利用模式生物秀丽隐杆线虫,考察8种人体必需氨基酸对衰老的影响。首先建立秀丽隐杆线虫寿命模型,以雷帕霉素为阳性对照药,分别考察8种必需氨基酸对线虫生存时间的影响。再用筛选出的氨基酸处理线虫21d,通过秀丽隐杆线虫-绿脓杆菌感染模型,考察氨基酸对线虫的抗感染能力的影响,利用实时荧光定量Real-Time RT-PCR方法检测氨基酸处理线虫后DAF-16/FOXO下游基因和免疫相关基因的表达水平。结果表明8种必需氨基酸中苏氨酸和异亮氨酸既能延长野生型线虫的寿命又能延长daf-16突变型线虫的寿命,同时还能增强秀丽隐杆线虫抗绿脓杆菌感染的能力,并提高免疫相关基因lys-7、clec-67的表达水平,而DAF-16/FOXO下游基因表达没有明显变化。因此苏氨酸和异亮氨酸能延长线虫寿命、提高抗感染能力,且对线虫寿命的延长作用不完全依赖于DAF-16/FOXO转录因子。  相似文献

与秀丽线虫有关的研究开始于20世纪60年代,秀丽线虫作为模式生物是第一个完成基因测序的多细胞生物,普遍应用于各种环境对生理和行为学研究中。空间环境以微重力、强辐射作为特点,对秀丽线虫的生理及行为产生很大影响。本文总结了微重力和辐射引起秀丽线虫运动能力、寿命、能量代谢等方面基因表达变化的研究成果。秀丽线虫与人类的序列相似性高达40%,空间环境对秀丽线虫行为及生长发育的研究为空间环境对人类健康影响研究提供数据支持,也为空间损伤修复研究提供理论基础。  相似文献

为了阐明铜(Cu)对秀丽隐杆线虫Caenorhabditis elegans长期作用的毒性效应,对实验室多代筛选的耐铜型秀丽隐杆线虫进行了寿命、衰老、发育、生殖和运动等生物学指标的研究.结果显示耐铜型秀丽隐杆线虫与野生型秀丽隐杆线虫相比其寿命缩短、衰老提前、个体发育受到抑制,且出现繁殖率降低、生殖能力减弱、运动行为存在障碍等一系列生理变化.本文为理解与阐明Cu的毒性效应提供了实验资料,有助于深入开展Cu毒性机理的研究.  相似文献

以秀丽隐杆线虫(Canorhabditis elegans)为模式生物研究山楂提取物(Haworth fruit extract,HFE)对其急性氧化损伤的保护作用及其可能的作用机制。饲喂线虫于含有不同浓度(0、25、50和100μg/m L)HFE的NGM(Nematode growth medium)培养基中,研究HFE对线虫急性应激耐受能力的影响。结果显示,饲喂HFE后,秀丽线虫表现出比正常组更高的寿命,并且在胡桃醌氧化应激、热应激及紫外辐射应激实验中寿命均明显延长,荧光显微镜观察发现HFE组线虫的脂褐素自发荧光明显减弱,并且与HFE浓度呈剂量依赖效应。HFE能够显著延长秀丽隐杆线虫的寿命,同时对多种氧化损伤具有较好的保护作用,改善机体的抗氧化能力,有效延缓衰老。  相似文献

探索了不同浓度蕨菜水提液对秀丽隐杆线虫整个生命周期生物学功能的影响,对蕨菜水提液的毒理性作出合理评价。实验以秀丽隐杆线虫为模式生物,通过给秀丽隐杆线虫喂食01 mL 6种不同浓度(00、02、04、06、08、10 g/mL)的蕨菜水提取液,观察不同浓度蕨菜水提取液对其生理指标(寿命、产卵数、运动性、细胞凋亡)的影响。研究结果表明:蕨菜能对秀丽隐杆线虫的生理指标产生较大影响,具体表现为随着蕨菜水提取液浓度增加,寿命逐渐缩短,产卵数逐渐减少,细胞凋亡数增加,三者均呈现出明显的浓度依赖性,而对运动性影响不大。  相似文献

秀丽隐杆线虫是研究衰老的重要模式生物,但目前对其免疫衰老的研究缺乏评价指标。建立了秀丽隐杆线虫-铜绿假单胞菌感染模型,考察了线虫抗感染免疫能力与衰老之间的关系,建立了线虫免疫衰老的评价指标。首先用铜绿假单胞菌感染线虫建立模型,将处于不同衰老程度的线虫用于感染实验,考察了线虫感染后的生存时间与衰老程度的关系;将感染后的生存时间作为抗感染免疫衰老的指标,其结果与线虫抗氧化能力和寿命指标的结果相互印证。结果显示,7 d、14 d、21 d龄的线虫感染后的生存时间依次减少,与抗氧化能力的衰退情况相符;与野生型相比,长寿线虫(daf-2突变线虫)感染后的生存时间延长,短寿线虫(daf-16突变线虫)感染后的生存时间缩短,线虫的抗感染免疫指标与寿命指标结果相符;能延长线虫寿命的化合物酪氨酸也延长了线虫感染后的生存时间。因此线虫-铜绿假单胞菌感染模型可以用于评价线虫的免疫衰老,感染后线虫的生存时间可作为免疫衰老的评价指标。  相似文献

作为模式生物,秀丽线虫(Caenorhabditis elegans)已经成功地用于许多生命过程的研究,尤其被广泛应用于现代发育生物学、行为与神经生物学、基因组学、正向和反向的遗传学研究中,近年来,秀丽线虫更成为了一个进行蛋白质组学研究的优良体系,诠释了基于基因组学和RNA干涉研究中的基因功能。许多比较蛋白质组学表达谱的建立可以更好地理解线虫在不同发育阶段、不同温度下基因的表达,在与人类神经疾病相关的疾病研究中,线虫对帕金森疾病、阿尔茨海默症、衰老与寿命、胰岛素通路都有所揭示。另外,线虫的亚蛋白质组学和翻译后修饰如糖基化和磷酸化也已经鉴定,其数据库也在不断地完善。本文介绍了秀丽线虫的蛋白质表达谱建立的历史,尤其是神经科学研究中的应用及翻译后修饰表达谱的建立等方面的研究现状,因此,结合其它分子生物学和基因工程技术,线虫蛋白质组学研究已成为提供一个新的全面的系统分析基因功能的重要工具,提示线虫是"蠕虫蛋白质组学"的一个丰富宝藏。  相似文献

Resveratrol and SRT1720 have been shown to act as sirtuin activators that may ameliorate type 2 diabetes and metabolic diseases in mice. Moreover, resveratrol extends lifespan in model organisms like C. elegans, N. FURZERI, and possibly D. melanogaster. The aim of the study was to test whether pharmacological concentrations of resveratrol and SRT1720 are capable of extending lifespan in a nematodal model organism for aging processes, the roundworm Caenorhabditis elegans. Several hundreds of adult C. ELEGANS roundworms were maintained on agar plates and fed E. COLI strain OP50 bacteria. Resveratrol (5 micromolar, 500 nanomolar) or SRT1720 (1 micromolar, 100 nanomolar) was applied to the agar to test whether they may promote longevity by quantifying survival in the presence and absence of the respective compounds. At a dose of 5 micromolar, which is pharmacologically relevant and 20 times lower than previously published concentrations, resveratrol significantly extends C. elegans lifespan by 3.6% (mean lifespan) and 3.4% (maximum lifespan). By unexpected contrast, SRT1720, which was previously proposed to be several hundred times more active than resveratrol, did not extend lifespan at none of the concentrations tested. Thus, in the model organisms C. elegans, resveratrol is capable of promoting longevity at a concentration that pharmacologically relevant and 20 times lower than previously published doses. The sirtuin activator SRT1720 did not extend lifespan, suggesting that in C. elegans, some relevant effects of resveratrol cannot be mimicked by SRT1720.  相似文献

重组荞麦胰蛋白酶抑制剂(recombinant buckwheat trypsin inhibitor,rBTI)是一种来源于荞麦Potato Ⅰ抑制剂家族的丝氨酸蛋白酶抑制剂,具有很好的生物活性及功能。先前的研究表明,rBTI在秀丽隐杆线虫(Caenorhabditis elegans)中具有很好的延长寿命的性质,但其具体的作用机制还不太清楚。本文的研究证明,rBTI能够调节转录因子DAF-16的转录活性,进而影响线虫的寿命,且该性质与其胰蛋白酶抑制活性密切相关。通过定点突变技术,分别对rBTI的45位、53位和44位氨基酸活性位点进行突变,获得了4种不同胰蛋白酶抑制活性的rBTI突变体,分别命名为rBTI-R45A,rBTI-R45F,rBTI-W53R和rBTI-P44T。经典模式生物秀丽隐杆线虫寿命检测实验显示,野生型rBTI可以明显延长C.elegans的寿命,且在0~10 μmol/L 范围内具有浓度依赖性。和未处理对照组相比,10 μmol/L 野生型rBTI延长寿命幅度可达到14.5%,但是突变体rBTI-R45A,rBTI-R45F和rBTI W53R均不同程度失去了延长寿命的功能。利用荧光显微观察及qRT-PCR等方法进一步研究发现,野生型rBTI 可增强寿命调控转录因子DAF-16的转录活性。与寿命检测实验结果一致,4种 rBTI突变体均不能使DAF-16转录活性增强。上述结果表明,在C.elegans中,rBTI可增强长寿因子DAF 16的转录活性,进而延长虫体寿命,且该功能的发挥依赖于其适当的胰蛋白酶抑制活性。本文的结果为进一步研究开发rBTI的功能提供了实验支持和理论基础。  相似文献

Extracellular polysaccharide (EPS) antigens produced by different mould species were purified and partially characterized. Purification included (NH4)2SO4 treatment, Sepharose CL-4B column chromatography and Con A-sepharose chromatography. The EPS of Penicillium digitatum, Mucor racemosus and Cladosporium cladosporioides showed high antigenic capacities. Immunologically the EPS were partially genus-specific, but cross-reactivity was observed. The EPS antigens produced by species of Penicillium, Aspergillus repens and Geotrichum candidum lost their immunological activity upon heating (100 degrees C) at pH 1.8, while the EPS antigen of M. racemosus, Rhizopus oligosporus and C. cladosporioides were stable under the same conditions. The dominant monosaccharides present in the EPS antigen were mannose, galactose and glucose. The EPS obtained from cultures of M. racemosus and R. oligosporus also contained rhamnose. In the EPS produced by Penicillium spp. and A. repens the galactose residues were determined to be immunodominant.  相似文献

Studies of mutations affecting lifespan in Caenorhabditis elegans show that mitochondrial generation of reactive oxygen species (ROS) plays a major causative role in organismal aging. Here, we describe a novel mechanism for regulating mitochondrial ROS production and lifespan in C .  elegans: progressive mitochondrial protein modification by the glycolysis-derived dicarbonyl metabolite methylglyoxal (MG). We demonstrate that the activity of glyoxalase-1, an enzyme detoxifying MG, is markedly reduced with age despite unchanged levels of glyoxalase-1 mRNA. The decrease in enzymatic activity promotes accumulation of MG-derived adducts and oxidative stress markers, which cause further inhibition of glyoxalase-1 expression. Over-expression of the C .  elegans glyoxalase-1 orthologue CeGly decreases MG modifications of mitochondrial proteins and mitochondrial ROS production, and prolongs C .  elegans lifespan. In contrast, knock-down of CeGly increases MG modifications of mitochondrial proteins and mitochondrial ROS production, and decreases C .  elegans lifespan.  相似文献

The superoxide dismutase mimetic EUK-8 has been reported to extend lifespan in the nematode Caenorhabditis elegans. However, in five trials administering EUK-8 in liquid culture with E. coli, and two trials using defined liquid medium, we observed no increase in C. elegans lifespan. Instead we saw a dose-dependent reduction of lifespan and fertility. We conclude that extension of C. elegans lifespan by EUK-8 may only occur under very particular culture conditions.  相似文献

The influencing factors of extracellular polysaccharide (EPS) produced from a strain of lactic acid bacteria (LAB L15) were studied by using the phenol-H2SO4 method. It was demonstrated that the strain produced EPS at the most amount when it was incubated for 40–48 h and when the pH value was 4 under 30°C. Glucose was the most suitable carbon source for LAB-producing EPS. The rough EPS was obtained from L15 culture after centrifugation, dialysis, deprotein, decoloration, and ethanol-precipitation. The sample was at least composed of two polysaccharides that were completely different in molecular weight and the amount.
