Background and Aims

Myrica rivas-martinezii is a critically endangered endemic of the laurel forest of the Canary Islands and co-occurs very close to M. faya. Some authors suggest that M. rivas-martinezii and M. faya are two morphs of the same species, so molecular markers were used to estimate the levels and structuring of genetic variation within and among natural populations in order to evaluate genetic relationships between these two congeners.

Methods

Six polymorphic microsatellite (simple sequence repeat, SSR) markers were used to determine the genetic diversity and the genetic relationship between both Myrica species.

Key Results

Most of the natural populations analysed were in Hardy–Weinberg equilibrium for both taxa. Analysis of molecular variance (AMOVA) for both species revealed that most of the genetic variability detected was contained within populations (92·48 and 85·91 % for M. faya and M. rivas-martinezii, respectively), which it is consistent with outcrossing and dioecious plants. Estimates of interpopulation genetic variation, calculated from FST and GST, were quite low in the two taxa, and these values did not increase substantially when M. rivas-martinezii and M. faya populations were compared. The UPGMA dendrogram based on Nei''s genetic distance clustered the populations by their island origin, independently of taxon. In fact, the mixture of individuals of both taxa did not appreciably disrupt the intrapopulational genetic cohesion, and only 3·76 % variation existed between species.

Conclusions

All the results obtained using molecular markers indicate clearly that both taxa share the same genetic pool, and they are probably the same taxa. Considering that M. rivas-martinezii is classified as at risk of extinction, there should be a change of focus of the current management actions for the conservation of this putatively endangered Canarian endemic.Key words: Canary Islands, conservation genetics, microsatellites, Myrica rivas-martinezii, Myrica faya, plant conservation  相似文献

The phytochemical information on Millettia dura Dunn, M. ferruginea (Hochst.) Baker and M. ferruginea subsp. darassana (Cufod.) J.B. Gillett was reviewed. All the three taxa elaborate mainly isoflavones (33 reported), occurring in the flowers, seeds/seed pods, stem bark and root bark. Out of the 33 isoflavones reported, some 19 (ca. 58%) contain prenyl at C-8 or its modification as 2,2-dimethylchromene ring at C-7/C-8, occurring in all the three taxa. Except for three isoflavones isolated from M. ferruginea subsp. darassana, all the isoflavones of these taxa are 5-deoxygenated. In these taxa, oxygenation at C-6 is a common feature, while isoflavones with C-8 oxygenation are rare, only three reported, and all of these from M. dura. There are 7 rotenoids reported from these taxa, and occur almost entirely in the seeds/seedpods of these plants. The major rotenoid with methylenedioxy group at C-2/C-3, millettone and its 12a-hydroxy derivative, millettosine, occur only in M. dura, this appears to distinguish M. dura from M. ferruginea.  相似文献

Phylogenies were inferred from nearly complete small subunit (SSU) 18S rDNA sequences of 12 species of Meloidogyne and 4 outgroup taxa (Globodera pallida, Nacobbus abberans, Subanguina radicicola, and Zygotylenchus guevarai). Alignments were generated manually from a secondary structure model, and computationally using ClustalX and Treealign. Trees were constructed using distance, parsimony, and likelihood algorithms in PAUP* 4.0b4a. Obtained tree topologies were stable across algorithms and alignments, supporting 3 clades: clade I = [M. incognita (M. javanica, M. arenaria)]; clade II = M. duytsi and M. maritima in an unresolved trichotomy with (M. hapla, M. microtyla); and clade III = (M. exigua (M. graminicola, M. chitwoodi)). Monophyly of [(clade I, clade II) clade III] was given maximal bootstrap support (mbs). M. artiellia was always a sister taxon to this joint clade, while M. ichinohei was consistently placed with mbs as a basal taxon within the genus. Affinities with the outgroup taxa remain unclear, although G. pallida and S. radicicola were never placed as closest relatives of Meloidogyne. Our results show that SSU sequence data are useful in addressing deeper phylogeny within Meloidogyne, and that both M. ichinohei and M. artiellia are credible outgroups for phylogenetic analysis of speciations among the major species.  相似文献

A taxonomic revision of Australian Macrobrachium identified three species new to the Australian fauna – two undescribed species and one new record, viz. M. auratumsp. nov., M. koombooloombasp. nov., and M. mammillodactylus(Thallwitz, 1892). Eight taxa previously described by Riek (1951) are recognised as new junior subjective synonyms, viz. M. adscitum adscitum, M. atactum atactum, M. atactum ischnomorphum, M. atactum sobrinum, M. australiense crassum, M. australiense cristatum, M. australiense eupharum of M. australienseHolthuis, 1950, and M. glypticumof M. handschiniRoux, 1933. Apart from an erroneous type locality for a junior subjective synonym, there were no records to confirm the presence of M. australe(Guérin-Méneville, 1838) on the Australian continent. In total, 13 species of Macrobrachiumare recorded from the Australian continent. Keys to male developmental stages and Australian species are provided. A revised diagnosis is given for the genus. A list of 31 atypical species which do not appear to be based on fully developed males or which require re-evaluation of their generic status is provided. Terminology applied to spines and setae is revised.  相似文献

The hoverfly genus Merodon Meigen, 1803 (Diptera, Syrphidae), which comprises more than 200 species is distributed in the Palaearctic and Afrotropical Region, but one species, Merodon equestris, is a pest of flower bulbs and has been invasive into New Zealand and North America. In this work, the Merodon aurifer species group, a member of the M. avidus-nigritarsis lineage, is revised. The diagnostic characters of the M. aurifer species group, and the morphological differences between M. aurifer Loew, 1862 and M. nudicorpus sp. n. are explained and illustrated, as well as their distribution map. A detailed study of the type material resulted in three new synonyms: M. distinctus Palma, 1863 is a junior synonym of M. aurifer; M. manicatus (Sack, 1938) and M. testacoides Hurkmans, 1993 are junior synonyms of M. testaceus.Lectotypes are designated for two taxa M. aurifer and M. manicatus instead of the erroneously designated types for these species in Hurkmans (1993).  相似文献

Essential oils from the leaves of three closely related taxa of Monardella (M. crispa, M. undulata var. undulata, M. undulata var. frutescens) were analysed by GC and GC-MS. All three taxa showed highly similar qualitative chromatographic profiles. Monardella crispa and M. undulata var. frutescens apparently show closer affinities qualitatively to each other than either taxon did to M. undulata var. undulata. The chemical data suggest, however, in addition to morphological and ecogeographical data, that M. undulata var.frutescens may represent a stabilized hybrid derivative between M. crispa and M. undulata var. undulata.  相似文献

An electrophoretic comparison of the nematodes Rugopharynx longibursaris and R. omega, both from Macropus rufogriseus in south-eastern Australia, revealed fixed genetic differences at 4.5% of the 23 enzyme loci examined. The electrophoretic data do not therefore reject the null hypothesis that the two taxa are conspecific. R. longibursaris was found in Tasmania and in the western mainland population of M. rufogriseus, while R. omega occurred only in the eastern mainland population. Implications for the taxonomic status of the western host population are considered. Specimens formerly assigned to R. omega, from Thylogale stigmatica from Queensland, were found to differ at 45.0% of enzyme loci from specimens from M. rufogriseus. Morphological examination revealed differences in the shape of the buccal capsule, the position of the deirid, the morphology of the spicule tip and the presence of a gubernaculum. A new species, R. sigma, is erected for specimens from T. stigmatica, T. thetis and T. calabyi.  相似文献

A survey of the secretory structures of leaflets has been carried out for five taxa of the genus Zornia J.F. Gmel. to assess possible taxonomic value of the glands, taking into account that these taxa, Zornia curvata Mohlenbr., Z. gemella Willd. ex Vogel, Z. glabra Desv., Z. latifolia Sm. and Z. reticulata Sm., have overlapping diagnostic characters. The leaflet secretory structures of the five studied taxa of Zornia were mucilage epidermis, mucilage cavities, and idioblasts secreting phenolic compounds. Mucilage epidermis is found in all five aforementioned taxa. Mucilage cavities are observed on both epidermal surfaces of the leaflets in four taxa, the exception being Z. glabra, whose cavities occur only on the abaxial surface. Idioblasts secreting phenolic compounds were detected only in the mesophyll of Z. latifolia and Z. reticulata. The joint occurrence of mucilage epidermis with mucilage cavities seems to be of unifying value for the five taxa analyzed. The position of mucilage cavities and the occurrence of secretory idioblasts in the mesophyll of the leaflets are of diagnostic value, providing a key to enable identification of the taxa studied. This study reveals that the secretory structures provide important information in support of systematic studies of the Leguminosae. In relation to the five taxa of Zornia studied, current results suggest recognition of four species: Z. curvata, Z. glabra, Z. latifolia, and Z. reticulata.  相似文献

In flora of Turkey, there are six species of Carthamus L: Carthamus dentatus (Forssk.) Vahl., C. glaucus M. Bieb. subsp. glaucus, C. lanatus L., C. tenuis (Boiss. & Blanche) Bornm., C. persicus Desf. ex Willd. and C. tinctorius L. Within these species, C. tinctorius L. is an alien species for the Turkish flora. The pollen grains belonging to six taxa collected from different locations of Anatolia and examined both under light microscope (LM) and under scanning electron microscope (SEM). As a result of these examinations and measurements, pollen grains of taxa are radially symmetrical, isopolar, oblate-spheroid, spheroid, tricolporate rarely tetracolporate, echinate. There are high similarities among taxa but some differences are recorded in size and spin length. According to cluster analyses of Carthamus species’ pollens, C. glaucus and C. tenuis have the most similar pollens. C. dentatus grouped with these two. C. persicus placed as the closest relative of C. tinctorius in the dendrogram.  相似文献

Pollen grains of 30 taxa of the genus Salvia, belonging to sections Salvia, Horminum, Drymosphace, Plethiosphace and Hemisphace from Turkey were examined by light (LM) and scanning electron microscopy (SEM). Detailed pollen morphological characteristics are provided for these taxa. Among the studied taxa, S. verticillata subsp. verticillata from sect. Hemisphace has the smallest pollen grains, and S. pachystachys from sect. Salvia possesses the largest ones. The basic shape of the pollen grains in most taxa is suboblate, oblate-spheroidal or prolate-spheroidal. However subprolate pollen grains are recorded for S. macrochlamys from sect. Salvia. The grains are hexacolpate in all taxa, but in S. recognita from sect. Salvia also octacolpate pollen was found. Three distinct exine sculpturing types exist, reticulate-perforate (the common type), reticulate-granulate and bireticulate. The reticulate-perforate and bireticulate sculpturing patterns can be divided into subtypes based on the number of perforations and the number of secondary lumina in each primary lumen. Pollen morphological characteristics of the taxa studied are compared and discussed on the basis of taxonomical concepts. In some cases, these characters are useful in distinguishing the sections. For instance, the presence of 1-2 large central secondary lumina per primary lumen is a significant character of sect. Horminum separating it from the other sections. As well, the presence of holes on colpus membrane ornamentation can be used as a diagnostic taxonomic character for sectional division between sect. Hemisphace and others. S. ballsiana from sect. Salvia is clearly distinct from the other taxa examined by its unique pollen morphology. Further, for several macromorphologically similar taxa pollen structures provide additional evidence to delimite them from each other.  相似文献

Random amplified polymorphic DNA (RAPD) fingerprinting was used to study species boundaries in six closely related NE Turkish Lilium (Liliaceae) taxa of the section Liriotypus. The investigated taxa were L. ciliatum, L. akkusianum, L. ponticum, L. kesselringianum, L. armenum, and L. szovitsianum. Of the 108 primers screened, 11 provided polymorphic and reproducible bands. A total of 93 polymorphic bands were scored for 122 individuals from 18 populations of the six Lilium taxa and principle coordinate analysis and neighbour-joining cluster analysis based on these RAPD profiles were performed. The results demonstrate a clear distinction between the two species L. ciliatum and L. akkusianum, and the other four species. While populations of the two species groups are found to be allopatrically distributed, the two species groups overlap in their geographical ranges. Analysis of molecular variance (AMOVA) indicated that nearly half of the total molecular variance is found within the individual populations and that the molecular variance among species is as high as the variance within the individual species, indicating that genetic differentiation of the species is rather weak.  相似文献

Delimitation of sections is controversial within the genus Cytisus L. (Fabaceae, Genisteae). A morphological study has been conducted on 19 taxa from sections Alburnoides, Spartopsis and Verzinum to clarify their discrimination. Thirty-five quantitative and qualitative characters were recorded on a maximum of 15 dry or living flowers per taxon. Three multiple correspondence factor analyses (MCFA) were performed on a matrix based on 22 of the 35 recorded morphological characters to (1) compare the variability within and between individuals and (2) distinguish groups among the studied taxa. MCFA showed that both flowers sampled from the same plant or different individuals could represent the morphological variability of a taxon. MCFA also clustered the 19 taxa into three groups corresponding to sections Alburnoides, Spartopsis and Verzinum as defined by Cristofolini and Troia (Taxon 44:733–746, 2006). However, floral morphology has not been sufficient to discriminate taxa within sections. A key of the three studied sections based on floral characters is given.  相似文献

Fourteen yeast isolates belonging to the Metschnikowia clade were isolated from the digestive tracts of lacewings (Neuroptera: Chrysopidae), soldier beetles and leaf beetles (Coleoptera: Cantharidae and Chrysomelidae), and a caddisfly (Trichoptera: Hydropsychidae). The insect hosts were associated with sugary substances of plants, a typical habitat for yeasts in this clade. Based on DNA sequence comparisons and phenetic characters, the yeasts were identified as Candida picachoensis, Candida pimensis, and four undescribed taxa. Among the undescribed taxa, three yeasts were distinguished from one another and from other described taxa by nucleotide differences in the ribosomal DNA repeat, which were sufficient to consider them as new species. Two of the novel yeast species are described as Metschnikowia noctiluminum (NRRL Y-27753T) and M. corniflorae spp. nov. (NRRL Y-27750T) based in part on production of needle-shaped ascospores, which are found in most Metschnikowia species. Sexual reproduction was not observed in the third new yeast, Candida chrysomelidarum sp. nov. (NRRL Y-27749T). A fourth isolate, NRRL Y-27752, was not significantly distinct from Metschnikowia viticola and Candida kofuensis to be described as a new species. Phylogenetic analysis of the D1/D2 loop sequences placed M. noctiluminum within the M. viticola clade, while C. chrysomelidarum was a sister taxon of Candida rancensis. Metschnikowia corniflorae was phylogenetically distinct from other new species and fell outside of the large-spored Metschnikowia group.  相似文献
