Conifers of the boreal zone encounter considerable combined stress of low temperature and high light during winter, when photosynthetic consumption of excitation energy is blocked. In the evergreen Pinus sylvestris L. these stresses coincided with major seasonal changes in photosystem II (PSII) organisation and pigment composition. The earliest changes occurred in September, before any freezing stress, with initial losses of chlorophyll, the D1-protein of the PSII reaction centre and of PSII light-harvesting-complex (LHC II) proteins. In October there was a transient increase in F0, resulting from detachment of the light-harvesting antennae as reaction centres lost D1. The D1-protein content eventually decreased to 90%, reaching a minimum by December, but PSII photochemical efficiency [variable fluorescence (Fv)/maximum fluorescence (Fm)] did not reach the winter minimum until mid-February. The carotenoid composition varied seasonally with a twofold increase in lutein and the carotenoids of the xanthophyll cycle during winter, while the epoxidation state of the xanthophylls decreased from 0.9 to 0.1 from October to January. The loss of chlorophyll was complete by October and during winter much of the remaining chlorophyll was reorganised in aggregates of specific polypeptide composition, which apparently efficiently quench excitation energy through non-radiative dissipation. The timing of the autumn and winter changes indicated that xanthophyll de-epoxidation correlates with winter quenching of chlorophyll fluorescence while the drop in photochemical efficiency relates more to loss of D1-protein. In April and May recovery of the photochemistry of PSII, protein synthesis, pigment rearrangements and zeaxanthin epoxidation occurred concomitantly. Indoor recovery of photosynthesis in winter-stressed branches under favourable conditions was completed within 3 d, with rapid increases in F0, the epoxidation state of the xanthophylls and in light-harvesting polypeptides, followed by recovery of D1-protein content and Fv/Fm, all without net increase in chlorophyll. Yellow-green foliage cultivars of four vegetables grown outdoors, i.e., Chinese mustard (Brassica rapa), Chinese kale (Brassica oleracea var. alboglabra), sweet potato (Ipomoea batatas) and Chinese amaranth (Amaranthus tricolor), had lower chlorophyll (Chl) (a+b) (29–36% of green cultivars of the same species), total carotenoids (46–62%) and ascorbate (72–90%) contents per leaf area. Furthermore, yellow-green cultivars had smaller photosystem II (PSII) antenna size (65–70%) and lower photosynthetic capacity (52–63%), but higher Chl a/b (107–156%) and from low (60%) to high (129%) ratios of de-epoxidized xanthophyll cycle pigments per Chl a content. Potential quantum efficiency of PSII (Fv/Fm) of all overnight dark-adapted leaves was ca. 0.8, with no significant difference between yellow-green and green cultivars of the same species. However, yellow-green cultivars displayed a higher degree of photoinhibition (lower Fv/Fm after illumination) when they were exposed to high irradiance. Although vegetables used in this study are of either temperate or tropical origin and include both C3 and C4 plants, data from all cultivars combined revealed that Fv/Fm after illumination still showed a significant positive linear regression with xanthophyll cycledependent energy quenching (qE) and a negative linear regression with photoinhibitory quenching (qI). Fv/Fm was, however, not correlated with nonphotochemical quenching (NPQ). Yet, a higher degree of photoinhibition in yellow-green cultivars could recover during the night darkness period, suggesting that the repair of PSII in yellow-green cultivars would allow them to grow normally in the field.  相似文献

Physiological and photosynthetic responses were investigated at three different depths of groundwater (DGW: 1.4, 2.4, and 3.4 m) in Elaeagnus angustifolia L., a locally adapted tree to the arid region in northwest China. Predawn leaf water potential and chlorophyll content declined gradually with the increasing DGW, whereas there was little effect on predawn variable-to-maximum chlorophyll fluorescence ratio F v/F m and leaf carotenoid compositions (xanthophyll cycle pool, neoxanthin, lutein, and β-carotene). Net photosynthetic rate (P n), quantum yield of PSII electron transport (ΦPSII), stomatal conductance (Gs), and intercellular CO2 concentration (Ci) declined obviously; however, P n decreased more than ΦPSII at deeper DGW. The photoinhibition of PSII at all three DGW occurred at midday in summer and increased as DGW increased. The ΔpH-dependent thermal dissipation and the level of de-epoxidation of the xanthophyll cycle at all three DGW reached their maxima at midday with the increase of light intensity. However, the fraction of functional PSII and light intensity at deeper DGW (2.4, 3.4 m) showed a negative correlation. This correlation suggested that most of violaxanthin was converted into zeaxanthin at midday, and the reversible inactivation of partial PSII reaction centers took place at deeper DGW. These results together suggest that both the xanthophyll cycle-dependent thermal dissipation and the reversible inactivation of partial PSII might have played important roles in avoiding the excess light-induced energy damage in leaves of this tree species at deeper DGW.  相似文献

The photosynthetic performances of regenerated protoplasts of Bryopsis hypnoides, which were incubated in seawater for 1, 6, 12, and 24 h, were studied using chlorophyll (Chl) fluorescence and oxygen measurements. Results showed that for the regenerated protoplasts, the pigment content, the ratios of photosynthetic rate to respiration rate, the maximal photosystem II (PSII) quantum yield (Fv/Fm), and the effective PSII quantum yield (ΦPSII) decreased gradually along with the regeneration progress, indicated that during 24 h of regeneration there was a remarkable reduction in PSII activity of those newly formed protoplasts. We assumed that during the cultivation progress the regenerated protoplasts had different photosynthetic vigor, with only some of them able to germinate and develop into mature thalli. The above results only reflected the photosynthetic features of the regenerated protoplasts at each time point as a whole, rather than the actual photosynthetic activity of individual aggregations. Further investigation suggested a relationship between the size of regenerated protoplasts and their viability. The results showed that the middle-sized group (diameter 20–60 μm) retained the largest number of protoplasts for 24 h of growth. The changes in Fv/Fm and ΦPSII of the four groups of differently sized protoplasts (i.e. < 20, 20–60, 60–100, and > 100 μm) revealed that the protoplasts 20–60 μm in diameter had the highest potential activity of the photosynthetic light energy absorption and conversion for several hours.  相似文献

We studied the developmental changes in photosynthetic and respiration rates and thermal dissipation processes connected with chloroplasts and mitochondria activity in etiolated wheat (Triticum aestivum L., var. Irgina) seedlings during the greening process. Etioplasts gradually developed into mature chloroplasts under continuous light [190 μmol(photon) m?2 s?1] for 48 h in 5-day-dark-grown seedlings. The net photosynthetic rate of irradiated leaves became positive after 6 h of illumination and increased further. The first two hours of de-etiolation were characterized by low values of maximum (Fv/Fm) and actual photochemical efficiency of photosystem II (PSII) and by a coefficient of photochemical quenching in leaves. Fv/Fm reached 0.8 by the end of 24 h-light period. During greening, energy-dependent component of nonphotochemical quenching of chlorophyll fluorescence, violaxanthin cycle (VXC) operation, and lipoperoxidation activity changed in a similar way. Values of these parameters were the highest at the later phase of de-etiolation (4–12 h of illumination). The respiration rate increased significantly after 2 h of greening and it was the highest after 4–6 h of illumination. It was caused by an increase in alternative respiration (AP) capacity. The strong, positive linear correlation was revealed between AP capacity and heat production in greening tissues. These results indicated that VXC in chloroplasts and AP in mitochondria were intensified as energy-dissipating systems at the later stage of greening (after 4 h), when most of prolamellar bodies converted into thylakoids, and they showed the greatest activity until the photosynthetic machinery was almost completely developed.  相似文献

Seasonal differences in the efficiency of open PSII units (F v/F m), leaf pigment composition and xanthophyll cycle conversion (Z+A)/(V+A+Z), leaf adenylate status, and photosynthetic capacity were investigated in Pinus ponderosa (Ponderosa pine) and Malva neglecta. In P. ponderosa, acclimation to winter involved a lower photosynthetic capacity, higher carotenoid to chlorophyll ratio, persistent reductions in F v/F m corresponding to persistent retention of Z+A, and no change in foliar ATP/ADP ratios. In contrast, M. neglecta characterized in winter exhibited higher rates of photosynthesis than in summer with no change in carotenoid to chlorophyll ratio, while small nocturnally persistent reductions in F v/F m were observed exclusively on colder winter nights when nocturnal retention of Z+A, and high ATP/ADP ratios were also present. Upon removal of winter-stressed leaves or needles from the field to room temperature, a portion of F v/F m relaxed within 15 min of warming and recovery was completed within 5 h in M. neglecta but required 100 h in P. ponderosa. In M. neglecta, the entire recovery of F v/F m correlated with decreases in the foliar ATP/ADP ratio, while in P. ponderosa this ratio remained unchanged. Possible ATP-dependent forms of sustained (Z+A)-dependent energy dissipation are discussed including a nocturnally retained pH gradient on cold winter nights. The slow recovery in pine involved not only retention of Z+A, but apparently also a persistent engagement of Z+A for energy dissipation via an unidentified mechanism. Tropical plants are sensitive to chilling temperatures above zero but it is still unclear whether photosystem I (PSI) or photosystem II (PSII) of tropical plants is mainly affected by chilling temperatures. In this study, the effect of 4°C associated with various light densities on PSII and PSI was studied in the potted seedlings of four tropical evergreen tree species grown in an open field, Khaya ivorensis, Pometia tomentosa, Dalbergia odorifera, and Erythrophleum guineense. After 8 h chilling exposure at the different photosynthetic flux densities of 20, 50, 100, 150 μmol m−2 s−1, the maximum quantum yield of PSII (F v /F m) in all of the four species decreased little, while the quantity of efficient PSI complex (P m) remained stable in all species except E. guineense. However, after chilling exposure under 250 μmol m−2 s−1 for 24 h, F v /F m was severely photoinhibited in all species whereas P m was relative stable in all plants except E. guineense. At the chilling temperature of 4°C, electron transport from PSII to PSI was blocked because of excessive reduction of primary electron acceptor of PSII. F v /F m in these species except E. guineense recovered to ~90% after 8 h recovery in low light, suggesting the dependence of the recovery of PSII on moderate PSI and/or PSII activity. These results suggest that PSII is more sensitive to chilling temperature under the moderate light than PSI in tropical trees, and the photoinhibition of PSII and closure of PSII reaction centers can serve to protect PSI.  相似文献

Seasonal differences in PSII efficiency (Fv/Fm), the conversion state of the xanthophyll cycle (Z + A)/ (V + A + Z), and leaf adenylate status were investigated in Euonymus kiautschovicus. On very cold days in winter, Fv/Fm assessed directly in the field remained low and Z + A high throughout day and night in both sun and shade leaves. Pre-dawn transfer of leaves from subfreezing temperatures in the field to room temperature revealed that recovery (increases in Fv/Fm and conversion of Z + A to violaxanthin) consisted of one, rapid phase in shade leaves, whereas in sun leaves a rapid phase was followed by a slow phase requiring days. The pre-dawn ATP/ADP ratio, as well as that determined at midday, was similar when comparing overwintering leaves with those sampled in the summer, although pre-dawn levels of ATP + ADP were elevated in all leaves during winter relative to summer. After a natural transition to warmer days during the winter, pre-dawn Fv/Fm and Z + A in shade leaves had returned to values typical for summer, whereas in sun leaves Fv/Fm and Z + A levels remained intermediate between the cold day in winter and the summer day. Thus two distinct forms of sustained (Z + A)-dependent energy dissipation were identified based upon their differing characteristics. The form that was sustained on cold days but relaxed rapidly upon warming occurred in all leaves and may result from maintenance of a low lumenal pH responsible for the nocturnal engagement of (Z + A)-dependent thermal dissipation exclusively on very cold days in the winter. The form that was sustained even upon warming and correlated with slow Z + A to violaxanthin conversion occurred only in sun leaves and may represent a sustained engagement of (Z + A)-dependent energy dissipation associated with an altered PSII protein composition. In the latter, warm-sustained form, uncoupler or cycloheximide infiltration had no effect on the slow phase of recovery, but lincomycin infiltration inhibited the slow increase in Fv/Fm and the conversion of Z + A to violaxanthin.  相似文献

Kalanchoë daigremontiana, a CAM plant grown in a greenhouse, was subjected to severe water stress. The changes in photosystem II (PSII) photochemistry were investigated in water‐stressed leaves. To separate water stress effects from photoinhibition, water stress was imposed at low irradiance (daily peak PFD 150 μmol m?2 s?1). There were no significant changes in the maximal efficiency of PSII photochemistry (Fv/Fm), the traditional fluorescence induction kinetics (OIP) and the polyphasic fluorescence induction kinetics (OJIP), suggesting that water stress had no direct effects on the primary PSII photochemistry in dark‐adapted leaves. However, PSII photochemistry in light‐adapted leaves was modified in water‐stressed plants. This was shown by the decrease in the actual PSII efficiency (ΦPSII), the efficiency of excitation energy capture by open PSII centres (Fv′/Fm′), and photochemical quenching (qP), as well as a significant increase in non‐photochemical quenching (NPQ) in particular at high PFDs. In addition, photoinhibition and the xanthophyll cycle were investigated in water‐stressed leaves when exposed to 50% full sunlight and full sunlight. At midday, water stress induced a substantial decrease in Fv/Fm which was reversible. Such a decrease was greater at higher irradiance. Similar results were observed in ΦPSII, qP, and Fv′/Fm′. On the other hand, water stress induced a significant increase in NPQ and the level of zeaxanthin via the de‐epoxidation of violaxanthin and their increases were greater at higher irradiance. The results suggest that water stress led to increased susceptibility to photoinhibition which was attributed to a photoprotective process but not to a photodamage process. Such a photoprotection was associated with the enhanced formation of zeaxanthin via de‐epoxidation of violaxanthin. The results also suggest that thermal dissipation of excess energy associated with the xanthophyll cycle may be an important adaptive mechanism to help protect the photosynthetic apparatus from photoinhibitory damage for CAM plants normally growing in arid and semi‐arid areas where they are subjected to a combination of water stress and high light.  相似文献

Drought and high-temperature stresses have been extensively studied; however, little is known about their combined impact on plants. In the present study, we determined the photosynthetic gas exchange, chlorophyll fluorescence, nitrogen level, and lipid peroxidation of the leaves of a perennial grass (Leymus chinensis (Trin.) Tzvel.) subjected to three constant temperatures (23, 29 and 32°C), and five soil-moisture levels (75–80%, 60–65%, 50–55%, 35–40% and 25–30% of field capacity, respectively). High temperature significantly decreased plant biomass, leaf green area, leaf water potential, photosynthetic rate (A), maximal efficiency of PSII photochemistry (F v/F m), actual PSII efficiency (ΦPSII), the activities of nitrate reductase (NR; EC 1.6.6.1) and glutamine synthetase (GS; EC 6.3.1.2), but markedly increased the ratio of leaf area to leaf weight (SLA), endopeptidase (EP; EC 3.4.24.11) activity, and malondialdehyde (MDA) content, especially under severe water stress conditions. The A and F v/F m were significantly and positively correlated with leaf-soluble protein content, and the activities of NR and GS. However, both photosynthesis parameters were significantly and negatively correlated with EP activity and MDA content (P < 0.05). It is suggested that high temperature, combined with severe soil drought, might reduce the function of PSII, weaken nitrogen anabolism, strengthen protein catabolism, and provoke lipid peroxidation. The results also indicate that severe water stress might exacerbate the adverse effects of high temperature, and their combination might reduce the plant productivity and distribution range of L. chinensis in the future.  相似文献

Dissipation of light energy absorbed by photosystem II (PSII) in assimilating shoots of an evergreen shrub Ephedra monosperma was investigated during its transition from the vegetative to frost-tolerant state under natural conditions of Central Yakutia. The dynamics of modulated chlorophyll fluorescence and carotenoid content was analyzed during seasonal decrease in ambient temperature. The seasonal cooling was accompanied by a stepwise decrease in photochemical activity of PSII (F v/F m = (F m ? F 0)/F m). The decrease in F v/F m occurred from the beginning of September to the end of October, when the temperature was lowered from 10 to ?8°C. During winter period the residual activity of PSII was retained at about 30% of the summer values. The seasonal decrease in temperature was accompanied by a significant stimulation of pH-independent dissipative processes in reaction centers and antenna of PSII. The increase in energy losses was paralleled by a proportional increase in zeaxanthin content on the background of decreasing content of violaxanthin and β-carotene as possible zeaxanthin precursors. At the same time, inhibition of light-induced non-photochemical quenching in the PSII antenna was observed. The results suggest that principal photoprotective mechanisms during seasonal lowering of temperature are: (1) inactivation of PSII and dissipation of excitation energy in PSII reaction centers and (2) zeaxanthin-mediated energy dissipation in the antenna complexes. The first mechanism seems to prevail at early stages of seasonal cooling, whereas both mechanisms are recruited from the onset of sustained freezing temperatures.  相似文献

We compared the responses of wild type (WT) and three mutants including npq1 (lutein-replete and violaxanthin deepoxidase-deficient), lut2 (lutein-deficient), and lut2-npq1 (double mutant) to high irradiance (HI, 2 000 μmol m−2 s−1) at both low (LT, 5 °C) and room (25 °C) temperature. Xanthophyll-dependent energy dissipation was highest in the WT, followed by the lut2, npq1, and npq1-lut2. At 25 °C the relative stress tolerance expressed by Fv/Fm was consistent with the energy dissipation capacity for the first 2 h of treatment. After 3–4 h, the Fv/Fm levels in lut2 and npq1 converged. Under combined LT and HI the relative tolerance sequence was in contrast to the energy dissipation capacity being WT > npq1> lut2 > lut2-npq1. There were little or no significant change in the contents of xanthophylls and carotenes or the chlorophyll (Chl) a/b ratio in any of the materials. Thus lutein (L) substitution possibly alters the conformation/organisation of L binding proteins to enhance damage susceptibility under HI at LT. The enhanced vulnerability is not compensated for the energy dissipation capacity in the lut2 background at LT. Members of the Rhodophyta present different carotenoid profiles. In a majority of the species, lutein constitutes >50% of the total carotenoid content, while in other species, it is replaced by zeaxanthin or antheraxanthin. Given that carotenoids have specific roles in photoprotection, different carotenoid profiles of red algae species could be related to their capacity to cope with photoinhibitory stress. Therefore, in the present work, the sensitivity to light stress of red algal species with different carotenoid profiles was investigated. Photoinhibition of photosynthesis induced by high‐light stress and the subsequent recovery in dim‐light conditions was measured using maximal PSII quantum efficiency (Fv/Fm). The degree of decrease and recovery of Fv/Fm and their respective kinetics were related to the carotenoid profile of the species. Although no relationship between sensitivity to high‐light stress and the carotenoid profile was observed, there were clear carotenoid profile‐related differences in the decrease and recovery kinetics. In species with zeaxanthin or antheraxanthin as the major carotenoid, Fv/Fm reduction and recovery was principally associated with slowly activated and relaxed processes. In contrast, in species with lutein as the major carotenoid, rapidly activated processes appear to play a major role in the down‐regulation of photosynthesis during light‐stress conditions. In these species, the repair of D1 is also important during light‐stress conditions. This finding could imply differential expression of mechanisms involved in photoprotection in red algae that seems to be related to the carotenoid profile of the species.  相似文献

The photosynthetic pigments and photochemical efficiency of photosystem 2 (PS2) were studied in four constitutive species (Achillea millefolium L., Festuca pseudovina Hack. ex Wiesb., Potentilla arenaria Borkh., and Thymus degenianus Lyka) of a semiarid grassland in South-eastern Hungary. Every species displayed typical sun-adapted traits and substantial plasticity in the composition and functioning of the photosynthetic apparatus. The contents of chlorophylls (Chls) and carotenoids (Cars) on a dry matter basis declined from May to July, however, the amount of total Cars on a Chl basis increased. This increase was the largest in Potentilla (48 %) and the smallest in Achillea (14 %). The pool of xanthophylls (VAZ) was between 25 % and 45 % of the total Car content and was larger in July than in May. The content of β-carotene increased by July, but lutein content did not change significantly. The Chl fluorescence ratio Fv/Fm was reduced by 3–10 % at noon, reflecting the down-regulation of PS2 in the period of high irradiance and high temperature. The occurrence of minimal values of ΔF/Fm’ showed close correlation to the de-epoxidation rate of violaxanthin. Hence in natural habitats these species developed a considerable capacity to dissipate excess excitation energy in the summer period in their photosynthetic apparatus through the xanthophyll cycle pool and a related photoprotective mechanism, when the photochemical utilization of photon energy was down-regulated.  相似文献

In the context of the search for future forestry species in Central Europe under climate change scenarios, the evergreen Mediterranean Quercus taxa Q. ilex ssp. ilex and Q. suber and the semideciduous hybrids. Q. × turneri and Q. × hispanica were studied in relation to their photochemical efficiency of photosystem II and to the activity of the xanthophyll cycle under Central European winter conditions. The evergreen taxa were remarkably insensitive to winter stress in the field and reacted towards frost periods with the phenomenon of “chronic photoinhibition”, i.e. a decrease in pre-dawn F v/F m and an increase in the deepoxidation state of the xanthophylls. Under dark and warmer conditions (room temperature), winter-acclimated leaves of the evergreen taxa except for Q. × turneri produced zea- and antheraxanthin and decreased F v/F m, possibly by creation of a chlororespiratory pH-gradient. It is suggested that the ability for dark violaxanthin deepoxidation may contribute to the winter hardiness of the evergreen taxa.  相似文献

This work aims to: (1) correlate photochemical activity and productivity, (2) characterize the flow pattern of culture layers and (3) determine a range of biomass densities for high productivity of the freshwater microalga Chlorella spp., grown outdoors in thin-layer cascade units. Biomass density, irradiance inside culture, pigment content and productivity were measured in the microalgae cultures. Chlorophyll-fluorescence quenching was monitored in situ (using saturation-pulse method) to estimate photochemical activities. Photobiochemical activities and growth parameters were studied in cultures of biomass density between 1 and 47 g L−1. Fluorescence measurements showed that diluted cultures (1–2 g DW L−1) experienced significant photostress due to inhibition of electron transport in the PSII complex. The highest photochemical activities were achieved in cultures of 6.5–12.5 g DW L−1, which gave a maximum daylight productivity of up to 55 g dry biomass m−2 day−1. A midday depression of maximum PSII photochemical yield (F v/F m) of 20–30% compared with morning values in these cultures proved to be compatible with well-performing cultures. Lower or higher depression of F v/F m indicated low-light acclimated or photoinhibited cultures, respectively. A hydrodynamic model of the culture demonstrated highly turbulent flow allowing rapid light/dark cycles (with frequency of 0.5 s−1) which possibly match the turnover of the photosynthetic apparatus. These results are important from a biotechnological point of view for optimisation of growth of outdoor microalgae mass cultures under various climatic conditions.  相似文献

The transfer of laboratory cultures of H. pluvialis to high irradiance outdoors caused a substantial decline in the maximum quantum yield of photosystem II (PSII), from 0.65 in the morning to 0.45 at midday, as measured by the ratio of variable to maximum fluorescence yields (Fv/Fm), and a steep rise in non-photochemical quenching (NPQ). Chlorophyll fluorescence induction curves of morning samples showed a clear I-step, reflecting a certain PSII heterogeneity. Single turnover flash measurements on samples taken from the outdoor photobioreactor in the middle of day showed an increase in the reoxidation time constant of the reduced plastoquinone QA , i.e., the time required for electron transfer from the primary plastoquinone acceptor of PSII QA to the secondary plastoquinone acceptor QB. Photosynthesis rates were almost constant during the day. Along with the increase in non-photochemical quenching, there was a slight increase in zeaxanthin and antheraxanthin contents and decrease in violaxanthin, showing the presence of an operative xanthophyll cycle in this microalga. A marked increase of secondary carotenoids was found at the end of the first day of exposure to sunlight, mainly astaxanthin monoester, which reached 15.5% of the total carotenoid content. Though cells turned reddish during the second day, the decline in the fluorescence parameter Fv/Fm in the middle of the day was less than during the first day, and there was no further increase in the value for NPQ. Similar behaviour was observed during the third day when the culture was fully red. After four days of exposure to sunlight, the dry weight reached 800 mg L–1 and the concentration of secondary carotenoids (81% astaxanthin monoester) reached 4.4% dry weight.  相似文献

Experimental ecology methods and chlorophyll fluorescence technology were used to study the effects of different concentrations of manganese (10−12– 10−4 mol L−1) on the growth, photosystem II and superoxide dismutase (SOD) activity of Amphidinium sp. MACC/D31. The results showed that manganese had a significant effect on the growth rate, fluorescence parameters (maximal photochemical efficiency of PSII (F v /F m ), photochemical quenching (qP) and non-photochemical quenching (NPQ)) in the exponential stage (days 1–3) and SOD activity of Amphidinium sp. (P < 0.05). F v/F m in the exponential stage in 10−12 mol L−1 manganese concentration was significantly lower whilst qP and NPQ significantly higher than those in the other concentrations. F v /F m (days 6–9) in 10−4 mol L−1 manganese was significantly higher than those in the other concentrations. F v /F m (days 3–6) increased with increased concentration of manganese from 10−12 to 10−4 mol L−1. The values of qP and NPQ decreased with decreased concentrations of manganese, except for those in days 4–6. F v /F m under each concentration increased earlier and decreased later with culture stage whilst NPQ decreased earlier and increased later. The SOD activity increased with increased concentration of manganese from 10−12 to 10−8 mol L−1. The SOD activity in 10−4 mol L−1 manganese was significantly higher than those in the other concentrations and in 10−12 mol L−1 manganese, it was significantly lower than those in the other concentrations.  相似文献

Near-isogenic lines of maize varying in their genes for flavonoid biosynthesis were utilized to examine the effects of foliar flavonoids and nutrient deficiency on maximum net photosynthetic rate (P N) and chlorophyll (Chl) fluorescence (Fv/Fm) in response to ultraviolet-B (UV-B) radiation. Plants with deficient (30 to 70 % lower N, K, Mn, Fe, and Zn) and sufficient nutrients were exposed to four irradiation regimes: (1) no UV-B with solar photosynthetically active radiation (PAR), (2) two day shift to ambient artificial UV-B, 8.2–9.5 kJ m−2 d−1 (21–25 mmol m−2 d−1); (3) continuous ambient artificial UV-B; (4) continuous solar UV-B in Hawaii 12–18 kJ m−2 d−1 (32–47 mmol m−2 d−1). The natural ratio of UVB: PAR (0.25–0.40) was maintained in the UV-B treatments. In the adequately fertilized plants, lines b and lc had higher contents of flavonoids and anthocyanins than did lines hi27 and dta. UV-B induced the accumulation of foliar flavonoids in lines hi27 and b, but not in the low flavonoid line dta or in the high flavonoid line lc. In plants grown on deficient relative to adequate nutrients, flavonoid and anthocyanin contents decreased by 30–40 and 40–50 %, respectively, and Chl a and Chl b contents decreased by 30 and 70 %, respectively. The UV-B treatments did not significantly affect P N and Fv/Fm in plants grown on sufficient nutrients, except in the low flavonoid lines dta and hi27 in which P N and Fv/Fm decreased by ∼15 %. P N, Fv/Fm, and stomatal conductance decreased markedly (20–30 %) in all lines exposed to UV-B when grown on low nutrients. The decrease in Fv/Fm was 10 % less in higher flavonoid lines b and lc. The photosynthetic apparatus of maize readily tolerated ambient UV-B in the tropics when plants were adequately fertilized. In contrast, ambient UV-B combined with nutrient deficiency significantly reduced photosynthesis in this C4 plant. Nutrient deficiency increased the susceptibility of maize to UV-B-induced photoinhibition in part by decreasing the contents of photoprotective compounds.  相似文献

The major light-harvesting complex (LHC) of the prasinophycean alga Mantoniella squamata is unique compared to other chlorophyll (Chl) a/b-binding LHC with respect to the primary protein structure and the pigmentation. Although the presence of Chl a, Chl b, a Chl c-type pigment and the xanthophylls neoxanthin, violaxanthin and prasinoxanthin was clearly determined, several carotenoids remained unidentified or were described controversially. We re-analysed the carotenoid composition and identified a new set of xanthophylls present in the LHC: uriolide, micromonol, micromonal and dihydrolutein. Additionally, one hydrophobic component was detected, presumably a xanthophyll. The pigment analysis in combination with quantitative protein determinations revealed a pigment-protein stoichiometry of 6 Chl a, 6 Chl b, 2 Chl c* and about 2 prasinoxanthin molecules per polypeptide. The other xanthophylls were present in sub-stoichiometric amounts. A comparison of results from LHC isolated either by sucrose density centrifugation or SDS-polyacryl gel electrophoresis revealed a decline in the amount of prasinoxanthin and a loss of violaxanthin using the latter preparation procedure, while the stoichiometric ratios of the other 6 xanthophylls remained constant. The fact that 8 different xanthophylls were found in the LHC of M. squamata can be explained best in terms of an oligomeric, presumably trimeric LHC organisation with subunits of heterogeneous pigmentation. Especially, the very stable assembly of most of the minor xanthophylls led to the assumption that these components play an important role in stabilisation and probably also in trunerisation of the LHC in vivo.
