Concentration of cytokinins (CKs) of zeatin-type (Z) and isopentyladenine-type (iP) were determined using an enzyme-linked immunosorbent assay (ELISA) technique in xylem sap and bulk leaf extract of the hemiparasiteMelampyrum arvense before and after attachment to the host (Capsella bursa pastoris). In all the samples Z-type were dominant, though the iP-type was also frequent. The results also demonstrate that in comparison with the unattached hemiparasite after attachment to the host the concentration of Z-type in xylem sap increased about 92 and 182 times in the light and dark phase, respectively, and that of the iP-type about 34 times in the two phases. The concentration of Z-type and iP-type in leaves expressed per dm3 of cell water was at the level of 10-7 M and 10-8 M, respectively in the unattached hemiparasite. After attachment, the concentration of CKs increased to 10-5 M and 10-6 M for Z-type and iP-type, respectively. In xylem sap the concentration of Z-type was at the level of 10-9 M and 10-7 M and in the unattached and attached hemiparasite, respectively. The concentrations of iP-type were 10-10 M and 10-8 M for the unattached and attached hemiparasite, respectively. The diurnal oscillation of CK concentration was evident in the unattached hemiparasite, but after attachment their level was nearly constant and independent of the diurnal cycle.  相似文献

Flowering, fruit set, embryological development, and pollination trials were Investigated In Santslum album L. Each ovary may have three to four ovules. Mlcrosporogenesls and megasporogenesls In the same flower were synchronized at the earlier stages of flower development. However, at anthesls, when pollen was mature, the magaspore had developed only to the stage of a one- to two-nucleus embryo sac. As the eight-nucleus embryo sac developed, some mamelon cells began to undergo programmed cell death, forming holes Into which the eight-nucleus embryo sacs extended, becoming "N" or "S" shaped. The development from a two-nucleus embryo sac to a matured eight-nucleus embryo sac lasted up to 10 d. Fruit-set from open pollination was less than 2%. The endosperm develops prior to division of the zygotic embryo and one to three embryos and endosperms were formed In the same fruit. A mature seed usually germinates to produce one seedling; however, two and three seedlings from one seed were also observed, albeit at a low frequency. Pollination trials showed that no seed sets when Inflorescences were covered with s bag; however, artificial pollination could Improve fruit set. Our pollinaUon trials and embryological studies proved that the flower of S. album Is dlchogamous and fruit set has high heterozygosity.  相似文献

The content of abscisic acid (ABA) in abaxial leaf epidermis of the host (Capsella bursa pastoris) and the unattached hemiparasiteMelampyrum arvense showed diurnal changes. ABA content increased during the light period and declined rapidly upon the darkening of leaves. In an attached hemiparasite the content of ABA in the epidermis was maintained at an almost constant level irrespective of the diurnal cycle. As compared with the maximum level in the host, at the end of the light phase the content of ABA in abaxial epidermis constituted about 70 % and 164 % in the unattached and attached hemiparasite, respectively. No significant changes in ABA content were recorded in adaxial epidermis. In all the samples abaxial/adaxial epidermis ABA content ratio was about 3.6:1 in light phase. In darkness this ratio decreased to about 1.1:1 in the host and the unattached hemiparasite and did not show significant change after attachment. ABA content ratio in mesophyll was 1:0.7:1.5 for the host, the unattached, and attached hemiparasite, respectively. In comparison with the host the concentration of ABA in xylem sap of the hemiparasite constituted about 31 % and 152 % for the unattached and attachedM. arvense, respectively.  相似文献

Gas exchange characteristics of a hemiparasiteMelampyrum arvense L. before and after attachment to the hostCapsella bursa pastoris (L.) Med. were compared. The net photosynthetic rates (PN) on a leaf area basis were extremely low and in comparison to the value obtained for the host were about 15 % and 23 % for the unattached and attached hemiparasite, respectively. Also the concentration of photosynthetic pigments was low (as compared with the host the content of chlorophylls was about 33 % and 49 % and of carotenoids about 38 % and 36 % in the unattached and attached hemiparasite, respectively). On the other hand the rates of respiration were high (about 1.8 and 2.6 times higher in the unattached and attached hemiparasite, respectively, than in the host). In darkness stomatal conductance (gS) of the host and the unattached hemiparasite was rapidly reduced to 10 % of the value obtained in light, gS of the attached hemiparasite was decreased only by about 30%. A total reduction of gS occurred at relative water content (RWC) of 85 %, 75% and 45 % for the unattached hemiparasite, the host, and the attached hemiparasite, respectively. The transpiration (E) rate in the preparasitic stage was very low, being 2.6 and 4.5 times smaller than in the host and the attached hemiparasite, respectively. In the attached hemiparasite WUE was 7.5 and 3 times poorer than in the host and in the preparasitic stage, respectively.  相似文献

The effect of cytokinins (CKs) and K+ on stomatal behaviour in darkness were studied in the root hemiparasite Melampyrum arvense before (the preparasitic stage) and after attachment to the host (Capsella bursa pastoris L. Med.). The solutes were applied with xylem stream. The stomatal apparatus of the attached hemiparasite was insensitive to externally supplied CKs and K+. Contrary to this finding, the stomatal aperture of hemiparasite in the preparasitic stage increased to about 25, 40 and 69% of the value obtained in light, respectively, after treatment with 200 mM KCl and 10-5 M zeatin riboside ([9R]Z), applied separately or together. CKs influenced K+ transport. The treatment with KCl and [9R]Z,separately or together, increased the content of K+ in guard cell pairs to about 32, 46 and 79 % of the value obtained in light, respectively. Other CKs had a smaller effect (45 - 16 %) in comparison with that of [9R]Z; isopentyladenine was nearly inactive. Small cyclic peptides exhibiting potent biological activity have great potential for anticancer therapy. An antiproliferative cyclic octapeptide, cyclosaplin was purified from somatic seedlings of Santalum album L. (sandalwood) using gel filtration and RP-HPLC separation process. The molecular mass of purified peptide was found to be 858 Da and the sequence was determined by MALDI-ToF-PSD-MS as ‘RLGDGCTR’ (cyclic). The cytotoxic activity of the peptide was tested against human breast cancer (MDA-MB-231) cell line in a dose and time-dependent manner. The purified peptide exhibited significant antiproliferative activity with an IC50 2.06 μg/mL. In a mechanistic approach, apoptosis was observed in differential microscopic studies for peptide treated MDA-MB-231 cells, which was further confirmed by mitochondrial membrane potential, DNA fragmentation assay, cell cycle analysis and caspase 3 activities. The modeling and docking experiments revealed strong affinity (kcal/mol) of peptide toward EGFR and procaspase 3. The co-localization studies revealed that the peptide sensitizes MDA-MB-231 cells by possibly binding to EGFR and induces apoptosis. This unique cyclic octapeptide revealed to be a favorable candidate for development of anticancer agents.  相似文献

The biosynthesis of the newly isolated polyamine, sym-homospermidine (NH(2)-[CH(2)](4)-NH-[CH(2)](4) -NH(2)), was studied by using radioactive amino acids. Arginine was the most effective precursor, being about 10 times as active as ornithine. Unlabelled agmatine and putrescine markedly inhibited the incorporation of [(14)C]arginine into homospermidine. Similarly the incorporation of ornithine was inhibited by unlabelled arginine and putrescine. gamma-Aminobutyraldehyde, the oxidation product of putrescine, was considered to be one of the intermediates in the biosynthesis of homospermidine. The biosynthesis may involve a Schiff-base formation of putrescine with gamma-aminobutyraldehyde and subsequent reduction. A limited synthesis of spermidine also takes place under these conditions.  相似文献

Nuclear ribosomal DNA (nrDNA) ITS sequences and partial sequences of three non-coding chloroplast DNA (cpDNA) introns and spacers were used to assess genetic variation within and among three presumed host races of the hemi-parasite Viscum album L. Currently, identification of host races occurs via the host trees, and morphological differences are minute at best. cpDNA and nrDNA ITS sequences revealed little sequence variation, but the variation found consistently supported the distinction of three host races. cpDNA and ITS sequences were not incongruent, as assessed by the incongruence length difference test. A combined analysis supported the sister group relationship between mistletoes from deciduous trees and fir.  相似文献

Mistletoes are bird dispersed, hemi-parasitic shrubs infecting a large number of woody host plants. Chloroplast fragment length polymorphisms were used to study genetic differentiation among presumed host races, population genetic structure, and to elucidate the postglacial migration history of mistletoe ( Viscum album ) across the entire natural distribution range in Europe. The populations sampled belong to four closely related taxa, three of which are widely distributed and differ in their host trees, whereas a fourth taxon is rare and endemic to the Island of Crete. The molecular analysis of chloroplast DNA variation supported the distinction of these four taxa. We further found evidence for phylogeographical structure in each of the three widely distributed host races. Independent of host race, mistletoe haplotypes from Turkey were distinct and distant from those found elsewhere in Europe, suggesting that highly differentiated populations, and possibly new taxa, exist at the range limit of the species.  相似文献

1. A hydroxyproline-containing protein was isolated from the soluble fraction of sandal leaves (Santalum album L.) and the purified protein was homogeneous by disc electrophoresis. 2. It is a glycoprotein containing 16% carbohydrate, the components of which were mainly arabinose, with only small amounts (about 5%) of galactose. The principal amino acids were glutamic acid, aspartic acid, glycine, alanine, arginine, lysine, proline and hydroxyproline, which together comprised 60% of the total. The number of acidic amino acids exceeds the number of basic amino acids. By Sephadex gel filtration, the approximate molecular weight was found to be about 63000. The ratio of residues of hydroxyproline to those of arabinose was 1:2. 3. The native protein is resistant to the action of several proteolytic enzymes. After partial hydrolysis with 0.1m-HCl, the protein became susceptible to attack by Pronase but remained resistant to collagenase.  相似文献

Changes of ABA levels in chilled rice (Oryza sativa L.) seedlings of two varieties were determined. On exposure to chilling, ABA concentration rapidly increased in the chilling-tolerant cultivar (cv. Tainung 67, TNG.67) but not in the chilling-sensitive cultivar (cv. Taichung Native 1, TN.1). Both detached shoots and roots of TNG.67 seedlings showed a significant ABA increase after exposure to chilling. TN.1 seedlings could not accumulate ABA under low temperature but well-watered status. Exogenous application of the ABA biosynthetic inhibitor, fluridone, reduced ABA accumulation, as well as survival ratio of chilled TNG.67 seedlings. Electrolyte leakage and leaf conductance were also increased by the inhibitor and the effects could be reversed by exogenously applied ABA. ABA concentrations in xylem sap of TNG.67 seedlings increased within 4 h after chilling, and this was temporally coincident with the reduction of leaf conductance. The roles of endogenous ABA in the tolerance of rice seedlings to chilling on a whole plant basis are discussed and suggested.  相似文献

Abstract

Viscum album L. ssp. album is semi-parasitic on deciduous trees and shrubs. In order to identify hosts and map the distribution of V. album ssp. album in Croatia and Slovenia, field research was carried out, and herbaria were surveyed. In Croatia and Slovenia, V. album ssp. album occurred on 59 taxa. In Croatia, there were 52 hosts (33 autochthonous and 15 allochthonous species, two cultivars and two hybrids). In Slovenia, there were 25 hosts (21 autochthonous and four allochthonous species). There were 18 hosts common to both countries, 34 hosts were found only in Croatia, and seven hosts only in Slovenia. The hosts belonged to 13 families. The majority of these (19 species) belong to the Rosaceae, followed by Salicaceae, Aceraceae, Betulaceae, Fagaceae, Juglandaceae, Tiliaceae, Hippocastanaceae, Ulmaceae, Oleaceae, Fabaceae, Moraceae and Viscaceae. All hosts have been previously recorded in the literature, except Alnus japonica (Thunb.) Steud., Amelanchier lamarckii F.G. Schroed. and Crataegus nigra Waldst. et Kit. The distribution of this mistletoe was scattered, due to the scattered distribution of hosts, local conditions, movement of bird-vectors, etc. A continuous distribution was found only in part of the distribution area of narrow-leaved ash (Fraxinus angustifolia Vahl).  相似文献

Santalum album L. (Indian sandalwood) is an economically important but vulnerable tropical tree species. Cultures were established via direct shoot regeneration from axillary buds on Murashige and Skoog (MS) medium supplemented with 2.5 mg L?1 6-benzylaminopurine (BAP). The shoots were multiplied using MS medium containing 1.0 mg L?1 BAP and 0.5 mg L?1 indole-3 acetic acid and rooted on half strength MS medium containing 1.0 mg L?1 indole-3 butyric acid. The rooted plantlets were hardened and acclimatized in greenhouse using soilrite® and cocopeat (1:1) mixture. The concentrations of photosynthetic pigments were analyzed and detected less under in vitro conditions (6.05 μg g?1 FW) as compared to the 4 weeks old hardened (6.91 μg g?1 FW) and 12 weeks old acclimatized plantlets (7.8 μg g?1 FW) under greenhouse (ex vitro) environment. The anatomical evaluation of plantlets at subsequent stages of propagation suggested that the in vitro raised plantlets possessed structural abnormalities such as underdeveloped cuticle, unorganized tissue systems, reduced mesophyll tissues, fewer vascular elements and mechanical tissues, and loosely arranged thin walled paranchymatous ground tissues, which were slowly repaired during ex vitro hardening and acclimatization process to validate the developmental adaptation of micropropagated plantlets for maximum survival in the field (98.0% survival rate). The findings could help in the optimization of high-frequency commercial micropropagation of S. album for year-round production, and supply of this economically prominent vulnerable plant species to the farmers and the industries that rely on it.

We have noticed Annona squamosa and Psidium guajava trees exhibiting heavy infection from the leafy mistletoe, Dendrophthoe falcata. The mistletoe has skipped its infection on other more common hosts, viz. Mangifera indica and Achras sapota growing in the close proximities to the infected hosts. The haustoria on each of the infected hosts were distinct. On A. squamosa, they occur as solitary unions while on P. guajava they exhibit a network of epicortical roots. The article illustrates these observations and discusses the possible explanation to such evidences.  相似文献

The levels of endogenous gibberellic acid equivalents, indole acetic acid, trans-zeatin and trans-zeatin riboside in bearing and non-bearing shoots of walnut (Juglans regia L.) were detected by high-performance liquid chromatography during vegetation period. The levels of endogenous hormones were analysed in four different shoot types of current season, bearing terminal shoot, non-bearing terminal shoot, bearing lateral shoot and non-bearing lateral shoot. Their average levels differed statistically at five sampling times from 25 May to 23 September. Relationships among changes in endogenous gibberellin, auxin and cytokinin contents were computed using correlation and regression analyses. Remarkable positive correlation coefficients were found among changes in gibberellin, auxin and cytokinin contents. Statistical findings indicated that changes in endogenous hormones exhibited a collective behaviour in walnut shoots during vegetation period.  相似文献
