This paper deals with the phylogenetic relationships in Sphagnum using the Wagner Groundplan-Divergence method. The main principle of the Wagner GPD method is grouping based on synapomorphy derived from outgroup comparison and ontogenetic analysis whenever possible. We make use of 21 available characters from gametophytes, estimate plesiomorphic conditions on the principle that characters found in the group under study and in most or all members of related groups are plesiomorphic, while those found only within the group are apomorphic, and finally arrange taxa according to shared derived features. In the Wagner tree diagrams, all characters can be placed on a Wagner tree so that the various relationships in the genus Sphagnum can be visualized. The results were as follows: 1. The genus Sphagnum can be divided into six sections, i.e. Sphagnum, Squarrosa, Acutifolia, Cuspidata, Subsecunda and Polyclada. They are almost consistent withe those derived from classical and numerical taxonomy of Sphagnum. 2. In Wagner tree diagrams, Section Sphagnum and Sect. Squarros were branched earlier, so they are primitive and the other four sections were last branched, so they are derived. Sphagnum magellanicum is a rather primitive species, whereus S. girgensohnii andS. fimbriatum are rather advanced ones in the 14 species of Sphagnum investigated.  相似文献

Peatlands can be classified into fens and bogs based on their hydrology. Development of fens to bogs is accompanied by the invasion of Sphagnum species. The purpose of this study was to determine how the decomposition process in fens is influenced by the transition from a vascular plant-dominated system to a Sphagnum -dominated system. We carried out a reciprocal litter bag experiment, using litter of Carex diandra , C. lasiocarpa , Sphagnum papillosum and S. squarrosum in a fen dominated by Sphagnum species and a fen without Sphagnum . Decomposition rate and nitrogen and phosphorus dynamics of the plant litter were measured in a field experiment for two years. Decomposition rate was highest for the Carex litter types and lowest for the Sphagnum litter types. Surprisingly, decomposition rates hardly differed between the two sites. Nutrient dynamics, however, showed a clear site-effect: In the Sphagnum site net mineralization was observed for all litter types whereas in the Carex site net immobilization was observed. These results show that carbon and nutrient cycles were coupled in a different way in a Sphagnum -dominated and a Carex -dominated site, respectively. Nutrient availability and adaptation of the microbial community to nutritional and other environmental conditions may be the main regulators of carbon and nutrient cycles in these peatlands.  相似文献

Haploid hybrid gametophytes are often present at low frequencies in sympatric populations of Sphagnum capillifolium and Sphagnum quinquefarium. We used intersimple sequence repeat (ISSR) markers and polymerase chain reaction-restriction fragment length polymorphism of the trnL(UAA) intron of the chloroplast genome to reveal the nuclear and chloroplast composition of mature hybrid gametophytes from natural populations and of gametophytes derived from spores of hybrid sporophytes collected in nature. Asymmetrical nuclear inheritance was found in the progeny of the hybrid sporophytes, indicating that only spores with a low level of recombination of parental genomes were viable. A similarly skewed nuclear composition was found among the naturally occurring hybrid gametophytes. All hybrid genomes contained a larger proportion of S. capillifolium ISSR markers, combined with only two to five S. quinquefarium markers together with a chloroplast haplotype derived from S. quinquefarium. In this way, a pattern resembling introgression is created within a single generation. Some individuals possessed nuclear genomes typical for S. capillifolium in combination with the chloroplast haplotype of S. quinquefarium, possibly indicating backcrossing. Our results indicate that hybridization between S. capillifolium and S. quinquefarium is relatively common, but the resistance of large parts of the genome against heterospecific genes maintains the genetic distinctness of the species. Further evolutionary and phylogenetic consequences of restricted interspecific gene exchange are discussed.  相似文献

It is commonly found that individual hybrid, polyploid species originate recurrently and that many polyploid species originated relatively recently. It has been previously hypothesized that the extremely rare allopolyploid peat moss Sphagnum troendelagicum has originated multiple times, possibly after the last glacial maximum in Scandinavia. This conclusion was based on low linkage disequilibrium in anonymous genetic markers within natural populations, in which sexual reproduction has never been observed. Here we employ microsatellite markers and chloroplast DNA (cpDNA)-encoded trnG sequence data to test hypotheses concerning the origin and evolution of this species. We find that S. tenellum is the maternal progenitor and S. balticum is the paternal progenitor of S. troendelagicum. Using various Bayesian approaches, we estimate that S. troendelagicum originated before the Holocene but not before c. 80 000 years ago (median expected time since speciation 40 000 years before present). The observed lack of complete linkage disequilibrium in the genome of this species suggests cryptic sexual reproduction and recombination. Several lines of evidence suggest multiple origins for S. troendelagicum, but a single origin is supported by approximate Bayesian computation analyses. We hypothesize that S. troendelagicum originated in a peat-dominated refugium before last glacial maximum, and subsequently immigrated to central Norway by means of spore flow during the last thousands of years.  相似文献

A seemingly obvious but sometimes overlooked premise of any evolutionary analysis is delineating the group of taxa under study. This is especially problematic in some bryophyte groups because of morphological simplicity and convergence. This research applies information from nucleotide sequences for eight plastid and nuclear loci to delineate a group of northern hemisphere peat moss species, the so-called Sphagnum subsecundum complex, which includes species known to be gametophytically haploid or diploid (i.e., sporophytically diploid-tetraploid). Despite the fact that S. subsecundum and several species in the complex have been attributed disjunct ranges that include all major continents, phylogenetic analyses suggest that the group is actually restricted to Europe and eastern North America. Plants from western North America, from California to Alaska, which are morphologically similar to species of the S. subsecundum complex in eastern N. America and Europe, actually belong to a different deep clade within Sphagnum section Subsecunda. One species often considered part of the S. subsecundum complex, S. contortum, likely has a reticulate history involving species in the two deepest clades within section Subsecunda. Nucleotide sequences have a strong geographic structure across the section Subsecunda, but shallow tip clades suggest repeated long-distance dispersal in the section as well.  相似文献

Phylogeographic studies of flora in species-rich south-western Australia point to complex evolutionary histories, reflecting patterns of persistence and resilience to climatic changes during the Pleistocene. We asked whether coastal areas of the mid-west and south, as well as granite outcrops and inland ranges, have acted as major refugia within this region during Pleistocene climatic fluctuations by analysing phylogeographic patterns in the shrub Calothamnus quadrifidus R.Br. (Myrtaceae). We determined variation in chloroplast DNA data for 41 populations across the geographic range. Relationships and major clades were resolved using parsimony and Bayesian analyses. We tested for demographic and spatial expansion of the major clades and estimated clade divergence dates using an uncorrelated, lognormal relaxed clock based on two conservative chloroplast mutation rates. Two distinct phylogeographic clades were identified showing divergence during the Pleistocene, consistent with other phylogeographic studies of south-west Australian flora, emphasising the impact of climatic oscillations in driving divergence in this landscape. The southern clade was more diverse, having higher haplotype diversity and greater genetic structure, while the northern clade showed evidence of fluctuation in population size. Regions of high haplotype diversity with adjacent areas of low diversity observed in each clade indicated the locations of two coastal refugia: one on the south coast and another along the mid-west coast. This is the first evidence for major Pleistocene refugia using chloroplast genetic data in a common, widespread species from this region.  相似文献

BACKGROUND AND AIMS: Ozone effects on peatland vegetation are poorly understood. Since stress responses are often first visible in cell ultrastructure, electron microscopy was used to assess the sensitivity of common peatland plants to elevated ozone concentrations. METHODS: Three moss species (Sphagnum angustifolium, S. magellanicum and S. papillosum), a graminoid (Eriophorum vaginatum) and two dwarf shrubs (Vaccinium oxycoccus and Andromeda polifolia), all growing within an intact canopy on peat monoliths, were exposed to a concentration of 0, 50, 100 or 150 ppb ozone in two separate growth chamber experiments simulating either summer or autumn conditions in central Finland. After a 4- or 5-week-long exposure, samples were photographed in a transmission electron microscope and analysed quantitatively using image processing software. KEY RESULTS: In the chlorophyllose cells of the Sphagnum moss leaves from the capitulum, ozone exposure led to a decrease in chloroplast area and in granum stack thickness and various changes in plastoglobuli and cell wall thickness, depending on the species and the experiment. In E. vaginatum, ozone exposure significantly reduced chloroplast cross-sectional areas and the amount of starch, whereas there were no clear changes in the plastoglobuli. In the dwarf shrubs, ozone induced thickening of the cell wall and an increase in the size of plastoglobuli under summer conditions. In contrast, under autumn conditions the cell wall thickness remained unchanged but ozone exposure led to a transient increase in the chloroplast and starch areas, and in the number and size of plastoglobuli. CONCLUSIONS: Ozone responses in the Sphagnum mosses were comparable to typical ozone stress symptoms of higher plants, and indicated sensitivity especially in S. angustifolium. The responses in the dwarf shrubs suggest stimulation of photosynthesis by low ozone concentrations and ozone sensitivity only under cool autumn conditions.  相似文献

Abstract

The distribution patterns of 18 Sphagnum species along base-richness and altitudinal gradients were studied in Bulgarian treeless wetlands which are noteworthy because of the edge-of-range occurrence of many mire species including Sphagnum. Of 483 spring and mire sites studied, 202 samples contained some Sphagnum species. The most common species were S. subsecundum (n=85), S. platyphyllum (46), S. contortum (41), S. teres (40) and S. capillifolium (26). The significance of Sphagnum responses to environmental gradients was tested by comparing generalized additive models against the null model. Many Sphagnum species displayed a significant response to the altitudinal gradient. Several species were clearly linked to low or to high altitudes, but the realized niche of other species was wide with respect to altitude. Most species significantly responded to water pH, both above and below the timberline. The same result was obtained for water conductivity below the timberline, whereas only a few species had a significant response to conductivity above the timberline. The highest water conductivity under which Sphagnum species occurred was 280 μS cm?1. Sphagnum contortum was the species occupying the mires with the highest mineral content. On the contrary, Sphagnum warnstorfii, one of the most calcitolerant species in many regions of Europe, often occurred in extremely mineral-poor mires above the timberline. Some other Sphagnum species growing in mineral-rich mires below the timberline, also inhabited extremely mineral-poor mires above the timberline. This could be explained by adaptation to local conditions during long-term isolation on mineral-poor bedrock or by changed competition pressure.  相似文献

Allopolyploidy is probably the most extensively studied mode of plant speciation and allopolyploid species appear to be common in the mosses (Bryophyta). The Sphagnum subsecundum complex includes species known to be gametophytically haploid or diploid, and it has been proposed that the diploids (i.e., with tetraploid sporophytes) are allopolyploids. Nucleotide sequence and microsatellite variation among haploids and diploids from Newfoundland and Scandinavia indicate that (1) the diploids exhibit fixed or nearly fixed heterozygosity at the majority of loci sampled, and are clearly allopolyploids, (2) diploids originated independently in North America and Europe, (3) the European diploids appear to have the haploid species, S. subsecundum, as the maternal parent based on shared chloroplast DNA haplotypes, (4) the North American diploids do not have the chloroplast DNA of any sampled haploid, (5) both North American and European diploids share nucleotide and microsatellite similarities with S. subsecundum, (6) the diploids harbor more nucleotide and microsatellite diversity than the haploids, and (7) diploids exhibit higher levels of linkage disequilibrium among microsatellite loci. An experiment demonstrates significant artifactual recombination between interspecific DNAs coamplified by PCR, which may be a complicating factor in the interpretation of sequence-based analyses of allopolyploids.  相似文献

I made a dispersal experiment with six Sphagnum (peat moss) species, representing a wide range of spore and capsule sizes. Spore deposition was recorded at nine distances up to 3.2 m during two four-day periods with sunny conditions in July and August.
Deposition patterns in all species fitted well to the inverse power law (deposition per unit area proportional to distanceb), with rates of decline b ranging from −1.84 to −2.35 among species (R2>0.99). However, even when these curves are extended to infinity for the four species with b<−2, they fail to explain all spores being dispersed (e.g. only 11% in S. squarrosum ). Difference in rates of decline b between the periods, despite similar horizontal wind speeds and directions, indicates the importance of thermal updrafts. Spore capsule diameter negatively correlated with the proportions of spores remaining in dehisced capsules (range 5–16%), being deposited within the colony (range 2–14%), and being deposited between the colony edge and the outer sampled perimeter (range 7–22%), probably because a larger capsule shoots the spores higher into the air, in effect meaning an increased initial release height. Spore diameter positively affected deposition outside the colony edge. The deposition curves together with observed long-distance colonisations, omnipresence in mires of many Sphagnum species and genetic evidence, suggest that Sphagnum spores regularly travel far.
Spatial extension of the empirical deposition curves at a regional scale indicate that with increasing isolation of target sites, the higher the proportion of spores would originate from sources farther away than the nearest sources. Given spatial patterns in the source populations, this would result in higher genetic (and species) diversity per coloniser with increasing isolation, thus counteracting differentiation.  相似文献

The class Sphagnopsida (Bryophyta) includes two genera: Ambuchanania and Sphagnum. Ambuchanania contains just one rare species known from two Tasmanian localities, but Sphagnum comprises a speciose clade of mosses that dominates many wetland ecosystems, especially in the boreal zone of the Northern Hemisphere. Recent phylogenetic analyses have resolved well-supported clades within Sphagnum, but polarizing Sphagnum evolution has been problematic because the genus is so isolated that it is difficult to determine homologies between morphological and/or molecular traits within Sphagnum with those of any potential outgroup. DNA sequences from 16 genomic regions representing the mitochondrial, chloroplast, and nuclear genomes (ca. 16 kilobases) were obtained from 24 species of Sphagnum plus one species each from Takakia and Andreaea in order to resolve a rooted phylogeny. Two tropical species, S. sericeum and S. lapazense, were resolved as sister to the rest of the genus and are extremely divergent from all other sphagna. The main Sphagnum lineage consists of two clades; one includes the sections Sphagnum, Rigida, and Cuspidata, and the other includes Subsecunda, Acutifolia, and Squarrosa. The placement of section Subsecunda is weakly supported, but other nodes are strongly supported by maximum parsimony, maximum likelihood, and Bayesian analyses. In addition to homogeneous Bayesian analyses, heterogeneous models were employed to account for different patterns of nucleotide substitution among genomic regions.  相似文献

This paper documents the occurrence of allotriploidy (having three differentiated genomes) in gametophytes of two Southern Hemisphere Sphagnum species ( S. australe, S . falcatulum ). The pattern of microsatellite alleles indicates that both species are composed of a complex of allodiploid and allotriploid gametophytes, with the latter resulting from two allopolyploidization events. No haploid ( n  =  x ) gametophytes were found for either species. The ploidal levels suggested by the pattern of microsatellite alleles were confirmed by flow cytometry and Feulgen DNA image densitometry. For both S. australe and S. falcatulum , the respective allodiploid plants (or their ancestors) are one of the parent species of the allotriploid plants. This is the first report of triploidy in Sphagnum gametophytes occurring in nature and also the first report of the presence of three differentiated genomes in any bryophyte. It is also the first report of intersectional allopolyploidy in Sphagnum , with S. australe appearing to have parental species from Sphagnum sections Rigida and Sphagnum, and S. falcatulum having parental species from Sphagnum sections Cuspidata and Subsecunda . In both species, the allotriploid cytotypes were the most prevalent cytotype on the South Island of New Zealand. The pattern of microsatellite alleles shows the presence of two genetically distinct populations of allodiploid S. australe , possibly indicating multiple origins of polyploidy for that allodiploid cytotype. Morphological evidence is also highly indicative of recurrent polyploidy in the allotriploid cytotype of S. falcatulum . Allopolyploidy has clearly played a major evolutionary role in these two Southern Hemisphere taxa. This study, in conjunction with other recent research, indicates that allopolyploidy is a common, if not the predominant, form of polyploidy in Sphagnum .  相似文献

Nitrogen fertilization reduces Sphagnum production in bog communities   总被引:3,自引:1,他引:2
The effects of increased nitrogen influx on Sphagnum growth and on interspecific competition between Sphagnum species were studied in a 3-yr experiment in mires situated in two areas with different rates of airborne N deposition. Sphagnum growth was recorded after various supplementary N influxes (0, 1, 3, 5 and 10 g m −2 yr−1) in hummocks and lawn communities. Sphagnum biomass production decreased with increasing N influx in both areas. After the first season at the low-deposition site, Sphagnum showed an increased growth in length with the intermediate N treatment, but in the second and third seasons the control treatment had the highest growth in length. Capitulum dry mass increased with increasing N influx. Sphagnum N concentration and N/P quotient were higher at the high- than at the low-deposition site. The low quotient at the low-deposition site, together with the initial growth increase with intermediate N supplements, indicates that growth was N-limited at this site, but our lowest N supplement was sufficient to reduce growth. The N treatments had no effect on interspecific competition between the Sphagnum species. This indicates that the species have similar responses to N. The species studied all occur naturally on ombrotrophic, N-poor sites and show low tolerances to increased N influx. Reduced Sphagnum production may affect the carbon balance, changing the mires from C sinks to sources.  相似文献

Experimental evidence for a persistent spore bank in Sphagnum   总被引:1,自引:0,他引:1
Spore capsules of four Sphagnum species were buried at different depths in peat on a bog. Spore viability was determined after 0, 1, 2 and 3 yr. Viability generally declined with time, but viable spores were still found at all depths after 3 yr. The light-coloured spores of S. balticum and S. tenellum retained their viability better than the darker spores of S. fuscum and S. lindbergii . Survival was highest under wet but aerobic conditions, but was also high under humid or periodically desiccated conditions. By contrast, most spores stored under wet, anaerobic conditions died within 2–3 yr. These results, and predictions from them, are not consistent with earlier results for spores of long-lived and dominant bryophytes, or for seeds of phanerogams of undisturbed wetlands and forests. There was no correlation between spore size and longevity across species, but the small spores from small capsules of S. balticum and S. tenellum generally showed higher viability than those from the medium-sized and large capsules of the same species. This suggests a positive intraspecific relationship between longevity and dispersal distance. There was an indication of conditional dormancy, controlled by weather, in Sphagnum spores. The experiments indicate that Sphagnum spores can form a long-term persistent spore bank under suitable conditions, with a half-life of between 1 and 20 yr (mean across species of 2.6 and 5.0 yr at two depths studied), and with potential values in individual spore capsules of several decades, or even of centuries. Sphagnum spores kept refrigerated showed 15–35% viable spores after 13 yr. The capacity to form a persistent spore bank that can be activated whenever favourable conditions occur might help explain the wide geographical distribution of many Sphagnum species in the boreal and temperate zones, where they have managed to colonize almost every suitable patch of acidic, nutrient-poor wetland.  相似文献

To analyze reasons for inconclusive results of earlier chloroplast phylogenies in the grass genus Hordeum, we established a genealogy of chloroplast haplotypes by sequencing the trnL-trnF region in 875 individuals, covering all 31 species of the genus. Although the outcomes of phenetic and parsimony analyses of 88 haplotypes were ambiguous, a network approach showed that in Hordeum ancient chloroplast types co-occur with their descendants. Moreover, we found up to 18 different chloroplast haplotypes within a single species and up to 6 species sharing single haplotypes. Persisting polymorphisms together with incomplete lineage sorting occurred preferentially in the rapidly speciating New World taxa of the genus, where ancient chloroplast types have survived for at least 4 Myr. Lineages-through-time plots and a high number of missing chloroplast haplotypes indicated far-reaching extinction of chloroplast lineages in Europe and particularly the Mediterranean. Survival of these lineages in East Asia and North America resulted in chloroplast relationships that markedly differed from nuclear estimations of species relationships. Thus, even for the deepest splits in the genus, reaching back more than 9 Myr, no safe phylogenetic inference from chloroplast data is possible in Hordeum. The chloroplast genealogy, however, revealed biogeographic patterns and indicated processes involved in speciation in Hordeum. We conclude that the described phenomena are not restricted to Hordeum and that the knowledge of the chloroplast relationships within a genus is indispensable to prevent misinterpretation of phylogeographic data within single species.  相似文献

Several complexes of species in Sphagnum (peat mosses) originated through hybridization and allopolyploidy, suggesting that these processes have played a major evolutionary role in this genus. The Sphagnum subsecundum complex includes gametophytically haploid and diploid species in North America. Analyses of 12 microsatellite loci and sequences from two plastid DNA markers show that the evolutionary history of this group is substantially more complex than previously thought. Two taxonomic species, Sphagnum lescurii and Sphagnum inundatum, include both haploid and diploid populations. Within each ploidal level, S. lescurii and S. inundatum are not genetically differentiated. The diploid taxa show patterns of fixed heterozygosity for the microsatellite markers, consistent with an allopolyploid origin. Diploid S. lescurii is an allopolyploid between haploid S. lescurii and (haploid) S. subsecundum. Sphagnum carolinianum is an allopolyploid between haploid S. lescurii and an unknown parent. We detected homoploid hybridization between the haploids Sphagnum contortum and S. subsecundum. Finally, we report three samples of diploid Sphagnum platyphyllum (otherwise haploid) that have an allopolyploid origin involving north‐eastern haploid S. platyphyllum and an unidentified taxon. © 2010 The Linnean Society of London, Biological Journal of the Linnean Society, 2010, 99 , 135–151.  相似文献

Climate oscillations and landscape heterogeneity make the historical population processes of organisms of the Qinghai–Tibet Plateau quite complex. We used chloroplast DNA sequences and amplified fragment length polymorphism (AFLPs) to study the phylogeography and genetic variation of the Qinghai–Tibet Plateau endemic Spenceria ramalana Trimen. The parsimony network contains two star-like evolutionary units, nested into two nested clades, coinciding with geographic population groups indicated by samova software. The central group shows genetic homogeneity, indicating a bottleneck or founder effect of population dynamics. Margin groups contribute more haplotype diversity (CT), indicating potential refugia. Several expansion events during several interglacial periods were detected. The latest expansion of the eastern population contributed to the modern population structure of the central area. Four AFLP clusters were detected. Higher CT, a relict pattern of haplotype distribution and DW values of marginal populations, confirmed the location of potential montane refugia. Relict distribution pattern, nested clade phylogeographic analysis conclusions, and restricted gene flow detected by AFLPs indicated an isolation pattern of the populations of montane areas. Heterogeneity of landscape of the western and southern montane areas could be the main barrier for gene flow. Moisture conditions and ecotype transformation might help to build the phylogeographic pattern of this species. Neither chloroplast DNA markers nor AFLP was able to differentiate the two varieties of this species.  相似文献

Most phylogeographic studies have used maximum likelihood or maximum parsimony to infer phylogeny and bootstrap analysis to evaluate support for trees. Recently, Bayesian methods using Marlov chain Monte Carlo to search tree space and simultaneously estimate tree support have become popular due to its fast search speed and ability to create a posterior distribution of parameters of interest. Here, I present a study that utilizes Bayesian methods to infer phylogenetic relationships of the cornsnake (Elaphe guttata) complex using cytochrome b sequences. Examination of the posterior probability distributions confirms the existence of three geographic lineages. Additionally, there is no support for the monophyly of the subspecies of E. guttata. Results suggest the three geographic lineages partially conform to the ranges of previously defined subspecies, although Shimodaira-Hasegawa tests suggest that subspecies-constrained trees produce significantly poorer likelihood estimates than the most likely trees reflecting the evolution of three geographic assemblages. Based on molecular support, these three geographic assemblages are recognized as species using evolutionary species criteria: E. guttata, Elaphe slowinskii, and Elaphe emoryi [phylogeographic, maximum likelihood, maximum parsimony, bootstrap, Bayesian, Markov chain Monte Carlo, cornsnake, Cytochrome b, geographic lineages, E. guttta, E. slowinskii, and E. emoryi].  相似文献
