Summary A new method of d(–)lactic acid production based on the aggregated form of growth of Bacillus laevolacticus in continuous culture in an anaerobic gaslift reactor is presented. With glucose as the substrate a bacterial dry weight of 25 g·1–1 and a lactic acid production rate of 13 g·1–1·h–1 was attained. In conventional glucose-limited chemostat cultures elevated levels of lactic acid stimulated specific lactic acid production while the formation rates of other end-products remained unaffected. In glucose-limited aggregated cultures lactic acid positively influenced the aggregation of cells, improving the volumetric production rate. It is concluded that lactic acid itself is a positive effector in the optimisation of lactic acid production with aggregated cultures of B. laevolacticus.Offprint requests to: J. P. de Boer  相似文献

A modified Rotating Biological Contactor (RBC) was used for the treatability studies of synthetic tapioca wastewaters. The RBC used was a four stage laboratory model and the discs were modified by attaching porous nechlon sheets to enhance biofilm area. Synthetic tapioca wastewaters were prepared with influent concentrations from 927 to 3600 mg/l of COD. Three hydraulic loads were used in the range of 0.03 to 0.09 m3·m–2·d–1 and the organic loads used were in the range of 28 to 306 g COD· m–2·d–1. The percentage COD removal were in the range from 97.4 to 68. RBC was operated at a rotating speed of 18 rpm which was found to be the optimal rotating speed. Biokinetic coefficients based on Kornegay and Hudson models were obtained using linear analysis. The combined effects of water activity (aw) and temperature on mycotoxin production by Penicilium commune (cyclopiazonic acid — CPA) and Aspergillus flavus (CPA and aflatoxins — AF) were studied on maize over a 14-day period using a statistical experimental design. Analysis of variance showed a highly significant interaction (P 0.001) between these factors and mycotoxin production. The minimum aw/temperature for CPA production (2264 ng g–1 P. commune, 709 ng g–1 A. flavus) was 0.90 aw/30 °C while greatest production (7678 ng g–1 P. commune, 1876 ng g–1 A. flavus) was produced at 0.98 aw/20 °C. Least AF (411 ng g–1) was produced at 0.90 aw/20 °C and most (3096 ng g–1) at 0.98 aw/30 °C.  相似文献

Styrene was degraded as sole source of carbon and energy by a selected bacterial community in a two-phase aqueous-organic medium (80%:20%, vol/vol). Silicone oil was used to solubilize styrene, which is sparingly soluble in water and to prevent its toxicity toward microorganisms. Preliminary studies with the mixed population in batch cultures indicate that the specific activity and the maximum growth rate at optimal 3H 6.0 were 46 mg·g–1·h–1 and 0.15 h–1, respectively. In pH-regulated chemostat cultures, styrene was degraded at dilution rates ranging from 0.05 to 0.20 h–1. Kinetic parameters and the proportion of each strain in the mixed culture were followed. At 0.20 h–1, only one strain as compared to four initially present, remained in the medium. This strain Pseudomonas aeruginosa, degrades styrene with a specific activity of 293 mg·g–1·h–1. Calli were obtained from Ginkgo biloba embryos grown on Murashige and Skoog (MS) medium. The G. biloba cells could grow on either MS or Gamborg B5 mineral salt medium supplemented with sucrose (3% and 2%, respectively) and naphthaleneacetic acid (NAA) and kinetin (K) in concentrations ranging from 0.1 to 2.0 mg·L–1. Best growth and maintenance of callus cultures were achieved using MS medium supplemented with 2 mg·L–1 NAA and 1 mg·L–1 K (N2K1MS). Light was required to maintain healthy growth of the callus tissue.In both MS and B5 based media, sucrose was hydrolyzed extracellularly before being taken up by Ginkgo cell suspension cultures. Specific growth rates of 0.13 d–1 and 0.08 d–1 were obtained in MS medium supplemented with 1 mg·L–1 NAA, 0.1 mg·L–1 K and 30 g·L–1 sucrose (N1K0.1MS) and B5 medium supplemented with the same growth regulator regime and 20 g·L–1 sucrose (N1K0.1B5) respectively. Complete phosphate and ammonium uptake was observed in 11 days when cultured in MS medium and 10 days and 4 days respectively when cultured in B5 medium. During the culture, G. biloba cells consumed only 64% and 29% of the nitrate content of N1K0.1MS and N1K0.1B5 media respectively. Maximum dry biomass concentrations were 13.4 g·L–1 and 7.9 g·L–1, and yields on carbohydrate were 0.39 and 0.45 in N1K0.1MS and N1K0.1B5 media respectively. An aroma-imparting mesophilic lactic starter (Lactococcus lactis ssp. lactis biovar. diacetylactis) was studied in batch culture in medium with 50 g·l–1 lactose and 2 g·l–1 citrate. The effect of pH on the physiology of growth and the production of flavour compounds was investigated with a mathematical model. The specific rates of growth and of lactose fermentation obeyed a law of non-competitive inhibition by lactic acid produced, inhibition increasing as the pH of the medium decreased. The pH thus acted indirectly by increasing the proportion of non-dissociated lactic acid, identified as the inhibiting form of lactic acid. The generalized model, taking into account the effect of pH, was tested using fermentations at pH controlled at different values (4.5–6.5), as well as with a fermentation conducted at non-regulated pH. These simulations supported the working hypotheses. The effect of pH on the fermentation of citric acid resulted in an increase in the maximal specific rate of citrate utilization, in the bioconversion yield, and in the constant of diacetyl and acetoin reduction at acid pH. The production of flavour compounds is a complex phenomenon resulting from the interaction of pH, citric acid concentration, and the physiological state of the cells. These results are discussed with respect to the use of this strain in the preparation of manufactured dairy products.  相似文献

An isolate of Isochrysis galbana rich in eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) has been grown as a chemostat culture at 20° C and pH 8.00 controlled by CO2 injection. From a low dilution rate (D) of 0.0024 h–1 to 0.0377 h–1, close to maximum growth, a decrease in EPA content from 5.21% dry weight (d.w.) to 2.80% d.w. was observed, although the percentage of EPA in the total fatty acids increased. Lipids were fractionated, EPA being the major fatty acid found in the glycolipid fraction, whereas in the neutral lipid fraction were mainly palmitic and palmitoleic acids. At the same time, the biomass concentration also decreased from 1015 mg·l–1 to 202 mg·l–1 over the range of Ds mentioned. Summary During tempeh fermentation, Rhizopus oligosporus produced polysaccharidases to degrade soya bean cell walls; the maximum activity for all polysaccharidases tested occurred 20–30 h after inoculation. R. oligosporus was also grown in a soya bean extract model medium to which glycerol was added to control water activity (a w). The overall activities of the major enzymes produced by the fungus, polygalacturonase, endocellulase and xylanase, appeared to be strongly influenced by a w. The production of enzymes as well as their specific activities were affected by a w. The optimum a w for polygalacturonase and xylanase activity coincided with that for mycelial growth, namely 0.99–1.00. Effect of water activity (a w) adjusted with NaCl or glycerol on the growth and metabolism ofLactobacillus helveticus var.pragensis at 40°C was demonstrated by growth curves (generation time) and changes in the degree of acidity of a milk medium expressed as lactic acid content. NaCl-regulateda w of 0.970 inhibited growth and acid production completely. The same, glycerol-regulateda w of 0.970 increased the generation time only from 33 min (a w=0.994) to 67 min which was less than the generation time at the highera w of 0.982 adjusted with NaCl (103 min). Glycerol-regulateda w of 0.970 did not decrease the acid production (2.6% lactic acid). Ata w of 0.951, the acid production was decreased by 39% compared with the values found in milk media with the original, unadjusteda w of 0.994, after the same time of incubation. Media witha w adjusted to the same values with NaCl or glycerol do not influence the growth and acid production ofL. helveticus in the same way. Summary The effect of various culture conditions on growth kinetics of an homofermentative strain of the lactic acid bacterium Streptococcus cremoris were investigated in batch cultures, in order to facilitate the production of this organism as a starter culture for the dairy industry. An optimal pH range of 6.3–6.9 was found and a lactose concentration of 37 g·l-1 was shown to be sufficient to cover the energetic demand for biomass formation, using the recommended medium. The study of the effect of lactic acid concentration on growth kinetics revealed that the end-product was not the sole factor affecting growth. The strain was characterized for its tolerance towards lactic acid and a critical concentration of 70 g·l-1 demonstrated. Embryogenic cultures and somatic embryos were obtained from immature zygotic embryos of oak (Quercus robur L.) cultured on a modified MS medium and WPM containing BAP (1 mg·l–1) and GA3 (1 mg·l–1) or BAP and IBA. Germination and conversion of oak somatic embryos into plantlets was achieved on WPM containing a reduced concentration of cytokinin. Linden (Tilia cordata Mill.) somatic embryos developed in embryogenic tissues initiated from immature zygotic embryos cultured on a modified MS medium supplemented with 2,4-D (0.3-2.0 mg·l–1). Germination of linden somatic embryos and plantlet formation occurred on MS medium containing a low concentration of IBA. Oak and linden plantlets produced from somatic embryos were successfully established in soil. The effect of water activity (aw) reduction on growth and acid and diacetyl production by three lactic streptococci was studied. In addition, the influence of low moisture conditions on several bacteria of significance in the fermentation of sauerkraut was examined. The minimal aw supporting growth of dairy lactics was 0.93 in a medium adjusted with glycerol. Media adjusted with sucrose generally were more inhibitory than those in which glycerol was the humectant. Titratable acidity, although not related to the type of humectant, did depend on the aw of the medium and was directly related to the extent of growth. Diacetyl concentration increased in cultures of reduced aw when the media were adjusted with both humectants; however, the effect was greatest with glycerol. A lactic strain associated with sauerkraut fermentation appeared to grow at a lower minimal aw in a glycerol-adjusted medium than in a system adjusted with NaCl; however, none of the sauerkraut organisms grew at aw levels of <0.95 when NaCl was the solute. Acid production appeared to be related to the presence and extent of growth at all of the aw levels studied.  相似文献

The purpose of these experiments is to determine the effects of foliar applications of benzylaminopurine (BAP) and gibberellic acid (GA) on tuber number production of seed potatoes. In field experiments conducted during 1989/90 cv. Mailén was used and BAP, 50 mg·l–1 was foliarly applied at (1) tuber initiation, 36 days after emergence (DAE); (2) 54 DAE; and, (3) 64 DAE. Under glasshouse conditions, in 1991/92 cv. Spunta was used and BAP 50 mg·l–1+GA 50 mg·l–1 were applied 30 and 37 days after planting/transplanting. In 1992 cv. Huinkul, Kennebec and Spunta were used and BAP 50 mg·l–1+GA 50 mg·l–1 and Biozyme (Techic SA), a commercial product with auxin (IAA, 32.2 mg·l–1), gibberellic acid (GA3, 32.2 mg·l–1) and cytokinins (zeatin, 83.2 mg·l–1) at 5 ml·l–1 were applied. In cv. Mailén, a higher tuber number in the seed fraction (<80 g) was found when BAP was applied at each of the three crop stages, while applications 54 and 62 DAE also increased tuber number in the 80–400 g fraction. As a result of BAP applications, tuber yield was also significantly increased. In the glasshouse experiments, cv. Spunta showed a significant increase in minituber production in 3 out of 4 cases, either if the mother plant came from in vitro generated plantlets or minitubers, or if GA + BAP or Biozyme was applied. It can be concluded that the use of these PGRs under both field and glasshouse conditions in cvs. Mailén and Spunta can result in increased tuber number in the seed fraction.  相似文献

The growth of the anaerobic acetogenic bacterium Acetobacterium woodii DSM 1030 was investigated in fructose-limited chemostat cultures. A defined medium was developed which contained fructose, mineral salts, cysteine · HCl and Ca pantothenate (1 mg · 1–1) supplied in a vitamin supplement. Growth at high dilution rates was dependent on the presence of CO2 in the gas phase. The max was found to be 0.16 h–1 and the fructose maintenance requirement was 0.1 to 0.13 mmol fructose · (g dry wt)–1 · h–1. A growth yield of 61 g dry wt · (mol fructose)–1, corrected for the cell maintenance requirement and for incorporation of fructose carbon into cell biomass, was determined from the fructose consumption. A corresponding growth yield of 69 g dry wt · (mol fructose)–1 was calculated from the acetate production assuming that fructose fermentation was homoacetogenic. A YATP of 12.2 to 13.8 g dry wt · (mol ATP)–1 was calculated from these growth yields using a value of 5 mol ATP · (mol fructose)–1 as an estimate of the amount of ATP synthesised from fructose fermentation. The addition of yeast extract (0.5 g · 1–1) to the medium did not influence the max or cell yield. Eicosapentaenoic (EPA) and docosahexaenoic (DHA) acid productivities from chemostat cultures of an isolate of Isochrysis galbana have been studied. The productivities reached in the interval of dilution rates between 0.0295 h–1 and 0.0355 h–1 were 1.5mg·1–1·h–1 for lipids, 300 g·1–1·h–1 for EPA and 130g1·1–1·h–1 for DHA. Furthermore, light attenuation by mutual shading, and agitation speed influences on growth and fatty acid composition were analysed. A model relating steady-state dilution rates to internal average light intensity has been proposed, the parameter values of which obtained by non-linear regression were: maximum specific growth rate (max)=0.0426 h–1; the affinity of cells to light (Ik) = 10.92 W·m–2; the exponent (n) = 5.13; regression coefficient (r 2)=0.9999. A method for leaf mesophyll protoplast isolation and plant regeneration of cucumber (Cucumis sativus L.) is described. Using an isolation solution complemented with 0.1 M glycine, 8.2·106 viable protoplasts were isolated from 1 g of fresh leaves. The effect of the growth substances indole-3-acetic acid, naphthalene acetic acid, 2,4,-dichlorophenoxy acetic acid, 6-benzylaminopurine, 2-isopentenyladenine and kinetin at concentrations from 0.5 to 5 mg·1–1 was studied using the multi-hanging drop technique. The optimal growth substance combination, namely 5 mg·1–1 naphthalene acetic acid and 3 mg·1–1 2-isopentenyladenine, together with agarose medium in a so-called bead culture resulted in a plating efficiency of 21%. Summary A procedure leading to the regeneration of whole plants from protoplasts of melon is described. Protoplasts were isolated from cotyledons and leaves of plants grown in vitro. After 14 days of culture, average viability and division rates were respectively 60% and 30% for the two organs, considering total initial protoplasts plated. The manipulation of the exogenous auxin / cytokinin balance in regeneration media enabled to direct morphogenesis towards somatic embryogenesis (1 mg·l–1 2,4-dichlorophenoxyacetic acid and 0.1 mg·l–1 6-benzylaminopurine) or caulogenesis (0.5 mg·l–1 6-benzylaminopurine and 0.5 mg·l–1 kinetin). Contrary to division ability, regeneration capacity was genotype-dependent under our conditions, but the two organs expressed similar division and regeneration capacities. Maltose was superior to sucrose for the development of caulogenic nodules into buds. Summary The effect of water activity (aw) on the growth and end-product formation of Lactobacillus viridescens SMRICC 174, Lactobacillus SMRICC 173 (homofermentative) and Brochothrix thermosphacta ATCC 11509T was studied. All strains orginated from meat or meat products. The aw was adjusted in the range 0.94–0.99 with NaCl or glycerol. A greater reduction in growth rates was found for L. viridescens and B. thermosphacta when aw was regulated with NaCl rather than with glycerol, the opposite was true for Lactobacillus 173. L. viridescens grew at aw >-0.94. At 0.94 aw B. thermosphacta was totally inhibited when NaCl was the solute and Lactobacillus 173 when glycerol was the solute. Only minor variations in the end-product formation of the Lactobacillus spp. were found at different aw values. In aerobic culture B. thermosphacta produced less l-lactic acid and more acetic acid as the aw was decreased with NaCl, while the yields were unaffected when glycerol was used.  相似文献

Aims: To model the effect of water activity (aw) and concentration of undissociated lactic acid (HLac) on the time to growth (TTG) and the growth/no growth boundary of acid‐adapted generic Escherichia coli, used as model organisms for Shiga toxin‐producing E. coli (STEC). Methods and Results: For each of two E. coli strains, the TTG in brain heart infusion broth at 27°C was estimated at 30 combinations of aw (range 0·945–0·995) and concentration of HLac (range 0–6·9 mol m?3) by using an automated turbidity reader. Survival analysis was used to develop a model predicting the TTG and the growth/no growth boundary. Conclusions: The present model can be used to predict the TTG and to indicate the growth/no growth boundary of acid‐adapted E. coli strains as a function of aw and concentration of HLac. Significance and Impact of the Study: Fermented food products have been implicated as sources of STEC in several outbreaks. The study results are relevant for modelling of growth of STEC in fermented food and can be used in microbiological risk assessments or in the design and validation of food‐production processes.  相似文献

Summary The production of -linolenic acid (GLA) by the fungus Mucor rouxii CBS 416.77 was studied on low budget nitrogen and carbon sources, i.e. rape meal, cocos expeller and two types of yeast extract (nitrogen sources), and starch, starch hydrolysate, beet molasses and cocos expeller (carbon sources). As references, Difco yeast extract and glucose were used. In flask cultivations the three yeast extracts were fully interchangeable, while the Difco yeast extract (the most expensive of those tested) gave a higher productivity of GLA in fermentor cultures (14 mg·l–1·h–1). The yield of lipids and GLA were increased in the order yeast extract < rape meal < cocos expeller. Thus the amount of lipid increased from 0.56 to 2.8 g·l–1, and that of GLA from 0.15 to 0.33 g·l–1. Use of beet molasses or cocos expeller as carbon sources gave poor growth. Starch and starch hydrolysate resulted in better productivity of GLA than glucose (4.7 and 4.9 compared to 3.4 mg·l–1·h–1).
