We have attempted to optimize the conditions under which a photoautotrophic soybean suspension culture line (SB-P; Horn et al. 1983) is grown. Magnesium, phosphate, and calcium concentrations were varied individually from one-tenth to five times the normal level found in the Murashige and Skoog (1962) recipe. After two subcultures, only phosphate at one-tenth the normal level caused the cells to show a substantial reduction in fresh and dry weight increase and chlorophyll level. Nitrate and ammonium levels were inversely varied in 20 millimolar increments of potassium nitrate and ammonium chloride. Neither N-source alone could support growth through two subcultures. A ratio of 40 millimolar potassium nitrate to 20 millimolar ammonium gave significantly better fresh and dry weight increases than did a ratio of 20:40 or 30:30 but the chlorophyll level was unchanged. The minor salts as a group resulted in a small improvement in growth when provided at twice the normal level.Indole-3-acetic acid at five milligrams per liter resulted in significantly better fresh and dry weight increases than did -naphthaleneacetic acid at any level but the final chlorophyll level was not changed. There was no correlation between growth and kinetin level and this resulted in the discovery that SB-P cells are cytokinin-autotrophic, as are heterotrophic SB cells, with regard to both growth and greening ability. Growing SB-P cells under a 14 h:10 h day:night photoperiod resulted in a slow but inevitable death. Increasing the carbon dioxide level to 10% for four weeks gave no increase in SB-P cell growth or chlorophyll level, but SB-P cells would not grow with carbon dioxide levels below 0.4%. The results clearly show that SB-P cells, despite their tenuous existence, are capable of adapting to a wide range of culture conditions. Body and egg volumes of 12 rotifer species of Lake Constance were calculated from more than 18 000 individually measured animals using geometric approximations. Body (V A) and egg (V E) volumes of various rotifer species have an allometric relationship described by: V E = 1272 · V A 0.3379, while the relation V E/V A for each species was constant ranging from 0.016 to 0.87. Dry weight of adult rotifers was assumed to be the sum of the egg dry weight plus a corrected weight increment from egg to adult. It was estimated with the formula W A = W E · (1 + a · F A/F E) where (a) was varied between 0 and 0.1. Egg dry weight content was assumed to be 35 % of fresh weight. The estimates of dry weight to wet weight ratios ranged from 0.57 % (Asplanchna priodonta) to 29.4 % (Kellicottia longispina). The allometric equation, y=gxh, was applied monthly to the relationships between two different dimensions of tree seedlings of hinoki cypress (Chamaecyparis obtusa) during a course of self-thinning from April 1990 to March 1991 to detect differences in biomass allocation among individuals. As the h-value in the allometry of crown length and seedling height was greater than unity for all seasons, crown ratio became greater as seedling height increased. Leaf weight ratio increased with increasing seedling size because the h-value in the allometry of leaf dry weight and whole seedling dry weight was greater than unity in every month. Therefore, smaller seedlings are disadvantageous to photosynthetic production by leaves. In contrast, the leaf area ratio was constant irrespective of seedling size because the h-value in the allometry of leaf area and whole seedling dry weight was nearly equal to unity in most seasons. In addition, because the h-value in the allometry of leaf area and leaf dry weight was less than unity in all seasons, specific leaf area decreased with an increase in leaf dry weight, indicating that smaller seedlings adapt to low light environments by possessing shade leaves. Root weight ratio decreased with increasing seedling size because the h-value in the allometry of root dry weight and whole seedling dry weight was less than unity in most seasons.  相似文献

Summary The effect of water stress on growth (fresh weight, dry weight), water relations (water saturation deficit, water potential, osmotic pressure), and proline metabolism in Phaseolus vulgaris were studied.Experimentally, water deficit was produced by reduced watering of the bean plants. This resulted in a decrease in water potential and leaf fresh and dry weight. Increases in the water saturation deficit and the osmotic pressure of the sap were, however, recorded. Water stress was also induced by treatment of the plants with polyethylene glycol, but its effects on the above mentioned parameters were different. In addition, necrosis of the foliage was observed. According to the present results, polyethylene glycol seems to be suitable only for the induction of short-termed water stress conditions.The effects of water stress on growth and on water relations of the plants were accompanied by a marked increase in the free amino acid content, especially that of the free proline content of the plants. The activities of the proline dehydrogenase and glutamate dehydrogenase were stimulated under water stress conditions, indicating that proline accumulation in water stressed plants is not attributed to an inhibited proline breakdown. The accumulated proline was metabolized rapidly once the water deficit of the plant was relieved by watering. In vitro screening procedure for osmotic tolerance in Prunus   总被引:1,自引:0,他引:1  
Significant growth differences (p<0.01) were observed for two micropropagated Prunus accessions after 14 days in culture when 685 mM mannitol was included in Quoirin and Lepoivre nutrient medium. While there was an 11% growth increase in fresh weight during the 28-day culture period for accession K537-067, explants from New Jersey Plumcot No. 3 increased fresh weight an average of 123%. Similar tests were conducted to determine the repeatability of this short term in vitro screening procedure. Explants of Mananna 2624 were subjected to two levels of mannitol, 275 mM and 550 mM, included in the Quoirin and Lepoivre nutrient medium. Three successive 28-day tests were conducted. Explants were examined at both 14 and 28 days after the onset of the experiment for net growth changes. Addition of mannitol to the nutrient medium at concentrations of 275 mM and 550 mM decreased explant fresh weights of Marianna 2624 to 36% and 28% of controls, respectively, at 28 days past initial culture. Initial fresh weight and fresh weight changes at day 14 were significantly different (p 0.05) between tests. No significant differences existed between tests with regard to weight changes at 28 days past initial culture. The effect of lettuce (Latuca sativa L.) germination and growth in nonsterilized potting compost of 0.1% and 1.0% w/w incorporation of fermenter biomass inocula of six strains of Trichoderma was investigated. Except for strains WT and T35 at 0.1 % w/w, all inocula inhibited germination. Biomass of strains WT, T35, 20, and 47 at 1.0% promoted shoot fresh weight, whereas strains TH1 and 8MF2 were inhibitory. In contrast, when biomass of strains WT, TH1, and 8MF2 was autoclaved and incorporated at 1%, shoot fresh weight was promoted, but the biomass of T35 was inhibitory. None of the strains incorporated at 0.1 % w/w increased shoot fresh weight, and autoclaved biomass of TH1, T35, and 20 incorporated at 0.1% w/w resulted in lower shoot fresh weights in comparison with uninoculated controls. The shoot dry weight of lettuce seedlings could be enhanced by germinating seeds in uninoculated compost and after five days' growth transferring them into WT-inoculated compost. Inoculum of strain TH1 when applied using this method was very inhibitory. A continuous culture apparatus is described, which allows cultivation of photosynthetic bacteria anaerobically in the light and semiaerobically in the dark at constant oxygen tensions. The growth-parameters 1. substrate concentration at half maximum growth rate (K s) and 2. yield (Y) forRhodopseudomonas capsulata are calculated.An automatic control system of the oxygen partial pressure in the culture medium is elaborated. It is shown, that the discontinuous regulation with a control unit in connection with two magnetic valves, which give short pulses of either oxygen-free gas or gas mixed with oxygen, is an economic, practicable and reliable method.The yield coefficientY during growth limited by ammonium sulfate is variable due to the synthesis of nitrogen independent metabolites, such as poly--hydroxybutyrate. The storage of poly--hydroxybutyrate in continuous culture is a function of both the actual concentration of ammonia and of theC/N ratio. At very low growth rates (1/6 µ m ) the poly--hydroxybutyrate content increases amounting to 33% of the dry weight.In semiaerobically dark grown cells (pO2: 5 mm Hg) the protein and bacteriochlorophyll content increased definitely on dry weight basis with increasing growth rates. In contrast, in anaerobic light cultures only a small increase of the bacteriochlorophyll level but no change of the protein content per dry weight of cells was observed at increasing growth rates.  相似文献

Rotifers (Brachionus plicatilis) grown atdifferent growth rate ( = 0.05–0.39 d–1)were analyzed for protein, lipid, fatty acids, aminoacids and free amino acids, and values are expressedin terms of individuals and dry weight. Increase ingrowth rate is equivalent with increased food rationof the individual rotifer, which responded by higheregg ratio. The protein content per individualrotifer increased by 60–80% when the growth rate increased, whereas the protein content per dryweight showed a slight, although insignificant,increase (p > 0.05). The lipid content perindividual was constant, whereas lipid per dryweight decreased when the growth rate increased. Theratio DHA/EPA decreased when the growth ratesincreased. The amino acids profile in percent oftotal amino acids showed low variation betweencultures maintained at different growth rates,whereas the values expressed in terms of amino acidper individual showed higher variation. The range ofvariation for free amino acids was more pronouncedthan for total amino acids.Short-term food enrichment of poorly fed rotifers( = 0.05 d–1) with balanced protein richdiet resulted in increased protein and lipid contentper rotifer. The protein content per dry weightshowed only minor changes whereas lipid per dryweight increased. Contrary, short term enrichmentwith a lipid rich diet resulted in increased lipidcontent per individual rotifer and per dry weight,whereas the protein content per individual remainedconstant and the protein content per dry weightshowed a slight decrease.Our experiments show that the amount of protein, wasquite variable in rotifers, and that feeding andgrowth condition were decisive factors affecting it.The range of variation was large enough to be animportant factor during first feeding of marinelarvae, and should therefore be considered infeeding larvae.  相似文献

Summary Sodium chloride reduced the growth of the wheat cultivar Armada. Although phenylmercuric acetate (PMA) also reduced the growth under non-saline conditions, in the presence of sodium chloride (100 mM) fresh weight, dry weight and leaf area per plant were promoted particularly when sprayed with PMA at 50 M. Both net photosynthesis and transpiration were reduced by PMA but the reduction was greater in the absence rather than the presence of NaCl. Furthermore PMA lowered shoot sodium contents and promoted the selectivity for K over Na under saline conditions. Any beneficial effects of PMA are the consequences of improved water relations, lowered ion content and increased leaf area for photosynthesis.  相似文献

The hypothesis tested in this paper is that, because the freshest water occurs in the largest soil pores (macropores), plants of low to moderate transpiration rate can survive in salinized soil because they preferentially extract water from macropores. The hypothesis predicts that a plant growing in a macroporous soil should have greater growth under a given salinity treatment than a similar plant growing in a soil with the same mineralogy but without macropores. This hypothesis was tested by growing bell pepper (Capsicum annuum) in the greenhouse in pots filled with either a commercial fritted clay (a highly macroporous soil) or the same clay ground to a finer texture and sieved to remove macropores and produce a microporous soil. The pots sat in pans filled with salt water. Half of the pots were irrigated once a day with fresh water and the other half received no fresh water. Plants growing in the macroporous soil had greater growth for a given salinity treatment than the plants growing in the microporous soil under both the irrigated and non-irrigated conditions. Under the irrigated condition for the highest salinity treatment, the non-reproductive fresh weight per plant, total dry weight per plant and fruit fresh weight per plant was 114 g, 12 g and 50 g, respectively, for the macroporous soil and 47 g, 4.5 g and 5 g, respectively, for the microporous soil. The results of this study provide evidence to suggest that a better understanding of what constitutes a good structure in a saline soil may aid us in our efforts to improve the management of saline soils. We suggest that it may be possible to increase the agricultural production on salinized land by no-tillage agriculture which preserves macroporosity. Possible obstacles could be the tendency of field saline-sodic soils to swell and the unavailability of relatively fresh irrigation water in areas with saline soils.  相似文献

A factorial experiment was conducted to determine the effect of aluminium (0 and 600M) and media (sand, and 1:1 sand:soil) on mycorrhizal (M) and non-mycorrhizal (NM) highbush blueberry plantlets. There were no differences in nutrient uptake and total plant dry weight between M and NM plantlets. However, more root growth, as determined by dry weight, was observed in M than NM plantlets. The plantlets growing in sand had more dry weight than did those in the soil medium. Although the root growth and shoot growth were reduced by the 600M Al treatment, the direct effect of Al on plantlet growth was not clear due to Al and P interactions. Plant nutrient uptake was reduced by high concentrations of Al, suggesting that high Al concentration limited the ability of roots to acquire most of the nutrients. Mycorrhizal cortical cell infection levels of 15–20% wene maintained in the roots in soil medium but decreased to about 5% over the 6 weeks of the experiment in the sand medium. Although M plantlets accumulated more Al in their roots, Al was readily transported to the leaf tissues of M and NM plantlets.  相似文献

Artemisinin was produced in differentiated shoot cultures of Artemisia annua L. but was undetected in callus or cell cultures. The growth regulators benzyladenine, kinetin, chlormequat, and daminozide, at concentrations which severely reduced rooting, reduced artemisinin production. A highly significant correlation (1% level) was observed between shoot artemisinin content and number of roots (r=0.775**), but shoot number and artemisinin content were unrelated (r=-0.198). Benzyladenine increased shoot proliferation at 0.5 and 5.0 M, but decreased root production at 0.5, 5.0, and 50 M. The highest levels of artemisinin production (0.287% DW) were obtained in hormone-free medium when root production was maximized. Removal of roots from shoots cultured in hormone-free liquid medium reduced shoot artemisinin by 53% and shoot arteannuin B by 60%. Hairy roots were obtained following inoculation of the stems of Lobelia inflata L. with Agrobacterium rhizogenes strain ATCC 15834. These hairy roots contained agropine and mannopine. In addition, lobeline was detected by HPLC and confirmed by mass spectrometry. Various media were tested for the growth of hairy roots as well as for the content of lobeline in hairy roots. The growth rate of hairy roots cultured in Nitsch and Nitsch's medium was approximately one third of those cultured in other media. The lobeline content of hairy roots (18–54 g/g dry weight) cultured in these media was the same order of magnitude compared with that of roots of L. inflata (24 g/g dry weight) cultivated in pots. Molar growth yield studies on chemostat cultures of Thiobacillus neapolitanus grown in thiosulfate-minerals medium have confirmed earlier observations that the dry weight increased linearly with the dilution rate. The observed increase can be explained neither by a change in cell composition nor by the observed excretion of organic compounds. The increase of the molar growth yield over the full range of growth rates, that is also observed in other obligate chemolithotrophs, was not found in the facultatively chemolithotrophic Thiobacillus A2, grown on thiosulfate or formate. The interpretation of the results in terms of maintenance energy requirement is discussed. The effects of 5-bromo-2-deoxyuridine (BUdR, thymidine analogue), AMO-1618 (2-isopropyl-4-dimethylamino-5-methylphenyl-1-piperidine carboxylate methyl chloride), a growth retardant, and p-chlorophenoxyisobutyric acid (PCIB, an antiauxin) on growth (dry weight increase) and fiber development in unfertilized cotton (Gossypium hirsutum L.) ovules grown in vitro have been studied. BUdR (5 M) causes about 70% inhibition of fiber production, with little effect on ovule growth, if applied during the first 6 d of culture in the presence of GA3 and IAA. AMO-1618, when used with GA3 alone, causes only a small reduction in both dry weight and fiber production, but when used with IAA alone reduces both fiber production and dry weight, the effect on the latter being predominant. In the presence of both IAA and GA3, AMO-1618 causes a small decrease in fiber production but a major decrease in dry weight. PCIB completely inhibits fiber growth but has little effect on dry weight, especially when GA3 is present. A laboratory experiment was performed to study the responsiveness of selected condition measures to starvation in herring, Clupea harengus, larvae in relation to temperature and ontogeny. The larvae at two intervals of development, i.e. stage 1 larvae with initial exogenous feeding and stage 2 larvae with established feeding prior to notochord flexion, were reared at three temperatures (5, 8 and 11°C) and subjected to sub-lethal durations of starvation. Temporal changes in standard length, dry weight, DNA concentration (% of dry weight), RNA concentration (% of dry weight), Fulton's condition factor (CF) and RNA/DNA were assessed and compared with fed controls. Starvation led to decreases in dry weight, CF, RNA concentration and RNA/DNA, while it led to an increase in DNA concentration. Higher responsiveness to starvation was observed at higher temperatures, and the magnitude of the changes was higher in stage 2 larvae. The shortest latency in starvation response was found with respect to RNA/DNA which was length independent in the size range studied. RNA/DNA was also significantly related to average DNA growth rate, and the model for DNA growth rate was, SGRDNA=4.49 RNA/DNA + 7.14 T – 0.42 T2 – 37.5 ; n=32, r2=0.85, p < 0.001). While the model seemed to adequately represent the average temperature dependent DNA growth, a relatively low classification success made it unsuitable for depicting individually starving larvae. Critical levels in DNA concentration can be used (2.2% for stage 1 larvae, 2.9% for stage 2 larvae) to differentiate starving larvae (after 3 – 5 days) from feeding larvae. RNA/DNA was the most sensitive and suitable condition index studied in detecting early starvation of herring larvae.  相似文献

Maltose-limited continuous culture of Aspergillus niger was carried out with potassium nitrate to investigate growth and glucoamylase formation characteristics. Glucoamylase production was dependent on the specific growth rate. The maximal amount of glucoamylase (units/l and U/g dry weight) was obtained at =0.08h–1, and the maximum specific rate of production (units/g/dry weight per hour) was at =0.2h–1. The maintenance coefficients (ms and mATP) were higher than for some other fungi. Maximal growth yields on substrate, oxygen and ATP (Yxsm, YxO2m and Yxam) were very efficient (high) and the value of Yxam, which cannot exceed the theoretical maximal value, is obtained when a P/O ratio of 1:1 is assumed. This indicates that biomass formation is energetically inexpensive and most of the expended energy has to be invested in the process of glucoamylase excretion.  相似文献

The aim of this study was to determine the plant growth-promoting potential of the nodule endophytic Pseudomonas brassicacearum strain Zy-2-1 when used as a co-inoculant of Medicago lupulina with Sinorhizobium meliloti under copper (Cu) stress conditions. Strain Zy-2-1 was capable of producing ACC deaminase activity, IAA and siderophores, and was able to grow in the presence of Cu2+ up to 2.0 mmol/L. Co-inoculation of S. meliloti with Zy-2-1 enhanced M. lupulina root fresh weight, total plant dry weight, number of nodules, nodule fresh weight and nitrogen content in the presence of 100 or 300 mg/kg Cu2+. In the presence of 500 mg/kg Cu2+, co-inoculation with S. meliloti and strain Zy-2-1 increased plant height, number of nodules, nodule fresh weight and nitrogen content in comparison to S. meliloti inoculation alone. Furthermore, a higher amount of Cu accumulation in both shoots and roots and a higher level of Cu translocation to shoots were observed in co-inoculated plants. These results demonstrate that co-inoculation of M. lupulina with S. meliloti and P. brassicacearum Zy-2-1 improves plant growth, nitrogen nutrition and metal extraction potential. This can be of practical importance in the remediation of heavy metal-contaminated soils.  相似文献
