Summary Cell-free translation of salivary gland RNA or of purified Balbiani ring RNA (75S) in a reticulocyte lysate system gives rise to high molecular weight translational products (HMTP). In addition to a common size (approx. 1×106 daltons) HMTP share imunogenic determinants with the giant secretory proteins of salivary glands. This suggests that HMTP correspond to in vivo secreted proteins and thus, corroborates the notion that 75S-RNA is the messenger for these proteins. The time course of HMTP synthesis and the lack of appearance of lower molecular weight components as translational products of 75S-RNA indicate that the synthesis of HMTP (and of secretory proteins) occurs in one piece by an uninterrupted process. HMTP are regarded the largest polypeptides so far synthesized in a cell-free system.  相似文献

In the yeast Cryptococcus albidus, the utilization of xylan as compared to xylose requires at least an inducible endoxylanase enzyme, secreted in the culture medium. The endoxylanase induction was monitored by immunoprecipitation of in vivo and in vitro synthesized products. The mature endoxylanase is a highly glycosylated enzyme with an apparent molecular weight of 48000. Upon chemical deglycosylation with trifluoromethanesulfonic acid, the molecular weight was reduced to 40000. Addition of tunicamycin to the culture medium resulted in the synthesis of a modified polypeptide having a molecular weight of 40000. Poly(A)-containing RNA isolated from the yeast was translated in the rabbit reticulocyte protein-synthesizing system. The appearance of a translatable xylanase mRNA was observed in xylan-grown cells but not in xylose-grown cells. The polypeptide identified as xylanase had a molecular weight of 44000. This suggests that the xylanase is synthesized as a precursor, containing a peptide signal sequence of 35 residues.  相似文献

An mRNA-dependent reticulocyte cell-free protein synthesizing system very efficient in the translation of myosin heavy-chain mRNA from a rat myogenic cell line is described. This system exhibits a high degree of fidelity with regard to the spectrum and relative proportion of the different proteins synthesized from a sample of cytoplasmic RNA as compared to the proteins synthesized in vivo by the cells from which the RNA is prepared. The main feature of this system is the use of a K+ and Cl- concentration similar to those of the reticulocyte cytoplasm. Using this system, myosin heavy chain, identified by low-salt precipitation, electrophoretic mobility, and partial peptide analysis, represents 17% of the total protein synthesis when cytoplasmic RNA from well-fused L6E9 cells is used. Furthermore, when RNA preparations from growing myoblasts, that when analyzed in other cell-free translational systems seem not to contain any myosin heavy-chain mRNA, are tested in the system reported here, they are proven to contain high amounts of translatable myosin heavy-chain mRNA.  相似文献

Southern bean mosaic virus RNA was translated in wheat embryo extracts and in nuclease-treated rabbit reticulocyte lysates. Four principal products were synthesized: two related products with molecular weights of 105,000 and 75,000, a product with a molecular weight of 29,000 that closely resembled coat protein, and a product with a molecular weight of 14,000. Their proportion depended on ionic conditions and the translational system used.  相似文献
