Metabolism of Glycine by Rumen Microorganisms   总被引:5,自引:4,他引:1       下载免费PDF全文
Rumen microorganisms rapidly metabolized glycine at rates varying from 0.014 to 0.241 mumole of glycine per ml per min. The main metabolic products were carbon dioxide, acetic acid, and ammonia; little glycine was incorporated into bacterial protein. Use of carboxyl or methylene-labeled glycine showed that the carbon dioxide came mainly from the carboxyl of glycine, whereas both carbons of acetic acid were derived partly from the methylene carbon of glycine and partly from carbon dioxide. The ratio of carbon-14 to nitrogen-15 in glycine isolated from the protein of rumen bacteria incubated in the presence of N(15)- and C(14)-labeled glycine indicated that most of the extracellular glycine incorporated into protein was incorporated without intervening deamination.  相似文献

1,6-缩水-β-D-吡喃葡萄糖是纤维素类物质热解的主要产物,黑曲霉突变株CBX-209能较好地利用该糖作为唯一的碳源和能源生长并产生有用的代谢产物柠檬酸,其效率与利用葡萄糖大致相当。利用葡萄糖氧化酶和竦根过氧化物酶复合系统测定证明该菌株不存在1,6-缩水-β-D-吡喃葡萄糖水解酶,采用快原子轰击质谱技术结合6-磷酸葡萄糖脱氢酶系统进行测定,结果表明经(NH4)2SO4沉淀或阴离子交换层析析处理后的无细胞提取液在加入ATP和Mg^2 条件下能直接催化1,6-缩水-β-D-吡喃葡萄糖合成6-磷酸葡萄糖,证明黑曲霉突变株中存在一个新酶,即1,6-缩水-β-D-吡喃葡萄糖激酶。该酶为诱导酶。  相似文献

In field experiment studies that tested the effects of three partly resistant cultivars of spring oilseed turnip rape ( Brassica campestris ), multiplication of clubroot was moderate. When initial soil inoculum levels were 47–72% of infected test plants in the bioassay, only 2.1–10% of the plants were infected after harvest. The yield of partly resistant lines was 5–10% higher compared with that of the susceptible cultivar SW Kulta, which on average yielded 1.73 t/ha. The average disease severity index (DSI) after harvest in these trials was 12.7 for the nonresistant cultivar SW Kulta and ranged between 2.2 and 6.2 for the partly resistant lines. The study demonstrated that in fields where the soil infestation level gives a DSI of less than 10, or when less than 20% of the bait plants are infected in the bioassay, it is possible to avoid the risk of severe yield losses from clubroot infections by integrating a partly resistant cultivar into the crop rotation.  相似文献

L-Alanyl-L-tyrosine and glycyl-L-phenylalanine labelled with 14C competed with each other and with the dipeptide antibiotic bacilysin for transport into Staphylococcus aureus NCTC 6571 in a medium which did not support growth. They also competed with other dipeptides and several tripeptides. The fast initial transport ofthe two labelled peptides appeared to show Michaelis-Menten kinetics. Neither was transported into a bacilysin-resistant mutant of S. aureus NCTC 6571, although tyrosine was taken up by the mutant as readily as it was by the parent strain. Uptake of alanyltyrosine or glycylphenylalanine was followed by rapid hydrolysis of the peptide and the excretion of tyrosine or phenylalanine. Glycine liberated from glycylphenylalanine was partly degraded and partly incorporated into the bacterial wall. The behaviour of these dipeptides paralleled the inactivation of bacilysin by suspensions of S. aureus and the appearance of its C-terminal amino acid, anticapsin, in the extracellular fluid.  相似文献

It was shown that soluble cytoplasmic proteins of rabbit cardiac muscle were able to penetrate into phospholipid bilayer during preincubation with liposomes. The incorporation of cytoplasmic proteins into the liposomic membrane induced the sodium permeability that was sensitive to neurotoxins (veratridine and tetrodotoxin). This effect was partly eliminated by addition of tetrodotoxin to the external medium and completely eliminated in the presence of tetrodotoxin on the both sides of the liposomic membrane. It was found also that some of the proteins incorporated into the liposomes become resistant to pronase digestion when the enzyme is added to external medium. At the same time pronase included in the internal volume of the proteoliposomes was able to digest partly proteoliposomic proteins. All these data support the ideas of the integral incorporation of soluble cytoplasmic proteins into the liposomic membrane and to the existence of cytoplasmic precursor of voltage-dependent sodium channel.  相似文献

In order to investigate how the flavor of fresh tea-leaves turn into that of green tea, six essential oils were obtained from chopped fresh tea-leaves, whole tea-leaves, and their residues after steaming, green tea and black tea by steam-distillation. They are then compared with each other in regard to their flavor, amount of I2-absorption, βγ-hexenol content and ultraviolet spectrum. It is concluded that the change mentioned above is produced partly by fleeing of βγ-hexenol and partly by some chemical change concerning βγ-hexenol.  相似文献

The acidic, partly folded states of bovine carbonic anhydrase II (BCAII) were used as an experimental system to study the interactions of partly denatured proteins with lipid membranes. The pH dependence of their interactions with palmitoyloleoyl phosphatidylcholine (POPC) and palmitoyloleoyl phosphatidylglycerol (POPG) membranes was studied. A filtration binding assay shows that acidic partly folded states of BCAII bind to POPC membranes. Fluorescence emission spectra from Trp residues of the bound protein are slightly shifted to shorter wavelength and can be quenched by a water-soluble quencher of fluorescence, indicating that the binding occurs without deep penetration of Trp residues into the membrane. The content of beta-structures of the protein in solution, as revealed by FT-IR spectroscopy, decreases in the partly folded states and the binding to POPC membrane occurs without further changes of secondary structure. In the presence of 0.1 M NaCl, a partly folded state self-aggregates and does not bind to POPC membrane. At acidic pH, BCAII binds to POPG membranes both at high and low ionic strength. The binding to the anionic lipid occurs with protein self-aggregation within the lipid-protein complexes and with changes in the secondary structure; large blue shifts in the fluorescence emission spectra and the decrease in the exposure to water-soluble acrylamide quencher of Trp fluorescence strongly suggest that BCAII penetrates the hydrocarbon domain in the POPG-protein complexes.  相似文献

Inclusion in agarose gel significantly affects the conformational dynamics of native and acidic partly folded states of tuna apomyoglobin, a single tryptophan containing protein, as documented by frequency domain fluorometry investigations. The heterogeneity of the tryptophanyl emission decay increases on gel inclusion compared to that observed for free-in-solvent protein at both neutral and acidic pH, thus suggesting that the interconversion rate among conformational substates is somewhat reduced. The observation that this effect is much more pronounced for the partly folded state suggests that confined environments such as those existing in the living cells might favor the sequential folding process avoiding that structured intermediates rapidly convert into less structured ones.  相似文献

Lactaptin, a human milk protein with a molecular weight of 8.6 kDa, is a fragment of human ?casein, which has cytotoxic activity toward mammalian cancer cells in vitro. RL2 is a recombinant analogue of lactaptin, which induces the apoptosis of human cancer cells in culture and suppresses the tumor growth in vivo. It has been shown earlier that RL2 penetrates into both human cancer and nonmalignized cells and binds to cytoskeletal structures. In this process, it induces the apoptosis of cancer cells and does not diminish the viability of normal cells. The mechanism of the penetration of RL2 into human cancer cells has been studied by flow cytometry and fluorescence microscopy using the inhibitors of different endocytosis pathways. It has been shown that RL2 penetrates into cells partly through lipid raft-mediated dynamin-independent pinocytosis and partly through direct penetration across the plasma cell membrane. An analysis of the primary structure of RL2 and the mechanism of its penetration into the cell suggests that it can be assigned to the class of cell-penetrating peptides.  相似文献

1. The influence of insulin on the metabolism of [1-14C]glucosamine by diaphragm muscle from normal rats and rats rendered diabetic with streptozotocin has been studied. 2. The glucosamine was converted into glucosamine 1-phosphate, glucosamine 6-phosphate, glycogen, lactate and small amounts of other unidentified intermediates. 3. Insulin increased the incorporation of 14C into glycogen in both the normal and diabetic muscle, but did not increase the formation of the glucosamine phosphate esters. 4. The 14C content in the glycogen was present partly as glucose and partly as glucosamine; there was significantly more [14C]glucose in the glycogen of the diabetic muscle than in that of the normal muscle.  相似文献

The H-2 complex of the mouse contains numerous class I genes of unknown function. These genes are here classified into families according to homology in the exons encoding the variable domains. There are one major and at least five minor families, whose members are partly clustered and partly interspersed on the mouse chromosome. DNA sequences show that not only Tla and Qa-2 but also other minor-family genes have intact coding domains. These may be expressible genes of novel types.  相似文献

Granular starches of nine botanical origins were exposed to low-pressure glow plasma generated in the air from either a high voltage DC supply or an AC inductor. Experiments with low energy air-plasma (10−2 Torr) and starches provided insight into some details of their surface structure. Starches were partly oxidized to carboxylic starches and partly depolymerized. Their affinity to plasma depended on their botanical origin. The structure of starch granules seemed to be a crucial factor, which determined the behaviour of starches in plasma. Lipids and/or proteins stabilized polysaccharides to plasma being preferentially decomposed.  相似文献

Taurine uptake into rat brain synaptosomal fractions appears to occur by two saturable transport processes and by bulk diffusion. The transport requires the presence of sodium ions. The dependence of the transport on temperature and cellular respiration implies that the uptake is an active process. The active process is specific for taurine and closely related amino acids. Brain regions differ in their ability to transport taurine. Uptake is not due to mitochondrial contamination of the synaptosomal fractions. However, glial contamination might partly contribute to the uptake. Kainic acid lesions of rat corpus striatum and cerebellum reduce taurine uptake implying that the uptake is, at least partly, into neurons.  相似文献

Chicken liver bile acid-binding protein (formerly known as chicken liver basic fatty acid-binding protein) binds to anionic lipid membranes acquiring a partly folded state [Nolan, V., Perduca, M., Monaco, H., Maggio, B., and Montich, G. (2003) Biochim. Biophys. Acta 1611, 98-106]. To understand the mechanisms of its interactions with membranes, we have investigated the presence of partly folded states in solution. Using fluorescence spectroscopy of the single Trp residue, circular dichroism in the far- and near-UV, Fourier transform infrared spectroscopy, and size-exclusion chromatography, we found that L-BABP was partly unfolded at pH 2.5 and low ionic strength, retaining some of its secondary structure. Addition of 0.1 M NaCl at pH 2.5 or decreasing the pH to 1.5 produced a more compact partly folded state, with a partial increase of secondary structure and none of tertiary structure. Fluorescence emission spectra of this state indicate that the Trp residue is within an environment of low polarity, similar to the native state. This environment is not produced by the insertion of the Trp into soluble aggregates as revealed by size-exclusion chromatography, fluorescence anisotropy, and infrared spectroscopy. The presence of partly folded states under acidic conditions in solution suggests the possibility that membrane binding of L-BABP occurs via this state.  相似文献

The results of progeny tests of males and females captured during two field trials of sex-linked translocation strains for genetic control of L. cuprina are presented. Males released as mature larvae survived to adulthood and mated with field females. However, the levels of genetic death introduced into the population were insufficient to suppress the native population. This was due partly to seasonal ineffectiveness of the release method, and partly to poor performance of the released males. On average, the mating competitiveness of the released males was only one-third that of field males, whereas their field-reared, translocation-bearing sons were fully competitive with native males.  相似文献

Summary An exhaustive study of the egg membranes of the oocytes of Trichiurus savala and Triacanthus brevirostris was made with particular reference to the origin and structure of the zona pellucida. The zona pellucida makes its first appearance as a deeply stained narrow zone of fine granular or amorphous homogeneous material around the oocytes, between the follicular epithelium and the vitelline membrane. This homogeneous substance is the product of the follicle cells. Very soon, radial striations in the zona pellucida get formed out of the homogeneous substance of the zona itself. The peripheral cytoplasm of the oocyte, by this time, also gets differentiated into a fibrillar layer. The follicle cells as well as the ooplasmic fibrillar layer send out a number of protoplasmic fibres towards each other into the canalicular passages of the radially striated zona. These fibres meet in the middle of the zona, differentiating it into two concentric zones, an outer—zona radiata externa and an inner-zona radiata interna. The former is purely follicular in origin, whereas the latter is partly follicular and partly ooplasmic in nature.  相似文献

In this paper, we prepared composite fibers via electrospinning from either W/O or O/W emulsion. SEM images demonstrate the beads-in-string structures in these fibers and proved this technique to be an effective method for microencapsulation. As a practical application, Ca-alginate microspheres, which serve as reservoirs for hydrophilic drugs, were prepared in a reverse emulsion and then incorporated into poly (l-lactic acid) (PLLA) fibers by electrospinning. With the bovine serum albumin (BSA) loaded into the microspheres, the beads-in-string structure thus entrapped hydrophilic proteins in hydrophobic polymeric matrix. In the in vitro release test, BSA, which was released from composite fibers, achieved prolonged release profiles and lower burst release rates than those from naked Ca-alginate microspheres. In comparison with other well-established techniques to prepare microcapsules, such as solvent evaporation and spray-drying techniques, emulsion electrospinning features partly competing, partly complementary characteristics. Extension to other emulsion systems will be able to fabricate new types of functional structures.  相似文献

A comparative study of the NAH and TOL catabolic plasmids was carried out to provide information for future genetic manipulation experiments involving these two plasmids. The plasmids were studied in a strain of P. putida and its mutant derivatives. The NAH and TOL plasmids were found to be incompatible. Under the conditions used in these experiments the TOL plasmid transferred into some strains into which NAH was unable to transfer. The use of mutants to remove certain catabolic activities encoded by the bacterial host cell facilitated the allocation of growth genotypes to the NAH and TOL plasmids. TOL encoded the degradation of benzoate, m-toluate and p-toluate, whereas NAH encoded the degradation of naphthalene and salicylate. The other plasmid-associated growth phenotypes were partly plasmid-specified and partly specified by the host cell. The pH optimum of the catechol 2,3-dioxygenase specified by the TOL plasmid was approximately 6.7, whereas that of the NAH-encoded enzyme was approximately 8.3.  相似文献
