The corpus luteum (CL) is a transient endocrine organ which can manifest a number of pathologic conditions such as cysts, inflammation, adhesions, dysfunction and neoplasia. Luteal and follicular cysts are the most commonly encountered abnormalities and need to be distinguished from cysts within a normal CL. Inflammatory lesions are also frequently encountered and can be caused by viral, bacterial, or iatrogenic causes. If inflammation is severe, adhesions and subfertility/infertility can result. Luteal dysfunction is a broad classification of another pathologic condition encountered in the cow. Generally this results in abnormal production of progesterone or abnormal luteal lifespan resulting in infertility. Neoplasms are relatively rare in the CL but include both primary and metastatic tumors. Understanding the pathologic conditions that occur within the CL will allow a more accurate clinical assessment of these very dynamic endocrine structures.  相似文献

The objective of this study was to investigate the presence of tumor necrosis factor (TNF)-alpha mRNA and TNF-alpha receptors in the bovine corpus luteum (CL) during the gestation period. TNF-alpha mRNA and TNF-alpha receptors were determined on bovine CL from pregnant cows at three stages: trimester I (fetal crown-rump length; 6-20 cm), trimester II (25-45 cm) and trimester III (50-80 cm). TNF-alpha mRNA was detected by an RT-PCR analysis in the CL of all stages of gestation. A Scatchard analysis revealed the presence of a high-affinity binding site (Kd; 5.1-6.9 nM) in the CL membranes collected at each stage of gestation. Furthermore, the concentrations of TNF-alpha receptors in the CL of trimesters I (24. 0 +/- 1.95 pmol/mg protein) and III (21.6 +/- 2.39 pmol/mg protein) of gestation were significantly higher than the concentration in trimester II (14.9 +/- 2.07 pmol/mg protein) (P < 0.05). These results indicate that TNF-alpha is locally produced and that TNF-alpha receptors are present in bovine CL during the gestation period, and suggest that TNF-alpha plays one or more roles as a paracrine factor in regulating bovine CL function during the entire gestation period.  相似文献

Neuron-specific enolase (NSE) is the key molecular marker for diffuse neuroendocrine system (DNES) cells, its expression in the pregnant corpus luteum (CL) of dairy goats was studied by the immunofluorescence method and the ultra structural characteristics of luteal cells were detected by the electron microscopy to identify the existence of DNES cells in the pregnant CL of dairy goats. Besides, the coding sequence of dairy goats NSE gene was cloned and its biological information was analyzed. Results revealed that NSE immunopositive cells exhibited widespread cytoplasmic staining throughout the whole pregnant CL. In addition, these cells showed typical characteristics of DNES cells in the electron microscopy. These results suggested that many DNES cells exist in the pregnant CL of dairy goats. Meanwhile, we identified the coding sequence of dairy goats NSE (GenBank Accession No. JN887466). Its nucleotide sequence homology was found to be 97.9%, 89.3%, 90% and 92.6%, respectively, compared with that of Bos taurus, Rattus norvegicus, Mus musculus and Homo sapiens, while the amino acid sequence homology was 99.1%, 97%, 97.2% and 98.2% respectively. These results first showed that the functional amino acids coded by the NSE gene were highly conserved in Caprine, B. taurus, R. norvegicus, M. musculus and H. sapiens. It was implied that the gene NSE in dairy goats had close homology to that of NSE of other species. Our findings demonstrated the possible existence of DNES cells in pregnant CL, providing new clue for further understanding of interactions between the neuroendocrine and reproductive systems. Characterization of gene sequence of dairy goats NSE will enable us to synthesize interference RNA for further study on the role of NSE on the formation, function and apoptosis of pregnant CL in dairy goats.  相似文献

A histomorphological investigation of the corpus luteum has been performed in 60 women with an unchanged menstrual cycle. Physiological changes of the corpus luteum are reliably reflected in the karyometry results, in correlation between immature, mature and distrophic luteocytes, their mitotic activity and in total thickness of the parenchymatous layer. The parameters taken into account are united into a total index of the corpus luteum activity, its functional significance is proved when a parallel estimation of microelements content (zinc and copper) is performed in the corpus luteum tissues. The morphometric indices used demonstrate that the menstrual corpus luteum is mainly formed at the expense of a combined hypertrophy of granulous and theca-lutein cells and maturation of the preceding cellular forms. Regressive changes are characterized by a decreasing volume of the nuclei and distrophic changes of luteocytes, which are especially manifested at the end of the menstrual cycle.  相似文献

Anestrous postpartum (PP) Hereford cows (n = 41) were used to compare corpora lutea (CL) from gonadotropin-releasing hormone (GnRH)-induced ovulation with CL from cycling cows. Postpartum cows were injected i.m. daily with 100 mg progesterone (P4) or oil on Days 25 through 28 PP and then given 200 micrograms GnRH i.m. on Day 30 PP. Corpora lutea were removed from one-half of the PP cows in the oil- and P4-treated groups 6.5 days after GnRH injection, and from the cycling cows 7 days after estrus. Intact PP cows were used to evaluate cycle length. Blood was collected daily from all PP cows from Day 25 PP through luteectomy and on Days 9, 11, and 13 post-GnRH from the oil- and P4-intact cows to determine short (SHORT) versus normal (NORM) luteal phases. Cycling cows were bled daily from estrus until CL removal NORM PP cows had higher (P less than 0.001) P4 levels than did SHORT PP cows from Day 7 through Day 13 post-GnRH, and more (P less than 0.05) P4-intact cows were NORM compared with oil-intact cows (45.5% vs. 14.3%, respectively). Corpora lutea from cycling cows were heavier (P less than 0.05) and had a higher luteinizing hormone (LH) receptor concentration (P less than 0.05), but CL P4 concentration did not differ from PP cows. Corpora lutea weight, LH receptor and P4 concentration, and in vitro P4 production were similar in the oil-and P4-treated PP cows. The corpus luteum (CL) is a site of intense angiogenesis. Within a short period, this is followed either by controlled regression of the microvascular tree in the non-fertile cycle, or maintenance and stabilisation of the new vasculature a conceptual cycle. The molecular regulation of these diverse aspects is examined. The CL provides a unique model system in which to study the cellular and molecular regulation of angiogenesis. Vascular endothelial growth factor (VEGF) has been found to have a major role in the CL. By targeting its action at specific stages of the luteal phase in vivo by antagonists, profound inhibitory effects on luteal angiogenesis and function are observed.  相似文献

The objectives of this experiment were to determine if subnormal levels of progesterone (P4) indicative of luteal insufficiency influence (1) pulsatile release of luteinizing hormone (LH), (2) the interval to the preovulatory surge of LH after removal of P4, and (3) the secretion of P4 during the estrous cycle subsequent to administration of subnormal levels of P4. On Day 5 (Day = 0 day of estrus) of the estrous cycle, cows received P4-releasing intravaginal devices (PRID) to produce normal (2 PRIDs; n = 7) or subnormal (0.5 PRID; n = 6) concentrations of P4. Five cows served as controls. On Day 10, serial blood samples were collected from all cows. Collection of blood samples was again initiated on Day 17 in cows receiving PRIDs. The PRIDs were removed and blood collection continued for 78 h. Daily blood samples were collected from all animals for 42 days subsequent to estrus (estrous cycles 1 and 2, respectively). During estrous cycle 1, mean concentration of P4 was lower (p less than 0.05) and frequency of pulses of LH was higher (p less than 0.05) in cows receiving subnormal P4 than in cows receiving normal P4 and control cows. Plasma concentrations of estradiol (E2) were higher (p less than 0.05) on Days 9-16 of estrous cycle 1 in cows receiving subnormal P4 than in cows receiving normal P4 or in control cows. Follicular development after treatment with a gonadotropin-releasing hormone agonist (buserelin) was compared in ovaries of postpartum cows bearing (CLO) or not bearing (NCLO) a corpus luteum (CL). In the first experiment, 16 cows on day 7 of the estrous cycle (day 0 of treatment) were treated either with saline or 8 μg of buserelin. Both ovaries were collected on day 3 or day 6 (n = 4 per group per day) and follicles over 1.57 mm in diameter were observed histologically. Compared with day 3 in the saline group, there was a greater decrease in the percentage of Class I total (1.57–3.67 mm; P < 0.08) and Class 1 atretic follicles (P < 0.04) but a greater increase in that of Class 2 total (3.68–8.57 mm; P < 0.06), Class 2 atretic (P < 0.04) and early atretic (P < 0.05) follicles on day 6 in the CLO than in the NCLO. In the buserelin group however, all follicular responses (except for Classes 1 and 2 nonatretic follicles, P < 0.08) were similar (P > 0.1) between CLO and NCLO within 3 days after treatment. In the second experiment, follicular responses in CLO and NCLO were compared by daily ultrasonography in cows that had (n = 6) or did not have (n = 4) a buserelin-induced ovulation. After buserelin treatment, the numbers of medium (5–10 mm) and large (over 10 mm) follicles were not different (P > 0.1) between the CLO and the NCLO whether ovulation occurred or not. Results indicate that treatment with buserelin overcame most of the local effects of the CL on the growth and atresia of ovarian follicles in postpartum cycling cows within a 6 day period and this occurred whether ovulation was induced or not.  相似文献

LH and prolactin were measured in plasma of 5 cows before and after enucleation of the persistent corpus luteum. Two cows returned to normal oestrous cycles following enucleation and did not differ from normal, cycling cows, with respect to hormone levels. Three other ones developed follicular cysts following enucleation and had decreased levels of LH and prolaction as compared with cycling cows. On the day of enucleation the fluctuations in LH-levels were higher than either before or after enucleation.  相似文献

A transrectal real-time B mode linear scanner was used to learn if the appearance of a cavity in the corpus luteum (CL) in cows causes infertility. Sixty-one cows that were confirmed to have ovulated after artificial insemination (AI) were used. Some of the cows that returned to estrus were examined two or three times; thus, a total of 78 observations were made. The rate of cavity appearance was 37.2% (29 78 ). In relation to the size of cavity, the rates of appearance were 30.8% of CL with cavities of 7 mm or greater and 24.4% with cavities of 10 mm or greater in diameter. It took more than 21 days for the cavity more than 10 mm to disappear. In the cows that were observed two or three times, the cavity did not tend to reappear. The mean period for return to estrus, mean serum progesterone concentration and mean pregnancy rate were not significantly different (P>0.05) for cows that had CL with or without a cavity. Our results suggest that the cavity mentioned above does not cause infertility.  相似文献

Binding of follicle stimulating hormone (FSH) to a crude membrane fraction of bovine corpus luteum (CL) has been detected. This binding meets the usual criteria for a receptor based on specificity, time course of reaction and association constant (Ka = 8.5 x 10(10)M(-1)). Physiological studies with CL removed from heifers at specific times after estrus indicate that day-6 CL had the highest FSH binding. However, a correlation with physiological function was not obvious since some functional mid-cycle CL were high in progesterone and luteinizing hormone (LH) receptor but had nondetectable FSH receptor. Conversely, some late-cycle CL had low progesterone and LH receptor but significant quantities of FSH receptor.  相似文献

There is a growing body of evidence that intercellular communication is important in the regulation of luteal function. Although the nature of the interactions between small and large luteal cells are not yet clear, it seems likely that they do exist. Many of the substances to which luteal cells respond, such as prostaglandins, growth factors, oxytocin and progesterone, are produced locally. These substances may act as paracrine factors to modulate the response of luteal cells to hormonal signals. Endothelial cells also produce factors that can modify steroidogenesis, and luteal cell-stimulation of endothelial cell proliferation is necessary for the extensive angiogenesis that occurs during luteinization Finally, bidirectional intercellular communication likely occurs between luteal cells and resident immune cells. Immune cells produce cytokines that can modify progesterone and prostaglandin synthesis by luteal cells. Cytokines may also have direct cytotoxic effects on luteal cells, and dead cells are then phagocytized by resident macrophages. Also, factors secreted by luteal cells can serve as chemoattractants for immune cells, and can enhance or suppress immune cell functions. There is little doubt that intercellular communication within the corpus luteum is very complex. One must remember, however, that nearly all evidence collected thus far is based on in vitro studies. Eventually, technology will allow for study of these interactions in vivo, and may lead to new methods for control of luteal function.  相似文献

One or two trophoblastic vesicles (0.4-2 mm diam.) from cow (Day 14) or ewe (Day 11-13) embryos without their disc were transferred, after culture for 24 h, into recipients. Each vesicle was transferred into the uterine horn ipsilateral to the CL by the cervical route in heifers and surgically in ewes on Day 12 of the oestrous cycle. In cows, daily measurements of plasma progesterone concentrations and checks for return to oestrus showed that the CL was maintained in 8 out of 12 recipients. These 8 cows had 25- to 37-day cycles while 4 recipient heifers returned to oestrus normally. Three recipients with an extended cycle were slaughtered. The dissected uterus showed that trophoblastic vesicles had developed in the uterine horns. In ewes, the serum progesterone curve, determined in each recipient, showed that the CL was maintained in 7 out of 12 recipients. These 7 ewes had 20- to 54-day cycles and the other 5 ewes had a normal cycle of 15-19 days comparable to that of 17.0 +/- 0.5 days for the 6 control ewes. Whenever the CL was maintained, high blood progesterone levels were followed by rapid luteolysis. In cattle and sheep, therefore, a trophoblastic vesicle transferred into the uterus can develop in vivo, secreting the embryonic signals when there is no embryonic disc control and transforming the cyclic CL into a CL of pregnancy in about 60% of the cases.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
