The synapse architecture of the simcipital and auditory cortex of the cat (fields 7 and 22 after M. O. Gurevich and oth., 1929) was studied electron microscopically. In the both areas of the cortex there are much more axo-dendritic synapses that axo-somatic ones. In the upper layers the synapses are more often formed on small dendrites and thorns, while in layers IV-VI they often occur on the main trunks of large dendrites. The synapses on small branches and thorns of dendrites contain spherical vesicles, and the synapses on on large dendrites are formed by the terminals of two kinds-with flattened and spherical vesicles. The amount of axo-somatic synapses increases towards the lower layers of the cerebral cortes. The synapses on the soma and apical dendrites of the pyramid neurons always contain flattened vesicles; on the stellate neurons there occur perisynaptic terminals with sperical vesicles as well.  相似文献

An attempt has been made to discriminate additional types of synapses than have been previously described in the nucleus accumbens septi of the cat, which can, according to Brockhaus (1942), justifiably be termed the fundus striati due to the fact that it possesses all of the morphological and some of the neurochemical features of the striatum. This was undertaken in order to correlate at least one type of synapse with each different afferent pathway. Nine distinct types of synapses could be differentiated electron microscopically: Type I: axo-spinous synapses with sparse, small, round vesicles which seemed to be the nigro-striatal endings (35%). Type II: axo-somatic or axo-dendritic en passant synapses containing small, round vesicles (3%). Type III: axo-spinous synapses filled with densely-packed, small, round vesicles displaying strong postsynaptic thickenings which seem to be cortico-striatal (17%). Type IV: large axo-spinous synapses with densely-arranged, small, round vesicles contacting larger spines branching off a pedicle (9%). Type V: axo-somatic or axo-dendritic synapses containing large pleomorphic vesicles, probably axon collaterals (1%). Type VI: axo-somatic or axo-dendritic synapses with elongated small vesicles (20 X 45 nm) (3%). Type VII: large axo-somatic or axo-dendritic synapses filled by densely-packed, small, round vesicles (11%). Type VIII: large axo-somatic or axo-dendritic synapses containing loosely-arranged, small, round vesicles (8%). Axo-dendritic synaptic profiles were quantified along the whole depth of the visual cortex of 10-day-old male and female rats. In both sexes the numerical density of synaptic profiles on spine-like structures was greater than the numerical density of synapses on dendritic shafts. Females had a significantly greater numerical density of synaptic profiles on spine-like structures, than did males at a distance of 200–400 and 500–600 μm from the pia surface, which corresponds to layers II–III and IV of the cortex, respectively. A small percentage (2%–4%) of spine-like structures received two presynaptic terminals. This type of double synapses was three times more abundant in females. No sex differences were found in the numerical density of synapses on dendritic shafts in any cortical layer. 1994 John Wiley & Sons, Inc.  相似文献

Under study was the morphology of synaptic terminations of the brain reticular formation in cats and dogs as well as that of the afferent nuclei of the cat's posterior columns. The neurons of the Goll's and Burdach's nuclei have a richly ramified dendritic network. In this connection the main mass of synapses is disposed on the dendrites and their different branchings. The dendrites of the multipolar cells of the reticular formation have the main type of branching, but the cells are distributed from the nerve cell body at a considerable distance (up to 50 mu and more). The synapses are observed at the total length of the dendrite, so the axo-dendritic contacts quantitatively prevail over axo-somatic ones. The morphological data are compared with physiological axo-somatic and axo-dendritic concepts of the role of synapses in conducting the nerve impulse.  相似文献

Ultrastructural features of thalamic afferent fibers were studied in the cat auditory cortex in the early stages (on the 4th day) of experimental degeneration produced by destruction of the medial geniculate body. A coordinate grid was used in conjunction with an electron micro-scope to study the topography of the degenerating elements over wide areas of sections, so that the density of degeneration could be determined quantitatively in different layers of the cortex. Degenerating axons were found in all layers. Most of the large (5–7 µ) degenerating axons are located in layer VI; their diameters were smaller in the upper layers of the cortex. Degenerative changes affecting synaptic terminals of thalamo-cortical afferents were of the "dark" type. Fibers of the geniculo-cortical tract were shown to terminate mainly in cortical layer IV. A few degenerating synapses were found in the molecular layer. Terminals with sperical synaptic vesicles are found mainly on the spines of dendrites where they form "asymmetrical" contacts. A few degenerating axo-somatic synapses were observed on stellate neurons in layer IV. The results are discussed in connection with electrophysiological investigations of the cat auditory cortex during stimulation of specific afferent fibers.A. A. 本文用免疫电镜方法对脊髓胶状质内GABA能神经元的突触联系进行了超微结构研究。结果表明;脊髓胶状质内有许多GABA能神经元胞体和末梢分布;标记的GABA能神经末梢可作为突触前成分与未标记的GABA形成输一树突触。未标记的末梢可与标记的GABA末梢形成输一轴突触。此外,标记的GABA能神经末梢还可作为突触前成分与标记的GABA能轴突、树突或胞体形成输-轴、轴-树或轴-体突触,即自调节突触。上述结果揭示:GABA能末梢可对脊髓胶状质内其它神经元产生抑制或脱抑制作用。值得注意的是胶状质内含GAnA的神经结构可形成各种形式的自调节突触,并借此实现其对脊髓功能的复杂调节。  相似文献

To integrate information from different presynaptic cell types, dendrites receive distinct patterns of synapses from converging axons. How different afferents in?vivo establish specific connectivity patterns with the same dendrite is poorly understood. Here, we examine the synaptic development of three glutamatergic bipolar cell types converging onto?a common postsynaptic retinal ganglion cell. We find that after axons and dendrites target appropriate synaptic layers, patterns of connections among these neurons?diverge through selective changes in the conversion of axo-dendritic appositions to synapses. This process is differentially regulated by neurotransmission, which is required for the shift from single to multisynaptic appositions of one bipolar cell type but not for maintenance and elimination, respectively, of connections from the other two types. Thus, synaptic specificity among converging excitatory inputs in the?retina emerges via differential synaptic maturation of axo-dendritic appositions and is shaped by neurotransmission in a cell type-dependent manner.  相似文献

The characteristics and distribution of benzodiazepine receptors in individual layers of the visual cortex of adult rats were examined with the 3H-flunitrazepam binding technique employed on intact tissue slices. The different visual cortical layers were separated by cutting serial cryocut sections horizontally to the cortical surface and collecting the slices from each individual cortical layer under anatomical control. Highest benzodiazepine receptor densities were found in layers IV and VI. A moderate receptor density was detected in layer V (80% of highest density). The lowest receptor binding was observed in cortical layers I and II/III, still representing 66% of the highest receptor density. Binding affinities varied slightly between layers with dissociation constants somewhat higher for layers IV to VI in comparison to layers I and II/III. The distinct laminar pattern of benzodiazepine receptors in rat visual cortex suggests a differential neuromodulatory significance of these receptors in each individual cortical layer.  相似文献

Intraocularly injected colchicine suppresses axonal transport within the developing rat's optic nerve throughout the critical period of visual system development. This results in a stunting of retinofugal terminals and relay neurons in the lateral geniculate nucleus. The present study focuses upon the effects of this unique form of developmental deprivation on the maturation of the visual cortex. Colchicine, in concentrations of from 10(-5) to 10(-2) M, was injected into the eyes of albino rats at birth or at 5, 10, or 15 days of age. Litters were killed at 5 to 50 days after this single injection, and the brains were processed for Nissl, rapid Golgi, histochemical, or electron microscopic analysis. The following results were obtained: Planimetry of coronal sections of the striate cortex revealed a reduction in the thickness of the cortex and in the ratio of neuropil area to neuronal soma area contralateral to the injected eye which was confined principally to layer IV, lower layer III, and upper layer V. This effect was inversely related to postnatal age at injection and directly proportional to colchicine concentration. A rapid Golgi analysis of 51 pairs of layer V pyramidal neurons in control and experimental cortex demonstrated a reduction in the number and size of spines along the portion of the apical dendrite passing through lower layer III and IV following colchicine administration at birth or 5 or 10 days of age but no significant change in the branching pattern of the entire dendritic arbor. Electron microscopy revealed a reduction in the number of small, asymmetric synaptic complexes with the result that the average size of remaining profiles was increased in layers III and IV. Histochemical analysis of cortical succinic dehydrogenase and cytochrome oxidase revealed a distinct band of intense enzyme activity in lower layers III and IV in normal cortex at 20-30 days of age. This band was significantly reduced in intensity after neonatal injection of colchicine as shown by densitometric measurements and comparison of experimental and control cortex. It is concluded that the geniculocortical projection, while not affected directly by colchicine administration, is altered by the secondary effects of axonal transport suppression, leading to an alteration in the establishment of cortical synaptic patterns and arborizations of their postsynaptic neurons whose dendrites are located in those layers recipient to this projection.  相似文献

Previous light microscopical studies have indicated that fibres from the ventrolateral thalamic nucleus (VL) establish direct axo-somatic and axo-dendritic presumed contacts with layers III and V neurones of the intact frontal cortex projecting to the striatum. Additional experiments provided evidence that this thalamo-fronto-striate pathway could be partly reconstructed by transplantation of embryonic frontal tissue into the damaged cortex. The present study was undertaken to validate these results at the ultrastructural level. Several months after the transplantation of fetal frontal tissue into the damaged frontal cortex of newborn rats, a retrograde neurotracer (subunit b of the cholera toxin) was used to label the grafted neurones projecting to the striatum whereas an anterograde neurotracer (Phaseolus vulgaris leuco-agglutinin) was used to label within the transplant, axons and terminations arising from the VL. The same injection procedures were applied to intact adult rats (control). The distribution of retrograde and anterograde labellings within the intact cortex and within the graft was examined at light and electron microscopic levels to identify the synaptic contacts. Our findings showed that labelled contacts were less numerous within the transplant than within the intact cortex but their synaptic organization was similar: asymmetrical synaptic axo-dendritic and axo-somatic contacts. This synaptic articulation is probably supplied by a thalamic excitatory input. These results provide ultrastructural evidence of the capacity of a frontal cortical transplant placed in damaged frontal cortex of newborn rats to help reconstruction of appropriate synaptic integration within the thalamofronto-striate system.  相似文献

Knowledge-based or top-down influences on primary visual cortex (area V1) are believed to originate from information conveyed by extrastriate feedback axon connections. Understanding how this information is communicated to area V1 neurons relies in part on elucidating the quantitative as well as the qualitative nature of extrastriate pathway connectivity. A quantitative analysis of the connectivity based on anatomical data regarding the feedback pathway from extrastriate area V2 to area V1 in macaque monkey suggests (i) a total of around ten million or more area V2 axons project to area V1; (ii) the mean number of synaptic inputs from area V2 per upper-layer pyramidal cell in area V1 is less than 6% of all excitatory inputs; and (iii) the mean degree of convergence of area V2 afferents may be high, perhaps more than 100 afferent axons per cell. These results are consistent with empirical observations of the density of radial myelinated axons present in the upper layers in macaque area V1 and the proportion of excitatory extrastriate feedback synaptic inputs onto upper-layer neurons in rat visual cortex. Thus, in primate area V1, extrastriate feedback synapses onto upper-layer cells may, like geniculocortical afferent synapses onto layer IVC neurons, form only a small percentage of the total excitatory synaptic input.  相似文献

We have carried out an electron microscopic investigation of retrogradely HRP-labeled nonpyramidal neurons in layers V and VI of the primary auditory cortex (AI), which are sources of transcallosal projections. We have established that on average 15.8±1.7% of the perikaryon surface of these cells is occupied by axo-somatic synapses. We detected in ultrathin sections from two to nine synapses on the profiles of the perikaryon of callosal neurons. All of these axo-somatic synapses are formed by axon terminals containing small flat synaptic vesicles and are characterized by symmetrical contacts. The length of the cross section of the contacts was on average 1.6±0.1 µm. The axon terminals of callosal fibers, antegradely labeled by the enzyme, form in the deep layers of the cortex asymmetrical synapses on the spines and stems of the dendrites. A possible functional significance of the axo-somatic synapses in the production of the impulse activity of callosal neurons in the deep layers of the AI region, is discussed.A. A. The rhesus monkey's visual cortex was studied on Golgi material. The terminal arborization of the geniculate fibres and non-specific vertical fibres have been analysed. The interneurons (intrinsic neurons) of the area described in detail and classified on the basis of their axonal and dendrite arborizations. The stellate neurons in layer IV are discussed.  相似文献

The cortex of one hemisphere in cats was isolated according to M.M. Khananashwili method by means of surgical section of its projection fibres which connect it with subcortical structures. In five cats of means of Nissl's method the neuronal status of different cortical layers in the field 17 was studied during long postoperative terms (from 9 months up to 2 years). In 9 cats the neurons of the same field were impregnated after Golgi-Bubenett method and studied during the same postoperative terms. Cytological investigation in the majority of neurons did not reveal any pathologic changes during different postoperative terms up to one year. In two years, slight neuronal changes were noted presented in poor staining and vacuolization of cytoplasm. As a result of deafferentation performed shortening and thickness of dendritic branching was noted, as well as decrease in the number of thorns of large pyramids in layers III, IV, and V of the isolated cortex. Dendrites and thorns in small and medium-sized pyramids in layers II, III, IV, V, and VI were preserved to a great extent. It is possible to conclude, on the fact of thorns preserving in the major number of the neurons, that the neocortex possesses an enormous intracortical connective system that is responsible for the high degree of functional neuronal activity in the isolated cortex.  相似文献

Morphological relationships between neuropeptide Y- (NPY) like and ghrelin-like immunoreactive neurons in the arcuate nucleus (ARC) were examined using light and electron microscopy techniques. At the light microscope level, both neuron types were found distributed in the ARC and could be observed making contact with each other. Using a preembedding double immunostaining technique, some NPY-immunoreactive axon terminals were observed at the electron microscope level to make synapses on ghrelin-immunoreactive cell bodies and dendrites. While the axo-somatic synapses were mostly symmetric in nature, the axo-dendritic synapses were both symmetric and asymmetric. In contrast, ghrelin-like immunoreactive (ghrelin-LI) axon terminals were found to make synapses on NPY-like immunoreactive (NPY-LI) dendrites although no NPY-like immunoreactive perikarya were identified receiving synapses from ghrelin-LI axon terminals. NPY-like axon terminals were also found making synapses on NPY-like neurons. Axo-axonic synapses were also identified between NPY- and ghrelin-like axon terminals. The present study shows that NPY- and ghrelin-LI neurons could influence each other by synaptic transmission through axo-somatic, axo-dendritic and even axo-axonic synapses, and suggests that they participate in a common effort to regulate the food-intake behavior through complex synaptic relationships.  相似文献

The types of dendro-dendritic synapses and their participation in the synaptic, organization of superficial layers of the quadrigeminum superior tubercles were studied electron microscopically. In addition to simple forms of dendro-dentritic synapses the reciprocal dendro-dendritic synapses were revealed. Presynaptic dendrites formed the synaptic fields and glomerules of the superficial grey layer. The terminals of optical, cortical fibres from the visual cortex and other types of terminals terminated on presynaptic dendrites.  相似文献

Quantitative electronmicroscopic studies have been made on the development of synapses in two modally different areas of the brain (V-VI layers of the visual and auditory cortex) in the rat and mouse Acomys cahirinus within first two weeks of their postnatal life. The density of synapses as well the relative amount of different types of synapses (symmetrical, asymmetrical, axo-spinal and synapses with large amounts of synaptic vesicles) were measured. It was shown that only in rats the development of synapses in the visual area usually is faster than in the auditory one.  相似文献

Summary The development of neurons and their synapses of the mouse motor cortex has been studied from the first postnatal day up to an age of three weeks both electronmicroscopically and with the Golgi method. Special attention has been paid to the maturation of the different cell types in the sixth cortical layer and their dendritic organization within this layer.The polymorph layer is subdivided into two zones: an internal (VIb) and an external one (VIa). In these zones six different cell types can be identified both electronmicroscopically and with the Golgi method: large, small and inverted pyramidal cells in VIa; horizontal cells, star cells and small pyramidal cells in VIb.Spines of apical dendrites of large pyramidal cells in sublayer VIa can be detected as early as the 6th postnatal day. About the ninth day the basal dendrites as well show emerging spines. Somatic spines are found only on the large pyramidal cells and disappear slowly towards the end of the 3rd postnatal week.The small pyramidal cells show developing spines on their apical dendrite in the first half of the second postnatal week. The final density and distribution of spines is reached by the stem dendrites towards the end of the second week, by the basal dendrites during the third week. The maturation process of the improperly orientated neurons occurs in time in between the large and the small pyramidal cells.The axo-somatic synapses appear in general at a later date than the axo-dendritic ones. In the horizontal cells axo-somatic synapses are visible already at the seventh postnatal day.At the end of the first week especially in layer VIb many immature neurons with an ovoid or round nucleus are present having little if any endoplasmic reticulum organised as ergastoplasm.Towards the end of the second week however most neurons in the polymorph layer have a well developed endoplasmic reticulum.Electronmicroscopical pictures reveal in outgrowing dendrites many enlargements filled with vesicles, these correspond to the varicosities seen in Golgi pictures. Stages of formation of different types of synapse between cells of the dissociated spinal cord and spinal ganglia of 12–14 day mouse embryos, in monolayer cultures, were studied electron-microscopically. The participation of cones of growth in the formation of different junctions between structures of the monolayer was traced. It was shown that the appearance of synaptic vesicles in the growing axon precedes the onset of membrane specialization in the region of contact between axon and target cell. Ultrastructural characteristics of axo-dendritic, axo-somatic, and axo-axonal synapses formed during growth are given. On the 24th day of culture structural complexes of axonal glomerulus type, incorporating axo-axonal and axo-dendritic synapses, were discovered. It is suggested that desmosomes participate in the formation of both chemical synapses and synapses of gap junction type.A. A.
