Assimilate distribution in leaves of Lolium temulentum was establishedby root absorption of [14C]sucrose and after exposure to 14CO2.Age determined the amount of carbon assimilated, with more labelbeing incorporated during expansion than at maturity. Duringsenescence 14C assimilation was much lower. Ethanol-solubleextracts from various tissues of root-labelled plants containedmost of the radioactivity chiefly in basic and acidic compounds.The neutral fraction was composed predominantly of sucrose. Sucrose was comparably labelled in leaves from plants fed equalamounts of either [14C]sucrose, glucose, or fructose and onlytraces of labelled monosaccharides appeared in extracts. Radioactive sucrose was translocated rapidly from mature leaveswhereas, in the expanding leaf, carbon incorporation was directedtowards growth and the greater proportion of label present atligule formation was in ethanol-insoluble material. Induced senescence, of a mature leaf fed during expansion, produceda rapid loss from the pool of insoluble 14C. This was accompaniedby a reduction in the contents of chlorophyll and soluble proteinand an accumulation of amino acids. The onset of senescencecaused changes in leaf sugar levels which were correlated withincreased rates of respiration.  相似文献

Warm temperatures (35°C day/30°C night) which inhibittuberization in potato (Solanum tuberosum L., cv. Sebago) increasedgibberellin activity in crude extracts from buds, but not frommature leaves, as determined by the lettuce hypocotyl bioassay.Changes in the growth of tubers and stolons indicate the occurrenceof basipetal movement of GA3 applied to the terminal bud ora mature leaf. 14C labelling from GA3 or mevalonic acid injectedjust below the terminal bud was recovered in the lower shoot,stolons and tubers, but the amount transported was greater atcool temperatures (20/15°C). When leaves of Nicotiana tabacum L. var. Xanthi were inoculatedwith TMV, local lesion formation gave enhanced ATP concentrationaccompanied by increased respiration. Phosphorus metabolismin healthy and infected leaves was investigated either by floatingleaf tissue on 32P1-phosphate buffer or by allowing intact leavesto take up 32P1-phosphate buffer through the petioles. Bothmethods gave increased 32P1 incorporation into inorganic andorganic acid-soluble phosphorus fractions of the infected leaf.Comparison of the specific activity of 32P1-phosphate buffersupplied with the specific activity of the -phosphate groupof ATP demonstrated that 50 per cent of the ATP in the healthyleaf compared with over 78 per cent of the ATP in the infectedleaf was metabolically active in respiration. As the ATP concentrationin the infected leaf is much greater than that in the healthyleaf, this means that the amount of ATP being utilized and resynthesizedas a result of respiratory metabolism in the infected leaf ismore than twice that in the healthy leaf. Pulse-labelling experimentsdemonstrated that the rate of ATP turnover was very similarin healthy and infected leaves; therefore the increased respirationin the infected leaf results from the larger ATP pool with aturnover rate of ATP similar to that in the healthy leaf. Withtobacco callus tissue cultures infected by TMV, where infectiondoes not result in local-lesion formation, phosphorus metabolismwas unaltered in the infected tissue. It is concluded that necroticlocal-lesion formation results from increased availability ofATP at sites of ATP utilization, and this is aggravated by over-productionof ATP in the infected leaf.  相似文献

The relation between tomato leaf acid phosphatase activity andleaf tissue P content has been examined, and a study made ofthe effects of leaf development, variation in nitrogen supply,and variation in the growing medium on this relationship. Tomatoplants were grown in sand and given various concentrations ofphosphate. Plants were also grown for an initial period in peatcontaining an adequate level of phosphate, then transferredto peat to which was added 0 or 2.3 kg superphosphate m–3and supplied with either 50 of 300 µg N ml–1. Expressed on a unit tissue f. wt basis, acid phosphatase activityin the control plants in sand (given 41 µg P mlminus;1)was highest in extracts from the expanding leaves and decreasedwith leaf maturity. However, when given a reduced supply ofphosphate, the enzyme activity in the more mature leaves wasequal to, or greater than, that in the expanding leaves. Thephosphatase activity increased first in the young, fully-expandedleaves and in the mature leaves (with 4.1 µg P ml–1),but did not increase in the expanding leaves until the supplywas restricted to 2.1 µg P ml–1. On closer examination,the increase in enzyme activity appeared to be associated withthe P level in the leaf tissues, the activity increasing whenthe level fell below about 0.25 per cent (g P per 100 g drywt tissue). The same relation was found with the plants grownin peat, and was independent of the concentration of nitrogensupplied to the plants. The fully expanded leaves showed the best enzyme response whenthe phosphate supply was restricted and the activity reflectedclosely the local levels of tissue P. The assay of the enzymein unpurified leaf extracts is simple and rapid, and could beused in a test to detect P-deficiency in tomato plants. Young plants of Banksia hookeriana were grown in acid-washedsand with adequate phosphate and water supply, and a proportionwere inoculated with Phytophthora cinnamomi. There were no majordifferences in growth between uninoculated and infected plants,but there was a large increase in uptake of 32P with increasingroot disease. In healthy plants 32P uptake was greatest in youngleaf tissue, but in diseased plants labelled phosphate was directedmore towards older leaves where the activity was almost twicethat of young leaves. Enhanced uptake with disease was ascribed to possible blockageof the ‘message’ or ‘signal’ of phosphatetranslocation from shoot to root, such that the diseased rootincorrectly treated the shoot as P deficient and increased Puptake. The combined application of 10–6 M adenine and 10–6M mevalonic acid to soya bean callus accelerated its growth.Two biologically active compounds that co-chromatographed withzeatin and isopentenyl adenine were extracted from this callus.Studies with labelled adenine and mevalonic acid indicated thatthe cytokinin-dependent soya bean callus incorporated only avery small amount of the radioactive precursors into the biologically-activecompounds, making it extremely difficult to determine whetherthese compounds were synthesized de novo or whether they aroseas by-products of tRNA turnover. As cytokinins do not accumulatein rapidly-growing cytokinin-dependent soya bean callus culturedon kinetin as a source of cytokinin it seems as if biosynthesisde novo occurs when the callus is supplied with adenine andmevalonic acid. Changes in the activity of adenine and guanine salvage in nucleotideand nucleic acid synthesis during the growth of Catharanthusroseus were investigated. Incorporation of [8-14C]adenine intoATP and ADP and that of [8-14C]guanine into GTP and GDP increasedmarkedly in the lag phase of cell growth and then sharply decreased.The incorporation into RNA from both precursors showed a similarpattern. 1. Both the acid-soluble fraction and the nucleic acid fraction of wheat embryos were extensively labelled after incubation for 6hr. in the presence of [8-(14)C]adenine. Subsequent incubation in the absence of labelled adenine resulted in no loss of radioactivity to the medium during a 48hr. period. Radioautography indicated that during this period there was a continuous increase in the radioactivity present in the acid-insoluble fractions of the root and leaf tissues relative to that present in the coleorhiza and coleoptile. 2. During incubation at 25 degrees there was a 26-fold increase in the activity of 3'-nucleotidase between 4hr. and 24hr.; the activities of enzymes hydrolysing AMP and IMP increased to a smaller extent. The activities of adenine phosphoribosyltransferase and hypoxanthine phosphoribosyltransferase increased three- to five-fold during incubation at 25 degrees for 24hr. 3. Adenosine kinase, inosine phosphorylase and 5-phosphoribosyl pyrophosphate synthetase activities were high in extracts from dry embryos and did not increase during 48hr. at 25 degrees . 4. The increase in 3'-nucleotidase activity was prevented by cycloheximide, cryptopleurine or incubation at 4 degrees , but not by actinomycin D; these treatments did not depress the activity of the other enzymes measured. 5. The results are discussed in relation to RNA translocation within the wheat embryo during germination.  相似文献

Translocation of Assimilates Within and Between Potato Stems   总被引:2,自引:0,他引:2  
Three aspects of translocation in potato were examined: (i)translocation within stems (ii) translocation between individualstems of a plant (iii) translocation between tubers followinginjection of 14C sucrose into a single daughter tuber. Assimilatesexported from single leaves of evenly illuminated potato stemsremained confined to the same side of the stem as the sourceleaf in a pattern consistent with the internal arrangement ofvascular bundles in the stem, and tubers borne on stolons verticallybelow the source leaf contained higher concentrations of 14Cthan those on the opposite side. Consequently 14C import intothe tubers bore little relationship to tuber growth rates. However,alteration of source/sink relations by pruning stems to a singlesouce leaf resulted in an even distribution of 14C throughoutthe vascular bundles of the stem and 14C import into the tubersbore a stronger relationship to tuber growth rates than to thephyllotactic relationship of the tubers with the source leaf. Labelling one stem of a potato plant resulted in little or nomovement of 14C into tubers on other unlabelled stems. However,removal of the unlabelled stems at ground level induced a significantmovement of 14C from the labelled stem to the tubers on unlabelledstems, this movement occurring via the mother tuber. Shadingthe unlabelled stems had less effect than stem removal. 14C sucrose injected into single daughter tubers was translocatedto other tubers on the same stem and also to tubers on a secondstem at the opposite end of the mother tuber. The sucrose wasconverted to starch in these tubers. The results favour the view that each potato stem functionsas an independent unit with potential for assimilate redistributionwithin a stem but with little or no carbon exchange occurringbetween stems, unless under severely altered source/sink patterns. To determine the metabolic profiles of purine nucleotides and related compounds in leaves and roots of tea (Camellia sinensis), we studied the in situ metabolic fate of 10 different (14)C-labeled precursors in segments from tea seedlings. The activities of key enzymes in tea leaf extracts were also investigated. The rates of uptake of purine precursors were greater in leaf segments than in root segments. Adenine and adenosine were taken up more rapidly than other purine bases and nucleosides. Xanthosine was slowest. Some adenosine, guanosine and inosine was converted to nucleotides by adenosine kinase and inosine/guanosine kinase, but these compounds were easily hydrolyzed, and adenine, guanine and hypoxanthine were generated. These purine bases were salvaged by adenine phosphoribosyltransferase and hypoxanthine/guanine phosphoribosyltransferase. Salvage activity of adenine and adenosine was high, and they were converted exclusively to nucleotides. Inosine and hypoxanthine were salvaged to a lesser extent. In situ (14)C-tracer experiments revealed that xanthosine and xanthine were not salvaged, although xanthine phosphoribosyltransferase activity was found in tea extracts. Only some deoxyadenosine and deoxyguanosine was salvaged and utilized for DNA synthesis. However, most of these deoxynucleosides were hydrolyzed to adenine and guanine and then utilized for RNA synthesis. Purine alkaloid biosynthesis in leaves is much greater than in roots. In situ experiments indicate that adenosine, adenine, guanosine, guanine and inosine are better precursors than xanthosine, which is a direct precursor of a major pathway of caffeine biosynthesis. (rice) seedlings growing in saline conditionsexhibit pronounced gradients in leaf sodium concentration whichis always higher in the older leaves than the younger ones.Individual leaf transpiration rates have been investigated todiscover whether movement of sodium in the transpiration streamis able to explain these profiles from leaf to leaf. The useof 14C labelled ethane diol to estimate transpiration was evaluatedby direct comparison with values obtained by gas exchange measurements.Ethane diol uptake was linearly related to the transpirationalvolume flow and accurately predicted leaf to leaf gradientsin transpiration rate in saline and non-saline conditions. 14C-ethanediol and 22NaCl were used to compare the fluxes of water andsodium into different leaves. The youngest leaf showed the highesttranspiration rate but the lowest Na accumulation in salineconditions; conversely, the older leaves showed the lower transpirationrates but the greater accumulation of Na. The apparent concentrationof Na in the xylem stream was 44 times lower into the youngerleaf 4 than into the older leaf 1. Exposure to NaCl (50 molm–3) for 24 h elicited an increase in endogenous ABA inthe oldest leaf only, but no significant changes occurred inthe younger leaves. In Pisum arvense, the amides and amino-acids normally suppliedto the shoot in the transpiration stream transfer carbon toprotein largely throught the amino-acids, aspartic acid (+asparagine),glutarnic acid (+glutamine), threonine, lysine, arginine, andproline. Carbon from carbon dioxide enters the protein of photosynthesizingtissues through an essentially complementary set of amino-acidsincluding glycine, alanine, serine, valine, and the aromaticamino-acids tyrosine, phenylalnine, and histidine. Young tissuesof the shoot synthesize certain amino-acids de novo by metabolismof sugars supplied from photosynthesizing leaves. Each mature leaf on a shoot contributes carbon to current synthesisof protein at the shoot apex. Sucrose accounts for more than90 per cent of the labelled carbon leaving any age of leaf whichhas been fed with 14CO2. Upper leaves supply labelled assimilatesdirectly to the shoot apex, and the radiocarbon from these assimilatesis subsequently incorporated into a wide range of amino-acidunits of protein. The majority of the labelled assimilates exportedfrom a lower leaf move downwards to the root and nodules and,in consequence, the amino-acids and amides associated with rootmetabolism are strongly represented among the compounds eventuallylabelled in the apical region of the shoot.  相似文献

The extent of starch turnover in pepper leaves in the lighthas been estimated. After leaves were labelled with 14CO2 atconstant specific activity for 4–7 h, the irradiance wasreduced to a level which caused no net change in the starchcontent of the leaf, and the supply of 14CO2 was removed. Therewas no significant change in specific activity of starch overthe following 6–10 h, thus there was no exchange of 14C-starchwith 12C-assimilates entering the chloroplasts. Soluble sugars were extracted by low speed centrifugation fromthe apoplast of leaves of barley (Hordeum distichum L.) infiltratedwith water. Infection of the leaf with the brown rust fungus(Puccinia hordeii) resulted in a reduction in the concentrationof sucrose, glucose and fructose in the apoplast. Sugars werepresent in an apoplastic space occupying 12 and 17 cm3 m–2of leaf area in healthy and infected tissue, respectively. Uptakeof hexoses by intercellular hyphae is suggested as a cause ofthis reduction. The pH of apoplastic sap extracted from rust-infectedleaves was increased to pH 7·3 from pH 6·6 incontrols. The effect of a reduced apoplastic sugar pool andincreased pH on export from infected leaves is discussed. A technique, using leaf disks, has been developed to study thepenetration of isotopically labelled compounds into leaves underconditions where there is no appreciable change in the concentrationof the external solution and no subsequent translocation. Inthis preliminary survey, the leaves of Phaseolus vulgaris andColeus Blumei were employed to investigate the entry of 2,4-dichlorophenoxyaceticacid (2,4-D), labelled in the carboxyl group with 14C. Over3 days there is no loss of 14C to the atmosphere from treatedleaves of Phaseolus. The rate of penetration is enhanced when(a) the leaves are young, (b) the water status is lowered, (c)the temperature is raised (Q10=2.3–2.8), and (d) a surface-activeagent is added to the external solution. Penetration is alsofavoured by a decrease in the pH, the relation indicating thatboth ions and molecules enter. Penetration is greater in thelight and prior illumination of the tissues positively affectsthe subsequent rate in the light, but not in the dark. In boththe light and the dark considerably more 2,4-D penetrates theabaxial surface of Phaseolus leaves. For Coleus an even greaterdifference between surfaces is found in the light but not inthe dark. For both species in the light the rates of entry intoboth surfaces are proportional to their respective stomataldensities. The simultaneous addition of indoleacetic acid tothe external solution caused more 2,4-D to enter Phaseolus leaves,but the addition of triiodobenzoic acid restricts entry. Therate of penetration remains constant over 24 hours and between0.1 and 200 mg./l. the rate is linearly related to concentration.Subsequent to entry, the 2,4-D is in a form which does not diffusefrom the tissue into buffer or exchange with unlabelled 2,4-D.Moreover, no outward movement takes place from treated tissuewhich has been frozen and thawed. These findings are discussedin relation to previous work on foliar penetration. It is concludedthat at least with Phaseolus penetration largely takes placethrough the guard cells and/or accessory cells.  相似文献

Leaf, stem, and root extracts of near-isogenic tomato plantscv. Craigella, resistant and susceptible to Verticillium albo-atrum,showed constitutive 1,3-ß-glucanase activity whichincreased following inoculation with the pathogen. Partiallypurified enzyme extracts were obtained by dialysing a 30–80%ammonium sulphate fraction of the tissue brei. The enzyme hadpH and temperature optima of 5?5 and 44 ?C respectively, withhigh activity between 50 and 60 ?C. The response to laminarinconcentration was linear between 1?2 and 7?5 mg ml–1.Root inoculation of susceptible plants with 106 propagules ml–1V. albo-atrum led to a umform 300 per cent increase in all steminternodes except the terminal one, which was 500 per cent ofthe controls. No spatial relationship of enzyme activity tothe localization of fungus within the stem was apparent. Petioles,leaves, and roots of susceptible infected plants similarly showedan increase in activity but less than that in stems. Changedlevels of stern enzyme activity at different times after inoculationwere associated with reductions in the number of vessels containinghyphae. Extracts of plants of the resistant isoline showed increasedglucanase activity over controls, but this was substantiallylower than that in susceptible plants and was associated withthe greatly reduced mycelial colonization in resistant plants. It is concluded that single gene resistance in tomato to Verticilliumis not associated with innately higher levels of 1,3-ß-glucanasein healthy plants. The increased activity in infected plantsis proportional to the overall quantity of pathogen in the plantor of pathogenic metabolites.  相似文献

The metabolism of [8-14C]adenine and [8-14C]hypoxanthine infour species of Camellia plants was investigated in relationto the synthesis of purine alkaloids, caffeine and theobromine.Young leaves of C. sinensis had the ability to synthesize caffeine,but in C. irrawadiensis, these labelled precursors were incorporatedinto theobromine, not caffeine. No synthesis of purine alkaloidscould be detected in C. japonica and C. sasanqua leaves. In both reproductive and vegetative plants of Lolium temulentumL., the export of 14C-labelled assimilates from each healthyleaf on the main shoot to terminal meristem, stem, tillers,and roots was measured each time a new leaf was expanded, fora period of 5 to 6 weeks. Some labelled assimilates moved fromeach leaf on the main shoot to every meristem in the same shoot,as well as to the tops and roots of adjacent organically attachedtillers. The terminal meristem of the reproductive shoot, which includedthe developing inflorescence, received 70–80 per centof the carbon assimilated by the emerged portion of the growingleaf, 15–25 per cent of the carbon assimilated by thetwo youngest expanded leaves, and 5–10 per cent of thatfrom each of the older leaves. A similar pattern of carbon supplyto the terminal meristem was found in vegetative shoots, exceptthat older leaves on young vegetative shoots supplied even lessof their carbon to the terminal meristem. The general conclusionis that developing leaves at the tip of the shoot receive aboutthe same proportion of carbon from each leaf as does a developinginflorescence. Young expanded leaves provided most labelled assimilates forstem growth; during both reproductive and vegetative growth,expanded leaves increased their export of labelled carbon tostem, and exported less of their 14C to roots and sometimesto tillers. In these reproductive and vegetative shoots, grown in a constantexternal environment, the major changes in the pattern of distributionof labelled assimilates appeared to be the result of increasedmeristematic activity in stem internodes; the development ofan inflorescence had no obvious direct effect on the carboneconomy of shoots.  相似文献

To examine 14CO2 fixation, potential translocation, and carbonflow among leaf chemical fractions of young developing leaves,the shoot tip of 24-leaf cottonwood (Populus deltoides Bartr.ex. Marsh) plants were cut off under water, placed in artificialxylem sap, and treated with 14CO2 in continuous and pulse-chaseexperiments. Additional leaves on whole plants were spot treatedon the lamina tip to follow export from the tip only. The analysedleaves ranged in age from leaf plastochron index(LPI) –5to 3, the spot treated leaves from LPI 2 to 5. After 30 minfixation, the specific activity in the lamina tip increasedlinearly with leaf age from LPI –5 to 1 (0.5 to 4.5 kBqmg–1). Specific activity in the lower lamina increasedslowly with leaf age and did not reach 500 kBq mg–1 untilLPI –1. Total 14CO2 fixed in the lower lamina exceededthat fixed in the tip by LPI –2 because of the large amountof tissue present in the lower lamina. Although the lamina tipfixed high levels of 14CO2, pulse-chase studies coupled withautoradiography indicated no vein loading or translocation fromthe tip until about LPI 4 or 5. The 14C fixed in both tip andlower lamina was incorporated at the site of fixation and wasnot distributed to younger tissue or translocated from the lamina.Although the percentage distribution (14C in each chemical fractioncompared with the total in all fractions) of 14C among certainchemical fractions, e.g. sugars, amino acids and proteins, indicatedthat the mesophyll of the tip was more mature than the lowerlamina, physiologically both leaf sectors were immature basedon the expected 14C distribution in mature tissue. Informationfrom this and other studies indicates that the extreme tip ofa developing cottonwood leaf first begins to export photosynthateabout LPI 4 or 5 on a 24-leaf plant. The first photosynthatetranslocated may be incorporated into the vascular tissues andmesophyll directly below the tip. However, as the tip continuesto mature photosynthate is translocated past the immature lowerlamina into the petiole and out of the leaf. Populus deltoides Bartr. ex. Adenine phosphoribosyltransferase activity was measured in extracts of soybean (Glycine max var. Acme) callus and of senescing barley leaves (Hordeum distichon c.v. Prior). The enzyme from soybean callus had Michaelis constants for adenine and 5-phosphoribosyl pyrophosphate of 1.5 and 7.5 μm respectively and was inhibited by AMP and stimulated by ATP. The presence of kinetin was found to considerably increase the activity of adenine phosphoribosyltransferase in extracts of soybean callus and senescing barley leaves.  相似文献
