Summary Restriction enzyme digestion of the ribosomal RNA genes of the nucleolar organisers of wheat has revealed fragment length polymorphisms for the nucleolar organiser on chromosome 1B and the nucleolar organiser on 6B. Variation between genotypes for these regions has also been demonstrated. This variation has been exploited to determine the recombination frequency between the physically defined nucleolar organiser on 1B (designatedNor1) and other markers; two loci,Glu-B1 andGli-B1 which code for endosperm storage proteins andRf3, a locus restoring fertility to male sterility conditioned byT. timopheevi cytoplasm.Gli-B1 andRf3 were located on the short-arm satellite but recombine with the nucleolar organiser giving a gene order ofNor1 — Rf3 — Gli-B1. Glu-B1 is located on the long arm of 1B but shows relatively little recombination withNor1, which is, in physical distance, distal on the short arm. This illustrates the discrepancy between map distance and physical distance on wheat chromosomes due to the distal localisation of chiasmata. The recombination betweenNor1 andRf3 indicates that, contrary to previous suggestions, fertility restoration is not a property of the nucleolar organiser but of a separate locus.  相似文献

Summary Four of 1,240 cultivated barley lines collected from different regions of the world and 3 of 120 lines of wild barley, Hordeum spontaneum C. Koch, carry spontaneous reciprocal translocations. Break-point positions and rearrangements in the interchanged chromosomes have been examined by both test crosses and Giemsa banding techniques. The four translocation lines in cultivated barley were all of Ethiopian origin and have the same translocation involving chromosomes 2 and 4. The breakpoints are at the centromeres of both chromosomes, resulting in interchanged chromosomes 2S+4S and 2L+4L (S=short arm, L=long arm). A wild barley line, Spont.II, also has translocated chromosomes 2 and 4 which are broken at the centromeres. The resultant chromosomes are, however, 2S+4L and 2L+4S. Another wild barley line, Spont.S-4, has interchanged chromosomes with breakpoints in the short arm of chromosome 3 and the long arm of chromosome 7. In addition, this line has a paracentric inversion in the short arm of chromosome 7 that includes a part of nucleolar constriction, resulting in two tandemly arranged nucleolar constrictions. The third wild barley line, Spont.S-7, has interchanged chromosomes with breakpoints in the long arms of both chromosomes 3 and 6. The translocated chromosome 3 is metacentric and the translocated chromosome 6 has a long arm similar in length to the long arm of chromosome 7.  相似文献

An Aegilops ventricosa Tausch (2n = 28, DvDvNvNv) subtelocentric chromosome added to wheat (Triticum aestivum L.) in a disomic addition line was found to carry the genes for resistance Yr17, Lr37, Sr38, and Cre5 already transferred onto chromosome 2AS of the wheat line VPM1. Previous works demonstrated that this Ae. ventricosa chromosome is translocated with respect to the standard wheat genome. The present investigations showed that this chromosome pre-existed in Ae. ventricosa and contains only chromatin specific to the N genome. Using biochemical markers and suitable cytogenetic materials including the monoisosomic addition line for the translocated long arm (6NvL-2NvS), its structure was defined as being 6NvSdel.6NvL-2NvS. It consists of a segment of the short arm 2Nv, containing the resistance genes, attached to a group 6 chromosome lacking a distal part of its short arm. The 2 re arrangements could already be present in Aegilops uniaristata Vis. (2n = 14, NN), the source of the Nv genome of Ae. ventricosa.  相似文献

Ten accessions of Russian wildrye, Psathyrostachys juncea (Fisch.) Nevski (2n = 2x = 14; NsNs), collected from different geographical regions were analyzed using the C-banding technique. C-banding pattern polymorphisms were observed at all levels, i.e., within homologous chromosome pairs of the same plant, among different individuals within accessions, between different accessions of the same geographic area, and among accessions of different origins. The seven homologous groups varied in the level of C-banding pattern polymorphism; chromosomes A, B, E, and F were more variable than chromosomes C, D, and G. The polymorphisms did not hamper chromosome identification in Ps. juncea, because each chromosome pair of the Ns genome had a different basic C-banding pattern and karyotypic character. A standard C-banded karyotype of Ps. juncea is proposed based on the overall karyotypes and C-bands in the 10 accessions. The C-bands on the Ns-genome chromosomes were designated according to the rules of nomenclature used in wheat. A deletion-translocation heterozygote of Russian wildrye was identified based on the karyotype and C-banding patterns established. The chromosome F pair consisted of a chromosome having the distal segment in the long arm deleted and a translocated chromosome having the distal segment of long arm replaced by the distal segment of the long arm of chromosome E. The chromosome E pair had a normal chromosome E and a translocated chromosome having the short arm and the proximal segment of the long arm of chromosome E and the distal segment of the long arm of chromosome F.  相似文献

Thinopyrum intermedium is a promising source of resistance to wheat streak mosaic virus (WSMV), a devastating disease of wheat. Three wheat germplasm lines possessing resistance to WSMV, derived from Triticum aestivum×Th. intermedium crosses, are analyzed by C-banding and genomic in situ hybridization (GISH) to determine the amount and location of alien chromatin in the transfer lines. Line CI15092 was confirmed as a disomic substitution line in which wheat chromosome 4A was replaced by Th. intermedium chromosome 4Ai?2. The other two lines, CI17766 and A29-13-3, carry an identical Robertsonian translocation chromosome in which the complete short arm of chromosome 4Ai?2 was transferred to the long arm of wheat chromosome 4A. Fluorescence in situ hybridization (FISH) using ABD genomic DNA from wheat as a probe and S genomic DNA from Pseudoroegneria stipifolia as the blocker, and vice versa, revealed that the entire short arm of the translocation was derived from the short arm of chromosome 4Ai?2 and the breakpoint was located at the centromere. Chromosomal arm ratios (L/S) of 2.12 in CI17766 and 2.15 in A29-13-3 showed that the translocated chromosome is submetacentric. Cytogenetic analysis of leukemic cells obtained at diagnosis from 122 patients with childhood acute lymphoblastic leukemia (ALL) disclosed chromosomal translocations in 36 cases. Two new nonrandom translocations were identified and found to be associated with specific immunophenotypes of the disease. The first, identified in 4 of 16 cases of T-cell ALL positive for sheep erythrocyte receptors (E+), involved the short arm (p) of chromosome 11 and the long arm (q) of chromosome 14 and was designated t(11;14) (p13;q13). The second, found in 7 of 23 cases with a pre-B-cell phenotype, involved the long arm of chromosome 1 and the short arm of chromosome 19; it was designated t(1;19) (q23;p13.3). A third abnormality involving a common breakpoint on chromosome 12 (band p 12) was also identified. These two new differentiation-specific translocations suggest a mechanism for aberrant expression of genes that influence lymphoid cell growth and development, as well as leukemogenesis.  相似文献

Summary One-dimensional sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) of reduced total protein extracts from the endosperm of hexaploid wheat revealed a new set of faintly-stained bands, having slower electrophoretic mobility than the high-molecular-weight (HMW) glutenin subunits. These new bands have been termed the E group of glutenin subunits. Analysis of aneuploid stocks of Chinese Spring wheat has shown that three of the E bands, in order of increasing electrophoretic mobility, are controlled by genes on the short arms of chromosomes 1B, 1A and 1D, respectively. The E bands are expressed only in the presence of the long arm of chromosome 1B indicating an interaction between two or more genes involved in their production in wheat endosperm. The gene on the short arm of chromosome 1D controlling an E subunit recombined freely with Tri-D1 and the centromere but not at all with Gli-D1, indicating additional complexity at the Gli-DI locus in wheat.  相似文献

The genes coding for the two classes of ribosomal RNA molecules, 5S RNA and 18+28S RNA, have been localized in the Norway rat (Rattus norvegicus). The 18+28S RNA cistrons are found on three chromosomes, at secondary constrictions on the short arms of chromosomes 3 and 12 and at the telomere of the short arm of chromosome 11. These sites were confirmed using the silver staining technique for nucleolar organizer regions. Two sites were found for the 5S RNA genes; one is closely linked to the 18+28S gene site on chromosome 12. The second site is at or near the telomere of the long arm of chromosome 19.  相似文献

Summary Using in situ hybridization techniques, we have been able to identify the translocated chromosomes resulting from whole arm interchanges between homoeologous chromosomes of wheat and rye. This was possible because radioactive probes are available which recognize specific sites of highly repeated sequence DNA in either rye or wheat chromosomes. The translocated chromosomes analysed in detail were found in plants from a breeding programme designed to substitute chromosome 2R of rye into commercial wheat cultivars. The distribution of rye highly repeated DNA sequences showed modified chromosomes in which (a) most of the telomeric heterochromatin of the short arm and (b) all of the telomeric heterochromatin of the long arm, had disappeared. Subsequent analyses of these chromosomes assaying for wheat highly repeated DNA sequences showed that in type (a), the entire short arm of 2R had been replaced by the short arm of wheat chromosome 2B and in (b), the long arm of 2R had been replaced by the long arm of 2B. The use of these probes has also allowed us to show that rye heterochromatin has little effect on the pairing of the translocated wheat arm to its wheat homologue during meiosis. We have also characterized the chromosomes resulting from a 1B-1R translocation event.From these results, we suggest that the observed loss of telomeric heterochromatin from rye chromosomes in wheat is commonly due to wheat-rye chromosome translocations.  相似文献

An alloplasmic wheat line carrying a zebra chromosome z5A was isolated from the derivatives of an Elymus trachycaulus x Triticum aestivum cv Chinese Spring hybrid. Chromosome z5A was named zebra because of its striped genomic in situ hybridization pattern. z5A consists of four chromosome segments derived from E. trachycaulus and four chromosome segments, including the centromere, from wheat. The short arm of z5A paired with the telocentric chromosome 1HtS of E. trachycaulus and the long arm with the long arm of normal 5A. z5A also carried several genetic markers derived from 1HtS. Chromosome 1Ht was the only E. trachycaulus chromosome found in the sib plants of a previous generation from which z5A was derived. Monosomic 5A and telocentric chromosome 5AL were also found in most of the sib plants. The zebra chromosome most probably originated from spontaneous multiple translocations between chromosomes 5A and 1HtS or 5A and 1Ht.  相似文献

The 1BL.1RS translocations between wheat (Triticum aestivum L.) and rye (Secale cereale L.) are widely used in bread wheat breeding programs, but all modern wheat cultivars with the 1BL.1RS have shown genetic vulnerability due to one rye source – a German cultivar, Petkus. We have developed, a new 1BL.1RS wheat-rye translocation line from the backcross of the F1 hybrid of wheat cv. Olmil and rye cv. Paldanghomil, both cultivars from Korea. The GISH technique was applied to identify the presence of rye chromatin in 467 BC1F6 lines selected from 77 BC1F5 lines. Only one line, Yw62–11, showed wheat-rye translocated chromosomes, with a somatic chromosome number of 2n=42. C-banding patterns revealed that the translocated chromosome was 1BL.1RS, showing prominent bands in the terminal and sub-terminal regions of the short arm as well as in the centromeric region and terminal region of the long arm. This new 1BL.1RS translocation line formed 21 bivalents like common wheat at meiotic metaphase I, thereby showing complete homology. Received: 28 February 2001 / Accepted: 17 April 2001  相似文献

Wheat (Triticum aestivum L.) deletion (del) stocks are valuable tools for the physical mapping of molecular markers and genes to chromosome bins delineated by 2 adjacent deletion breakpoints. The wheat deletion stocks were produced by using gametocidal genes derived from related Aegilops species. Here, we report on the origin, structure, and behavior of a highly rearranged chromosome 1BS-4. The cytogenetic and molecular marker analyses suggest that 1BS-4 resulted from 2 breakpoints in the 1BS arm and 1 breakpoint in the 1BL arm. The distal segment from 1BS, except for a small deleted part, is translocated to the long arm. Cytologically, chromosome 1BS-4 is highly stable, but shows a unique meiotic pairing behavior. The short arm of 1BS-4 fails to pair with a normal 1BS arm because of lack of homology at the distal ends. The long arm of 1BS-4 only pairs with a normal 1BS arm within the distal region translocated from 1BS. Therefore, using the 1BS-4 deletion stock for physical mapping will result in the false allocation of molecular markers and genes proximal to the breakpoint of 1BS-4.  相似文献

Wheat scab (Fusarium Head Blight, FHB) is a destructive disease in the warm and humid wheat-growing areas of the world. Finding diverse sources of FHB resistance is critical for genetic diversity of resistance for wheat breeding programs. Leymus racemosus is a wild perennial relative of wheat and is highly resistant to FHB. Three wheat- L. racemosus disomic addition (DA) lines DA5Lr#1, DA7Lr#1 and DALr.7 resistant to FHB were used to develop wheat- L.racemosus translocation lines through irradiation and gametocidal gene-induced chromosome breakage. A total of nine wheat-alien translocation lines with wheat scab resistance were identified by chromosome C-banding, GISH, telosomic pairing and RFLP analyses. In line NAU614, the long arm of 5Lr#1 was translocated to wheat chromosome 6B. Four lines, NAU601, NAU615, NAU617, and NAU635, had a part of the short arm of 7Lr#1 transferred to different wheat chromosomes. Four other lines, NAU611, NAU634, NAU633, and NAU618, contained translocations involving Leymus chromosome Lr.7 and different wheat chromosomes. The resistance level of the translocation lines with a single alien chromosome segment was higher than the susceptible wheat parent Chinese Spring but lower than the alien resistant parent L. racemosus. At least three resistance genes in L. racemosus were identified. One was located on chromosome Lr.7, and two could be assigned to the long arm of 5Lr#1 and the short arm of 7Lr#1.  相似文献

Powdery mildew is one of the serious diseases of wheat (Triticum aestivum L., 2n = 6 × = 42, genomes AABBDD). Rye (Secale cereale L., 2n = 2 × = 14, genome RR) offers a rich reservoir of powdery mildew resistant genes for wheat breeding program. However, extensive use of these resistant genes may render them susceptible to new pathogen races because of co-evolution of host and pathogen. Therefore, the continuous exploration of new powdery mildew resistant genes is important to wheat breeding program. In the present study, we identified several wheat-rye addition lines from the progeny of T. aestivum L. Mianyang11 × S. cereale L. Kustro, i.e., monosomic addition lines of the rye chromosomes 4R and 6R; a disomic addition line of 6R; and monotelosomic or ditelosomic addition lines of the long arms of rye chromosomes 4R (4RL) and 6R (6RL). All these lines displayed immunity to powdery mildew. Thus, we concluded that both the 4RL and 6RL arms of Kustro contain powdery mildew resistant genes. It is the first time to discover that 4RL arm carries powdery mildew resistant gene. Additionally, wheat lines containing new wheat-rye translocation chromosomes were also obtained: these lines retained a short arm of wheat chromosome 5D (5DS) on which rye chromosome 4R was fused through the short arm 4RS (designated 5DS-4RS·4RL; 4RL stands for the long arm of rye chromosome 4R); or they had an extra short arm of rye chromosome 4R (4RS) that was attached to the short arm of wheat chromosome 5D (5DS) (designated 4RS-5DS·5DL; 5DL stands for the long arm of wheat chromosome 5D). These two translocation chromosomes could be transmitted to next generation stably, and the wheat lines containing 5DS-4RS·4RL chromosome also displayed immunity to powdery mildew. The materials obtained in this study can be used for wheat powdery mildew resistant breeding program.  相似文献

The genomic DNA of wheatgrass (Agropyron intermedium (Host) P.B. = Elytrigia intermedia (Host) Nevski = Thinopyrum intermedium (Host) Barkworth and Dewey) was labelled with biotin-16-dUTP as a probe, and genomic DNA of common wheat ( Triticum aestivum L. ) "Chinese Spring" was used for blocking. Wheat-wheatgrass line 33 was examined by fluorescence in situ hybridization (FISH) technique. The terminal regions of a pair of chromosomes showed green fluorescent signals. It has been concluded that chromosome segrnents containing alien genes of wheatgrass are located at the terminal regions of wheat chromosomes in wheat-wheatgrass line 33, and the translocated segments were small. Wheat-wheatgrass line 33 has been proved to be a translocation line with chromosome segments of wheatgrass translocated to the terminal regions of wheat chromoSomes.  相似文献

Genotyping was performed for the leaf rust-resistant line 73/00i (Triticum aestivum x Aegilops speltoides). Fluorescence in situ hybridization (FISH) with probes Spelt1 and pSc119.2 in combination with microsatellite analysis were used to determine the locations and sizes of the Ae. speltoides genetic fragments integrated into the line genome. Translocations were identified in the long arms of chromosomes 5B and 6B and in the short arm of chromosome 1B. The Spelt1 and pSc119.2 molecular cytological markers made it possible to rapidly establish lines with single translocation in the long arms of chromosomes 5B and 6B. The line carrying the T5BS x 5BL-5SL translocation was highly resistant to leaf rust, and the lines carrying the T6BS x 6BL-6SL translocation displayed moderate resistance. The translocations differed in chromosomal location from known leaf resistance genes transferred into common wheat from Ae. speltoides. Hence, it was assumed that new genes were introduced into the common wheat genome from Ae. speltoides. The locus that determined high resistance to leaf rust and was transferred into the common wheat genome from the long arm of Ae. speltoides chromosome 5S by the T5BS x 5BL-5SL translocation was preliminarily designated as LrAsp5.  相似文献

利用APAGE、荧光原位杂交技术和RFLP标记,对导入黑麦(SecalecerealeL.)多小穗等性状创制的小麦新种质10_A进行了分子标记检测。APAGE分析发现,10_A与其他1RS/1BL易位系一样,含有1RS的醇溶蛋白标记位点Gld1B3。以黑麦基因组总DNA作探针,用中国春(Triticumaestivumcv.ChineseSpring)基因组DNA作封阻,与10_A根尖细胞有丝分裂染色体进行荧光原位杂交。结果表明,黑麦的1RS易位到10_A中。用25个RFLP探针进行Southern分析,进一步发现10_A的1BS特异限制性片段发生丢失,代之以黑麦1RS的特异限制性片段,而位于其他染色体上的特异限制性片段未发生缺失。据此认为,多小穗小麦新种质10_A属于1RS/1BL易位系。同时还讨论了10_A在小麦遗传改良中的利用情况。  相似文献

A new CMS system designated as ‘msH1’ has been reported in bread wheat using the cytoplasm of H. chilense. While testing this system in different wheat backgrounds, a highly fertile line with chromosome number 42 plus an extra acrocentric chromosome was obtained. The extra chromosome did not pair with any wheat chromosome at meiosis, and progeny from this line which lack the acrocentric chromosome showed pollen abortion and male sterility. In order to establish the origin of this chromosome, FISH using H. chilense genomic DNA as probe was used and showed that it had originated from H. chilense chromosome(s). The novel chromosome did not possess sequences similar to wheat rDNA; however, the probe pSc119.2 from S. cereale containing the 120 bp family was found to occur at the end of its long arm. Data obtained from FISH and EST molecular markers confirm that the long arm of the acrocentric chromosome is indeed, the short arm of chromosome 1Hch from H. chilense. We suggest that the novel chromosome originated from a deletion of the distal part of the long arm of chromosome 1Hch. Neither the 1HchS short arm, nor the whole chromosome 1Hch restores pollen fertility of the alloplasmic wheat. Therefore, the restorer gene on the acrocentric chromosome must be located on the retained segment from the hypothetical 1HchL, while some pollen fertility inhibitor could be present on the deleted 1HchL distal segment. Disomic addition of the acrocentric chromosome was obtained and this line resulted fully stable and fertile.  相似文献

Bread wheat chromosome 3A has been shown to contain genes/QTLs controlling grain yield and other agronomic traits. The objectives of this study were to generate high-density physical and genetic-linkage maps of wheat homoeologous group 3 chromosomes and reveal the physical locations of genes/QTLs controlling yield and its component traits, as well as agronomic traits, to obtain a precise estimate of recombination for the corresponding regions and to enrich the QTL-containing regions with markers. Physical mapping was accomplished by 179 DNA markers mostly representing expressed genes using 41 single-break deletion lines. Polymorphism survey of cultivars Cheyenne (CNN) and Wichita (WI), and a substitution line of CNN carrying chromosome 3A from WI [CNN(WI3A)], with 142 RFLP probes and 55 SSR markers revealed that the extent of polymorphism is different among various group 3 chromosomal regions as well as among the homoeologs. A genetic-linkage map for chromosome 3A was developed by mapping 17 QTLs for seven agronomic traits relative to 26 RFLP and 15 SSR chromosome 3A-specific markers on 95 single-chromosome recombinant inbred lines. Comparison of the physical maps with the 3A genetic-linkage map localized the QTLs to gene-containing regions and accounted for only about 36% of the chromosome. Two chromosomal regions containing 9 of the 17 QTLs encompassed less than 10% of chromosome 3A but accounted for almost all of the arm recombination. To identify rice chromosomal regions corresponding to the particular QTL-containing wheat regions, 650 physically mapped wheat group 3 sequences were compared with rice genomic sequences. At an E value of E < or = 10(-5), 82% of the wheat group 3 sequences identified rice homologs, of which 54% were on rice chromosome 1. The rice chromosome 1 region collinear with the two wheat regions that contained 9 QTLs was about 6.5 Mb.  相似文献

The genetic map of chromosome 5B has been constructed by using microsatellite (SSR) analysis of 381 plants from the F2 population produced by cross of the Chinese Spring (CS) and Renan cultivars. Initially, 180 SSR markers for the common wheat 5B chromosome have been used for analysis of these cultivars. The 32 markers able to detect polymorphism between these cultivars have been located on the genetic map of chromosome 5B. Cytogenetic mapping has involved a set of CS 5B chromosome deletion lines. Totally, 51 SSR markers have been located in ten regions (deletion bins) of this chromosome by SSR analysis of these deletion lines. Five genes—TaCBFIIIc-B10, Vrn-B1, Chi-B1, Skr, and Ph1—have been integrated into the cytogenetic map of chromosome 5B using the markers either specific of or tightly linked to the genes in question. Comparison of the genetic and cytogenetic maps suggests that recombination is suppressed in the pericentromeric region of chromosome 5B, especially in the short arm segment. The 18 markers localized to deletion bins 5BL16-0.79-1.00 and 5BL18-0.66-0.79 have been used to analyze common wheat introgression lines L842, L5366-180, L73/00i, and L21-4, carrying fragments of alien genomes in the terminal region of 5B long arm. L5366-180 and L842 lines carry a fragment of the Triticum timopheevii 5GL chromosome, while L73/00i and L21-4 lines, a fragment of the Aegilops speltoides 5SL chromosome. As has been shown, the translocated fragments in these four lines are of different lengths, allowing bin 5BL18-0.66-0.79 to be divided into three shorter regions. The utility of wheat introgression lines carrying alien translocations for increasing the resolution of cytogenetic mapping is discussed.  相似文献
