Human nails are an attractive matrix for assessing environmental exposures to toxic elements as well as the deficiency of essential elements since it is noninvasively collected and can be stored at room temperature. However, reference or baseline levels in nonexposed and healthy populations are not yet established. Then, the aims of this study were (a) to estimate background values for essential and toxic elements in nails from a healthy young population in southern Brazil and (b) to evaluate possible correlations between essential and toxic elements in nails. Furthermore, this report took part of a recent study of background values in the hair of a young population. Thus, correlations between chemical elements in nails and hair were also evaluated. Nails were collected from healthy children (n?=?126), and element concentrations were determined by inductively coupled plasma mass spectrometry. This study provides new and reliable reference concentrations for toxic and essential elements in children's nails. The results can be used as reference values for epidemiological or clinical investigations based on trace elements in nails. Additionally, we also showed weak to strong correlations between Cu, Mg, Mn, Se, and Zn levels in nails and hair.  相似文献

Lung cancer is seriously threatening human health and exposure to trace metals is the most important aetiology for lung cancer. Selected essential/toxic metals (Ca, Mg, Na, K, Fe, Zn, Cu, Sr, Li, Co, Mn, Ni, Cr, Cd and Pb) are measured in the scalp hair and nails of lung cancer patients and controls by atomic absorption spectrophotometric method employing nitric acid-perchloric acid-based wet digestion. Average concentrations of Pb, Cd, Mn, Co and Cu are found to be significantly higher (p?<?0.05) in the scalp hair and nails of lung cancer patients compared with the controls, however, appreciably higher concentrations of Zn, Ca, Na, Mg and Cr are noted in the scalp hair of the controls. Most of the metal levels reveal higher dispersion and asymmetry in the scalp hair/nails of the patients compared with the controls. Average metal levels are also compared to investigate probable differences based on sex, abode, food and smoking habits. The correlation study shows significantly diverse mutual variations of the metals in the scalp hair and nails of the patients and controls. Considerable variations in the metal levels are also noted for various stages and types of lung cancer (adenocarcinoma, squamous cell carcinoma, large cell carcinoma and small cell lung cancer). Multivariate apportionment of the metals in the scalp hair and nails of the patients and controls are also significantly diverse. The study reveals considerably divergent variations in the metal levels in lung cancer patients in comparison with healthy subjects.  相似文献

Inductively coupled plasma-mass spectrometry (ICP-MS) is a sensitive analytical method to detect the total concentrations of elements in biological samples, but it is unable to identify molecules that can bind to metals, and for this reason it is vital to combine this method's use with other biochemical techniques. Therefore, in order to identify elements complexed to specific proteins, a very relevant combination of bidimensional electrophoresis followed by ICP-MS was used. Protein spots from gels were excised and submitted directly to element detection, a method not reported before. This report focused on the use of plasma from people with laryngeal carcinoma. Most elements were below detection level, with only Cr and Pb being observed in all samples. Although the relationship between metals and laryngeal cancer was not conclusive, it is possible to affirm that the methodology utilized here is successful and has the advantage of determining to which proteins the elements bind.  相似文献

A change in the normal concentration of essential trace elements in the human body might lead to major health disturbances. In this study, hair samples were collected from 115 human subject, including 55 healthy people and 60 patients with prostate cancer. The concentrations of 20 trace elements (TEs) in these samples were measured by inductively coupled plasma-mass spectrometry. A support vector machine was used to investigate the relationship between TEs and prostate cancer. It is found that, among the 20 TEs, 10 (Mg P, K, Ca, Cr, Mn, Fe. Cu, Zn, and Se) are related to the risk of prostate cancer. These 10 TEs were used to build the prediction model for prostate cancer. The model obtained can satisfactorily distinguish the healthy samples from the cancer samples. Furthermore, the cross-validation by leaving-one method proved that the prediction ability of this model reaches as high as 95.8%. Laryngeal cancer is a significant disease worldwide, which presents an increasing incidence. Two contrasting ideas of the immune system role during cancer development are accepted: (1) it fights tumor cells, and (2) it aids tumor progression. Thus, there is no clear understanding about the immune response in laryngeal cancer. Furthermore, since tobacco is the main cause of laryngeal cancer and it contains various carcinogenic components, including metallic elements, these may play a role on cancer development. Plasmas of patients with laryngeal cancer and of healthy smokers were evaluated by 2D gel electrophoresis and mass spectrometry. Proteins were detected on every gel around pH 4.0–10.0 from molecular mass of 10–60 kDa. Few differences were found among cancer and control patients. However, three spots gathered between pI 7.3 and 7.6 with different molecular masses appeared exclusively in cancer profiles. From ten spots identified, six correspond to immune system components, including the three differential ones. The latter were observed only in cancer patients. The presence of several trace elements in the identified proteins was determined by inductively coupled plasma mass spectrometry, where chromium was increased in all proteins analyzed from patients with cancer. This study reinforces the importance of the immune response as target in the understanding and treatment of laryngeal cancer and the possibility that chromium is important in the carcinogenic progress.  相似文献

The aim of this study was to establish background values for toxic and essential elements in hair, interelement correlations and the differences in levels between genders in a healthy young population from Southern Brazil. Hair samples (n = 167) were collected from healthy students aged 12–18 years. Trace element concentrations in hair were determined by inductively coupled plasma mass spectrometry. The study provided relatively low values for toxic elements and balanced concentrations for the essential elements in the adolescents' hair with reliable reference data. Interestingly, this study also demonstrated statistical correlations considered newfound between the elements in hair. Hair mercury levels were influenced by gender; with males presenting higher values. The overall findings of the present study, with respect to the estimated chemical elements, are of prime importance in the evaluation of reference values for determining environmental effects on children living in urban areas.  相似文献

In the research, the contents of heavy metals, Al, Cd and Pb, in the hair of individuals who reside in similar environmental conditions were spectrometrically determined with the use of atomic emission spectrometry–inductively coupled plasma spectrophotometer. The relation to their tobacco smoking habit, age and sex was established. It was observed that the level of all three determined elements was generally higher in the hair of smokers in comparison to hair of non-smokers in both younger and older age groups. In addition, it has been observed that, in the group of elderly people over 50 years old, there was an increase in the content of aluminium, cadmium and lead both in smokers and non-smokers, irrespective of their sex. The sex-related differences in the content of the investigated elements were not unidirectional, and only in few cases did they reveal statistical significance.  相似文献

In order to improve the monitoring system, watching influence of toxic metals on human health in industrial plants, indicator properties of different biosubstances were compared. Four types of samples (whole blood, plasma, urine, and hair) from 263 workers of the “Khimprom” chemical plant (Novocheboksarsk, Russia) were subjected to multielement analysis by ICP-AES/ICP-MS. 19–25 chemical elements, including main toxic metals (Cd, Hg, Pb, etc.) were determined. The results were calculated with regard to workers’ individual data on occupational exposure to chemical elements. Hair was found to be the most sensitive to toxic and conditionally toxic trace metals: Pb, Mn, Cr, Be, Ni, while occupational contact with macro elements (Na, P), trace metalloids (Si, B) and some other metals (Zn) was not reflected in hair. Whole blood relatively weakly indicated a moderate occupational level of metals except Pb and Mn, but effectively reflected deficiencies of essential elements: I, Cr, and shifts in K/Na ratio, which are likely to be secondary effects of harmful occupational factors. Blood plasma reflected only contact with Be, P; urine – only with Ni. In both whole blood and plasma the changes for the absolute majority of elements were similar. Thus, hair analysis is useful for monitoring the occupational exposure to toxic and conditionally toxic chemical elements, while a general estimation of occupational harmful influence on mineral metabolism requires simultaneous investigation of two biosubstances: hair and whole blood, or hair and blood plasma, with whole blood being more preferable. Trace element analysis of human hair has the potential to reveal retrospective information about an individual's nutritional status and exposure. As trace elements are incorporated into the hair during the growth process, longitudinal segments of the hair may reflect the body burden during the growth period. We have evaluated the potential of human hair to indicate exposure or nutritional status over time by analysing trace element profiles in single strands of human hair. The hair strands from five healthy and occupationally unexposed subjects were cut into 1-cm long segments starting from the scalp. By using high-resolution inductively coupled plasma mass spectrometry (HR-ICP-MS), we achieved profiles of 12 elements in single strands of human hair, namely, Ag, As, Au, Cd, Cu, Hg, Fe, Pb, Se, Sr, U and Zn. We have shown that trace element analysis along single strands of human hair can yield information about essential and toxic elements, and for some elements, can be correlated with seasonal changes in diet and exposure. The information obtained from the trace element profiles of human hair in this study substantiates the potential of hair as a biomarker.  相似文献

The incidence of rheumatoid arthritis (RA) has been associated with cigarette smoking. The aim of our study was to assess the trace essential and toxic metals, cadmium (Cd), lead (Pb), and zinc (Zn), in scalp hair samples of 32 Irish and 46 Pakistani smokers and non-smokers RA male patients with age range 42-56 years. For comparison purpose, the scalp hair samples of 27 Irish and 55 Pakistani non-RA male subjects of the same age group were collected. The concentrations of trace and toxic elements were measured by inductive coupled plasma atomic emission spectrophotometer and atomic absorption spectrophotometer prior to microwave-assisted acid digestion. The validity and accuracy of the methodology was checked using certified reference materials and using conventional wet acid digestion method on the same certified reference materials (CRMs). The recovery of all studied elements was found to be in the range of 97.5-99.7% of certified reference values of CRMs. The results of this study showed that the mean values of Cd and Pb were significantly higher in scalp hair samples of both smoker and non-smoker RA patients than in referents (P < 0.001), whereas the concentration of Zn was lower in the scalp hair samples of smokers and non-smokers rheumatoid arthritis patients. The deficiency of Zn and the high exposure of Cd and Pb as a result of cigarette smoking may be synergistic risk factors associated with rheumatoid arthritis.  相似文献

Head and neck cancers are one of the most frequent cancers worldwide. This paper attempts to evaluate disturbances of homeostasis of the necessary elements (calcium, magnesium, zinc, copper, iron, manganese) and changes in the levels of toxic metals (lead, cadmium, cobalt, chromium VI) in hair of patients with head and neck cancers, as well as people without a diagnosed neoplastic disease. In order to quantify the necessary elements and toxic metals, a method using ICP-MS and ICP-OES techniques had been developed and validated. The studies have shown that patients with head and neck cancer used to drink alcohol and smoked much more frequently than healthy individuals, both in the past and presently. Statistically significant differences in concentrations of average metal content in the group of patients with head and neck cancers compared to the control group were confirmed. Significant differences in metal content between the group of patients with head and neck cancers and healthy individuals were found which enabled distinguishing between the study groups. To this end, a more advanced statistical tool, i.e. chemometrics, was used. The conducted research analyses and the use of advanced statistical techniques confirm the benefits of using alternative material to distinguish the patients with head and neck cancers from the healthy individuals.  相似文献

Toxic levels of heavy metals and low levels of essential minerals have been suggested to play a critical role in the pathogenesis of autism spectrum disorders (ASD). This study documents the levels of heavy metals and essential minerals in hair samples of children with ASD in Muscat, the urbanized capital of Oman, Muscat. The study included 27 children with ASD and 27 matched non-ASD controls. Parental interviews were held and dietary intake questionnaires completed in conjunction with the collection of hair samples. Analysis of heavy metals and essential minerals was carried out by inductively coupled plasma mass spectrometry. Chi-square analysis and non-parametric Fisher’s exact tests were used to assess statistical significance. Children with ASD had significantly higher levels of all 11 analyzed heavy metals in their hair samples (P?<?0.05), ranging from 150 to 365 % of control levels. ASD children also had significantly higher levels of essential minerals sulfur, sodium, magnesium, potassium, zinc, and iron, but lower levels of calcium and copper in their hair samples. This study corroborates data from previous studies in different parts of the world indicating the presence of elevated levels of heavy metals and selective depletion of essential minerals in the hair of children with ASD.  相似文献

BackgroundHuman biomonitoring studies of trace elements in biological fluids are mostly limited to a certain number of elements or biological materials. In this study, we describe the significant extension of a biomonitoring to 73 elements being present in concentration ranges from ng/L to g/L in clinically relevant specimens such as blood, serum, erythrocytes and urine.MethodsThe samples were collected from 102 occupationally non-exposed inhabitants of northern Germany. The elements were determined either by inductively coupled plasma tandem mass spectrometry (ICP-MS/MS) in the low concentration range or by inductively coupled plasma optical emission spectrometry (ICP-OES) for essential trace elements and electrolytes.ResultsMean values and selected percentiles of element concentrations are presented for all sample materials. From the results, we calculated the distribution of elements between plasma and blood cells. Application of ICP-MS/MS improves selectivity and accuracy in the determination of elements that are strongly spectrally interfered, such as Cr, Ge, Pd or Ti in blood samples.ConclusionsThis publication provides very valuable information for occupational or environmental hygienists, toxicologists and clinical chemists due to the particularly high number of determined elements and presented concentration ranges.  相似文献

To reveal the role of key elements present in the hair of breast cancer patients on cancer development, the levels of a number of elements in scalp hair samples of 82 people including healthy individuals, people suffering from benign breast disease, and breast cancer patients were measured by PIXE analysis. Pellets of hair samples were prepared and bombarded by 2.2 MeV proton beam of a 3-MV Van de Graaff accelerator. The number of incident ions hitting the sample was indirectly measured using the RBS spectrum of a thin Ag film placed in the beam path. The concentrations of S, Cl, K, Ca, Fe, and Cu in the hair of healthy individuals were in agreement with those observed in the hair of hyperplasia and cancer patients within standard deviations. However, a lower average level of zinc was found in samples from hyperplasia and breast cancer patients. Strong positive correlations were found between iron and potassium as well as between calcium and potassium in the cancer patients. These results could be of significance in the screening for breast cancer.  相似文献

This prospective study was designed to compare the hair levels of 36 elements in 52 patients with stage III breast cancer to those of an equal number of healthy individuals. Principal component and cluster analysis were used for source of identification and apportionment of heavy metals and trace elements in these two groups. A higher average level of iron was found in samples from patients while controls had higher levels of calcium. Both patients and controls had elevated levels of tin, magnesium, zinc, and sodium. Almost all element values in cancer patients showed higher dispersion and asymmetry than in healthy controls. The analysis of trace elements in biological samples will extend our understanding of the impact that environmental exposure to these elements has on human health. Measuring arsenic content in nails has proven useful in studies evaluating the chronic body burden of arsenic. In this study, we developed methodology with inductively coupled plasma-mass spectrometry (ICP-MS) for the determination of total arsenic in nails. We assessed the utility of the washing procedures for removing surface contamination. Four types of preanalysis treatments (water bath, sonication, water bath plus sonication, and control) after sample decomposition by nitric acid were compared to evaluate the digestion efficiencies. In addition, we studied the stability of the solution over 1 wk and the effect of acidity on the arsenic signal. Arsenic content in the digested solution was analyzed by using Ar-N2 plasma with Te as the internal standard. The results suggest that washing once with 1% Triton Χ-100 for 20 min for cleaning nail samples prior to ICP-MS analysis is satisfactory. Repeated measurement analysis of variance revealed that there was no significant difference among the various sample preparation techniques. Moreover, the measurements were reproducible within 1 wk, and acidity seemed to have no substantial influence on the arsenic signal. A limit of detection (on the basis of three times the standard deviation of the blank measurement) of 7 ng As/g toenail was achieved with this system, and arsenic recoveries from reference materials (human hair and nails) were in good agreement (95–106% recovery) with the certified/reference values of the standard reference materials. ICP-MS offers high accuracy and precision, as well as highthroughput capacity in the analysis of total arsenic in nail samples.  相似文献

Our work aimed at extending the search for the trace elements (TE) abnormalities in patients with lung cancer and in healthy controls who smoke, and also for evidence of a possible association between lung cancer and TE. The analysis of the hair from patients with Stage-IIIB non-small cell lung cancer (group 1) and healthy controls (group 2) were analyzed using the inductively coupled plasma mass spectrometry technique in order to obtain information on the correlation between the lung cancer patients and healthy controls. Sixty-seven one-hair samples in group 1 were individually collected before chemoradiotherapy. For comparison, 74 hair samples were collected from group 2. In group 1, the trace elements present at the highest levels were measured to be Ca, Zn, Sn, Na and Mg, respectively, and they were quantified as 68.2, 53.2, 33.9, 23.3, and 28.9?μg.kg(-1), respectively. In group 2, the trace elements present at the highest levels were Zn, Mg, Ca, Fe, and Se, respectively, and they were quantified as 109.7, 31.9, 30.8, 25.0, and 20.1?μg.kg(-1). In group 1, the highest levels of Ca, Sn, and Na were 2.03, 1.06, and 1.01 times higher, respectively, compared with group 2. In group 2, Zn, Mg, Fe, and Se were 2, 1.01, 2.7, and 1.6 times higher, respectively, compared with group 1. When the levels of trace elements were compared between groups 1 and 2 using Student's t test, the levels of Ag, Au, Be, Bi, Ca, Cd, Ce, Co, Cr, Cu, Fe, Ga, Hg, K, Ni, Rb, Rh, Sb, Sc, Ti, V, and Zn were found to be statistically different (p?相似文献

Tumorous and adjacent non-tumorous paired biopsies from 38 patients with colorectal cancer were analyzed by inductively coupled plasma mass spectrometry and inductively coupled plasma optical emission spectrometry after low-volume microwave digestion. 18 elements were investigated: Ag, Al, Ca, Cd, Co, Cr, Cu, Fe, K, Mg, Mn, Mo, Ni, P, Pb, S, Se and Zn. Different chemometric tools were used for data evaluation: Wilcoxon signed rank test, Hieratical clustering analysis, principal component analysis (PCA) and linear discriminant analysis (LDA). With the exception of Al, tumours were observed to have significantly more elevated concentrations of essential elements as compared to non-tumours. On the contrary, elements considered potentially carcinogenic such as Cr, Ni, Mo or Co do not display significant differences. When PCA was applied, different components were obtained for tumorous and non-tumorous tissues. When LDA was applied for the elements studied (including essential and non-essential elements) about 90% of cases were correctly classified.  相似文献

Concentrations of 14 trace elements (Bi, Cd, Co, Cs, Cu, Hg, Mn, Pb, Rb, Sb, Sn, Sr, Tl, and Zn) were determined by inductively coupled plasma mass spectrometry (ICP-MS) in 120 whole-blood and 121 plasma samples of 56 patients with angiographically documented coronary heart disease (CHD). One serum and two wholeblood reference materials were analyzed for quality control. At baseline, patients had elevated Co plasma as well as diminished Cu blood concentrations compared to healthy adults. The Zn concentrations in whole blood were below or at the lower end of the normal range, but the concentrations in plasma were elevated. All other trace elements were within the normal concentration ranges for healthy adults. After initial investigations, patients were randomly assigned to an experimental group (N = 27) and to a usual care group (N = 29). Experimental group patients were prescribed a lifestyle program that included a low-fat diet and a weekly moderate exercise. Patients were examined at baseline, after 6 and 12 mo for clinical assessment and fasting venous blood samples. No significant time-course changes in concentrations of trace elements in blood and plasma during the clinical treatment in both groups of patients could be observed. The experimental group patients lost weight and had lower blood pressure after 12 mo compared to baseline. The interventional therapy reduced the need for further revascularization procedures.  相似文献
