The following new species of Apioninae weevils (Coleoptera: Brentidae) are described from Dominican amber. Toxorhynchus leptorhinusn. sp., Toxorhynchus robustusn. sp., Toxorhynchus amphioculusn. sp., Toxorhynchus atriartusn. sp., Toxorhynchusatriantenusn. sp. and Toxorhynchus hispaniolicusn. sp. are members of the Toxorhynchus decoloratum species group. Toxorhynchus microsomusn. sp., Toxorhynchus stenelytrusn. sp., Toxorhynchus dominicanusn. sp., Toxorhynchus convexusn. sp., Toxorhynchus fuscocorpusn. sp., Toxorhynchus hispidulusn. sp. and Toxorhynchus convexoculusn. sp. are members of the Toxorhynchus sordidum species group. A new subtribe, Stenapiina n. subtribe of the tribe Toxorhynchini, is erected. New species Apionion homochrononn. sp.,Apionion kallimosumn. sp.,Apionion formoculusn. sp. and Stenapion levigatumn. sp. are described. The dominate species of the genus Toxorhynchus and especially those in the Toxorhynchus decoloratum and Toxorhynchus sordidum species groups in Dominican amber are now absent from the West Indies. Abstract

Context: Garcinia hombroniana Pierre, known as manggis hutan in Malaysia is a rich source of xanthones and benzophenones.

Objectives: This study was aimed to isolate and characterize potential cholinesterase inhibitors from the extracts of G. hombroniana bark and investigate their interactions with the enzymes.

Materials and methods: The dichloromethane extract afforded five triterpenoids which were characterized by NMR and mass spectral techniques. Cholinesterase inhibitory assay and molecular docking were performed to get insight of the inhibitory activity and molecular interactions of the compounds. The compounds were also tested for their antioxidant capacity.

Results: The isolated triterpenoids were identified as: 2β-hydroxy-3α-O-caffeoyltaraxar-14-en-28-oic acid (1), taraxerol (2), taraxerone (3), betulin (4) and betulinic acid (5). Compound 1 was the most active dual inhibitor of both AChE and BChE. Compound 1 also showed good antioxidant activities.

Conclusion: Compound 1 had dual and moderate inhibitory activity on AChE and BChE worthy for further investigations.  相似文献

Tob belongs to the anti-proliferative Tob/BTG family. The level of Tob throughout the cell cycle is regulated by the SCF (Skp1/Cullin/F-box protein)Skp2 ubiquitin ligase (E3) complex. Here, we show that Coronin7 (CRN7) is also involved in Tob degradation. We identified CRN7 as a Tob-interacting molecule. A sequence containing two of the six WD motifs in the middle of CRN7 was responsible for the interaction. CRN7 enhanced the polyubiquitination of Tob in vitro, and overexpression of CRN7 promoted proteasome-dependent degradation of Tob. Furthermore, CRN7 interacted with Cullin1 and Roc1 to form a novel SCF-like E3 complex, suggesting that Tob protein is regulated by multiple ubiquitination machineries.

The extreme environments of South Africa mines were investigated to determine microbial community structure and biomass in the deep subsurface. These community parameters were determined using phospholipid fatty acid (PLFA) technique. Air, water and rock samples were collected from several levels and shafts in eight different mines. Biomass estimates ranged over nine orders of magnitude. Biofilm samples exhibited the highest biomass with quantities ranging from 10 3 to 10 7 pmol PLFA g ?1 . Rock samples had biomass ranging from 10 3 to 10 6 pmol PLFA g ?1 . Mine service waters and rock fracture waters had biomass estimates ranging from 10 0 to 10 6 pmol PLFA L ?1 . Air samples biomass values ranged from 10 ?2 to 10 0 pmol PLFA L ?1 . The biomass estimates were similar to those estimates for other deep subsurface sites. Redundancy analysis of the PLFA profiles distinguished between the sample types, where signature lipid biomarkers for aerobic and anaerobic prokaryotes, sulfate-and metal-reducing bacteria were associated with biofilms. Rock samples were enriched in 18:1 ω 9 c , 18:2 ω 6, br17:1s and br18:1s, which are indicative of microeukaryotes and metal- reducing bacteria. Air samples were enriched with 22:0, 17:1, 18:1, and a polyunsaturated fatty acid. Service waters had monounsaturated fatty acids. Fracture waters contained i17:0 and 10Me18:0 which indicated gram-positive and other anaerobic bacteria. When the fracture and service water sample PLFA responses to changes in environmental parameters of temperature, pH, and anion concentrations were analyzed, service waters correlated with higher nitrate and sulfate concentrations and the PLFAs 18:1 ω 7 c and 16:1 ω 7 c . Dreifontein shaft 5 samples correlated with chloride concentrations and terminally branched saturated fatty acids and branched monounsaturated fatty acids. Kloof, Tau Tona, and Merriespruit fracture waters aligned with temperature and pH vectors and 18:0, 20:0 and 22:6 ω 3. The redundancy analysis provided a robust method to understand the PLFA responses to changes in environmental parameters.  相似文献

The following new genus and seven new species of Cossonine weevils (Curculionidae: Cossoninae) are described from Dominican amber: Acamptus exilipes Poinar and Legalov, n. sp. (similar to A. rigidus), Caulophilus camptus Poinar and Legalov, n. sp. (similar to C. bennetti), Caulophilus elongatus Poinar and Legalov, n. sp. (similar to C. swensoni), Caulophilus ruidipunctus Poinar and Legalov, n. sp., Dryotribus pedanus Poinar and Legalov, n. sp. (similar to D. amplioculus), Ogygius obrieni Poinar and Legalov, n. gen., n. sp. (similar to the genus Apotrepus), and Stenotrupis pumilis Poinar and Legalov, n. sp. (similar to S. breviscapus). A key to the Dominican amber species of the genus Caulophilus is provided. A significant amount of nuclear p53 is found associated with the nuclear matrix in cells that were exposed to genotoxic stress. In this study we identified Scaffold attachment factor B1 (SAFB1), a nuclear matrix-associated protein that binds the scaffold or matrix attachment regions (S/MARs) of genomic DNA, as a novel p53-interacting protein. SAFB1 was able to associate with p53 through its C-terminal domain, while significant co-localization of the two proteins was observed in cells treated with 5-fluorouracil or mithramycin. Binding of p53 to SAFB1 had a significant functional outcome, since SAFB1 was shown to suppress p53-mediated reporter gene expression. These data suggest that nuclear matrix-associated proteins may play a critical role in regulating p53 localization and activity.

ABSTRACT

Three new species of Parhydraenida Balfour-Browne, 1975 are described from southeastern Brazil: P. plesioformis, sp. n.; P. proboscis sp. n.; P. unicornis sp. n. The genus Parhydraenida is recorded from Minas Gerais for the first time, and P. hygropetrica Perkins, 1980 is recorded from Rio de Janeiro for the first time. We describe one secondary sexual character, which enables, for the first time, unambiguous distinction of males and females. Habitus photographs as well as line drawings of the male and female genitalia of all three new species, and of P. hygropetrica are provided.
