared to diabetic animals suggesting that mitochondria from diabetics are unable to initiate protein synthesis at a rate comparable to control. Phenotypic changes observed in diabetic muscle mitochondria included a 36% decrease in the content of cytochromes aa3 and a 27% decrease in cytochrome b, both established as containing mitochondrial translation products in lower eucaryotes. State 3 respiration with glutamate as substrate decreased by 27% and uncoupler-stimulated respiration decreased by 23% in the diabetic mitochondria. By contrast, the specific activities of NADH and succinate dehydrogenases, established as products of cytoplasmic protein synthesis in lower eucaryotes, were not decreased in skeletal muscle mitochondria from the diabetic animals. These results suggest that the considerable muscular atrophy observed in diabetics may involve decreases in both cytoplasmic and mitochondrial protein synthesis, the latter reflected in profound changes in the respiratory chain. By contrast, comparison of kidney mitochondria from control and diabetic rats revealed no differences in the rates of protein synthesis in vitro, nor in the mitochondrial translation products, which corresponded closely to liver and skeletal muscle translation products. Similarly, the mitochondrial content of cytochromes b, c + c1, and aa3, the specific activity of succinate dehydrogenase, the rate of state 3 respiration, and the recovery of mitochondria from kidney homogenates did not differ in control and diabetic animals. Kidney mitochondria are thus like liver mitochondria in being relatively unaffected by insulin deprivation.  相似文献

Mitochondrial reactive oxygen species are implicated in the etiology of multiple neurodegenerative diseases, including Parkinson disease. Mitochondria are known to be net producers of ROS, but recently we have shown that brain mitochondria can consume mitochondrial hydrogen peroxide (H2O2) in a respiration-dependent manner predominantly by the thioredoxin/peroxiredoxin system. Here, we sought to determine the mechanism linking mitochondrial respiration with H2O2 catabolism in brain mitochondria and dopaminergic cells. We hypothesized that nicotinamide nucleotide transhydrogenase (Nnt), which utilizes the proton gradient to generate NADPH from NADH and NADP+, provides the link between mitochondrial respiration and H2O2 detoxification through the thioredoxin/peroxiredoxin system. Pharmacological inhibition of Nnt in isolated brain mitochondria significantly decreased their ability to consume H2O2 in the presence, but not absence, of respiration substrates. Nnt inhibition in liver mitochondria, which do not require substrates to detoxify H2O2, had no effect. Pharmacological inhibition or lentiviral knockdown of Nnt in N27 dopaminergic cells (a) decreased H2O2 catabolism, (b) decreased NADPH and increased NADP+ levels, and (c) decreased basal, spare, and maximal mitochondrial oxygen consumption rates. Nnt-deficient cells possessed higher levels of oxidized mitochondrial Prx, which rendered them more susceptible to steady-state increases in H2O2 and cell death following exposure to subtoxic levels of paraquat. These data implicate Nnt as the critical link between the metabolic and H2O2 antioxidant function in brain mitochondria and suggests Nnt as a potential therapeutic target to improve the redox balance in conditions of oxidative stress associated with neurodegenerative diseases.  相似文献

The supply of nitrate nitrogen caused a marked increase in the rate of respiration of Japanese mint. Sodium azide strongly inhibited the rate of respiration at all the nitrate levels, but the inhibition was more marked at higher levels. Besides this, inhibition caused by azide treatment was less marked in older and nitrogen deficient tissues than in younger ones at higher nitrogen levels. The addition of sodium diethyl dithio-carbamate (DIECA), an inhibitor of copper containing enzymes resulted in an increase in the respiration of mint leaves which increased further with an increase in the nitrate supply. The same concentration of DIECA which stimulate the respiration of leaves caused an inhibition in the respiration of roots. The inhibition was greater at lower levels and decreased consistently as the supply of nitrate increased. The sensitivity of root respiration to DIECA observed with varying levels of nitrate indicated that unlike the leaf, the roots contain copper-containing enzymes which get decreased as the nitrate supply is increased. An increased supply of nitrogen up to 16 me NO3 - -N was associated with an increase in respiratory quotient.  相似文献

A series of nine complexes of uranyl nitrate with some Schiff bases derived from 4-aminoantipyrine and certain carbonyl compounds, such as benzaldehyde, 2-nitrobenzaldehyde, 3-nitrobenzaldehyde, 4- methylbenzaldehyde, 4-N,N-dimethylaminobenzaldehyde, 2-hydroxybenzaldehyde, 2-hydroxy-1-naphthaldehyde, acetylacetone and benzoylacetone have been synthesized. These complexes have been characterized by elemental analysis, molecular weight determination and IR spectral, conductance and magnetic studies. From these studies they can be formulated as [UO2L2(NO3)2], in which the first five ligands (in the order given above) and nitrate ions are coordinated bidentately, while the last four ligands (which have either a phenolic hydroxyl group or a side-chain carbonyl group as an additional site) act as terdentate ligands, and nitrate ions are coordinated monodentately. Hence the proposed general formula for the complexes suggests that the uranyl ion has a coordination number of eight in addition to the two oxygen atoms which have already been bonded to the U(VI) species.  相似文献

The substrate-dependent O2 uptake by sycamore (Acer pseudoplatanus L.) cell mitochondria in the presence of ADP and limiting Pi concentrations has been measured. The Pi concentration for half-maximum O2 uptake rate was found to be in the range 20 to 50 micromolar for all the substrates tested. 31P NMR of intact sycamore cells indicated that the Pi concentration in the cytoplasm was in the range 5 to 6 millimolar, approximately 100-fold higher than the Pi concentration required for maximum O2 uptake rates by isolated mitochondria. When sycamore cells were transferred to a culture medium devoid of Pi, the cytoplasmic Pi concentration decreased from 6 to less than 3 millimolar, but the intact cell respiration remained practically constant for at least 4 days. These results strongly suggest that, in vivo, the respiration rate of sycamore cells is not limited by the quantity of Pi supplied to the mitochondria.  相似文献

The construction and operation of a device for continuous measurement of CO2 release by suspensions of respiring mitochondria is described. A combination of this device with a Clark-type O2 electrode was used for simultaneous measurement of respiration and of CO2 release by spinach and pea leaf mitochondria with glycine as substrate. Both mitochondrial preparations showed high rates of respiration and high respiratory control ratios. The addition of oxaloacetate not only inhibited O2 uptake substantially, but also greatly stimulated glycine oxidation as monitored by CO2 release. In spinach leaf mitochondria, the maximal rates of glycine oxidation thus obtained, were two times higher than the rate of glycine oxidation required at average rates of photorespiration. It is concluded from these results that under saturating conditions the capacity of glycine oxidation by intact mitochondria exceeds the capacity of glycine-dependent respiration.  相似文献

Myoglobin (Mb) is reported in biochemistry and physiology textbooks to act as an O2 reservoir and to facilitate O2 diffusion from capillaries to mitochondria, to sustain cellular respiration. Recently, it has been proposed that Mb is an intracellular scavenger of bioactive nitric oxide (NO), regulating its level in the skeletal and cardiac muscle and thereby protecting mitochondrial respiration, which is impaired by NO. This novel function of Mb is based on the rapid and irreversible reaction of ferrous oxygenated Mb (MbO2) with NO yielding ferric oxidized Mb (metMb) and nitrate (NO3). The efficiency of this process, which is postulated to depend on the superoxide (O2) character acquired by O2 once bound to the heme iron, may be enhanced by intramolecular diffusion of NO trapped momentarily into cavities of the protein matrix. O2 can also react with ferrous nitrosylated Mb (MbNO), albeit very slowly, leading to metMb and NO3. The O2-dependent NO-detoxification process may be considered to be pseudo-enzymatic given that metMb obtained by the primary reaction of MbO2 with NO is reduced back to ferrous Mb by a specific metMb-reductase, and can therefore repeat a cycle of NO conversion to harmless nitrate.  相似文献

The Tl+-induced opening of the MPTP in Ca2+-loaded rat liver mitochondria energized by respiration on the substrates succinate or glutamate plus malate was recorded as increased swelling and dissipation of mitochondrial membrane potential as well as decreased state 4, or state 3, or 2,4-dinitrophenol-stimulated respiration. These effects of Tl+ increased in nitrate media containing monovalent cations in the order of Li+ < NH4+ ≤ Na+ < K+. They were potentiated by inorganic phosphate and diminished by the MPTP inhibitors (ADP, CsA, Mg2+, Li+, rotenone, EGTA, and ruthenium red) both individually and more potently in their combinations. Maximal swelling of both non-energized and energized Ca2+-loaded mitochondria in rotenone-free media is an indication of Ca2+ uptake driven by respiration on mitochondrial endogenous substrates. It is suggested that Tl+ (distinct from Cd2+, Hg2+, and other heavy metals and regardless of the used respiratory substrates) can stimulate opening of the MPTP only in the presence of Ca2+. We discuss the possible participation of Ca2+-binding sites, located near the respiratory complex I and the adenine nucleotide translocase, in inducing opening of the MPTP.  相似文献
