PGI2 and 6-keto-PGF were converted to 6-methoxime-PGF (6-MeON-PGF) by treatment with methoxyamine HCl in acetate buffer. The formed 6-MeON-PGF was measured by radioimmunoassay. Antisera were raised in rabbits after immunization against 6-MeON-PGF-BSA conjugate. Diluted 1:20.000 to bind 50% of the tracer (3H-6-MeON-PGF, 100 Ci/mmol), the antiserum cross reacted 0.8% with PGE2, 1% with PGF and less than 0.2% with PGD2, PGF, PGF and TXB2. The radioimmunoassay was used to estimate release of PGI2 and 6-keto-PGF from chopped rabbit renal medulla and cortex incubated in Krebs-Ringer bicarbonate buffer (37°C, 30 min). The 6-keto-PGf radioimmunoassay was validated in biological samples by mass fragmentography. The chopped medulla (n=5) released 38±9 ng/g/min and the cortex (n=5) 4.7±2.0 ng/g/min, while the release of immunoreactive PGE2 (iPGE2) and iPGF was 171±26 and 74±13 ng/g/min from the medulla and 4.3±1.3 and 2.7±0.3 ng/g/min from the cortex, respectively. The results confirm previous findings, which indicate that in the renal medulla prostaglandin endoperoxides are mainly transformed to prostaglandins, while in the cortex transformation to PGI2 seems to be of greater importance.  相似文献

The structures of (H2tmen)[PtCl4], (1), (H2tmen = N,N,N′,N′-tetramethylethylenediammonium), [triclinic, P ; A = 7.344(3), B = 8.345(3), C = 6.216(2) Å, α = 84.53(3), β = 109.22(3), γ = 69.43(3)°, Z = 1] and (H2tmen)[PtCl6], (2), [monoclinic, P21/a; A = 14.409(4), B = 12.736(7), C = 8.601(3) Å, β = 99.58(3)°, Z = 4] were determined from diffractometric data by Patterson and Fourier methods and refined by full-matrix least-squares to R = 0.027 and 0.039 for (1) and (2) respectively. In both cases the anions and cations are joined in polymeric chains through hydrogen bonds involving the protonated nitrogens and the co-ordinated chlorine atoms. The square-planar [PtCl4]2− and octahedral [PtCl6]2− anions are centrosymmetric; the H2tmen2+ cations are centrosymmetric in (1) with a N-C-C-N dihedral angle of exactly 180°, while in (2) the dihedral angle is 166°. The different symmetry of the organic moieties/Ci and C1 in (1) and (2) respectively] results in a different i.r. spectrum which is more complex the lower the symmetry; moreover the spectrum of a KBr pellet of (2) changes with time, finally resembling that of (1) and of the (H2tmen)Cl2 salt.  相似文献

X-ray crystal analyses of divalent copper, cobalt and calcium complexes of monoanionic (3-hydroxy-5-(hydroxymethyl)-2-methylisonicotinic acid) 5-phosphate (L1C8H9NO7P) revealed the chemical compositions of Cu ---L·3H2O(1), Co ---L·5H2O(2) and Ca·L2·7H2O (3) and the coordination structures which depend on the coordination abilities and chemical properties of the respective metal ions. Although 1 and 2 crystals showed similar features, i.e., presence of the metal ion at the crystallographic center of symmetry and octahedral six-coordination, the patterns of coordination with the ligand molecules differed. While direct coordination to the L carboxyl oxygen was observed in 1 crystals, all ligation positions in 2 crystals were occupied by water molecules. On the other hand, 3 crystals formed a pentagonal bipyramidal structure (seven-coordination), where oxygens of L phosphates and water molecules coordinated to the calcium ion. Each of the complex structures showed characteristic molecular packing depending on the pattern of coordination to the respective metal ion. L is monoanionic in all complex crystals, where the phosphate and carboxyl groups are deprotonated and pyridine nitrogen is protonated, and is neutralized by each metal ion. Neuropsin (Mr25 032) is a serine protease expressed in the limbic system of mouse brain. It has been implicated in various neurological processes including formation of memory and may be important as a drug target in the treatment of epilepsy. The recombinant protein was produced using a baculovirus expression system and was purified. Two crystal forms were obtained by a hanging-drop vapor-diffusion method with polyethylene glycol. Preliminary X-ray crystallographic analysis revealed that crystal form I belongs to triclinic space groupP1 with unit cell dimensionsa= 97.16 Å,b= 97.12 Å,c= 46.75 Å and α = 99.17°, β = 99.77°, γ = 117.35°. Self-rotation function analysis of these data of form I indicates the position of a noncrystallographic threefold axis. There are six molecules in the crystallographic asymmetric unit. Crystal form II also belongs to triclinic space groupP1 but has unit cell dimensions ofa= 38.40 Å,b= 55.16 Å,c= 65.37 Å and α = 95.38°, β = 89.98°, γ = 110.46° with two molecules in the crystallographic asymmetric unit. Form II has a noncrystallographic twofold axis. Intensity data to 3.1 Å resolution for form I and to 2.2 Å resolution for form II have been collected.  相似文献

Lamellar single crystals of a high temperature polymorph of synthetic dextran were prepared at temperatures ranging from 120 to 200°C in a mixture of water and polyethylene glycol. Individual crystals with lath-like shapes gave well resolved electron diffraction diagrams from which the reciprocal unit cell parameters a*, b* and γ* could be measured. The direct cell parameters were then determined from a series of electron diffraction diagrams obtained by sequential tilting of the crystal about the b* axis. This gave a = 0·922 ± 0·001 nm, b = 0·922 ± 0·001 nm, c (chain axis) = 0·78 ± 0·01 nm, α = γ = 90° and β = 91·3° ± 0·5°. The crystal symmetry was P21 with b as the unique monoclinic axis. These data coupled with the observed density of the crystals, indicated that the unit cell contained two antiparallel dextran chains of two residues each. When the crystals were grown at temperatures between 90 and 120°C, a percentage of crystals containing both low and high temperature polymorphs were obtained. These mixed crystals had most likely grown in syntaxy.  相似文献

The thermal coefficient of expansion of egg lecithin bilayer thickness, αd1, was measured as a function of its cholesterol content up to mole ratio lecithin/cholesterol of 1:1, and over the temperature range 0–40 °C. At all cholesterol contents αd1 changes abruptly at approximately 12 °C indicating a structural transition at this temperature. Above 12 °C, αd1 decreases monotonically from −2·10−3 for pure egg lecithin to −1·10–3 at mole ratio 1:1. Below 12 °C αd1 is walways higher than above 12 °C and shows a sharp, anomalously high value of −6·10−3 at the mole ratio 2:1. The results have been interpreted as the movement of cholesterol into the bilayer or the formation of lecithin-cholesterol “complexes” at temperatures below 12 °C. Similar studies with phosphatidylinositol containing cholesterol showed no structural transition and lysolecithin containing cholesterol behaved differently giving two lamellar phases in equilibrium.  相似文献

We have obtained two new crystal forms of theAscarismajor sperm protein (MSP) that mediates amoeboid cell motility in nematode sperm. We obtained crystals with C2 symmetry from bacterially expressed α-MSP witha= 216.5 Å,b= 38.6 Å,c= 32.5 Å, γ = 93.1° and also crystals with P21symmetry from native β-MSP witha= 63.1 Å,b= 91.7 Å,c= 72.5 Å, γ = 91.3°. A full native data set has been collected for each crystal form using synchrotron radiation. Both crystal forms diffract to 2 Å and are suitable for high-resolution structural investigation.  相似文献

High pressure liquid chromatography (HPLC) using 4′ × 1/8″ columns of a pellicular silica support (Corasil-II) allows identification of prostaglandins diastereomers based on their characteristic retention relative to a standard, PGE2 in this study. Surprisingly this simple method allows separation of PGE2, PGE1, and PGEo (dihydro-PGE1) or PGF2α, PGF1α, and PGFoα without resort to silver nitrate impregnated stationary phases. Even more subtle distinctions such as that between 13,14-dihydro-PGF2α, PGF1α and 5,6- -PGF2α are possible by HPLC. The differential refractometer detector used throughout can also be used for quantitation. This is illustrated by a study of the relative rates of degradation of natural PGF2α (an oil at the temperature employed, 41°C) and racemic PGF2α (mp. 63°) on exposure to air.  相似文献

The structures of bis(1H+,5H+-S-methylisothiocarbonohydrazidium) di-μ-chlorooctachlorodibismuthate(III) tetrahydrate: (C2H10N4S)2(Bi2Cl10)· 4H2O (compound [I]) and of tris(1H+-S-methylisothiocarbonohydrazidium) esachlorobismuthate(III): (C2H9N4S)3(BiCl5.67I0.33) (compound [II]) were determined from single crystal X-ray diffractometer data. Both compounds crystallize as triclinic (P ); crystals [I] with Z = 1 formula unit in a cell of constants: A = 10.621(3), B = 9.989(5), C = 7.439(3) Å, α = 88.31(2), β = 84.51(2), γ = 68.88(2)°, final R = 0.0427 for 2229 unique reflections with I 2σ(I); crystals [II] with Z = 2 and cell dimensions: A = 14.109(4), B = 12.209(9), C = 8.206(7) Å, α = 103.54(3), β = 104.95(2), γ = 81.96(2)°, final R = 0.0411 for 3637 unique reflections (1 2σ(I)). The structure of [I] is built up of diprotonated organic cations, water molecules and dinuclear centrosymmetric [Bi2Cl10]4− anions held together by N-HCl, N-HO, O-HCl hydrogen bonds and Van der Waals interactions. The [Bi2Cl10]4− complex consists of two edge-sharing octahedra in which three pairs of bonds of similar length are observed (Bi-Clav = 2.602(5), 2.712(4), 2.855(5) Å). The structure of [II] consists of monoprotonated cations and [BiCl5.67I0.33]3− anions held together by a tridimensional network of hydrogen bonds. Each bismuth atom is octahedrally surrounded by six chlorine atoms, one of which is statistically substituted by a iodine atom.  相似文献

The compound [Cu2(bipy)2(OH)2](C4O4)·5.5H2O, where bipy and C4O42− correspond to 2,2′-bipyridyl and squarate (dianion of 3,4-dihydroxy-3-cyclo- butene-1,3-dione) respectively, has been synthesized. Its magnetic properties have been investigated in the 2–300 K temperature range. The ground state is a spin-triplet state, with a singlet-triplet separation of 145 cm−1. The EPR powder spectrum confirms the nature of the ground state.Well-formed single crystals of the tetrahydrate, [Cu2(bipy)2(OH)2](C4O4)·4H2O, were grown from aqueous solutions and characterized by X-ray diffraction. The system is triclinic, space group P , with a = 9.022(2), b = 9.040(2), c = 8.409(2) Å, α = 103.51(2), β = 103.42(3), γ = 103.37(2)°, V = 642.9(3) Å3, Z = 1, Dx = 1.699 g cm−3, μ(Mo Kα) = 17.208 cm−1, F(000) = 336 and T= 295 K. A total of 2251 data were collected over the range 1θ25°; of these, 1993 (independent and with I3σ(I)) were used in the structural analysis. The final R and Rw residuals were 0.034 and 0.038 respectively. The structure contains squarato-O1, O3-bridged bis(μ-hydroxo)bis[(2,2′-bipyridyl)copper(II)] units forming zigzag one-dimensional chains. Each copper atom is in a square-pyramidal environment with the two nitrogen atoms of 2,2′-bipyridyl and the two oxygen atoms of the hydroxo groups building the basal plane and another oxygen atom of the squarate lying in the apical position.The magnetic properties are discussed in the light of spectral and structural data and compared with the reported ones for other bis(μ-hydroxo)bis[(2,2′-bipyridyl)copper(II)] complexes.  相似文献

Axis determination in early Drosophila embryos is controlled, in part, by regulation of translation of mRNAs transcribed in maternal cells during oogenesis. The Pumilio protein is essential in posterior determination, binding to hunchback mRNA in complex with Nanos to suppress hunchback translation. In order to understand the structural basis of RNA binding, Nanos recruitment, and translational control, we have crystallized a domain of the Drosophila Pumilio protein that binds RNA. The crystals belong to the space group P63 with unit cell dimensions of a = b = 94.5 Å, c = 228.9 Å, α = β = 90°, γ = 120° and diffract to 2.6 Å with synchrotron radiation. We show that the purified protein actively binds RNA and is likely to have a novel RNA binding fold due to a very high content of α-helical secondary structure.  相似文献

Uranyl complexes of glycine, β-alanine and γ-aminobutanoic acid were prepared and characterized. All those studied or examined contain the aminoacids in the zwitterionic form binding the metal through the ionized carboxyl group. The structure of the title compound was determined by X-ray crystallography and refined to R=6.6%. The crystals are triclinic, space group P1, Z = 2, with a = 11.966(5), b = 12.054(5), c = 10.581(5) Å, α = 70.88(3)°, β = 109.89(3)°, and γ = 120.72(3)°. The uranyl group is equatorially bonded to the bidentate carboxylate of three molecules of the organic ligand forming a distorted hexagonal bipyramidal coordination geometry around the metal. U---O(equatorial) distances are in the range 2.24–2.48 Å.  相似文献

The preparation and X-ray structure of [Ag(9-EtGH-N7)2]NO3·H2O(9-EtGH=neutral 9-ethylguanine) is reported. The compound crystallizes in the triclinic system, space group P with a=7.063(6), b=7.153(3), c=11.306(10) Å, α=83.36(6), β=76.66(7), γ=81.44(6)°. The cation is centrosymmetric with Ag(I) coordinated via two N7 positions and Ag---N7 bond lengths of 2.11(1) Å. Applying 109Ag NMR spectroscopy, complex formation constants for both the 1:1 complex (log β1=0.6) and the title compound (log β2=1.6) in Me2SO have been determined.  相似文献

Cycloaddition reactions with α,β-unsaturated carbene complexes of the Fischer-type bearing the carbene carbon atom and the double bond incorporated in the same ring are described. Pentacarbonyl(2H-benzopyran-2- ylidene)chromium(0) complexes (2a-c) and pentacarbonyl(4-methoxy-3,3-dimethyl-2-oxacyclopentylidene)- chromium(0) (3) show a rather low reactivity towards 1,3-dipoles and 1,3-dienes. The reactions with diazomethane are regioselective but not chemoselective; compounds 2 and 3 show two sites of attack: the α,β carbon-carbon and the carbon-metal double bond. The crystal and molecular structures of 2a and 3 have been elucidated by single crystal X-ray analysis. Crystals of 2a are monoclinic, space group P21/c, a=7.614(3), b=14.033(3), c=12.766(3) Å, β=95.24°, V=1358.3(7) Å Z=4; crystals of 3 are triclinic, space group P , a=6.553(1), b=9.408(1), c=10.620(1) Å α=92.70(1), β=92.30(1), γ=92.12(1)°, V=653.0(1), Å3, Z=2. Final agreement indices for 2a and 3 are R=0.034 and 0.033, respectively. Vibrational properties of the Cr(CO)5 moiety were interpreted by FT-IR and FT-Raman spectroscopy. Electronic spectra and π electron distribution were interpreted by resonance Raman spectroscopy.  相似文献

Receptors for α2-macroglobulin-proteinase complexes have been characterized in rat and human liver membranes. The affinity for binding of 125I-labelled α2-macroglobulin · trypsin to rat liver membranes was markedly pH-dependent in the physiological range with maximum binding at pH 7.8–9.0. The half-time for association was about 5 min at 37°C in contrast to about 5 h at 4°C. The half-saturation constant was about 100 pM at 4°C and 1 nM at 37°C (pH 7.8). The binding capacity was approx. 300 pmol per g protein for rat liver membranes and about 100 pmol per g for human membranes. Radiation inactivation studies showed a target size of 466 ± 71 kDa (S.D., n = 7) for α2-macroglobulin · trypsin binding activity. Affinity cross-linking to rat and human membranes of 125I-labelled rat α1-inhibitor-3 · chymotrypsin, a 210 kDa analogue which binds to the α2-macroglobulin receptors in hepatocytes (Gliemann, J. and Sottrup-Jensen, L. (1987) FEBS Lett. 221, 55–60), followed by SDS-polyacrylamide gel electrophoresis, revealed radioactivity in a band not distinguishable from that of cross-linked α2-macroglobulin (720 kDa). This radioactivity was absent when membranes with bound 125I-α1-inhibitor-3 complex were treated with EDTA before cross-linking and when incubation and cross-linking were carried out in the presence of a saturating concentration of unlabelled complex. The saturable binding activity was maintained when membranes were solubilized in the detergent 3-[(3-cholamidopropyl)dimethylammonio]profane sulfonate (CHAPS) and the size of the receptor as estimated by cross-linking experiments was shown to be similar to that determined in the membranes. It is concluded that liver membranes contain high concentrations of an approx. 400–500 kDa α2-macroglobulin receptor soluble in CHAPS. The soluble preparation should provide a suitable material for purification and further characterization of the receptor.  相似文献

The structures of (4-bipyH)2[(μ-4-bipy)Nd2(NO3)8(H2O)4]·3(4-bipy) (4-bipy = 4,4′-bipyridine; P21/c, a = 18.723(10), b = 10.720(6), c = 18.027(10) Å, β = 94.43(5)°, Z = 2; R = 0.066 for 4931 (diffractometer data) and of a second monoclinic form of [Ho(NO3)3(H2O)3]·2(4-bipy) (P21/c, a = 15.830(10), b = 21.44(3), c = 15.70(3) Å, β = 100.4(2)°, Z = 8; R = 0.091 for 2335 film data) are reported. In the first compound pairs of Nd atoms are bridged across a crystal inversion centre by a 4-bipy ligand, and 10-coordination is completed by one monodentate NO3, three bidentate NO3, and two H2O ligands, with bond lengths Nd---N 2.70, Nd---OH2(av.) 2.44, Nd---O(NO3, av.) 2.56 Å. The second compound has a variant of the previously-reported monoclinic [Y(NO3)3(H2O)3]·2(4-bipy) structure, with doubling of the unit cell on a but with essentially no change in the geometry and orientation of the nine-coordinate complex. In both compounds the non-coordinated, non-protonated 4-bipy N atoms form hydrogen bonds with ligand H2O.  相似文献

The molecular conformation of the monoclinic crystalline polymorph of prostaglandin A1 has been determined by X-ray diffraction techniques. The space group is P21 with a = 13.637 (2), b = 7.567 (1), I c = 10.576 (2) Å, β = 107.37 (3)°; Dc = 1.073 g·cm−3 for Z = 2. The molecular conformation is characterized by the nearly parallel arrangement of the C1–C7 and C13–C20 side chains, with a general flattening of the overall structure when compared with the orthorhombic polymorph. The cyclopentenone moiety assumes a C8 envelope conformation with C8 and O9 displaced +0.29 Å and −0.18 Å from the C9–C10=C11–C12 plane respectively. Concerted, small variations of the torsion angles, primarily about the C8–C12, C14–C15 and C16–C17 bonds, bring the monoclinic and orthorhombic conformations into coincidence.  相似文献

An X-ray structural analysis of bis-2,2′,N,N′-bipyridyl ketone cobalt(III) nitrate dihydrate, CoC22H20N4O4+· NO3·2H2O,Mr=559.38 g/mol, P , a=8.862(2), b=16.195(3), c=8.772(2) Å, α=103.54(2), β=95.74(3), γ=105.07°, V=1164.4(4) Å3, Z=2, Dx=1.595 g/cm3, Mo Kα radiation (λ=0.71073 Å), μ=7.8 cm−1 and R=0.079, revealed a Co(III) cation in a slightly distorted octahedral environment. The structure reveals that the ligand di-2-pyridyl ketone (dpk) has undergone a hydration reaction across the ketone double bond and one of the hydrate oxygen atoms coordinated to the metal forming a tridentate chelate. This new Co(dpk-hydrate)2+ complex displays the least distorted geometry yet reported for either 1:1 or 1:2 (metal:ligand) complexes. A geometry optimization using the INDO model Hamiltonian as implemented in the program ZINDO was performed on the title complex with the Co3+ modeled as a singlet. The result of the computation corroborates the geometry of the title complex as that expected for Co3+.  相似文献

A water-soluble glucuronan “protuberic acid”, [α]d22 −83.6° and purified from Kobayashia Nipponica, and its physicochemical properties were investigated.The purified protuberic acid was homogeneous as shown by zone electrophoresis, gel filtration over Sepharose 4B, and ultracentrifugation. The sedimentation coefficient was 1.8 S and its intrinsic viscosity was 1.1 dl/g. By gel filtration the molecular weight was estimated to be about 170 000. The results of periodate oxidation, methylation analysis, and partial acid hydrolysis indicated that this acidic polysaccharide has a linear structure of mainly 1,4-linkages and containing an acid-labile linkage. Reduced protuberic acid, [α]d22 −44°, is also described.  相似文献

The compound (HOCH2CH2S) ) (1) has been prepared by the reaction of antimony(III) isopropoxide and 2-mercaptoethanol in a 1:2 molar ratio. Reaction of 1 with MOCH3 (where M = Li, Na and K) yields bimetallic products of the type, M[(OCH2CH2S) )]. All these derivatives have been characterized by elemental analysis, IR, NMR (1H and 13C) spectra and molar conductivity measurements. Crystals of 1 are triclinic, space group P , with a = 6.449(2), b = 10.285(2), c = 13.494(1) Å, α = 78.08(1), β = 75.99(1), γ = 71.54(2)°, V = 815.48 Å3, Z = 4, Dcalc = 2.239 g cm−3, (Mo Kα) λ = 0.7107 Å, μ = 3.55 mm−1, F(000) = 528, T = 295 K, final R = 0.0189 for 2344 reflections. One of the two mercaptoethanol moieties in 1 forms a five-membered chelate ring with antimony, Sb(1)---O(11) = 2.023(2) Å and Sb(1)---S(11) = 2.434(1) Å, while the other is bonded through the S atom only, Sb(1)---S(12) = 2.434(1) Å. The angles between these primary bonds with a mean value of 90.2° suggest a basically pyramidal, or pseudo tetrahedral structure if the stereochemically active lone pair is included in the coordination sphere. Two molecules are linked by intermolecular hydrogen bridges. The presence of weak intermolecular secondary bonding, Sb(1)---O(12) = 2.567(3) Å, in the complex indicates that the overall coordination polyhedron is best described in terms of a distorted trigonal bipyramidal arrangement.  相似文献
