Phospholipids activate cathepsin D   总被引:1,自引:0,他引:1  
Total lipids as well as phospholipids extracted from the mitochondrial-lysosomal fraction of porcine adrenal cortex activated the lysosomal cathepsin D of this tissue 30- and 40-fold, respectively, with bovine serum albumin as the substrate. Phosphatidic acid, phosphatidyl ethanolamine, phosphatidyl serine, phosphatidyl inositol, phosphatidyl glycerol and cardiolipin were found to activate greatly the cathepsin D. The degree of activation ranged from 6-fold by phosphatidyl ethanolamine to 40-fold by cardiolipin at 1 mM, respectively. These results strongly point to the importance of phospholipids in intracellular protein degradation by lysosomal cathepsin D.  相似文献

1. The predominant lipids of nerve cords, ganglion and brain from horseshoe crabs were cholesterol (11% of lipid) and phospholipid (81% of lipid). 2. Major phospholipids were phosphatidyl ethanolamine and phosphatidyl choline with lesser amounts of phosphatidyl serine and phosphatidyl inositol and sphingomyelin. 3. The phospholipid fraction was characterized by a high content of plasmalogen, i.e. alk-1-enyl acyl phosphatides, so that 42% of the ethanolamine phosphatides were the plasmalogen, phosphatidal ethanolamine. 4. Phosphatidyl choline and phosphatidyl ethanolamine were high in polyunsaturation with 20:4 and 20:5 major fatty acids. Sphingomyelin had predominantly long chain saturated fatty acids. 5. Cerebrosides and gangliosides, which are associated with vertebrate nerve tissues, were absent from nerves of horseshoe crabs.  相似文献

A lipoid anticoagulant (LA) from human brain tissue was shown to possess a higher anticoagulant activity than previously obtained preparations. It has been established that out of four phospholipids present in LA, only phosphatidyl serine inhibits the coagulant activity of plasma. In isolated hemocoagulating systems LA and phosphatidyl serine were shown to inhibit prothrombin conversion catalyzed by thrombokinase and to exert the antithrombic action in the system thrombin--fibrinogen. Kinetic study of phosphatidyl serine- and LA-induced inhibition of thrombine formation and the thrombine--fibrinogen reaction by conjugated inhibition. Similar kinetic behaviour, observed upon inhibition of both processes by phosphatidyl serine and LA suggest that phosphatidyl serine is a main anticoagulant agent of LA.  相似文献

A study was conducted on the in vivo incorporation of l -[14C]-serine into the lipids and proteins of the various subcellular fractions of the developing rat brain before and during the stage of active myelination. The total radioactivity in the various fractions at 12 days of age was higher than that at 3 days, while the radioactive specific activity was reversed. The specific activities of the proteins and lipids were higher at 3 days of age with the exception of the subcellular fraction containing myelin. At both ages the lipids of the various cellular fractions had similar specific activities, a finding that suggests a common source for lipid biosynthesis. Incorporation of radioactivity into the various phospholipids was in the following order: phosphatidyl serine > phosphatidyl ethanolamine > phosphatidal serine > sphingomyelin and phosphatidyl choline. Of all the phospholipids, the plasmalogens increased most in total radioactivity during the period when meylination was most active. Serine-containing phospholipids appear to be most tightly bound to proteins. The brain mitochrondrial fraction contained most of the phosphatidyl serine decarboxylase activity with some activity in the nuclei. Biosynthesis of phosphatdyil ethanolamine through decarboxylation of phosphatidyl serine could take place in rat brain. Four unidentified radioactive metabolites were found in the acid-soluble fraction in addition to l -[14C]serine.  相似文献

The positional distribution of the fatty acids in the major phospholipids of bovine retina rod outer segments was determined. Phosphatidyl ethanolamine and phosphatidyl serine have mostly saturated acids in the 1-position and docosahexaenoic acid in position 2. These phospholipids contain 94 and 79%, respectively, of polyun-saturated acids in the 2-position. Phosphatidyl choline contains mostly saturated acids in the 1-position, but has significant quantities of palmitic in the 2-position along with docosahexaenoic acid. The levels of docosahexaenoic acid in rod outer segment phospholipids are among the highest yet reported for membrane phospholipids, amounting to 23% in phosphatidyl choline, 39% in phosphatidyl ethanolamine, and 45% in phosphatidyl serine.  相似文献

Thin-layer chromatographic analyses were made of phospholipids extracted from adult Echinostoma revolutum (Trematoda) and from the nonnutrient medium in which worms were maintained. Identity of phospholipids was based on comparison with authentic lipid standards and on specific chemical detection tests. The most abundant phospholipids in the extract were phosphatidyl choline, phosphatidyl ethanolamine and phosphatidyl serine; lesser amounts of cerebrosides and lysophosphatidyl choline were detected also. Phospholipids detected in the incubation medium were essentially as described for extracts.  相似文献

Influence of NSE on lipid composition of metastases and the neighbouring conditionally normal lung tissue in mice with Lewis carcinoma was investigated. The processes of peroxidation in investigated tissues were also studied. It was shown that under the influence of NSE the high level of antioxidant activity in the metastases was decreased, while in the neighbouring conditionally normal lung tissue the catalase activity was increased. The content of the thiobarbituric acid-reactive substances in comparison with animals which were not fed by NSE was decreased. The development of carcinoma was accompanied by significant decrease of cholesterol level and by the increase of unsaturated fatty acids esterified in membrane phospholipids in both the metastases and the neighbouring conditionally normal lung tissue. An analysis of the phospholipid spectra shows that under tumor growth in investigated tissues the high-level lysophosphatidylcholine (LPC) was observed. The content of phosphatidyl choline (PC), phosphatidyl ethanolamine (PE), phosphatidyl serine (PS) was found to be significantly lower than in the lung of intact animals. It was found that administration of NSE to tumor-bearing mice contributed to the increase of cholesterol level, to the decrease of omega-6/omega-3 ratio polyunsaturated fatty acids of total phospholipids. NSE modulated the phospholipid membrane composition in both the metastases and the neighbouring conditionally normal lung tissue.  相似文献

Pure phosphatidyl ethanolamine and lecithin from egg yolks were fed to rats in saline or in olive oil and the changes in individual phospholipids in the intestinal wall, liver, and plasma of the animals were studied. Ingestion of olive oil alone produced increased levels of all phospholipid fractions in each of the three tissues. Feeding phosphatidyl ethanolamine in saline resulted in slightly increased plasma phospholipids, but levels of liver total phospholipids were greatly reduced; when phosphatidyl ethanolamine was fed with olive oil, liver phospholipids were again reduced but this reduction was confined to the phosphatidyl ethanolamine and phosphatidic acid fractions. Feeding lecithin alone did not produce significant changes in levels of plasma or tissue phospholipids. The results suggest that liver phospholipid synthesis is depressed by feeding phosphatidyl ethanolamine; in the presence of olive oil, hepatic synthesis of phosphatidyl ethanolamine seems to be more selectively inhibited.  相似文献

Summary The lipid content and composition from an axolemma-rich preparation isolated from squid retinal axons was analyzed.The lipids, which accounted for 45.5% of the dry weight of this membrane, were composed of 22% cholesterol, 66.7% phospholipids and 5.2% free fatty acids. The negatively charged species phosphatidyl ethanolamine (37%), phosphatidyl serine (10%) and lysophosphatidyl ethanolamine (4%) made up 51% of the phospholipids. The amphoteric phosphatidyl choline and sphingomyelin accounted for 39% and 4%, respectively.The relative distribution of fatty acids in each of the isolated phospholipids was studied. The most remarkable feature of these phospholipids was the large proportion of long-chain polyunsaturated fatty acids. The 226 acyl chain accounted for 37% in phosphatidyl ethanolamine, 21.7% in phosphatidyl choline, 17.5% on phosphatidyl serine and 20.3% in sphingomyelin (all expressed as area %).The molar fraction of unsaturated fatty acids reached 65% in phosphatidyl ethanolamine and 42.0 and 44.8% in phosphatidyl choline and phosphatidyl serine, respectively. The double bond index in these species varied between 1.0 and 2.6.The lipid composition of the axolemma-rich preparation isolated from squid retinal axons appears to be similar to other excitable plasma membranes in two important features: (a) a low cholesterol/phospholipid molar ratio of 0.61; and (b) the polyunsaturated nature of the fatty acid of their phospholipids.This particular chemical composition may contribute a great deal to the molecular unstability of excitable membranes.The preceding papers of this series were published inArchives of Biochemistry and Biophysics.  相似文献

The phospholipid content of the homogenate, nuclei and chromatin of rat thymus was being studied during three months after fractionated gamma-irradiation (2 Gy X 3 at a week interval). The number of phospholipids in the total fraction of phosphatidyl choline + phosphatidyl serine and phosphatidyl ethanolamine in the nuclei and chromatin of rat thymus was shown to decrease 60 min following the last exposure. In a month the phospholipid content in the nuclei and chromatin increased up to the control level keeping it throughout the entire period of observation.  相似文献

Abstract— A significant increase in arachidonate was noted in the total phospholipids of brain of chicks with nutritional antioxidant-deficiency and encephalomalacia. After thin-layer chromatography of the brain lipids, this increase in arachidonate was found to be restricted to the phosphatidyl serine fraction. Significant decreases in docosahexaenoate and docosapentaenoate were noted in the phosphatidyl ethanolamine fraction. The changes in fatty acid composition of phospholipids in chick brain are comparable to those previously observed in phospholipids of skeletal muscle, liver and testes of the rat.  相似文献

A comparison has been made between the unsaturation of plasma-membrane phospholipids,present in the human erythrocyte, rat liver, mouse liver and a rapidly growing rat hepatoma. Of the double bonds present in the hydrocarbon chains of the membrane phospholipids,onethird is contributed by sphingomyelin plus phosphatidyl choline and the remainder by phosphatidyl serine, ethanolamine and inositol. Assuming that the phospholipids are asymmetrically distributed in the two leaflets of the bilayer in general, the consequences of this asymmetry in combination with cholesterol content and fatty acid distribution on plasma membrane organization and function are discussed. It is suggested, that the organizational disposition of plasma membrane components other than phospholipids is at least related if not dependent upon the latter's asymmetric distribution in the bilayer.  相似文献

A glycosphingolipid (galactosyl ceramide) has been synthesized which has a spin label covalently attached near the methyl end of the fatty acid chain. This is to our knowledge the first glycolipid spin label to be reported. It is being used to study glycosphingolipid behaviour in lipid bilayers — especially with a view to potential differences from phospholipids. Like phospholipids it assumes a random distribution in fluid lipid bilayers but tends to be excluded from regions rich in phosphatidyl serine in the face of a Ca2+-induced lateral phase separation.  相似文献

1. 1. Lipid peroxidation, superoxide dismutase (SOD) activity, ascorbic acid (AsA) and individual phospholipid contents in liver of fresh water cat fish Heteropneustes fossilis were measured after exposure to different temperatures (25, 27, 32, 37°C) at various times (1–4 h).
2. 2. Lipid peroxidation and superoxide dismutase activity were significantly increased with increases in temperature at various times.
3. 3. Ascorbic acid content was depleted when temperature was increased.
4. 4. After temperature exposure, phosphatidyl inositol was increased while phosphatidyl choline, phosphatidyl serine and phosphatidyl ethanolamine were depleted. Phosphatidic acid level did not change.
5. 5. The findings indicated an increased oxidative stress in liver following increases in temperature at various times. Concurrent with the increase in lipid peroxidation, superoxide dismutase activity and ascorbic acid from the liver of fish varied. It is suggested that depletion of major individual phospholipids following temperature exposure could be due to superoxide created oxidative stress in the liver.

The effect of different phospholipids on the functional activity of highly purified cytochrome P-450 used as a co-substrate of cumene hydroperoxide was examined. At the molar ratio of phospholipids to cytochrome P-450 that was equal to 30, phosphatidyl serine, phosphatidyl inositol, the total fraction of microsomal phospholipids, and lysophosphatidyl choline increased the hydroxylation rate of aniline and naphthalene. The effect of the above phospholipids on the rate of naphthalene oxidation was much more pronounced. Phosphatidyl choline and sphingomyelin in similar large quantities did not exert a stimulating effect on the reactions studied. The kinetic parameters of aniline oxidation in the systems containing phospholipids that produced an activating effect were investigated.  相似文献

Abstract The effect of phospholipids on chitin synthase activity has been studied with digitonin-solubilized and partially purified preparations from Coprinus cinereus . When cholate was used as detergent, it inhibited enzyme activity, but this inhibition was reversed by increasing concentrations of phospholipids. Preincubation with cholate and phospholipid caused irreversible loss of activity. When sonicated with solubilized enzyme preparation, dimyristoyl phosphatidyl choline strongly stimulated activity, while dioleoyl phosphatidyl choline was inhibitory. The Arrhenius plot of the effect of temperature on enzyme activity contained breaks, characteristic of a membrane-bound enzyme. It is suggested that chitin synthase requires an annulus of phospholipids for activity.  相似文献

It was reported that unsaturated long chain fatty acids, such as arachidonic acid, oleic acid and docosahexaenoic acid inhibit the binding between progesterone and estrogen receptors and steroid hormones. Most of the long chain fatty acids are contained in phospholipids within the cells. The effect of phospholipids on the binding between R5020 and progesterone receptors was studied. Phosphatidyl ethanolamine and sphingomyelin had no effect on binding, but phosphatidyl inositol and phosphatidyl serine inhibited the binding 53% and 34% respectively. The effect of phosphatidyl inositol on the binding between R5020 and progesterone receptors was dose dependent. Scatchard analysis revealed that the addition of phospholipid markedly decreased the number of binding sites from 1398 fmol/mgp to 258 fmol/mgp, but the dissociation constant was little affected.  相似文献

Germinating non-dormant (ND) embryos of wild oat incorporate [3H]glycerol into phospholipid, and a 250% increase in total extractable phospholipid occurs within 72 h. During germination, leveles of phosphatidyl inositol showed the greatest change, increasing approximately 5-fold.Imbibed dormant (D) embryos of the wild oat also incorporate [3H]gycerol into phospholipids, but there is no net synthesis. A continuous turnover of membrane phospholipids could be demonstrated in pulse chase experiments, and although the proportions of most phospholipids does not change, there was a decrease of 50% in phosphatidyl serine.The half-life of [3H]glycerol in the extracted phospholipids of D and ND embryos varies between 35 and 57 h, and in membrane fractions separated on sucrose density gradients the half-lives vary between 26 and 56 h.D embryos induced to germinate with GA and ND embryos in which germination is repressed by ABA show similar phospholipid changes to ND and D embryos respectively, with the exception that the proportion of phosphatidyl serine remained unchanged in the ND-ABA embryos.It is concluded that the continual turnover of membranes of imbibed dormant embryos is consistent with the maintenance of cellular integrity determining the longevity of the seed under natural conditions.Abbreviations D dormant - ND nondormant - ABA abscisic acid - GA gibberellic acid (GA3)  相似文献

A lipid-containing neurophysin fraction was isolated and purified from bovine posterior pituitary glands by acid extraction and affinity chromatography on a heparin-Sepharose 4B column. This lipid-rich fraction was found to be composed of noncovalent aggregates of neurophysin proteins and phospholipids such as phosphatidyl choline, phosphatidyl ethanolamine, phosphatidyl serine and sphingomyelin. The lipid-containing neuophysin was delipidated by treatment with choloform-methanol. The resultant apoproteins were characterized as bovine neuroions were developed for the reaggregation of purified bovine neurophysin-I and -II with lipids extracted from bovine posterior pituitary and hypothalamus and with synthetic lecithin. The resultant neurophysin lipid complexes have been shown to band upon isopycnic centrifugation at densities different from those of the respective purified bovine neurophysins.  相似文献

The phospholipid composition of Micrococcus denitrificans was unusual in that phosphatidyl choline (PC) was a major phospholipid (30.9%). Other phospholipids were phosphatidyl glycerol (PG, 52.4%), phosphatidyl ethanolamine (PE, 5.8%), an unknown phospholipid (5.3%), cardiolipin (CL, 3.2%), phosphatidyl dimethylethanolamine (PDME, 0.9%), phosphatidyl monomethylethanolamine (PMME, 0.6%), phosphatidyl serine (PS, 0.5%), and phosphatidic acid (0.4%). Kinetics of 32P incorporation suggested that PC was formed by the successive methylations of PE. Pulse-chase experiments with pulses of 32P or acetate-1-14C to exponentially growing cells showed loss of isotopes from PMME, PDME, PS, and CL with biphasic kinetics suggesting the same type of multiple pools of these lipids as proposed in other bacteria. The major phospholipids, PC, PG, and PE, were metabolically stable under these conditions. The fatty acids isolated from the complex lipids were also unusual in being a simple mixture of seven fatty acids with oleic acid representing 86% of the total. Few free fatty acids and no non-extractable fatty acids associated with the cell wall or membrane were found.  相似文献
