Treatment of Trollius ledebouri seeds with gibberellins A4+A7 promotes germination. The efficacy of the treatment is dependent upon the duration of imbibition in distilled water prior to GA4+7 application. Presoaking increases both the final percentage germination attained and also its rate of achievement. No presoaking effect is exhibited by seeds induced to germinate by testa removal in the absence of GA4+7. Active washing of Trollius seeds enhances the presoaking effect and the eluent from washed seeds is inhibitory to germination. The results support the hypothesis that the presoaking effect exhibited by Trollius is the result of the leaching of a germination inhibitor from the seeds which is antagonistic to GA4+7. Additionally, treatment of Trollius seeds with the gibberellin-biosynthesis inhibitor (2-chloroethyl)-trimethylammonium chloride (CCC) prior to testa removal retards germination. The inhibitory effect of CCC on germination is overcome by GA4+7. Although CCC inhibits embryo growth during the presoaking of intact seeds, it does not affect the increased sensitivity of presoaked seeds to GA4+7. Therefore, although endogenous gibberellins may be involved in the germination process, they do not contribute to the presoaking phenomenon. The expansion of isolated endosperm tissue is not affected by CCC. However, the chemical markedly inhibits endosperm expansion in intact seeds and implicates the embryo as both the site of production of the germination inhibitor and of gibberellin. Using lettuce (Lactuca sativa L., cv. Grand Rapids) embryos in osmotica, we have demonstrated that when the growth rates of the embryonic axes of seeds treated with red (R) or far-red (FR) light are equalized, the axes of R-treated seeds develop a 3.4-bar decrease in water potential (paper No. III).As axial growth begins, reserve protein and phytin decrease rapidly, concomitant with increases in reducing sugars, -amino nitrogen, and inorganic and esterified soluble phosphates. However, no differences between the axes of R-and FR-treated seeds are found with respect to the changes in these compounds, indicating that these changes arise as a result of growth and are not under immediate phytochrome control. Little change in the total lipid content is found in either treatment. The axes of FR-treated seeds hydrolyze endogenous sucrose at a greater rate thant those of R-treated seeds. Axes of R-treated seeds accumulate K+ and Na+ to a greater extent than those of FR-treated seeds. When potassium salts are added to the incubation medium, R induces increased K+ uptake by the axis and greater medium acidification by the axis. Malate and other organic acids and acidic amino acids increase at equal rates in both treatments, indicating that inorganic anions may also be taken up to balance the ionic charges. The results are compatible with the assumption that changes in the osmotic and pressure potentials of the embryonic axes of R-treated seeds are the result of a phytochrome-stimulated proton pump which, in whole dormant seeds, would initiate water-potential changes allowing the embryos to overcome the mechanical restraint of the surrounding seed layers, resulting in germination.Abbreviations FR far-red light - PEG polyethylene glyeol 4000 - Pfr far-red-absorbing form of phytochrome - R red light III=Carpita et al. 1979  相似文献

The germination of seeds of tomato [Lycopersicon esculentum (L.) Mill.] cv. Moneymaker has been compared with that of seeds of the gibberellin-deficient dwarf-mutant line ga-1, induced in the same genetic background. Germination of tomato seeds was absolutely dependent on the presence of either endogenous or exogenous gibberellins (GAs). Gibberellin A4+7 was 1000-fold more active than commercial gibberellic acid in inducing germination of the ga-1 seeds. Red light, a preincubation at 2°C, and ethylene did not stimulate germination of ga-1 seeds in the absence of GA4+7; however, fusicoccin did stimulate germination independently. Removal of the endosperm and testa layers opposite the radicle tip caused germination of ga-1 seeds in water. The seedlings and plants that develop from the detipped ga-1 seeds exhibited the extreme dwarfy phenotype that is normal to this genotype. Measurements of the mechanical resistance of the surrounding layers showed that the major action of GAs was directed to the weakening of the endosperm cells around the radicle tip. In wild-type seeds this weakening occurred in water before radicle protrusion. In ga-1 seeds a similar event was dependent on GA4+7, while fusicoccin also had some activity. Simultaneous incubation of de-embryonated endosperms and isolated axes showed that wild-type embryos contain and endosperm-weakening factor that is absent in ga-1 axes and is probably a GA. Gibberellin (GA) metabolism from GA12-aldehyde was studied in cell-free systems from 2-d-old germinating embryos of barley. [14C]- or [17-2H2]Gibberellins were used as substrates and all products were identified by combined gas chromatography-mass spectrometry. Stepwise analysis demonstrated the conversion of GA12-aldehyde via the 13-deoxy pathway to GA51 and via the 13-hydroxylation pathway to GA29, GA1 and GA8. In addition, GA3 was formed from GA20 via GA5. We conclude that the embryo is capable of producing gibberellins that can induce -amylase production in the aleurone layer. There was no evidence for 12- or 18-hydroxylation and GA4 was neither synthesised nor metabolised by the system. In a carrot (Daucus carota L.) cell line lacking the ability to undergo somatic embryogenasis, and in carrot and anise (Pimpinella anisum L.) cell lines in which embryogenesis could be regulated by presence or absence of 2,4-dichlorophen-oxyacetic acid (2,4-D), in the medium (+2,4-D=no embryogenesis,-2,4-D=embryo differentiation and development), the levels of endogenous gibberellin(s) (GA) were determined by the dwarfrice bioassay, and the metabolism of [3H]GA1 was followed. Embryos harvested after 14 d of subculture in-2,4-D had low levels (0.2–0.3 g g-1 dry weight) of polar GA (e.g. GA1-like), but much (3–22 times) higher levels of less-polar GA (GA4/7-like); GA1, GA4 and GA7 are native to these cultures. Conversely, the undifferentiated cells in a non-embryogenic strain, and proembryos of an embryogenic strain (+2,4-D) showed very high levels of polar GA (2.9–4.4 g g-1), and somewhat reduced levels of less-polar GA. Cultures of anise undergoing somatic embryo development (-2,4-D) metabolized [3H]GA1 very quickly, whereas proembryo cultures of anise (+2,4-D) metabolized [3H]GA1 slowly. The major metabolites of [3H]GA1 in anise were tentatively identified as GA8-glucoside (24%), GA8 (15%), GA1-glucoside (8%) and the 1(10)GA1-counterpart (2%). Thus, high levels of a GA1-like substance and a reduced ability to metabolize GA1 are correlated with the absence of embryo development, while lowered levels of GA1-like substance and a rapid metabolism of GA1 into GA8 and GA-conjugates are correlated with continued embryo development. Exogenous application of GA3 is known to reduce somatic embryogenesis in carrot cultures; GA4 was found to have the same effect in anise cultures. A gibberellin (GA) C-20 hydroxylase that catalyses the conversion of GA53 to GA44 was purified from developing pea embryos by ammonium-sulfate precipitation, gel filtration and anion-exchange column chromatography. The purification was about 270-fold and 15% of the enzymic activity was recovered. The relative molecular mass was 44000 by Sephadex G-200 gel filtration. The apparent Michaelis constant was 0.7 M and the isoelectric point was 5.6–5.9. The enzymic activity was optimal at pH 7.0 2-Oxoglutarate and ascorbate were required for activity. Low concentrations of Fe2+ stimulated the reaction, but externally added Fe2+ was not essential, even in the most purified preparation. Catalase and bovine serum albumin also stimulated. Dithiothreitol preserved the activity during purification but was not needed during incubation. In fact, the simultaneous presence of dithiothreitol and Fe2+ in the incubation mixture was inhibitory to the purified enzyme. The cofactor requirements are typical for those of 2-oxoglutarate-dependent dioxygenases.When the incubation time was long enough, GA53 was converted to both GA44 and GA19. The proportions of these two products remained constant throughout the purification, but this does not necessarily mean that their formations is catalysed by a single enzyme. Sodium dodecyl sulfatepolyacrylamide gel electrophoresis showed that the final preparation contained several proteins. The influence of several plant growth regulators on the growth of the embryonic axes from red- and far-red-(R- and FR-)treated lettuce (Lactuca sativa L., cv. Grand Rapids) seeds was examined; as shown previously, the water potential of the axes from R-treated seeds has been lowered by 3.5–5.6 bars compared to that in axes from FR-treated ones. Kinetin and abscisic acid (ABA), when included in the incubation medium, reduced the elongation of the axes whereas fusicoccin stimulated it; however, these effects were the same in axes of both R- and FR-treated seeds. In contrast, elongation of axes from FR-treated seeds was stimulated by gibberellic acid (GA3, but elongation of axes from R-treated ones was not affected by this hormone. The endogenous levels of abscisic acid (ABA), cytokinins (CKs) and gibberellins (GA1/GA3 combined) in Leucospermum glabrum embryos were monitored in axes and cotyledons separately during normal germination. Plant growth substance changes were correlated with known morphological, structural and ultrastructural events in the embryo of Proteaceae. The effect of exogenous application of 6-benzyladenine (BA) and GA4+7 under three known dormancy-enforcing environmental conditions were studied in L. glabrum and L. cordifolium. The endogenous levels of the hormone classes GAs and CKs changed phasically during normal germination under a single alternating temperature regime. GA1/GA3 levels increased in cotyledons within 3 d of hydration while at the same time initial CK levels decreased. Following this transient peak GAs fell to a low level throughout the germinative period. Subsequently the CKs, Z and ZR, and to a lesser extent their dihydro-derivatives, appeared in both the axes and the cotyledons as fluctuating, transient peaks. Early increases in GAs are thought to control the induction of the germination process. The CK pattern suggests that CKs control at least three major processes of germination sensu stricto following induction: 1) early mobilization of protein and lipid reserves in the axis and later in cotyledons, 2) cotyledon expansion which causes the endotesta to split permitting radicle protrusion and 3) later, radicle growth.Our results indicate that dormancy in intact Leucospermum seeds is enforced by embryo anoxia, regulated by the impermeable exotesta. In addition synthesis of or tissue sensitizing to both hormone classes GAs and CKs depends on moderately low temperature as the primary environmental requirement. For GA synthesis a secondary, daily pulse of high temperature is required. The physiological and biochemical consequences of treating Le (tall) and le (dwarf) pea seedlings with varying quantities of the gibberellins [3H]GA20 and GA1 have been investigated. Although the percentage uptake of these compounds from the site of application on the 3 stipules was low and most of the applied GA remained unmetabolised in situ, the quantitative relationship between GA translocation and GA dosage was found to be linear for GA1 but saturating for GA20. The movement of the GAs and their subsequently produced metabolites was mainly acropetal. They accumulated in greatest quantity in the apical extremities of the shoot. Overall, the extent to which GA20 was metabolished in le seedlings was considerably less than in Le pea seedlings. Although all le tissues contained significantly less [3H]GA1 than their Le counterparts, phenotypic effects of the le mutation were apparent only on internode and tendril development. Increased tissue growth, consequent upon GA treatment, was also apparent only in the internodes and tendrils of le plants. For internodes, GA1 content determined the mid-logarithmic-phase growth rate and, consequently, final length. The gibberellin (GA) 2-hydroxylases in mature and immature seeds of Pisum sativum have been partially purified and characterised. The enzymes are unstable when stored below pH 7.0 or in the absence of a thiol reagent. The optimum assay pH is between 7.4 and 7.8 and activity is dependent upon the presence of -ketoglutarate, Fe2+ and ascorbate. The 2-hydroxylase activities for GA1, GA4, GA9 and GA20 are chromatographically inseparable and correspond to a protein of Mr 44000. The rate of GA 2-hydroxylation varies according to substrate and some evidence indicates that the 2-hydroxylase activities for GA1 and GA4 and for GA9 and GA20 may reside in different proteins. During pea seed maturation, the specific activity of the enzyme(s) increases dramatically and reaches a maximum at a time when endogenous GA9, GA20, GA29 and GA51 are also at their greatest concentration. This correlation is not the result of substrate induction of enzyme activity. Since the GA 2-hydroxylases operate at maximal rate at low substrate concentrations they are incapable of rapidly 2-hydroxylating excessive quantities of (exogenously applied) GA1 or GA20. 为探明华重楼离体胚培养及植株再生的基本体系,该文以华重楼离体幼胚为试验材料,以MS培养基为基本培养基,研究不同光照、不同浓度梯度组合的植物生长调节剂对华重楼离体幼胚萌发、成苗的影响。结果表明:培养到60 d时,暗培养条件下华重楼离体幼胚的生长率和萌发率分别比光培养条件下高45.25%、19.17%,故暗培养比光培养更有利于华重楼离体幼胚生长发育。当GA3浓度相同时,离体幼胚萌发所需时间随IAA浓度增加而延长。不同浓度的GA3都可促进离体幼胚萌发,促进作用由强到弱依次为5 mg·L-1 GA3>1 mg·L-1 GA3> 10 mg·L-1 GA3。在黑暗条件下,优选得到最适华重楼离体幼胚生长发育配方为1/2 MS+30 g·L-1 蔗糖+7 g·L-1琼脂+0.5 g·L-1 活性炭+5 mg·L-1GA3+1 mg·L-1 IAA。此配方可诱导华重楼离体幼胚在2个月左右萌发,萌发率达50%需80 d左右。在华重楼成熟胚出苗培养时发现,高浓度的植物生长调节剂会抑制华重楼成熟胚生长发育,高浓度GA3甚至会导致华重楼成熟胚死亡。优选得到最适华重楼成熟胚出苗配方为MS+30 g·L-1 蔗糖+6.2 mg·L-1 硼酸+7 g·L-1 琼脂+0.7 g·L-1 活性炭+0.5 mg·L-1 2,4-D+1.5 mg·L-1 IAA + 1.5 mg·L-1 ZT+5mg·L-1 GA3,诱导成熟胚出苗需43 d左右,75 d左右可形成真叶。  相似文献

Grand Rapids lettuce (Lactuca sativa L.) seeds were stored in an imbibed state for up to two years. Embryos dissected from stored seeds showed a progressive loss with time in their ability to germinate on polyethylene glycol (PEG) solutions. Little germination of dissected embryos from one-month imbibed seeds occurred on-6 bar PEG but only after four months of storage did the dissected embryos fail to germinate on-4 bar PEG. After two years storage 30% of dissected embryos still were able to germinate on-2 bar PEG. This loss of germination potential, which may be a symptom of the development of an embryo dormancy, could be reversed by N6-benzyladenine (BA) and red light (R) applied together or separately to dissected embryos. Two weeks of chilling of 12-month imbibed seeds restored sensitivity to R and a 48-h BA pretreatment prior to R resulted in germination rates similar to those of seeds emerging from primary dormancy. There was loss of embryo control of endo--mannanase activity after two weeks of storage even though the endosperms themselves retained their capacity for enzyme synthesis for six more weeks. Eventually, then, endo--mannanase synthesis is not possible because of inherent changes in both the embryo and endosperm, although each tissue undergoes changes at its own rate. Oxygen uptake by embryos dissected from two-month imbibed seeds did not increase to the same extent as embryos dissected from freshly imbibed seeds. In intact seeds germinating from a skotodormant state, oxygen uptake increased at a time coincident with radicle protrusion, but did not achieve the levels of uptake of those seeds germinating from a primary dormant state. Abscisic-acid (ABA) levels were determined in triticale 6A190 kernels at various stages of development from anthesis to maturity. ABA reached a maximum at ca. 22 d post-anthesis and declined rapidly 12 d later. Associated with drying of the kernel at maturity there was a rapid increase in the endogenous level of -amylase, apparently based upon de-novo synthesis. Simultaneously there were visible signs of degradation of the large starch grains in the starchy endosperm. Regulation of -amylase production in the kernel by exogenous gibberellic acid (GA3) was only evident in the almost mature kernel (30–40 d after anthesis) and then only if these kernels were first dried artificially. Furthermore, little -amylase mRNA could be detected prior to kernel maturity and water loss. Thus, the high levels of gibberellin (GA) that have been found early in kernel development in cereals do not appear to control the later production of -amylase and onset of kernel germination in the ear of triticale. However, the presence of high levels of ABA until maturity could prevent early germination and premature production of -amylase. Kernels of triticale 6A190 are characteristically shrivelled and non-dormant at maturity. Growth parameters were determined for tall (rht3) and dwarf (Rht3) seedlings of wheat (Triticum aestivum L.). Plant statures and leaf length were reduced by 50% in dwarfs but root and shoot dry weights were less affected. Leaves of dwarf seedlings had shorter epidermal cells and the numbers of cells per rank in talls and dwarfs matched the observed relationships in overall length. Talls grew at twice the rate of dwarfs (2.3 compared with 1.2 mm h-1). [3H]Gibberellin A1 ([3H]GA1) was fed to seedlings via the third leaf and metabolism was followed over 12 h. Immature leaves of tall seedlings transferred radioactivity rapidly to compounds co-chromatographing with [3H]gibberellin A8 ([3H]GA8) and a conjugate of [3H]GA8, whereas leaves of dwarf seedlings metabolised [3H]GA1 more slowly. Roots of both genotypes produced [3H]GA8-like material at similar rates. Isotopic dilution studies indicated a reduced 2-hydroxylation capacity in dwarfs, but parallel estimates of the endogenous GA pool size, obtained by radioimmunoassay, indicated a 12–15 times higher level of GA in the dwarf immature leaves. Elongation growth and gibberellin (GA9) metabolism in excised hypocotyls of lettuce (Lactuca sativa L. cv. Arctic) were investigated. Exogenously supplied GA9 stimulates elongation of hypocotyl sections and this response is intermediate between that elicited by GA1 or GA20 and GA4/7 mixture. Although uptake of radioactivity from [3H]GA9 increases with time, this gibberellin does not accumulate in the tissue but is rapidly converted to a compound with HPLC properties resembling those of [3H]GA20. After 2 h incubation in [3H]GA9, the presumptive GA20 represents 90% of the acidic ethyl acetate-soluble radioactivity in the tissue. Radioactivity is also associated with an acidic butanol-soluble fraction containing two components resolvable by HVE. The major component is similar in electrophoretic properties to a GA-glucosyl ether while the other compares to a GA-glucosyl ester. Conversion of [3H]GA9 to its [3H]GA20-like metabolite is reduced by addition of carrier GA9 or GA4/7 at concentrations as low as 1 M, while GA1, GA3 and L-proline are without effect. Formation of the GA20-like compound can be blocked by the addition of 2,2-dipyridyl, and this inhibitory effect of dipyridyl can be reversed by addition of Fe2+. At 200 M dipyridyl, elongation growth as well as [3H]GA9 metabolism are reduced by 80%. The biosynthetic steps from gibberellin A12-aldehyde (GA12-aldehyde) to C19-GAs were studied by means of a cell-free system from the embryos of immature Phaseolus vulgaris seeds. Stable-isotope-labeled GAs were used as substrates and the products were identified by gas chromatography-mass spectrometry. Gibberellin A12-aldehyde was converted to GA4 via non-hydroxylated intermediates and to GA1 via 13-hydroxylated intermediates. 13-Hydroxylation took place at the beginning of the pathway by the conversion of GA12-aldehyde to GA53-aldehyde. The conversion of GA20 to GA5 and GA6 was also shown but no 2-hydroxylating activity was found. Endogenous GAs from embryos and testas of 17-dold seeds were re-examined by gas chromatography-selected ion monitoring using stable-isotopelabeled GAs as internal standards. Gibberellins A9, A12, A15, A19, A23, A24, and A53 were identified for the first time in P. vulgaris, in addition to GA1, GA4, GA5, GA6, GA8, GA17, GA20, GA29, GA37, GA38 and GA44, which were previously known to occur in this species. The levels of all GAs, except the 2-hydroxylated ones, were greater in the embryos than in the testas. Exogenous plant growth regulators are known to increase the efficiency of interspecific and intergeneric crosses. In vitro floret culture provides a defined system for assessing the importance of various plant growth regulators on the determinants of haploid production efficiency (seed set, embryos per seeds, and plants per embryos) in Hordeum vulgare × Hordeum bulbosum crosses. The individual and combined effects of three plant growth regulators (2,4-D, GA3 and kinetin) on in vitro seed growth, embryo development and haploid production efficiency were tested in floret culture of the cross H. vulgare, cultivar Klages × H. bulbosum. All treatments, except kinetin alone, produced larger seeds and more embryos/100 seeds than the control (no plant growth regulator). 2,4-D alone was superior to GA3 alone in haploid production efficiency (70.6 vs. 51.5) as measured by the number of plants regenerated/100 florets pollinated. Although kinetin +2,4-D+GA3 produced the largest seeds and embryos, no advantage over 2,4-D alone was observed in haploid production efficiency. 2,4-D alone or kinetin +2,4-D are recommended for the purpose of barley haploid production in floret culture using the bulbosum method.Abbreviations 2,4-D 2,4-dichlorophenoxyacetic acid - GA3 gibberellic acid  相似文献

Gibberellins (GAs) control a wide range of physiological functions in plants from germination to flowering. The cellular mechanisms by which gibberellic acid (GA3) acts have been most extensively studied in the cereal aleurone. In this tissue, alterations in cellular calcium are known to be important for the primary response to GA, which is the production and secretion of hydrolytic enzymes. The extent to which cytosolic Ca2+ mediates the early events in GA action, however, is not known. In order to address this question, changes in cytosolic Ca2+ in wheat (Triticum aestivum L. cv. Inia) aleurone cells that occur rapidly after treatment with GA were characterized. In addition, GA-induced changes were compared with changes induced by three environmental stimuli that are known to modify the GA response: osmotic stress, salt (NaCl), and hypoxia. The Ca2+-sensitive dye fluo-3 was used to photometrically measure cytosolic Ca2+. It was found that GA3 induced a steady-state increase in cytosolic Ca2+ of 100–500 nM. This increase was initiated within a few minutes of treatment with GA and was fully developed after 30–90 min. The changes in cytosolic Ca2+ that were induced by GA were distinct from those induced by mannitol, NaCl, or hypoxia. Mannitol caused a steady-state decrease whereas NaCl and hypoxia both increased cytosolic Ca2+. In the case of NaCl this increase was transient but for hypoxia the increase was prolonged as long as hypoxic conditions were maintained. Gibberellin-induced changes in cytosolic Ca2+ were not induced by the inactive GA, GA8, nor did the GA-insensitive wheat mutant, D6899, respond to active GA3 with altered cytosolic Ca2+. It is concluded that changes in cytosolic Ca2+ are an early and integral part of the GA response in aleurone cells. Germinating non-dormant (ND) embryos of wild oat incorporate [3H]glycerol into phospholipid, and a 250% increase in total extractable phospholipid occurs within 72 h. During germination, leveles of phosphatidyl inositol showed the greatest change, increasing approximately 5-fold.Imbibed dormant (D) embryos of the wild oat also incorporate [3H]gycerol into phospholipids, but there is no net synthesis. A continuous turnover of membrane phospholipids could be demonstrated in pulse chase experiments, and although the proportions of most phospholipids does not change, there was a decrease of 50% in phosphatidyl serine.The half-life of [3H]glycerol in the extracted phospholipids of D and ND embryos varies between 35 and 57 h, and in membrane fractions separated on sucrose density gradients the half-lives vary between 26 and 56 h.D embryos induced to germinate with GA and ND embryos in which germination is repressed by ABA show similar phospholipid changes to ND and D embryos respectively, with the exception that the proportion of phosphatidyl serine remained unchanged in the ND-ABA embryos.It is concluded that the continual turnover of membranes of imbibed dormant embryos is consistent with the maintenance of cellular integrity determining the longevity of the seed under natural conditions.Abbreviations D dormant - ND nondormant - ABA abscisic acid - GA gibberellic acid (GA3)  相似文献
