Pantoea (formerly Enterobacter) agglomerans YS19 is a dominant diazotrophic endophyte isolated from rice (Oryza sativa cv. Yuefu) grown in a temperate-climate region in west Beijing, China. In vitro adsorption and invasion of YS19 on host plant root were studied in this research. Adsorption of YS19 on rice seedling roots closely resembled the Langmuir adsorption and showed a higher adsorption quantity than the control strains Paenibacillus polymyxa WY110 (a rhizospheric bacterium from the same rice cultivar) and Escherichia coli HB101 (a general model bacterium). Adsorption dynamics study revealed high rates and a long duration of the YS19-rice root adsorption process. Adsorption of YS19 was mainly observed on the root hair, though which it enters the plant. This in vitro adsorption study revealed an apparent strong interaction between YS19 and rice at the early endophyte-host recognition stage.  相似文献

为研究内生细菌对宿主植物侵染定殖的机理和其共生生物学作用 ,对水稻内生优势成团泛菌 (Pantoeaagglomerans)YS19与绿色荧光蛋白 (GFP)标记的YS19B ::gfp菌株的生长动力学进行了比较研究 ,探讨了成团泛菌YS19B ::gfp的标记稳定性和荧光性质 .标记菌株与野生型菌株相比 ,最大比生长速率和最大生物量仅减小 12 4 %和 6 % ,代时延长 14 0 % .成团泛菌YS19B ::gfp在指数期连续传代培养 10 0代后 ,GFP标记的保持率为 89 1% ,建立了标记菌株在有标记丢失存在时的生长动力学模型 :dX+ dt =μ+ (1-p)X+ ,解析出细胞分裂时标记丢失的概率p =9 75 6× 10 -7,确定了方程的模型参数 .标记菌株的荧光光谱在激发波长为 4 0 0nm时 ,最大发射波长为 5 0 8nm ,与供体菌株完全相同 .在LB培养基上生长时 ,成团泛菌YS19B ::gfp的GFP产生时间在指数期末期到稳定期较快 ,并于培养至 2 0h时达到最高 ,同时单位菌体生物量的荧光强度也达到最大 .结果说明 ,在GFP标记后成团泛菌YS19B ::gfp的生长仅受到较小影响 ,不致对成团泛菌的生理活动造成大的改变 ,同时由于该菌对宿主的侵染能力比其它内生细菌要强得多 ,因而该菌对植物的侵染活性影响也较小 ,该菌仍然可以保持其内生优势地位 .该标记的稳定性比较高 ,荧光产生正常 ,很适  相似文献

Pantoea agglomerans is characterized by the formation of multicellular symplasmata. One unanswered question regarding this bacterium is how these structures are formed. In this study, the rice diazotrophic endophyte P. agglomerans YS19 was selected for exploration of this theme. YS19 was labeled with green fluorescent protein and the resulting recombinant YS19::gfp was observed to grow only slightly more slowly (a decrease of 5.5%) than the wild-type strain, and to show high GFP label stability (label loss rate 8.9218 x 10(-6) per generation, nearly reaching the generally accepted spontaneous mutation rate for most bacteria). YS19::gfp resembled the wild-type YS19 in symplasmata formation and growth profiles. Based on associated cultivation of both strains by mixing their individually cultivated single cells, symplasmata were formed and composed of both YS19::gfp and YS19, suggesting that YS19 formed symplasmata via aggregation, not proliferation, of the original single cells.  相似文献

Pantoea (formerly Enterobacter) agglomerans YS19 is an endophytic diazotrophic bacterium isolated from rice (Oryza sativa cv. Yuefu) grown in temperate climatic regions in west Beijing (China). The bacterium forms aggregate structures called `symplasmata'. A symplasmatum is a multicellular aggregate structure in which several (at least two) to hundreds of individual cells tightly bind together. The studies on the symplasmata formation of YS19 showed that there were two growth stages for YS19, including the single cell stage existing before exponential growth phase and the symplasmata forming stage starting at the early stationary growth phase in liquid GY (glucose yeast extract) medium or at the end of the exponential growth phase in liquid LB (Luria-Bertani) medium. There was a correlation between symplasmata formation and bacterial growth phase. When the medium was acidified, the cell growth rate was affected by the low pH of the medium, but the time required for symplasmata formation was not influenced by it. YS19 also formed symplasmata on agar medium, where more symplasmata were formed than in liquid medium. The volume of individual constitutional cells of symplasmata was sharply decreased by more than a half in comparison with that of the single cells existing before symplasmata formation. On all the media tested, YS19 formed symplasmata in most of the cell growth phases. The genome DNA/DNA homology between P. agglomerans YS19 and type strain P. agglomerans JCM1236T (ATCC27155T) was determined as 90.1%, confirming its membership of P. agglomerans. In order to investigate the phylogenetic relationships of YS19 at the intraspecific, intrageneric and super-generic level, the 16S rDNA similarities between strain YS19 and 17 other strains of Pantoea and 4 representatives of the closely related genera were analyzed. All the strains of Pantoea were clustered into 5 groups, and YS19 was clustered in a unique branch. The 16S rDNA similarity between YS19 and type strain JCM1236T was 93.9%, much lower than the generally accepted value (=97%) for members of the same species, indicating that the 16S rDNA of YS19 has a distinct molecular characteristic.  相似文献

Pantoea agglomerans YS19 is a diazotrophic endophyte isolated from rice (Oryza sativa cv. Yuefu) grown in a temperate-climatic region in west Beijing (China). The colonization of YS19 on host rice was studied in this paper. It was revealed that YS19 colonizes in all the tissues of rice seedlings, including roots (dominantly at elongation regions, lateral root junctions, root hairs and root caps), stems and leaves. More YS19 colonizes in stem and leaves (1.40 × 105 CFU mg−1 fresh weight) than that in roots (3.60 × 104 CFU mg−1). Symplasmata, a kind of adaptive structure of the strain for its endophytic living, were repeatedly observed to form inside root or stem cortex parenchyma tissues, as well as on leaf surfaces and also rhizoplanes. A novel matrix protein (SPM43.1) with its expression paralleling to the formation of symplasmata was captured, whose meaning in structural construction of symplasmata was also discussed.  相似文献

AIMS: To investigate the ability of Azospirillum sp., a facultative endophitic diazotrophic bacterium, to release plant growth regulators (PGR) such as polyamines, ethylene, indoleacetic acid and amino acids in both combined-N and N-free cultures. METHODS AND RESULTS: The presence of those substances was analysed by HPLC. Azospirillum sp. is capable of releasing PGR and amino acids into the culture medium. CONCLUSIONS: The type and quantity of the released substances varied, depending on the presence of combined-N in the medium. SIGNIFICANCE AND IMPACT OF THE STUDY: A better knowledge of PGR produced by Azospirillum sp. has been gained.  相似文献

植物内生菌由于其独特的生态学地位而广受关注,近年来有关植物内生菌与宿主相互作用的研究取得了很大进展.本文综述了植物内生菌通过分泌促生物质、拮抗病原菌等实现与宿主共生互作,同时植物为内生菌提供适宜的黏附表面,使其形成以生物薄膜(biofilm)为主要形式的多细胞聚集体结构以更好地适应周围的生存环境,从而更加高效地对植物产生促生作用.本文论述了内生菌在与植物的互作中形成的多细胞聚集结构在抵抗非生物胁迫方面的独特生理及生态学意义,结合水稻内生成团泛菌YS19形成多细胞聚集体symplasmata现象及其生物学效应,对未来有关植物内生菌的研究方向提出了一些看法.  相似文献

An understanding of hereditary endophytic fungi, and the effects on grass persistence strategies (i.e. relative investment in sexual reproduction and vegetative growth) under natural conditions may help to predict how some alpine ecosystems will respond to environmental change. Grass persistence and endophyte maintenance in host populations are closely related, but could become independent due to endophyte loss mechanisms. We used native grass and endophyte populations to test the hypothesis that fungal endophytes manipulate grass persistence strategies to secure endophyte maintenance in plant populations. Two conditions were required to verify this hypothesis: 1) the fungus caused alterations in host plant strategies; and 2) plant phenotypic changes induced by the fungal endophyte increased endophyte transmission. We compared symbiotic (S) and non‐symbiotic (NS) persistence strategies of Festuca eskia (Poaceae), an alpine grass infected by the asexual form of the fungal endophyte Epichloë festucae. We characterised endophyte transmission efficiency, and described vegetative growth and sexual reproduction in a field population that naturally supports approximately 50% S plants. We built a demographic model to estimate plant vegetative growth rates. A correlation between plant persistence strategy, and fungal maintenance was evaluated by increasing soil resource levels. Under natural conditions, S and NS plants exploited different persistence strategies in the same population; S plants exhibited greater vegetative growth than their NS counterparts, while maintaining the same reproductive output. In response to higher soil resource levels, S plants shifted in persistence strategies and phenology, whereas NS plants maintained the same strategies. Therefore, results suggested the fungal endophyte fine‐tuned host persistence strategies according to soil resource level. Finally, we found no direct relationship between the changes induced by fungal endophyte and endophyte transmission. Consequently, fungal endophytes affected host persistence strategies, but did not directly increase endophyte transmission.  相似文献

AIMS: This study aims at evaluating the ability of Beijerinckia derxii, a free-living nitrogen (N)-fixing bacterium frequently isolated from tropical soils, to release certain plant growth regulators [indoleacetic acid (IAA), ethylene, polyamines] and amino acids into the growth medium. METHODS AND RESULTS: The production of those substances was compared using both cultures in which nitrogenase was active (N-free medium) and cultures in which nitrogenase was repressed (combined-N cultures). Those cultures were grown under agitation and in absence of agitation. Total IAA production was higher in agitated, N-free cultures but specific production was greater in combined-N cultures under agitation. Putrescine and spermidine were detected under all conditions tested. Ethylene was produced in both N-free and combined-N cultures. A greatest diversity of amino acids was released in N-free cultures. CONCLUSIONS: There was no inhibition of the production of the analysed substances under conditions where nitrogenase was inactive. SIGNIFICANCE AND IMPACT OF THE STUDY: Beijerinckia derxii is potentially a producer of plant-active substances; its presence in the natural environment suggests that this bacterium may contribute to the development of other living organisms.  相似文献

The response of the stem of a resistant host (Impatiens baslamina) to infection by the parasitic flowering plant Cuscuta japonica was studied with light and electron microscopy. The intra- and interfascicular cambial cells in the host stem first reacted to the penetrating upper haustorium by dividing, and the differentiation of the host xylem (vascular) tissues proceeded toward interfascicular areas from vascular bundles. When the host vascular tissue was invaded by the endophyte (haustorial portion in the host stem), the host xylem was displaced, and host vessels became occluded with parenchyma cells, resulting in tyloses. As the parasitism progressed, areas of the host stem penetrated by the endophyte became swollen via secondary growth and cell division in the parenchymatous cortex, pith, and interfascicular areas. During this intrusion by the endophyte, darkly stained necrotic reactions were detected at the interface between the host tissue and the invading endophyte. The results suggested that in the host tissues penetrated by the parasite, the formation of secondary tissue and swellings caused by active cell division of ground tissue and host vessel occlusion by tyloses constitute the host structural defense against the parasite.  相似文献

The present paper reports the establishment of rice cell suspension culture system, including callus induction and proliferation, isolation of single cells and small aggregates, cell suspension culture and callus re-formation, as well as regeneration of plantlets. The results have been obtained as follows: 1. The compositions of the different media used for callus induction, callus proliferation, cell suspension and plant regeneration are summarized in Table 1.2. Two kinds of disifectants, mercuric chloride and sodium hypochlorite, were used for surface sterilization of brown rice. The percentage of callus formation and callus yields were much higher when sodium hypochlorite was used (Fig. 3). We suggest that the disinfactant is one of the important factors that affect callus formed at the initial stage has an influence upon subsequent isolation of cells and suspension culture and even plant regeneration. 3. Table 3 shows that addition of yeast extract to the medium improves callus yield greatly and the efficiency of callus formation to a lesser extent. 4. Both medium Ⅱ (modified B5 medium) and N6 medium were suitable for cell suspension culture, but medium II was more effective for cell growth and callus re-formation (Fig. 4 and Table 4). 5. Effect of 2, 4-D on cell growth was tested at the concentration range among 0, 10-6, 10-5, 10-4 to 10-3 M. The results indicated that 10-5 M of 2,4-D was most effective for induction of rice callus. It has also been found that absence of 2,4-D increased callus re-formation in suspension culture, but no plant regeneration was observed. 6. By using 7% sucrose in differentiation medium, for all the three varieties, the plant regeneration frequency was raised up to 3 or 4 times than those of the 3% ones (Table 6). Occurrence of albino plants is often reported as one of the problems in rice anther culture. It is, however, no problem in seed-derived rice cell culture.  相似文献

Aims

The main objective of this study was to test the hypothesis that isolating and characterizing bacterial suspensions (undefined consortia) from the root/rhizosphere of rice will contribute to the selection of mixtures of rhizobacteria with better growth-promoting activity of rice plants.

Methods

Bacterial consortia were obtained from roots/rhizosphere soil samples of rice plants grown under upland and irrigated production systems. Those undefined consortia were subjected to five consecutive passes every 7 days in NFb (N-free broth) semisolid medium. Thereafter, strains of each growth-promoting consortia were isolated by plating on three different culture media. Then, undefined consortia, as well as mix and single bacterial strains, were characterized in terms of indoleacetic acid production, nitrogen fixation capacity, and growth promotion of rice plants.

Results

Of the 72 consortia analyzed, 41.7 % and 50.0 % increased nitrogenase activity and the production of indolic compounds, respectively, after 5 continuous passes in NFB medium. Three undefined consortia, 11 single strains and 5 strain mixtures, exhibited plant growth promotion in rice plants under greenhouse conditions.

Conclusions

Continuous enrichment in Nfb medium of undefined consortia from root/rhizosphere soil is a good strategy for the selection of plant growth-promoting bacteria for rice plants.  相似文献

Growth of a filamentous fungus endophyte, Neotyphodium, and its host plant, tall fescue, Festuca arundinacea, was examined during the seed germination process under pseudo-microgravity [correction of micrgravity] generated by three dimensional (3D-) clinorotation. The shoot growth of tall fescue infected with the endophyte was remarkably suppressed on a 3D-clinostat compared with that of the ground control. Without being infected, shoot growth of tall fescue was not strongly affected by the 3D-clinorotation. Many aggregated hyphae were observed in the plant seed incubated for 1-day on the 3D-clinostat [correction of clinost] than in those kept on the ground. These results indicate that the clinorotation induces responses in the endophyte and its host plant different from those under normal gravity.  相似文献

Embryogenic callus was induced from immature embryos of Angelica sinensis cultured on Murashige and Skoog (MS) basal medium. Embryogenic callus growth was more rapid on MS basal medium than on B5 or White medium. Embryogenic callus was used to establish a suspension culture and somatic embryos and germinating embryos developed during the culture. A shaking speed of 80 rpm was found to be optimal for establishing suspension cultures, while 100 rpm produced more somatic embryos and germinating embryos with an initiation cell density of 0.2 ml packed cell volume/25 ml medium. Adding 0.3% agar to the liquid medium also stimulated the formation of somatic and germinating embryos. While no plant growth regulators were needed for culture initiation and plant regeneration, the addition of 0.5–1 mg/l 2,4-dichlorophenoxyacetic acid was needed to maintain the embryogenic suspension culture by preventing embryo germination. Forty percent of the germinating embryos survived after culturing on filter paper moistened with liquid half-strength MS medium containing 3% sucrose. Endophytic fungi have been shown to increase tolerance of hosts to biotic and abiotic stresses and in some cases alter growth and development of plants. In this article we evaluate some effects that clavicipitaceous endophytes have on development and physiology of plant tissues. We postulate that oxidative stress protection is the fundamental underlying benefit conferred by many endophytes, accounting for frequently observed enhanced disease resistance, drought tolerance, heavy metal tolerance and tolerance to numerous additional oxidative stresses. We hypothesize that endophyte-mediated oxidative stress protection of the host is the result of at least two processes, including: (1) secretion of reactive oxygen species (ROS) from endophytic mycelia into plant cells; and (2) secretion of auxin from endophytic mycelia into plant cells. Both processes result in an increase in ROS in plant tissues; and stimulate plant tissues to increase activities of antioxidant systems. Auxin is suggested to function in suppression of plant cell death and may be important in maintaining the endophyte–plant symbiosis.  相似文献

In the 'F' horizons of acid mor-humus soils of heathland ecosystems, mycorrhizal roots of the dominant ericaceous species form a large fraction of the soil biomass. Rapid turnover of these roots provides the potential for recycling of substantial amounts of nitrogen contained in their fungal and plant components. Here, we first determine the amount of N in the biomass of ericoid roots growing in heathland and show it to constitute a large proportion of total soil N. In order to assess the accessibility of this N to ericaceous plants, experiments were then conducted using aseptically produced shoot and root necromass of Vaccinium macrocarpon Ait., the roots being grown with or without mycorrhizal colonization. These materials were provided as sole nitrogenous substrates in growth experiments using the ericoid mycorrhizal fungus Hymenoscyphus ericae (Read) Korf & Kernan in pure culture and V. macrocarpon in the mycorrhizal (M) or non-mycorrhizal (NM) condition as test organisms. The experiments were designed to test the hypothesis that the N contained in these substrates can be mobilized by the mycorrhizal endophyte. The ability of the endophyte to utilize the substrates was determined by measuring fungal yields and by assessing the presence of its extra-cellular protease and chitinase enzymes. Transfer of N to the host by the endophyte was determined through measurements of plant yield and tissue N contents. H. ericae produced a significantly greater yield on shoot and mycorrhizal root necromass than on non-mycorrhizal root necromass. The extra-cellular enzymes protease and chitinase were produced by the fungus when grown on the M root necromass. The fungus also transferred N to the host plant, up to 76% of N contained in the substrate being found in M plants whereas less than 5% was present in their NM counterparts.  相似文献

The biology of the Striga species will be briefly described. Pot experiments indicate that damage to the host plant is very much greater than is explained by removal of photosynthates. A phytotoxin is almost certainly involved, which causes decreased shoot growth of the host plant but which also directly or indirectly encourages increased root growth. The significance of this change in root/shoot ratio will be discussed in relation to host resistance and various control measures.  相似文献

Plants simultaneously associate with multiple microbial symbionts throughout their lifetimes. To address the question of whether the effects of simultaneous symbionts are contingent on the specific identities, we conducted a greenhouse experiment manipulating the presence and identities of arbuscular mycorrhizal fungi (AMF) and fungal endophytes on the shared host grass Elymus hystrix. Each plant host was inoculated with one of two AMF species having varying effects on host growth, or a sterile soil control. Further, we used naturally occurring endophyte‐infected (E+) and uninfected (E–) individuals from two populations of the endophyte Epichloë elymi that varied in their interaction with E. hystrix. We then measured responses of plants, AMF, and fungal endophytes. Overall, we found that the combined effects of AMF and fungal endophytes on plant growth were additive, reflecting the mutualistic quality of each symbiont independently interacting with host plants. However, fungal endophyte infection differentially altered hyphal colonization of the two AMF species and the identity of the coinfecting AMF species affected fungal endophyte fitness traits. The results of this study demonstrate that the outcome of interspecific symbiotic interactions varies with partner identity such that the effects of simultaneous symbioses can not be generalized.  相似文献
