The ultrastructure of the Malpighian tubules of the adult desert locust, Schistocerca gregaria, is described. Male and female adults possess about 233 tubules, which empty proximally into the midgut-ileal region of the alimentary canal by way of 12 ampullae. The tubules vary from 10 mm to 23 mm in length. About one third of them are directed anteriorly, attaching distally at the caeca, while the remainder are directed posteriorly, attaching to other tubules, the rectum or large tracheal trunks adjacent to the hindgut. The Malpighian tubules from all locations examined consist of three ultrastructurally distinct regions: proximal, middle, and distal, referring to their position relative to the midgut. All cell types possess ultrastructural features characteristic of ion transporting tissue, i.e., elaboration of the basal and apical membranes and a close association of these membranes with mitochondria. The distal and proximal segments are short (1.5-1.7 mm) and heavily tracheated, and each is composed of a single, distinct cell type. The middle region is the longest segment of the Malpighian tubule and is composed of two distinct cell types, primary and secondary. Both cell types are binucleate. The more numerous primary cells have large nuclei, contain laminate concretions in membrane-bound vacuoles, and possess large microvilli that contain mitochondria. The secondary cells are smaller and possess smaller nuclei. The microvilli are reduced and lack mitochondria. Secondary cells do not contain laminate concretions. The possible compartmentalization of ion and fluid transport function based on segmentation in the Malpighian tubules is discussed.  相似文献

Subcellular changes in the embryo and endosperm of Atropa belladonna were studied at four developmental stages. The endosperm cells turn to storage cells much earlier than those of the embryo, which matures later. Cells of the cotyledon and radicle are very similar in structure. The young cells contain large osmiophilic spherosomes. The cytoplasm is filled with ribosomes but dictyosomes are very rare. Some proplastids, containing starch, and mitochondria are present in the early developmental stages but do not occur in the dormant cells. During ripening, the vacuoles of the endosperm cells and embryo develop into protein bodies. They become filled with protein material without any recognisable transport mechanism. Protein bodies have several electron-translucent globoid cavities and the protein mass contains a roundish or crystalline body. This body does not stain with potassium iodide but with periodic acid Schiff-reagent and protein stains, indicating that it contains glycoproteins. The embryo and endosperm cells of ripe Atropa seeds are very similar and filled with protein bodies and small spherosomes.  相似文献

During spermatogenesis in the crab, Carcinus maenas, mitochondria in the developing spermatids degenerate. In close association with mitochondrial fragments in late spermatids, an elaborate lattice-like complex of fused membranes is formed and lies in a position adjacent to the acrosome. Mature sperm possess no mitochondria, but a whorl of membranes is present alongside the acrosome in the diffuse nucleus. To ascertain whether or not cytochrome c oxidase activity is lost as the mitochondria disintegrate, differentiating Carcinus maenas spermatids and mature sperm have been studied cytochemically. Cytochfome c oxidase activity was localised by means of the osmophilic compound 3,3'-diaminobenzidine (DAB). Cytochrome c oxidase activity is confined to the cristae of mitochondria in the testis. As spermatids mature most mitochondria are lost. A few mitochondrial fragments may be caught up within the lamellar complex. While they are recognisalbe as mitochondria they retain cytochrome c oxidase activity. The lamellar complex does not show this enzyme activity. These results therefore suggest that the mature sperm of Carcinus maenas do not contain the enzymes normally incorporated in the mitochondrial membrane, capable of oxidative phosphorylation.  相似文献

Distinguishable patterns of cytoplasmic membrane senescence in cotyledon tissue of Phaseolus vulgaris have been elucidated by examining the behavior of four microsomal enzymes—NADH-cytochrome C reductase, NADPH-cytochrome C reductase, glucose-6-phosphatase and 5′-nucleotidase during germination. For young cotyledon tissue, specific activities for the phosphatases were similar for rough and smooth microsomal fractions, but both cytochrome C reductases were 2–3 times more concentrated in the smooth fraction. These proportionalities changed with increasing age. As senescence becomes more intense the enzyme activities change independently of one another. These changes do not appear to be influenced by the presence or absence of ribosomes on the membranes. Parallel analyses of phospholipid levels in the isolated fractions revealed that loss of microsomal enzyme activity correlates with an ultimate dismantling of the membranes into their macromolecular constituents. The data have been interpreted as indicating that functionally distinct membranes or regions of the same membrane are differentially sensitive to senescence.  相似文献

1. Adrenocortical mitochondria contain filamentous or saccular internal membranes and are probably the sites of lipide elaboration or accumulation. They are numerous in the stimulated gland and scanty after hypophysectomy. There is evidence that they are expendable: light cells contain numbers of irregularly vacuolated and broken mitochondria with disorganized internal membranes. 2. Openings have been observed in some mitochondrial limiting membranes, particularly in the stimulated hamster and rat adrenal cortices, but to a lesser extent in the unstimulated glands. 3. Similarities between mitochondrial and Golgi membranes suggest a possible mode of mitochondrial formation. 4. These findings have been interrelated and discussed.  相似文献

The respiration of isolated peanut (Arachis hypogea) embryos has been studied with dry and wet embryos and mitochondria prepared after various times of imbibition. Dry seeds respire slowly, apparently via a respiratory chain which is deficient in cytochrome c. Cytochrome c-deficient mitochondria have been prepared from the embryos up to 16 hours following imbibition. These mitochondria can metabolize reduced nicotinamide adenine dinucleotide and succinate, without respiratory control by ADP, but they do phosphorylate. Added cytochrome c increases both respiration and phosphorylation of these embryonic mitochondria. When growth starts, mitochondria appear which are similar to those isolated from other mature plant tissues; they have respiratory control and can actively metabolize succinate, malate, and reduced nicotinamide adenine dinucleotide. These latter mitochondria contain a concentration of cytochrome c comparable to that found in mitochondria isolated from other mature plant tissues. It is suggested that the earliest type of mitochondria may be required to control respiration in the dry and the recently wetted embryo.  相似文献

Summary Ultrastructure of the granulosa lutein cells of the raccoon from throughout pregnancy has been described. The lutein cells often from epithelial cords which are separated by the connective tissues, capillaries and lymphatics. Based on the arrangements and modifications of the cytoplasmic organelles and inclusions, three types of lutein cells have been recognized. The type I lutein cells predominantly contain tubular, agranular endoplasmic reticulum, juxtanuclear Golgi complexes, a few round to rod-shaped mitochondria, some free ribosomes, and occasional lipid droplets. Occasionally the tubular cristae of mitochondria and tubular smooth endoplasmic reticulum appear contiguous. The type II cells contain abundant lace-like and/or stacked fenestrated endoplasmic reticulum cisternae that frequently form membranous whorls, some tubular, agranular endoplasmic reticulum, mitochondria, and lipid droplets. Mitochondria are usually small, but unusual large ones also occur. The small, rod-to round-shaped mitochondria usually have tubular cristae; but the large, oval, elongate, and cup shaped mitochondria possess tubular, lamellar, plate like, and whorl-like cristae. The plasma membranes of the cells are complexly elaborated and folded, especially when apposing each other. In favorable sections, strands of fenestrated cisternae appose the folds of the plasma membranes. In general, the amount of cytoplasmic organelles and inclusions vary greatly in the cells. The type III cells predominantly contain lipid droplets and sparse cytoplasmic organelles. The type I and II cells are found throughout pregnancy, but the type III cells are observed from mid gestation to term. The cytological features of type I and II cells suggest that they probably secrete most of the steroids, whereas the type III cells primarily store lipids.This research was supported by UPSHS grant AM-11376 and NIH contract 69-2136.  相似文献

The vascular bundle sheath cells of sugar cane contain starch-storing chloroplasts lacking grana, whereas the adjacent mesophyll cells contain chloroplasts which store very little starch and possess abundant grana. This study was undertaken to determine the ontogeny of these dimorphic chloroplasts. Proplastids in the two cell types in the meristematic region of light-grown leaves cannot be distinguished morphologically. Bundle sheath cell chloroplasts in tissue with 50% of its future chlorophyll possess grana consisting of 2-8 thylakoids/granum. Mesophyll cell chloroplasts of the same age have better developed grana and large, well structured prolamellar bodies. A few grana are still present in bundle sheath cell chloroplasts when the leaf tissue has 75% of its eventual chlorophyll, and prolamellar bodies are also found in mesophyll cell chloroplasts at this stage. The two cell layers in mature dark-grown leaves contain morphologically distinct etio-plasts. The response of these two plastids to light treatment also differs. Plastids in tissue treated with light for short periods exhibit protrusions resembling mitochondria. Plastids in bundle sheath cells of dark-grown leaves do not go through a grana-forming stage. It is concluded that the structure of the specialized chloroplasts in bundle sheath cells of sugar cane is a result of reduction, and that the development of chloroplast dimorphism is related in some way to leaf cell differentiation.  相似文献

The structural changes of mitochondria that occur during oogenesis and early embryogenesis in the rabbit have been examined with the electron microscope. Mitochondria of oogonia are both elongate and oval and contain a variable number of cristae which may or may not traverse the longitudinal axis of the organelle. When oogonia differentiate into oocytes, mitochondria become spheroidal and their cristae are sparse when compared with those found in the ellipsoidal organelles of concomitantly maturing follicle cells. As differentiation proceeds, the cristae of the mitochondria display varied configurations. For example, many display an arch-like arrangement in several regions of the organelle whereas others contain a pair of concentric membranes closely associated with limiting membrane of the mitochondrion. Mitochondria of stages from the fertilized egg to the morula possess the same internal structure as those of young oocytes. As the morula differentiates into a blastocyst there is a gradual increase in the size of the mitochondria and an increase in the number of cristae. We believe that the number and modifications of cristae indicate stages of mitochondriogenesis and the level of enzymatic activity in which this organelle is engaged during oogenesis and early embryogenesis.  相似文献

电镜观察结果表明,幼龄箭舌豌豆根瘤侵染细胞的细胞质较少,中央是一些体积较大的液泡。细胞质中侵入线经常可见,由侵入线释放出来的细菌均有细菌周膜。这些细菌只位于细胞质中,不出现在液泡里面。成熟根瘤中的侵染细胞与此不同,它们中有大量的成熟侵染细胞,细胞质丰富,里面充满大量细菌,中央常有一个大液泡。当中央液泡发育到一定程度时,位于其附近的细菌可通过液泡膜内吞、液泡膜与细菌周膜融合及液泡膜破裂3种途径进入液泡,后一种途径常伴有寄主细胞质。液泡中的细菌绝大部分裸露在外,只有个别细菌具有细菌周膜且多位于液泡膜的破损处附近,因此细菌周膜可能是原来就有的。  相似文献

Some characteristics of the mitochondria of hepatocytes and of three hepatoma cell lines have been compared. By means of stereologic analysis of electron micrographs of cross-sections through cells the volume of mitochondria per unit volume of cell cytoplasm and the surface areas of the mitochondrial envelope and cristae membranes have been measured. The relative mitochondrial volume in the cytoplasm decreases with increasing growth rate but the surface area of outer and cristae membranes per unit volume of mitochondria is not altered. The internal organization of hepatoma mitochondria, however, differs distinctly from that of normal liver mitochondria as evident from electron micrographs; the hepatoma cells contain mitochondria in which parallel cristae appear to cross the whole mitochondrial profile unlike the irregular, short cristae seen in normal liver mitochondria. Furthermore, in the fast-growing hepatoma cells the mitochondrial matrix appears less dense than in the hepatocyte. Hepatoma cells contain less organized rough endoplasmic reticulum than normal liver cells and the spatial relationship of the mitochondria to the rough cisternae, seen in the hepatocyte, is absent in the fast-growing hepatoma cell lines. It is concluded that hepatoma cells have fewer mitochondria than normal liver cells, but that the organelles have a normal content of inner membranes.  相似文献

Cerebral and branchial eyes of serpulid polychaetes have been studied by electron microscopy for the first time. In one species both eye types possess a novel ultrastructure. The receptor cells of the simple cerebral ocelli are rhabdomeric and display a close structural and functional relationship between ciliary rootlets and mitochondria. The receptors of the compound branchial eyes contain both a rhabdom and a stack of photosensitive ciliary membranes.  相似文献

Summary The fine structural changes in cotyledon cells of germinatingPelargonium seeds are studied on the first to fifth day of seedling emergence. Initially there is a rapid change in the cell fine structure, marked most conspicuously by the progressive liberation of the lipid and protein food reserves and the formation of an extensive thylakoid system within the plastids, but gradually the cells start to senesce. The subcellular changes in mutant cells are similar except that the plastids lack the usual thylakoid system and develop only deranged prolamellar bodies. They may store starch and they possess plastoglobuli, but seem not to contain plastid ribosomes. In rare mixed cells normal and mutant plastids remain quite distinct.  相似文献

There have been several reports describing paracrystalline arrays in the intermembrane space of mitochondria. On closer inspection these structures appear to be junctions of two adjoining membranes. There are two types. They can be formed between the outer and inner mitochondrial membranes (designated outer-inner membrane junctions) or between two cristal membranes (intercristal membrane junctions). In rat heart, adjoining membranes appeared associated via a central dense midline approximately 30 Å wide. In rat kidney, the junction had a ladder-like appearance with electron-dense "bridges" approximately 80 Å wide, spaced 130 Å apart, connecting the adjoining membranes. We have investigated the conditions which favor the visualization of such structures in mitochondria. Heart mitochondria isolated rapidly from fresh tissue (within 30 min of death) contain membrane junctions in approximately 10–15% of the cross sections. This would indicate that the percentage of membrane junctions in the entire mitochondrion is far greater. Mitochondria isolated from heart tissue which was stored for 1 h at 0°–4°C showed an increased number of membrane junctions, so that 80% of the mitochondrial cross sections show membrane junctions. No membrane junctions are observed in mitochondria in rapidly fixed fresh tissue or in mitochondria isolated from tissue disrupted in fixative. Thus, the visualization of junctions in the intermembrane space of mitochondria appears to be dependent upon the storage of tissue after death. Membrane junctions can also be observed in mitochondria from other stored tissues such as skeletal muscle, kidney, and interstitial cells from large and small intestine. In each case, no such junctions are observed in these tissues when they are fixed immediately after removal from the animal. It would appear that most studies in the literature in which isolated mitochondria from tissues such as heart or kidney were used were carried out on mitochondria which contained membrane junctions. The presence of such structures does not significantly affect normal mitochondrial function in terms of respiratory control and oxidative phosphorylation.  相似文献

Chromatographic analyses have indicated that aspartate and glutamate constitute from 50–70% of the total free amino acids in freshly isolated mitochondria. Radioactive tracer studies indicate that while the l-isomers of glutamate and aspartate are rapidly accumulated by mitochondria, the d-isomers of these amino acids do not penetrate the mitochondrial membrane. The action of two inhibitory compounds, 1-fluoro-2,4-dinitrobenzene (Sanger's reagent) and tannic acid, on the transport of l-glutamate and l-aspartate has been examined. A marked inhibition of l-glutamate transfer by 1-fluoro-2,4-dinitrobenzene is observed. A corresponding effect on the transport of either l-aspartate or the anionic substrate, succinate has not been found. Tannic acid, an agent previously known to inhibit certain carrier-mediated solute fluxes in mitochondria, is shown also to inhibit the uptake of both l-glutamate and l-aspartate. These findings are consistent with the view that the mitochondrial membranes of rat liver cells contain distinct, stereospecific transport mechanisms for aspartate and glutamate.  相似文献

The rapid freezing and vacuum dehydration of tissue has been employed to study the intracellular distribution of diffusible substances at the electron microscopic level. Hower, the ultrastructural detail of freeze-dried tissue is difficult to retain. Consequently, the ultrastructural preservation of freeze-dried smooth muscle has not been sufficient to permit satisfactorily definition of intracellular organelles. Therefore, determinations of the intracellular distribution of soluble ions have not been achieved in freeze-dried smooth muscle. In this study a freeze drying method is presented which provides more satisfactory definition of intracellular organelles than has been available in the past. Using this merens have been preserved. When these muscles are compared to convention preparer of surface vesicles that are observed, and the mitochondria are extremely electron opaque in the freeze-dried tissue. The smooth muscles which have been examined do not have the inherent contrast of other types of tissue nor do they contain the different types of mitochondria that have been observed in nonmuscle tissue.  相似文献

The effect of endogenous cytokinins on the pattern of palisade cell division post-germination does not depend on the conditions of cotyledon development -in planta (attached to seedlings) or in vitro (isolated from dry zucchini seeds and cultured on water). In cotyledons originating from 4-day-old seedlings (experimental system 1), exogenous cytokinin temporarily (in the first 2 day of cultivation) enhanced post-mitotic cell enlargement of palisade cells, mainly due to enhanced water uptake and use of cell storage compounds, all of which lead to cotyledon senescence. Cytokinin is not able to resume the completed palisade cell division on day 5. As a result, the number of cells and the final areas of treated and control cotyledons are quite similar. By contrast, the effects of cytokinin on cotyledons isolated from dry seeds (experimental system 2) are better expressed, promoting an increase in number of palisade cells accompanied by additional cotyledon area enlargement. However, the prolonged post-mitotic cell expansion in control cotyledons compensates for the reduced speed of cell growth and division activity and decreases differences in final cotyledon area between treatments. The results define cell division as the primary target of cytokinin stimulation in cotyledon tissues competent for division, and determine the temporal patterns of palisade cell cycling related to cotyledon age. This knowledge permits a better choice of experimental system to study effects on cell proliferation and cell growth, as well as cell enlargement and senescence-related events using physiologically homogeneous material.  相似文献

Summary Cortical cells in the apical region of excised radicles from cotton (Gossypium hirsutum L. var. M-8) seeds of differing moisture content (6, 12, or 16%) were examined in thin sections of vapor-fixed tissue and in freeze-etched replicas of unfixed tissue with transmission electron microscopy (TEM). The structure of the plasma membrane, as revealed in chemically-fixed tissue, had the tripartite feature characteristic of unit membranes. Regions of the plasma membrane appearing as discontinuities or breaks were shown by stereo tilting to represent an optical artifact caused by image superposition of curved regions of the plasma membrane contained within the thickness of a thin section. In freeze-etched preparations two complementary-type images of the plasma membrane were consistently observed. The asymmetric distribution of membrane-associated particles (MAPS) in the P and E fracture faces suggests that the structure of plasma membranes in cells of dry seeds resembles that of plasma membranes in germinating seeds. The physiological significance of these findings is that leaching of ions and small molecules during seed imbibition does not appear to involve passage through disorganized membranes in the radicle.  相似文献

Spermatogonia and both generations of spermatocytes of Tenebrio molitor possess conventional bipolar spindles with only few aster MTs. Spindles in metaphase spermatogonia are surrounded by fenestrated two-layered cisternae and do not contain intraspindle membranes. In metaphase spermatocytes, a spindle envelope is missing, but intraspindle membranes are abundant. Mitochondria form long threads lateral to the nucleus in prophase I of meiosis. The elongated mitochondria also align parallel to the spindle apparatus in prometaphase I. As a consequence, the spindles reside in a cage formed of mitochondria. This arrangement may guarantee proper bisection of the chondriome during division. Cells are tightly packed during spermatogonial divisions and in prophase I, but large intercellular spaces develop when the first meiotic spindle assembles. Then, cytoplasmic bridges which persist between the cells as a result of incomplete cytokinesis appear as slender tubes. Anti-tubulin immunofluorescence using an antibody against acetylated α-tubulin revealed intense acetylation throughout spermatogonial mitosis but a low degree of α-tubulin acetylation in meiotic spindles prior to telophase. This may indicate a high microtubule turnover in meiosis.  相似文献

The fine structure of the prothoracic glands of Spodoptera littoralis was investigated during the first half of the last larval instar. The secretory cells have two types of mitochondria, micromitochondria and macromitochondria. The micromitochondria have rounded to elongated profiles and sometimes branch. They contain lamellar, tubular and also tubulo-vesicular cristae. The macromitochondria appear generally rounded or oval and possess tubular cristae. Many regular parallel membranes appear within macromitochondria. Favorable sections show tubular structures packed in honeycomb fashion. The mitochondrial cristae are in connection with the tubular structures. Honeycomb and parallel membranes increase in number as the size of the macromitochondria increases.  相似文献
