Extracellular ATP acts as a signal that regulates a variety of cellular processes via binding to P2 purinergic receptors (P2 receptors). We herein investigated the effects and signaling pathways of ATP on glucose uptake in C(2)C(12) skeletal muscle cells. ATP as well as P2 receptor agonists (ATP-gamma S) stimulated the rate of glucose uptake, while P2 receptor antagonists (suramin) inhibited the stimulatory effect of ATP, indicating that P2 receptors are involved. This ATP-stimulated glucose transport was blocked by specific inhibitors of Gi protein (pertusiss toxin), phospholipase C (U73122), protein kinase C (GF109203X), and phosphatidylinositol (PI) 3-kinase (LY294002). ATP stimulated PI 3-kinase activity and P2 receptor antagonists blocked this activation. In C(2)C(12) myotubes expressing glucose transporter GLUT4, ATP increased basal and insulin-stimulated glucose transport. Finally, ATP facilitated translocation of GLUT1 and GLUT4 into plasma membrane. These results together suggest that cells respond to extracellular ATP to increase glucose transport through P2 receptors.  相似文献

Hydrogen sulfide (H2S), a gasotransmitter, is formed from l-cysteine by multiple enzymes including cystathionine-γ-lyase (CSE). We have shown that an H2S donor, NaHS, causes hyperalgesia in rodents, an effect inhibited by knockdown of Cav3.2 T-type Ca2+ channels (T-channels), and that NaHS facilitates T-channel-dependent currents (T-currents) in NG108-15 cells that naturally express Cav3.2. In the present study, we asked if endogenous and exogenous H2S participates in regulation of the channel functions in Cav3.2-transfected HEK293 (Cav3.2-HEK293) cells. dl-Propargylglycine (PPG), a CSE inhibitor, significantly decreased T-currents in Cav3.2-HEK293 cells, but not in NG108-15 cells. NaHS at 1.5 mM did not affect T-currents in Cav3.2-HEK293 cells, but enhanced T-currents in NG108-15 cells. In the presence of PPG, NaHS at 1.5 mM, but not 0.1–0.3 mM, increased T-currents in Cav3.2-HEK293 cells. Similarly, Na2S, another H2S donor, at 0.1–0.3 mM significantly increased T-currents in the presence, but not absence, of PPG in Cav3.2-HEK293 cells. Expression of CSE was detected at protein and mRNA levels in HEK293 cells. Intraplantar administration of Na2S, like NaHS, caused mechanical hyperalgesia, an effect blocked by NNC 55-0396, a T-channel inhibitor. The in vivo potency of Na2S was higher than NaHS. These results suggest that the function of Cav3.2 T-channels is tonically enhanced by endogenous H2S synthesized by CSE in Cav3.2-HEK293 cells, and that exogenous H2S is capable of enhancing Cav3.2 function when endogenous H2S production by CSE is inhibited. In addition, Na2S is considered a more potent H2S donor than NaHS in vitro as well as in vivo.  相似文献

The potential for C4 photosynthesis was investigated in five C3-C4 intermediate species, one C3 species, and one C4 species in the genus Flaveria, using 14CO2 pulse-12CO2 chase techniques and quantum-yield measurements. All five intermediate species were capable of incorporating 14CO2 into the C4 acids malate and aspartate, following an 8-s pulse. The proportion of 14C label in these C4 products ranged from 50–55% to 20–26% in the C3-C4 intermediates F. floridana Johnston and F. linearis Lag. respectively. All of the intermediate species incorporated as much, or more, 14CO2 into aspartate as into malate. Generally, about 5–15% of the initial label in these species appeared as other organic acids. There was variation in the capacity for C4 photosynthesis among the intermediate species based on the apparent rate of conversion of 14C label from the C4 cycle to the C3 cycle. In intermediate species such as F. pubescens Rydb., F. ramosissima Klatt., and F. floridana we observed a substantial decrease in label of C4-cycle products and an increase in percentage label in C3-cycle products during chase periods with 12CO2, although the rate of change was slower than in the C4 species, F. palmeri. In these C3-C4 intermediates both sucrose and fumarate were predominant products after a 20-min chase period. In the C3-C4 intermediates, F. anomala Robinson and f. linearis we observed no significant decrease in the label of C4-cycle products during a 3-min chase period and a slow turnover during a 20-min chase, indicating a lower level of functional integration between the C4 and C3 cycles in these species, relative to the other intermediates. Although F. cronquistii Powell was previously identified as a C3 species, 7–18% of the initial label was in malate+aspartate. However, only 40–50% of this label was in the C-4 position, indicating C4-acid formation as secondary products of photosynthesis in F. cronquistii. In 21% O2, the absorbed quantum yields for CO2 uptake (in mol CO2·[mol quanta]-1) averaged 0.053 in F. cronquistii (C3), 0.051 in F. trinervia (Spreng.) Mohr (C4), 0.052 in F. ramosissima (C3-C4), 0.051 in F. anomala (C3-C4), 0.050 in F. linearis (C3-C4), 0.046 in F. floridana (C3-C4), and 0.044 in F. pubescens (C3-C4). In 2% O2 an enhancement of the quantum yield was observed in all of the C3-C4 intermediate species, ranging from 21% in F. ramosissima to 43% in F. pubescens. In all intermediates the quantum yields in 2% O2 were intermediate in value to the C3 and C4 species, indicating a co-function of the C3 and C4 cycles in CO2 assimilation. The low quantum-yield values for F. pubescens and F. floridana in 21% O2 presumably reflect an ineffcient transfer of carbon from the C4 to the C3 cycle. The response of the quantum yield to four increasing O2 concentrations (2–35%) showed lower levels of O2 inhibition in the C3-C4 intermediate F. ramosissima, relative to the C3 species. This indicates that the co-function of the C3 and C4 cycles in this intermediate species leads to an increased CO2 concentration at the site of ribulose-1,5-bisphosphate carboxylase/oxygenase and a concomitant decrease in the competitive inhibition by O2.Abbreviations PEP phosphoenolpyruvate - PGA 3-phosphoglycerate - RuBP ribulose-1,5-bisphosphate  相似文献

Oxidative stress has an important role in neurodegenerative diseases and cerebral ischemic injury. It is reported that d-β-hydroxybutyrate (DβHB), the major component of ketone bodies, is neuroprotective in recent studies. Therefore, in the present work the neuroprotective effects of DβHB on H2O2-induced apoptosis mediated by oxidative stress was investigated. PC12 cells were exposed to H2O2 with different concentrations of H2O2 for different times after DβHB pretreatment. MTT assay, apoptotic rates, intracellular reactive oxygen species (ROS) level, GSH content, mitochondrial membrane potential (MMP) and caspase-3 activity were determined. The results showed that DβHB inhibited the decrease of cell viability induced by H2O2 in PC12 cells. DβHB decreased the apoptotic rates induced by H2O2. The changes of intracellular ROS, GSH, MMP and caspase-3 activity due to H2O2 exposure were partially reversed in PC12 cells. So DβHB inhibited the apoptosis of PC12 cells induced by H2O2 via inhibiting oxidative stress.  相似文献

Separation of important chemical feedstocks, such as C2H6 from natural gas, can greatly benefit the petrochemical industry. In this paper, the grand canonical Monte Carlo method has been used to study the adsorption and separation of CH4 and C2H6 in zeolites, isoreticular metal-organic framework-1 (IRMOF-1) and zeolitic imidazolate frameworks (ZIFs) with different topology, including soadlite, gmelinite and RHO topologies. Compared with mordenite zeolite and IRMOF-1, ZIFs and mordenite framework inverted (MFI) zeolite have better separation performance for C2H6/CH4 mixtures at different mole fractions of C2H6. From the study of equilibrium snapshots and density distribution profiles, adsorption sites could be grouped as (1) sites with strong interactions with adsorbent and (2) sites with strong interactions with surrounding adsorbates. The gas molecules occupied the first site and then went on to occupy the second site. In CH4/C2H6 mixture adsorption/separation, the adsorption of CH4 was confined by the existence of C2H6. Due to energetic effect, C2H6 selectivity was affected by temperature at a low-pressure range, but did not change as much in a high-pressure range because of packing effect in micropore. In binary adsorption, large C2H6 molecules favour sites with strong adsorbent interactions. At high pressures, packing effects played an important role and it became easy for small CH4 molecules to access the sites with strong adsorbate interactions.  相似文献

Insulin stimulates the mobilization of glucose transporter 4 (GLUT4) storage vesicles to the plasma membrane, resulting in an influx of glucose into target tissues such as muscle and fat. We present evidence that CLIP-associating protein 2 (CLASP2), a protein previously unassociated with insulin action, is responsive to insulin stimulation. Using mass spectrometry-based protein identification combined with phosphoantibody immunoprecipitation in L6 myotubes, we detected a 4.8-fold increase of CLASP2 in the anti-phosphoserine immunoprecipitates upon insulin stimulation. Western blotting of CLASP2 immunoprecipitates with the phosphoantibody confirmed the finding that CLASP2 undergoes insulin-stimulated phosphorylation, and a number of novel phosphorylation sites were identified. Confocal imaging of L6 myotubes revealed that CLASP2 colocalizes with GLUT4 at the plasma membrane within areas of insulin-mediated cortical actin remodeling. CLASP2 is responsible for directing the distal end of microtubules to the cell cortex, and it has been shown that GLUT4 travels along microtubule tracks. In support of the concept that CLASP2 plays a role in the trafficking of GLUT4 at the cell periphery, CLASP2 knockdown by siRNA in L6 myotubes interfered with insulin-stimulated GLUT4 localization to the plasma membrane. Furthermore, siRNA mediated knockdown of CLASP2 in 3T3-L1 adipocytes inhibited insulin-stimulated glucose transport. We therefore propose a new model for CLASP2 in insulin action, where CLASP2 directs the delivery of GLUT4 to cell cortex landing zones important for insulin action.  相似文献

Light-induced changes in the fluorescence of the pH-indicating dyes pyranine or 5-(and 6-)carboxy-2, 7-dichlorofluorescein (CDCF) which had been fed to leaves were examined to monitor cellular pH changes. After short-term feeding of pyranine (pK 7.3) to leaves of Amaranthus caudatus L., a NAD-malic-enzyme-type C4 plant, vascular bundles and surrounding cells became fluorescent. Fluorescence emission from mesophyll cells required longer feeding times. In CO2-free air, pyranine fluorescence increased much more on illumination after mesophyll cells had become fluorescent than when only the vascular bundles and the bundle sheath of Amaranthus leaves had been stained. After short feeding times and in the absence of actinic illumination, CO2 decreased pyranine fluorescence very slowly in Amaranthus and rapidly in C3 leaves. After prolonged feeding times, the extent of the light-dependent increase in pyranine fluorescence was several times greater in different C4 plants than in C3 species. The kinetics of the fluorescence changes were also remarkably different in C3 and C4 plants. Carbon dioxide (500 l · l–1) suppressed the light-induced increase in pyranine fluorescence more in C4 than in C3 leaves. Light-dependent changes in light scattering, which are indicative of chloroplast energization, and in 410-nm transmission, which indicate chloroplast movement, differed kinetically from those of the changes in pyranine fluorescence. Available evidence indicated that light-dependent changes in pyranine fluorescence did not originate from the apoplast of leaf cells. Microscopic observation led to the conclusion that, after prolonged feeding times or prolonged incubation, changes in pyranine fluorescence emitted from C4 leaves reflect pH changes mainly in the cytosol of mesophyll cells. A transient acidification reaction indicated by quenching of pyranine fluorescence in the dark-light transient and not observed in C3 species is attributed to the carboxylation of phosphoenolpyruvate. After short feeding times and in the absence of actinic illumination, CO2 (250 l l–1) decreased pyranine fluorescence very slowly in Amaranthus and more rapidly in C3 leaves. After prolonged feeding times, both the rate and the extent of CO2-dependent quenching of pyranine fluorescence increased, but the increase was insufficient to indicate the presence of highly active carbonic anhydrase in the compartment from which pyranine fluorescence was emitted. In contrast to pyranine, CDCF (pK 4.8) did not increase but rather decreased its fluorescence on illumination of an Amaranthus leaf, indicating acidification of an acidic compartment, most probably the vacuole of green leaf cells. The pattern of the acidification reaction was similar in C4 and C3 leaves. The remarkably large extent of the light-dependent increase in pyranine fluorescence from leaves of C4 species and its slow kinetics are proposed to be caused by an alkalization of the cytosol which in the absence of CO2 is larger in the mesophyll than in the bundle sheath. It gives rise to deprotonation of dye originally located in the mesophyll and, in addition, of dye which diffuses from the bundle sheath into the mesophyll following a pH gradient. We examined the expression of the major H2S-producing enzymes, cystathionine-β-synthase (CBS) and cystathionine-γ-lyase (CSE). CBS was ubiquitously distributed in the mouse pancreas, but CSE was found only in the exocrine. Freshly isolated islets expressed CBS, while CSE was faint. However, high glucose increased the CSE expression in the beta-cells. l-Cysteine or NaHS suppressed islet cell apoptosis with high glucose, and increased glutathione content in MIN6 beta-cells. Pretreatment with l-cysteine improved the secretory responsiveness following stimulation with glucose. The CSE inhibitor dl-propargylglycine antagonized these l-cysteine effects. We suggest HS may function as an ‘intrinsic brake’ which protects beta-cells from glucotoxicity.  相似文献

A scientific explanation for the beneficial role of vitamin D supplementation in the lowering of glycemia in diabetes remains to be determined. This study examined the biochemical mechanism by which vitamin D supplementation regulates glucose metabolism in diabetes. 3T3L1 adipocytes were treated with high glucose (HG, 25 mm) in the presence or absence of 1,25-dihydroxyvitamin D3 (1,25(OH)2D3) (25, 50 nm), the active form of vitamin D. 1,25(OH)2D3 treatment caused significant up-regulation of GLUT4 total protein expression and its translocation to cell surface, and an increase in glucose uptake as well as glucose utilization in HG-treated cells. 1,25(OH)2D3 also caused cystathionine-γ-lyase (CSE) activation and H2S formation in HG-treated adipocytes. The effect of 1,25(OH)2D3 on GLUT4 translocation, glucose utilization, and H2S formation was prevented by propargylglycine, an inhibitor of CSE that catalyzes H2S formation. Studies using antisense CSE also demonstrated the inhibition of GLUT4 translocation as well as glucose uptake and utilization in 1,25(OH)2D3-supplemented CSE-siRNA-transfected adipocytes compared with controls. 1,25(OH)2D3 treatment along with insulin enhanced GLUT4 translocation and glucose utilization compared with either insulin or 1,25(OH)2D3 alone in HG-treated adipocytes. 1,25(OH)2D3 supplementation also inhibited monocyte chemoattractant protein-1 and stimulated adiponectin secretion in HG-treated adipocytes, and this positive effect was prevented in propargylglycine-treated or CSE-knockdown adipocytes. This is the first report to demonstrate that 1,25(OH)2D3 up-regulates GLUT4 translocation and glucose utilization and decreases inflammatory markers, which is mediated by CSE activation and H2S formation in adipocytes. This study provides evidence for a novel molecular mechanism by which 1,25(OH)2D3 can up-regulate the GLUT4 translocation essential for maintenance of glucose metabolism.  相似文献

Summary Ethylene (C2H4) accumulation in flooded soil was related to oxygen (O2), redox potential (Eh), and flooding rate. The water status response of tobacco (Nicotiana, tabacum L.) to these conditions was evaluated from stem diameter, relative water content, leaf water potential, and C2H4 content of leaf tissue. Treatments were: flooded with either 0,5, or 15 cm of water per day for 6 days. By the third day, O2 in the soil decreased to less than 9% in treatments flooded with 5 or 15 cm of water. When O2 in the soil air was less than 9% and redox potential (Eh) was less than +150 mv, most of the soil air samples contained some C2H4 and 16% contained more than 6 ppm. Very little C2H4 was present in soil air when O2 exceeded 9%. Tobacco leaf C2H4 peaked 3 days after flooding and then declined to the preflooding level a day later, one day ahead of the rapid increase in soil C2H4. Wilting developed progressively beginning with the rise of C2H4 in the soil; leaf water potential, stem diameter, and relative leaf water content all were decreased. The quantum yield for CO2 uptake was measured in C3 and C4 monocot species from several different grassland habitats. When the quantum yield was measured in the presence of 21% O2 and 340 cm3 m-3 CO2, values were very similar in C3 monocots, C3 dicots, and C4 monocots (0.045–0.056 mole CO2 · mole-1 quanta absorbed). In the presence of 2% O2 and 800 cm3 m-3 CO2, enhancements of the quantum yield values occurred for the C3 plants (both monocots and dicots), but not for C4 monocots. A dependence of the quantum yield on leaf temperature was observed in the C3 grass, Agropyron smithii, but not in the C4 grass, Bouteloua gracilis, in 21% O2 and 340 cm3 m-3 CO2. At leaf temperatures between 22–25°C the quantum yield values were approximately equal in the two species.  相似文献

Species in the Laxa and Grandia groups of the genus Panicum are adapted to low, wet areas of tropical and subtropical America. Panicum milioides is a species with C3 photosynthesis and low apparent photorespiration and has been classified as a C3/C4 intermediate. Other species in the Laxa group are C3 with normal photorespiration. Panicum prionitis is a C4 species in the Grandia group. Since P. milioides has some leaf characteristics intermediate to C3 and C4 species, its photosynthetic response to irradiance and temperature was compared to the closely related C3 species, P. laxum and P. boliviense and to P. prionitis. The response of apparent photosynthesis to irradiance and temperature was similar to that of P. laxum and P. boliviense, with saturation at a photosynthetic photo flux density of about 1 mmol m-2 s-1 at 30°C and temperature optimum near 30°C. In contrast, P. prionitis showed no light saturation up to 2 mmol m-2 s-1 and an optimum temperature near 40°C. P. milioides exhibited low CO2 loss into CO2-free air in the light and this loss was nearly insensitive to temperature. Loss of CO2 in the light in the C3 species, P. laxum and P. boliviense, was several-fold higher than in P. milioides and increased 2- to 5-fold with increases in temperature from 10 to 40°C. The level of dark respiration and its response to temperature were similar in all four Panicum species examined. Carbon-isotope ratios were examined as 13C values in several C3, C4, and C3–C4 Flaveria species, and compared to predicted 13C, values generated from theoretical models. The measured 13C values were within 4 of those predicted from the models. The models were used to identify factors that contribute to C3-like 13C values in C3–C4 species that exhibit considerable C4-cycle activity. Two of the factors contributing to C3-like 13C values are high CO2 leakiness from the C4 pathway and pi/pa values that were higher than C4 congeners. A marked break occurred in the relationship between the percentage of atmospheric CO2 assimilated through the C4 cycle and the 13C value. Below 50% C4-cycle assimialtion there was no significant relationship between the variables, but above 50% the 13C values became less negative. These results demonstrate that the level of C4-cycle expression can increase from, 0 to 50% with little integration of carbon transfer from the C4 to the C3 cycle. Spartina maritima (Curtis) Fernald, Spartina densiflora Brong, Arthrocnemum perenne (Miller) Moss, and Arthrocnemum fruticosum (L.) Moq are very frequent halophytes on the coasts of SW Europe. The first two are perennial Gramineae with C4 metabolism; the last two are perennial Chenopodiaceae with C3 metabolism. Controlled garden experiments were carried out with the four species to compare their physiological response, i.e., water potential (Ψ), net photosynthetic rate (PN), transpiration rate (E), stomatal conductance (gs), intercellular CO2 concentration (Ci), and chlorophyll fluorescence of photosystem (PS) 2 under saline and non-saline conditions. S. maritima behaves as an osmoconformer species, the other three as osmoregulators. In the four species, PN, E, and gs improved following freshwater irrigation. The variations in PN might be related with biochemical changes (which appear not to affect PS2), but not with significant stomatal fluctuations, which are associated with a lower water use efficiency in the case of Arthrocnemum. The species were segregated into two groups (not depending on their C3 or C4 photosynthetic pathway), in relation with the topographic level of this species in natural conditions: the relative responses of PN in S. maritima and A. perenne were lower than those of S. densiflora and A. fruticosum. The salt-tolerance index supports such segregation. S. densiflora demonstrated the best competitive possibilities against salt-tolerant glycophytes, with its more flexible response in saline or brackish environments, which explains its spreading along the rivers draining into the estuaries of the SW Iberian Peninsula. The present study found that serum H2S level, H2S production rate, CSE mRNA and CSE protein levels were increased in CVB3-induced myocarditis. dl-proparglygylcine (PAG), an irreversible CSE inhibitor, decreased the infected myocardium titers on postinfection day 4, while NaHS, a H2S donor, alleviated myocardial injury and necrosis, inflammatory cell infiltration and interstitial edema on postinfection day 10. These data reveal that the CSE/H2S pathway is upregulated in the heart in a murine model of CVB3-induced myocarditis and that inhibition of endogenous H2S is beneficial to treatment early in the disease while administration of exogenous H2S is protective to infected myocardium during the later stage.  相似文献

Hydrogen peroxide (H2O2), an oxidizing agent, has been widely used as a disinfectant. Recently, because of its reactive properties, H2O2 has also been used as a tooth bleaching agent in dental care. This is a cause for concern because of adverse biological effects on the soft and hard tissues of the oral environment. To investigate the influence of H2O2 on odontoblasts, the cells producing dentin in the pulp, we assessed cellular viability, generation of reactive oxygen species (ROS), alkaline phosphatase (ALP) activity, and nodule formation of an odontoblastic cell line (MDPC-23) after treatment with H2O2, and compared those with the effects on preosteoblastic MC3T3-E1 cells. Cytotoxic effects of H2O2 began to appear at 0.3 mmol/L in both MDPC-23 and MC3T3-E1 cells. At that concentration, the accumulation of intracellular ROS was confirmed by a fluorescent probe, DCFH-DA. Although more ROS were detected in MDPC-23, the increasing pattern and rate are similar between the two cells. When the cells were treated with H2O2 at concentrations below 0.3 mmol/L, MDPC-23 displayed a significant increase in ALP activity and mineralized bone matrix, while MC3T3-E1 cells showed adverse effects of H2O2. It is known that ROS are generally harmful by-products of aerobic life and represent the primary cause of aging and numerous diseases. These data, however, suggest that ROS can induce in vitro cell differentiation, and that they play a more complex role in cell physiology than simply causing oxidative damage.  相似文献

Light-dependent CO-evolution by the green leaves of C3 and C4 plants depends on the CO2/O2 ratio in the ambient atmosphere. This and other physiological responses suggest that CO-evolution is a byproduct of photorespiration. At CO2/O2 ratios up to 10-3, the ratio of CO evolved: CO2 fixed in photosynthesis is significantly higher in C3 than in C4 plants. This discrepancy disappears when a correction is made for the CO2-concentrating mechanism in C4 photosynthesis, by which CO2-concentration at the site of ribulose-bis-phosphate carboxylase/oxygenase in the bundle sheaths is raised significantly as compared to the ambient atmosphere. Since the oxygenase function of this enzyme is responsible for glycolate synthesis, i.e., the substrate of photorespiration, this result seems to support the conclusion that CO-evolution is a consequence of photorespiration. Most species of the genus Salsola (Chenopodiaceae) that have been examined exhibit C4 photosynthesis in leaves. Four Salsola species from Central Asia were investigated in this study to determine the structural and functional relationships in photosynthesis of cotyledons compared to leaves, using anatomical (Kranz versus non-Kranz anatomy, chloroplast ultrastructure) and biochemical (activities of photosynthetic enzymes of the C3 and C4 pathways, 14C labeling of primary photosynthesis products and 13C/12C carbon isotope fractionation) criteria. The species included S. paulsenii from section Salsola, S. richteri from section Coccosalsola, S. laricina from section Caroxylon, and S. gemmascens from section Malpigipila. The results show that all four species have a C4 type of photosynthesis in leaves with a Salsoloid type Kranz anatomy, whereas both C3 and C4 types of photosynthesis were found in cotyledons. S. paulsenii and S. richteri have NADP- (NADP-ME) C4 type biochemistry with Salsoloid Kranz anatomy in both leaves and cotyledons. In S. laricina, both cotyledons and leaves have NAD-malic enzyme (NAD-ME) C4 type photosynthesis; however, while the leaves have Salsoloid type Kranz anatomy, cotyledons have Atriplicoid type Kranz anatomy. In S. gemmascens, cotyledons exhibit C3 type photosynthesis, while leaves perform NAD-ME type photosynthesis. Since the four species studied belong to different Salsola sections, this suggests that differences in photosynthetic types of leaves and cotyledons may be used as a basis or studies of the origin and evolution of C4 photosynthesis in the family Chenopodiaceae.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

Cytosolic and vacuolar pH changes caused by illumination or a changed composition of the gas phase were monitored in leaves of the NAD malic-enzyme-type C4 plant Amaranthus caudatus L. and the C3 plant Vicia faba L. by recording changes in the fluorescence of pH-indicating dyes which had been fed to the leaves. Light-dependent cytosolic alkalization and vacuolar acidification were maximal in the mesophyll cells under high-fluence-rate illumination and in the absence of CO2. Under the same conditions, measurements of light scattering and electrochromic absorption changes at 518 nm revealed maximum thylakoid energization. The results show an intimate relationship between the energization of the photosynthetic apparatus by light, an increase in cytosolic pH and a decrease in vacuolar pH. This was true for both the C4 and the C3 plant, although kinetics, extent and even direction of cytosolic pH changes differed considerably in these plants, reflecting the differences in photosynthetic carbon metabolism. Darkening produced rapid acidification in Vicia, but not in Amaranthus. Continued alkalization in Amaranthus is interpreted to be the result of the decarboxylation of a C4 intermediate and the release of liberated CO2. In the presence of CO2, energy consumption by carbon reduction decreased thylakoid energization, cytosolic alkalization and vacuolar acidification. Under low-fluence-rate illumination, thylakoid energization and light-dependent cytosolic and vacuolar pH changes were decreased in CO2-free air compared with thylakoid energization and pH changes in 1% oxygen/99% nitrogen not only in the C3 plant, but also in Amaranthus. Since oxygenation of ribulose bisphosphate initiates energy-consuming photorespiratory reactions in 21% oxygen, but not in 1% oxygen, this shows that photorespiratory reactions are active not only in the C3 but also in the C4 plant in the absence of external CO2. Photorespiratory conditions appeared to decrease energization not only in the chloroplasts, but also in the cytosol.
