In Dunaliella tertiolecta, D. bioculata and D. viridis the activitiesof phosphoenolpyruvate carboxylase and carbonic anhydrase werehigher in the cells grown in ordinary air (low-CO2 cells) thanin those grown in air enriched with 1–5% CO2 (high-CO2cells), whereas in Porphyridium cruentum R-1 there was no differencein phosphoenolpyruvate carboxylase activity between these twotypes of cells. Apparent Km(NaHCO3) values for photosynthesisin low-CO2 cells of all species tested were smaller than thosein high-CO2 cells. Most of the 14C was incorporated into 3-phosphoglycerate,sugar mono- and di-phosphates during the initial periods ofphotosynthetic NaH14CO3 indicating that both types of cellsin D. tertiolecta are C3 plants. The relationship among gibberellins, CCC, vernalization, and photoperiod in the flowering response of radish, Raphanus sativus L., cv. Miyashige-sofuto, was studied. The optimal condition for flowering was vernalization and a 16-hour photoperiod; GA3 had no additional effect. Gibberellin A3 (60 μg total) was not able to induce flowering in nonvernalized plants grown on 8-hour days, but it did increase the percentage of nonvernalized plants that flowered under long days from 60 to 100.

Gibberellin content of vernalized seedlings increased within the first 24 hours after seedlings were transferred to the greenhouse. Content reached a peak in the first 4 days after transfer and thereafter remained constant. Essentially no gibberellin was found in 2 day-old non-vernalized (control) seedlings of comparable size to the vernalized ones. Gibberellin content in the controls reached a peak on the fourth day of growth in the greenhouse; thereafter, it decreased steadily.

Bolting was inhibited slightly by CCC when applied during vernalization; it was almost completely inhibited when CCC was applied after seed vernalization. Extraction experiments revealed that CCC actually reduced the gibberellin content when applied during or after vernalization. The dwarfing agent, however, had essentially no effect on flowering. We concluded that gibberellins likely play a direct role in bolting of `Miyashige-sofuto' radish, but probably are not directly functional in initiating flowering.

Removal of the blade from the leaf subtending the first flowerbud on Cyclamen persicum ‘Swan Lake’ plants causedthe petiole of that leaf to senesce, but had no effect on thegrowth of the flower peduncle in the debladed petiole's axil.A 10 mg NAA l–1 application generally had no effect onpetiole senescence, peduncle elongation or flowering date whenapplied to the cut end of the petiole after blade removal. A25 mg GA3 l–1 application or a combination of 25 mg GA3l–1 application or a combination of 25 mg GA3 l–1plus 10 mg NAA l–1 delayed petiole senescence and enhancedpeduncle elongation and subsequent flowering. The long-day plant Lemna gibba G3 flowers normally in E medium(Hoagland-type medium plus 30 µM EDTA) but in 0.5 H mediumthere is no flowering. Ammonium is present in 0.5 H medium andis known to inhibit flowering in L. gibba G3, but even in NH4+-free0.5 H medium there is virtually no flowering under continuouslight. Increasing the phosphate concentration of the NH4+-free0.5 H medium from 1.15 ITIM to 12 or 16 mM results in substantialflowering. Decreasing the EDTA concentration from 850 µIMto 250 µM, or raising the nitrate concentration from 4mM to 12 mM, results in only a small increase in flowering.If the decrease in EDTA and increase in nitrate are combinedwith the increase in phosphate, however, the flowering responseis nearly as good as that obtained using E medium. Thus, withthese three changes the inhibitory effect of NH4+free 0.5 Hmedium for flowering in L. gibba G3 is almost completely reversed In the above studies flowering was not limited by daylength.When plants were grown on E medium under an 11 hour daylengthwhere flowering is limited by daylength, decreasing the phosphateconcentration in the medium reduced flowering, but increasingthe phosphate concentration in the medium did not stimulateflowering. Thus, when flowering is limited by daylength, highphosphate will not cause flowering, but a certain level of phosphateappears to be necessary for the expression of photoinductionunder long days. The occurrence and endogenous level of various plant hormoneswere measured for the short-day plants Lemna paucicostata 151and 381 and the long-day plant Lemna gibba G3 to determine whetherany of them are involved in the photoperiodic control of flowering.ABA, IAA, GA1, GA29, GA34, GA53, trans- and cis-zeatin, trans-and cis-ribosyl zeatin, N6-(2-isopentenyl) adenine and N6-(2-isopentenyl)adenosine were definitely detected in each species, while GA4was only detected in L. gibba G3 and GA20 was only detectedin L. paucicostata 151. The endogenous levels of ABA and IAAwere in the range of 1–7 ng/g fr wt and were not significantlydifferent in vegetative and flowering plants. The endogenousgibberellin levels were generally higher in Lemna grown underlong-day rather than short-day conditions. The endogenous cytokininlevels were almost the same in both flowering and vegetativeplants of L. paucicostata 151 and 381. In L. gibba G3, however,the level of cis-ribosyl zeatin, N6-(2-isopentenyl) adenineand N6-(2-sopentenyl) adenosine were higher in vegetative thanin flowering plants. These results indicate that there is not necessarily a directrelation between endogenous plant hormone levels and flowering,and that the chemical basis for the photoperiodic control offlowering cannot be explained solely by changes in hormone levels.The possibility remains, however, that one or more of the planthormones has some influence of secondary importance on the floweringprocess in Lemna. Chicory plants (Cichorium intybus L. var foliosum cv Flash) were tested with and without a 4-week-long cold treatment for in vivo and in vitro flowering potential every 2 weeks during the growing season. One hundred percent of the plants harvested 112 days or later after sowing and then vernalized flowered in vivo. In vitro, no vernalization was needed to initiate flowering-stems on chicory explants taken from roots of 100 days old and older. 5-Azacytidine, a DNA demethylation agent, increased the flowering percentage on explants from young, vernalized roots but could not induce more than 15% flowering on young, nonvernalized roots. The greater flowering potential of chicory root explants in vitro when compared to plants of the same age tested in vivo was clearly established. This result suggests that some negative control on flowering was removed when root explants were excised and the main plant body discarded. The purpose of this study was to examine the effect of prolongedbed rest (BR) on the peak isometric force(Po) and unloaded shorteningvelocity (Vo)of single Ca2+-activated musclefibers. Soleus muscle biopsies were obtained from eight adult malesbefore and after 17 days of 6° head-down BR. Chemicallypermeabilized single fiber segments were mounted between a forcetransducer and position motor, activated with saturating levels ofCa2+, and subjected to slacklength steps. Vowas determined by plotting the time for force redevelopment vs. theslack step distance. Gel electrophoresis revealed that 96% of the pre-and 87% of the post-BR fibers studied expressed only the slow type Imyosin heavy chain isoform. Fibers with diameter >100 µm made uponly 14% of this post-BR type I population compared with 33% of thepre-BR type I population. Consequently, the post-BR type I fibers(n = 147) were, on average, 5%smaller in diameter than the pre-BR type I fibers(n = 218) and produced 13% lessabsolute Po. BR had no overalleffect on Po per fibercross-sectional area(Po/CSA), even though halfof the subjects displayed a decline of 9-12% inPo/CSA after BR. Type Ifiber Voincreased by an average of 34% with BR. Although the ratio of myosinlight chain 3 to myosin light chain 2 also rose with BR, there was nocorrelation between this ratio andVo for either thepre- or post-BR fibers. In separate fibers obtained from the originalbiopsies, quantitative electron microscopy revealed a 20-24%decrease in thin filament density, with no change in thick filamentdensity. These results raise the possibility that alterations in thegeometric relationships between thin and thick filaments may be atleast partially responsible for the elevatedVo of the post-BRtype I fibers.

Negative linear relations were detected (P < 0·005)between the rate of progress from sowing to panicle initiationand CO2 concentration (210-720 µmol CO2 mol-1 air) fortwo genotypes of sorghum [Sorghum bicolor (L.) Moench]. Relationsbetween CO2 concentration and the rate of progress from sowingto first flowering were also negative in soyabean [Glycine max(L.) Merrill] (P < 0·025), but positive in cowpea[Vigna unguiculata (L.) Walp.] (P < 0·025), albeitthat in both grain legumes sensitivity was much less than insorghum. Thus CO2 elevation does not delay flowering in allshort-day species. The considerable effect of CO2 concentrationon times to panicle initiation resulted in large differencesamong the sorghum plants at this developmental stage; with increasein CO2 concentration, plants were taller with slightly moreleaves and more pronounced apical extension. At the same timeafter sowing however, sorghum plants were heavier (P < 0·05)at 210 than at 360 µmol CO2 mol-1 air. In contrast, relationsbetween the dry masses of the soyabean and cowpea plants andCO2 concentration were positive and curvilinear (P < 0·05).It is suggested that the impact of global environmental changecould be severe for sorghum production in the semi-arid tropics.Copyright1995, 1999 Academic Press Sorghum bicolor (L.) In the sweet pea (Lathyrus odoratus L.) genes Dnl andDrh controlthe production of a graft-transmissible substance which delaysflowering and promotes outgrowth of basal laterals. Seed vernalizationpromotes flowering and reduces lateral outgrowth in intact plantsand grafted scions of genotype DniDnl, suggesting that vernalizationreduces output of the Dni system, possibly by disrupting therelationship between chronological and plastochronic age. Whenlateral outgrowth and floral abortion are used as indicatorsof inhibitor levels, it can be shown that vernalized Dni plantspossess more inhibitor but initiate flower buds at a lower nodethan unvernalized dn plants. This supports the suggestion thatin regard to floral initiation vernalization also alters thesensitivity of the shoot apex to the flowering hormone(s). InLathyrus odoratus an hormonally based vernalization responseof considerable magnitude can be shown for day-neutral (dndn)lines, supporting the suggestion that vernalization also influencesthe level of a flower promotor. Duckweed, Lemna minor L., grown on inorganic nutrient solutionscontaining either NH4+ or NO3 as nitrogen source wasallowed to assimilate [1-14C]- or [2-14C]glycollate during a20 min period in darkness or in light. The incorporation ofradioactivity into water-soluble metabolites, the insolublefraction, and into the CO2 released was measured. In additionthe extractable activity of phosphoenolpyruvate carboxylasewas determined. During the metabolism of [2-14C]glycollate in darkness, as wellas in the light, NH4+ grown plants evolved more 14CO2 than NO3grown plants. Formate was labelled only from [2-14C]glycollateand in NH4+ grown plants it was significantly less labelledin light than in darkness. In NO3 grown plants formateshowed similar radioactivity after dark and light labelling.The radioactivity in glycine was little influenced by the nitrogensource. Amounts of radioactivity in serine implied that thefurther metabolism of serine was reduced in darkness comparedwith its metabolism in the light under both nitrogen regimes.In illuminated NH4+ plants, serine was labelled through a pathwaystarting from phosphoglycerate. After [1-14C]glycollate feedingNH4+ grown plants contained markedly more radioactive aspartateand malate than NO3 plants indicating a stimulated phosphoenolpyruvatecarboxylation in plants grown on NH4+. Lines representing a range of internode length and floweringgenotypes in Pisum sativum L. were grown in 8 h of daylightfollowed by either 16 h of darkness or incandescent light. Thestem elongation response index (RI = length in 24 h ÷length in 8 h) was least in the very short internode nana types,which are grossly deficient in gibberellins (GAs), and the verylong internode slender types, which behave as if saturated withGAs. The common tall (genotype Le) and dwarf (le) types (lepartially blocks conversion of GA20 to the active form, GA1)were all markedly responsive but the peak RI (based on the mostresponsive internode) was less in tall lines (1.79 to 2.78)than in dwarf lines (2.32 to 5.01) and the peak RI tended tooccur about three to four internodes earlier in tall than indwarf lines. The cry8 mutation reduced the RI. (Duplicate lengthloci La and Cry are probably concerned with GA reception.) Amongle dwarf lines, genotype La cry8, was generally less responsivethan La Cry, La cryc and la Cry. Data from crosses showed thaton either an le La or le la background cry8 segregates had alower RI than cry8 segregates. On an le la background, cry8plants were shorter than cryc plants, cry8 was partially dominantto cry8 and segregation was clear only in long days. On an lela background, cryc plants were shorter than cryc plants, cry8was partially dominant to cry8 and segregation was clear inlong or short days. The very high peak RI (5.0) of the microcryptodwarfline, L57, appeared to result, in part, from a marked foreshorteningof internodes 4 to 10 in the 8 h regime. In the 24 h regimeL57 (lm) had a fairly similar growth pattern to normal (Lm)cryptodwarf types. The peak RI tended to occur at a lower internode in early thanlate flowering lines, especially among dwarf types, and genotypeswith a day neutral flowering habit (genotype sn or dne) wereless responsive than their photoperiodic counterparts (Sn Dne). White fluorescent light, given as a daylength extension, wasmuch less effective than incandescent light at stimulating stemelongation suggesting control through the phytochrome equilibrium(Ptr/Ptotal). Gametophytic self-incompatibility, a mechanism that preventsinbreeding in some families of flowering plants, is mediatedby the products of a single genetic locus, the S-locus. Theproducts of the S-gene in the female sexual tissues of Nicotianaalata are an allelic series of glycoproteins with RNase activity.In this study, we report on the microheterogeneity of N-linkedglycosylation at the four potential N-glycosylation sites ofthe S2-glycoprotein. The S-glycoproteins from N.alata containfrom one to five potential N-glycosylation sites based on theconsensus sequence Asn-Xaa-Ser/Thr. The S2-glycoprotein containsfour potential N-glycosylation sites at Asn27, Asn37, Asn138and Asn150, designated sites I, n, IV and V, respectively. SiteIII is absent from the S2-glycoprotein. Analysis of glycopeptidesgenerated from the S2-glycoprotein by trypsin and chymotrypsindigestions revealed the types of glycans and the degree of microheterogeneitypresent at each site. Sites I (Asn27) and IV (Asn138) displaymicroheterogeneity, site II (Asn37) contains only a single typeof N-glycan, and site V (Asn150) is not glycosylated. The microheterogeneityobserved at site I on the S2-glycoprotein is the same as thatobserved at the only site, site I, on the Srglycoprotein (Woodwardet al., Glycobiology, 2, 241-250, 1992). Spinach plants grown without molybdenum lack nitrate reductaseand when plants are deprived of nitrate existing activity islost. Transfer of molybdenum-deficient plants to a solutioncontaining (NH4)299MoO4) or nitrate-starved plants to NaNO3solution induced enzyme activity in 24 hr. After purificationby selective adsorption, precipitation and disc electrophoresis,the protein from molybdenum-deficient plants given 99Mo showedradioactivity only where nitrate reductase was revealed on theacrylamide gel. Molybdenum was similarly selectively concentratedinto the enzyme as a result of induction by nitrate in plantsgrown with sub-optimal molybdenum supply in order to minimizeeffects of isotope dilution on measurement of 99Mo incorporation. There was no exchange in vitro between 99Mo and purified activeenzyme in the resting state over 18 hr at 4°C, or with functioningenzyme held at room temperature for 24 hr. There was evidenceeither for possible in vivo exchange of 99Mo andenzyme boundMo or for slight synthesis of fresh enzyme under conditionsof net loss of enzyme in nitrate starved plants. Five NADH2 and two NADPH2 reactive diaphorases which could beseparated by electrophoresis were present in extracts. Onlyone of these having strong NADH2 and weak NADPH2 activity wasdirectly associated with nitrate reductase. The same complexalso showed the only benzyl viologen (BV.) reactive nitratereductase. Nitrate reductase in spinach is therefore considered to be amolybdenum-dependant and molybdenum-containing protein in whichNADH2 (with weak NADPH2) and BVelectron donor functions anddiaphorase/reductase activities remain closely associated duringpurification and electrophoresis. The techniques provide a simple means for the production andpurification of enzyme containing radioactively labelled Moapplicable to investigations on the structure of the enzyme. Lemna paucicostata, strain LP6, does not ordinarily flower underany photoperiodic schedule, when grown in the modified Bonner-Devirianmedium supplemented with 10–4 M EDTA and 1% sucrose (thisis a medium which is otherwise satisfactory for short day inductionof flowering of other strains of this duckweed). However, when the ferric citrate concentration in the culturemedium containing EDTA was raised 10-fold over that in the normalmedium, a low but significant flowering could be initiated inthis duckweed, irrespective of the length of the photoperiod.A similar flowering response was obtained when ferric citratewas replaced by ferrous sulphate or ferric chloride, therebyindicating that the inductive effect of higher level of ferriccitrate on flowering in strain LP6 is due to its iron component. Some flowering in this strain could be induced even in mediumcontaining normal level of iron, provided the level of EDTA,itself, was raised to 5?10–4 (5% flowering) or to 10–3M (12% flowering), but replacement of EDTA by EDDHA led to trulyremarkable effects. With EDDHA, flowering could be induced at very high levels (90%)under all photoperiodic regimes tried. Cells of Dunaliella tertiolecta which had been grown in ordinaryair (low-CO2 cells) had high carbonic anhydrase (CA) activityon the cell surface and mainly utilized HCO3 for photosynthesis.When CA activity on the cell surface was inhibited by Diamoxor subtilisin, the cells utilized CO2. When bovine CA was added,the subtilisin-treated low-CO2 cells utilized mainly HCO3.When grown in air containing 2% CO2, the cells had low CA activityon the cell surface, and preferred CO2 to HCO3. Kineticanalysis of these results indicated that low-CO2 cells of D.tertiolecta absorb CO2 which was converted from HCO3via the CA located on the cell surface. The level of benzoic acid was measured in Lemna gibba G3 grownon M and E media under inductive and non-inductive daylengths.Benzoic acid was slightly higher in plants grown on M mediumbut there was no difference in the benzoic acid levels in floweringand vegetative plants. When L. gibba G3 was grown under continuouslight on 1/10 M medium or 1/2 H medium there was virtually noflowering, but addition of benzoic acid to either medium ledto a substantial flowering response. In both cases this floweringresponse was inhibited by the plant hormones IAA, GA3, ABA andzeatin, with IAA and GA3 being the least inhibitory and ABAbeing the most inhibitory. This same pattern of inhibition wasseen when L. gibba G3 was grown on M medium under continuouslight, conditions that lead to photoinduction of flowering.These results leave open the possibility that endogenous benzoicacid may interact with other factors to influence the floweringresponse in L. gibba G3. A long-term, steady-state 13CO2 assimilation system at a constantCO2 concentration with a constant 13C abundance was designedand applied to quantitative investigations on the allocationof photoassimilated carbon in nodulated soya bean (Glycine maxL.) plants. The CO2 concentration in the assimilation chamberand its 13C abundance were maintained constant with relativevariances of less than ±0.5 per cent during an 8-h assimilationperiod. At the termination of 8-h 13CO2 assimilation by plantsat early flowering stage, the currently assimilated carbon relativeto total tissue carbon (measured by the degree of isotopic saturation)were for young leaves (including flower buds), 13.9 per cent;mature leaves, 15.7 per cent; stems+petioles, 5.9 per cent;roots, 5.4 per cent and nodules, 6.9 per cent, 48 h after theend of the 13CO2 assimilation period, they were 12.3, 7.5, 7.4,6.8 and 6.1 per cent, respectively. The treatment with a highconcentration of nitrate in the nutrient media significantlydecreased the allocation of 13C into nodules. Experiments on13CO2 assimilation by plants at the pod-filling stage were alsoconducted. Labelling by 13C was weaker than at the early floweringstage, but an intense accumulation of 13C into reproductiveorgans was observed. Leaves of three C4 plants, Setaria italica, Pennisetum typhoides,and Amaranthus paniculatus possessed five- to ten-fold higheractivities of a (Na+-K+)-dependent ATPase than those of twoC3 plants, Oryza sativa and Rumex vesicarius. Na+-K+ ATPasefrom leaves of Amarathus exhibited an optimal pH of 7?5 andan optimal temperature of 35 ?C. It required 40 mM K+ and 80mM Na+ for maximal activity. Ouabain partially inhibited (Na+-K+)-dependentATPase activity in leaves of C4 plants. Ouabain also blockedthe movement of label from initially formed C4 acids into endproducts in leaves of only C4 plants, Setaria and Amaranthusbut not in a C3 plant, Rumex. We propose that Na+-K+ ATPasemay mediate transfer of energy during active transport of C4acids from mesophyll into the bundle sheath.  相似文献

A comparison of the activity and properties of the enzyme phosphoenolpyruvatecarboxylase (PEPC) was made for plants of Sedum telephium L.grown under low (70 µmol m–2 s–1) or high(500µmol m–2 s–1) PPFD and subjected to varyingdegrees of water stress. Under well-watered conditions onlyplants grown under high PPFD accumulated titratable acidityovernight and the extractable activity of PEPC was almost 2-foldhigher in these plants than in plants grown under low PPFD.Increasing drought stress resulted in a substantial increasein the activity of PEPC extracted both during the light anddark periods and a decrease in the sensitivity to inhibitionby malic acid. The magnitude of these changes was determinedby the severity and duration of drought and by light intensity.A comparison of the kinetic properties of PEPC from severelydroughted plants revealed that plants droughted under high PPFDhad a lower Km for PEP than plants under low PPFD. Additionof 2·0 mol m–3 malate resulted in an increase inthe Km for PEP, with plants draughted under low PPFD havinga significantly higher Km in the presence of malic acid comparedto those under high PPFD. Response to the activator glc-6-P,which lowered the Km for PEP, also varied between plants grownunder the two light regimes. Under well-watered conditions PEPCextracted from plants under high PPFD was more sensitive toactivation by glc-6-P than those under low PPFD. After the severedrought treatment, however, the Km for PEP in the presence ofglc-6-P was similar for enzyme extracted from plants grown underboth light regimes. Soluble sugars and starch were depletedovernight and were both possible sources of substrate for PEPC.With increasing drought, however, the depletion of starch relativeto soluble sugars increased under both light regimes. The propertiesof PEPC and the characteristics of carbohydrate accumulation/depletionare discussed in relation to the regulation of CAM in S. telephiumgrown under different light and watering regimes. Abscisic acid (ABA) at 1 x 10–4 M or 3 x 10–4 Mwas applied to the apical buds of Chenopodium rubrum plantsexposed to different photoperiodic treatments and showing differentpatterns of floral differentiation. Stimulation of growth inwidth of the apical meristem of the shoot and/or inhibitionof growth in length was obtained under all photoperiodic treatments.This change of growth pattern was followed by different effectson flowering. In non-induced plants grown under continuous light ABA stimulatedpericlinal divisions in the peripheral zone and the initiationof leaves as well as the growth in width of bud primordia. Inplants induced by two short days reduced growth of the meristemcoincided with ABA application. Longitudinal growth of the meristemwas inhibited in this case and only a temporary stimulationof inflorescence formation took place. In plants induced ata very early stage, ABA exerted a strong inhibitory effect onflowering. A permanent and reproducible stimulatory effect onflowering was obtained in plants induced by three sub-criticalphotoperiodic cycles if ABA was applied to apices released fromapical dominance. In this case formation of lateral organs andinternodes was promoted by ABA and was followed by stimulatedinflorescence formation. Gibberellic acid (GA2) at 1x 10–4M or 3 x 10–4 M brought about a similar effect on floweringas ABA, although the primary growth effect was different, i.e.GA2 stimulated longitudinal growth. The effects of ABA and GA2 on floral differentiation have beencompared with earlier results obtained from auxin and kinetinapplications. These growth hormones are believed to regulateflowering by changing cellular growth within the shoot apex.Depending on the actual state of the meristem identical growthresponses may result in different patterns of organogenesisand even in opposite effects on flowering.
