Several genes can alter the yellowing processes that normallyoccur during pod development and monocarpic senescence in soybean.CytG and d1 + d2 cause the leaves and seeds to stay green atmaturity. G blocks yellowing of the seed coat but not the leavesor embryos. By contrast, another gene, y3, causes earlier yellowingof the leaves. This paper examines the effects of these geneson photosynthesis and some related parameters of the senescencesyndrome in near-isogenic backgrounds (cv. Clark). Neither cytGnor d1 d2 delays the decline in photosynthetic rate during monocarpicsenescence relative to Clark; while Gd1 d2 does. Again, y3 causesan earlier decrease. Similarly, neither cytG nor d1 d2 altersthe decline in stomatal conductance and transpiration, whichoccurs at the end of podfill; however, Gd1 d2 delays it andy3 advances it. Neither cytG nor d1 d2 prevents the rise inintercellular CO2 during podfill, while Gd1 d2 does, and y3promotesit. These changes in intercellular CO2 may reciprocally reflectthe status (maintenance for Gd1 d2 and early loss for y3) ofthe photosynthetic enzymes. Gd1 d2, d1 d2 and cytG do not appreciablyaffect leaf blade abscission; however, y3 prevents it. Gd1 d2increases the dry weight seed yield, and y3 decreases it. Gd1d2 inhibits several components in the senescence process, whilecytG seems to affect mainly chlorophyll loss and y3 may actmore broadly than just accelerating senescence. Mistletoes usually have slower rates of photosynthesis thantheir hosts. This study examines CO2assimilation, chlorophyllfluorescence and the chlorophyll content of temperate host–parasitepairs (nine hosts parasitized by Ileostylus micranthus and Carpodetusserratus parasitized by Tupeia antarctica). The hosts of I.micranthus had higher mean annual CO2assimilation (3.59 ±0.41 µmol m-2 s-1) than I. micranthus(2.42 ± 0.20µmol m-2 s-1), and C. serratus(2.41 ± 0.43 µmolm-2 s-1) showed higher CO2assimilation than T. antarctica(0.67± 0.64 µmol m-2 s-1). Hosts saturated at significantlyhigher electron transport rates (ETR) and light levels thanmistletoes. The positive relationship between CO2assimilationand electron transport suggests that the lower CO2assimilationrates in mistletoes are a consequence of lower electron transportrates. When photosynthetic rates, ETR and chlorophyll a /b ratioswere adjusted for photosynthetically active radiation, hostsdid not have significantly higher CO2assimilation (3.21 ±0.37 µmol m-2 s-1) than mistletoes (2.54 ± 0.41µmol m-2 s-1), but still had significantly higher ETRand chlorophyll a / b ratios. The electron transport rates,saturating light and chlorophyll a / b ratios of sun leavesfrom mistletoes were similar to host shade leaves. These responsesindicate that in comparison with their hosts, mistletoe leaveshave the photosynthetic characteristics of the leaves of shadeplants. Photosynthesis by developing embryos of oilseed rape (Brassica napus L.)   总被引:1,自引:0,他引:1  
The aim of this study was to assess the photosynthetic potentialof developing seeds of oilseed rape (Brassica napus L.) andto compare photosynthetic properties of embryo plastids withthose of leaf chloroplasts from the same species. Measurementsof CO2-dependent O2 evolution show that developing seeds ofB. napus are photosynthetically active in vitro. Essentially,all of the photosynthetic activity of the developing seed isaccounted for by the embryo. The rate of photosynthesis by developingembryos increased until the onset of desiccation, after whichit declined, so that by maturity embryos were no longer photosyntheticallyactive. Photosynthetic activity was positively correlated withchlorophyll content throughout development. Comparison of thephotosynthetic characteristics of leaf and embryo chloroplastsrevealed that rates of uncoupled electron transport were 2.5-foldgreater in those from the embryo. Light-saturated rates of CO2-dependentO2 evolution, per unit chlorophyll, and CO2 saturation pointswere similar for chloroplasts from both tissues. However, light-saturationpoints and chlorophyll a/b ratios were lower for embryo thanfor leaf choroplasts. Embryos and embryo chloroplasts also containedconsiderably less ribulose 1,5-bisphosphate carboxylase/oxygenaseprotein per unit total protein, than leaves. Although excisedembryos were capable of high rates of CO2-dependent O2 evolution(90–100 mol mg–1 chlorophyll h–1) under asaturating photosynthetic photon flux density (PPFD), low transmittanceof light through the silique wall (30%), together with the highPPFD required to achieve light compensation points in developingseeds (500 mol m–2 s–1), suggests that photosynthesisin vivo is unlikely to make a net contribution to carbon economyunder normal environmental conditions. Trends in several photosynthetic parameters and their responseto changed growth light were followed for 15 d in leaves ofyoung birch saplings using a rapid-response gas exchange measuringequipment. These in vivo measurements were compared to biochemicalassays that were made from the same leaves after the gas exchangestudies. The measurements were made on leaves that were selectedprior to the study and were at that time of similar age. Forthe first 7 d the photosynthetic parameters were followed fromthe growth conditions of moderate light (200 µmol m–2s–1; referred to as controls later in the text). On day7 some of the saplings were transferred to grow either underhigh (450 µmol m–2 s–1; referred to as highlight plants) or low (75 µmol m–2 s–1; referredto as low light plants) light and the capability of the preselectedleaves for acclimation was followed for 6 d. For comparison,at the end of the experiment the measurements were made on bothcontrols and on young leaves that had developed under high andlow light. Generally the in vivo measured rate of CO2 uptake (gross photosynthesis)both at 310 ppm CO2 and 2000 ppm CO2 corresponded very wellto the biochemically determined CO2 fixation capacity in vitroafter rapid extraction (measured as the initial and total activityof Rubisco, respectively). However, if the flux of CO2 intothe chloroplasts was limited by the closure of the stomata,as was the case of the high light plants, then the in vitromeasured Rubisco activity was greater than the in vivo measuredCO2 uptake. Vmax, calculated from the mesophyll conductanceat 1% O2, exceeded the initial activity of Rubisco (assayedat saturating RuBP and CO2) constantly by 60%. The catalyticactivity of Rubisco in birch leaves was overall very low, evenwhen calculated from the total activity of Rubisco (Kcat 0.63–1.18 s–1), when compared to herbaceous C3 species. Signs of light acclimation were not observed in most of thephotosynthetic parameters and in chloroplast structure whenmature birch leaves were subjected to changes in growth lightfor 6 d. However, the change of the growth light either to highor low light caused day-to-day fluctuations in most of the measuredphotosynthetic parameters and in the case of the high lightplants signs of photoinhibition and photodestruction were alsoobserved (decrease in the amount of chlorophyll and increasein chlorophyll a/b ratio). As a result of these fluctuationsthese plants achieved a new and lower steady-state conditionbetween the light and dark reactions, as judged from the molarratio of RuBP to Rubisco binding site. Changes in carbon fixation rate and the levels of photosyntheticproteins were measured in fourth leaves of Lolium temulentumgrown until full expansion at 360 µmol quanta m–2s–1 and subsequently at the same irradiance or shadedto 90 µmol m–2 s–1. Ribulose-1,5-bisphosphatecarboxylase/oxygenase (Rubisco), light-harvesting chlorophylla/b protein of photosystem II (LHCII), 65 kDa protein of photosystemI (PSI), cytochrome f (Cytf) and coupling factor 1 (CF1) declinedsteadily in amount throughout senescence in unshaded leaves.In shaded leaves, however, the decrease in LHCII and the 65kDa protein was delayed until later in senescence whereas theamount of Cyt f protein decreased rapidly following transferto shade and was lower than that of unshaded leaves at the earlyand middle stages of senescence. Decreases in the Rubisco andCF1 of shaded leaves occurred at slightly reduced rates comparedwith unshaded leaves. These results indicate that chloroplastproteins in fully-expanded leaves are controlled individually,in a direction appropriate to acclimate photosynthesis to agiven irradiance during senescence. A midday depression in net photosynthesis and in stomatal conductancewas observed when leaves of well-watered Vitis vinifera plantswere subjected to a diurnal pattern of variation in leaf temperatureand leaf-to-air water vapour pressure difference similar toa summer day, while photon flux density was kept constant at1450 µmolm–2 s–1,. When leaves were kept atconstant leaf temperature (22.5°C) and leaf-to-air watervapour presure difference (8.5 Pa kPa–1) at the same lightintensity, stomata opened with the onset of illumination andmaximal conductance and photosynthesis values were observedabout 1 h later. Subsequently, conductance and photosynthesisdecreased gradually. Leaf water potential never dropped below{macron}0.3 MPa. Leaves kept under constant environmental conditionsshowed an afternoon decline in photosynthesis at high internalCO2, in carboxylation efficiency and in maximum conductanceas well as an increase in stomatal sensitivity to CO2. Whenthe photon flux density during the day was reduced to 750 µmolm–2 s–1, the afternoon depression in gas exchangerates was attenuated. To evaluate the possible effects of highlight stress on changes in chloroplastic behaviour we comparedlight response curves of photosynthesis determined with an oxygenelectrode, in the morning and in the afternoon after the plantswere exposed to either high or moderate photon flux densities.A significant depression in photosynthetic capacity was foundby this method in high light treated leaves, but not in leavespreviously exposed to moderate photon flux density. Apparentquantum yield decreased in the afternoon, particularly afterexposure to high light. Maximum chlorophyll a fluorescence at22°C was reduced and the quenching of fluorescence afterreaching the peak was slower in the afternoon than in the morning,especially in high light-treated leaves. Changes in the patternsof chlorophyll fluorescence kinetics were observed after lighttreatment, i.e. in the afternoon, with oscillations either absent(after high light) or significantly reduced (after moderatelight) in comparison to the morning. The significance of theseresults is discussed and it is suggested that a direct inhibitoryeffect of high light at the chloroplast level provides the bestinterpretation for the observed afternoon decline in photosyntheticrate. Chloroplast preparations from the young primary leaves of Phaseolusvulgaris L. cv. Canadian Wonder carry out the DNA-dependentincorporation of UTP into RNA at rates between 8 and 14 pmolUTP µg–1 chlorophyll h–1. It is estimatedthat 90% of the activity was localized in the chloroplasts.The incorporation proceeded for between 20 and 30 min at 35°C. The maximum rates of RNA synthesis were attained atpH 8.3, in the presence of 15 mM MgCl2. Chloroplasts were alsoactive, to a lesser extent, with 1.5 mM MnCl2. The simultaneouspresence of MnCl2 and MgCl2 resulted in inhibition of activity.Nuclear material prepared from young P. vulgaris leaves incorporatedUTP at a rate of about 12 pmol UTP µg–1 DNA h–1.On a chloroplast (Tritonsoluble) DNA basis chloroplast activitywas over 40-fold that of nuclei. Methods of solubilizing chloroplastRNA polymerase were explored. Yields of over 75% were achieved,but methods suitable for one species were not always successfulwhen applied to another. The highest yields of the P. vulgarisenzyme were obtained using EDTA and KCl. All methods resultedin solubilization of DNA. RNA synthesis by the soluble P. vulgarisenzyme proceeded for more than 40 min at 35 °C.  相似文献

Rates of net photosynthesis of the flag leaves of 15 genotypesof wheat and related species were measured throughout theirlife, using intact leaves on plants grown in the field. At thestage when rates were maximal, they were in general highestfor the diploid species, intermediate for the tetraploidspeciesand lowest for Triticum aestivum (means of 38, 32 and 28 mgCO2 dm–2 h–1 respectively). Rates were stronglynegatively correlated with leaf area, leaf width and the meanplan area per mesophyll cell and positvely correlated with stomatalfrequency and number of veins per mm of leaf width. The differencesamong species in these attributes were mainly related to ploidylevel. It was not possible to determine the relative importanceof each anatomical feature, though the changes in stomatal frequencyhad only slight effects on stomatal conductance and the observeddifferences in rates of photosynthesis were much greater thanwould be expected from those in stomatal conductance alone. There was genetic variation in rates of light dependent oxygenevolution of isolated protoplasts and intact chloroplasts butno difference attributable to ploidy. The mean rate, 91 µmolO2 mg–1 chlorophyll h–1, equivalent to 3.9 mg CO2mg-1chlorophyll h-1 was considerably less than the rate of photosynthesisin comparable intact leaves, which was 7.2 mg CO2 mg–1chlorophyll h–1. The total above-ground dry matter yields were least for thewild diploids T. urartu and T. thauodar and the wild tetraploidT. dicoccoides, but the other wild diploids produced as muchdry matter as the hexaploids. The prospects of exploiting differences in photosynthetic ratein the breeding of higher yielding varieties are discussed. Changes in anatomical and physiological features, includingchanges in amount per unit area of anthocyanin and chlorophyll,in leaves of seedling mango (Mangifera indica L. cv. Irwin)trees were determined to understand what controls the rate ofphotosynthesis (Pn) at various stages of development. The youngleaves of seedling trees contained high concentrations of anthocyanin.During enlargement of leaves, the disappearance of anthocyaninand the accumulation of chlorophyll occurred concomitantly;the anthocyanin content began to decrease markedly once theleaf area had reached a maximum. During the early period ofleaf development, the thickness of mesophyll tissue decreasedtemporarily, but when the length of the leaf reached half thatof a mature leaf, the mesophyll began to thicken again. Smallstarch grains appeared in the chloroplasts of the young leavesand chloroplast nucleoids (ct-nuclei) were distributed throughoutthe chloroplasts. When leaves matured, ct-nuclei were displacedto the periphery of chloroplasts because of the accumulationof large starch grains. Compared with young leaves, green andmature leaves contained greater concentrations of ribulose bisphosphatecarboxylase-oxygenase (RuBisCO) protein. The results of immunocytochemicalexamination of RuBisCO under the light microscope reflectedthe results of electrophoresis measurements of RuBisCO. Stands of spring wheat grown in open-top chambers (OTCs) wereused to assess the individual and interactive effects of season-longexposure to elevated atmospheric carbon dioxide (CO2 and ozone(O3) on the photosynthetic and gas exchange properties of leavesof differing age and position within the canopy. The observedeffects were related to estimated ozone fluxes to individualleaves. Foliar chlorophyll content was unaffected by elevatedCO2 but photosynthesis under saturating irradiances was increasedby up to 100% at 680 µmol mol–1 CO2 relative tothe ambient CO2 control; instantaneous water use efficiencywas improved by a combination of increased photosynthesis andreduced transpiration. Exposure to a seasonal mean O3 concentration(7 h d–1) of 84 nmol mol–1 under ambient CO2 acceleratedleaf senescence following full expansion, at which time chlorophyllcontent was unaffected. Stomatal regulation of pollutant uptakewas limited since estimated O3 fluxes to individual leaves werenot reduced by elevated atmospheric CO2, A common feature ofO3-treated leaves under ambient CO2 was an initial stimulationof photosynthesis and stomatal conductance for up to 4 d and10 d, respectively, after full leaf expansion, but thereafterboth variables declined rapidly. The O3-induced decline in chlorophyllcontent was less rapid under elevated CO2 and photosynthesiswas increased relative to the ambient CO2 treatment. A/Ci analysessuggested that an increase in the amount of in vivo active RuBisCOmay be involved in mitigating O3-induced damage to leaves. Theresults obtained suggest that elevated atmospheric CO2 has animportant role in restricting the damaging effects of O3 onphotosynthetic activity during the vegetative growth of springwheat, and that additional direct effects on reproductive developmentwere responsible for the substantial reductions in grain yieldobtained at final harvest, against which elevated CO2 providedlittle or no protection. Photosynthetic characteristics of detached green barley leavesafter 72 h of treatment with 0·2 mol m–3 of thepyridazinone herbicides SAN 6706, SAN 9785 and SAN 9789 werestudied. For comparison, the effects of 0·01 mol m–3diuron were also investigated. Pyridazinone herbicides causedonly a slight reduction of the total carotene content of thebarley leaves. The total chlorophyll content, as well as thelinolenic to linoleic acid ratio of chloroplast glycerolipids,however, remained unchanged. Diuron treatment caused total inhibitionof electron transport, as revealed by fast fluorescence inductionof leaves and the Hill reaction activity of chloroplasts. The14CO2-nxation by the leaves and the light-induced fluorescencequenching were also completely inhibited in vivo by diuron.Pyridazinone herbicides left 20–40% of the 14CO2-fixationfound in the control, in spite of the fact that their fast fluorescenceinduction tracings showed inhibition in the electron transport.Chloroplasts isolated from the leaves treated with pyridazinoneswere found to be highly active in mediating the ferricyanide-dependentHill reaction. In order to test the ability of pyridazinonesto inhibit photosynthetic electron transport in vivo, their‘prompt’ effect on fluorescence was also investigated.It is concluded that pyridazinone herbicides can readily andrapidly enter the chloroplasts and inhibit the photosyntheticelectron transport in vivo. The differences between the long-termeffects of pyridazinones and those of diuron suggest differencesin the inhibitory effectiveness on the various photosyntheticparameters between the two herbicide groups. It is suggestedthat pyridazinones can leave the chloroplasts during isolationowing to the loose binding onto the thylakoid membranes. Young tomato plants were exposed to two weeks of chilling undernon-photoinhibiting or mild photoinhibiting conditions. Thedevelopment of the leaves was studied under chilling and controlconditions by measuring several physiological parameters. Agradual decrease of the efficiency of the photosynthetic apparatuswith maturation and ageing occurred in unchilled plants. Thiswas reflected by gradual changes in CO2-saturated photosynthesisand protein and rubisco contents. Except for senescing leaves,a correlation close to 1 : 1 was observed between maximum rubiscoactivity and CO2-saturated photosynthesis. Chlorophyll (Chl)contents and photochemical chlorophyll fluorescence quenchingshowed strong decreases only in the last phase of senescencein the oldest leaves. In plants chilled under non-photoinhibitingconditions (10C, 100–150 µE m–2 s–1or 6C, 30–50 µE m-2 s–1), a similar patternof ageing was observed, and no indications were found for aninduction of protein or rubisco degradation by chilling. Sincethese plants stopped growing in the cold, they revealed lowertotal photosynthetic capacities than unchilled plants of thesame size. When the chilling conditions were mildly photoinhibitory(6C, 100–150 µE m–2 s–1), a much strongerdepression of rubisco activity and photosynthetic capacity wasfound in all leaves, which was partly reversible in the youngones. This decrease in CO2fixation capacity, in turn, led toa higher susceptibility of the chilled plants to photoinhibitionat 20C. It is concluded that the decrease of both photosyntheticcapacity and growth after long-term chilling in tomato is aconsequence of the preceeding ageing and senescing of the leavesduring chilling, in contrast to chilling-tolerant species withthe ability for acclimation to low temperatures. Acclimation of fully developed leaves of Bischofia javanicaBlume to shadelight was examined. Seedlings were grown undersimulated daylight (1000 µmol m–2 s–1), thentransferred to a simulated shadelight (40 µmol m–2s–1). When a high-light leaf was transferred to low light, large negativenet photosynthetic rates (Pm) were recorded. This decrease wasrapid, but within 7 d the rate increased and became equal tothe low-light control leaf. These changes in photosynthesisdid not follow the pattern of changes in stomatal conductance(gs). Transfer to the low light resulted in a dramatic decreasein leaf weight per unit area (Lw), and most of the decreasesin Lw occurred within 3 d of transfer when the Pm of the transferredleaf was well below that of the low-light control leaf. There was a significant decrease in chlorophyll a in the transferredleaf without an appreciable change in chlorophyll b resultingin a large decrease in the chlorophyll a to chlorophyll b ratio.Leaf chlorophylls per unit area were higher in the transferredleaf than the low-light control leaf. Maximum photosyntheticrate in the transferred leaf was decreased by 40% compared tothat for the high-control leaf, but was almost at the same extenthigher than the low-light control leaf The results are discussedin the context of carbon gain capacity of its seedlings underlight-limiting forest understorey habitats. The photosynthetic capacity of newly expanded leaves of vernalizedor non-vernalized plants of S24 perennial ryegrass (Lolium perenneL.), grown in long or short photoperiods, was measured in twoexperiments. In the first, leaves were protected from shadingduring development, while in the second, the natural shade ofneighbouring tillers in a sward was allowed. In the first experiment there was little effect of vernalization,day length or flowering, and leaves in all treatments had photosyntheticrates at 250 W m–2 of between 28 and 32 mg CO2 dm–2h–1.In the second experiment the photosynthetic rate of successiveleaves fell as sward leaf area increased. This downward trendwas reversed, however, in flowering tillers in the vernalizedlong-day treatment, while in the other treatments, which didnot flower, photosynthetic capacity continued to fall. It isconcluded that the leaves of reproductive tillers have highphotosynthetic capacities because stem extension carries themto the top of the canopy where they are well illuminated duringexpansion. The relationship between chlorophyll a (Chl a) and primary productivity(PP) in the uppermost water layer and the water column-based(0–15 m) integral values of those variables were examinedusing measurements taken in Lake Kinneret (Israel) from 1990to 2003. In 81% of all Chl a profiles examined, the distributionwas fairly uniform within the entire 0–15 m water column,and 12.3% of instances showed a prominent subsurface maximum,when the lake phytoplankton was dominated by the dinoflagellatePeridinium gatunense. Chl a can be reliably estimated by remotesensing techniques in the productive and turbid water of LakeKinneret, since Chl a concentration at surface layers can beextrapolated to the entire water column. Light vertical attenuationcoefficient average for wavelengths from 400 to 700 nm, Kd,ranged from 0.203 to 1.954 m–1 and showed high degreeof temporal variation. The maximal rate of photosynthetic efficiency,PBopt [average 3.16 (±1.50)], ranged from 0.25 to 8.85mg C m–3 h–1 mg Chl a–1. Using measured dataof Chl a, PBopt, and light as an input, a simple depth-integratedPP model allowed plausible simulation of PP. However, a lackof correlation between photosynthetic activity and temperature(or other variable with remotely sensed potential) renders theuse of models that require input of photosynthetic efficiencyto calculate integrated PP of little value in the case of productiveand turbid Lake Kinneret.  相似文献

Activities of photosynthetic and photorespiratory enzymes viz.,ribulose bisphosphate carboxylase, phosphoenol pyruvate carboxylaseand glycolate oxidase from jute (Corchorus olitorius L.; cv.JRO 632) leaves were compared with those from maize (C4) andsunflower (C3) leaves. The photosynthetic CO2 fixation products,the release of 14CO2 in light and dark following photosynthesisin 14CO2, chlorophyll a: b ratio, gross leaf photosyntheticrate and dry matter production rate were also studied. The regulation of senescence by oxygen-concentration, lightirradiance and H2O2 has been studied in leaf segments of Avenasativa L. cv. Suregrain. The development of the components of the senescence process,for example chlorophyll breakdown, proteolysis (as soluble aminoacids), hydroperoxides (as malondi-aldehyde) and permeability(as conductivity) is accelerated in light as the O2-tensionincreases. In darkness, 0.3% O2 accelerates increases in hydroperoxides,permeability and proteolysis and delays the chlorophyll break-down,but 0.0005% O2 delays all the components studied. In every casethe hydroperoxide content, permeability and proteolysis areclosely related. Any treatment inducing an increase in membranepermeability causes chlorophyll bleaching (photo-oxidation)if leaf segments are then treated with light in an atmospherecontaining oxygen. Light has a modulating effect on the senescenceprocess. An irradiance lower or higher than 40 W m–2 hasan accelerating effect on the senescence process. Stomatal conductances were measured in ageing leaves of Commelinacommunis L. as plants developed; stomatal responses to CO2 andabscisic acid (ABA) in epidermal strips of C. communis takenfrom ageing leaves of developing plants and in epidermal stripsfrom the same-aged leaves (the first fully-expanded leaf) ofdeveloping plants were also monitored. Stomatal behaviour wascorrelated with parallel measurements of photosynthesis andleaf ABA concentrations. Stomatal conductance in intact leavesdecreased from a maximum of 0-9 cm s– 1 at full leaf expansionto zero about 30 d later when leaves were very senescent. Conductancesdeclined more slowly with age in unshaded leaves. Photosynthesisof leaf slices also declined with age from a maximum at fullleaf expansion until about 30 d later when no O2 exchange wasdetectable. Exogenously applied ABA (0.1 mol m– 3) didnot affect respiration or photosynthesis. In epidermal stripstaken from ageing leaves the widest stomatal apertures occurredabout 10 d after full leaf expansion (just before floweringbegan) and then decreased with age; this decrease was less dramaticin unshaded leaves. The inhibitory effects of ABA on stomatalopening in epidermal strips decreased as leaves aged and wasgreater in the presence of CO2 than in its absence. When leaveswere almost fully-senescent stomata were still able to open.At this stage, guard cells remained healthy-looking with greenchloroplasts while mesophyll cells were senescing and theirchloroplasts were yellow. Similar data were obtained for stomatain epidermal strips taken from the same-aged leaves of ageingplants. The inhibitory effects of ABA on stomatal opening alsodecreased with plant age. In ageing leaves both free and conjugated ABA concentrationsremained low before increasing dramatically about 30 d afterfull leaf expansion when senescence was well advanced. Concentrationsof free and conjugated ABA remained similar to each other atall times. It is concluded that the restriction of stomatal movements inintact leaves as the leaves and plants age is due mainly toa fall in photosynthetic capacity of the leaves which affectsintracellular CO2 levels rather than to an inherent inabilityof the stomata to function normally. Since stomatal aperturein epidermal strips declines with plant and leaf age and stomatabecome less responsive to ABA (while endogenous leaf ABA levelsremain fairly constant until leaf senescence) it is suggestedthat some signal, other than ABA, is transmitted from the leafor other parts of the plant to the stomata and influences theirbehaviour. 14CO2 assimilation was studied with diploid, tetraploid, hexaploidspecies of the genera Triticum and their wild relatives Aegilops.Attached mature leaves of 3–4 weekold plants were allowedto undergo photosynthesis under air at ambient temperature.The pattern of distribution of 14C was notably similar in Triticumand Aegilops species whatever the level of ploidy. Sucrose wasthe sink for photosynthetic carbon. 14C for sucrose synthesis was supplied either through the glycolatepathway by glycolate, the product of the photorespiration orby the Calvin cycle intermediates exported into the cytoplasm.Depending on the species, the glycolate pathway provided 40to 75%of the sucrose 14C. The higher labeling of sucrose was associated with the greaterparticipation of the glycolate pathway in the wild diploid (DD)A. squarrosa and in the cultivated hexaploid (AABBDD) T. aestivum.The results suggest that the expression of the male D genomeis dominant over the female AB genome in T. aestivum. In T. aestivum under ambient conditions lowering (low temperature)or hindering (1% O2 ) photorespiration, sucrose labeling decreased,but serine and glycine labeling was favoured. We propose thatin wheat leaves, the role of photorespiration is to drain artof the carbon exported from the chloroplast as glycolate, towardssucrose synthesis. (Received March 16, 1979; )  相似文献

Gabaculine, a potent inhibitor of the biosynthesis of aminolaevulinicacid in plants, was supplied via the roots to seedlings of Loliumtemulentum and chloroplast development along the age gradientof the emergent leaf was followed. Exposure to the inhibitorprevented the development of an organized photosynthetic membranesystem and a noticeable accompanying feature was the appearanceof aggregates of osmiophilic globules even in young plastids.In untreated leaves these were only seen to any extent in chloroplastsundergoing senescence. Though chlorophyll formation was inhibitedby gabaculine, levels of carotenoid in treated tissue were somewhatelevated. However, there was a selective depletion ofß-carotenewhich, in situ, is closely associated with the photosyntheticreaction centre. Amounts of two plastid-localized tetrapyrrole-bindingproteins, light-harvesting chlorophyll protein of photosystem-2(LHCP-2) and cytochrome f, were determined. There was a closecorrespondence in inhibition between LHCP-2 and its prostheticgroup with the greatest decrease, compared to leaves of untreatedplants, being at the leaf base. In contrast, formation of theapoprotein and haem components of cytochrome f was apparentlynot so tightly coupled. Amounts of apoprotein were similar tothose of untreated controls, but haem was concentrated in theregion proximal to the base and declined along the age gradient,being absent from the leaf tip.
