A simple method for preparation of D-rhamnose   总被引:1,自引:0,他引:1  
A rapid procedure for the preparation of D-rhamnose from bacterial lipopolysaccharide (LPS) has been developed. It involves purification of LPS from Pseudomonas syringae pv. phaseolicola by phenol extraction and hydrophobic interaction chromatography (HIC), followed by mild hydrolysis and cleavage of the O-antigen into D-fucose and D-rhamnose. The monosaccharides were separated by column chromatography, and D-rhamnose recovered after filtration over Sephadex-LH 20.  相似文献

A simple and quick method for the preparation of small unilamellar vesicles (SUV) was developed. SUV are spontaneously formed by swelling of the specially prepared phospholipid film in water/buffer. Normally, large multilamellar vesicles (MLV) are formed when a phospholipid film is dissolved in water. To prevent the formation of multilamellar structures we used the slightly charged phospholipids which exhibit infinite swelling while the formation of large structures was prevented by the deposition of the phospholipid film on the support with small surfaces. These two requirements were met by mixing a small amount of ionic detergent into phospholipid which was deposited on microcrystals. The size and size distribution of the produced vesicles depend on the size and homogeneity of the microcrystals. When 1.5 wt% of cetyltetramethylammonium bromide (CTAB) in egg yolk phosphatidylcholine was deposited on zeolite X microcrystals with crystallite sizes of approx. 0.4 μm a homogeneous population of vesicles with average diameter 21.5 nm was obtained.  相似文献

A simple and rapid method for preparing plasma membranes from isolated cells or tissues is described. The membranes were characterised (a) biochemically by an analysis of specific marker enzymes, (b) by quantitation of cell surface receptors, and (c) immunologically by their ability to elicit specific allogeneic responses from cytotoxic T cells in secondary in vitro stimulations. Based on both biochemical and immunologic criteria, plasma membranes prepared by the method described here are of equal or greater 'purity' compared to those prepared by two other methods that are most widely used to date and the yields are several-fold higher.  相似文献

A crude extract from hog submaxillary gland was found to synthesize guanosine diphosphate (GDP)-fucose, when incubated with fucose, ATP and GTP, the two enzymatic steps (fucose
fucose-1-phosphate
GDP-fucose) proceeding without noticeable side reactions. Column chromatography on Dowex 1 (HCO3?), gel filtration on Sephadex G-15 and preparative paper chromatography gave pure GDP-fucose in an overall yield of 81%. The sugar nucleotide was shown to be active as a glycosyl donor in fucose-incorporating systems.  相似文献

A new simple preparation method for NaK-ATPase-rich membrane fragments   总被引:3,自引:0,他引:3  
A method for the isolation of highly active membrane bound NaK-ATPase without detergents in quantity from the electric organ of the electric eel (Electrophorus electricus) is described. This method consists of the homogenation of electric organ with an isotonic solution containing sucrose, histidine, EDTA, and arginine, and of the separation of the higher active membrane fraction from the microsomal fraction by density gradient centrifugation. The enzyme has a specific activity of about 20 μmol Pi/min/mg at 37°C, and 13 μmol Pi/min/mg at 30°C. Although it is not as pure as the detergent-treated enzyme preparation based on the level of phosphorylated protein, ouabain binding, or sodium dodecyl sulfate-polyacrylamide gel electrophoresis, its enzyme activity is comparable to that of the purified enzymes. This preparation is very stable and is able to change its medium by Sephadex chromatography without any loss of enzyme activity and protein content. This preparation is also expected to keep the original characteristics as well as the enzyme in the tissue.  相似文献

A simple method for preparing bulk quantities of tRNA from chick embryo has been developed. In this method chick embryos were homogenized in a buffer of pH 4.5, followed by deproteinization with phenol. The aqueous layer was allowed to separate under gravity. The resulting aqueous layer, after two more phenol treatments, was directly passed through a DEAE-cellulose column and the tRNA eluted therefrom with 1 m NaCl. The tRNA prepared by this method was as active as the one prepared at neutral pH.  相似文献

A simple mechanical procedure that has been developed for the large-scale preparation of intact mitochondria from yeast, is also applicable to the extraction of organelles from other organisms having cell walls. A procedure for the isolation of large quantities of pure mitochondrial DNA from these mitochondria is described. In Schizosaccharomyces pombe, further purification of the mitochondria by urografin isopycnic centrifugation leads to 50% recovery of whole cell respiration activity in a vesicular fraction of respiratory chain enzymes, with NADH oxidase activity usually greater than 10 μmol of electrons/min/mg of protein. The method has the advantage of rapidity and low cost and it is extremely healthy for the operator.  相似文献
