Chloroplast DNA (cpDNA) and mitochondrial DNA (mtDNA) of malesterile tobacco plants obtained by fusion of Nicotiana tabacumprotoplasts and X-irradiated N. debneyi protoplasts were analyzed.Digestion of cpDNA isolated from ten male sterile lines withfour restriction endonucleases (EcoRI, XhoI, SmaI and HindIII)indicated that these lines possessed either one or the otherparental chloroplast genome. Neither mixture of two types ofcpDNA nor unique restriction fragments were detected in anyof the cases examined. The genetic constitution of chloroplastgenomes identified by restriction analysis of cpDNA showed goodagreement with that based on isoelectric focusing of the largesubunit of the Fraction I protein. The mtDNA from five fusion-derivedmale sterile plants showed banding patterns quite differentfrom each other and from the parental plants. Each plant exhibitednew restriction fragments not found in the parental species.These findings indicate that recombinational events in the mitochondrialgenomes take place rather frequently in the mixed cytoplasmsafter protoplast fusion, whereas the mixed chloroplasts becomesegregated to homogeneity. This study was done to confirm our previous observation withthe pattern of changes in electron transport composition inducedby an imbalance of the electron transport state. Contents ofphotosystem (PS) I and II complexes and their antennae and Cytb6/f complex were determined for systems of cyanobacterium SynechocystisPCC 6714 of different PS I/PS II ratios. The results indicatedthat (1) the observed changes in the PS I/PS II ratio are not-dueto regulation of the activities of the respective PS's but tochanges in their contents, (2) the molar ratio between PS IIand Cyt b6/f complexes was fairly constant when marked changesoccurred in the PS I content, and (3) the PS II and Cyt b6/fcontents per cell remained fairly constant while the PS I contentchanged markedly. These findings agree with our previous observationwith autotrophic cells of Anacystis nidulans Tx 20 and supportour argument that in cyanobacterial and red algal electron transportsystems, the content of the terminalcomponent(s), such as PSI complex, is regulated in order to maintain a balance betweenthe electron influx by PS II action to the system and the effluxby PS I action from it. Using Toyopearl and cyclohexane: cyclohexanol solvent, fourCl-containing Chls were separated from 36Cl-labeled cells ofthe blue-green, Plectonema boryanum. In normally grown cells,all four Cl-containing chlorophylls amounted to less than 1/2,000of the total Chi and about 1/50 of P700, values much lower thanpreviously reportedcontents of Chi RC I, and varied from algato alga. The level of Cl-containing Chi was markedly enhancedwhen the cells were poisoned with methyl viologen. The quantitative composition of thylakoid components such asthe photosystem (PS) I/PS II ratio in the cyanobacterial photosyntheticsystem is regulated in response to the light regime. The regulationoccurs as changes in PS I content due to control of either PSI formation or decomposition. In order to determine which ofthese two is controlled in this regulation, experiments wereperformed to determine the light-induced PS I decrease in cellsof Synechocystis PCC 6714 under conditions where protein synthesiswas suppressed, i.e. the incubation without a nitrogen sourcefor cell growth or with chloramphenicol. The results revealedthat light-induced PS I decrease did not occur when synthesisof the thylakoid system was suppressed by incubation withouta nitrogen source or by the addition of chloramphenicol, indicatingthat (1) the thylakoid composition is regulted in the processof thylakoid formation and (2) the regulation is achieved bythe control of PS I formation. A population of Pseudotachea litturata (Pfeiffer, 1851) fromTarifa (Càdiz, Spain) has been studied. The morphologicalresults are compared with those from P. splendida, Iberus gualtierianus,I. alonensis, I. marmoralus, I. guiraoanus and four speciesof the genus Cepaea using, as an exploratory method, the Wagnerparsimony procedure and 18 characters of the shell, genitalsystem and karyotype have been analysed. According to this methodit seems that the taxonomical position of P. litturata in thegenus Pseudotachea is confirmed, and agrees with the phylogeneticalrelationships in this group of species. The genus Cepaea seemsto be well established, although two species groups can be distinguished:C. nemoralis—C. hortensis and C. syluatica—C. vindobonensis.These differ mainly in chromosome number, diverticulum lengthand degree of shell polymorphism. Although the present resultsdo not allow us to clarify the current taxonomical problemswithin the genus Iberus, the species studied seem to belongto a natural group (Received 15 September 1987; accepted 1 January 1988)  相似文献

The experimental susceptibility of Portugala inchoata, Euomphalia(M.) brigantina, Ponentina ponentina, Hygromia (P.) cantabricacantabrica and Hygromia (P.) cantabrica covadongae to Neostrongyluslinearis larvae were studied for the first rime. The total numberof molluscs used in the infections was 207 and they were killedin series between day 6 and SO post-infection. In order to definethe suitability of the molluscs as intermediate hosts for thisparasite, the number of L-I that penetrated the foot and developedto L-I, L-Il and L-III has been taken into account, as wellas the first days on which the different stages were observed One way analysis of variance showed statistically significantdifferences for the penetration and development of the larvaeamong the species of molluscs examined Statistically significant differences concerning the susceptibilityof the adult and young examples of P. inchoata and E. Short-term and long-term movements of the patellid limpet Patellaflexuosa Quoy & Gaimard were studied on an intertidal rockyshore where a mytilid, Hormomya mutabilis (Gould), formed anextensive mussel zone. At low tide, all the limpets were found restingwithin gaps in the mussel zone, which were formed after massdislodgement of the mussels by waves. The foraging areas ofall the marked limpets were restricted to within the gaps duringfour 24h diving observations. All of these limpets displayedhoming behaviour after short excursions with a maximum averagedistance of 6.7 cm from their homes. During short-term observationperiods of about 4–5 days, no marked limpets moved fromone gap to another gap across the mussel bed of H. mutabilis.During long-term observation periods of one month, however,10–30% of marked limpets shifted their homes to differentgaps that had a lower limpet density. The shell length of theseemigrants was, in general, significantly smaller than that ofnon-emigrants. The percentage of emigrants tended to be higherin summer and autumn than in winter and spring. The movementof P. flexuosa within patchy habitats is discussed with a focuson the difference in site fidelity between small and large limpets. The plastids of dark-grown resting cells of Euglena gracilisKlebs var. bacillaris Cori undergo only limited developmentwhen illuminated at the developmental threshold for light intensity7 foot-candles (ft-c) (27 µW/cm2). In the present work,we have found that these low intensity cells have substantialamounts of electron transport components such as ferredoxin-NADPreductase and Cyt c-552 but only trace amounts of the majorantenna components such as the light-harvesting Chl-proteincomplex (LHCP), the LHCP oligomer, CP la, Chi b and the 26.5kDa apo-LHCP; CP I and CPa are at levels comparable to the electrontransport components. Exposure of the low intensity cells tonormal light intensity causes large increases in major antennacomponents and small increases in electron transport components.The kinetics of accumulation of the antenna components Chi band apo-LHCP during greening of dark-grown resting cells atnormal intensities are the same as for Chi a. The low intensitywild-type cells strongly resemble mutants of Euglena low inChi b grown at normal intensities in lacking major antenna components. Effects of yeast extract on the inhibitory effects of diethylstilbestrol(DES) were examined in Neurospora crassa The inhibition by DESof mycelial growth and lightinduced phase-shifting of the conidiationrhythm was overcome by the concomitant addition of yeast extractin which the active agents were substances of low molecularweight. However, shifting of the phase of the conidiation rhythmby DES was not affected by addition of the yeast extract. Theseresults suggest that DES exerts an effect at two different sites,at least, in Neurospora: one site being the system which connectsthe perception of light to the clock mechanism and is relatedto mycelial growth; the other site being the system which controlsthe clock mechanism itself. Hydrogen peroxide was formed in isolated cell walls from Marchantiapolymorpha L. in the presence of MnCl2 by either NADH or NADPHoxidation. This reaction was stimulated by phenols such as 2,4-dichlorophenolor p-coumarate, suggesting a reaction similar to that proposedfor the last step of lignification in higher plant cells, althoughbryophytes have been reported to be devoid of lignin. Gould and Lewontin use San Marco, Venice, to criticise the adaptationist program in biology. Following their lead, the architectural term “spandrel” is now widely used in biology to denote a feature that is a necessary byproduct of other aspects of the organism. I review the debate over San Marco and argue that the spandrels are not necessary in the sense originally used by Gould and Lewontin. I conclude that almost all the claims that Gould makes about San Marco are wrong and that it is reasonable to view the architectural spandrel as an adaptation. The spandrels example has not provided a good illustration of why adaptive explanations should be avoided. In fact, it can be used as an example of how adaptive explanations can be dismissed even when there is evidence in their favour. Seasonal and ontogenetic changes in natural development timewere studied for Eodiaptomus japonicus in Lake Biwa in 1986and 1987. Wild individuals in a certain developmental stagewere collected, fed on natural food and examined until theyhad moulted twice. Natural development times fluctuated irrespectiveof temperature from May to October. Food deficiency delayeddevelopment in all feeding stages, and food availability probablydetermined natural development time. Serious food limitationraised mortality in copepodid stage I. In November developmentwas delayed even with enough food. The development of E.japonicuswas almost isochronal except for a short prefeeding naupliarstage I and a long copepodid stage V.  相似文献

My general aim is to clarify the foundational difference between Stephen Jay Gould and Richard Dawkins concerning what biological entities are the units of selection in the process of evolution by natural selection. First, I recapitulate Gould’s central objection to Dawkins’s view that genes are the exclusive units of selection. According to Gould, it is absurd for Dawkins to think that genes are the exclusive units of selection when, after all, genes are not the exclusive interactors: those agents directly engaged with, directly impacted by, environmental pressures. Second, I argue that Gould’s objection still goes through even when we take into consideration Sterelny and Kitcher’s defense of gene selectionism in their admirable paper “The Return of the Gene.” Third, I propose a strategy for defending Dawkins that I believe obviates Gould’s objection. Drawing upon Elisabeth Lloyd’s careful taxonomy of the various understandings of the unit of selection at play in the philosophy of biology literature, my proposal involves realizing that Dawkins endorses a different understanding of the unit of selection than Gould holds him to, an understanding that does not require genes to be the exclusive interactors.  相似文献

The contents of phytochrome I and II in crude extracts fromembryonic axes of Pisum sativum cv. Alaska seeds were immunochemicallydetermined using purified pea phytochrome I and II as standards.We have produced and used three different types of mouse monoclonalanti-pea phytochrome antibodies (mAP) such as one reacting preferentiallywith phytochrome I, one with phytochrome II, and one with bothI and II. Phytochrome II was separated from I in the samplesusing immobilized column chromatography with mAPl. The amountsof two phytochrome species were quantitatively measured withwestern blotting and ELISA. Ca. 0.2 µg /axis of phytochromeI and ca. 0.05 µg /axis of phytochrome II were detectedby ELISA after imbibition for 12 h in the dark, though smallamounts of both were detected in dry axes. Ca. 0.05 µg/axis each of phytochrome I and II were detected by ELISA afterimbibition for 12 h in the light, and the results were confirmedby western blotting. This study showed that phytochrome II isnot green-tissue-specific, being also found in dark-imbibedembryonic axes, and that although light significantly lowersthe content of phytochrome I in the axis, it does not significantlyaffect that of phytochrome II. Extensive series of Amphidromus Albers, 1850 were collectedfrom low-altitude areas (30–500 m above sea level)in Thailand, Laos, Malaysia, Singapore and Indonesia between1993 and 2004. Data on shell colour, genital and spermatophoremorphology, and radular structure are presented. Five speciesof Amphidromus (Amphidromus) are reviewed, of which four occurin Thailand. Intraspecific variation between populations ofAmphidromus (Amphidromus) atricallosus (Gould, 1843) is considered;three subspecies are accepted and a new subspecies described.Subspecies of A. (A.) inversus (Müller, 1774) and A. (A.)schomburgki (Pfeiffer, 1861) are reconsidered, and a new subspeciesof the latter is described. The hydrobiid gastropod genus Potamopyrgus is shown to be representedin Australia by the New Zealand species P. antipodarum (Gray).It is widely distributed in south eastern Australia and Tasmaniafollowing its introduction about the middle of the last century.The Australian populations of Potamopyrgus were known, incorrectly,as P. niger, because of a misinterpretation of Paludina nigraQuoy & Gai-mard, 1835. Paludina nigra, here placed in thegenus Fluvidona, is redescribed and a neotype designated. Crocodylus porosus is a mound-nesting crocodilian in which incubationtemperature influences the rate of embryonic development, theprobability that embryos will survive to hatching, post-hatchinggrowth rates and the probability of hatchlings surviving to2 yr of age. Similar responses have been described in Alligatormississippiensis (Joanen et al., 1987) and C. niloticus (Hutton,1987), and they reflect a suite of "non-sexual" effects of incubationtemperature. Temperature-dependent sex determination allocatessex on the basis of these "non-sexual" effects. In C. porosus,it results in maleness being assigned to embryos with high probabilitiesof surviving and good potential for post-hatching growth. Withinthe limits of survival, effects of the moisture environmenton embryological development rate and hatchling fitness seemminor relative to those of the temperature environment. Reptilian orders have either obligate oviparity (chelonians,crocodilians and rhynchocephalians) or facultative oviparity(squamates), depending on the extent of embryonic developmentwithin the oviducts. The distinction is equally one betweenembryos which are buffered from thermal effects within a female'sbody (facultative oviparity) and those that are not (obligateoviparity). Facultative oviparity and internal thermal bufferingmay be the primitive condition within the Class Reptilia, andthe "shell-less" eggs of extant squamates may reflect the originalamniote egg. Obligate oviparity, which also exists in birds,appears to have been a specialized development, and is a blindend in the evolution of viviparity among vertebrates. The significanceof thermal buffering being lost in obligate oviparous reptilesremains unclear.  相似文献

Seed priming may enhance establishment success of cool-seasonrange grasses which must compete with annual weeds for earlyspring moisture. Previous priming studies have confirmed germinationrate enhancement for these species but relative treatment effectsunder field-temperature conditions have not been assessed. Weprimed seeds of thickspike wheatgrass [Elymus lanceolatus(Scribn.and J. G. Smith) Gould], bluebunch wheatgrass [Pseudoroegneriaspicata(Pursh) Löve], Sandberg bluegrass (Poa sandbergiiVasey.) and bottlebrush squirreltail [Elymus elymoides(Raf.)Swezey] and evaluated their relative emergence rate in threesoil types as a function of spring-planting date. Germinationresponse was simultaneously evaluated in laboratory germinatorsthat were programmed to simulate the field-temperature regimeat planting depth. Seed priming enhanced both germination andemergence rate with the greatest effect occurring during theearlier, cooler planting dates. Total emergence and emergencerate in the field were lower than for the equivalent germinationresponse in the laboratory. Thermal-germination response wasmodelled and predictions developed for evaluating potentialgermination under late winter/early spring soil-temperatureregimes. Regulation of the assembly of the photosystem I (PS I) complexin response to the light regime in the photosynthetic systemof cyanophytes was studied in Synechocystis PCC 6714. The relationshipbetween the assembly of the PS I complex and synthesis of Chla was examined by model experiments in which synthesis of Chla was controlled by two inhibitors, gabaculine (GAB) and 2,2'-dipyridyl(DP). Both inhibitors caused a change to a lower ratio of PSI to PS II even under light that normally induces a high ratioof PS I to PS II. The change in stoichiometry induced by theseinhibitors was suppressed when protein synthesis was inhibitedby chloram-phenicol, similarly to the change in the stoichiometryinduced by light that excites mainly PS I (PS I light). Comparisonof the levels of PS I, PS II and Cyt b6-f complexes per cellindicated that a selective suppression of the assembly of thePS I complex was induced by the inhibitors: the stoichiometricrelationship among PS I, PS II and Cyt b6-f complexes was identicalto that induced by PS I light or white light of high intensity.GAB induced a decrease in size of the phycobilisome also, whileDP did not, similarly to PS I light. The results indicate thatthe ratio of PS I to PS II can be changed by the control ofsynthesis of Chl a. They also suggest that control of the synthesisor supply of Chl a probably exerted at site(s) in or after theprocess of the Mg-protoporphyrin branch, is involved in themechanism of regulation of the assembly of the PS I complexin cyanophytes.
