Differences in ammonium net uptake by the roots of beech (Fagus sylvatica L.) and spruce (Picea abies (L.) Karst) trees between day and night were examined during the growing seasons in 1995 and 1996 using the depletion technique. In addition, diurnal courses of ammonium net uptake of both species were analysed in five sets of uptake experiments in May and September 1997 and were related (1) to the content of carbohydrates, organic acids and total soluble non protein N (TSNN) in the fine roots, and (2) to xylem flow densities and soil temperature. During the growing seasons 1995 and 1996, ammonium net uptake of beech was significantly lower during the night than during the day at 5 of 8 dates of measurement. On average, uptake rates during the night amounted to 50% of the uptake rates during the day. In spruce, the mean values of ammonium net uptake rates determined were similar between day and night during both growing seasons. In beech, the assessment of diurnal courses showed highest ammonium uptake rates during noon and in the afternoon and minima at midnight. In May 1997, comparable, but less pronounced diurnal patterns of ammonium uptake were observed in spruce, whereas in September 1997, ammonium uptake by spruce was constant during the day. Since no distinct differences in carbohydrate and organic acid contents in fine roots were observed during the diurnal courses and since the addition of sucrose into the artificial soil solutions root tips were exposed to did not alter ammonium uptake, depression of uptake by C- and/or energy limitation during night could be excluded. The TSNN contents in the fine roots of beech (May and September 1997) and spruce (May 1997) showed a diurnal pattern inverse to ammonium uptake. It is concluded that the enrichment of TSNN compounds during night that is apparently caused by a reduction of xylem transport is responsible for the down-regulation of ammonium net-uptake.  相似文献

In numerous locations in Europe spruce trees are exposed to high loads of nitrogen. The present study was performed to characterize the distribution of nitrogen compounds under these conditions. For this purpose Norway spruce ( Picea abies [L.] Karst.) trees were cultivated under close-to-natural conditions of a forest understory in soil from an apparently nitrogen-limited field site in the Black Forest either with, or without supplementation of nitrogen as ammonium nitrate. After 11 and 20 months, growth, total nitrogen contents of the biomass, and total soluble non-proteinogenic nitrogen compounds (TSNN, i.e. nitrate, ammonium, soluble proteinogenic and non-proteinogenic amino compounds) in needles, xylem sap and phloem exudate were analysed. After 20 months of growth, N-fertilization had slightly enhanced the biomass of current-, but not of 1-year-old shoots. At both harvests, total N-content of 1-year-old needles was increased by N-fertilization, whereas current-year needles were not significantly affected. By contrast, TSNN was elevated by N-fertilization in both current-year and 1-year-old needles. The increase in TSNN was mainly attributed to an accumulation of arginine. Xylem sap analysis showed that the increase in TSNN of the needles was a consequence of enhanced nitrogen assimilation of the roots rather than the shoot. Since also TSNN in phloem exudates was enhanced, it appears that N-fertilization elevates the cycling pool of amino compounds in young Norway spruce trees. However, this pool seems to be subject to metabolic interconversion, since mainly glutamine and aspartate are transported in the xylem from the roots to the shoot, but arginine accumulated in the needles and the phloem.  相似文献

Abstract: We assessed the role of water availability as a factor regulating the ability of beech seedlings to cope with competitive interference for nitrogen resources by an early successional species (Rubus fruticosus). A glasshouse experiment was performed with two levels of interference (beech with and without R. fruticosus ) and three levels of irrigation (high, intermediate, none). 15N uptake and partitioning of both species, and composition of N pools in leaves, roots and phloem of beech, were determined. Under all irrigation regimes, 15N uptake by beech seedlings decreased when grown together with R. fruticosus. R. fruticosus had higher 15N uptake rates than beech, under all water supply levels. When irrigation was reduced, a substantial decrease in 15N uptake of beech seedlings and a concurrent increase in 15N uptake by R. fruticosus were observed. Interference by R. fruticosus and low irrigation also affected the 15N partitioning in beech seedlings and resulted in reduced allocation of 15N to the roots. The combination of competitive interference and lack of irrigation led to an increase in soluble non-protein N in roots and leaves of beech, due to protein degradation. This response was attributed to an increase in levels of amino acids serving as osmoprotectants under these conditions. The concentration of proline in leaves of beech was negatively correlated to shoot water potential. A competition-induced reduction of total N in leaves of beech under high and intermediate irrigation was found. These results illustrate (1) the advantage of R. fruticosus in terms of N uptake when compared to young beech, particularly under inadequate water supply, and (2) the changes in N composition of beech seedlings in order to cope with reduced soil water and interference by R. fruticosus.  相似文献

To investigate the short‐term consequences of direct competition between beech and sycamore maple on root N uptake and N composition, mycorrhizal seedlings of both tree species were incubated for 4 days (i.e. beech only, sycamore maple only or both together) in an artificial nutrient solution with low N availability. On the fourth day, N uptake experiments were conducted to study the effects of competition on inorganic and organic N uptake. For this purpose, multiple N sources were applied with a single label. Furthermore, fine roots were sampled and analysed for total amino acids, soluble protein, total nitrogen, nitrate and ammonium content. Our results clearly show that both tree species were able to use inorganic and organic N sources. Uptake of inorganic and organic N by beech roots was negatively affected in the presence of the competing tree species. In contrast, the presence of beech stimulated inorganic N uptake by sycamore maple roots. Both the negative effect of sycamore maple on N uptake of beech and the positive effect of beech on N uptake of sycamore maple led to an increase in root soluble protein in beech, despite an overall decrease in total N concentration. Thus, beech compensated for the negative effects of the tree competitor on N uptake by incorporating less N into structural N components, but otherwise exhibited the same strategy as the competitor, namely, enhancing soluble protein levels in roots when grown under competition. It is speculated that enhanced enzyme activities of so far unknown nature are required in beech as a defence response to inter‐specific competition.  相似文献

Mixed spruce-beech plantations grown in large open-top chambers (OTC) were used to study consequences of elevated CO2, nitrogen-deposition and soil type on plant internal nitrogen and sulphur cycling of juvenile beech (Fagus sylvatica L.) and spruce (Picea abies Karst.) in a competitive situation. Processes of re-cycling as a consequence of protein turnover during leaf senescence in autumn were of further interest. For this purpose, phloem sap was collected in September 1998 and analysed for the composition and concentrations of organic and inorganic nitrogen and sulphur compounds. The phloem exudate of spruce showed higher total soluble non-protein nitrogen (TSNN) concentration on calcareous soil than on acidic soil, independent of the treatment. N-fertilization increased the N-concentration of phloem exudate significantly on both soil types, mainly by an increase of Arg and Gln concentrations. Elevated CO2 slightly increased TSNN on calcareous, but not on acidic soil. The combination of elevated CO2 and augmented N-deposition induced a further increase of TSNN on calcareous soil, but caused a lower N-effect on TSNN on acidic soil. Arg, the main TSNN component in phloem exudate, mediated this effect. Since Arg is considered to be a major nitrogen storage compound, it is concluded that in autumn elevated CO2 and augmented N-deposition, influence storage of N rather than N-supply of spruce. An effect of elevated CO2 and augmented N-deposition on GSH and sulphate concentrations in phloem exudate of spruce was not observed on acidic soil. On calcareous soil augmented N-deposition enhanced, elevated CO2 decreased phloem exudate GSH contents. In combination, elevated CO2 compensated the positive effect of N-deposition. The effects of elevated CO2 and augmented N-deposition on phloem sap N- and S-contents described above were not observed for beech trees. Apparently, elevated CO2 and augmented N-deposition did not affect plants internal S and N cycling of beech grown in spruce-beech plantations.  相似文献

A new concept illustrated by a corresponding mathematical model of nitrate metabolism regulation is proposed. The model is based on root nitrate compartmentation in several functional pools: storage, metabolic and a mobile pool which is intended for translocation to shoots. Data on nitrate uptake, compartmentation, reduction in intact roots and translocation to shoots were obtained on steady-state wheat seedlings grown at 25 and 12 degrees C in the root zone. The net uptake, influx/efflux ratio, mobile pool size and translocation changed depending on the medium temperature. The oscillations of the net uptake rate, nitrate tissue concentration were revealed and the effect of temperature on these changes was demonstrated. The scheme of regulation is based on the idea that net uptake through nitrate influx/efflux is under the control of the nitrate the mobile pool whose size was dependent on the nitrate translocation into shoots. The mathematical model is represented by a system of ordinary differential equations simplified according to the time hierarchy of reactions. It has a limit cycle at definite values of the parameters. The model postulates the mechanism of a positive feed-back regulation of the transfer of newly absorbed nitrate into translocated pool formed in the root cortex. To assess the physiological performance of drought-sensitive European beech ( Fagus sylvatica L.) under the dry Mediterranean climate prevailing at its southeastern distribution limit in Europe, we analyzed seasonal changes in carbon, nitrogen and water balance of naturally grown adult trees. We determined the foliar C and N contents, delta13C and delta18O signatures, total soluble non-protein nitrogen compounds (TSNN) in xylem, leaves, and phloem, as well as leaf water potential and photosynthetic quantum yield in northern Greece during 2003. Tissue sampling was performed in May, July, and September, while field measurements were conducted regularly. Climatic conditions for the 2003 growing season fall within the typical range of the studied area. The N- and C-related parameters displayed distinct seasonal courses. TSNN was highest in May in all tissues, and asparagine (Asn) was then the most abundant compound. Thereafter, TSNN decreased significantly in all tissues and both its concentration and composition remained constant in July and September. In both months, glutamate (Glu) prevailed in leaves, gamma-aminobutyric acid (GABA) in phloem exudates from twigs and trunks, and arginine (Arg) in the xylem sap, where loading with amino acids was rather low during that period, amounting to only 0.8 micromol N ml-1 in September. Highest total foliar N and C contents were detected in May, and the elevated abundance of nutrients as well as an increased foliar delta13C signature at the beginning of the growing season is attributed to remobilization processes. The signatures of delta18O, quantum yield and leaf water potentials varied only slightly throughout the growing season. Although summer precipitation at the study site was considerably lower compared to what is usual for typical central European beech forests, no intensive drought responses of the physiological apparatus were detected in the studied beech trees. This suggests efficient internal regulation mechanisms, constantly ensuring a favourable physiological status under the relatively dry Mediterranean climate.  相似文献

Nitrous oxide (N2O) emission estimates from forest ecosystems are based currently on emission measurements using soil enclosures. Such enclosures exclude emissions via tall plants and trees and may therefore underestimate the whole-ecosystem N2O emissions. Here, we measured plant-mediated N2O emissions from the leaves of potted beech (Fagus sylvatica) seedlings after fertilizing the soil with 15N-labelled ammonium nitrate (15NH4(15)NO3), and after exposing the roots to elevated concentrations of N2O. Ammonium nitrate fertilization induced N2O + 15N2O emissions from beech leaves. Likewise, the foliage emitted N2O after beech roots were exposed to elevated concentrations of N2O. The average N2O emissions from the fertilization and the root exposure experiments were 0.4 and 2.0 microg N m(-2) leaf area h(-1), respectively. Higher than ambient atmospheric concentrations of N2O in the leaves of the forest trees indicate a potential for canopy N2O emissions in the forest. Our experiments demonstrate the existence of a previously overlooked pathway of N2O to the atmosphere in forest ecosystems, and bring about a need to investigate the magnitude of this phenomenon at larger scales.  相似文献

Abstract Roots of sterile-grown, intact 6-day-old seedlings of Ricinus communis possess at least two independent active amino acid uptake systems, one for neutral and one for basic amino acids. The kinetics of uptake of L-proline and L-arginine, which were taken as representative substrates for the two systems, are biphasic. At low concentrations (0.01–0.5 mol m?3) Michaelis -Menten kinetics prevail, changing to a linear concentration dependence at higher substrate concentrations (1–50 mol m?3). L-glutamate uptake velocity is linear over the whole substrate concentration range. For comparison the uptake kinetics of nitrate and ammonium were determined as well as interactions among the different nitrogen sources. The Km value for nitrate uptake was 0.4 mol m?3, and for ammonium 0.1 mol m?3. The uptake capacity for nitrate or ammonium was approximately the same as for amino acids. The interaction between the uptake systems for organic and inorganic nitrogen is small. Two hypotheses for the physiological significance of amino acid uptake by roots were considered: (i) Uptake of amino acids from the soil-determination of amino acids in soil and in soil water indicates that they might contribute 15–25% to the nitrogen nutrition of the plant. (ii) Amino acid uptake systems of root cells serve primarily as retrieval of amino acids delivered from the phloem- it was found that 14C L-glutamine, which was delivered to the cotyledon and transported to the root via the phloem, was not lost by the roots, whereas it appeared in the bathing medium if L-glutamine was applied externally to the root to compete for the uptake sites; this suggests that an apoplastic pool of amino acids in the root exists due to their efflux from the phloem.  相似文献

To analyze the effects of elevated carbon dioxide concentration (PCO2) on the mass flow of reduced nitro- gen (N) in the phloem and xylem of trees, juvenile beech (Fagus sylvatica L.) and spruce (Picea abies (L.) Karst.) were grown in phytotrons and exposed to ambient and elevated PCO2 (plus 687.5 mg/m^3 CO2) for three growing seasons. Elevated PCO2 significantly decreased the mass flow of N from the shoot to roots of beech by significantly reducing the concentration of soluble amino compounds in the phloem, even if the area of conductive phloem of cross-sectional bark tissue was significantly increased, because of less callus deposition in the sieve elements. In spruce, the downward mass flow of reduced N also tended to be decreased, similar to that in beech. Resembling findings in the phloem, N mass flow from roots to shoot in both tree species was significantly diminished owing to significantly reduced concentrations of amino compounds in the xylem and a lower transpiration rate. Therefore, the mass flow of reduced N between shoots and roots of trees was mainly governed by the concentrations of soluble amino compounds in the phloem and xylem in relation to the loading of reduced N in both long-distance transport pathways.  相似文献

Sulfur nutrition of plants is largely determined by sulfate uptake of the roots, the allocation of sulfate to the sites of sulfate reduction and assimilation, the reduction of sulfate to sulfide and its assimilation into reduced sulfur-containing amino acids and peptides, and the allocation of reduced sulfur to growing tissues that are unable to fulfill their own demand for reduced sulfur in growth and development. Association of the roots of pedunculate oak (Quercus robur L.) and beech (Fagus sylvatica L.) trees with ectomycorrhizal fungi seems to interact with these processes of sulfur nutrition in different ways, but the result of these interactions is dependent on both the plant and the fungal partners. Mycorrhizal colonisation of the roots can alter the response of sulfate uptake to sulfate availability in the soil and enhances xylem loading and, hence, xylem transport of sulfate to the leaves. As a consequence, sulfate reduction in the leaves may increase. Simultaneously, sulfate reduction in the roots seems to be stimulated by ectomycorrhizal association. Increased sulfate reduction in the leaves of mycorrhizal trees can result in enhanced phloem transport of reduced sulfur from the leaves to the roots. Different from herbaceous plants, enhanced phloem allocation of reduced sulfur does not negatively affect sulfate uptake by the roots of trees. These interactions between mycorrhizal association and the processes involved in sulfur nutrition are required to provide sufficient amounts of reduced sulfur for increased protein synthesis that is used for the enhanced growth of trees frequently observed in response to ectomycorrhizal association. During the growing season of the exceptionally dry and warm year 2003, we assessed seasonal changes in nitrogen, carbon and water balance related parameters of mature naturally grown European beech (Fagus sylvatica L.) along a North–South transect in Europe that included a beech forest stand in central Germany, two in southern Germany and one in southern France. Indicators for N balance assessed at all four sites were foliar N contents and total soluble non-protein nitrogen compounds (TSNN) in xylem sap, leaves and phloem exudates; C and water balance related parameters determined were foliar C contents, δ13C and δ18O signatures. Tissue sampling was performed in May, July and September. The N related parameters displayed seasonal courses with highest concentrations during N remobilization in May. Decreased total foliar N contents as well as higher C/N ratios in the stands in central Germany and southern France compared to the other study sites point to an impaired N nutrition status due to lower soil N contents and precipitation perception. TSNN concentrations in leaves and phloem exudates of all study sites were in ranges previously reported, but xylem sap content of amino compounds in July was lower at all study sites when compared to literature data (c. 1 μmol N mL−1). In September, TSNN concentrations increased again at the two study sites in southern Germany after a rain event, whereas they remained constant at sites in central Germany and southern France which hardly perceived precipitation during that time. Thus, TSNN concentrations in the xylem sap might be indicative for water balance related N supply in the beech trees. TSNN profiles at all study sites, however, did not indicate drought stress. Foliar δ13C, but not foliar C and δ18O followed a seasonal trend at all study sites with highest values in May. Differences in foliar δ13C and δ18O did not reflect climatic differences between the sites, and are attributed to differences in altitude, photosynthesis and δ18O signatures of the water sources. Except of low TSNN concentrations in the xylem sap, no physiological indications of drought stress were detected in the trees analysed. We suppose that the other parameters assessed might not have been sensitive to the drought events because of efficient regulation mechanisms that provide a suitable physiological setting even under conditions of prolonged water limitation. The uniform performance of the trees from southern France and central Germany under comparably dry climate conditions denotes that the metabolic plasticity of mature beech from the different sites studied might be similar.  相似文献

The inhibitory effect of NH4+ on net NO3- uptake has been attributed to an enhancement of efflux and, recently, to an inhibition of influx. To study this controversy, we devised treatments to distinguish the effects of NH4+ on these two processes. Roots of intact barley (Hordeum vulgare L.) seedlings, uninduced or induced with NO3- or NO2-, were used. Net uptake and efflux, respectively, were determined by following the depletion and accumulation in the external solutions. In roots of both uninduced and NO2- -induced seedlings, NO3- efflux was negligible; hence, the initial uptake rates were equivalent to influx. Under these conditions, NH4+ had little effect on NO3- uptake (influx) rates by either the low- or high-Km uptake systems. In contrast, in plants preloaded with NO3-, NH4+ and its analog CH3NH3+ decreased net uptake, presumably by enhancing NO3- efflux. The stimulatory effect of NH4+ on NO3- efflux was a function of external NH4+ and internal NO3- concentration. These results were corroborated by the absence of any effect of NH4+ on NO2- uptake unless the roots were preloaded with NO2-. In this case NH4+ increased efflux and decreased net uptake. Hence, the main effect of NH4+ on net NO3- and NO2- uptake appears to be due to enhancement of efflux and not to inhibition of influx.  相似文献

采用营养液水培方法,以"雪美"品种甜瓜(Cucumis melo L.)为材料,研究了外源脯氨酸(Proline)对盐胁迫下甜瓜幼苗叶片和根系硝酸还原的影响。结果表明:(1)盐胁迫提高了甜瓜幼苗叶片和根系内铵态氮(NH4+-N)和可溶性蛋白含量;降低了硝态氮(NO-3-N)含量和硝酸还原酶(nitrate reductase,NR)活性。(2)外源施用脯氨酸明显地提高了盐胁迫下甜瓜幼苗叶片和根系内NO-3-N和可溶性蛋白含量;降低了盐胁迫下甜瓜幼苗叶片和根系内NH+4-N含量;增强了盐胁迫下甜瓜幼苗体内NR活性。研究结果表明,外源脯氨酸可以通过调节甜瓜幼苗体内硝酸还原酶活性和氮化合物含量来缓解盐胁迫对甜瓜幼苗植株的伤害。  相似文献

Summary Decreases in the concentrations of nitrogen, phosphorus, potassium, calcium and magnesium, in the shoots of wheat seedlings soon after the start of waterlogging were mainly attributed to an inhibition of ion uptake and transport by roots in the oxygen deficient soil. There was a small net accumulation of nitrogen, phosphorus and potassium by the aerial tissues, principally the tillers rather than the main shoot. By contrast, calcium and magnesium accumulated in both tillers and main shoot. With waterlogging, nitrogen, phosphorus and potassium were translocated from the older leaves to the younger growing leaves, and in the case of nitrogen this was associated with the onset of premature senescence. Calcium and magnesium were not translocated from the older leaves, the younger leaves acquiring these cations from the waterlogged soil. The promotion of leaf senescence by waterlogging was counteracted by applications of nitrate or ammonium to the soil surface, or by spraying the shoots with solutions of urea, but the beneficial effects on shoot growth were small.The role of mineral nutrition in relation to waterlogging damage to young cereal plants is discussed.  相似文献

The interactions between the assimilation and transport of nitrogenand carbon were investigated in barley and spinach leaves. Bothplants were fumigated with NH3 (1 mg m–3 and the contentof amino acids, sucrose and carbon intermediates of amino acidmetabolism were analysed in the leaves, apoplast and phloemsap. The following changes took place in the C- and N-metabolismof barley leaves during 5 h of fumigation with NH3 (a) The contentsof amino acids, especially glutamine, largely increased andthe contents of sucrose, 2-oxoglutarate, phosphoenolpyruvate,and glycerate-3-phosphate declined. (b) A decrease in the phophoenolpyruvatecontent was accompanied by an increased activity of phosphoenolpyruvatecarboxylase. (c) The altered cytosolic concentrations of aminoacids and sucrose during NH3 fumigation correlated with similarchanges in the apoplast and phloem sap. The altered percentageof each amino acid relative to the total amino acid concentrationin the cytosol, caused by NH3 fumigation, is reflected in theapoplast and the phloem sap. The results indicate that the concentrations of amino acids in the cytosol determine their concentrationsin the phloem. The shoots of fireweed (Chamerion angustifolium (L.) Holub) and common flax (Linum usitatissimum L.) were infused with 50 mM KNO3 solution to compare the influence of nitrate on photosynthesis and assimilate export from leaves in plants with the symplastic and apoplastic phloem loading, respectively. The infusion of nitrate in the shoots of both plant species lowered 14CO2 fixation and enhanced the assimilate transport in the upward direction. Irrespective of the phloem loading type, the incorporation of 14C into sucrose decreased in nitrate-treated seedlings exposed to assimilation for short (3 min) periods. However, when shoots were sampled 3 h after 14CO2 fixation, the content of 14C-labeled sucrose was higher in treated plants than in control seedlings infused with water. In fireweed, in contrast to flax, a similar temporal pattern was also characteristic for 14C incorporation into oligosaccharides. Within 3 h after nitrate infusion into the fireweed apoplast, the mitochondria and the cell vacuolar system underwent ultrastructural changes indicative of the increase in cytosolic osmotic pressure. At the same time, we observed accumulation of fibrillar inclusions in cell vacuoles of vascular bundles. It is concluded that the mechanisms of nitrate influence on photosynthesis and sugar export in leaves of symplastic and apoplastic plants are similar to a certain extent and involve the blocking of pores in phloem tubes, initiated by the NO-signaling system.  相似文献
